Abstract
본 발명은 살리드로사이드(salidroside) 및 베툴린(betulin)을 유효성분으로 포함하는 유방암 예방 또는 치료를 위한 조성물에 관한 것으로, 본 발명의 살리드로사이드 및 베툴린가 2:1 내지 1:2의 중량비율로 혼합된 혼합물을 포함하는 조성물은 인간유래 유방암 세포주인 MCF-7의 세포증식을 억제함으로써 유방암을 예방 또는 치료하는 효과를 나타낸다. 따라서, 살리드로사이드 및 베툴린의 혼합물을 이용하여 유방암의 예방 또는 치료를 위한 약학적 조성물 및 유방암의 개선 또는 예방을 위한 기능성 식품 조성물로서 유용하게 사용될 수 있을 것으로 기대된다.
Description
본 발명은 살리드로사이드(salidroside) 및 베툴린(betulin)을 유효성분으로 포함하는 유방암 예방 또는 치료를 위한 조성물에 관한 것으로, 보다 구체적으로는 살리드로사이드 및 베툴린을 2:1 내지 1:2의 중량비율로 혼합한 혼합액을 유효성분으로 포함하는 유방암 예방 또는 치료를 위한 약학적 조성물 또는 유방암 개선 또는 예방을 위한 식품 조성물에 관한 것이다.
종양은 비정상적이고 비제어성이며 무질서한 세포증식의 산물이다. 이러한 종양이 파괴적인 성장성, 침윤성 및 전이성이 있을 때 악성종양, 즉 암으로 분류된다. 현재, 암은 주로 3가지 치료법, 즉 외과적 수술, 방사선치료 및 화학요법 중 1가지 또는 이들의 조합을 통해 치료되고 있다. 외과적 수술은 질병 조직을 대부분 제거하는 것을 포함한다. 이러한 외과적 수술은 특정 부위, 예컨대 유방, 결장 및 피부에 위치한 종양을 제거하는 데에는 효과적이지만, 척추와 같이 일부 구역에 있는 종양을 치료하거나 백혈병과 같은 분산성 종양 질환을 치료하는 데는 사용할 수 없다.
화학요법은 세포 복제 또는 세포 대사를 붕괴시키며, 흔히 유방, 폐 및 정소의 암을 치료하는데 많이 사용되는데, 종양 질병을 치료하는데 사용되는 전신성 화학요법의 부작용은 암 치료를 받는 환자들에게 가장 문제가 된다. 이러한 부작용 중에서 멀미와 구토는 가장 일반적이며 심각한 부작용이다. 화학요법에 의한 부작용은 환자의 생명에 큰 영향을 미치며, 치료에 대한 환자의 순응성을 급격하게 변화시킬 수 있다. 또한, 화학치료제와 관련된 부작용으로는 일반적으로 이러한 약물의 투여시 주의해야 하는 주용량 제한 독성(DLT)이 있다. 예를 들어, 점막염은 여러 항암제, 예컨대 항대사물질 세포 독소제인 5-플루오로우라실, 메토트렉세이트 및 항종양 항생제(예, 독소루비신) 등에 대한 주용량 제한 독성이다. 이러한 화학요법 유래의 부작용 중 대부분은 심한 경우 입원을 요하거나 통증을 치료하기 위해 진통제를 필요로 하기도 한다. 또한, 종래의 항암제들은 정상세포까지 죽이는 경우가 많았기 때문에, 최근에는 정상세포에는 작용하지 않고 암세포에만 작용할 수 있는 새로운 기전의 항암제에 대한 연구가 진행되고 있다.
유방암은 미국에서는 여성암 중 1위, 우리나라에서는 3위를 차지하는 가장 보편적인 암으로 매년 약 2만 명의 여성들이 유방암에 걸리며, 한국을 포함한 동양인에서는 발생이 다소 낮은 편이었지만, 식생활의 변화와 환경의 변화로 인해 한국인 여성에서의 발생률은 매년 증가하고 있는 추세이다. 유방암을 일으키는 여러 가지 요인으로는 연령, 이른 초경, 늦은 폐경, 신장, 폐경 후 비만, 유방암 가족력, 방사선 노출, 경궁피임약, 호르몬 대체 치료법, 유방 조직의 밀도, 유전자 변이 및 유방의 양성종양과 같은 요인들이 유방암의 발생 위험을 증가시키며 그 중에서도 식생활에 의한 유방암 발생이 전체 유방암 발생률 중 약 35%를 차지한다고 알려져 있다.
많은 연구자들은 종양 면역치료법(Cancer immunotherapy)이 전통적인 종양 치료법에 비하여 많은 장점이 있을 것으로 판단하여 면역치료법에 대한 연구가 활발해지고 있다. MCF-7 세포는 사람의 유선과 유방 암 세포로서 바닥에 붙어 자라는 부착(monolayer) 세포로서, 비교적 화학치료 및 방사선 치료에 저항성을 나타내는 유방암 세포주로 보고되었다(Kumar et al., 2007; Mougel et al., 2004).
MCF-7 세포를 활용한 논문으로는, "마늘 성분인 Diallyl Disulfide의 유방암 세포주(MCF-7)에 대한 세포사멸 유도기전에 관한 연구"(박해린 외 8인, 대한외과학회지, 2001.)와 "강황 추출물이 유방암 세포주 MCF-7 증식 억제에 미치는 영향"(조수인 외 4인, 대한본초학회지 2006.)을 들 수 있다.
이에 본 발명자들은 인간유래의 유방암 세포주인 MCF-7을 이용하여 세포증식을 억제할 수 있는 성분을 개발하기 위해 계속 연구를 진행하던 중 살리드로사이드(salidroside)와 베툴린(betulin)을 특정 중량비율로 혼합할 경우 유방암 세포주 MCF-7의 세포증식을 현저하게 억제할 수 있다는 사실을 발견함으로써 본 발명을 완성하였다.
따라서, 본 발명에서 해결하고자 하는 기술적 과제는 유방암을 예방 또는 치료할 수 있는 약학적 조성물을 제공하기 위한 것이다.
또한, 본 발명에서 해결하고자 하는 다른 기술적 과제는 유방암을 개선 또는 예방할 수 있는 식품 조성물을 제공하기 위한 것이다.
상기한 기술적 과제를 해결하기 위하여, 본 발명에서는 살리드로사이드 및 베툴린을 유효성분으로 포함하는 유방암 예방 또는 치료를 위한 약학적 조성물을 제공한다.
또한, 상기한 다른 기술적 과제를 해결하기 위하여, 본 발명에서는 살리드로사이드 및 베툴린을 유효성분으로 포함하는 유방암 개선 또는 예방을 위한 식품 조성물을 제공한다.
본 발명의 바람직한 하나의 구현예에 따르면 살리드로사이드 및 베툴린은 2:1 내지 1:2의 중량비율, 특히 1:1의 중량비율로 혼합하는 것이 바람직하다.
본 발명의 유방암 예방 또는 치료를 위한 조성물의 유효성분인 살리드로사이드는 백색분말의 성상을 가지며, 분자식 C14H20O7을 가지며, 화학적으로 합성하거나 시판하는 것을 구입하여 사용할 수 있다.
본 발명의 유방암 예방 또는 치료를 위한 조성물의 유효성분인 베툴린은 백색분말의 성상을 가지며, 분자식 C30H50O2을 가지며, 화학적으로 합성하거나 시판하는 것을 구입하여 사용할 수 있다.
본 발명에서는 인간유래의 유방암 세포주인 MCF-7을 이용하여 살리드로사이드를 시험물질 1로서 설정하고, 베툴린을 시험물질 2로서 설정하여 세포주에 대한 억제 효과를 평가한 다음 살리드로사이드 및 베툴린의 조성비를 산출하여 누드마우스를 이용한 항암시험에 적용하고자 실시하였다.
본 발명의 하나의 구체적인 실시양태에 따르면, 1차 시험에서 시험물질 1인 살리드로사이드 및 시험물질 2인 베툴린의 적용농도를 100 μg/mL을 최고농도로 설정하였고, 그 이하의 농도는 공비 2로 50, 25, 12.5, 6.25, 3.13 및 1.57 μg/mL로 설정하였다. 음성대조군은 부형제를 처리하였고 시험물질 처리 후 24시간째에 MTT assay를 실시하였다. 그 결과, 시험물질 1인 살리드로사이드의 IC20, IC50, IC70은 각각 4, 25 및 50μg/mL, 시험물질 2인 베툴린은 1.5, 4 및 15μg/mL로 산출되었다.
본 발명의 하나의 구체적인 실시양태에 따르면, 2차 시험에서는 1차 시험의 결과에 따라서 시험물질의 적용농도를 3.13, 6.25, 12.5, 25, 50 μg/mL 농도의 시험물질 1 (살리드로사이드)에 1.5 μg/mL의 시험물질 2인 베툴린을 각각 혼합하여 처리하였고, 0.94, 1.88, 3.75, 7.5, 15 μg/mL 농도의 시험물질 2 (베툴린)에 4 μg/mL의 시험물질 1인 살리드로사이드을 각각 혼합하여 처리하였다. 그 결과, 시험물질 1의 농도가 4μg/mL, 시험물질 2의 농도가 3.75 μg/mL인 혼합한 처리군에서 만족할만한 결과인 세포증식 억제율이 86.1 %로 산출되었다.
본 발명의 하나의 구체적인 실시양태에 따르면, 3차 시험에서는 2차 시험의 결과에 따라서 0.125, 0.25, 0.5, 1, 2, 4 및 8 μg/mL 농도의 시험물질 1인 살리드로사이드 및 베툴린을 각각 농도별로 혼합처리하여 MTT assay를 실시한 결과, 음성대조물질 처리 시의 세포증식 억제율을 0.0 %로 볼 때, 각각 4.9, 19.5, 34.9, 44.7, 71.5, 90.5 및 93.7%로 산출되었다. 50 % 세포증식 억제 농도를 산출한 결과, 1.09 μg/mL (살리드로사이드 : 1.09 μg/mL, 베툴린: 1.09 μg/mL)으로 산출되었다.
본 발명의 하나의 구체적인 실시양태에 따르면, 인간유래의 유방암 세포주인 MCF-7을 이용하여 시험물질 1인 살리드로사이드 및 시험물질 2인 베툴린의 세포주에 조성비를 평가한 결과, 시험물질 1과 시험물질 2의 조성비는 1:1로 판정되었으며 세포에 시험물질 1 및 시험물질 2를 1:1로 병용처리하였을 때 50 % 세포증식억제 농도는 약 1.09 μg/mL (살리드로사이드: 1.09 μg/mL, botulin: 1.09 μg/mL )로 산출되었다.
본 명세서에서 사용되는 용어 "치료"는 개체에서 (a) 질환 또는 질병의 발전의 억제 (b) 질환 또는 질병의 경감 및 (c) 질환 또는 질환의 제거를 의미한다.
본 명세서에서 사용되는 용어 "개체"는 본 발명의 상기 조성물을 투여하여 증상이 호전될 수 있는 질환을 가진 인간을 포함한 원숭이, 소, 말, 돼지, 양, 개, 고양이, 래트, 마우스, 침팬지 등의 포유동물을 의미한다.
본 발명의 살리드로사이드 및 베툴린의 혼합물은 인간유래 유방암 세포주인 MCF-7의 세포증식을 억제함으로써 유방암을 예방 또는 치료하는 효과를 나타낸다.
따라서, 본 발명의 살리드로사이드 및 베툴린의 혼합물을 유효성분으로 포함하는 조성물은 유방암을 예방 또는 치료하기 위한 용도로 유용하게 사용할 수 있다.
본 발명의 조성물에 유효성분으로 포함되는 살리드로사이드 및 베툴린의 혼합물은 전체 조성물의 총 중량을 기준으로 0.001 내지 50 중량%, 바람직하게는 0.01 내지 30 중량%, 보다 바람직하게는 0.01 내지 10 중량%의 함량으로 포함될 수 있다. 상기 살리드로사이드 및 베툴린의 혼합물의 함량이 0.001 중량% 미만일 경우 MAO-A 저해 활성이 미약하고, 50 중량%를 초과하는 경우 함량 증가에 따른 효과 증가가 비례적이지 않아 비효율적일 수 있으며, 제형상의 안정성이 확보되지 않는 문제가 있다.
본 발명의 하나의 구체적 용도로서, 본 발명의 조성물은 유방암을 예방 또는 치료하기 위한 약학적 조성물로 사용될 수 있다.
본 발명에 있어서, 약학적 조성물의 투여 경로는 이들로 한정되는 것은 아니지만 구강, 정맥내, 근육내, 동맥내, 골수내, 경막내, 심장내, 경피, 피하, 복강내, 비강내, 장관, 국소, 설하 또는 직장이 포함된다.
본 발명의 조성물이 약학적 조성물로 사용되는 경우, 유효성분으로서 살리드로사이드 및 베툴린의 혼합물 이외에 약학적으로 허용되는 담체를 추가로 포함할 수 있다. 본 발명의 약학적 조성물에 포함되는 약학적으로 허용되는 담체는 제제시에 통상적으로 이용되는 것으로서, 탄수화물류 화합물(예: 락토스, 아밀로스, 덱스트로스, 수크로스, 솔비톨, 만니톨, 전분, 셀룰로스 등), 아카시아 고무, 인산 칼슘, 알기네이트, 젤라틴, 규산 칼슘, 미세결정성 셀룰로스, 폴리비닐피롤리돈, 셀룰로스, 물, 시럽, 염 용액, 알코올, 아라비아 고무, 식물성 기름(예: 옥수수 기름, 목화 종자유, 두유, 올리브유, 코코넛유), 폴리에틸렌 글리콜, 메틸 셀룰로스, 메틸히드록시 벤조에이트, 프로필히드록시 벤조에이트, 활석, 스테아르산 마그네슘 및 미네랄 오일 등을 포함하나, 이에 한정되는 것은 아니다. 본 발명의 약학적 조성물은 상기 성분들 이외에 윤활제, 습윤제, 감미제, 향미제, 유화제, 현탁제, 보존제 등을 추가로 포함할 수 있다. 적합한 약학적으로 허용되는 담체 및 제제는 Remington's Pharmaceutical Sciences (19th ed., 1995)에 상세히 기재되어 있다.
상기 약학적 조성물은 통상의 방법에 따라 산제, 과립제, 정제, 캡슐제, 현탁액, 에멀젼, 시럽제, 에어로졸 등의 경구형 제형, 외용제, 좌제 및 멸균 주사용액 등의 형태로 제형화하여 사용될 수 있다.
상기 약학적 조성물은 유방암을 예방 또는 치료할 수 있는데, 치료방법은 상기 약학적 조성물을 약학적 유효량으로 쥐, 생쥐, 가축, 인간 등의 포유동물 내에 다양한 경로로 투여하는 것을 포함한다. 상기 투여방법은 모든 방식으로 이루어질 수 있는데, 예를 들어, 경구, 직장 또는 정맥 내 주입, 복강 내 투여, 근육 내 투여, 또는 뇌혈관내 투여 등으로 투여될 수 있다.
상기 약학적 조성물의 투여량은 제제화 방법, 투여 방식, 환자의 연령, 체중, 성별, 병적 상태 등의 요인에 따라 달라질 수 있으며, 당업자에 의하여 적절하게 선택될 수 있다. 일반적으로 본 발명의 약학적 조성물은 성인 기준으로 0.001-100 ㎎/kg의 양을 일일 1회 내지 수회로 나누어 투여할 수 있다. 다만, 상기 투여량은 어떠한 면으로든 본 발명의 범위를 한정하는 것이 아니다.
본 발명의 다른 하나의 구체적 용도로서, 본 발명의 조성물은 유방암을 예방 또는 개선하기 위한 식품 조성물로 사용될 수 있다.
본 발명에서 사용되는 용어 "개선"은 개체에서 질환 또는 질병의 증세가 호전되는 모든 행위를 의미한다.
본 명세서에서 용어 "기능식품"은 건강기능식품에 관한 법률 제6727호에 따른 인체에 유용한 기능성을 가진 원료나 성분을 사용하여 제조 및 가공한 식품을 의미하며, "기능"이라 함은 인체의 구조 및 기능에 대하여 영양소를 조절하거나 생리학적 작용 등과 같은 보건 용도에 유용한 효과를 얻을 목적으로 섭취하는 것을 의미한다.

본 발명의 식품 조성물은 상술한 성분 외에 여러가지 영양제, 비타민, 전해질, 풍미제, 착색제, 펙트산 및 그의 염, 알긴산 및 그의 염, 유기산, 보호성 콜로이드 증점제, pH 조절제, 안정화제, 방부제, 글리세린, 알콜, 탄산음료에 사용되는 탄산화제 등을 함유할 수 있다. 그 밖에 본 발명의 식품 조성물은 천연 과일쥬스, 음료 및 야채 음료의 제조를 위한 과육을 함유할 수 있다. 이러한 성분은 독립적으로 또는 조합하여 사용할 수 있다.
상기 식품의 종류에는 특별한 제한이 없다. 본 발명의 살리드로사이드 및 베툴린의 혼합물을 첨가할 수 있는 식품의 예로는 육류, 소세지, 빵, 쵸코렛, 캔디류, 스넥류, 과자류, 피자, 라면, 기타 면류, 껌류, 아이스크림류를 포함한 낙농제품, 각종 스프, 음료수, 차, 드링크제, 알콜 음료 및 비타민 복합제 등이 있으며, 통상적인 의미에서의 건강식품을 모두 포함한다.
본 발명의 식품 조성물이 드링크제로 제조되는 경우에는 본 발명의 살리드로사이드 및 베툴린의 혼합물 이외에 구연산, 액상과당, 설탕, 포도당, 초산, 사과산, 과즙 등을 추가로 포함시킬 수 있다.
또한, 본 발명의 식품 조성물은 식품 첨가물을 추가로 포함할 수 있으며, 식품 첨가물로서의 적합여부는 다른 규정이 없는 한 식품의약품안정청에 승인된 식품첨가물공전의 총칙 및 일반시험법 등에 따라 해당 품목에 관한 규격 및 기준에 의하여 판정한다.
상기 식품첨가물공전에 수재된 품목으로는 예를 들어, 케톤류, 글리신, 구연산 칼륨, 니코틴산, 계피산 등의 화학적 합성품; 결정 셀룰로오스, 구아검 등의 천연 첨가물; L-글루타민산나트륨 제제; 면류 첨가 알칼리제; 보존료제제; 타르색소제제 등의 혼합 제제류 등을 들 수 있다.
한편, 본 발명의 살리드로사이드 및 베툴린의 혼합물은 세포독성이 없는 무해한 물질이다. 따라서, 본 발명의 살리드로사이드 및 베툴린의 혼합물은 장기간 사용시에도 안심하고 사용할 수 있으며, 특히 상기한 바와 같은 약학적 또는 식품 조성물에 안전하게 사용할 수 있다.

본 명세서에 첨부되는 다음의 도면들은 본 발명의 바람직한 실시예를 예시하는 것이며, 전술한 발명의 내용과 함께 본 발명의 기술사상을 더욱 이해시키는 역할을 하는 것이므로, 본 발명은 그러한 도면에 기재된 사항에만 한정되어 해석되어서는 아니 된다.
도 1은 살리드로사이드의 인간유래 유방암 세포주 MCF-7의 세포증식 억제효과를 나타낸 것이다.
도 2는 베툴린의 인간유래 유방암 세포주 MCF-7의 세포증식 억제효과를 나타낸 것이다.
도 3은 살리드로사이드와 베툴린의 혼합비율에 따른 인간유래 유방암 세포주 MCF-7의 세포증식 억제효과를 나타낸 것이다.
도 4는 베툴린과 살리드로사이드의 혼합비율에 따른 인간유래 유방암 세포주 MCF-7의 세포증식 억제효과를 나타낸 것이다.
도 5는 살리드로사이드와 베툴린의 혼합비율에 따른 인간유래 유방암 세포주 MCF-7의 세포증식 억제효과를 나타낸 그래프이다.
이하, 본 발명의 이해를 돕기 위하여 실시예 등을 들어 상세하게 설명하기로 한다. 그러나, 본 발명에 따른 실시예들은 여러 가지 다른 형태로 변형될 수 있으며, 본 발명의 범위가 하기 실시예들에 한정되는 것으로 해석되어서는 안 된다. 본 발명의 실시예들은 당업계에서 평균적인 지식을 가진 자에게 본 발명을 보다 완전하게 설명하기 위해 제공되는 것이다.
재료 준비
본 발명에서는 인간유래 유방암 세포주 MCF-7을 한국세포주 은행으로부터 입수하여 사용하였다.
(1) Complete medium
세포계대에서 시험물질 처리 전 단계까지 사용될 배양액을 하기 표 1과 같이 조성하였다. Complete medium은 100 mL 당 FBS (fetal bovine serum), Penicillin-Streptomycin (10,000 units/mLof penicillin and 10,000 μg/mL of streptomycin)과 RPMI 1640을 혼합하여 사용하였다.
(2) Serum-free medium
시험물질 처리 단계 이후에 사용될 배양액을 하기 표 2와 같이 조성하였다.
(3) 세포주의 준비
세포를 세포배양용 플라스크에 넣고 37 ℃, 5 % CO2 incubator (MCO-20AIC, Sanyo, Japan)에서 배양한다. 배양 종료 후 세포를 수거하여 원심분리 튜브에 넣고 원심분리 (1,000 rpm, 5 분)를 실시하였다. 원심분리 후 상층액은 제거하고 pellet에 새로운 배지를 넣어 5 × 104cells/mL의 세포부유액을 만들었다.
<참조예 1> MTT assay
96 well plate에 5 × 104 cell/mL 농도로 준비된 세포부유액을 각 well에 200 μL씩 분주 (1 × 104 cells/well)하였다. 37℃, 5 % CO2 incubator에서 24 시간 배양하였다.
배양완료 후, 각 well의 배양액을 제거하였다. 시험물질의 각 농도별 조제물을 각각의 well에 200 μL씩 넣는다. 음성대조물질은 부형제인 주사용수를 처리하였다. 각 농도당 2 well씩 넣었다. 37℃, 5 % CO2 incubator에서 24 시간 배양하였다.
배양완료 후, MTT solution (3-(4,5-dimethylthiazol-2yl)-2,5-diphenyl-2H-tetrazolium bromide, 5 mg/mL, Lot No.: MKBJ2415V, Sigma-Aldrich Co., U.S.A.)을 각 well에50 μL씩 첨가하였다. 37℃, 5 % CO2 incubator에서 4 시간 배양하였다. 배양완료 후, 각 well의 배양액을 제거하였다.
각 well에 150 μL의 DMSO (Dimethyl sulfoxide) 용액을 넣어서 세포를 용해시켰다. ELISA reader를 이용하여 570 nm에서 흡광도 (optical density; OD)를 측정하였다. 음성대조군의 흡광도를 기준으로 각 농도별 시험물질을 처리한 well의 흡광도를 다음 계산식에 대입하여Inhibition rate 를 계산하였다. 세포증식 억제가 관찰되는 경우에는 회귀분석법을 이용하여 20%, 50% 및 70% 세포증식 억제농도 (IC20, IC50, IC70)를 계산하여 평가하였다.

<실시예 1> 살리드로사이드 및 베툴린의 조성비율 설정
(1) 1차 시험
시험물질 1인 살리드로사이드(salidroside) 및 시험물질 2인 베툴린(betulin)을 vial (20 mg)에 1 mL의 부형제인 DMSO를 넣고 vortex mixer로 교반하여 용해시켜 stock solution (20000 μg/mL, Stock solution)을 조제하였다. Stock solution은 공비 2로 단계 희석하여 총 7 농도 (10000, 5000, 2500, 1250, 625, 313, 157 μg/mL)로 사용시 조제하였다.
조제된 조제물은 세포에 처리하기 직전에 Serum-free medium 으로 각 농도를 100배 희석하여 조제하였다.
(2) 2차 시험
1차 시험에서 각 시험물질의 세포증식 억제율이 20 % (IC20)및 70 % (IC70)의 농도를 기준으로 하기 표 3과 같은 조성농도로 조제하였다. 즉, 시험물질 1을 기준 (IC70)으로 병용조제 시에는 시험물질 2의 IC20 농도를 고정하고, 시험물질 2를 기준 (IC70)으로 병용조제 시에는 시험물질 1의 IC20 농도를 고정하여 조제하였다.
상기에서 조제된 조제물은 세포에 처리하기 직전에 Serum-free medium 으로 각 농도를 100배 희석하여 조제하였다.
(3) 3차 시험
2차 시험에서 얻어진 조성표로 수행한 결과를 기준으로 시험물질 1의 IC20을 고정했을 때와 시험물질 2의 IC20을 고정했을 때의 조성농도 중 병용효과가 좋은 조성농도를 선택하여 하기 표 4와 같이 조제하였다.
조제된 조제물은 세포에 처리하기 직전에 Serum-free medium 으로 각 농도를 100배 희석하여 조제하였다.
<실시예 1> 1차 시험
하기 표 5와 같이 1차 시험 처리농도를 설정하고, 참조예 1에 따라 MTT assay 분석을 실시하였다.
그 결과는 하기 표 6에서 보듯이, 시험물질 1인 살리드로사이드를 0, 1.57, 3.13, 6.25, 12.5, 25, 50, 100 μg/mL 농도로 MTT assay를 실시한 결과, 음성대조물질 처리 시의 세포증식 억제농도를 0.0 %로 볼 때 각각 8.8, 13.4, 34.5, 34.7, 45.0, 74.0 및 94.6%로 나타냈다. 이때 IC20, IC50 및 IC70은 각각 4, 25 및 50μg/mL로 산출되었다. 도 1은 살리드로사이드의 인간유래 유방암 세포주 MCF-7의 세포증식 억제효과를 나타낸 것이다.
또한, 시험물질 2인 베툴린을 0, 1.57, 3.13, 6.25, 12.5, 25, 50, 100 μg/mL 농도로 MTT assay를 실시한 결과, 하기 표 7에서 보듯이, 음성대조물질 처리 시의 세포증식 억제율을 0.0 %로 볼 때 각각 19.1, 33.5, 46.9, 69.8, 79.7, 84.7, 88.7 %로 나타냈다. 이때 IC20, IC50 및 IC70은 각각 1.5, 4 및 15μg/mL로 산출되었다. 도 2는 베툴린의 인간유래 유방암 세포주 MCF-7의 세포증식 억제효과를 나타낸 것이다.
<실시예 2> 2차 시험
하기 표 8과 같이 2차 시험 처리농도를 설정하고, 참조예 1에 따라 MTT assay 분석을 실시하였다.
그 결과는 하기 표 9에서 보듯이, 음성대조물질 처리 시의 세포증식 억제율을 0.0 %로 볼 때, 3.13, 6.25, 12.5, 25, 50 μg/mL 농도의 시험물질 1 (살리드로사이드)에 1.5 μg/mL의 시험물질 2인 베툴린을 각각 혼합하여 처리했을 때의 세포증식 억제율은 각각 28.1, 49.5, 67.2, 80.1, 93.1%, 50 μg/mL 농도의 시험물질 1인 살리드로사이드의 단독처리군은 82.6 %, 1.5 μg/mL 농도의 시험물질 2인 베툴린 단독처리군은 29.7 %로 산출되었다. 도 3은 살리드로사이드와 베툴린의 혼합비율에 따른 인간유래 유방암 세포주 MCF-7의 세포증식 억제효과를 나타낸 것이다.
또한, 하기 표 10에서 보듯이, 음성대조물질 처리 시의 세포증식 억제율을 0.0 %로 볼 때, 0.94, 1.88, 3.75, 7.5, 15 μg/mL 농도의 시험물질 2 (베툴린)에 4 μg/mL의 시험물질 1인 살리드로사이드을 각각 혼합하여 처리하였을때의 세포증식 억제율은 각각 37.2, 68.4, 86.1, 94.9, 92.8 %, 15 μg/mL농도의 시험물질 2인 베툴린의 단독처리군은 76.3 %, 4 μg/mL농도의 시험물질 1인 살리드로사이드 단독처리군은 17.2 %로 산출되었다. 도 4는 베툴린과 살리드로사이드의 혼합비율에 따른 인간유래 유방암 세포주 MCF-7의 세포증식 억제효과를 나타낸 것이다.
<실시예 3> 3차 시험
하기 표 11과 같이 3차 시험 처리농도를 설정하고, 참조예 1에 따라 MTT assay 분석을 실시하였다.
그 결과 하기 표 12에서 보듯이, 음성대조물질 처리 시의 세포증식 억제율을 0.0 %로 볼 때, 0.125, 0.25, 0.5, 1, 2, 4 및 8 μg/mL 농도의 시험물질 1인 살리드로사이드 및 시험물질 2인 베툴린을 각각의 농도로 혼합처리하였을때의 세포증식 억제율은 각각 4.9, 19.5, 34.9, 44.7, 71.5, 90.5 및 93.7로 산출되었다. 50 % 세포증식 억제 농도를 산출한 결과, 1.09 μg/mL (살리드로사이드 : 1.09 μg/mL, 베툴린: 1.09 μg/mL)으로 산출되었다. 도 5는 살리드로사이드와 베툴린의 혼합비율에 따른 인간유래 유방암 세포주 MCF-7의 세포증식 억제효과를 나타낸 그래프이다.
결론적으로 인간유래의 유방암 세포주인 MCF-7을 이용하여 시험물질 1인 살리드로사이드 및 시험물질 2인 베툴린의 세포주에 조성비를 평가한 결과, 시험물질 1과 시험물질 2의 조성비는 1:1로 판정되었으며 세포에 시험물질 1 및 시험물질 2를 1:1로 병용처리하였을 때 50 % 세포증식억제 농도는 약 1.09 μg/mL (살리드로사이드: 1.09 μg/mL, botulin: 1.09 μg/mL )로 산출되었기 때문에 in vivo 항암시험을 실시시 참고하여 용량을 설정하면 될 것으로 사료된다.
<제제예 1> 산제의 제조
살리드로사이드 및 베툴린(1:1 중량비) 20 ㎎
유당 100 ㎎
탈크 10 ㎎
상기의 성분들을 혼합하고 기밀포에 충진하여 산제를 제조한다.
<제제예 2> 정제의 제조
살리드로사이드 및 베툴린(1:1 중량비) 10 ㎎
옥수수전분 100 ㎎
유당 100 ㎎
스테아린산 마그네슘 2 ㎎
상기의 성분들을 혼합한 후 통상의 정제의 제조방법에 따라서 타정하여 정제를 제조한다.
<제제예 3> 캡슐제의 제조
살리드로사이드 및 베툴린(1:1 중량비) 10 ㎎
결정성 셀룰로오스 3 ㎎
락토오스 14.8 ㎎
마그네슘 스테아레이트 0.2 ㎎
통상의 제조방법에 따라 상기의 성분을 혼합하고 젤라틴 캡슐에 충전하여 캡슐제를 제조한다.
<제제예 4> 주사제의 제조
살리드로사이드 및 베툴린(1:1 중량비) 10 ㎎
만니톨 180 ㎎
주사용 멸균 증류스 2974 ㎎
Na2HPO4, 12H20 26 ㎎
통상의 주사제의 제조방법에 따라 1 앰플당(2㎖) 상기의 성분 함량으로 제조한다.
<제제예 5> 액제의 제조
살리드로사이드 및 베툴린(1:1 중량비) 20 ㎎
이성화당 10 g
만니톨 5 g
정제수 적량
통상의 액제의 제조방법에 따라 정제수에 각각의 성분을 가하여 용해시키고 레몬향을 적량 가한 다음 상기의 성분을 혼합한 다음 정제수를 가하여 전체 100㎖로 조절한 후 갈색병에 충진하여 멸균시켜 액제를 제조한다.
<제제예 6> 건강 식품의 제조
살리드로사이드 및 베툴린(1:1 중량비) 1000 ㎎
비타민 혼합물 적량
비타민 A 아세테이트 70 ㎍
비타민 E 1.0 ㎎
비타민 B1 0.13 ㎎
비타민 B2 0.15 ㎎
비타민 B6 0.5 ㎎
비타민 B12 0.2 ㎍
비타민 C 10 ㎎
비오틴 10 ㎍
니코틴산아미드 1.7 ㎎
엽산 50 ㎍
판토텐산 칼슘 0.5 ㎎
무기질 혼합물 적량

상기의 비타민 및 혼한물의 조성비는 비교적 건강식품에 적합한 성분을 바람직한 예로 혼합 조성하였지만, 그 배합비를 임의로 변형실시 하여도 무방하며, 통상의 건강식품제조방법에 따라 상기의 성분을 혼합한 다음, 과립을 제조하고, 통상의 방법에 따라 건강식품 조성물 제조에 사용할 수 있다.
<제제예 7> 건강 음료의 제조
살리드로사이드 및 베툴린(1:1 중량비) 1000 ㎎
구연산 1000 ㎎
올리고당 100 ㎎
매실농축액 2 g
타우린 1 g
정제수를 가하여 전체 900㎖
통상의 건강음료 제조방법에 따라 상기의 성분을 혼합한 다음, 약 1 시간동안 85℃에서 교반 가열한 후, 만들어진 용약을 여과하여 멸균된 2ℓ용기에 취득하여 밀봉 멸균한 뒤 냉장 보관한 다음 본 발명의 건강음료 조성물 제조에 사용한다.
본 발명의 살리드로사이드 및 베툴린의 혼합물은 인간유래 유방암 세포주인 MCF-7의 세포증식을 억제함으로써 유방암을 예방 또는 치료하는 효과를 나타낸다. 따라서, 살리드로사이드 및 베툴린의 혼합물을 이용하여 유방암의 예방 또는 치료를 위한 약학적 조성물 및 유방암의 개선 또는 예방을 위한 기능성 식품 조성물로서 유용하게 사용될 수 있을 것으로 기대된다.
Claims (7)
- 살리드로사이드(salidroside) 및 베툴린(betulin)을 유효성분으로 포함하는 것을 특징으로 하는 유방암의 예방 또는 치료를 위한 약학적 조성물.
- 제 1 항에 있어서,상기 살리드로사이드(salidroside) 및 베툴린(betulin)를 2:1 내지 1:2의 중량비율로 포함하는 것을 특징으로 하는 유방암의 예방 또는 치료를 위한 약학적 조성물.
- 제 2 항에 있어서,상기 살리드로사이드(salidroside) 및 베툴린(betulin)를 1:1의 중량비율로 포함하는 것을 특징으로 하는 유방암의 예방 또는 치료를 위한 약학적 조성물.
- 제 1 항에 있어서,상기 살리드로사이드(salidroside) 및 베툴린(betulin)를 조성물의 총 중량을 기준으로 하여 0.001 내지 50 중량%의 양으로 포함하는 것을 특징으로 하는 유방암의 예방 또는 치료를 위한 약학적 조성물.
- 살리드로사이드(salidroside) 및 베툴린(betulin)을 유효성분으로 포함하는 것을 특징으로 하는 유방암의 개선 또는 예방을 위한 식품 조성물.
- 제 5 항에 있어서,상기 살리드로사이드(salidroside) 및 베툴린(betulin)를 2:1 내지 1:2의 중량비율로 포함하는 것을 특징으로 하는 유방암의 개선 또는 예방을 위한 식품 조성물.
- 제 6 항에 있어서,상기 살리드로사이드(salidroside) 및 베툴린(betulin)를 1:1의 중량비율로 포함하는 것을 특징으로 하는 유방암의 개선 또는 예방을 위한 식품 조성물.
