本発明のADAMTS5遺伝子の定量方法は、ADAMTS5遺伝子の量を測定する工程(以下、「ADAMTS5遺伝子量測定工程」と称することがある)を少なくとも含み、必要に応じて更にその他の工程を含む。
前記ADAMTS5遺伝子量測定工程は、定量PCRにより、試料中のADAMTS5遺伝子の量を測定する工程である。
前記ADAMTS5遺伝子量測定工程では、前記試料中における前記ADAMTS5遺伝子のmRNAの量が定量される。
前記試料としては、特に制限はなく、目的に応じて適宜選択することができ、例えば、末梢血、関節滑膜、関節液、これらからRNAを抽出した試料、前記RNAをcDNAとした試料などが挙げられる。これらの中でも、末梢血、前記末梢血からRNAを抽出した試料、前記RNAをcDNAとした試料が、採取が簡便である点で、好ましい。
前記試料の由来としては、特に制限はなく、目的に応じて適宜選択することができるが、ヒトが好ましい。
前記末梢血などからRNAを抽出する方法としては、特に制限はなく、公知の方法を適宜選択することができる。
前記RNAからcDNAを調製する方法としては、特に制限はなく、公知の方法を適宜選択することができる。
前記ADAMTS(a disintegrin and metalloproteinase with thrombospondin motifs)5は、ADAMTSファミリーに属する軟骨破壊に関連する酵素である。
前記ADAMTS5遺伝子の塩基配列は公知であり、その塩基配列は、GenBank(NCBI)などの公共データベースを通じて容易に入手することができる。例えば、ヒトADAMTS5遺伝子の塩基配列は、NCBI accession number NM_007038.3で入手可能である。
前記定量PCRは、本発明のADAMTS5遺伝子が含まれる領域を増幅する増幅用プライマー対(以下、「ADAMTS5遺伝子用プライマー対」と称することがある)を用いて行う。
前記ADAMTS5遺伝子用プライマー対は、下記配列番号:2で表される塩基配列からなるフォワードプライマーと、下記配列番号:3で表される塩基配列からなるリバースプライマーとからなる。

前記ADAMTS5遺伝子の検出用プローブとしては、特に制限はなく、目的に応じて適宜選択することができるが、下記配列番号:4で表される塩基配列からなるプローブが好ましい。

前記定量PCRとしては、前記ADAMTS5遺伝子用プライマー対を用いる限り、特に制限はなく、目的に応じて適宜選択することができるが、蛍光プローブを用いたリアルタイムPCRが好ましく、前記ADAMTS5遺伝子の検出用プローブを用いたリアルタイムPCRがより好ましい。
前記定量PCRでは、ADAMTS5遺伝子の絶対量、及び後述するコントロール遺伝子に対する相対量(ADAMTS5遺伝子の量/コントロール遺伝子の量)の少なくともいずれかを定量することができる。
前記リアルタイムPCRに用いる前記リバースプライマーの量としては、特に制限はなく、目的に応じて適宜選択することができるが、最終濃度が300nM程度が好ましい。
前記リアルタイムPCRに用いる前記プローブの量としては、特に制限はなく、目的に応じて適宜選択することができるが、最終濃度が75nM程度が好ましい。
前記(2)95℃、15秒間、次いで、60℃、60秒間のサイクル数としては、特に制限はなく、目的に応じて適宜選択することができるが、50サイクル程度が好ましい。
前記検量線法は、検量線を作成し、その検量線からサンプルの定量を行う方法である。
前記検量線法のコントロールDNAの鋳型として用いるDNAとしては、濃度が既知であれば、特に制限はなく、目的に応じて適宜選択することができ、例えば、健常人から採取し、必要に応じて精製したADAMTS5の全cDNAや、前記ADAMTS5 cDNAを組み込んだプラスミドなどが挙げられる。
前記その他の工程としては、本発明の効果を損なわない限り、特に制限はなく、目的に応じて適宜選択することができ、例えば、コントロール遺伝子(「内部標準として用いる遺伝子」と称することもある)の量を測定する工程(以下、「コントロール遺伝子量測定工程」と称することがある)などが挙げられる。
前記コントロール遺伝子量測定工程は、定量PCRにより、試料中のコントロール遺伝子の量を測定する工程である。
前記コントロール遺伝子量測定工程では、前記試料中における前記コントロール遺伝子のmRNAの量が定量される。
前記試料は、上記した<ADAMTS5遺伝子量測定工程>の項目における<<試料>>と同様である。
前記コントロール遺伝子としては、特に制限はなく、目的に応じて適宜選択することができ、例えば、β-アクチン遺伝子、GAPDH(glyceraldehyde-3-phosphate dehydrogenase)遺伝子などが挙げられる。これらの中でも、β-アクチン遺伝子が、好ましい。
前記β-アクチン遺伝子の塩基配列は公知であり、その塩基配列は、GenBank(NCBI)などの公共データベースを通じて容易に入手することができる。例えば、ヒトβ-アクチン遺伝子の塩基配列は、NCBI accession number NM_001101.2で入手可能である。
以下、前記コントロール遺伝子として、前記β-アクチン遺伝子を用いる場合の定量PCRについて説明する。
前記β-アクチン遺伝子の定量PCRに用いるプライマー対としては、特に制限はなく、目的に応じて適宜選択することができるが、以下のβ-アクチン遺伝子が含まれる領域を増幅する増幅用プライマー対(以下、「β-アクチン遺伝子用プライマー対」と称することがある)が好ましい。
前記β-アクチン遺伝子の定量PCRに用いるプローブとしては、特に制限はなく、目的に応じて適宜選択することができるが、以下のβ-アクチン遺伝子の検出用プローブが好ましい。
前記β-アクチン遺伝子の定量PCRに用いるプライマー対と、プローブとの組合せとしては、特に制限はなく、目的に応じて適宜選択することができるが、以下のβ-アクチン遺伝子用プライマー対と、以下のβ-アクチン遺伝子の検出用プローブとの組合せが好ましい。

前記レポーター色素と、クエンチャー色素との組合せは、上記した<ADAMTS5遺伝子量測定工程>の-ADAMTS5遺伝子の検出用プローブ-の項目において記載したものと同様である。
前記β-アクチン遺伝子の定量PCRとしては、特に制限はなく、目的に応じて適宜選択することができるが、蛍光プローブを用いたリアルタイムPCRが好ましい。
前記β-アクチン遺伝子の定量PCRにより、β-アクチン遺伝子の絶対量を定量することができる。また、上記ADAMTS5遺伝子の前記試料における相対量の定量に用いることができる。
前記β-アクチン遺伝子のリアルタイムPCRに用いる前記リバースプライマーの量としては、特に制限はなく、目的に応じて適宜選択することができるが、最終濃度が300nM程度が好ましい。
前記β-アクチン遺伝子のリアルタイムPCRに用いる前記プローブの量としては、特に制限はなく、目的に応じて適宜選択することができるが、最終濃度が50nM程度が好ましい。

本発明のプライマー対とプローブとの組合せは、定量PCRによる試料中のADAMTS5遺伝子の定量に用いるプライマー対とプローブとの組合せである。
前記プライマー対は、上記した(ADAMTS5遺伝子の定量方法)の<ADAMTS5遺伝子量測定工程>の-ADAMTS5遺伝子用プライマー対-の項目に記載のものと同様である。
前記プローブは、上記した(ADAMTS5遺伝子の定量方法)の<ADAMTS5遺伝子量測定工程>の-ADAMTS5遺伝子の検出用プローブ-の項目に記載のものと同様である。
本発明のキットは、定量PCRによる試料中のADAMTS5遺伝子の定量に用いるキットであって、本発明のプライマー対を少なくとも含み、必要に応じて更にプローブなどのその他の構成を含む。
前記プライマー対は、上記した本発明のADAMTS5遺伝子用プライマー対である。
前記その他の構成としては、本発明の効果を損なわない限り、特に制限はなく、目的に応じて適宜選択することができ、例えば、ADAMTS5遺伝子の検出用プローブ、コントロール遺伝子が含まれる領域を増幅する増幅用プライマー対(以下、「コントロール遺伝子用プライマー対」と称することがある)、コントロール遺伝子の検出用プローブ、PCRに用いる試薬などが挙げられる。
前記プローブは、上記した(ADAMTS5遺伝子の定量方法)の<ADAMTS5遺伝子量測定工程>の-ADAMTS5遺伝子の検出用プローブ-の項目に記載のものと同様である。
前記コントロール遺伝子は、上記した(ADAMTS5遺伝子の定量方法)の<その他の工程>の<<コントロール遺伝子量測定工程>>の-コントロール遺伝子-の項目に記載のものと同様であり、好ましい遺伝子も同様である。
ADAMTS5遺伝子のcDNAとして、配列番号:1で表される塩基配列からなるDNAをプラスミドに挿入後、制限酵素処理をすることにより調製した。前記配列番号:1で表される塩基配列は、ADAMTS5(NCBI Reference Sequence: NM_007038.3)の1,914番目~2,343番目(430bp)の塩基に相当する。
ADAMTS5遺伝子が含まれる領域を増幅する増幅用プライマー対及びADAMTS5遺伝子の検出用プローブとして、以下のものを化学合成により調製した。
・ フォワードプライマー
5’-CTAAGCCCTGGTCCAAATGC-3’ (配列番号:2)
・ リバースプライマー
5’-CCAGCAAACAGTTACCATGGC-3’ (配列番号:3)
・ プローブ
5’-FAM-TTCAGCCACCATCACAGAATTCCTGGA-TAMRA-3’ (配列番号:4)
前記配列番号:4で表されるプローブは、5’末端がFAM(レポーター色素)で修飾されており、3’末端がTAMRA(クエンチャー色素)で修飾されている。
β-アクチン遺伝子のcDNAとして、配列番号:5で表される塩基配列からなるDNAをプラスミドに挿入後、制限酵素処理をすることにより調製した。前記配列番号:5で表される塩基配列は、β-アクチン(NCBI Reference Sequence: NM_001101.2)の3番目~469番目(467bp)の塩基に相当する。
β-アクチン遺伝子が含まれる領域を増幅する増幅用プライマー対及びβ-アクチン遺伝子の検出用プローブとして、以下のものを化学合成により調製した。
・ フォワードプライマー
5’-CCAGCTCACCATGGATGATG-3’ (配列番号:6)
・ リバースプライマー
5’-CCTTGCACATGCCGGAG-3’ (配列番号:7)
・ プローブ
5’-FAM-CCGCGCTCGTCGTCGACAAC-TAMRA-3’ (配列番号:8)
前記配列番号:8で表されるプローブは、5’末端がFAM(レポーター色素)で修飾されており、3’末端がTAMRA(クエンチャー色素)で修飾されている。
本発明のプライマー対及びプローブを用いて、下記試験例1-1~1-4のリアルタイムPCRを行った。
<試験例1-1>
コントロールDNAとして、前記調製例1で調製したADAMTS5遺伝子のcDNAを、1.0μg/mL、1.0×10-3μg/mL、又は1.0×10-6μg/mLとなるように希釈したものを使用した。
プライマー対及びプローブとして、前記調製例2で調製したADAMTS5遺伝子用プライマー対(配列番号:2、3)及びプローブ(配列番号:4)を用いた。
リアルタイムPCRは、ABI PRISM 7000(サーモフィッシャーサイエンティフィック社製)を用い、表1-1に記載の反応試薬、及び以下に記載の反応時間の反応条件で行った。
以下の順に行った。
(1) 95℃、30秒間
(2) サイクル数:50
条件:95℃、15秒間、次いで、60℃、60秒間
前記増幅曲線の結果から、コントロールDNAの濃度を縦軸に、Ct値(Thresholdに相当するサイクル数)を横軸にとり、検量線を作成した。
健常人のcDNAを2種(以下、「サンプル1」、「サンプル2」と称することがある)用意し、これを2倍希釈し、サンプルとした。
プライマー対及びプローブとして、前記調製例2で調製したADAMTS5遺伝子用プライマー対(配列番号:2、3)及びプローブ(配列番号:4)を用いた。
リアルタイムPCRの反応条件は、前記試験例1-1におけるコントロールDNAを前記サンプル1又は2に代えた以外は、前記試験例1-1と同様にしてリアルタイムPCRを行った。
コントロールDNAとして、前記調製例3で調製したβ-アクチン遺伝子のcDNAを、1.0μg/mL、1.0×10-3μg/mL、又は1.0×10-6μg/mLとなるように希釈したものを使用した。
プライマー対及びプローブとして、前記調製例4で調製したβ-アクチン遺伝子用プライマー対(配列番号:6、7)及びプローブ(配列番号:8)を用いた。
リアルタイムPCRは、ABI PRISM 7000(サーモフィッシャーサイエンティフィック社製)を用い、表1-2に記載の反応試薬、及び以下に記載の反応時間の反応条件で行った。
以下の順に行った。
(1) 95℃、30秒間
(2) サイクル数:50
条件:95℃、15秒間、次いで、60℃、60秒間
前記増幅曲線の結果から、コントロールDNAの濃度を縦軸に、Ct値(Thresholdに相当するサイクル数)を横軸にとり、検量線を作成した。
健常人のcDNAを2種(サンプル1、サンプル2)用意し、これを50倍希釈し、サンプルとした。
プライマー対及びプローブとして、前記調製例4で調製したβ-アクチン遺伝子用プライマー対(配列番号:6、7)及びプローブ(配列番号:8)を用いた。
リアルタイムPCRの反応条件は、前記試験例1-3におけるコントロールDNAを前記サンプル1又は2に代えた以外は、前記試験例1-3と同様にしてリアルタイムPCRを行った。
市販のプライマー対及びプローブ(MGBプローブ)を用いて、下記試験例2-1~2-4のリアルタイムPCRを行った。
<試験例2-1>
コントロールDNAとして、前記調製例1で調製したADAMTS5遺伝子のcDNAを、1.0μg/mL、1.0×10-3μg/mL、又は1.0×10-6μg/mLとなるように希釈したものを使用した。
プライマー対及びプローブとして、サーモフィッシャーサイエンティフィック社製のADAMTS5遺伝子用プライマー対及びプローブセット(カタログ番号 Hs00199841_m1;プローブは、MGBプローブ;プライマー及びプローブの配列は未公開)を用いた。
リアルタイムPCRは、ABI PRISM 7000(サーモフィッシャーサイエンティフィック社製)を用い、表2-1に記載の反応試薬、及び以下に記載の反応時間の反応条件で行った。
以下の順に行った。
(1) 50℃、30秒間
(2) 95℃、10分間
(3) サイクル数:50

リアルタイムPCRは、ABI PRISM 7000(サーモフィッシャーサイエンティフィック社製)を用い、表2-2に記載の反応試薬、及び以下に記載の反応時間の反応条件で行った。
以下の順に行った。
(1) 50℃、30秒間
(2) 95℃、10分間
(3) サイクル数:50
条件:95℃、15秒間、次いで、60℃、60秒間

リアルタイムPCRの反応条件は、前記試験例2-3におけるコントロールDNAを前記サンプル1又は2に代えた以外は、前記試験例2-3と同様にしてリアルタイムPCRを行った。
したがって、本発明のプライマー対及びプローブの組合せを用いることにより、β-アクチン mRNA量に対するADAMTS5 mRNA量の相対比を、従来の高コストであるMGBプローブを用いる場合よりも低コストで、かつ同一の性能で定量できることが示された。
前記調製例2で調製したADAMTS5遺伝子用プライマー対(配列番号:2、3)及びプローブ(配列番号:4)と、前記市販のADAMTS5遺伝子用プライマー対及びプローブセット(サーモフィッシャーサイエンティフィック社製、カタログ番号 Hs00199841_m1;プローブは、MGBプローブ;プライマー及びプローブの配列は未公開)とについて、リアルタイムPCRの性能の比較を以下のようにして行った。
前記試験例1-1と同様にしてリアルタイムPCRを行った。なお、各濃度のコントロールDNAについて、リアルタイムPCRを5回行い、それぞれについてCt値を求めた。結果を表4に示す。
前記試験例2-1と同様にしてリアルタイムPCRを行った。なお、各濃度のコントロールDNAについて、リアルタイムPCRを5回行い、それぞれについてCt値を求めた。結果を表4に示す。
したがって、本発明のプライマー対及びプローブの組合せを用いることにより、ADAMTS5 mRNA量の絶対量を、従来のMGBプローブを用いる場合よりも高い感度で検出できることが示された。

本発明のプライマー対、プライマー対とプローブとの組合せ、及びキットは、本発明のADAMTS5遺伝子の定量方法などに好適に利用可能である。
具体例としては、内在性コントロールとしてβ-アクチンを用いた場合における、ADAMTS5のmRNAの発現量(ADAMTS5のmRNAの発現量/β-アクチンのmRNAの発現量)が、0.6×1/25より小さい場合には、関節リウマチに対してインフリキシマブが有効であると評価することができる。また、内在性コントロールとしてβ-アクチンを用いた場合における、ADAMTS5のmRNAの発現量(ADAMTS5のmRNAの発現量/β-アクチンのmRNAの発現量)が、4.0×10-4以上の場合には、関節リウマチに対してアダリムマブが有効であると評価することができる。

- 定量PCRにより、試料中のADAMTS5遺伝子の量を測定する工程を含み、
前記ADAMTS5遺伝子が含まれる領域を増幅する増幅用プライマー対として、下記配列番号:2で表される塩基配列からなるプライマーと、下記配列番号:3で表される塩基配列からなるプライマーとを用いることを特徴とするADAMTS5遺伝子の定量方法。
5’-CTAAGCCCTGGTCCAAATGC-3’ (配列番号:2)
