Definitions
- the present invention relates to a pharmaceutical composition for preventing or treating malignant hyperthermia comprising a NAADP antagonist.
- the present invention also relates to a composition for skin regeneration comprising a cADRR or NAADP antagonist.
- Malignant hyperthermia is a disease caused by the use of anesthetics, especially inhaled anesthetics (Halothane, Isoflurane, etc.) and succinycholine-based anesthetics during surgery, and the genetic disease of the ryanodine receptor (RYR) in skeletal muscle It is a disease that occurs in a patient who dies due to an increase in body temperature due to an irreversible increase in the oxygen consumption rate of the skeletal muscle of the human body.
- the malignant hyperthermia is a disease that occurs in about 1 person per 100,000 people have been reported many cases worldwide.
- RYR inhibitor Dantrolene is used clinically as the only drug, but due to its low water solubility, there is a limitation that a large amount of drug is required. In addition, the prognosis is also reported to be poor, it is urgent to develop a treatment for malignant hyperthermia.
- Photoaging or exogenous aging refers to aging that occurs when the skin is always exposed to the external environment. Due to the use of cooling and warming devices, it is difficult to maintain proper moisture of the skin due to the dry indoor environment, and it is also difficult for the skin to maintain normal moisture due to wind and chemicals. In particular, since most of them are exposed to ultraviolet light, it can be avoided a great deal, which is a big difference from endogenous aging.
- the main cause of such photoaging is known to be due to the active oxygen generated when sunlight (ultraviolet rays). Ultraviolet rays contained in the sun's rays can be divided into UV-C, UV-B and UV-A according to their wavelength range.
- the UV-A (320 ⁇ 400nm) is not absorbed by the ozone layer, the wavelength range corresponds to 320 ⁇ 400 nm (0.32 ⁇ 0.40 ⁇ m) and less energy than UV-B, but may burn the skin.
- UV-B The major UV region that burns skin is UV-B, but UV-A not only makes the skin bulge, but also acts on the skin's immune system and can cause long-term skin damage from skin aging.
- UV-B (280-320 nm) is mostly absorbed by the ozone layer, but some reach the ground surface. UV-B reaching a very small amount of earth corresponds to a wavelength range of 0.28 ⁇ 0.32 ⁇ m, and can burn the skin of animals including humans and penetrate the skin tissue, thereby inducing damage. UV-B is sometimes attracting attention as a cause of skin cancer, and the cause of skin cancer has been reported to be mostly associated with exposure to the sun's rays and UV-B. UV-B, on the other hand, activates provitamin D in the skin and converts it into vitamin D, which is essential for the human body.
- UV-C (100 ⁇ 280nm) is completely absorbed by the ozone layer, the wavelength range is 0.20 ⁇ 0.29 ⁇ m, causing harmful effects on life, causing chromosomal variation, killing single-celled organic matter, harming the cornea of the eye.
- NAADP cADPR The NAADP cADPR and is known to induce the release of Ca + 2 ions from the endoplasmic reticulum, these antagonists act thereof and competitively inhibits its function. Many of the actions of these cADPR and NAADP antagonists are known, but their use for skin regeneration is unknown.
- Dantrolin which is a conventional treatment for malignant hyperthermia, requires a large amount of drugs due to its low water solubility, and its prognosis is also reported to be poor.
- an object of the present invention is to provide a pharmaceutical composition for preventing or treating new malignant hyperthermia.
- composition for skin regeneration comprising a cADPR antagonist and / or a NAADP antagonist.
- the present invention provides a pharmaceutical composition for preventing or treating malignant hyperthermia comprising a NAADP antagonist.
- a pharmaceutical composition for preventing or treating malignant hyperthermia comprising a compound represented by Formula 1 below:
- X is halogen and R is OH Or OC 1-5 alkyl.
- the inventors of the present invention have completed the present invention by discovering that cADPR and NAADP antagonists can promote skin regeneration. Therefore, the means for solving the above problems are as follows.
- composition for skin regeneration comprising at least one compound selected from a compound represented by Formula 1 and a compound represented by Formula 2 below:
- A is halogen, NH 2 , OC 1-5 alkyl group or C 1-5 alkyl group; X is halogen and R is OH Or OC 1-5 alkyl.
- composition for treating skin diseases comprising the composition for skin regeneration of 2 above.
- a cosmetic composition comprising the composition for skin regeneration of 2 above.
- a food composition comprising the composition for skin regeneration of 2 above.
- the pharmaceutical composition for the prevention and treatment of malignant hyperthermia comprising the NAADP antagonist of the present invention, it is possible to lower irreversible oxygen utilization in skeletal muscle of patients with malignant hyperthermia hereditary disease, thereby effectively preventing and treating malignant hyperthermia.
- the pharmaceutical composition for preventing or treating malignant hyperthermia of the present invention can be used even after the onset of malignant hyperthermia, and can replace the conventional malignant hyperthermia therapeutic agent because it can suppress abnormal oxygen utilization of cells at a lower concentration than the conventional malignant hyperthermia treatment agent. It can be used stably and can be effectively used in related industries.
- composition for skin regeneration of the present invention can protect the skin from skin cell damage induced by UV-B of ultraviolet light.
- the ability to recover and regenerate the skin from such damage is excellent, there are no other side effects, the skin regeneration composition of the present invention can be used as a skin external preparation, pharmaceutical compositions, cosmetic compositions and food compositions.

- Figure 2 shows the intracellular oxygen utilization rate according to the treatment of dantroline and NED-19 in RYR gene mutant cells (B2435H) showing malignant hyperthermia response with isoflurane (ISO) treatment in human B cells with malignant hyperthermia (%) O 2 ) is a diagram showing the change (*, P ⁇ 0.05 versus ISO treatment level. #, P ⁇ 0.01 versus ISO treatment level.One-way ANOVA.Error bars denote sd).
- FIG 3 is a diagram showing the effect of inhibiting intracellular oxygen utilization (% O 2 ) following NED-19 treatment after inducing malignant hyperthermia response with isoflurane (ISO) treatment in human B cells with malignant hyperthermia (B2435H).
- ISO isoflurane
- the present invention provides a pharmaceutical composition for preventing or treating malignant hyperthermia, including a NAADP antagonist.
- NAADP nicotinic acid adenin dinucleotide phosphate
- NADP nicotinamide adenine dinucleotide phosphate
- NAADP antagonists refer to compounds that are similar in structure to NAADP and can bind to receptors on NAADP, but have little functional activity on their own. That is, the NAADP antagonist can competitively bind NAADP to the receptor to which NAADP binds, thereby preventing the action of NAADP, thereby inhibiting the intracellular response induced by NADDP.
- prevention means blocking the occurrence before the disease occurs, or early detection and timely treatment of the disease.
- treatment includes the cure of the disease as well as the partial cure, improvement and alleviation of the condition.
- the NAADP antagonist is a compound having a structure of formula (1).
