본 발명은 모새나무(Vaccinium
bracteatum Thunb.) 추출물을 유효성분으로 포함하는 우울증 예방 및 개선용 건강 기능성 식품 조성물 또는 약학적 조성물에 관한 것으로서, 보다 구체적으로는 천연원료인 모새나무 열매 추출물을 이용하여 독성 및 부작용 없이 안전하게 사용될 수 있는 우울증 예방 및 개선용 약학조성물 및 건강식품 조성물에 관한 것이다.
우울증은 의욕 저하와 우울감을 주요 증상으로 하여 다양한 인지 및 정신 신체적 증상을 일으켜 일상 기능의 저하를 가져오는 질환을 말한다. 우울장애는 감정, 생각, 신체 상태, 그리고 행동 등에 변화를 일으키는 심각한 질환으로 한 개인의 전반적인 삶에 영향을 준다. 우울증은 일시적인 우울감과는 다르며 개인적인 약함의 표현이거나 의지로 없앨 수 있는 것이 아니다.
우울장애는 매우 흔한 정신질환의 하나로, 국가에 따른 비교에서 유병률의 차이를 많이 보인다. 미국이나 유럽, 뉴질랜드 등은 주요 우울장애 평생 유병률이 10.1%~16.6%로 높은 수준을 보이는데 비하여, 한국이나 중국을 비롯한 비서구권국가에서는 5% 이하의 낮은 수준의 유병률을 보인다.
2011년 보건복지부의 정신질환상태 역학조사에서는 주요우울장애 평생유병률이 6.7%, 일년유병률이 3.1%로서 2006년 역학연구에 비하여 다소 높은 수준의 유병률을 보이나, 서구권 국가에 비하여 낮은 수준이며, 비서구권 국가들과는 비슷한 수준이거나 다소 높은 수준이었다. 우울증의 분명한 원인에 대해서는 아직 명확하지 않으나 다른 정신 질환과 같이 다양한 생화학적, 유전적 그리고 환경적 요인이 우울증을 야기할 수 있다. 우울증은 다른 질환과는 달리 전문가의 적절한 치료를 받는다면 상당한 호전을 기대할 수 있고 이전의 정상적인 생활로 돌아가는 것이 가능하다.
한편 모새나무(Vaccinium
bracteatum Thunb.) 는 진달래목의 쌍떡잎 속씨식물로 해변의 산지에서 자란다. 높이가 1~3m로, 작은가지는 회갈색에서 회색이고 거의 털이 없다. 잎은 어긋나고 두꺼우며 타원형 또는 긴 타원형으로 두꺼운 가죽같은 질감을 갖는다. 가장자리에 잔 톱니가 있으며 뒷면 밑에 작은 선점(腺點)이 있으며, 꽃은 6월에 피고 홍백색이 돌며 밑으로 처지는 총상꽃차례에 종처럼 생긴 꽃이 10여 개씩 달리고 포(苞)가 남아 있다. 열매는 장과로 둥글고 흰가루로 덮이며 지름 6mm 정도로 10월에 익으며 먹을 수 있다.
최근 현대인의 생활수준이 향상됨에 따라 부작용이 거의 없고 자연에서 채취가 가능한 천연자원에 대해서 관심이 높아지고 있다. 특히, 인체에 필요한 영양소를 우수하게 함유하거나 질병의 예방과 회복에 특이하게 도움을 주는 천연 기능성물질을 함유한 건강식품에 대한 연구가 활발히 진행됨에 따라 모새나무 또한 그 기능에 대한 조사가 실시되고 있으나, 아직까지는 영양학적 가치나 생리활성 기능성에 대한 관련정보가 부족한 실정이다.
본 발명은 생리활성기능에 대해서는 확실하게 알려지지 않은 천연자원인 모새나무의 기능성 식품 조성물 및 약학조성물을 제공하기 위해, 모새나무 열매로부터 유효성분을 추출하여 독성 및 부작용 없이 안전하게 사용될 수 있는 모새나무 열매 추출물을 유효성분으로 포함하는 우울증 예방 및 개선용 건강 기능성 식품조성물 및 약학 조성물을 제공하고자 한다.
본 발명은 우리나라 천연자원인 모새나무 열매 추출물을 이용하여 독성 및 부작용 없이 안전하게 사용될 수 있는 우울증 예방 및 개선용 건강 기능성 식품조성물 및 약학 조성물을 제공한다. 상기 목적을 달성하기 위하여, 본 발명은 모새나무 열매추출물을 유효성분으로 포함하는 우울증 예방 및 개선용 건강 기능성 식품조성물 및 약학 조성물을 제공한다.
우울증 예방 및 개선 기능을 갖는 약학조성물 및 기능성 식품 조성물의 추출방법은 모새나무 열매에 증류수를 가하고, 환류추출기를 이용하여 100 ℃에서 온도가 올라간 후 3시간 동안 가열, 추출하였다.
상기 물, 메탄올, 에탄올, 프로판올, 이소프로판올, 부탄올 또는 이들의 혼합용매 중 어느 하나에서 가용한 추출물을 유효성분으로 포함하는 추출물은 0.01 내지 99.9 중량%의 양으로 포함하는 우울증 예방 및 개선용 건강 기능성 식품조성물 및 약학 조성물을 제공한다.
상기 조성물은 정제 및 캡슐제, 연질 캡슐제, 과립제, 액제 또는 음료 중에서 선택된 하나 이상의 형태로 제조될 수 있다.
본원발명의 모새나무 열매 추출물은 사지를 전혀 움직이지 않는 부동시간을 감소시켜 항우울 효과가 비교군인 에스시탈로프람 옥살산염보다 뛰어남을 확인하였으며, 동물실험에서 모새나무 열매 추출물, 에스시탈로프람 옥살산염을 투여했을 때, 모새나무 열매 추출물 투여군에서 대조군보다 노르에피네프린 (노르아드레날린)의 농도를 증가시키는 경향성을 보임으로서 모새나무 열매 추출물은 노르에피네프린 (노르아드레날린) 농도를 증가시켜 항우울제제로서의 효과이 있다는 것을 확인하였다.

1. 모새나무 열매 열수 추출물의 제조
도 1는 모새나무 열매 추출물을 얻는 과정을 나타낸다. 모새나무 열매 2.0 kg을 증류수 30L를 가하고, 환류추출기를 이용하여 100 ℃에서 온도가 올라간 후 3시간동안 가열, 추출 하였다. 얻어진 추출물을 감압 필터 후 감압 농축하여 107.02 g의 농축액을 얻었다.
2. 모새나무 열매의 극성 용매, 비극성 용매 가용 분획물 제조
도 2에 도시된 바와 같이 상기 모새나무 열매 열수 추출물로부터 계통분획을 실시하여 모새나무 열매 추출물의 분획물 (15 g)을 제조하는 단계를 구체적으로 설명하면 다음과 같다. 농축액을 물로 충분히 현탁하여 2배 부피의 헥산으로 1차 용매이행을 3회 수행하여 물 층과 hexane ext. (41.8 mg) 로 분리하였다. 물 층을 다시 2배 부피의 클로로포름으로 2차 용매이행을 3회 수행하여 CHCl3 ext. (79.9 mg)를 확보하였다. 에틸아세테이트로 3차 용매이행을 3회 수행하여 EtOAc ext. (312.4 mg) 를 확보하였다. 마지막으로 n-부탄올로 3차 용매이행을 3회수행하여 n-BuOH ext. (5g)을 확보하였다.
3. 실험동물 및 사육
모새나무 추출물의 항우울 효과 측정을 위한 실험동물로서 생후 6주령 된 수컷 ICR 마우스를 ㈜샘타코 (SAMTACO, Korea)부터 구입하여 동물 사육실에서 일전한 조건 (온도: 22±2℃, 습도:50±5%, 명암: 12시간 light/dark cycle)으로 일주일간 적응시킨 후 사용하였다.
4. 모새나무 추출물을 이용한 항우울 동물행동실험
수컷 ICR 마우스에 본 발명의 모새나무 추출물을 경구투여 하였으며, 비교군으로는 항우울제로 사용되는 에스시탈로프람 옥살산염을 경구투여하였다. 이들은 실험에 따라 6일 투여하였다.
약물투여 전 예비실험 (pre-swim)으로 원통모양의 수조 (직경 20 cm, 높이 40 cm)에 25℃ 내외의 수돗물을 실린더 바닥에서부터 15 cm까지 부은 후, 생쥐를 집어넣고 15분간 강제로 수영을 시키고, 물에서 건져 올려 마른 수건으로 닦고 사육상자로 돌려보냈다. 약물 투여 6일 후에 본 시험 (post-swim)을 시행하였으며, 실린더에 생쥐를 넣고 6분 동안 강제 수영을 시행하면서 비디오 촬영을 하였다. 기록된 영상에서 최초 1분간을 제외하고, 나머지 5분 동안 동물의 행동을 3가지로 분류하여 분석하였는데, 동물이 얼굴을 포함한 상체의 일부분만 수면 위로 드러낸 채 약간의 움직이만 나타낼 뿐 물위에 떠 있는 행동 (부동자세, immobility behavior), 실린더 주위를 수평방향으로 움직이면서 앞발과 뒷발을 물장구를 치는 행동 (수영, swimming behavior), 벽을 향해 앞발을 물 밖으로 차올리는 다소 격렬하게 움직이면서 벽을 긁는 행동 (기어오르기, climbing behavior)으로 구분하여 각각의 시간을 측정하였다.
도 3은 본 발명의 모새나무 열매 추출물을 6일간 투여한 흰쥐에 대한 강제수영시험법에 있어서, 부동 시간 (immobility time), 수영 시간 (swimming time), 기어오른 시간 (climbing time)을 대조군, 에스시탈로프람 옥살산염 투여군 생쥐와 비교한 그래프를 나타낸다. 도 3 에 도시된 바와 같이, 각 약물을 6일 투여 후 강제 수영시키는 동안 부동시간은 대조군 (245.48±4.05초) 과 비교하여 볼 때 비교군인 에스시탈로프람 옥살산염은 169.65±7.15초, 모새나무 열매 추출물 (100 및 200mg/kg p.o)은 각각 114.98±9.26초, 118.29±6.35초로 유의성 있는 감소를 보였다 (P < 0.01 and P < 0.001). 그리고 수영 시간은 대조군 (51.82±3.38초)과 비교하여 볼 때 비교군인 에스시탈로프람 옥살산염은 128.16±6.65초, 모새나무 열매 추출물 (100 및 200mg/kg p.o)은 각각 177.07±9.79초, 172.23±6.72초로 유의성 있게 증가를 보였다 (P < 0.01 and P < 0.001).
상기에서 살펴본 바와 같이, 본 발명의 모새나무 열매 추출물은 사지를 전혀 움직이지 않는 부동시간을 감소시키므로 항우울 효과가 비교군인 에스시탈로프람 옥살산염보다 뛰어남을 확인하였다.
5. 모새나무 추출물을 이용한 혈장 내 노르에피네프린 (norepinephrine, NE) 및 코르티코스테론 (corticosterone, COR) 측정
정상적인 상태에서 우울증과 관련된 신경전달물질인 NE와 스트레스 호르몬인 COR 의 농도 변화에 미치는 영향은 각 약물 6일 투여 후 생쥐로부터 혈장 (plasma)를 분리하여 Abnova ELISA kit를 사용하여 제조사의 지시에 따라 측정하였다.
도 4은 본 발명의 모새나무 열매 추출물을 투여한 생쥐에 대한 혈액에서 신경전달물질의 증가유무에 있어서 대조군, 에스시탈로프람 옥살산염 투여군 생쥐와 비교한 그래프를 나타낸다. 정상적인 상태에서 NE와 COR 의 변동에 미치는 모새나무 열매 추출물의 항우울 효능을 검증하였다. 우울한 기분이 느껴지는 이유는 뇌의 신경세포에서 외부로 분비되는 세로토닌 또는 노르에피네프린 (노르아드레날린)의 양이 적기 때문이다. 모새나무 열매 추출물을 통한 노르에피네프린 신경전달물질의 증가유무를 확인한 결과, 도 4에 (A) 에 도시된 바와 같이, 정상동물에게 모새나무 열매 추출물 (100 및 200 mg/kg), 에스시탈로프람 옥살산염 (10mg/kg)을 투여했을 때, 모새나무 열매 추출물 100 및 200 mg/kg 투여군에서 대조군보다 NE의 농도를 증가시키는 경향성을 보였다. 따라서 모새나무 열매 추출물은 NE 농도를 증가시킴으로 항우울제제로서의 효능이 있다는 것을 의미한다.
또한, 도 4의 (B)에 도시된 바와 같이, 모새나무 열매 추출물 (100 및 200 mg/kg) 투여군은 스트레스 호르몬인 코르티코스테론 수치가 대조군과 비교하여 볼 때 유의성 있게 감소하였고, 대조군에 비해 비교군인 에스시탈로프람 옥살산염은 감소 경향은 보였으나 유의성은 없었다. 따라서 스트레스를 느낄 때 코르티코스테론의 수치가 증가한다고 알려져 있는데, 모새나무 열매 추출물이 스트레스성 우울증 모델에서도 항우울효과를 나타낼 것을 시사한다.
6. 모새나무 추출물을 이용한 뇌신경세포에서 스트레스 완화 활성 확인
SH-SY5Y cell (neuroblastoma, human dopaminergic neuronal cell)을 1% antimicotics/antibiotics와 10% FBS를 함유한 MEM 배지에서 배양하고 96-well plate에 세포수가 105 cell/㎖가 되도록 seeding 한 후 24시간 배양하였다.
모새나무 열매 추출물이 신경세포에 미치는 세포독성을 확인하기 위하여 농도별로 처리하였다. 모새나무열매 추출물 처리 24시간 후, MTT assay로 세포 생존율을 측정하였다.
도 5은 본 발명의 모새나무 열매 추출물이 SH-SY5Y 세포 (neuroblastoma, human dopaminergic neuronal cell) 에 미치는 세포독성 정도를 나타내는 그래프를 나타낸다.
세포독성정도는 모새나무 열매 추출물의 cytotoxicity를 MTT를 이용하여 측정하여 사용농도의 최대안전범위를 설정하였다. 사용 세포주는 SH-SY5Y 세포로서 모새나무 열매 열수 추출물 농도 1, 3, 10, 30, 100 μg/ml 로 처리하여 세포독성 확인을 수행하였으며, 1, 3, 10, 30 μg/ml 에서는 control (0 μg/ml)과 비교하여 유의적인 차이가 발견되지 않았다. 하지만 100 μg/ml 농도에서는 control 군과 비교하여 유의적 차이는 보이지 않았지만 세포 생존도가 감소하였다. 본 실험의 결과로 30 μg/ml 내에서 실험을 수행하였다
모새나무 열매 추출물이 산화적 스트레스로 인한 신경세포 사멸에 미치는 효과를 확인하기 위하여 먼저, 과산화수소 사용 농도결정을 위하여, 과산화수소 (H2O2)을 농도별로 24시간 처리하여 세포독성을 확인하여 50-60% 세포독성을 보이는 농도를 결정하여 실험에 사용하였다. 모새나무열매 추출물의 신경세포 보호효과 확인실험은 실험 세포주(SH-SY5Y cell line)를 96well microplate에 분주하여 24시간 배양한 다음, 모새나무열매 추출물을 2시간 처리하고 10 μM H2O2을 처리하여 24시간 배양시켰다. 24시간 후, 세포 생존율 (cell viability)는 MTT assay를 이용하여 측정하였다.
도 6은 과산화수소 처리를 통한 산화적 스트레스 유도에 대한 모새나무열매 추출물의 신경세포 보호효과를 나타낸 그래프를 나타낸다. 도 6A은 과산화수소 사용농도 결정을 위하여 사용 세포주인 SH-SY5Y 세포에 H2O2 농도별로 처리하여 세포독성 확인한 결과, 10 μM 에서는 control (0 μg/ml)과 비교하여 세포 생존도가 50-60% 유의적 세포독성을 보이므로 실험에 사용하였다. 도 6B 는 과산화수소 (H2O2) 10 μM 과 함께, 모새나무 열매의 열수추출물을 처리 하였을 때, 농도 0.3, 1, 3, 10 μg/ml에서 각각 54.75 ±2.85 %, 52.18 ±5.05 %, 69.01 ±1.48 %, 80.24 ±1.14 % 로 세포생존도가 유의적으로 증가한 것을 확인하였다 (P<0.05 또는 P<0.01).
모새나무 추출물의 정신적 스트레스에 대한 저항 활성을 세포수준에서 확인하기 위하여 스트레스 호르몬인 코르티코스테론이 유발하는 신경세포 손상에 보호효과를 확인하기 위하여 실험하였다. 코르티코스테론 (corticosterone)을 농도별로 24시간 처리하여 세포독성을 확인하여 50-60% 세포독성을 보이는 농도를 결정하여 실험에 사용하였다. 모새나무열매 추출물의 보호효과실험은 실험 세포주(SH-SY5Y cell line)를 96well microplate에 분주하여 24시간 배양한 다음, 모새나무열매 추출물을 2시간 처리하고 1mM 코르티코스테론을 처리하여 24시간 배양시켰다. 24시간 후, 세포 생존율 (cell viability)는 MTT assay를 이용하여 측정하였다.
도 7은 정신적 스트레스 호르몬인 Corticosterone 처리를 통한 세포 내 스트레스 유발 및 모새나무열매 추출물의 농도에 따른 신경세포 보호효과를 나타낸 그래프를 나타낸다. 스트레스 호르몬인 코르티코스테론 (corticosterine )의 사용 농도를 결정하기 위하여, SH-SY5Y 세포에 corticosterone 농도 50, 100, 300, 500, 1000 μM 로 처리하여 세포독성 확인을 수행하였으며, 300, 500, 1000 μM 에서는 control (0 μg/ml)과 비교하여 세포 생존도가 감소하였으며, 50-60% 세포독성을 보이는 1mM을 실험에 사용하였다. 도 7B 는 corticosterone과 함께, 모새나무 열매의 열수추출물을 처리 하였을 때, 농도 0.3, 1, 3, 10 μg/ml에서 각각 63.28 ±0.42 %, 66.55 ±2.21 %, 86.91 ±4.27 %, 89.08 ±5.47 % 로 세포생존도가 유의적으로 증가한 것을 확인하였다 (P<0.05 또는 P<0.01).
7. 모새나무 추출물을 이용한 5-HT1A 수용체 또는 5-HT2A 수용체 유전자 발현된 세포주에서 세포내 Ca2
+ 활성 측정
CHO-K1 세포주 (Chinese Hamster Ovary Cell) 를 ATCC로부터 구입하고 배양액 (RPMI 1640, fetal bovine serum 10%, penicilin 100 IU/ml, streptomycine 100 μg/ml)을 사용하여 96-well black wall/clear bottom (BD Falcon)에 세포를 분주한 후 37℃, 5% CO2 incubator에서 24시간 배양하였다. 세로토닌 1A 수용체 (5-HT1A receptor) 또는 세로토닌 2A 수용체 (5-HT2A receptor) 각각 cDNA를 플라스미드 트랜스펙션 시약인 lipofectamine 2000을 이용하여 48시간동안 세포내 일시적으로 발현시킨 세포를 실험에 사용하였다.
형광염색 전에 세포는 HEPES-buffered solution (155 mM NaCl, 2mM CaCl2, 1mM MgCl2, 3 mM KCl, 10 mM HEPES, 10 mM Glucose, pH 7.4) 으로 1회 세척한 다음, 세포내 칼슘농도를 측정하기 위한 형광염료로 Fura-2/AM을 최종 5 μM 농도로 처리하여 37℃, 5% CO2 incubator에서 빛이 차단된 상태에서 60분간 반응시켰다. 60분 후 HEPES-buffered solution로 1회 세척한 다음 세포내 칼슘농도 측정은 high-throughput system (HTS) system을 이용하여 340/380nm 빛이 선택적으로 노출되도록 한 다음, 디지털 형광 분석기에 의해 분석된 340/380nm ratio 값을 측정하였다. 세포내에 존재하는 Fura-2/AM은 Ca2
+과 결합한 상태에서는 340 nm에서 excitation되고, 결합하지 않은 상태에서는 380 nm에서 excitation되므로, 340/380 nm ratio값의 증가는 세포내 Ca2
+과 결합한 fura-2/AM이 많아졌다는 의미가 된다.
세로토닌 (5-HT) 100 μM을 처리하여 얻은 값을 control 로 사용하였고, 모새나무 열매 추출물을 1, 3, 10, 30, 100 μg/ml 농도로 1분간 전 처리한 후 5-HT 100 μM을 처리하여 세포내 칼슘양의 변화를 확인하였다. 참고로 도 8와 도 9에서 각 항목의 에러바 위에 표기된 숫자는 실험에 사용된 세포의 수를 의미한다.
도 8은 본 발명의 모새나무 열매 추출물이 5-HT1A 수용체 유전자가 발현된 세포주에서 5-HT 에 의해 유도된 세포내 Ca2+의 증가를 농도 의존적으로 증가시키는 효과를 나타낸 그래프를 나타낸다. 모새나무 열매 추출물이 5-HT1A 수용체에서 세로토닌에 의해 유도되는 세포내 Ca2
+ 의 증가에서 세로토닌과 비교하여 세포내 Ca2+을 활성화시켜 5-HT1A 수용체의 부분적인 효현제작용제로써 효과를 확인하였다. 도 8에 나타낸 바와 같이 세로토닌 100 μM을 처리한 control 보다 모새나무 열매 추출물 1, 3, 10, 30, 100 μg/ml에서 각각 10.31±6.86%, 47.45±1.45%, 62.73±2.37%, 67.08±1.36%, 88.09±3.50% 로 5-HT1A 수용체-특이적 세로토닌에 의해 유도된 세포내 Ca2
+ 이 농도 의존적으로 활성화되는 것을 확인하였다.
8. 모새나무의 용매분획물을 이용한 5-HT2A 수용체 유전자 발현된 세포주에서 세포내 Ca2
+ 활성 측정

세로토닌 (5-HT) 100 μM을 처리하여 얻은 값을 control 로 사용하였고, 모새나무 열매 추출물의 헥산, 클로로포름, 에틸아세테이트, 부탄올 및 물 분획물을 10 μg/ml 농도로 1분간 전 처리한 후 5-HT 100 μM을 처리하여 세포내 칼슘양의 변화를 확인하였다.
도 9는 본 발명의 모새나무 열매 추출물이 5-HT2A 수용체 유전자가 발현된 세포주에서 5-HT 에 의해 유도된 세포내 Ca2+의 증가를 농도 의존적으로 억제하는 효과를 나타낸 그래프를 나타낸다. 도 9에 의하면 모새나무 열매 추출물이 5-HT2A 수용체에서 세로토닌에 의해 유도되는 세포내 Ca2
+ 의 증가에서 세로토닌과 비교하여 세포내 Ca2
+을 농도 의존적으로 억제되는 것을 확인할 수 있었다. 도 9에 나타낸바와 같이 세로토닌 100 μM을 처리한 control 보다 모새나무 열매 추출물 1, 3, 10, 30, 100 μg/ml에서 각각 29.15±0.81%, 37.53±0.49%, 42.09±0.92%, 54.39±0.55%, 59.03±0.48% 로 5-HT2A 수용체-특이적 세로토닌이 유도하는 세포내 Ca2
+ 농도를 억제하였다.
도 10은 본 발명의 모새나무 열매 추출물의 용매분획물이 5-HT2A 수용체 유전자가 발현된 세포주에서 5-HT 에 의해 유도된 세포내 Ca2+의 증가를 억제하는 효과를 나타낸 그래프를 나타낸다. 도 10에서 각 항목의 에러바 위에 표기된 숫자는 실험에 사용된 세포의 수를 의미한다. 도 10에 의하면 모새나무 열매 추출물의 용매 분획물에 대한 5-HT2A 수용체에서 세로토닌에 의해 유도되는 세포내 Ca2
+ 의 증가에서 세로토닌과 비교하여 세포내 Ca2
+을 농도 억제활성에 대한 결과를 나타낸다. 도 10에 나타낸 바와 같이 모새나무 열매 추출물의 용매분획물의 헥산, 클로로포름, 에틸아세테이트, 부탄올 및 물 분획물 중 에틸아세테이트 및 부탄올 분획물이 5-HT2A 수용체에서 세로토닌에 의해 유도되는 세포내 Ca2
+ 증가에 대한 저해활성이 가장 높았다.
9. 모새나무 열매 추출물을 이용한 약제 또는 건강식품 제조
모새나무 열매 추출물을 유효성분으로 하는 면역 증강용 약학적 조성물 또는 건강식품 조성물은 정제 및 캡슐제, 연질 캡슐제, 과립제, 액제 형태로 제조될 수 있다. 또 다른 적절한 실시 형태에 따르면, 상기 조성물은 음료 첨가제로 제조할 수 있다. 상기 면역 증강용 약제 또는 건강식품은 상기 모새나무 열매 추출물을 유효성분으로 포함하는 조성물이 0.01 내지 99.9중량%로 포함되도록 산제, 과립제, 정제, 캡슐제, 현탁액, 에멀젼, 시럽, 에어로졸, 경피제, 좌제 또는 멸균 주사용 액으로 제형화하여 제조할 수 있다.
멸균주사액의 경우, 상기 약학 조성물이 0.01 내지 99.9중량%로 포함하도록 하고 정제수 또는 포도당을 99.9 내지 0.01 중량%를 혼합하여 제조 가능하며, 캡슐제의 경우, 상기 약학조성물을 동결 건조하여 0.01 내지 99.9중량%로 포함하도록 하고, 비타민제, 칼슘제을 99.9 내지 0.01 중량%를 혼합하여 제조 가능하다.
상기 제조된 약학조성물의 1일당 투여량은 상기 추출물이 10 내지 2000mg/kg 체중 포함되는 함량으로 제공하며, 또한 상기 약학 조성물을 0.01 내지 99.9중량%로 포함하는 우울증 증상 예방 또는 개선용 건강기능성 식품으로도 제조 가능하다.
본 발명은 모새나무(Vaccinium
bracteatum Thunb.) 열매 추출물을 유효성분으로 포함하는 우울증 원인과 증상 질환의 개선을 위한 건강기능성 식품조성물을 제공함으로써, 제조 원료를 자연에 서식하는 식물로 대체함으로 제조생산단가 절감과 산업화를 통한 수입대체 및 수출효과를 기대할 수 있어 산업상 이용가능성이 있다.

- 제1항의 건강기능식품 조성물이 0.01 내지 99.9 중량%의 양으로 포함되는 것을 특징으로 하는 우울증 예방 또는 개선용 건강기능성 식품.
- 제3항에 있어서, 건강기능성 식품은 정제, 캡슐제, 연질 캡슐제, 과립제, 액제 중에서 선택되는 어느 하나의 형태로 제조되는 것을 특징으로 하는 우울증 예방 또는 개선용 건강기능성 식품.
