Abstract
본 명세서에는 유효성분으로 인삼 꽃대 추출물을 포함하는 세포 재생용 조성물로서, 상기 추출물은 물, C1-C5 알코올 또는 C1-C5 알코올 수용액을 이용하여 추출한 추출물이며, 상기 조성물은 건강 식품, 화장료 또는 약학 조성물로 사용될 수 있고, 상기 조성물은 세포 재생을 촉진시키므로 피부 재생 또는 상처 치유 효과가 우수하다.
Description
본 명세서는 유효성분으로 인삼 꽃대 추출물을 포함하는 세포 재생용 조성물에 관한 것이다.
기존에는 인삼근, 인삼 열매 또는 인삼 꽃 등의 추출물에 대한 연구는 많았다. 예를 들어, 인삼 꽃등을 이용한 미백용 화장료 조성물에 대한 연구, 피부 질환의 연구가 있었다.
그러나, 인삼 수확 후 버려지는 인삼꽃대 추출물에 대한 연구는 없었으며, 특히 상기 인삼꽃대 추출물에 대한 세포 재생 연구는 전혀 없었다.

상기와 같은 문제점에 착안하여, 본 발명의 발명자들은 인삼 꽃대 추출물에 관한 연구를 하여 상기 인삼 꽃대 추출물에 세포 재생 효과가 있음을 알게 되어 본 발명에 이르게 되었다.

일반적으로 인삼이라 함은 인삼의 뿌리부분인 '인삼근'을 말하는 것으로서 약재로 사용되거나 음식 등으로 조리되어 섭취되어 온 부분도 뿌리 부분인 '인삼근'이다. 인삼(Panax ginseng C.A. Meyer)은 오가피과 인삼 속에 속하는 식물로 한국, 중국 및 일본 등지에서 질병을 예방하고 수명을 연장시킬 목적으로 사용되어 왔으며, 지금까지 알려진 인삼의 효능 및 효과는 탈모 예방 및 육모 촉진 효능을 비롯하여 항암활성, 면역기능 조절작용, 항당뇨 작용, 간기능 항진효능, 심혈관 장애 개선, 혈압조절 작용, 갱년기 장애 개선, 항스트레스 및 항피로 작용, 항산화 활성 및 노화억제 효능 등이 있다.
인삼꽃대는 통상적으로 약재로써 사용되지 않았을 뿐만 아니라 식품의 형태로도 섭취한 이력은 찾아보기 어렵다. 인삼꽃대는 씨를 심어 4년째 되는 5월 ~ 6월경 나오는데 인삼 근의 원활한 성장을 위해 만개하기 전인 6월경에 인삼 꽃과 함께 제거되어 버려진다.
인삼의 대표적 생리활성 성분인 진세노사이드(Ginsenoside)는 인삼의 지상 및 지하부에 고르게 분포되어 있으며, 특히 인삼근(뿌리), 인삼엽 및 인삼 열매 등 부위에 따라 진세노사이드 함량뿐만 아니라 조성도 다른 것으로 알려져 있다. 이러한 조성의 차이는 다양한 효능의 있고 없음에 차이를 만들고 각기 다른 부위별 효능 특성의 원인이 된다. 일례로, 인삼 꽃 추출물이 인삼 근 추출물과 비교할 때 당뇨병의 예방과 치료에 약 23배 이상의 효과를 보이며 탁월한 효과를 가지는 것으로 확인된 바 있다.
본 명세서에서는 그간 버려지고 사용되지 않았던 인삼꽃대 부분의 세포 재생 효능을 평가하여 인삼꽃대 추출물이 세포 재생 효과를 가짐을 밝혀냈다.
이하 본 발명에 대하여 상세히 설명한다.

본 발명의 일측면에서, 상기 조성물은 산제, 과립제, 정제, 캡슐제, 현탁액, 에멀젼, 시럽, 에어로졸, 및 외용제 중 어느 하나의 형태로 제형화된 조성물을 제공한다.
본 발명의 일측면에서, 상기 조성물은 화장료 조성물일 수 있다. 본 명세서의 화장료 조성물은 당업계에서 통상적으로 제조되는 어떠한 제형으로도 제조될 수 있으며, 예를 들어, 용액, 현탁액, 유탁액, 페이스트, 겔, 크림, 로션, 파우더, 비누, 계면활성제-함유 클린싱, 오일, 분말 파운데이션, 유탁액 파운데이션, 왁스 파운데이션 및 스프레이 등으로 제형화될 수 있으나, 이에 한정되는 것은 아니다.
상기 인삼 꽃대 추출물을 화장료 조성물로 사용 시 투여량 결정은 당업자의 수준 내에 있으며, 조성물의 1일 투여 용량은 투여하고자 하는 대상의 연령, 건강상태, 합병증 등의 다양한 요인에 따라 달라지지만, 성인을 기준으로 할 때 일반적으로는 상기 인삼 꽃대 추출물을 1 내지 500mg/kg, 바람직하게는 30 내지 200 mg/kg을 1일 1 내지 2회 분할하여 투여할 수 있으며, 상기 투여량은 어떠한 방법으로도 본 발명의 범위를 한정하는 것이 아니다
본 발명의 일 측면에서 상기 조성물은 식품 조성물일 수 있으며 예를 들어, 각종 식품류, 음료, 껌, 차, 비타민 복합제, 건강보조 식품류 등이 있고, 분말, 과립, 정제, 캡슐 또는 음료인 형태로 사용될 수 있다. 각 제형의 식품 조성물은 유효 성분 이외에 해당 분야에서 통상적으로 사용되는 성분들을 제형 또는 사용 목적에 따라 당업자가 어려움 없이 적의 선정하여 배합할 수 있으며, 다른 원료와 동시에 적용할 경우 상승 효과가 일어날 수 있다.
이때, 식품 또는 음료 중에 포함되는 유효성분의 양은 일반적으로 건강식품 조성물인 경우에는 전체 식품의 0.1 내지 60 중량%를 포함할 수 있고, 건강음료 조성물은 100ml를 기준으로 0.002 내지 50g, 일측면에서는 0.02 내지 10g을 포함할 수 있다.
상기 인삼 꽃대 추출물을 식품 조성물로 사용 시 투여량 결정은 당업자의 수준 내에 있으며, 이의 1일 투여 용량은 예를 들어 0.1mg/kg/일 내지 5000mg/kg/일, 보다 구체적으로는 50 mg/kg/일 내지 500 mg/kg/일이 될 수 있으나, 이에 제한되지 않으며, 투여하고자 하는 대상의 연령, 건강 상태, 합병증 등 다양한 요인에 따라 달라질 수 있다.
일실시예에서 상기 조성물은 본 발명이 목적으로 하는 주 효과를 손상시키지 않는 범위 내에서 주 효과에 상승 효과를 줄 수 있는 다른 성분 등을 함유할 수 있다. 예를 들어, 물성 개선을 위하여 향료, 색소, 살균제, 산화방지제, 방부제, 보습제, 점증제, 무기염류, 유화제 및 합성 고분자 물질 등의 첨가제를 더 포함할 수 있다. 그 외에도, 수용성 비타민, 유용성 비타민, 고분자 펩티드, 고분자 다당 및 해초 엑기스 등의 보조 성분을 더 포함할 수도 있다. 상기 성분들은 제형 또는 사용 목적에 따라서 당업자가 어려움 없이 적의 선정하여 배합할 수 있으며, 그 첨가량은 본 발명의 목적 및 효과를 손상시키지 않는 범위 내에서 선택될 수 있다. 예를 들어, 상기 성분들의 첨가량은, 조성물 전체 중량을 기준으로, 0.01-5 중량%, 보다 구체적으로는 0.01-3 중량% 범위일 수 있다.
본 발명의 일측면에서, 상기 조성물은 약학 조성물일 수 있다. 약학 제형의 제조에 있어서, 활성 성분을 담체와 함께 혼합 또는 희석하거나, 용기 형태의 담체 내에 봉입시키는 것이 바람직하다. 담체가 희석제로 사용 되는 경우에는 활성 성분에 대한 담체, 부형제 또는 매질(medium)로 작용하는 고형, 반고형 또는 액상의 물질일 수 있다. 따라서, 제형은 정제, 환제, 분제, 사쉐, 엘릭시르, 현탁제, 유제, 용액제, 시럽제, 에어로졸, 연질 또는 경질 젤라틴 캅셀제, 멸균 주사제, 멸균 분제 등의 형태일 수 있다. 적합한 담체, 부형제 및 희석제의 예로는, 락토로스, 덱스트로스, 수크로스, 솔비톨, 만니톨, 칼슘 실리케이트, 셀룰로오스, 메틸 셀룰로오스, 미정질 셀룰로오스, 폴리비닐피롤리돈, 물, 메틸하이드록시벤조에이트, 프로필하이드록시벤조에이트, 탈크, 마그네슘 스테아레이트 및 광물유를 들 수 있다. 제형은 충진제, 항응집제, 윤활제, 습윤제, 향료, 유화제, 방부제 등을 추가로 포함할 수 있다. 본 발명의 약학 조성물은 포유동물에 투여된 후 활성성분의 신속, 지속 또는 지연된 방출을 제공할 수 있도록 당업계에 잘 알려진 방법을 사용하여 제형화될 수 있다. 본 발명의 약학 조성물은 경구, 경피, 피하, 정맥, 복강, 근육, 국소도포, 첩포 및 이온토포레시스 (iontophoresis)를 포함한 여러 경로를 통해 투여될 수 있고, 이 중에서 국소 적용 및 경구투여가 바람직할 수 있다. 사람의 경우, 활성 화합물의 통상적인 1일 투여량은 0.1 내지 100mg/kg체중, 바람직하게는 5 내지 70mg/kg체중의 범위일 수 있고, 1회 또는 수회로 나누어 투여할 수 있다. 그러나, 활성 성분의 실제 투여량은 치료할 질환, 투여 경로, 환자의 연령, 성별 및 체중, 및 질환의 중증도 등의 여러 관련 인자에 비추어 결정되어야 하는 것으로 이해되어야 하며, 따라서, 상기 투여량은 어떠한 방법으로도 본 발명의 범위를 한정하는 것은 아니다.
본 발명에 따른 조성물의 제형은 용액, 유화물, 점성형 혼합물, 타블렛, 분말 등의 다양한 형태일 수 있으며, 이는 단순 음용, 주사 투여, 스프레이 방식 또는 스퀴즈 방식 등의 다양한 방법으로 투여될 수 있다.
이하, 실시예를 통하여 본 발명을 더욱 상세히 설명하고자 한다. 이들 실시예는 오로지 본 발명을 예시하기 위한 것으로, 본 발명의 범위가 이들 실시예에 의해 제한되는 것으로 해석되지 않는 것은 당업계에서 통상의 지식을 가진 자에게 있어서 자명할 것이다.
실시예
1 :
인삼꽃대
추출물의 제조
인삼꽃대 추출물의 경우 인삼꽃대를 채취하여 열매부분을 분리하여 제거한 다음 일광건조 또는 열풍건조 하여 인삼꽃대 건조원료로 제조하였다. 그 후 인삼 꽃대 건조물 1kg에 70% 에탄올를 가하여 환류 추출한 다음 여과하고 40~45에서 감압 농축하여 사용하였다.
상기 과정을 통해 얻은 인삼 꽃대 추출물을 하기 실험예에 사용하였다.
비교예
1 내지 4
비교예 1로 금산에서 수확한 인삼을 찌고 말려서 제조된 홍삼을 실시예 1과 동일한 추출방법으로 추출하여 사용하였다.
비교예 2로 상기 실시예 1과 동일한 추출방법으로 추출하되 원료를 인삼 꽃대 대신 인삼 꽃을 사용하였다.
비교예 3으로 상기 실시예 1과 동일한 추출방법으로 추출하되 원료를 인삼 꽃대 대신 인삼 뿌리를 사용하였다.
비교예 4로 상기 실시예 1과 동일한 추출방법으로 추출하되 원료를 인삼 꽃대 대신 인삼 열매를 사용하였다.
상기 비교예 1 내지 4를 하기 실험예들에 사용하였다.
실험예
1 : 인삼 꽃대 추출물의 진세노사이드 분석
상기 실시예 1의 인삼 꽃대 추출물과 상기 비교예 3 내지 4의 인삼 뿌리 추출물, 인삼 열매 추출물의 진세노사이드를 건강식품공전의 가이드에 따라 분석하였다.
상기 실험에 따른 인삼 꽃대, 인삼 뿌리 및 인삼 열매의 진세노사이드 분석 결과는 하기 표 1과 같았다.

표 1에서와 같이 본 발명 실시예 1의 인삼 꽃대 추출물은 인삼 뿌리 및 인삼 열매 추출물과 진세노사이드 성분에 있어 차이가 있음을 확인하였다. 따라서, 인삼 뿌리 및 인삼 열매 추출물과 본원발명의 인삼 꽃대 추출물은 진세노사이드의 성분 함량에 있어 상이한 물질임을 확인하였다.
실험예
2 : Cell migration assay 실험
상기 실시예 1(70% 에탄올 추출물)의 인삼 꽃대 추출물의 세포 재생 효과 및 상처 치유 효과를 확인하기 위해 cell migration assay 실험을 하였다.
실험은 하기 방법에 따라 수행하였다.
ATCC에서 구입한 각질 세포인 HaCaT를 95% 이상이 되도록 디쉬(dish)에 배양한 후 엘로우 팁(Yellow tip)으로 긁어 wound를 만들었다. 상기 wound가 생긴 디쉬에 2% FBS(fetal bovine serum, 웰진)를 처리 후, 하기 표 2와 같은 시료를 처리하여, 시간 경과에 따른 회복 정도를 이동거리로 측정하여 정량화 하였다. 이동거리는 현미경을 이용하여 배지를 측정하였고 Toupview 프로그램을 통해 100배 비율을 유지하며 같은 부위를 측정하여 비교하였다.
양성 대조군으로 시료를 처리하지 않은 10% FBS포함 배지를 양성대조군으로 하였으며, 음성 대조군으로 FBS 및 시료를 처리하지 않은 배지를 음성대조군으로 하였고, 일반 대조군으로 시료를 처리하지 않은 2% FBS포함 배지를 일반대조군으로 하였다.

실험결과는 도 1 및 도 2에 도시하였다.
실험결과 도 1 및 도 2에서 보는 것처럼 각질 세포에 wound를 가한 후 이동거리를 측정한 결과 비교예 1, 2 및 4에 비해 실시예 1의 인삼 꽃대 추출물의 이동 거리가 우세하였으며, 특히 48시간이 지난 후에는 실시예 1의 인삼 꽃대가 양성 대조군만큼 회복되는 경향을 보여주었다. 따라서, 실시예 1의 인삼 꽃대 추출물은 세포재생 효과가 우수하였다. 또한, 이로 인해 상기 실시예 1의 인삼 꽃대 추출물은 상처 회복 또는 피부 재생에 효과가 있음을 확인하였다.
실험예
3 :
CCK8
assay 실험
상기 실시예 1(70% 에탄올 추출물)의 인삼 꽃대 추출물의 CoCl2 처리 후 생존력 평가를 위해 CCK8 assay 실험을 하였다.
실험은 하기 방법에 따라 수행하였다.
각질 세포인 HaCaT cell line에서 CoCl2를 500um 처리 후 하기 표 3과 같은 효능 물질을 처리하여 생존력을 평가하였다. 96 well microplate에 HaCaT cel을 5 × 104 cells/well을 배양액(DMEM 10% FBS, P/S 1%) 100 μl에 넣어서 seeding한다. 16시간 후 효능물질(표3)과 CoCl2(500um)를 동시 처리한 후, CO2 incubator에서 24시간 배양한다. 세포배양액을 제거하고 PBS로 2회 세척하고 Cell Counting Kit-8 10 μl와 배양액 100 μl 섞은 후 well에 넣어 반응 시켜준다. CO2 incubator에서 1시간 배양 후 microplate reader를 활용하여 450 nm 흡광도를 측정한다. 일반 대조군으로 CoCl2를 처리하지 않은 HaCaT cell line을 사용하였다.

실험결과 도 3에서와 같이 생존력 평가시 CoCl2 처리군의 경우 40% 가량 생존율이 감소하였지만 인삼 꽃대 추출물을 처리한 경우(실시예 1) 약 91%의 생존율을 보였다. 또한, 실시예 1의 인삼 꽃대 추출물은 비교예 1, 2 및 4에 비해 우수한 생존율을 보였다.
이하, 본 발명에 따른 조성물의 제형예를 설명하나, 이는 본 발명을 한정하고자 함이 아니며, 단지 구체적으로 설명하고자 함이다.
[제형예 1] 환제의 제조
실시예 1을 30 중량%, 옥수수전분 30 중량%, 글리세린 20 중량% 및 솔비톨 분말 20 중량%를 혼합하고, 제환기를 사용하여 환을 제조하였다. 내용물의 최종 중량은 3.5g 이었다.
[제형예 2] 정제의 제조
실시예 1의 추출물 30 중량%, 유당 20.5 중량%, 덱스트린 20 중량%, 말티톨 분말 20 중량% 및 자일리톨 분말 7 중량%를 혼합하고, 유동층 건조기를 이용하여 과립화한 다음, 슈거 에스테르(sugar ester) 2.5 중량%를 첨가하여 타정기로 타정하였다. 내용물의 최종 중량은 2g이었다.
[제형예 3] 과립제의 제조
실시예 1의 추출물 30 중량%, 자일리톨 5 중량% 및 이소말트 65 중량%를 혼합하고, 유동층 과립기를 사용하여 과립으로 성형한 후에 포에 충진하였다. 내용물의 최종 중량은 2g이었다.
[제형예 4] 주사제의 제조
하기 표 4에 기재된 조성에 따라 통상적인 방법으로 주사제를 제조하였다.

Claims (11)
- 유효성분으로 인삼 꽃대 추출물을 포함하는 세포 재생용 조성물.
- 제 1항에 있어서,상기 추출물은 물, C1-C5 알코올 또는 C1-C5 알코올 수용액을 이용하여 추출한 추출물인, 조성물.
- 제 2항에 있어서,상기 C1-C5 알코올 수용액의 농도는 40% 내지 90% (v/v)인, 조성물.
- 제 2항에 있어서,상기 C1-C5 알코올은, 메탄올, 에탄올, 이소프로필알코올, n-프로필알코올, n-부탄올 및 이소부탄올로 구성된 군으로부터 선택되는 어느 하나 이상을 이용한, 조성물.
- 제 2항에 있어서,상기 추출물은 60 내지 80%(v/v) 농도의 에탄올 수용액을 이용하여 추출한 추출물인, 조성물.
- 제 1항에 있어서,상기 인삼 꽃대는 인삼 열매가 제거된 것인, 조성물.
- 제 1항에 있어서,상기 조성물은 상처 회복용인, 조성물.
- 제 1항에 있어서,상기 조성물은 피부 재생을 촉진시키는, 조성물.
