Description
(式中、R1及びR2は一緒になって、それが結合する窒素原子と共に、置換されてもよい飽和ヘテロ環基を表す。ただし、(1)次の(a)または(b)であるときはR1及びR2は一緒になって、それが結合する窒素原子と共に、置換されてもよい飽和ヘテロ環基を表すことはない。(a) R5、R6、R7およびR8が全て水素原子である。(b)R5、R6、R7およびR8のうち、1~2個がそれぞれ独立してハロゲン原子であり、残りが水素原子である。XはO、S、NR10、又はCR11R12を表す。他の記号は当該公報参照。)
即ち、本発明は式(I)の化合物又はその塩、並びに、式(I)の化合物又はその塩、及び賦形剤を含有する医薬組成物に関する。

なお、当該医薬組成物は式(I)の化合物又はその塩を含有する認知機能障害、統合失調症陰性症状、CIAS、パーキンソン病、アルツハイマー病、ハンチントン病、うつ病、ADHD、又は、薬物依存の予防及び/又は治療剤を包含する。
また、本発明は、
(1)認知機能障害、統合失調症陰性症状、CIAS、パーキンソン病、アルツハイマー病、ハンチントン病、うつ病、ADHD、又は、薬物依存の予防及び/又は治療用医薬組成物の製造のための式(I)の化合物又はその塩の使用、
(2)認知機能障害、統合失調症陰性症状、CIAS、パーキンソン病、アルツハイマー病、ハンチントン病、うつ病、ADHD、又は、薬物依存の予防及び/又は治療のための式(I)の化合物又はその塩の使用、
(3)認知機能障害、統合失調症陰性症状、CIAS、パーキンソン病、アルツハイマー病、ハンチントン病、うつ病、ADHD、又は、薬物依存の予防及び/又は治療のための式(I)の化合物又はその塩、及び、

「アルキル」及び「アルキレン」とは、いずれも直鎖又は分枝状の飽和炭化水素鎖を意味し、それぞれ一価基、二価基を意味する。
(1) ある態様として、R1、R2及びR3が、それぞれ同一又は異なって、H又はハロゲンである式(I)の化合物又はその塩。
(2) ある態様として、R1がHである式(I)の化合物又はその塩。
(3) ある態様として、R2がH又はハロゲンである式(I)の化合物又はその塩。又はある態様として、R2がH、F又はClである式(I)の化合物又はその塩。又はある態様として、R2がHである式(I)の化合物又はその塩。

また、式(I)の化合物には、不斉炭素原子を有する場合があり、これに基づく光学異性体が存在しうる。本発明は、式(I)の化合物の光学異性体の分離されたもの、あるいはそれらの混合物も包含する。
「ポジティブアロステリックモジュレーター(PAM)」とは、PAM作用を有する化合物を意味する。例えば、試験例1において、ドパミン用量反応曲線を左方シフトさせる化合物を意味する。
PAMは、それ自体では受容体機能を増強しないが、リガンド存在下において、受容体機能を増強する。
「統合失調症陰性症状」とは統合失調症における陰性症状である。
「統合失調症認知機能障害(Cognitive impairment associated with schizophrenia、CIAS)」とは、統合失調症に伴う認知機能障害である。
式(I)の化合物及びその塩は、その基本構造あるいは置換基の種類に基づく特徴を利用し、種々の公知の合成法を適用して製造することができる。その際、官能基の種類によっては、当該官能基を原料から中間体へ至る段階で適当な保護基(容易に当該官能基に転化可能な基)に置き換えておくことが製造技術上効果的な場合がある。このような保護基としては、例えば、Peter G. M. Wuts, 及びTheodora W. Greene著、「Greene's Protective Groups in Organic Synthesis」, 第4版, 2006年に記載の保護基等を挙げることができ、これらの反応条件に応じて適宜選択して用いればよい。このような方法では、当該保護基を導入して反応を行ったあと、必要に応じて保護基を除去することにより、所望の化合物を得ることができる。
また、式(I)の化合物のプロドラッグは、上記保護基と同様、原料から中間体へ至る段階で特定の基を導入、あるいは得られた式(I)の化合物を用いてさらに反応を行うことで製造できる。反応は通常のエステル化、アミド化、脱水等、当業者に公知の方法を適用することにより行うことができる。
以下、式(I)の化合物の代表的な製造法を説明する。各製法は、当該説明に付した参考文献を参照して行うこともできる。なお、本発明の製造法は以下に示した例には限定されない。

式(I)の化合物は、化合物(1)及び化合物(2)から製造できる。
本反応では、化合物(1)と化合物(2)とを等量又は一方を過剰量用い、これらの混合物を、縮合剤の存在下、反応に不活性な溶媒中、冷却下~加熱下、好ましくは-20℃~60℃において、通常0.1時間~5日間撹拌する。溶媒は例えばトルエン等の芳香族炭化水素類、ジクロロメタン等のハロゲン化炭化水素類、THF等のエーテル類、DMF、NMP、DMSO、EtOAc、MeCN又は水、及びこれらの混合物が挙げられる。縮合剤は、HATU、WSC、WSC・HCl、DCC、CDI、DPPA等が挙げられる。添加剤(例えば、HOBt)を用いることが反応に好ましい場合がある。本反応をTEA、DIPEA若しくはNMM等の有機塩基、又はK2CO3、Na2CO3若しくはKOH等の無機塩基の存在により、反応が更に円滑に進行する場合がある。

化合物(I-1)は、化合物(3)から製造できる。
本反応は水酸基のハロゲン化である。反応に不活性な溶媒中、化合物(3)にPPh3及びハロゲン源を加え、通常20℃~100℃、0.5時間~8時間撹拌する。溶媒は例えばTHF等のエーテル系溶媒等が挙げられる。ハロゲン源は、例えば、ビス(2-メトキシエチル)アミノサルファートリフルオリド、CCl4、CBr4、I2が挙げられる。なお、PPh3とCCl4を用いたCl化はいわゆるAppel反応である。

原料化合物(2a)は、化合物(4)及び化合物(5a)から製造できる。原料化合物(2b)は、化合物(4)及び化合物(5b)から製造できる。脱離基は、例えばHal(以下、HalはCl、Br又はIを示す)、MsO基、TsO基等が挙げられる。Pg1は、例えばベンジル基、BOC基が挙げられる。
本反応では、化合物(4)と化合物(5a)とを等量若しくは一方を過剰量用い、これらの混合物を、反応に不活性な溶媒中、又は無溶媒下、冷却下~加熱還流下、好ましくは0℃~80℃において、通常0.1時間~5日間撹拌する。溶媒は例えば芳香族炭化水素類、エーテル類、ハロゲン化炭化水素類、DMF、DMSO、EtOAc、MeCN及びこれらの混合物が挙げられる。TEA、DIPEA若しくはNMM等の有機塩基、又はKOtBu、K2CO3、Na2CO3若しくはNaOH等の無機塩基の存在により、反応が更に円滑に進行する場合がある。

本反応は原料製法1と同様の方法で行うことができる。化合物(6)を反応に不活性な溶媒中、化合物(7)を加え、塩基の存在下、通常室温~加温し、1時間~2日撹拌する。塩基は、通常無機塩基が用いられ、例えばK2CO3、Na2CO3、Cs2CO3若しくはKOH等が挙げられる。溶媒は例えばMeCN等が挙げられる。

化合物(4)は、化合物(8)及び化合物(9)から製造できる。
本反応では、化合物(8)と化合物(9)を反応に不活性な溶媒中、100℃以上又は還流条件下で、12時間~3日間撹拌する。溶媒は例えばトルエン等の芳香族炭化水素類等が挙げられる。

化合物(13)は、化合物(11)及び化合物(12)から製造できる。
脱離基Lvは、例えばハロゲンや-OSO2CF3が挙げられる。
本反応は、反応に不活性な溶媒中、化合物(11)及び化合物(12)に、塩基の存在下、パラジウムなどの金属とPPh3などの配位子を触媒として加え、通常20℃~140℃、5時間~4日間撹拌する。触媒は例えばPd2dba3等が挙げられる。塩基は例えばビス(トリメチルシリル)ナトリウムアミドやNaOtBu等が挙げられる。溶媒は例えばジオキサン等が挙げられる。いわゆるバックワルド・ハートウィグ クロスカップリング(Buchwald-Hartwig Cross Coupling)である。

化合物(2c)は、化合物(13)の還元により製造できる。
本反応は、通常のニトロ基の還元方法であり、塩酸や酢酸等の酸溶媒中、化合物(13)に鉄又はスズ等を加え、通常50℃~140℃で、0.5時間から3日間撹拌する。
(式中、(RP、RP')は、(イミダゾリル、イミダゾリル)、(Cl3C-O-、-O-CCl3)、(Cl、-O-CCl3)(Cl、-O-フェニル)、(Cl、-O-パラニトロフェニル)の組み合わせを示す。)
化合物(16)は、化合物(14)及び化合物(15)から製造できる。
本反応では化合物(14)を反応に不活性な溶媒中、等量若しくは過剰量の化合物(15)を加え、この混合物を、冷却下~加熱下、好ましくは50℃~150℃で0.1時間~1日程度撹拌する。溶媒は、例えばハロゲン化炭化水素類、芳香族炭化水素類、エーテル類、DMF、DMSO、EtOAc、MeCN又はこれらの混合溶媒が挙げられる。化合物(15)はカルボニル化試薬ともいい、例えば、CDI、ジホスゲン、トリホスゲンだけでなく、クロロギ酸 4-ニトロフェニル、クロロギ酸フェニルも挙げられる。中間体であるフェニルカルボナート体が安定である場合(例えば、クロロギ酸 4-ニトロフェニル又はクロロギ酸フェニルを用いた場合)には、これを一旦単離し、再度反応を行うこともできる。

化合物(1)は、化合物(16)及び化合物(17)から製造できる。
化合物(1)のPg3エステル体は、化合物(16)及び化合物(17)から製造できる。脱離基は、例えばHalの他に、MsO基、TsO基等を用いることもできる。保護基Pg3は例えばMe基、Et基、tBu基等が挙げられる。
本反応では、化合物(16)と化合物(17)とを等量若しくは一方を過剰量用い、これらの混合物を、塩基の存在下、撹拌する。溶媒は、MeCN等が挙げられる。TBAB等の相間移動触媒の存在により、反応がより円滑に進行する場合がある。
化合物(1)は、得られた化合物(1)のPg3エステル体から、Pg3を脱保護し製造する。脱保護は例えばPeter G. M. Wuts, 及びTheodora W. Greene著、「Greene's Protective Groups in Organic Synthesis」, 第4版, 2006年を参照して行うことができる。
単離、精製は、抽出、分別結晶化、各種分画クロマトグラフィー等、通常の化学操作を適用して行われる。
各種の異性体は、適当な原料化合物を選択することにより製造でき、あるいは異性体間の物理化学的性質の差を利用して分離することができる。例えば、光学異性体は、ラセミ体の一般的な光学分割法(例えば、光学活性な塩基又は酸とのジアステレオマー塩に導く分別結晶化や、キラルカラム等を用いたクロマトグラフィー等)により得られ、また、適当な光学活性な原料化合物から製造することもできる。
本明細書に記載される粉末X線回折パターンは、結晶の同一性認定においては、データの性質上、結晶格子間隔や全体的なパターンが重要であり、回折角及び回折強度は結晶成長の方向、粒子の大きさ、測定条件によって多少の誤差が生じうるものであるから、厳密に解されるべきではない。本明細書中、粉末X線回折パターンにおける回折角(2θ(°))は、当該測定法において通常許容される誤差範囲を考慮して解釈され、ある態様としては±0.2°の誤差範囲を取りうる。また、例えば、賦形剤との混合物の状態で測定した場合、賦形剤由来のピークの近傍に存在しベースラインの傾き上にあるピークでは、見かけ上、ピークが±0.3°程度でシフトする場合がありうる。

アッセイ用緩衝液=1mM IBMXを含有するαMEM。
希釈用緩衝液=1mM IBMX及び0.8mMアスコルビン酸を含有するαMEM。
ポジティブアロステリックモジュレート作用(PAM作用)の評価
本発明化合物のPAM作用は、ドパミン用量反応曲線の左方シフトの割合により評価した。
左方シフトとは、用量の対数を横軸(X軸)とし反応(本試験ではcAMP濃度)を縦軸(Y軸)としたドパミンの用量反応曲線が、PAMの添加によりX軸の負の方向に平行移動したことを意味する。
ヒトのドパミンD1受容体(アクセッション番号:NM_000794.3)のコード配列はPCRにより増幅し、pEF-BOSベクターにサブクローニングした。得られた構築物はCHO(dhfr-)細胞(ATCC番号:CRL-9096)にリポフェクタミン2000(Thermo Fisher Scientific Inc.、リポフェクタミンは登録商標)を用いてトランスフェクションした。安定的にトランスフェクションされたクローンは100nM MTXを用いた選択により得られた。得られた細胞クローンは、10% FBS、ペニシリン及びストレプトマイシンを含有するαMEMで維持した。
(1) 本発明化合物を添加したドパミンの用量反応曲線
被験薬物を添加したドパミンの用量反応曲線は下記の実験を行い、作成した。
被験薬物の段階希釈溶液はアッセイ用緩衝液を用いて調製した(濃度0.49~40μM、3倍希釈)。黒色384ウェルプレート(Corning Inc.)に、上記の被験薬物の段階希釈溶液を2.5μLずつ分注した。
ヒトのドパミンD1受容体安定発現CHO細胞を約5000個/ウェルの密度で各ウェル5μLずつ播種し、プレート用遠心機(05PR-22、Hitachi Koki Co., Ltd.、800rpm)で遠沈後に10分間静置した。
ドパミンの段階希釈溶液は、ドパミン(Sigma-Aldrich Co. LLC)のDMSO溶液に希釈用緩衝液を用いて調製した(濃度0.018~13.3μM、3倍希釈)。プレートの各ウェルにドパミンの段階希釈溶液を2.5μLずつ分注した。
プレートシェーカー(mx-5、Sanko Junyaku Co., Ltd.)を用いてプレートを撹拌後、室温で15~20分間静置した(最終濃度:被験薬物0~10μM、ドパミン0~3.3μM、3倍希釈)。
cAMPの測定には、LANCE Ultra cAMP キット(PerkinElmer Inc.、LANCEは登録商標)を用いた。上述のプレートの各ウェルに、キット試薬Eu-cAMP tracer液及びキット試薬ULight-anti-cAMP液(ULightは登録商標)をそれぞれ5μL添加し、反応を停止した。プレートは室温で1時間インキュベートした後、プレートリーダー(2103 Multilabel Plate Reader(登録商標Envision)、PerkinElmer, Inc.)を用い、TR-FRET法で665 nm及び620 nmにおける蛍光強度を測定した(励起波長:320 nm, 蛍光波長:665 nm, 620 nm)。各ウェルの蛍光強度比 [(665 nmでの蛍光強度)/(620 nmでの蛍光強度) × 104] を算出し、解析に用いた。
対照群として、ドパミンを添加しないウェルを0%とし、ドパミン最終濃度3.3μMのウェルを100%とした。
(2)ドパミンの用量反応曲線
ドパミンの用量反応曲線は、被験薬物を無添加にして(1)と同様に実験を行い、作成した。
上記試験の結果、本実施例化合物を添加したドパミンの用量反応曲線(1)が、ドパミンの用量反応曲線(2)を基準として、左方シフトすることが判明した。被験薬物の濃度を変えて各シフト値を算出し、ドパミンの用量反応曲線が用量依存的に左方にシフトしていることも確認した。
なお、ドパミン用量が0の場合、被験薬物はアゴニスト作用を示さなかった。
以上より、本実施例化合物がPAM作用を有することが判明した。
活性の値は、活性の指標として用いたシフト値で表した。
シフト値とは、被験薬物1.1 μMにおけるドパミンのEC50値が、被験薬物無添加におけるドパミンのEC50値を基準として、何倍強力になったかを示す値である。
シフト値=(被験薬物無添加におけるドパミンのEC50値)/(被験薬物1.1 μMにおけるドパミンのEC50値)。
従って、被験薬物の値が1より大きければ、被験薬物を添加することによりドパミンの用量反応曲線が左方シフトしたことを示す。ドパミンのEC50値はそれぞれのドパミンの用量反応曲線を用いて算出した。
表中、No.は化合物番号、Exは実施例化合物番号、Dat1はシフト値を示す。
Y字迷路試験(Y-maze試験)
本発明化合物の認知機能障害に対する改善効果を、自発交替行動の実験系であるY字迷路試験を用いて評価した。
Y字迷路は一本の走路の長さが40 cm、高さが13 cm、床の幅が3 cm、上部の幅が10cmの3本の走路がそれぞれ120度でY字状に接合する迷路を用いた。
5~6週齢のddY系雄性マウス(n=8 Japan SLC, Inc.)に、Y字迷路試験開始の1時間前に被験薬物を単回経口投与(0.01、0.03、0.1および0.3 mg/kg。0.5%メチルセルロースにて懸濁)し、さらに、Y字迷路試験開始の20分前に認知機能障害を引き起こすNMDA受容体拮抗薬であるMK-801(Sigma-Aldrich Co. LLC)を0.15 mg/kgの用量で腹腔内投与した。
なお、対照群のマウスには、被験薬物の代わりにvehicle(0.5 %メチルセルロース)を用い、さらに、MK-801ではなく生理食塩水を用いた。
MK-801対照群のマウスには、被験薬物の代わりにvehicle(0.5 %メチルセルロース)を用いた。
上述のマウスをY字迷路におけるいずれかの走路の末端に置いた後に8分間自由に探索させた。マウスが走路進入した回数を数えて、これを総進入数とした。異なる3本の走路に連続して進入した回数(例えば、3本のアームをそれぞれa、b、cとした際に、進入した走路の順番がabccbacabの場合は4とカウントした)を自発交替行動数とした。自発交替行動率は、計算式:
自発交替行動率= [ 自発交替行動数/(総進入数 - 2) ] × 100
で算出した自発交替行動率を自発交替行動の指標とした。
この指標値が高い程、短期記憶が保持されていたことを示す。
測定値は群毎に平均値±標準誤差で表した。対照群とMK-801対照群との有意差検定はStudentのt検定で行った。また、被験薬物投与群とMK-801対照群との有意差検定は、Dunnett型多重比較検定で行い、被験薬物の学習障害改善作用を判定した。各検定においてP<0.05であれば有意差があると判断した。
注意セットシフト課題(Attentional set-shifting task)
本発明化合物を注意セットシフト課題試験により評価した。
ある手がかりがあれば餌報酬を得るという課題を学習させた後に、その手がかりを次元内にシフトさせた(例えば、手がかりを形から形に変更した)場合、正常動物も、脳の前頭前野に異常を有する病態動物(以下、病態動物という)も手がかりを弁別し餌報酬を得ることができる。しかし、ある手がかりで餌報酬を得る課題を学習させた後に、手がかりを次元外へシフトさせた(例えば、手がかりを形から香りに変更した)場合には、正常動物は餌の手がかりを弁別できるが、病態動物では弁別できない。本試験はこの行動を応用している。
1.試験装置及び動物
試験装置は、ギロチンドアで仕切られた、スタートエリアとチョイスエリアからなるフィールドを用いた。チョイスエリアは、スタートエリアから見て、左と右のエリアが同じ大きさになるように一部仕切られている。左右のエリアに、内容物の入った容器を1個ずつ置き、その中に餌報酬(シリアル片、General Mills Inc.)を埋めた。各試験(試験のスタートから正解又は不正解まで)ごとに左右の容器はランダムに入れ替えた。スタートエリアに置いたラットは、ギロチンドアを開けるとチョイスエリアに移動し、左右のチョイスエリア間を自由に行動することができた。
病態モデルは、亜慢性PCPを投与したラットを用いた。5週齢の雄性ロングエバンスラット(Japan SLC, Inc.)に2 mg/mL/kgのPCP生理食塩水溶液を1日2回で7日間腹腔内投与して作製した。7日または8日のウォッシュアウト期間の後にテストを行った。ウォッシュアウト期間中に摂餌を制限した。
ギロチンドアを開けて試験を開始し、4分間自由に行動させた後、ラットをスタートエリアに戻した。その後、左右のチョイスエリアにそれぞれ1個ずつ容器を置いた。容器はアルファドライ(Shepherd Specialty Papers、アルファドライは登録商標)で満たし、容器の最上部に1個と容器の底に1個の合計2個の餌報酬を置いた。試験を開始し、餌報酬(4個)を完食したことを確認後、ラットをホームケージに戻した。
馴化と同じ日に、トレーニングとして単純弁別(SD1)課題を行った。一方の容器はプラスチックビーズで満たし、縁にナツメグのアロマオイルを塗布した。他方の容器は木製ビーズで満たし、縁にレモンのアロマオイルを塗布した。前者の容器にのみ餌報酬を埋め、試験を開始した。(1)正解として餌報酬を完食した後、又は、(2)不正解の容器を掘削した直後、のいずれかの時にラットをスタートエリアに戻した。ラットが5回連続で正解するまで訓練した。
トレーニングの翌日、内容物または香りを手がかりとしてラットに餌報酬を探索させた。内容物と香りの組み合わせによる6つの弁別課題を行った。各弁別課題で5回連続正解した場合、次の課題に進めた。試験は表の1番目(SD2)から6番目(EDR)まで順に行った。CD終了後、ラットに被験薬物(0.001、0.003および0.01mg/kg, 0.5%メチルセルロースにて懸濁)を単回経口投与した。なお、対照群のラットには被験薬物の代わりにvehicle(0.5%メチルセルロース)を投与した。
表3は各課題における内容物と香りの組み合わせを表した。表4は具体例(一例)を表した。表中の下線は正解(餌報酬が入った容器の手がかり)を示した。
例数は各群6例である。5回連続正解するまでの施行回数について、統計解析を行った。対照群とPCP対照群についてStudentのt検定を行った。EDでのみ有意差が認められ(P<0.05)、PCP対照群において遂行機能障害が検出された。また、被験薬物投与群とPCP対照群についてDunnett型多重比較検定を行い、被験薬物の遂行機能障害改善作用を判定した。弁別課題の1番目(SD2)から4番目(IDR)まで、及び6番目(EDR)では有意差は認められず、弁別課題の5番目(ED)でのみ有意差(P<0.05)が認められた。
投与は錠剤、丸剤、カプセル剤、顆粒剤、散剤、液剤等による経口投与、又は、関節内、静脈内、筋肉内等の注射剤、坐剤、点眼剤、眼軟膏、経皮用液剤、軟膏剤、経皮用貼付剤、経粘膜液剤、経粘膜貼付剤、吸入剤等による非経口投与のいずれの形態であってもよい。
経口投与のための液体組成物は、薬剤的に許容される乳濁剤、溶液剤、懸濁剤、シロップ剤又はエリキシル剤等を含み、一般的に用いられる不活性な希釈剤、例えば精製水又はEtOHを含む。当該液体組成物は不活性な希釈剤以外に可溶化剤、湿潤剤、懸濁剤のような補助剤、甘味剤、風味剤、芳香剤、防腐剤を含有していてもよい。

2θ(°)=粉末X線回折における特徴的ピークの回折角を表す。粉末X線回折は、RINT-TTRII(RIGAKU社)を用い、管球:Cu、管電流:300 mA、管電圧:50 kV、サンプリング幅:0.020°、走査速度:4°/分、波長:1.54056Å、測定回折角範囲(2θ):2.5~40°の条件で測定した。
NaOH (5 g)、水 (7 mL)、TBAB (50 mg)とDMF (25 mL)の混合物に1-(4-フェニル-1H-イミダゾール-2-イル)メタンアミン一塩酸塩 (1 g)を氷冷下加え、更に1,3-ジブロモプロパン (1.98 g)のDMF (25 mL)溶液をゆっくり加えてから、室温で終夜撹拌した。その後、不溶物をろ過で除き、トルエンで洗浄し、ろ液を減圧下濃縮した。残渣をシリカゲルCC (MeOH-クロロホルム)で精製し、2-フェニル-6,7,8,9-テトラヒドロ-5H-イミダゾ[1,2-a][1,4]ジアゼピン (200 mg)を得た。
NaOH (1.1 g)、水 (2 mL)、TBAB (15 mg)とDMF (5 mL)の混合物に1-(4-フェニル-1H-イミダゾール-2-イル)メタンアミン一塩酸塩 (300 mg)を氷冷下加え、10分間同温で撹拌した。更に2-(クロロメチル)オキシラン (230 μL)のDMF (5 mL)溶液を加え、室温で18時間、40℃で5時間撹拌した。その後、不溶物をろ過で除き、トルエンで洗浄し、ろ液を減圧下濃縮した。残渣をシリカゲルCC (MeOH-クロロホルム)で精製して、2-フェニル-6,7,8,9-テトラヒドロ-5H-イミダゾ[1,2-a][1,4]ジアゼピン-6-オール (85.3 mg)を得た。
NaOH (1.0 g)、水 (3 mL)、TBAB (17 mg)とDMF (10 mL)の混合物に1-[4-(4-フルオロフェニル)-1H-イミダゾール-2-イル)メタンアミン二塩酸塩 (446 mg)を氷冷下加え、同温で10分撹拌し、更に1-ブロモ-3-クロロプロパン (250 μL)のDMF (10 mL)溶液を加え、室温で1時間、50℃で終夜撹拌した。その後不溶物をろ過で除き、トルエンで洗浄し、ろ液を減圧下濃縮した。残渣をシリカゲルCC (MeOH-クロロホルム)で精製して、2-(4-フルオロフェニル)-6,7,8,9-テトラヒドロ-5H-イミダゾ [1,2-a][1,4]ジアゼピン (20.8 mg)を得た。
({4-[4-(トリフルオロメチル)フェニル]-1H-イミダゾール-2-イル}メチル)カルバミン酸tert-ブチルエステル (3 g)のDMF (45 mL)溶液に、1-ブロモ-3-クロロプロパン (0.95 mL)とK2CO3 (3.64 g)を加え、60℃で終夜撹拌した。その後、反応液をセライトろ過して、固体をDMF (15 mL)で洗浄した。ろ液に氷冷下KOtBu (1.97 g)を加え、アルゴン雰囲気下、同温で2時間撹拌した。その後反応液に水を加え、EtOAcで抽出し、有機層を水、brineで順次洗浄した後に、MgSO4で乾燥し、減圧下濃縮した。残渣をシリカゲルCC (EtOAc-ヘキサン)で洗浄して2-[4-(トリフルオロメチル)フェニル]-6,7-ジヒドロ-5H-イミダゾ[1,2-a][1,4]ジアゼピン-8(9H)-カルボン酸tert-ブチルエステル (1.26 g)を得た。
4-ベンジル-1-(5-フルオロ-2-ニトロフェニル)-1,4-ジアゼパン-2-オン (440 mg)の酢酸 (9 mL)溶液に鉄 (286 mg)を加えて80℃で3時間撹拌した。その後反応液をセライトろ過してろ液をEtOAcで希釈し、有機層を飽和重曹水、brineで順次洗浄して、MgSO4で乾燥し、減圧下濃縮した。残渣をシリカゲルCC (EtOAc-ヘキサン)で精製し、2-ベンジル-8-フルオロ-2,3,4,5-テトラヒドロ-1H-[1,4]ジアゼピノ[1,2-a]ベンゾイミダゾール (150 mg)を得た。
4-ベンジル-1,4-ジアゼパン-2-オン (500 mg)のジオキサン (15 mL)溶液に2-ブロモ-4-フルオロ-1-ニトロベンゼン (538 mg)、Pd2dba3 (67 mg)、キサントホス (127 mg)とCs2CO3 (1 g)を加え、アルゴン雰囲気下100℃で終夜撹拌した。その後EtOAcで希釈して、有機層を水、brineで順次洗浄し、Na2SO4で乾燥し、減圧下濃縮した。残渣をシリカゲルCC (EtOAc-ヘキサン)で精製し、4-ベンジル-1-(5-フルオロ-2-ニトロフェニル)-1,4-ジアゼパン-2-オン (440 mg)を得た。
[(tert-ブトキシカルボニル)アミノ]酢酸2-オキソ-2-[4-(トリフルオロメチル)フェニル]エチルエステル (3 g)のトルエン (30 mL)溶液に酢酸アンモニウム (3.2 g)を加え、終夜還流した。その後、反応液に水を加え、EtOAcで抽出した。有機層をbrineで洗浄し、Na2SO4で乾燥し、減圧下濃縮した。残渣をシリカゲルCC (EtOAc-ヘキサン)で精製し、 ({4-[4-(トリフルオロメチル)フェニル]-1H-イミダゾール-2-イル}メチル)カルバミン酸tert-ブチルエステル (2.6 g)を得た。
N-(tert-ブトキシカルボニル)グリシン (3 g)のMeCN (60 mL)溶液に2-ブロモ-1-[4-(トリフルオロメチル)フェニル]エタノン (4.57 g)とK2CO3 (2.4 g)を加え、室温で1時間撹拌した。その後、反応液をセライトろ過して、ろ液を減圧下濃縮し、[(tert-ブトキシカルボニル)アミノ]酢酸2-オキソ-2-[4-(トリフルオロメチル)フェニル]エチルエステル (6 g)を得た。
2-[4-(トリフルオロメチル)フェニル]-6,7-ジヒドロ-5H-イミダゾ[1,2-a][1,4]ジアゼピン-8(9H)-カルボン酸tert-ブチルエステル (1.25 g)のジクロロメタン (12.5 mL)溶液に、4M塩化水素/ジオキサン溶液 (4.1 mL)を加え、室温で終夜撹拌した。その後減圧下濃縮し、2-[4-(トリフルオロメチル)フェニル]-6,7,8,9-テトラヒドロ-5H-イミダゾ[1,2-a][1,4]ジアゼピン塩酸塩 (1.1 g)を得た。
2-ベンジル-8-フルオロ-2,3,4,5-テトラヒドロ-1H-[1,4]ジアゼピノ[1,2-a]ベンゾイミダゾール (140 mg)のEtOH (5.5 mL)溶液に10% Pd/C (50 mg)を加え、水素雰囲気下、終夜撹拌した。その後反応液をセライトろ過して、ろ液を減圧下濃縮し、8-フルオロ-2,3,4,5-テトラヒドロ-1H-[1,4]ジアゼピノ[1,2-a]ベンゾイミダゾール (97 mg)を得た。
2-アミノ-3,4-ジフルオロフェノール (6.9 g)とジオキサン (70 mL)との混合物に室温で、CDI (10 g)を加え、60℃で2時間撹拌した。反応混合物に1M 塩酸水溶液を加え、EtOAcで希釈後、有機層をbrineで洗浄した後、Na2SO4で乾燥し、減圧下濃縮した。残渣をシリカゲルCC (EtOAc-ヘキサン)で精製し、4,5-ジフルオロ-1,3-ベンゾオキサゾール-2(3H)-オン (3.65 g)を得た。
4,5-ジフルオロ-1,3-ベンゾオキサゾール-2(3H)-オン (100 mg)とMeCN (2 mL)との混合物に室温で、K2CO3 (105 mg)及びブロモ酢酸tert-ブチルエステル (100 μL)を加え、50℃で2.5時間撹拌した。反応混合物にEtOAcを加え、固体をろ取し、EtOAcで洗浄後、減圧下乾燥した。残渣をシリカゲルCC (EtOAc-ヘキサン)で精製し、(4,5-ジフルオロ-2-オキソ-1,3-ベンゾオキサゾール-3(2H)-イル)酢酸tert-ブチルエステル (180 mg)を得た。
4-フルオロ-1,3-ベンゾオキサゾール-2(3H)-オン (27.2 g)とMeCN (500 mL)との混合物に室温で、K2CO3 (35.2 g)及びブロモ酢酸tert-ブチルエステル (27.6 mL)を加え、60℃で終夜撹拌した。反応混合物を室温まで放冷し、不溶物をろ別してろ液を減圧下濃縮した。得られた残渣にヘキサン-EtOAc (9:1)混合溶媒を加えて生じた固体をろ取し、ヘキサン-EtOAc (9:1)混合溶媒で洗浄し、(4-フルオロ-2-オキソ-1,3-ベンゾオキサゾール-3(2H)-イル)酢酸tert-ブチルエステル (42.6 g)を得た。
(4,5-ジフルオロ-2-オキソ-1,3-ベンゾオキサゾール-3(2H)-イル)酢酸tert-ブチルエステル (166 mg)のジクロロメタン (2 mL)溶液に室温でTFA (400 μL)を加え、同温で3時間撹拌した。反応混合物を減圧下濃縮し、ヘキサンを加えた。固体をろ取し、ヘキサンで洗浄後、減圧下乾燥し、(4,5-ジフルオロ-2-オキソ-1,3-ベンゾオキサゾール-3(2H)-イル)酢酸 (128 mg)を得た。
(4-フルオロ-2-オキソ-1,3-ベンゾオキサゾール-3(2H)-イル)酢酸tert-ブチルエステル (5 g)とジクロロメタン (14 mL)の混合物に室温でTFA (7 mL)を加え、同温で終夜撹拌した。反応混合物を減圧下濃縮し、得られた固体をろ取し、ジエチルエーテルで洗浄後、減圧下乾燥し、(4-フルオロ-2-オキソ-1,3-ベンゾオキサゾール-3(2H)-イル)酢酸 (3.65 g)を得た。
(4,5-ジフルオロ-2-オキソ-1,3-ベンゾオキサゾール-3(2H)-イル)酢酸 (80 mg)のジクロロメタン (1 mL)混合物に、HATU (140 mg)とDIPEA(120 μL)を加え、2-フェニル-6,7,8,9-テトラヒドロ-5H-イミダゾ[1,2-a][1,4]ジアゼピン-6-オール(80 mg)のジクロロメタン (2 mL)混合物を加え、室温で2時間撹拌した。反応液をクロロホルムで希釈後、飽和重曹水を加えた。得られた固体をろ取し、水とクロロホルムで洗浄し、4,5-ジフルオロ-3-[2-(6-ヒドロキシ-2-フェニル-6,7-ジヒドロ-5H-イミダゾ[1,2-a][1,4]ジアゼピン-8(9H)-イル)-2-オキソエチル]-1,3-ベンゾオキサゾール-2(3H)-オン (64 mg)を得た。ろ液を水、brineで順次洗浄した後に、Na2SO4で乾燥し、減圧下濃縮した。残渣をシリカゲルCC (MeOH-クロロホルム)で精製し、4,5-ジフルオロ-3-[2-(6-ヒドロキシ-2-フェニル-6,7-ジヒドロ-5H-イミダゾ[1,2-a][1,4]ジアゼピン-8(9H)-イル)-2-オキソエチル]-1,3-ベンゾオキサゾール-2(3H)-オン (9.7 mg)を得た。
(4-フルオロ-2-オキソ-1,3-ベンゾオキサゾール-3(2H)-イル)酢酸 (656 mg)のジクロロメタン (20 mL)混合物に、HATU (1.29 g)とDIPEA (1.9 mL)を加え、室温で3分撹拌した。次に2-[4-(トリフルオロメチル)フェニル]-6,7,8,9-テトラヒドロ-5H-イミダゾ[1,2-a][1,4]ジアゼピン塩酸塩 (1 g)を加え、室温で終夜撹拌した。反応液をEtOAcで希釈後、有機層を水、飽和重曹水、brineで順次洗浄した後に、MgSO4で乾燥し、減圧下濃縮した。残渣をシリカゲルCC (MeOH-クロロホルム)で精製した後に、更にシリカゲルCC (EtOAc-ヘキサン)で精製して4-フルオロ-3-(2-オキソ-2-{2-[4-(トリフルオロメチル)フェニル]-6,7-ジヒドロ-5H-イミダゾ[1,2-a][1,4]ジアゼピン-8(9H)-イル}エチル)-1,3-ベンゾオキサゾール-2(3H)-オン (1.1 g)を得た。
上記の方法で製造した4-フルオロ-3-(2-オキソ-2-{2-[4-(トリフルオロメチル)フェニル]-6,7-ジヒドロ-5H-イミダゾ[1,2-a][1,4]ジアゼピン-8(9H)-イル}エチル)-1,3-ベンゾオキサゾール-2(3H)-オンについて(1)又は(2)の結晶化を行った。
(1)4-フルオロ-3-(2-オキソ-2-{2-[4-(トリフルオロメチル)フェニル]-6,7-ジヒドロ-5H-イミダゾ[1,2-a][1,4]ジアゼピン-8(9H)-イル}エチル)-1,3-ベンゾオキサゾール-2(3H)-オン (230 mg)のEtOH (1.6 mL)溶液を室温で3時間撹拌した。析出した沈澱をろ取して減圧下乾燥し、4-フルオロ-3-(2-オキソ-2-{2-[4-(トリフルオロメチル)フェニル]-6,7-ジヒドロ-5H-イミダゾ[1,2-a][1,4]ジアゼピン-8(9H)-イル}エチル)-1,3-ベンゾオキサゾール-2(3H)-オンのA01結晶(A01 form) (51 mg)を得た。
(2) 4-フルオロ-3-(2-オキソ-2-{2-[4-(トリフルオロメチル)フェニル]-6,7-ジヒドロ-5H-イミダゾ[1,2-a][1,4]ジアゼピン-8(9H)-イル}エチル)-1,3-ベンゾオキサゾール-2(3H)-オン (39.5 g)のEtOH (80 mL)溶液を室温で30分撹拌した。その後、徐々に減圧下濃縮して得られた固体を乾燥し、4-フルオロ-3-(2-オキソ-2-{2-[4-(トリフルオロメチル)フェニル]-6,7-ジヒドロ-5H-イミダゾ[1,2-a][1,4]ジアゼピン-8(9H)-イル}エチル)-1,3-ベンゾオキサゾール-2(3H)-オンのA02結晶(A02 form) (36 g)を得た。
(5-クロロ-2-オキソ-1,3-ベンゾオキサゾール-3(2H)-イル)酢酸(230 mg)のジクロロメタン(10 mL)溶液に室温で、2,3,4,5-テトラヒドロ-1H-[1,4]ジアゼピノ[1,2-a]ベンゾイミダゾール(160 mg)、WSC・HCl(220 mg)およびHOBt(105 mg)を加え、室温で12時間撹拌した。反応液に水を加え、クロロホルムで抽出した。有機層をbrineで洗浄した後、Na2SO4で乾燥し、減圧下濃縮した。残渣をシリカゲルCC(MeOH-クロロホルム)で精製した後、得られた粗生成物をEtOAc-ヘキサンで固体化して5-クロロ-3-[2-(4,5-ジヒドロ-1H-[1,4]ジアゼピノ[1,2-a]ベンゾイミダゾール-2(3H)-イル)-2-オキソエチル]-1,3-ベンゾオキサゾール-2(3H)-オン(70 mg)を得た。
(5-クロロ-2-オキソ-1,3-ベンゾオキサゾール-3(2H)-イル)酢酸 (530 mg)のジクロロメタン (15 mL)溶液にオキサリルクロライド (1.18 g)と触媒量のDMFを氷冷下加え、室温で3時間撹拌した。反応液を減圧下濃縮して、残渣をジクロロメタン (15 mL)に溶かし、8,9-ジメチル-2,3,4,5-テトラヒドロ-1H-[1,4]ジアゼピノ[1,2-a]ベンゾイミダゾール (400 mg)とTEA (1.6 mL)を氷冷下加え、室温で12時間撹拌した。その後反応液を減圧下濃縮し、残渣をシリカゲルCC (MeOH-クロロホルム)で精製して、EtOAc-ヘキサンで固体化して、5-クロロ-3-[2-(8,9-ジメチル-4,5-ジヒドロ-1H-[1,4]ジアゼピノ[1,2-a]ベンゾイミダゾール-2(3H)-イル)-2-オキソエチル]-1,3-ベンゾオキサゾール-2(3H)-オン (453 mg)を得た。
4,5-ジフルオロ-3-[2-(6-ヒドロキシ-2-フェニル-6,7-ジヒドロ-5H-イミダゾ[1,2-a][1,4]ジアゼピン-8(9H)-イル)-2-オキソエチル]-1,3-ベンゾオキサゾール-2(3H)-オン (40 mg)のジクロロメタン(1 mL)の混合物に、氷冷下、ビス(2-メトキシエチル)アミノサルファートリフルオリド (80 μL)を加え、同温で2時間、室温で19時間撹拌した。反応液に水を加え、クロロホルムで抽出した。有機層をbrineで洗浄した後に、Na2SO4で乾燥し、減圧下濃縮した。残渣をシリカゲルCC (MeOH-クロロホルム)で精製して、4,5-ジフルオロ-3-[2-(6-フルオロ-2-フェニル-6,7-ジヒドロ-5H-イミダゾ[1,2-a][1,4]ジアゼピン-8(9H)-イル)-2-オキソエチル-1,3-ベンゾオキサゾール-2(3H)-オン (11.1 mg)と4,5-ジフルオロ-3- [2-オキソ-2-(2-フェニル-7H-イミダゾ[1,2-a][1,4]ジアゼピン-8(9H)-イル)エチル]-1,3-ベンゾオキサゾール-2(3H)-オン (17.3 mg)を得た。

- R5が、H、
R6が、置換されていてもよいフェニルである、請求項1に記載の化合物又はその塩。 - ----が、単結合である請求項2に記載の化合物又はその塩。
- R4が、Hである請求項3に記載の化合物又はその塩。
- R1が、H、
R2が、H又はハロゲン、
R3が、ハロゲンである請求項4に記載の化合物又はその塩。 - R6が、同一又は異なった1~4個のRA1で置換されていてもよいフェニル、
RA1が、-OH、-O-低級アルキル、-CN、ハロゲン、低級アルキル、ハロ低級アルキル、低級アルキレン-O-低級アルキル、-C(=O)-低級アルキル、及び-C(=O)-低級アルキレン-CNからなる群より選択される基である請求項5に記載の化合物又はその塩。 - R6が、1個のRA2で置換されていてもよいフェニルであり、
RA2が、ハロゲン、低級アルキル、及びハロ低級アルキルからなる群より選択される基である請求項6に記載の化合物又はその塩。 - R2が、Hである請求項7に記載の化合物又はその塩。
- R6が、その結合手からみてパラ位に1個のRA2が置換されているフェニルである請求項8に記載の化合物又はその塩。
- 下記の群から選択される、請求項1に記載の化合物又はその塩。
4-フルオロ-3-(2-オキソ-2-{2-[4-(トリフルオロメチル)フェニル]-6,7-ジヒドロ-5H-イミダゾ[1,2-a][1,4]ジアゼピン-8(9H)-イル}エチル)-1,3-ベンゾオキサゾール-2(3H)-オン、
4-フルオロ-3-{2-[2-(2-フルオロフェニル)-6,7-ジヒドロ-5H-イミダゾ[1,2-a][1,4]ジアゼピン-8(9H)-イル]-2-オキソエチル}-1,3-ベンゾオキサゾール-2(3H)-オン、
3-[2-(4,5-ジヒドロ-1H-[1,4]ジアゼピノ[1,2-a]ベンゾイミダゾール-2(3H)-イル)-2-オキソエチル]-4,5-ジフルオロ-1,3-ベンゾオキサゾール-2(3H)-オン、
4-フルオロ-3-[2-(8-フルオロ-4,5-ジヒドロ-1H-[1,4]ジアゼピノ[1,2-a]ベンゾイミダゾール-2(3H)-イル)-2-オキソエチル]-1,3-ベンゾオキサゾール-2(3H)-オン、

3-{2-[2-(4-クロロフェニル)-6,7-ジヒドロ-5H-イミダゾ[1,2-a][1,4]ジアゼピン-8(9H)-イル]-2-オキソエチル}-4-フルオロ-1,3-ベンゾオキサゾール-2(3H)-オン。 - 下記の群から選択される、請求項10に記載の化合物又はその塩。
4-フルオロ-3-(2-オキソ-2-{2-[4-(トリフルオロメチル)フェニル]-6,7-ジヒドロ-5H-イミダゾ[1,2-a][1,4]ジアゼピン-8(9H)-イル}エチル)-1,3-ベンゾオキサゾール-2(3H)-オン、
4-フルオロ-3-{2-[2-(2-フルオロフェニル)-6,7-ジヒドロ-5H-イミダゾ[1,2-a][1,4]ジアゼピン-8(9H)-イル]-2-オキソエチル}-1,3-ベンゾオキサゾール-2(3H)-オン、
4,5-ジフルオロ-3-[2-オキソ-2-(2-フェニル-6,7-ジヒドロ-5H-イミダゾ[1,2-a][1,4]ジアゼピン-8(9H)-イル)エチル]-1,3-ベンゾオキサゾール-2(3H)-オン、
4-フルオロ-3-{2-[2-(3-フルオロフェニル)-6,7-ジヒドロ-5H-イミダゾ[1,2-a][1,4]ジアゼピン-8(9H)-イル]-2-オキソエチル}-1,3-ベンゾオキサゾール-2(3H)-オン、及び
3-{2-[2-(4-クロロフェニル)-6,7-ジヒドロ-5H-イミダゾ[1,2-a][1,4]ジアゼピン-8(9H)-イル]-2-オキソエチル}-4-フルオロ-1,3-ベンゾオキサゾール-2(3H)-オン。 - 4-フルオロ-3-(2-オキソ-2-{2-[4-(トリフルオロメチル)フェニル]-6,7-ジヒドロ-5H-イミダゾ[1,2-a][1,4]ジアゼピン-8(9H)-イル}エチル)-1,3-ベンゾオキサゾール-2(3H)-オンである、請求項10に記載の化合物又はその塩。
- 3-{2-[2-(4-クロロフェニル)-6,7-ジヒドロ-5H-イミダゾ[1,2-a][1,4]ジアゼピン-8(9H)-イル]-2-オキソエチル}-4-フルオロ-1,3-ベンゾオキサゾール-2(3H)-オンである、請求項10に記載の化合物又はその塩。
- 請求項1に記載の化合物又はその塩、及び製薬学的に許容される賦形剤を含有する医薬組成物。
- ドパミンD1受容体ポジティブアロステリックモジュレーターである請求項14に記載の医薬組成物。
- 認知機能障害、統合失調症陰性症状、統合失調症認知機能障害(CIAS)、パーキンソン病、アルツハイマー病、ハンチントン病、うつ病、注意欠陥多動性障害(ADHD)、又は、薬物依存の予防及び/又は治療用医薬組成物である請求項14に記載の医薬組成物。
- 認知機能障害、統合失調症陰性症状、統合失調症認知機能障害(CIAS)、パーキンソン病、アルツハイマー病、ハンチントン病、うつ病、注意欠陥多動性障害(ADHD)、又は、薬物依存の予防及び/又は治療用医薬組成物の製造のための請求項1に記載の化合物又はその塩の使用。
- 認知機能障害、統合失調症陰性症状、統合失調症認知機能障害(CIAS)、パーキンソン病、アルツハイマー病、ハンチントン病、うつ病、注意欠陥多動性障害(ADHD)、又は、薬物依存の予防及び/又は治療のための請求項1に記載の化合物又はその塩の使用。
