本発明で使用される酸化剤は、反応が進行する限りは、いずれの酸化剤でもよい。対応する原料化合物(スルフィド誘導体)を目的化合物(スルホキシド誘導体)に酸化できる酸化剤を用いることができる。本発明で使用される酸化剤の例は、無機過酸化物(例えば、過酸化水素、尿素-過酸化水素付加体等)等を含むが、これらに限定されるものではない。安全性、反応性、選択性および経済効率等の観点から、好ましい酸化剤は過酸化水素である。酸化剤は、単独でまたは任意の割合の2種以上の組み合わせで使用してもよい。

本発明における触媒は、金属-配位子錯体である。金属-配位子錯体は、金属化合物と配位子から調製することができる。加えて、金属-配位子錯体は、金属化合物および配位子以外の成分を含んでいてもよい。したがって、金属-配位子錯体は、金属化合物配位子を含む。

本発明の方法における一般式(4)で表される安息香酸化合物の使用量は、反応が進行する限りは、いずれの量でもよい。一つの態様において、収率および経済効率等の観点から、本発明における一般式(4)で表される安息香酸化合物の使用量としては、一般式(2)で表されるスルフィド誘導体(原料化合物)1モルに対して、0.1~20mol%、好ましくは3~20mol%、より好ましくは3~15mol%、さらに好ましくは3~10mol%、特に好ましくは3~5mol%を例示することができる。さらに、別の態様において、上記と同様の観点から、本発明における一般式(4)で表される安息香酸化合物の使用量としては、一般式(2)で表されるスルフィド誘導体(原料化合物)1モルに対して、好ましくは1~20mol%、より好ましくは1~15mol%、さらに好ましくは1~10mol%、特に好ましくは1~5mol%もまた例示することができる。しかしながら、本発明における一般式(4)で表される安息香酸化合物の使用量は、目的と状況に応じて、当業者により適宜調整されることができる。
金属化合物および配位子として一般式(3)で表される化合物、並びに一般式(4)で表される安息香酸化合物を所定の溶媒中撹拌して、触媒溶液を調製することができる。本発明の触媒溶液の調製における溶媒としては、下記の「本発明の反応における溶媒」の溶媒が挙げられる。本発明の触媒溶液の調製における温度は、特に制限されないが、触媒の安定性等の観点から、0℃~50℃、好ましくは15~30℃(室温)の範囲を例示できる。本発明の触媒溶液の調製における時間は、特に制限されないが、触媒の安定性等の観点から、10分~12時間、より好ましくは10分~2時間、さらに好ましくは10分~1時間の範囲を例示できる。その具体例は30分、1時間および2時間、好ましくは30分および1時間、さらに好ましくは30分を含む。しかしながら、反応が進行する限りは、触媒溶液を調製する方法は、当業者により適宜選択かつ調整されることができる。
一般式(2)で表されるスルフィド誘導体および上記で調製した触媒溶液並びに酸化剤を所定の溶媒中反応させることにより、一般式(1)で表されるスルホキシド誘導体を製造することができる。しかしながら、反応が進行する限りは、本発明の方法は実施例および上記に示される触媒溶液を先に調製する方法に限定されるものではない。原料、試薬、溶媒等を加える順番は、当業者により適宜選択かつ調整されることができる。
反応の円滑な進行等の観点から、本発明の反応は溶媒の存在下で実施することが好ましい。本発明の反応における溶媒は、反応が進行する限りは、いずれの溶媒でもよい。

本発明の反応時間は、特に制限されない。一つの態様においては、収率の向上、副生成物の抑制、および経済効率等の観点から、0.5時間~120時間、好ましくは1時間~72時間、より好ましくは1時間~48時間、さらに好ましくは1時間~24時間の範囲を例示できる。別の態様においては、上記と同様の観点から、6時間~120時間、好ましくは8時間~72時間、より好ましくは8時間~48時間、さらに好ましくは8時間~24時間の範囲もまた例示できる。しかしながら、本発明の反応時間は当業者により適切に調整されることができる。
本明細書中、用語「反応収率」と用語「収率」は、それぞれ下記の意味を有する。
本発明では、反応混合物の有機層を下記のHPLC分析条件(A)により分析して、反応収率を求めた。本明細書では、反応収率を目的化合物のHPLC面積百分率で示す。
本発明における収率は、一般式(2)で表されるスルフィド誘導体(原料化合物)のモル数に対する、得られた一般式(1)で表されるスルホキシド化合物(目的化合物)のモル数から計算することができる。すなわち、本発明における収率は、以下の式で表される:

スクリューキャップ付きバイアルにFe(acac)3(1.8mg、0.005mmol)、式(3-4)の化合物(1.0mg、0.005mmol)、酢酸ナトリウム(2.1mg、0.025mmol)、およびジクロロメタン(1mL)を加えた。混合物を室温で30分間撹拌した。
(2)標題化合物の製造
5-トリフルオロメチルチオペンチル-[4-クロロ-2-フルオロ-5-(2,2,2-トリフルオロエチルチオ)フェニル]エーテル(215.4mg、0.500mmol)をジクロロメタン(1.0mL)に溶解した。そこに上記(1)で調製した触媒溶液を加えた。混合物を0℃に冷却した。そこに30%過酸化水素(113.4mg、1.0mmol)を加えた。混合物を0℃で16時間撹拌した。反応混合物の有機層をHPLC(面積百分率)により分析した。その結果、反応混合物中の溶媒等を除く成分は以下の通りであった;
5-トリフルオロメチルチオペンチル-[4-クロロ-2-フルオロ-5-(2,2,2-トリフルオロエチルスルフィニル)フェニル]エーテル(目的化合物):19%、
5-トリフルオロメチルチオペンチル-[4-クロロ-2-フルオロ-5-(2,2,2-トリフルオロエチルチオ)フェニル]エーテル(原料化合物):79%、
5-トリフルオロメチルチオペンチル-[4-クロロ-2-フルオロ-5-(2,2,2-トリフルオロエチルスルホニル)フェニル]エーテル(過剰酸化による副生成物):0%。

5-トリフルオロメチルチオペンチル-[4-クロロ-2-フルオロ-5-(2,2,2-トリフルオロエチルスルホニル)フェニル]エーテル(過剰酸化による副生成物):0%。
5-トリフルオロメチルチオペンチル-[4-クロロ-2-フルオロ-5-(エチルスルフィニル)フェニル]エーテルの製造
スクリューキャップ付きバイアルにFe(acac)3(1.8mg、0.005mmol)、式(3-4)の化合物(1.0mg、0.005mmol)、式(4-1)の化合物(2,6-ジメトキシ安息香酸ナトリウム;5.1mg、0.025mmol)、およびジクロロメタン(1mL)を加えた。混合物を室温で30分間撹拌した。
(2)標題化合物の製造
5-トリフルオロメチルチオペンチル-[4-クロロ-2-フルオロ-5-(エチルチオ)フェニル]エーテル(188.4mg、0.500mmol)をジクロロメタン(1.0mL)に溶解した。そこに上記(1)で調製した触媒溶液を加えた。混合物を0℃に冷却した。そこに30%過酸化水素(113.4mg、1.0mmol)を加えた。混合物を0℃で24時間撹拌した。反応混合物の有機層をHPLC(面積百分率)により分析した。その結果、反応混合物中の溶媒等を除く成分は以下の通りであった;
5-トリフルオロメチルチオペンチル-[4-クロロ-2-フルオロ-5-(エチルスルフィニル)フェニル]エーテル(目的化合物):39%、
5-トリフルオロメチルチオペンチル-[4-クロロ-2-フルオロ-5-(エチルチオ)フェニル]エーテル(原料化合物):55%、
5-トリフルオロメチルチオペンチル-[4-クロロ-2-フルオロ-5-(エチルスルホニル)フェニル]エーテル(過剰酸化による副生成物):0%。
生成物のLC/MS(目的化合物);Exact Mass:392.0295、実測値(ポジティブ):393.0370
メチルフェニルスルホキシドの製造
スクリューキャップ付きバイアルにFe(acac)3(1.8mg、0.005mmol)、式(3-4)の化合物(1.0mg、0.005mmol)、式(4-1)の化合物(2,6-ジメトキシ安息香酸ナトリウム;5.1mg、0.025mmol)、およびジクロロメタン(1mL)を加えた。混合物を室温で30分間撹拌した。
(2)標題化合物の製造
チオアニソール(62.1mg、0.500mmol)をジクロロメタン(1.0mL)に溶解した。そこに上記(1)で調製した触媒溶液を加えた。混合物を0℃に冷却した。そこに30%過酸化水素(113.4mg、1.0mmol)を加えた。混合物を0℃で20時間撹拌した。反応混合物の有機層をHPLC(面積百分率)により分析した。その結果、反応混合物中の溶媒等を除く成分は以下の通りであった;
メチルフェニルスルホキシド(目的化合物):16%、
チオアニソール(原料化合物):77%、
メチルフェニルスルホン(過剰酸化による副生成物):7%。
副生成物のLC/MS;Exact Mass:156.0、実測値(ポジティブ):157.1
5-トリフルオロメチルチオペンチル-[4-クロロ-2-フルオロ-5-(2,2,2-トリフルオロエチルスルフィニル)フェニル]エーテルの製造
5-トリフルオロメチルチオペンチル-[4-クロロ-2-フルオロ-5-(2,2,2-トリフルオロエチルチオ)フェニル]エーテル(215.4mg、0.500mmol)をアセトニトリル(1.0mL)に溶解した。そこにタングステン酸ナトリウム二水和物(8.2mg、0.025mmol)を加えた。混合物を室温で撹拌しながら、そこに30%過酸化水素(113.4mg、1.0mmol)を加えた。混合物を室温で15時間撹拌した。反応混合物の有機層をHPLC(面積百分率)により分析した。その結果、反応混合物中の溶媒等を除く成分は以下の通りであった;
5-トリフルオロメチルチオペンチル-[4-クロロ-2-フルオロ-5-(2,2,2-トリフルオロエチルスルフィニル)フェニル]エーテル(目的化合物):61%、
5-トリフルオロメチルチオペンチル-[4-クロロ-2-フルオロ-5-(2,2,2-トリフルオロエチルチオ)フェニル]エーテル(原料化合物):32%、
5-トリフルオロメチルチオペンチル-[4-クロロ-2-フルオロ-5-(2,2,2-トリフルオロエチルスルホニル)フェニル]エーテル(過剰酸化による副生成物):2%。
(2)追加の過酸化水素を用いた標題化合物の製造
上記で得られた反応混合物に、30%過酸化水素(113.4mg、1.0mmol)を加えた。混合物を室温で23時間撹拌した。反応混合物の有機層をHPLC(面積百分率)により分析した。その結果、反応混合物中の溶媒等を除く成分は以下の通りであった;
5-トリフルオロメチルチオペンチル-[4-クロロ-2-フルオロ-5-(2,2,2-トリフルオロエチルスルフィニル)フェニル]エーテル(目的化合物):79%、
5-トリフルオロメチルチオペンチル-[4-クロロ-2-フルオロ-5-(2,2,2-トリフルオロエチルチオ)フェニル]エーテル(原料化合物):0%、
5-トリフルオロメチルチオペンチル-[4-クロロ-2-フルオロ-5-(2,2,2-トリフルオロエチルスルホニル)フェニル]エーテル(過剰酸化による副生成物):12%。
本明細書において上述したように、本発明の方法は経済的であり、環境にも優しく、高い工業的な利用価値を有する。
特に、本発明の方法では、スルホン誘導体への過剰酸化を回避して、目的とするスルホキシド誘導体を選択的に製造することができる。
したがって、本発明には除去が難しい副生成物のスルホン誘導体を除去する必要がないという利点がある。
Claims (17)
- 一般式(1):
R1が、C1~C10アルキル基、
C3~C6シクロアルキルC1~C6アルキル基、ここで当該C3~C6シクロアルキル基部分はハロゲン原子、C1~C4アルキル基、C1~C4アルコキシ基またはC1~C4ハロアルキル基によりモノ置換またはポリ置換されていてもよい、
フェニルC1~C6アルキル基、ここで当該フェニル基部分はハロゲン原子、C1~C4アルキル基、C1~C4アルコキシ基、C1~C6ハロアルキル基、シアノ基またはニトロ基によりモノ置換またはポリ置換されていてもよい、
C1~C4アルコキシC2~C10アルキル基、
C1~C4ハロアルコキシC2~C10アルキル基、
C1~C4アルキルチオC2~C10アルキル基、
C1~C4アルキルスルフィニルC2~C10アルキル基、
C1~C4アルキルスルホニルC2~C10アルキル基、
C1~C4ハロアルキルチオC2~C10アルキル基、
C1~C4ハロアルキルスルフィニルC2~C10アルキル基、
C1~C4ハロアルキルスルホニルC2~C10アルキル基、
C1~C6ハロアルキル基、
C3~C6シクロアルキルC1~C6ハロアルキル基、ここで当該C3~C6シクロアルキル基部分はハロゲン原子、C1~C4アルキル基、C1~C4アルコキシ基またはC1~C6ハロアルキル基によりモノ置換またはポリ置換されていてもよい、
フェニルC1~C6ハロアルキル基、ここで当該フェニル基部分はハロゲン原子、C1~C4アルキル基、C1~C4アルコキシ基、C1~C4ハロアルキル基、シアノ基またはニトロ基によりモノ置換またはポリ置換されていてもよい、または
C1~C4ハロアルキルチオC1~C6ハロアルキル基であり;
R2およびR3は、それぞれ独立して、水素原子、ハロゲン原子またはC1~C4アルキル基である。)で表されるスルホキシド誘導体の製造方法であって、一般式(2):
で表されるスルフィド誘導体を、金属化合物および配位子としての一般式(3):
R4およびR5は、それぞれ独立して、水素原子、ハロゲン原子、C1~C6アルキル基、フェニルC1~C6アルキル基、C6~C10アリール基、シアノ基、ニトロ基またはC1~C6アルコキシ基であり;
R6は、C1~C4アルキル基、シアノ基、ニトロ基、カルボキシ基、C1~C4アルコキシカルボニル基、C1~C4アルキルカルボニル基、ヒドロキシC1~C4アルキル基、C1~C4アルコキシC1~C4アルキル基、アミノC1~C4アルキル基、シアノC1~C4アルキル基、ニトロC1~C4アルキル基、カルボキシC1~C4アルキル基またはC1~C4アルコキシカルボニルC1~C4アルキル基であり;
R71およびR72は、それぞれ独立して、水素原子、C1~C6アルキル基、フェニルC1~C6アルキル基またはC6~C10アリール基である。但し、R71およびR72の両方が水素原子である場合を除く。)
で表される化合物を含む金属-配位子錯体である触媒、ならびに一般式(4):
A1がC1~C2アルコキシ基であり;
A2が水素原子であり;
A3が水素原子またはC1~C2アルコキシ基であり;
A4が水素原子であり;
A5がC1~C2アルコキシ基であり;
Mは、水素原子、アルカリ金属原子またはアルカリ土類金属原子であり;そして
nは1または2である。)
で表される安息香酸化合物の存在下で、酸化剤と反応させることを特徴とする方法。 - R1が、C1~C10アルキル基、
C3~C6シクロアルキルC1~C6アルキル基、ここで当該C3~C6シクロアルキル基部分はハロゲン原子またはC1~C4アルキル基によりモノ置換またはポリ置換されていてもよい、
フェニルC1~C6アルキル基、ここで当該フェニル基部分はハロゲン原子またはC1~C4アルキル基によりモノ置換またはポリ置換されていてもよい、
C1~C4アルコキシC2~C10アルキル基、
C1~C4ハロアルコキシC2~C10アルキル基、
C1~C4アルキルチオC2~C10アルキル基、
C1~C4アルキルスルフィニルC2~C10アルキル基、
C1~C4アルキルスルホニルC2~C10アルキル基、
C1~C4ハロアルキルチオC2~C10アルキル基、
C1~C4ハロアルキルスルフィニルC2~C10アルキル基、
C1~C4ハロアルキルスルホニルC2~C10アルキル基、
C1~C6ハロアルキル基、
C3~C6シクロアルキルC1~C6ハロアルキル基、ここで当該C3~C6シクロアルキル基部分はハロゲン原子またはC1~C4アルキル基によりモノ置換またはポリ置換されていてもよい、または
フェニルC1~C6ハロアルキル基、ここで当該フェニル基部分はハロゲン原子またはC1~C4アルキル基によりモノ置換またはポリ置換されていてもよい、である、請求項1に記載の方法。 - R1が、C1~C10アルキル基、
C1~C4アルコキシC2~C10アルキル基、
C1~C4ハロアルコキシC2~C10アルキル基、
C1~C4アルキルチオC2~C10アルキル基、
C1~C4アルキルスルフィニルC2~C10アルキル基、
C1~C4アルキルスルホニルC2~C10アルキル基、
C1~C4ハロアルキルチオC2~C10アルキル基、
C1~C4ハロアルキルスルフィニルC2~C10アルキル基、
C1~C4ハロアルキルスルホニルC2~C10アルキル基、または
C1~C6ハロアルキル基である、請求項1に記載の方法。 - R1がC1~C4ハロアルキルチオC2~C10アルキル基であり;そして
R2およびR3が、それぞれ独立して、ハロゲン原子またはC1~C4アルキル基である、請求項1に記載の方法。 - R1が5-トリフルオロメチルチオペンチル基または6-トリフルオロメチルチオヘキシル基であり;そして
R2がフッ素原子でありかつR3が塩素原子であるか、またはR2およびR3がメチル基である、請求項1に記載の方法。 - R1が5-トリフルオロメチルチオペンチル基であり;
R2がフッ素原子であり;そして
R3が塩素原子である、請求項1に記載の方法。 - R1が6-トリフルオロメチルチオヘキシル基であり;そして
R2およびR3がメチル基である、請求項1に記載の方法。 - 金属化合物が鉄化合物であり;
R4が水素原子であり;
R5が水素原子またはハロゲン原子であり;
R6がヒドロキシメチル基であり;そして
R71およびR72が、それぞれ独立して、水素原子またはC1~C6アルキル基である(但し、R71およびR72の両方が水素原子である場合を除く。)、請求項1~7のいずれか1項に記載の方法。 - 金属化合物が鉄化合物であり;
R4が水素原子であり;
R5が水素原子、塩素原子または臭素原子であり;
R6がヒドロキシメチル基であり;そして
R71がメチル基でありかつR72がメチル基であるか、またはR71が水素原子でありかつR72がイソプロピル基である、請求項1~7のいずれか1項に記載の方法。 - 金属化合物が鉄化合物であり;
R4が水素原子であり;
R5が水素原子または塩素原子であり;
R6がヒドロキシメチル基であり;そして
R71がメチル基でありかつR72がメチル基であるか、またはR71が水素原子でありかつR72がイソプロピル基である、請求項1~7のいずれか1項に記載の方法。 - 金属化合物が鉄化合物であり;
R4が水素原子であり;
R5が水素原子であり;
R6がヒドロキシメチル基であり;
R71がメチル基であり;そして
R72がメチル基である、請求項1~7のいずれか1項に記載の方法。 - 金属化合物が鉄化合物であり;
