Abstract
효소 필름 및 그를 포함하는 고감도 및 선택성을 갖는 바이오 센서에 관한 것으로, 기존의 적층 방식이 아닌, 효소와 전극 전체를 하이드로다이나믹 공정을 이용함으로써 효소 일체형 필름을 구현할 수 있을 뿐만 아니라, 스티커를 접착하듯이 타겟 전극 상에 용이하게 목적하는 형태로 전사시킬 수 있는 효과가 있다. 또한, 상기 효소 필름을 포함하는 바이오 센서는 전기 화학적인 활성도가 높고, DET 기반의 시료 내 분석물의 검출이 가능한 효과가 있다.
Description
효소 필름 및 그를 포함하는 고감도 및 선택성을 갖는 바이오 센서에 관한 것이다.
삶의 질 향상과 의료 기술의 발달로 인해 인간의 기대 수명은 꾸준히 증가하고 있는 추세이며 노령 인구 증가에 따른 건강 상태의 자가 진단에 대한 관심 및 투자가 활발히 이루어지고 있다.
자가 진단을 위한 바이오 센서 분야에서 침습형으로 피를 뽑아내는 방식이 아닌 환자에게 착용된 센서가 스스로 환자의 체액 (침, 땀, 눈물 등)을 이용하여 글루코오스 농도를 지속적으로 측정하고 이를 분석한 정보가 무선통신 등을 통해 외부 기기로 전달 가능한 웨어러블 바이오 센서(wearable biosensor)가 주목을 받고 있다. 눈물 등의 체액상에 있는 글루코오스 농도는 일반 혈액상의 농도보다 10~20배 낮기 때문에, 낮은 농도의 글루코오스 레벨을 효과적으로 측정하는 웨어러블 바이오 센서를 개발하기 위해서는 혈액상의 글루코오스 농도를 측정하는 센서보다 높은 감도를 지닌 센서가 요구된다. 또한, 사람의 신체에 부착되어야 하기 때문에 패터닝이 가능한 물질을 이용하여 센서의 크기를 대폭 축소하여야 하며, 휘어짐 (flexibility), 투명도 (transparency), 표면 부착성 (surface adhesion) 측면에서도 특성이 향상되어야 한다.
향상된 특성을 갖는 바이오 센서를 제작하기 위한 노력으로서, 센서 표면 상에 고정된 시약의 산화 환원 상태의 변화를 매개체 없이 직접적으로 감지하는 3세대 바이오 센서 (3rd generation biosensor)가 주목을 받고 있다. 3세대 바이오 센서 개념을 이용한 글루코오스 센서의 경우 직접적 전자 이동(Direct electron transfer, DET)이 일어남에 따라 방해물질인 AA 와 UA를 효과적으로 배제할 수 있다. 또한, 글루코오스 옥시다아제가 글루코오스와 일어나는 반응을 어떠한 매개체 없이 직접적으로 DET에 의해 바로 전극상으로 전달할 수 있기 때문에 센서의 효과나 정확도 면에서도 발전될 수 있다.
그러나, 기존에 보고된 바이오 센서의 경우, 복잡한 공정을 요구하는 나노 물질 등을 이용하여 센서 플랫폼을 구축하여야 하기 때문에, 재현성이 떨어지고, 센서의 감도 조절이 어려워 센서의 제작 공정에 많은 비용을 필요로 한다. 이뿐만이 아니라, 전극을 형성시킨 후 적층 공정을 사용하여 폴리머 전해질을 기판에 코팅한 후 효소를 그 위에 적층하는 방식으로 구현되었다. 이 경우, 패터닝된 기판 상에 전해질 코팅 공정과 효소 적하 공정을 진행하였으므로, 이들을 국소화시키는 것이 어렵고 공정이 복잡한 단점이 있었다. 즉, 층의 개념(layer-by-layer)으로 효소 등을 전사하여야 했기 때문에, 패턴이 형성된 마스크를 사용하여 이들을 대면적으로 전사하는 것이 어려웠다.
이에 본 발명자들은 상기의 문제점을 해결하기 위하여 노력한 결과, 효소 전극 전체를 하이드로다이나믹 공정을 이용함으로써, 효소 일체형의 전극을 프리스탠딩한 필름 형태로 구현하고, 이것이 효소로부터 전자를 직접 주고 받는 제3 세대 바이오 센서에 유용하게 사용가능하고, 전극 내 효소의 안정성이 우수한 것을 확인함으로써 본 발명을 완성하였다.
일 양상은 효소와 전극 전체를 하이드로다이나믹 공정을 이용해서 구현한 효소 필름 및 그를 제조하는 방법을 제공하는 것이다.
또 다른 양상은 전사하고자 하는 전극 상에 용이하게 목적하는 형태로 전사시킬 수 있는 상기 효소 필름과 전사용 필름을 포함하는 효소 스티커 및 그를 사용하여 효소를 포함하는 전극을 제조하는 방법을 제공하는 것이다.
다른 양상은 상기 효소 필름을 포함하는 바이오 센서 및 그를 포함하는 생체 정보 검출용 웨어러블 디바이스를 제공하는 것이다.
일 양상은 그래피틱 물질; 상기 그래피틱 물질에 결합된, 상기 그래피틱 물질에 결합능을 갖는 펩티드 또는 파지; 및 효소를 포함하는 효소 필름을 제공한다.
본 명세서에서 용어 "필름"이라 함은 일정한 폭과 두께를 갖는 물질을 의미할 수 있으며, 예를 들면, 시트, 웹, 막, 또는 이들의 복합 구성체를 포함하는 개념으로 이해될 수 있다.
또한 본 명세서에서 용어 "효소 필름" 또는 "효소 전극 필름"이라 함은 전극에 효소가 일체형으로 결합된 필름, 즉 효소 일체형 필름을 의미할 수 있다. 본 명세서에서 "효소 일체형"은 센서의 전극의 구성에 있어서, 효소 또는 효소를 전극 상에 고정화하기 위한 물질, 그를 보호하기 위한 보호 물질 등이, 레이어바이레이어(layer-by-layer)로 적층되는 것과는 구별된 개념으로, 효소 자체가 전극과 하나의 복합체로서 존재하는 것을 의미할 수 있다. 따라서, 상기 효소 필름은 상기 효소가 그래피틱 물질 상에 단순 적층된 것이 아닐 수 있다. 상세하게는 상기 효소는 상기 그래피틱 물질의 네트워크 구조 상에 내에 및/또는 아래에 형성된 것일 수 있다. 예를 들면, 상기 그래피틱 물질의 네트워크 구조에 일체형으로 효소를 형성시키는 것은 투석 고정에 의한 것일 수 있다. 또한, 예를 들면, 상기 그래피틱 물질은 네트워크 구조를 포함할 수 있고, 상기 효소는 상기 네트워크 구조 상에, 상기 네트워크 구조 내에 및/또는 상기 네트워크 구조 아래에 포함되어 있는 것일 수 있다. 또한, 상기 네트워크 구조는 상기 그래피틱 물질과 상기 펩티드의 결합 복합체 또는 상기 그래피틱 물질과 상기 파지의 결합 복합체를 통해 이루어진 것일 수 있다. 따라서, 상기 그래피틱 물질의 내부 구조는 퍼콜레이트 네트워크(percolated network) 구조를 갖는 것일 수 있다. 본 발명에서 용어 "퍼콜레이트 네트워크(percolated network)"는 무작위적 전도성 또는 비전도성 연결로 구성된 격자 구조를 의미할 수 있다.
일 구체예에 있어서, 상기 그래피틱 물질과 결합하는 펩티드 또는 파지는 바이오 접착제로서의 역할을 수행하는 것일 수 있다. 본 명세서에서 "바이오 접착제"는 상기 펩티드 또는 파지가 그래피틱 물질 간의 결합을 촉진하여 그래피틱 물질의 구조화/안정화에 기여하는 것을 의미할 수 있다. 즉, 이는 그래피틱 물질의 기능화와 구별된 개념으로써, 그래피틱 물질의 상기의 네트워크 구조의 형성을 가능하게 하는 것을 의미할 수 있다. 또한, 상기 그래피틱 물질은 전하가 없어 바이오 센서 등을 위한 효소를 부착하기 위해서 별도의 전해질을 코팅하여야 하나, 그래피틱 물질에 결합능을 갖는 펩티드 또는 파지는 전하를 가지므로 별도의 전해질의 코팅이 없이도 효소가 그래피틱 물질의 네트워크 구조 내에 일체형으로 결합될 수 있다. 따라서, 상기 파지 또는 펩티드는 복수의 그래피틱 물질의 접합(junction)을 형성하는 것일 수 있다. 또한, 상기 펩티드는 두 개의 펩티드가 링커로 연결되어 두 개의 그래피틱 물질을 서로 연결하는 것일 수 있다. 상세하게는 복수의 그래피틱 물질의 네트워크 구조 내에서 상기 링커로 연결된 두 개의 펩티드는 각각 하나의 그래피틱 물질과 결합하는 것일 수 있다. 상기 링커는 펩티드 링커일 수 있다. 상기 펩티드 링커는 당업계에 공지된 다양한 링커를 이용할 수 있으며, 예를 들어, 복수의 아미노산으로 이루어진 링커일 수 있다. 일 구체예에 따르면, 상기 링커는 예를 들어, 1 내지 10개 또는 2 내지 8개의 임의의 아미노산으로 이루어진 폴리펩티드일 수 있다. 상기 펩티드 링커는 Gly, Asn 및 Ser 잔기를 포함할 수 있으며, Thr 및 Ala과 같은 중성 아미노산들도 포함될 수 있다. 펩티드 링커에 적합한 아미노산 서열은 당업계에 공지되어 있다.

본 명세서에서 용어 "그래피틱 물질(graphitic materials)"은, 탄소원자가 육각형 모양으로 배열되어 있는 표면, 즉 그래피틱 표면(graphitic surface)을 지니는 물질을 의미할 수 있으며, 그래피틱 표면을 포함하는 물질이라면 물리적, 화학적 성질, 구조적 특성에 상관없이 그래피틱 물질에 포함될 수 있다. 상기 그래피틱 물질은 예를 들면 그래핀 시트, 고배향성 열분해흑연(HOPG) 시트, 단겹 탄소나노튜브(Single-walled carbon nanotube) 및 이겹 탄소나노튜브(double-walled carbon nanotubes), 다겹탄소나노튜브(multi-walled carbon nanotube) 등의 탄소나노튜브 또는 풀러린(fullerene)을 포함할 수 있다. 상기 그래피틱 물질은 금속성, 반도체성 혹은 혼성되어 있는 물질일 수 있으며, 예를 들면 그래핀 시트 및 단겹 탄소나노튜브의 혼합물을 사용할 수 있다.

상기 그래피틱 물질에 결합하는 펩티드는 펩티드의 라이브러리를 통해 선별될 수 있으며, 예를 들면, 파지 디스플레이 기법을 통해 선별될 수 있다. 파지 디스플레이 기법을 통해 펩티드가 유전적으로 파지의 외피 단백질에 연결, 삽입 또는 치환되어 파지의 외부에 디스플레이되고, 펩티드는 비리온 내의 유전 정보에 의해 암호화될 수 있다. 디스플레이된 단백질 및 그를 암호화하는 DNA에 의해 다양한 변이체의 단백질을 스크리닝하여 선별할 수 있으며, 그를 "바이오패닝(biopanning)"이라고 부른다. 요약하여 바이오패닝 기법은 다양한 변이체가 디스플레이된 파지를 고정화된 타겟(예를 들면, 그래피틱 물질)과 반응시키고, 결합하지 않은 파지를 세척한 후, 파지와 타겟 사이의 결합 상호작용을 파괴하여 특이적으로 결합된 파지를 용리(elution)하는 방법을 포함한다. 용리된 파지의 일부는 DNA 시퀀싱 및 펩티드 식별을 위하여 남겨두고, 나머지는 인 비보(in vivo)상에서 증폭하고 다음 라운드를 위한 서브 라이브러리를 만들어 상기 과정을 반복할 수 있다.

이하 본 발명을 실시예를 통하여 보다 상세하게 설명한다. 그러나, 이들 실시예는 본 발명을 예시적으로 설명하기 위한 것으로 본 발명의 범위가 이들 실시예에 한정되는 것은 아니다.
[제조예 1] 파지를 이용한 GOx 엔자임 필름의 제조
본 발명의 일 구현예로서, GOx 엔자임 필름을 하기의 방법으로 제조하였다.
콜로이드 용액의 제조
먼저, 증류수에 계면 활성제로 소듐 콜레이트(sodium-cholate)를 2% w/v의 농도로 첨가한 수용액을 제조한 다음, 그래피틱 물질로 탄소나노튜브(제조사:Nanointegris, SuperPure SWNTs, solution 형태, 농도: 250 μg/ml)를 48시간 동안 투석하여 단겹탄소나노튜브를 소듐 콜레이트로 안정화시켜 콜로이드 용액을 제조하였다.
이때 탄소나노튜브(CNT)의 평균 길이를 1 μm, 평균 지름을 1.4 nm로 가정했을 때, 상기 콜로이드 용액에 포함된 단겹탄소나노튜브 개수의 계산식은 다음과 같다.

단겹탄소나노튜브 개수(개/ml) = 농도 ug/ml X 3 x 1011 CNT
상기 수학식에 의하면 상기 콜로이드 용액에 포함된 단겹탄소나노튜브 개수는 (7.5X1013 )개/ml임을 알 수 있다.
파지의 제조
그래피틱 표면과 강한 결합력을 지닌 M13 파지인, 그래피틱 표면과 강한 결합력을 갖는 펩티드인 SWAADIP(서열번호 5)가 디스플레이된 M13파지 (GP1) 및 NPIQAVP(서열번호 6)가 디스플레이된 파지(GP2)를 하기의 방법으로 제조하였다.
우선, M13KE 벡터(NEB, product#N0316S, 서열번호 13)의 1381번째 염기쌍(base pair)인 C를 G로 부위특이적 변이(site-directed mutation)하여 M13HK 벡터를 제작하였다.
이때 상기 M13KE 벡터(NEB, product#N0316S)는 7222bp의 DNA로 이루어진 클로닝 벡터(Cloning vector M13KE)로서, 유전자 정보는 인터넷 사이트 (https://www.neb.com/~/media/NebUs/Page%20Images/Tools%20and%20Resources/Interactive%20Tools/DNA%20Sequences%20and%20Maps/Text%20Documents/m13kegbk.txt)에 개시되어 있다. 그리고 상기 부위특이적 변이에 사용한 올리고뉴클레오티드(oligonucleotides)의 염기서열은 다음과 같다:
5'-AAG GCC GCT TTT GCG GGA TCC TCA CCC TCA GCA GCG AAA GA-3'(서열번호 14), 및
5'-TCT TTC GCT GCT GAG GGT GAG GAT CCC GCA AAA GCG GCC TT-3'(서열번호 15).
상기 제작된 M13HK 벡터에 제한효소 BspHI (NEB, product# R0517S) 및 BamHI 제한효소(NEB, product#R3136T)를 이용하여 파지 디스플레이 p8 펩티드 라이브러리를 제작하였다.
본 발명의 일 실시예로서 상기 파지 디스플레이 p8 펩티드 라이브러리의 제조에 사용된 올리고뉴클리오티드의 염기서열은 다음과 같다:
5'- TTA ATG GAA ACT TCC TCA TGA AAA AGT CTT TAG TCC TCA AAG CCT CTG TAG CCG TTG CTA CCC TCG TTC CGA TGC TGT CTT TCG CTG CTG -3'(서열번호 16), 및
5'- AAG GCC GCT TTT GCG GGA TCC NNM NNM NNM NNM NNM NNM NNM NCA GCA GCG AAA GAC AGC ATC GGA ACG AGG GTA GCA ACG GCT ACA GAG GCT TT -3'(서열번호 17).
상기 제조된 파지 디스플레이 p8 펩티드 라이브러리의 염기서열은 4.8 X 107 pfu(plaque form unit) 가지의 다양성을 가지며 각각의 서열당 1.3 X 105 개 정도의 복제수(copy number)를 지녔다.
그 다음, 상기 제조된 파지 디스플레이 p8 펩티드 라이브러리를 바이오 패닝(bio-panning) 방법에 의해 그래피틱 표면에 결합시켜 본 발명의 바이오 물질로 사용할 펩티드가 디스플레이 된 파지를 선별하였다. 바이오 패닝 방법은 구체적으로 하기와 같다.
먼저, 그래피틱 표면을 갖는 물질인 HOPG(highly oriented pyrolytic graphite, SPI, product#439HP-AB)를 실험 전에 테이프로 떼어내어 깨끗한(fresh) 표면을 얻어 샘플 표면의 산화 등으로 인한 결함을 최소화시켰다. 이때 HOPG 기판은 입자크기(grain size)가 100?m 이하의 비교적 큰 HOPG 기판을 사용하였다.
그리고 상기 제조한 4.8 X 1010 가지(4.8 X 107 가지 다양성, 각 서열마다 복제수 1000개)의 파지 디스플레이 p8 펩티드 라이브러리를 100 μl의 TBS(Tris-Buffered Saline) 완충액에 준비한 다음 HOPG 표면과 1시간 동안 100rpm으로 진탕배양기(shaking incubator)에서 반응(conjugating)시켰다. 1 시간 후 용액을 제거한 다음, TBS에서 10번 반복 세척(washing)하였다. 세척된 HOPG 표면에 산성 완충액으로서 pH 2.2의 Tris-HCl을 8분 동안 반응시켜 약하게 반응하는 펩티드를 용리(elution)한 후, 미드-로그(mid-log) 상태인 XL-1 blue E. coli 배양물(culture)을 30분 동안 용리시켰다. 용리된 배양물의 일부는 DNA 시퀀싱(DNA sequencing) 및 펩티드 식별(peptide identification)을 위하여 남겨두고 나머지는 증폭(amplification)해서 다음 라운드를 위한 서브 라이브러리(sub-library)를 만들었다. 이때 만들어진 서브 라이브러리를 이용하여 상기의 과정을 반복하였다. 한편, 남겨둔 플라크(plaque)는 DNA를 분석하여 p8 펩티드 서열을 구하고, 이때 얻어진 서열을 분석하여 그래피틱 물질에 강한 결합능을 갖는 펩티드 서열인 SWAADIP(서열번호 5)이 디스플레이 된 파지(GP1) 및 NPIQAVP(서열번호 6)가 디스플레이된 파지(GP2)를 얻었다.
파지를 이용하는 GOx 엔자임 필름의 제조
상기 제조예 1에서 제조한 콜로이드 용액과 상기 그래피틱 물질의 표면과 강한 결합력을 지닌 M13 파지(GP1)를 포함하는 파지용액을 32:4의 몰비율로 혼합하였다.
그 다음, 투석을 위하여 반투과성 멤브레인(semipermeable dialysis membrane; SpectrumLab, MWCO 12,000~14,000, product # 132 700) 튜브 안에 상기 혼합물을 각각 넣은 다음, 상기 각 멤브레인 튜브를 상기 3차 증류수에 대하여 12시간 투석을 진행하였다. 그 다음, 1 mL의 1%의 polyethyleneimine(PEI)을 멤브레인 튜브 내에 첨가하여 2차 투석 공정을 12시간 동안 진행하였다. 그 다음, 250 mg/mL의 GOx의 효소를 1mL 추가적으로 첨가하여 3차 투석공정을 12시간 동안 진행하였다. 3차 투석 공정을 완료하여 반투과성 멤브레인을 따라서 GOx 엔자임 필름을 형성시켰다.
도 1은 형성된 GOx 엔자임 필름을 투석용 멤브레인에서 떼어낸 사진 및 그의 주사 현미경 사진을 나타낸 것이다. 상기 공정을 통해서 제작된 엔자임 필름은 약 5 x 5 cm 의 크기를 가지고, 엔자임 필름의 주사현미경 사진을 보면, 엔자임 필름에 효소가 균일하게 잘 incorporation 되어 있음을 알 수 있다.
[비교예 1] 일반적 효소 전극의 제조
상기의 파지를 이용한 엔자임 필름을 제조하는데 사용되는 조건과 같은 조건으로 GOx 엔자임과 SWNT를 준비하고 혼합한 후 24 시간 인큐베이션하여 물리적으로 흡착 시킨 후, 파지와 혼합하여 3차 증류수에 대하여 투석 공정을 진행하였고, 그 결과를 도 2에 나타내었다.
도 2는 비교예에 따른 효소 전극의 제조 결과를 나타낸 도면이다.
도 2에서 나타낸 바와 같이 엔자임 필름이 성공적으로 형성되지 않았음을 확인할 수 있다. 이는 SWNT 표면과 GOx 간의 물리적 흡착을 통하여 GOx가 SWNT의 그래피틱 표면을 덮었기 때문이다. 즉, 바이오 접착제인 M13 파지가 π-π 결합을 통하여 대면적 전자시트 합성을 진행하여야 하는 SWNT의 부위를 GOx가 이미 확보하여, M13 파지와 SWNT간의 효율적 π-π 결합이 저하됨으로 인해 대면적 필름의 형성이 제한된 것이다.
[제조예 2] 파지 유래 펩티드를 이용한 GOx 엔자임 필름의 제조
그래피틱 표면과 특이적으로 결합하는 파지 유래 펩티드를 화학적으로 합성하여 (㈜ 펩트론사) 1 mg/mL 농도로 준비하였다. 파지 유래 펩티드는 구체적으로 3가지를 사용하였다. 펩티드 서열번호 9(ADSWAADIPDPA), 두 개의 펩티드 서열번호 9를 링커로 연결한 펩티드 서열번호 10, 및 파지의 외피 단백질의 일부 서열(KAA)을 더 포함하는 두 개의 펩티드를 링커로 연결한 펩티드 서열번호 11을 사용하였다. 두 개의 펩티드 서열을 링커로 연결하여 각각의 펩티드가 하나의 그래피틱 물질에 결합하고 결과적으로, 두 개의 그래피틱 물질을 연결할 수 있게 하였다. 준비된 상기 서열번호 10의 펩티드 800 μL와 상기 제조예 1에 기재된 방법으로 준비한 800 μL의 SWNT (3.0 × 1014 개/mL) 콜로이드 용액을 혼합한 후 반투과성 멤브레인(semipermeable dialysis membrane; SpectrumLab, MWCO 12,000~14,000, product # 132 700) 튜브에 넣고 3차 증류수에 대하여 12시간 동안 1차 투석을 진행하였다. 그 다음, 1 mL의 1%의 polyethyleneimine(PEI)을 멤브레인 튜브 내에 첨가하여 2차 투석 공정을 12 시간 동안 진행하였다. 그 다음, 1 mL의 250 mg/mL의 GOx를 추가적으로 멤브레인 튜브 내에 첨가하여 3차 투석공정을 12 시간 동안 진행하였다. 3차 투석 공정을 완료하여 반투과성 멤브레인을 따라서 GOx 엔자임 필름을 형성시켰다.
도 3은 형성된 GOx 엔자임 필름을 멤브레인에서 떼어낸 사진을 나타낸 것이다. 도 3에 나타낸 바와 같이, 제조예 2에 따른 공정을 통해서 제작된 GOx 엔자임 필름이 대면적으로 형성됨을 알 수 있다.
[제조예 3] 파지 유래가 아닌 펩티드를 이용한 GOx 엔자임 필름의 제조
그래피틱 표면과 효율적으로 결합하는 것으로 알려진 펩티드 (YYACAYY)(서열번호 12)를 상기 제조예 2와 동일한 방법으로 펩티드를 합성하여 상기 제조예 2와 동일한 방법으로 GOx 엔자임 필름을 형성시켰다.
도 4는 형성된 GOx 엔자임 필름을 멤브레인에서 떼어낸 사진을 나타낸 것이다. 도 4에 나타낸 바와 같이, 제조예 3에 따른 공정을 통해서 제작된 GOx 엔자임 필름이 대면적으로 형성됨을 알 수 있다.
[제조예 4] 파지를 이용한 LOx 엔자임 필름의 제조
상기 제조예 1에 기재한 바와 같이 제조한 콜로이드 용액과 상기 그래피틱 표면과 강한 결합력을 지닌 M13 파지(GP1)를 포함하는 파지용액을 32:4 의 몰비율로 800 μL의 SWNT (3.0 × 1014 개/mL)와 혼합하였다. 그 다음 GOx 대신에 1 mL의 50 mg/mL의 LOx를 사용한 것만을 제외하고는 제조예 1과 동일한 방법으로 LOx 엔자임 필름을 제조하였다.
도 5는 형성된 LOx 엔자임 필름을 멤브레인에서 떼어낸 사진 및 그의 주사 현미경사진을 나타낸 것이다. 도 5에 나타낸 바와 같이, 제조예 5에 따른 공정을 통해서 제작된 LOx 엔자임 필름은 대면적이며, 효소 필름의 주사현미경 사진을 통하여 엔자임 필름에 효소가 균일하게 잘 incorporation 되어 있음을 알 수 있다.
[제조예 5] 파지를 이용한 HRP 엔자임 필름의 제조
LOx 대신에 1 mL의 125 mg/mL의 HRP(horseradish peroxidase)를 사용한 것만을 제외하고는 상기 제조예 4와 동일한 방법으로 HRP 엔자임 필름을 제조하였다.
도 6은 형성된 HRP 엔자임 필름을 멤브레인에서 떼어낸 사진 및 그의 주사 현미경 사진을 나타낸 것이다. 도 6에 나타낸 바와 같이, 제조예 5에 따른 공정을 통해서 제작된 HRP 엔자임 필름은 대면적이며, 효소 필름의 주사현미경 사진을 통하여 엔자임 필름에 효소가 균일하게 잘 incorporation 되어 있음을 알 수 있다.
[제조예 6] 파지를 이용한 catalase 엔자임 필름의 제조
LOx 대신에 1 mL의 250 mg/mL의 catalase를 사용한 것만을 제외하고는 상기 제조예 4와 동일한 방법으로 catalase 엔자임 필름을 제조하였다.
도 7은 형성된 catalase 엔자임 필름을 멤브레인에서 떼어낸 사진 및 그의 주사 현미경 사진을 나타낸 것이다. 도 7에 나타낸 바와 같이, 제조예 6에 따른 공정을 통해서 제작된 catalase 엔자임 필름은 대면적이며, 효소 필름의 주사현미경 사진을 통하여 엔자임 필름에 효소가 균일하게 잘 incorporation 되어 있음을 알 수 있다.
[제조예 7] 파지를 이용한 laccase 엔자임 필름의 제조
LOx 대신에 1 mL의 250 mg/mL의 laccase를 사용한 것만을 제외하고는 상기 제조예 4와 동일한 방법으로 laccase 엔자임 필름을 제조하였다.
도 8은 형성된 laccase 엔자임 필름을 멤브레인에서 떼어낸 사진 및 그의 주사 현미경 사진을 나타낸 것이다. 도 8에 나타낸 바와 같이, 제조예 8에 따른 공정을 통해서 제작된 laccase 엔자임 필름은 대면적이며, 효소 필름의 주사현미경 사진을 통하여 엔자임 필름에 효소가 균일하게 잘 incorporation 되어 있음을 알 수 있다.
[제조예 8] 엔자임 스티커의 제조
상기의 제조예 1 내지 7에서 형성된 엔자임 필름을 포함하는 멤브레인 튜브 내의 용액을 제거한 후, 엔자임 필름을 멤브레인에서 떼어내지 않고, 멤브레인 상에서 건조시켜 엔자임 스티커를 제조하였고, 그 결과를 도 9에 나타내었다.
도 9에 나타낸 바와 같이, 엔자임 스티커는 약 20~30 μm 두께의 멤브레인 상에 약 1 μm 두께의 엔자임 필름이 건조된 상태로 존재하기 때문에 아주 얇은 두께를 가지며, 높은 유연성을 가짐을 알 수 있다.
[제조예 9] 엔자임 스티커를 이용한 제 3세대 글루코스 센서의 제조
상기의 제조예 8에서 제조된 엔자임 스티커를 원하는 모양으로 자른 후 물에 약 1분간 적신 후 원하는 전극 상에 전사시킨다. 그 다음, 핀셋 등으로 멤브레인을 제거하고 상온에서 필름을 건조시켜 원하는 전극 상에 효소 전극을 구현하였다. 즉, 엔자임 필름을 스티커를 접착하듯이 타겟 전극 상에 용이하게 목적하는 형태로 전사시킬 수 있었다.
상세하게는, 도 10에 나타낸 바와 같이, GOx 엔자임 스티커를 사무용 천공기를 사용하여 5 mm 지름의 원모양으로 제작한 후 1분간 물에 적신 후 SPE 전극상에 원하는 위치에 전사시켰다. 그 다음, 멤브레인 막을 핀셋을 사용하여 제거한 후 상온 건조시켜 GOx 엔자임 필름이 집적된 GOx 바이오 센서를 제작하였다.
[제조예 10] 두 종류 이상의 엔자임 스티커를 이용한 다중분석 바이오 센서의 제조
제조예 8에서 제조된 GOx 엔자임 스티커와 LOx 엔자임 스티커를 사무용 천공기를 사용하여 5 mm 지름의 원모양으로 제작하였다. 우선 GOx 엔자임 스티커를 1분간 물에 적신 후 SPE 전극상에 원하는 위치에 전사시켰다. 그 다음, 핀셋 등으로 멤브레인을 제거하고 상온에서 필름을 건조시켰다. 그 다음, 같은 방법으로 LOx 엔자임 스티커를 이용하여 원하는 다른 SPE 전극 상에 LOx 엔자임 전극을 구현하였고, 그 결과를 도 11에 나타내었다. 도 11은 다중 분석용 바이오 센서의 사진을 나타낸다.
[실험예 1] GOx 엔자임 필름의 전기화학적 활성도 측정
제조예 1에서 제조한 GOx 엔자임 필름을 멤브레인에서 떼어 낸 후 스텐실 마스크 공법을 통하여 상용적으로 구매가 가능한 금 전극 (SPE 250BT, DropSens)에 전사하였다. 이후, 10 mM K3(Fe[CN]6) (244023, Sigma Aldrich)가 포함된 10 mM PBS buffer (pH = 7.4, 79383, Sigma Aldrich) 용액에서 성능 실험을 진행하였다. 전압 주사 범위는 -0.2 ~ 0.6 V이며, 전압 주사 속도(scan rate)는 200 mV/s으로 유지하였고, 그 결과를 도 12에 나타내었다.
도 12에 나타난 바와 같이, 엔자임 필름을 포함한 SPE 250BT 금 전극에서 측정된 10 mM K3(Fe[CN]6) (244023, Sigma Aldrich)의 산화/환원 전류는 엔자임 필름이 사용되지 않은 금 전극에 비해 약 73% 증가 하였으며, quasi-reversible potential 이 약 181 mV 감소한 것으로 나타났다. 상기의 결과로 비파괴적으로 합성된 엔자임 필름은 높은 전기화학적 활성을 갖는 SWNT를 기반으로 하기 때문에 높은 전기 화학적 반응성을 가짐을 알 수 있다.
또한, GOx 엔자임 필름을 포함하는 전극을 10 mM PBS buffer (pH = 7.4, 79383, Sigma Aldrich) 용액에서 -0.6 V ~ -0.2 V의 음의 전압으로 200 mV/s의 속도로 전압을 주사하였고, 그 결과를 도 13에 나타내었다.
도 13에 나타난 바와 같이, GOx 엔자임 필름을 포함하는 전극은 Ag/AgCl 레퍼런스 전극 (3 M KCl saturated, PAR, K0260) 대비 약 -400 mV 범위에서 강한 산화 환원 피크가 Cyclic voltammetry (CV) 상에 나타남을 알 수 있다. 이는 GOx 내의 산화환원 센터 (FAD redox center)가 SWNT 전자시트와 효율적/직접적으로 전기적 쌍을 형성하기 때문에 직접적인 전자 주고 받음(Direct-Electron-Transfer, DET)이 아래 반응식과 같이 일어났음을 의미한다.
FAD + 2H+ + 2e- -> FADH2
또한, 주사 속도를 20 mV/s에서 순차적으로 4000 mV/s까지 변화시키며 CV를 측정하였고, 그 결과를 도 14에 나타내었다.
도 14에 나타난 바와 같이, 20 mV/s에서 순차적으로 4000 mV/s까지 전압 주사 속도를 증가 시켰을 때도 산화 환원 전류가 직선적으로 증가함을 보이며, 200 mV/s 과 400 mV/s의 50 사이클의 고속 반복 주행에서도, 효소의 이탈 및 소재의 degradation 없이 안정하게 소재가 구동됨을 보임으로서, 엔자임 필름이 수용액 상에서도 높은 안정성을 보임을 알 수 있다. 이러한 반응을 바탕으로 엔자임 필름에 존재하는 엔자임이 전극과 효율적으로 전자를 직접 주고 받을 수 있음을 알 수 있다.
[실험예 2] GOx 엔자임 필름의 글루코스에 대한 반응성 평가 1
제조예 1에서 제조한 GOx 엔자임 필름을 포함하는 금전극 (SPE 250BT, DropSens)을 10 μM 내지 1000 μM의 글루코스가 포함된 10 mM PBS buffer (pH = 7.4, 79383, Sigma Aldrich) 용액 상에서 200 mV/s의 속도로 전압을 주사하면서 CV를 측정하였고, 그 결과를 도 15에 나타내었다.
도 15에 나타난 바와 같이, -0.2 V에서 -0.6 V사이로 전압을 인가한 상태에서, 10mM PBS buffer 상에 포함된 글루코스 농도를 증가시켰을 때, 환원 전류가 직선적으로 양의 방향으로 증가하는 형태를 보임을 알 수 있다. 이렇게 측정된 글루코스 센서의 감도는 약 ~67 μA / mM cm2로 측정되었다. 상기의 결과로, DET 기반의 GOx 엔자임 필름은 비칩습적 방식으로 채취가 가능한 사람의 체액 (땀, 눈물, 침 등) 내에 포함되어 있는 글루코스 농도 범위 (100 μM ~ 500 μM) 에서 환원 전류의 범위가 높은 민감도를 가지고 직선적으로 변하기 때문에, 3세대 DET 기반의 웨어러블 바이오 센서의 전자 소재로서 사용가능함을 알 수 있다.
[실험예 3] GOx 엔자임 필름의 글루코스에 대한 반응성 평가 2
제조예 1에서 제조한 GOx 엔자임 필름의 글루코스 반응에 대한 선택성에 대하여 평가하였다. 구체적으로, 1mM의 글루코스만 첨가된 시약 및, 2 mM의 아스코르브산(Ascorbic Acid, AA), 요산 (Uric Acid, UA), 아세트아미노펜(Acetaminophen,AP)의 방해물질도 첨가된 시약을 사용하여 -0.6 V에서 0.6 V까지 200 mV/s의 주사 속도로 센서를 구동하였다. 또한, 10 mM PBS, 10 mM PBS 중 10 mM 글루코스, PBS 중 인간 혈청(~11 mM), 및 인공 눈물 중 10 mM 글루코스에 대해서도 상기 실험예 2와 동일하게 CV를 측정하였고, 그 결과를 도 16에 나타내었다.
도 16에 나타난 바와 같이, 일 구체예에 따른 GOx 엔자임 필름 기반의 3세대 바이오 센서는 방해물질 첨가 시에 양의 전압에서 강한 산화 전류가 측정 됨에도 불구하고, 음의 전압 영역에서는 그 변화가 최소화 되어 AA, UA, AP 등의 방해물질의 첨가 유무와 관계 없이, 안정적인 구동을 보임을 알 수 있다. 따라서 GOx 엔자임 필름 기반의 3세대 바이오 센서가 마이크로몰 농도의 낮은 농도에서 높은 감도를 보일 뿐만 아니라 전기 화학적 활성을 갖는 방해 물질이 존재 하였을 때에도 높은 선택성을 지님을 알 수 있다. 또한, 일 구체예에 따른 센서는 10 mM PBS 용액에서뿐만 아니라, 인간 혈청 또는 인공 눈물 등 실제 생리학적 환경에서도 안정적으로 구동함을 보임으로써, 침/눈물/땀 등의 비침습적 방식으로 채집이 가능한 생채 용액에서 구동이 가능함을 알 수 있다.
[실험예 4] GOx 엔자임 필름 기반 바이오 센서의 균일도 평가
제조예 1에서 제조된 GOx 엔자임 필름을 도 17의 왼쪽 패널에 나타난 바와 같이 대략적으로 6부분으로 나누어 6개의 SPE 250BT 금 전극상에 각각 전사하여 6개의 독립적인 바이오 센서를 제조하고 그 특성을 측정하였다. 센서의 측정은 0 mM 의 글루코스가 포함된 10 mM PBS 용액과 1 mM 의 글루코스가 포함된 용액 내에서 -0.6 V ~ -0.2 V 사이로 전압을 인가하여 환원 전류의 변화량을 측정하였고, 그 결과를 도 17의 오른쪽 패널에 나타내었다. 그 결과. 도 17에 나타낸 바와 같이, 6개의 샘플 모두 약 ~65 μA / mM cm2의 감도를 보였으며, 최대 감도와 최소 감도를 보인 샘플의 차이가 약 8% 이내인 것으로 보아, 대면적으로 형성된 GOx 엔자임 필름이 높은 균일성을 보이는 것을 알 수 있다.
[실험예 5] GOx 엔자임 필름 기반 바이오 센서의 안정성 평가
제조예 1에서 제조된 GOx 엔자임 필름을 10 mM PBS 버퍼 용액 내에서 4 ℃ 냉장 보관하여, 간헐적으로 SPE 250BT 금 전극에 전사 하여 30일간의 감도 변화를 측정 하였다. 센서의 측정은 0 mM 의 글루코스가 포함된 10 mM PBS 용액과 1 mM 의 글루코스가 포함된 10mM PBS 용액상에서 -0.6 V ~ -0.2 V 사이로 전압을 인가하여 환원 전류의 변화량을 측정하였고, 그 결과를 도 18에 나타내었다.
도 18에 나타낸 바와 같이, 30일간 약 4%의 감도 저하를 보임으로 일 구체예에 따른 GOx 엔자임 필름 기반 바이오 센서는 높은 안정성을 보임을 확인할 수 있었다.
[실험예 6] 파지 유래 펩티드를 이용하여 제조한 GOx 엔자임 필름의 글루코스 반응성 평가
제조예 2에서 제조한 GOx 엔자임 필름에 대해 실험예 2와 동일한 방법으로 CV를 측정하였고, 그 결과를 도 19에 나타내었다.
도 19에 나타낸 바와 같이, Ag/AgCl 레퍼런스 전극 대비 약 -400 mV 범위에서 강한 산화 환원 피크가 Cyclic voltammetry (CV) 상에 나타남을 알 수 있다. 상기의 결과는 GOx 내의 산화환원 센터 (FAD redox center)가 파지 유래 펩티드를 이용하여 제조한 엔자임 필름 내에서도 SWNT와 직접적인 전자 주고 받음 (Direct-Electron-Transfer, DET)이 효율적으로 일어남을 의미한다. 또한, 센서의 감도는 약 ~50 μA/mM cm2으로 측정되었다.
[실험예 7] 파지 유래가 아닌 펩티드를 이용하여 제조한 GOx 엔자임 필름의 글루코스 반응성 평가
제조예 3에서 제조한 GOx 엔자임 필름에 대해 실험예 2와 동일한 방법으로 CV를 측정하였고, 그 결과를 도 20에 나타내었다.
도 20에 나타난 바와 같이, Ag/AgCl 레퍼런스 전극 대비 약 -400 mV 범위에서 강한 산화 환원 피크가 Cyclic voltammetry (CV) 상에 나타남을 알 수 있다. 상기의 결과는 GOx 내의 산화환원 센터 (FAD redox center)가 파지 유래가 아닌 펩티드를 이용하여 제조한 엔자임 필름 내에서도 SWNT와 직접적인 전자 주고 받음 (Direct-Electron-Transfer, DET)이 효율적으로 일어남을 의미한다. 또한, 센서의 감도는 약 ~55 μA / mM cm2으로 측정되었다.
[실험예 8] LOx 엔자임 필름의 락테이트에 대한 반응성 평가
10 mM PBS 버퍼에 보관된 제조예 4의 LOx 엔자임 필름을 금전극 (SPE 250BT, DropSens)에 전사시켜 상온 건조를 시켰다. 글루코스가 포함된 용액 대신에 10 μM 에서 2500 μM의 락테이트를 포함하는 10 mM PBS buffer (pH = 7.4, 79383, Sigma Aldrich) 용액을 사용한 것만을 제외하고는 실험예 2와 동일한 방법으로 CV를 측정하였고, 그 결과를 도 21에 나타내었다.
도 21에 나타낸 바와 같이, Ag/AgCl 레퍼런스 전극 대비 약 -400 mV 범위에서 강한 산화 환원 피크가 Cyclic voltammetry (CV) 상에 나타남을 알 수 있다. 상기의 결과는 LOx 내의 산화환원 센터 (FAD redox center)가 엔자임 필름 내의 SWNT와 직접적인 전자 주고 받음 (Direct-Electron-Transfer, DET)이 효율적으로 일어남을 의미한다. 또한 점차적으로 락테이트 농도를 증가 시켰을 때, 환원 전류는 양의 방향으로 농도 증가에 따라 선형적으로 증가하였으며, 이렇게 측정된 센서의 감도는 약 ~70 μA / mM cm2으로 측정 되었다.
[실험예 9] HRP 엔자임 필름의 과산화수소에 대한 반응성 평가
10 mM PBS 버퍼에 보관된 제조예 5에서 제조된 HRP 엔자임 필름을 금전극 (SPE 250BT, DropSens)에 전사시켜 상온 건조를 시켰다. 글루코스가 포함된 용액 대신에 10 μM 에서 1000 μM의 과산화수소를 포함하는 10 mM PBS buffer (pH = 7.4, 79383, Sigma Aldrich) 용액을 사용한 것만을 제외하고는 실험예 2와 동일한 방법으로 CV를 측정하였고, 그 결과를 도 22에 나타내었다.
도 22에 나타낸 바와 같이, Ag/AgCl 레퍼런스 전극 대비로 약 -450 mV 범위에서 강한 산화 환원 피크가 Cyclic voltammetry (CV) 상에 나타남을 알 수 있다. 상기의 결과는 HRP 내의 산화환원 센터 (Heme redox center)가 엔자임 필름 내의 SWNT와 직접적인 전자 주고 받음 (Direct-Electron-Transfer, DET)이 효율적으로 일어남을 의미한다. 또한 점차적으로 H2O2 농도를 증가 시 환원 전류는 음의 방향으로 농도 증가에 따라 선형적으로 증가하였으며, 센서의 감도는 약 ~ 310 μA / mM cm2으로 측정되었다. GOx나 LOx 처럼 엔자임의 산화환원 센터가 FAD인 경우와 달리 HRP의 경우 효소의 산화 환원 센터가 Heme으로 구성되어 있는데 Heme의 산화환원 센터는 H2O2 첨가 시 엔자임 반응에 의해 산화가 되므로, 전기화학적으로 순환 전류 인가 시 환원 전류의 증가로 나타난다. 따라서, HRP가 환원되어야 하는 양은 H2O2가 첨가되는 양에 따라 비례하며, 환원 전류가 음의 방향으로 증가하게 된다.
[실험예 10] Catalase 엔자임 필름의 과산화수소에 대한 반응성 평가
10 mM PBS 버퍼에 보관된 제조예 6에서 제조한 calatase 엔자임 필름을 금전극 (SPE 250BT, DropSens)에 전사시켜 상온 건조를 시켰다. 글루코스가 포함된 용액 대신에 10 μM 에서 1000 μM의 과산화수소를 포함하는 10 mM PBS buffer (pH = 7.4, 79383, Sigma Aldrich) 용액을 사용한 것만을 제외하고는 실험예 2와 동일한 방법으로 CV를 측정하였고, 그 결과를 도 23에 나타내었다.
도 23에 나타낸 바와 같이, Ag/AgCl 레퍼런스 전극 대비 약 -450 mV 범위에서 강한 산화 환원 피크가 Cyclic voltammetry (CV) 상에 나타남을 알 수 있다. 상기의 결과는 catalase 내의 산화환원 센터 (redox center)가 엔자임 필름 내의 SWNT와 직접적인 전자 주고 받음 (Direct-Electron-Transfer, DET)이 효율적으로 일어남을 의미한다. 또한 점차적으로 H2O2 농도를 증가 시켰을 때, 환원 전류는 음의 방향으로 농도 증가에 따라 선형적으로 증가하였으며, 센서의 감도는 약 ~ 230 μA / mM cm2으로 측정되었다.
[실험예 11] Laccase 엔자임 필름의 도파민에 대한 반응성 평가
10 mM PBS 버퍼에 보관된 제조예 7에서 제조한 laccase 엔자임 필름을 금전극 (SPE 250BT, DropSens)에 전사시켜 상온 건조를 시켰다. 글루코스가 포함된 용액 대신에 10 μM 에서 1000 μM의 도파민(dopamine)을 포함하는 10 mM PBS buffer (pH = 7.4, 79383, Sigma Aldrich) 용액을 사용한 것만을 제외하고는 실험예 2와 동일한 방법으로 CV를 측정하였고, 그 결과를 도 24에 나타내었다.
도 24에 나타낸 바와 같이, Ag/AgCl 레퍼런스 전극 대비 약 -350 mV 범위에서 강한 산화 환원 피크가 Cyclic voltammetry (CV) 상에 나타남을 알 수 있다. 상기의 결과는 laccase 내의 산화환원 센터 (copper redox center)가 엔자임 필름 내의 SWNT와 직접적인 전자 주고 받음 (Direct-Electron-Transfer, DET)이 효율적으로 일어남을 의미한다. 또한 점차적으로 도파민 농도를 증가시켰을 때, 산화 전류는 양의 방향으로 농도 증가에 따라 선형적으로 증가하였으며, 센서의 감도는 약 ~ 820 μA / mM cm2으로 측정되었다.
[실험예 12] GOx 엔자임 스티커를 이용한 바이오 센서의 특성 평가
제조예 9에서 제조된 엔자임 기반 바이오 센서 전극을 10 mM PBS 버퍼 용액 내에서 -0.6 V ~ -0.2 V 사이 전압 범위에서 200 mV/s의 속도로 전압을 주사하면서 CV를 30번 반복 측정하였고, 그 결과를 도 25에 나타내었다. 또한, 일 구체예에 따른 GOx 엔자임 스티커를 이용한 바이오 센서의 민감도 값을 통상적인 스텐실 마스크 공법(layer-by-layer)으로 제조된 GOx 기반 바이오 센서와 비교하여 민감도 값을 측정하였고, 그 결과를 도 26에 나타내었다.
도 25에 나타낸 바와 같이, Ag/AgCl 레퍼런스 전극 대비 약 -400 mV 범위에서 강한 GOx 산화 환원 피크가 Cyclic voltammetry (CV) 상에 나타남을 알 수 있다. 또한, 일 구체예에 따른 엔자임 스티커를 사용하여 제작된 바이오 센서는 30번의 전압 반복 주사에서도 산화-환원 전류가 변화 없이 안정적으로 구동됨을 알 수 있다. 또한, 글루코스 농도 증가 시에 환원 전류가 양의 방향으로 증가함을 알 수 있다.
도 26에 나타난 바와 같이, 엔자임 스티커 공법으로 제작된 GOx 바이오센서는 약 ~63 μA / mM cm2의 감도를 보였다. 이 민감도 값은 스텐실 마스크 공법으로 제작된 GOx 기반 바이오센서의 감도 (~67 μA / mM cm2)와 약 6%의 차이만을 나타낸다. 상기의 결과로, 일 구체예에 따른 엔자임 스티커 개념을 이용하면, 제3세대 바이오 센서를 임의의 기판에 효율적으로 구현할 수 있으면서도, 기존 제3세대 바이오 센서와 민감도의 차이가 없음을 알 수 있다.
[실험예 13] GOx, LOx 엔자임 스티커를 이용한 다중 바이오 센서의 특성 평가
제조예 10에서 제조된 GOx, LOx 엔자임 스티커 기반 다중 바이오 센서의 작업 전극에 -0.4 V를 인가하고 각 전극에서 나오는 전류를 측정하였고, 그 결과를 도 27에 나타내었다.
도 27에 나타낸 바와 같이, 글루코스를 첨가 시에는 GOx 엔자임 스티커가 전사된 전극에서는 양의 방향으로의 전류의 증가가 관찰된 반면에, LOx 엔자임 스티커가 전사된 작업 전극에서는 전류의 증가가 관찰되지 않았다. 또한, 락테이트가 첨가 되었을 때에는 LOx 엔자임 스티커가 전사된 전극에서는 전류 증가가 관찰되었으나, GOx 엔자임 스티커가 전사된 전극에서는 전류의 증가가 관찰되지 않았다. 또한 동시에 글루코스와 락테이트를 첨가하였을 때는 동시에 두 작업전극에서 전류의 증가가 목격되었다. 이는 각 GOx 엔자임 스티커와 LOx 엔자임 스티커가 전사된 전극이 각각의 기판에서만 효소반응에 의한 전류 증가가 유도됨을 의미하고, 따라서, 일 구체예에 따른 센서는 높은 선택성뿐만 아니라, 다중 작업 전극 기반의 Multiple array 바이오 센서로서도 사용가능함을 알 수 있다.
Claims (20)
- 기판; 및그래피틱 물질, 상기 그래피틱 물질에 결합된, 상기 그래피틱 물질에 결합능을 갖는 펩티드 또는 파지, 및 상기 그래피틱 물질의 네트워크 구조 상에, 및 구조 내에 형성된 효소를 포함하는 효소 필름; 을 포함하는 바이오 센서.
- 청구항 1에 있어서, 상기 효소 필름은 효소 일체형인 것인 바이오 센서.
- 청구항 2에 있어서, 상기 효소는 상기 그래피틱 물질의 네트워크 구조 상에, 내에, 및 아래에 형성된 것인 바이오 센서.
- 청구항 1에 있어서, 상기 파지는 파지의 외피 단백질 또는 그 단편에 디스플레이된, 그래피틱 물질에 결합능을 갖는 펩티드를 포함하는 것인 바이오 센서.
- 청구항 1에 있어서, 상기 그래피틱 물질과 상기 펩티드 또는 상기 파지의 복합체가 양전하 또는 음전하를 띠도록 양전하 고분자 또는 음전하 고분자로 개질된 것인 바이오 센서.
- 청구항 1에 있어서, 상기 펩티드는 α-헬릭스 또는 β-시트의 구조로 이루어진 것인 바이오 센서.
