Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung von substituierten Phenylalanin Derivaten gemäß Formel (Ia), insbesondere ein Verfahren zur Herstellung von 3-[5-(4-{[(2 S )-2-({[trans-4-(Aminomethyl)cyclohexyl]carbonyl}amino)-3-{4'-[(trans-4-hydroxycyclohexyl)carbamoyl]-2'-methylbipheny1-4-yl}propanoyl]amino}phenyl)-4/H-1,2,4-triazol-3-yl]-2,2,3,3 tetrafluorpropansäure gemäß Formel (Ib).
Description
HERSTELLVERFAHREN FÜR PHENYLALANIN-DERIVATE
Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung von substituierten Phenylalanin- Derivaten gemäß Formel (Ia), insbesondere ein Verfahren zur Herstellung von 3-[5-(4- {[(25)-2- ({[ira«s-4-(Aminomethyl)cyclohexyl]carbonyl}amino)-3-{4'-[(/ra«Ä-4-hydroxycyclohexyl)- carbamoyl]-2'-methylbiphenyl-4-yl}propanoyl]amino}phenyl)-4//-l,2,4-triazol-3-yl]-2,2,3,3- tetrafluo rop ansäure gemäß Formel (Ib).
Verbindungen der Formel (Ia)
in welcher R1 für Wasserstoff, Fluor, Chlor, Methyl, Methoxy oder Trifluormethyl steht, R ' für Fluor oder Methyl steht,
R3 für Cyclohexyl, über ein Kohlens toffatom gebundenes Pyrrolidinyl oder über ein Kohlenstoffatom gebundenes Piperidinyl steht, wobei Cyclohexyl substituiert ist mit einem Substituenten ausgewählt aus der Gruppe bestehend aus Hydroxy, Amino und Ci-C3-Alkylamino, und wobei Pyrrolidinyl und Piperidinyl substituiert sein können mit 1 bis 2 Substituenten unabhängig voneinander ausgewählt aus der Gruppe bestehend aus Oxo und Methyl.
Bei den Verbindungen der Formel (Ia) und (Ib) handelt es sich um Inhibitoren des Blutgerinnungsfaktors Xla. die sich insbesondere zur Behandlung und/oder Prophylaxe von thrombotischen beziehungsweise thromboembolischen Erkrankungen eignen.
Ein allgemeines Syntheseverfahren zur Herstellung von substituierten Phenylalanmderivaten wird in JP 2014227401 beschrieben. Die Verbindungen der vorliegenden Erfindung unterscheiden sich von den Verbindungen aus JP 2014227401 in den Resten R6 und R8 der Verbindungen der Formel (Ic). Der Substituent R6 ist in JP 2014227401 als kleiner Substituent, wie Halogen, Ci-C6-Alkyl und Ci-Ce-Alkoxy beschrieben. Der Substituent R6 ist in der vorliegenden Erfindung ein substituiertes Phenyl. Der Substituent R8 ist in JP 2014227401 ein bizyklischer Heterozyklus. In der vorliegenden Erfindung ist R8 ein ΛΗ- 1 ,2,4-Triazol-3 -yl-2,2,3 ,3 -tetrafluorpropansäure Rest.
Das folgende Syntheseschema 1 veranschaulicht die Synthese von Verbindungen der Formel (Ic) gemäß dem in JP 2014227401 beschriebenen Verfahren:

Gemäß dem in Schema 1 beschriebenen Synthe s everfahren können damit Verbindungen der Formel (Ic) ausgehend von entsprechend substituierten Phenylalaninderivaten der Formel (II) hergestellt werden, und zwar je nach Verfügbarkeit der entsprechenden Ausgangsverbindungen gemäß der Syntheseschritte a) - d) oder e) - h). Finale Schutzgruppenabspaltung im Schritt i) liefert dann die Endverbindungen der Formel (Ic).
Diese Syntheseverfahren weisen jedoch allgemein bei der Herstellung von Verbindungen der Formel (Ia) und speziell bei der Herstellung von 3 - [ 5 - (4- { [ (25) -2 - ( { [ ira«s-4- ( Aminomethyl) - cyclohexyl]carbonyl}amino)-3-{4'-[(irawÄ-4-hydroxycyclohexyl)carbamoyl]-2'-methylbiphenyl-4- yl}propanoyl]amino}phenyl)-4i - 1 ,2,4-triazol-3 -yl] -2,2,3 ,3 -tetrafluorpropansäure gemäß Formel (Ib) verschiedene Nachteile auf. Als besonders nachteilig erweist sich die Einführung des 3-[5-(4- Aminophenyl)-4_ί/-l,2,4-triazol-3-yl]-2,2,3,3-tetrafluo ropansäureesters unter Verwendung von Syntheseschritt d).
So wird bei der Herstellung von Verbindungen der Formel (Ia) bzw. (Ib), ausgehend von enantiomererrreinen Ausgangsmaterialen der Formel (V), nach Amidkupplung im Syntheseschritt d) stets eine partielle bis vollständige Epimerisierung bei den Syntheseprodukten (IX) beobachtet. Auch der Einsatz spezieller Amidkupplungsreagenzien, die bevorzugt für Amidkupplungen von leicht zu epimerisierenden Ausgangsverbindungen eingesetzt werden können, wie beispielsweise Ethylcyan(hydroxyimino)acetat (Oxyma) in Chem. Eur. J. 2009, Vol. 15, 9394 und 2,4,6- Tripropyl-l,3,5,2,4,6-trioxatriphosphinan-2,4,6-trioxid (T3P) in Org. Lett. 201 1, Vol. 13, 5048 beschrieben, liefert bei der Synthese von Verbindungen der Formel (Ia) und (Ib) keine ausreichende Stereokontrolle und/oder Ausbeute an gewünschter Verbindung.
In JP 2014227401 werden keine Aussagen zur Epimerisierung der Verbindungen gemacht. Weiterhin ist nicht erwähnt, ob einer der Synthesesequenzen a) - d) oder e) - h) Vorteile bietet.
Daraus ergibt sich die Aufgabe der vorliegenden Erfindung, ein verbessertes stereoselektives Verfahren zur Herstellung von Verbindungen der Formel (Ia), insbesondere von 3-[5-(4- {[(25)-2- ({[ira«Ä-4-(Aminomethyl)cyclohexyl]carbonyl}amino)-3-{4'-[(trans-4-hydroxycyclohexyl)- carbamoyl]-2'-methylbiphenyl-4-yl}propanoyl]amino}phenyl)-4/ -l,2,4-triazol-3-yl]-2,2,3,3- tetrafluorprop ans äure gemäß Formel (Ib) in technischem Maßstab bereit zu stellen.
Überraschenderweise wurde nun gefunden, dass sich durch die Wahl geeigneter Reaktionsbedingungen und den Einsatz spezieller Schutzgruppen während der Synthese die Verbindungen der Formel (Ia) bzw. (Ib) in guten Ausbeuten und hoher stereochemischer Reinheit herstellen lassen. Diese Aufgabe wird gemäß der vorliegenden Erfindung, wie in Syntheseschema 2 und 3 gezeigt, gelöst. Syntheseschema 2 veranschaulicht die einzelnen Schritte anhand der Verbindung der Formel (Ib) und Syntheseschema 3 veranschaulicht die einzelnen Schritte anhand der Verbindung der Formel (Ia):
Zur Herstellung von 3-[5-(4-{[(25)-2-({[ira«5-4-(Aminomethyl)cyclohexyl]carbonyl}amino)-3-{4'- [(iraKS-4-hydroxycyclohexyl)carbamoyl]-2'-methylbiphenyl-4-yl}propanoyl]amino}phenyl)-4 l,2,4-triazol-3-yl]-2,2,3,3-tetraf uo ropansäure (Ib) werden L-Brom-Phenylalaninderivate der
Formel (X) mit Boronsäurederivaten der Formel (XI) zu entsprechenden Produkten (XII) umgesetzt. Anschließend erfolgt die Einführung von 3-[5-(4-Aminophenyl)-4 -l,2,4-triazol-3-yl]- 2,2,3 ,3 -tetrafluorpropansäurederivaten (XIII) und man gelangt zu Amidverbindungen der Formel (ΧΓν). Abspaltung der Aminschutzgruppe (R9) und der Silyl-Gruppe (R11) (Syntheseschritt 1) in Syntheseschema 2) liefert Verbindungen der Formel (XV). Wenn die Amins chutzgrupp e (R9) tert- Butoxycarbonyl ist, erfolgt die Abspaltung der Aminschutzgruppe (R9) und der Silyl-Gruppe (R11) in einem Syntheseschritt mit einer Säure. Wenn die Aminschutzgruppe (R9) 9- Fluorenylmethoxycarbonyl oder B enzyloxy c arbonyl ist, erfolgt die Abspaltung der Aminschutzgruppe (R9) und der Silyl-Gruppe R11 in zwei Schritten. Die Silyl-Gruppe R11 wird unter dem Fachmann bekannten Bedingungen abgespalten, um zu den Verbindungen der Formel (XV) zu gelangen. Unter dem Einsatz entsprechender trans-Λ- (Aminomethyl)cyclohexancarbonsäurederivate (XVI) werden die Verbindungen der Formel (XV) zu Verbindungen der Formel (XVII) weiter umgesetzt. Abspaltung der Aminschutzgruppe liefert dann Verbindungen der Formel (XVIII). Bei der Umsetzung von Verbindungen der Formel (XVII), in denen R13 tert-Butoxycarbonyl ist, mit Trifluoressigsäure erhält man Ester der Formel (XIX) als besonders gut isolierbare Zwischenstufen (Syntheseschritt p) in Syntheseschema 2). Ausgehend von Verbindungen der Formel (XVIII) bzw. (XIX) erhält man nach Esterspaltung die Verbindung der Formel (Ib).

ln einem alternativen Verfahren wird zuerst der Ester in Verbindungen der Formel (XVII) gespalten und man erhält Verbindungen der Formel (XXXVI). Durch Abspaltung der Aminschutzgruppe in Verbindungen der Formel (XXXVI) erhält man die Verbindung der Formel (Ib). In Verbindungen der Formel (XXXVI) ist bevorzugt R13 tert-Butoxycarbonyl. R12 für Methyl, Ethyl oder iso-Propyl steht, und
R13 für 9-Fluorenylmethoxycarbonyl, tert-Butoxycarbonyl oder B enzyloxycarbonyl steht.
Wobei bevorzugt in den Verbindungen des Syntheseschema 2, des Syntheseschema 2.1 und des Syntheseschema 3
R9 für tert-Butoxycarbonyl steht,
R10 für -B(OH)2 steht,
R11 für tert-Butyl(dimethyl)silyl steht,
R12 für Methyl steht, und
R13 für tert-Butoxycarbonyl steht.
Wobei in den Verbindungen des Syntheseschema 3
R1 für Wasserstoff, Fluor, Chlor, Methyl, Methoxy oder Trifluormethyl steht, R- für Fluor oder Methyl steht,
R3 für Cyclohexyl, über ein Kohlenstoffatom gebundenes Pyrrolidinyl oder über ein Kohlenstoffatom gebundenes Piperidinyl steht, wobei Cyclohexyl substituiert ist mit einem Substituenten ausgewählt aus der Gruppe bestehend aus Hydroxy, Amino und C i -C3 - Alkylamino,
und wobei Pyrrolidinyl und Piperidinyl substituiert sein können mit 1 bis 2 Substituenten unabhängig voneinander ausgewählt aus der Gruppe bestehend aus Oxo und Methyl.
Der Substituent R; kann gegebenenfalls während der Synthese eine Schutzgruppe tragen, die zu gegebener Zeit während der Synthese unter dem Fachmann bekannten Bedingungen abgespalten wird. Diese Schutzgruppe ist beispielsweise tert-Butyl(dimethyl)silyl, Trimethylsilyl oder Triethylsilyl.
Die Herstellung der in Syntheseschema 2 beschriebenen Ausgangsverbindungen der Formel (XI) und (XIII) kann entsprechend folgender Syntheseschemata 4 und 5, ausgehend von käuflichen bzw. dem Fachmann bekannten Ausgangsverbindungen, erfolgen:

Sofern die erfindungsgemäßen Verbindungen in tautomeren Formen vorkommen können, umfasst die vorliegende Erfindung sämtliche tautomere Formen.
Folgende zwei Darstellungsweisen (A) und (B) eines 1 ,4-disubstituierten Cyclohexylderivats sind äquivalent zueinander und gleichbedeutend und in beiden Fällen deskriptiv für ein trans-\ ,A- disubsituiertes Cyclohexylderivat.

Dies gilt insbesondere für das Strukturelement des Tranexamsäureamids, beispielsweise ~N-[(trans- 4- { [(ieri-Butoxycarbonyl)amino]methyl } cyclohexyl)carbonyl und trans-Λ- ( Aminomethyl) - cyclohexyl] carbonyl} . Weiterhin gilt dies auch für das Strukturelement des trans-A- Hydroxycyclohexylamins, beispielsweise in {trans-A -Hydroxy cyclohexyl) c arbamoyl . In der vorliegenden Erfindung wird für das Tranexamsäureamid die Darstellung (A) bevorzugt verwendet.
Folgende drei Tautomer-Darstellungsweisen (C), (D) und (E) eines Triazolderivate s sind äquivalent zueinander und gleichbedeutend und in allen Fällen deskriptiv für ein 1 ,4- disubsituiertes Triazolderivat.

Dies gilt insbesondere für folgende Strukturelemente : li/-l,2,4-triazol-3-yl, li/-l,2,4-triazol-5-yl, AH- 1 ,2,4-triazol-3 -yl und 4H-l ,2,4-triazol-5-yl. Y1 und Y2 sind dabei unterschiedliche Substituenten.
Als Salze sind im Rahmen der vorliegenden Erfindung physiologisch unbedenkliche Salze der erfin- dungsgemäßen Verbindungen bevorzugt. Umfasst sind aber auch Salze, die für pharmazeutische Anwendungen selbst nicht geeignet sind aber beispielsweise für die Isolierung oder Reinigung der erfindungsgemäß en Verbindungen verwendet werden können.
Physiologisch unbedenkliche Salze der erfindungsgem äß en Verbindungen umfassen Säure - additionssalze von Mineralsäuren, Carbonsäuren und Sulfonsäuren, z.B. Salze der Chlor-
wasserstoffsäure, Bromwasserstoffsäure, Schwefelsäure, Phosphorsäure, Methansulfonsäure, Ethan- sulfonsäure, Toluolsulfonsäure, Benzolsulfonsäure, Naphthalindisulfonsäure, Essigsäure, Trifluor- essigsäure, Propionsäure, Milchsäure, Weinsäure, Äpfelsäure, Zitronensäure, Fumarsäure, Maleinsäure und Benzoesäure. Physiologisch unbedenkliche Salze der erfindungsgemäßen Verbindungen umfassen auch Salze üblicher Basen, wie beispielhaft und vorzugsweise Alkalimetallsalze (z.B. Natrium- und Kaliumsalze), Erdalkalisalze (z.B. Calcium- und Magnesiumsalze) und Ammoniumsalze, abgeleitet von Ammoniak oder organischen Aminen mit 1 bis 16 C -Atomen, wie beispielhaft und vorzugsweise Ethylamin, Diethylamin, Triethylamin, Ethyldiisopropylamin, Monoethanolamin, Diethanolamin, Triethanolamin, Dicyclohexylamin, Dimethylaminoethanol, Prokain, Dibenzylamin, N-Methyl- morpholin, Arginin, Lysin, Ethylendiamin, N-M ethylpiperidin und Cholin.
Als Solvate werden im Rahmen der Erfindung solche Formen der erfindungsgemäß en Verbindungen bezeichnet, welche in festem oder flüssigem Zustand durch Koordination mit Lösungsmittelmolekülen einen Komplex bilden. Hydrate sind eine spezielle Form der Solvate, bei denen die Koordination mit Wasser erfolgt.
Gegenstand der Erfindung ist ein Verfahren zur Herstellung der Verbindung (Ib), oder ihrer Salze, ihrer Solvate oder der Solvate ihrer Salze, dadurch gekennzeichnet, dass Verbindungen der Formel
R12 für Methyl, Ethyl oder iso-Propyl steht, mit einer Base zu der Verbindung der Formel
Bevorzugt sind Verbindungen der Formel (XVIII), in welcher R12 für Methyl steht.
Die Umsetzung erfolgt in inerten Lösungsmitteln in einem Temperaturbereich von Raumtemperatur bis 60°C bei Normaldruck.
Inerte Lösungsmittel sind Dichlormethan, T etrahydrofuran, 2-Methyl-tetrahydrofuran, Dioxan, Methanol, Ethanol oder Isopropanol oder Gemische der Lösungsmitteln, oder Gemische der Lösungsmittel mit Wasser, bevorzugt ist ein Gemisch aus T etrahydrofuran und Wasser.
Basen sind Natriumhydroxid, Lithiumhydroxid, Kaliumhydroxid, Cäsiumcarbonat, Natriumc arbonat, Kaliumc arbonat, bevorzugt ist Natriumhydroxid oder Lithiumhydroxid.
Die Verbindungen der Formel (XVIII) können hergestellt werden, indem von Verbindungen der Formel
R12 für Methyl, Ethyl oder iso-Propyl steht, und
R13 für 9-Fluorenylmethoxycarbonyl, tert-Butoxycarbonyl oder Benzyloxycarbonyl steht, die Schutzgruppe R13 abgespalten wird.
Bevorzugt sind Verbindungen der Formel (XVII), in welcher R12 für Methyl steht und R13 für tert- Butoxycarbonyl steht.
Im Falle von 9 -Fluor enylmethoxyc arbonyl erfolgt die Umsetzung in inerten Lösungsmitteln in Gegenwart von Piperidin in einem Temperaturbereich von Raumtemperatur bis 60°C bei Normaldruck.
Inerte Lösungsmittel sind Tetrahydrofuran, Dimethylformamid, Dimethylacetamid oder 1 -Methyl - 2-pyrrolidon (NMP), bevorzugt ist Dimethylformamid.
Im Falle von tert-Butoxy c arbonyl erfolgt die Umsetzung in inerten Lösungsmitteln in Gegenwart einer Säure in einem Temperaturbereich von Raumtemperatur bis 60°C bei Normaldruck.
Inerte Lösungsmittel sind Dichlormethan, Tetrahydrofuran oder Dioxan, bevorzugt ist Dioxan.
Säure ist Chlorwasserstoff in Dioxan.
Im Falle von Benzyloxycarbonyl erfolgt die Umsetzung in inerten Lösungsmitteln mit Wasserstoff in Gegenwart von Palladium auf Kohle bei Raumtemperatur bis 40°C bei Normaldruck bis 3 bar.
Inerte Lösungsmittel sind Tetrahydrofuran, Dioxan, Dimethylformamid, Methanol, Ethanol oder Isopropanol, oder Gemische der Lösungsmittel, bevorzugt ist Ethanol oder Dimethylformamid.
Die Verbindungen der Formel (XVII) können hergestellt werden, indem Verbindungen der Formel
R12 für Methyl, Ethyl oder iso-Propyl steht, mit Verbindungen der Formel
in welcher R13 für 9-Fluorenylmethoxycarbonyl, tert-Butoxycarbonyl oder Benzyloxycarbonyl steht, in Gegenwart eines Amidkupplungsre agenze s umgesetzt werden.
Bevorzugt sind Verbindungen der Formel (XV), in welcher R für Methyl steht.
Bevorzugt sind Verbindungen der Formel (XVI), in welcher R13 für tert-Butoxycarbonyl steht.
Die Umsetzung erfolgt in inerten Lösungsmitteln, in Gegenwart einer Base, in einem Temperaturbereich von 10°C bis 50°C bei Normaldruck, bevorzugt ist Raumtemperatur.
Als Amidkupplungsreagenzien eignen sich hierbei ^-(S-Dimethylaminoisopropy^-A^'-ethylcarbo- diimid-Hydrochlorid (EDC), Carbonyldiimidazol, Ethyl- cy an(hydro y-imino) ac etat (Oxyma), (1- Cyano-2-ethoxy-2-oxoemylidenaminooxy)dimethylamino-mo holino-carbeniurn- hexafluorophosphat (COMU), 2,4,6-Tripropyl-l,3,5,2,4,6-trioxatriphosphinan-2,4,6-trioxid (T3P), oder Mischungen aus diesen, bevorzugt ist ( 1 -Cyano-2-ethoxy-2-oxoethylidenaminooxy) dimethylamino-niorpholino-carbenium-hexafluorophosphat (COMU).
Basen sind Natriumcarbonat, Kaliumcarbonat, Natriumhydrogencarbonat, Kaliumhydrogen- carbonat, Triethylamin, N-M e&ylmorpholin, N-Methylpiperidin, 2,2,6,6-Tetramethylpiperidin oder Diisopropylethylamin, bevorzugt ist 2 ,2 , 6 ,6 -T etramethylpiperidin oder Diisopropylethylamin.
Inerte Lösungsmittel sind Tetrahydrofuran, 2-Methyl-tetrahydrofuran, 1 -Methyl-2-pyrrolidon, Dioxan, Dimethylformamid oder Acetonitril, oder Gemische der Lösungsmittel, bevorzugt ist Tetrahydrofuran, Dimethylformamid oder 1 -Methyl-2-pyrrolidon.
Die Verbindungen der Formel (XVI) sind bekannt oder lassen sich nach bekannten Verfahren aus den entsprechenden Ausgangsverbindungen synthetisieren.
Die Verbindungen der Formel (XV) können hergestellt werden, indem von Verbindungen der Formel
in welcher für 9-Fluorenylmethoxycarbonyl, tert-Butoxycarbonyl oder B enzyloxyc arbonyl steht, für tert-Butyl(dimethyl)silyl, Trimethylsilyl oder Tri ethyl silyl steht,
und
R12 für Methyl, Ethyl oder iso-Propyl steht, die Schutzgruppen R9 und R11 abgespalten werden.
Bevorzugt sind Verbindungen der Formel (XIV), in welcher R9 für tert-Butoxycarbonyl steht, R11 für tert-Butyl(dimethyl)silyl steht und R12 für Methyl steht.
Im Falle von 9 -Fluor enylmethoxyc arbonyl erfolgt die Umsetzung in zwei Stufen:
Die Abspaltung der Silyl-Gruppe erfolgt in inerten Lösungsmitteln in Gegenwart von Tetrabutylammoniumfluorid in einem Temperaturbereich von Raumtemperatur bis 50°C bei Normaldruck.
Inerte Lösungsmittel sind Tetrahydrofuran, Dioxan und Dimethylformamid, oder Gemische der Lösungsmittel, bevorzugt ist Dimethylformamid.
Die Abspaltung der 9-Fluorenylmethoxycarbonyl-Gruppe erfolgt in inerten Lösungsmitteln in Gegenwart von Piperidin in einem Temperaturbereich von Raumtemperatur bis 60°C bei Normaldruck.
Inerte Lösungsmittel sind Tetrahydrofuran, Dimethylformamid, Dimethylacetamid oder 1 -Methyl - 2-pyrrolidon, bevorzugt ist Dimethylformamid.
Im Falle von tert-Butoxycarbonyl erfolgt die Umsetzung in einer Stufe in inerten Lösungsmitteln in Gegenwart einer Säure in einem Temperaturbereich von Raumtemperatur bis 60°C bei Normaldruck.
Inerte Lösungsmittel sind Dichlormethan, Tetrahydrofuran oder Dioxan, bevorzugt ist Dioxan. Säure ist Chlorwasserstoff in Dioxan.
Im Falle von Benzyloxycarbonyl erfolgt die Umsetzung in zwei Stufen:
Die Abspaltung der Silyl-Gruppe erfolgt in inerten Lösungsmitteln in Gegenwart von Tetrabutylammoniumfluorid in einem Temperaturbereich von Raumtemperatur bis 50°C bei Normaldruck.
Inerte Lösungsmittel sind Tetrahydrofuran, Dioxan und Dimethylformamid, oder Gemische der Lösungsmittel, bevorzugt ist Dimethylformamid.
Die Abspaltung der B enzyloxyc arbonyl -Gruppe erfolgt in inerten Lösungsmitteln mit Wasserstoff in Gegenwart von Palladium auf Kohle bei Raumtemperatur bis 40°C bei Normaldruck bis 3 bar.
Inerte Lösungsmittel sind Tetrahydrofuran, Dioxan, Dimethylformamid, Methanol, Ethanol oder Isopropanol, oder Gemische der Lösungsmittel, bevorzugt ist Ethanol oder Dimethylformamid. Die Verbindungen der Formel (XIV) können hergestellt werden, indem Verbindungen der Formel
R9 für 9-Fluorenylmethoxycarbonyl, tert-Butoxycarbonyl oder B enzyloxyc arbonyl steht, und R11 für tert-Butyl(dimethyl)silyl, Trimethylsilyl oder Triethylsilyl steht, mit Verbindungen der Formel
R 1 für Methyl, Ethyl oder iso-Propyl steht, in Gegenwart eines Amidkupplungsre agenze s umgesetzt werden.
Bevorzugt sind Verbindungen der Formel (XII), in welcher R9 für tert-Butoxy c arbonyl steht und R11 für tert-Butyl(dimethyl)silyl steht.
Bevorzugt sind Verbindungen der Formel (XIII), in welcher R12 für Methyl steht.
Die Umsetzung erfolgt in inerten Lösungsmitteln, in Gegenwart einer Base, in einem Temperaturbereich von 10°C bis 50°C bei Normaldruck, bevorzugt ist Raumtemperatur.
Als Amidkupplungsreagenzien eignen sich hierbei iV-(3-Dimethyiaminoisopropyl)-7V'-ethylcarbo- diimid-Hydrochlorid (EDC), Carbonyldiimidazol, Ethyl- cy an(hydroxy-imino) ac etat (Oxyma), (1- Cyano-2-ethoxy-2-oxoethylidenaminooxy)dimethylamino-morpholino-carbenium- hexafluorophosphat (COMU), 2,4,6-Tripropyl-l,3,5,2,4,6-trioxatriphosphinan-2,4,6-trioxid (T3P), oder Mischungen aus diesen, bevorzugt ist ( 1 -Cyano-2-ethoxy-2-oxoethylidenaminooxy) dimethylamino-morpholino-carbenium-hexafluorophosphat (COMU).
Basen sind Natriumcarbonat, Kaliumcarbonat, Natriumhydrogencarbonat, Kaliumhydrogen- carbonat, Triethylamin, N-M ethylmo holin, 7V-Methylpiperidin, 2,2,6,6-Tetramethylpiperidin oder Diisopropylethylamin, bevorzugt ist 2 ,2 , 6 ,6 -T etramethylpiperidin oder Diisopropylethylamin. Inerte Lösungsmittel sind Tetrahydrofuran, 2-Methyl-tetrahydrofuran, 1 -Methyl-2-pyrrolidon, Dioxan, Dimethylformamid oder Acetonitril, oder Gemische der Lösungsmittel, bevorzugt ist Tetrahydrofuran, Dimethylformamid oder 1 -Methyl-2-pyrrolidon.
Die Verbindungen der Formel (XIII) können analog dem im Beispielteil beschriebenen Verfahren aus den entsprechenden Ausgangsverbindungen hergestellt werden. Die Verbindungen der Formel (XII) können hergestellt werden, indem Verbindungen der Formel
R9 für 9-Fluorenylmethoxycarbonyl, tert-Butoxycarbonyl oder B enzyloxyc arbonyl steht, mit Verbindungen der Formel
in welcher
R10 -B(OH)2, einen Boronsäure -Ester, bevorzugt Boronsäurepinakolester, oder
steht, R11 für tert-Butyl(dimethyl)silyl, Trimethylsilyl oder Triethylsilyl, unter Suzuki-Kupplungsbedingungen umgesetzt werden.
Bevorzugt sind Verbindungen der Formel (X), in welcher R9 für tert-Butoxycarbonyl steht.
Bevorzugt sind Verbindungen der Formel (XI), in welcher R10 für -B(OH)2 steht und R1! für tert-
Butyl(dimethyl)silyl steht. Die Umsetzung erfolgt in inerten Lösungsmitteln, in Gegenwart eines Katalysators, in Gegenwart eines Zusatzreagenzes, bevorzugt in einem Temperaturbereich von 50°C bis 150°C bei Normaldruck bis 3 bar.
Katalysatoren sind Dichlorbis(triphenylphosphin)palladium, Tetrakistriphenylphosphin- palladium(O), Palladium(II)acetat/Triscyclohexylphosphin, Tris(dibenzylidenaceton)dipalladium, Bis-(diphenylphosphanferrocenyl)-palladium-(II)-chlorid, 1 ,3 -Bis(2,6-diisopropylphenyl)imidazol- 2-yliden( 1 ,4-napthtochinon)palladiumdimer, Allyl(chlor)-( 1 ,3 -dimesityl- 1 ,3 -dihydro-2H-imidazol- 2-yliden)palladium, Palladium(II)acetat/Dicyclohexyl-(2',4',6'-triisopropyl-biphenyl-2-yl)-phosphin oder [1,1 -Bis-(diphenylphosphino)-ferrocen] -palladium(II)chlorid-Monodichlormethan-addukt, bevorzugt sind Tetrakistriphenylphosphin-palladium(O) und [1,1 -Bis-(diphenylphosphino)- ferrocen] -palladium(II)chlorid.
Zusatzreagenzien sind Natriumacetat, Kaliumacetat, Cäsiumcarbonat, Kaliumc arbonat, Natrium- c arbonat oder Kaliumphosphat, bevorzugt ist Natriumcarbonat.
Inerte Lösungsmittel sind Dioxan, Tetrahydrofuran, 2-Methyl-tetrahydrofuran, 1,2- Dimethoxyethan, Dimethylformamid, Dimethylacetamid, 1 -Methyl-2-pyrrolidon oder Acetonitril, bevorzugt ist Dioxan oder Tetrahydrofuran.
Die Verbindungen der Formeln (X) und (XI) können analog dem im Beispielteil beschriebenen Verfahren aus den entsprechenden Ausgangsverbindungen hergestellt werden.
Gegenstand der Erfindung ist weiterhin ein Verfahren zur Herstellung der Verbindung (Ib), oder ihrer Salze, ihrer Solvate oder der Solvate ihrer Salze, dadurch gekennzeichnet, dass Verbindungen der Formel
R12 für Methyl, Ethyl oder iso-Propyl steht, mit einer Base zu der Verbindung der Formel
Bevorzugt sind Verbindungen der Formel (XIX), in welcher R12 für Methyl steht.
Die Umsetzung erfolgt in inerten Lösungsmitteln in einem Temperaturbereich von Raumtemperatur bis 60°C bei Normaldruck.
Inerte Lösungsmittel sind Dichlormethan, T etrahydrofuran, 2-Methyl-tetrahydrofuran, Dioxan, Methanol, Ethanol oder Isopropanol oder Gemische der Lösungsmitteln, oder Gemische der Lösungsmittel mit Wasser, bevorzugt ist ein Gemisch aus T etrahydrofuran und Wasser.
Basen sind Natriumhydroxid, Lithiumhydroxid, Kaliumhydroxid, Cäsiumcarbonat, Natriumcarbonat, Kaliumc arbonat, bevorzugt ist Natriumhydroxid oder Lithiumhydroxid.
Die Verbindungen der Formel (XIX) können hergestellt werden, indem Verbindungen der Formel
R12 für Methyl, Ethyl oder iso-Propyl steht, und R13 für tert-Butoxyc arbonyl steht, mit Trifluoressigsäure umgesetzt werden.
Bevorzugt sind Verbindungen der Formel (XVIIa), in welcher R12 für Methyl steht.
Die Umsetzung erfolgt in inerten Lösungsmitteln in einem Temperaturbereich von Raumtemperatur bis 60°C bei Normaldruck. Inerte Lösungsmittel sind Dichlormethan, Tetrahydrofuran oder Dioxan, bevorzugt ist Dioxan.
Die Verbindungen der Formel (XVIIa) sind eine Teilmenge der Verbindungen der Formel (XVII).
Gegenstand der Erfindung ist weiterhin ein Verfahren zur Herstellung der Verbindung (Ib), oder ihrer Salze, ihrer Solvate oder der Solvate ihrer Salze, dadurch gekennzeichnet, dass in Verbindungen der Formel
R13 für 9-Fluorenylmethoxycarbonyl, tert-Butoxycarbonyl oder Benzyloxycarbonyl steht, die Schutzgruppe R13 abgespalten wird. Bevorzugt sind Verbindungen der Formel (XXXVI), in welcher R13 für tert-Butoxycarbonyl steht.
Im Falle von tert-Butoxy c arbonyl erfolgt die Umsetzung in inerten Lösungsmitteln in Gegenwart einer Säure in einem Temperaturbereich von Raumtemperatur bis 60°C bei Normaldruck.
Inerte Lösungsmittel sind Dichlormethan, Tetrahydrofuran oder Dioxan, bevorzugt ist Dioxan.
Säure ist Chlorwasserstoff in Dioxan. Die Verbindungen der Formel (XXXVI) können hergestellt werden, indem Verbindungen der Formel
R12 für Methyl, Ethyl oder iso-Propyl steht, und
R13 für 9-Fluorenylmethoxycarbonyl, tert-Butoxycarbonyl oder Benzyloxycarbonyl steht, mit einer Base umgesetzt werden.
Bevorzugt sind Verbindungen der Formel (XVII), in welcher R12 für Methyl steht und R13 für tert- Butoxycarbonyl steht.
Die Umsetzung erfolgt in inerten Lösungsmitteln in einem Temperaturbereich von Raumtemperatur bis 60°C bei Normaldruck.
Inerte Lösungsmittel sind Dichlormethan, T etrahydrofuran, 2-Methyl-tetrahydrofuran, Dioxan, Methanol, Ethanol oder Isopropanol oder Gemische der Lösungsmitteln, oder Gemische der Lösungsmittel mit Wasser, bevorzugt ist ein Gemisch aus T etrahydrofuran und Wasser.
Basen sind Natriumhydroxid, Lithiumhydroxid, Kaliumhydroxid, Cäsiumcarbonat, Natriumcarbonat, Kaliumc arbonat, bevorzugt ist Natriumhydroxid oder Lithiumhydroxid.
Gegenstand der Erfindung ist weiterhin ein Verfahren zur Herstellung der Verbindung (Ia), oder ihrer Salze, ihrer Solvate oder der Solvate ihrer Salze, dadurch gekennzeichnet, dass Verbindungen der Formel

R3 für Cyclohexyl, über ein Kohlenstoffatom gebundenes Pyrrolidinyl oder über ein Kohlenstoffatom gebundenes Piperidinyl steht, wobei Cyclohexyl substituiert ist mit einem Substituenten ausgewählt aus der Gruppe bestehend aus Hydroxy, Amino und C i -C3- Alkylamino, und wobei Pyrrolidinyl und Piperidinyl substituiert sein können mit 1 bis 2 Substituenten unabhängig voneinander ausgewählt aus der Gruppe bestehend aus Oxo und Methyl,
R12 für Methyl, Ethyl oder iso-Propyl steht, und

Inerte Lösungsmittel sind Dichlormethan, Tetrahydrofuran oder Dioxan, bevorzugt ist Dioxan.
Säuren sind Trifluore s sigs äure oder Chlorwasserstoff in Dioxan, bevorzugt ist Trifluoressigsäure. Im Falle von Benzyloxycarbonyl erfolgt die Umsetzung in inerten Lösungsmitteln mit Wasserstoff in Gegenwart von Palladium auf Kohle bei Raumtemperatur bis 40°C bei Normaldruck bis 3 bar.
Inerte Lösungsmittel sind Tetrahydrofuran, Dioxan, Dimethylformamid, Methanol, Ethanol oder Isopropanol, oder Gemische der Lösungsmittel, bevorzugt ist Ethanol oder Dimethylformamid.
Die Verbindungen der Formel (XXXIII) können hergestellt werden, indem Verbindungen der Formel
(XXXII),
in welcher für Wasserstoff, Fluor, Chlor, Methyl, Methoxy oder Trifluormethyl steht, für Fluor oder Methyl steht,
R ' für Cyclohexyl, über ein Kohlenstoffatom gebundenes Pyrrolidinyl oder über ein Kohlenstoffatom gebundenes Piperidinyl steht, wobei Cyclohexyl substituiert ist mit einem Substituenten ausgewählt aus der Gruppe bestehend aus Hydroxy, Amino und C i -C3- Alkylamino, und wobei Pyrrolidinyl und Piperidinyl substituiert sein können mit 1 bis 2 Substituenten unabhängig voneinander ausgewählt aus der Gruppe bestehend aus Oxo und Methyl, und

Basen sind Natriumcarbonat, Kaliumcarbonat, Natriumhydrogencarbonat, Kaliumhydrogen- carbonat, Triethylamin, N-M emylmorpholin, iV-Methylpiperidin, 2,2,6,6-Tetramethylpiperidin oder Diisopropylethylamin, bevorzugt ist 2 ,2 , 6 ,6 -T etramethylpiperidin oder Diisopropylethylamin.
Inerte Lösungsmittel sind Tetrahydrofuran, 2-Methyl-tetrahydrofuran, 1 -Methyl-2-pyrrolidon, Dioxan, Dimethylformamid oder Acetonitril, oder Gemische der Lösungsmittel, bevorzugt ist Tetrahydrofuran, Dimethylformamid oder 1 -Methyl-2-pyrrolidon.
Die Verbindungen der Formel (XVI) sind bekannt oder lassen sich nach bekannten Verfahren aus den entsprechenden Ausgangsverbindungen synthetisieren.
Die Verbindungen der Formel (XXXII) können hergestellt werden, indem von Verbindungen der Formel
R1 für Wasserstoff, Fluor, Chlor, Methyl, Methoxy oder Trifluormethyl steht, R- für Fluor oder Methyl steht,
R3 für Cyclohexyl, über ein Kohlenstoffatom gebundenes Pyrrolidinyl oder über ein Kohlenstoffatom gebundenes Piperidinyl steht, wobei Cyclohexyl substituiert ist mit einem Substituenten ausgewählt aus der Gruppe bestehend aus Hydroxy, Amino und C i -C3- Alkylamino, und wobei Pyrrolidinyl und Piperidinyl substituiert sein können mit 1 bis 2 Substituenten unabhängig voneinander ausgewählt aus der Gruppe bestehend aus Oxo und Methyl,

Säuren sind Trifluoressigsäure oder Chlorwasserstoff in Dioxan, bevorzugt ist Chlorwasserstoff in Dioxan.
Im Falle von B enzyloxy c arbonyl erfolgt die Umsetzung in inerten Lösungsmitteln mit Wasserstoff in Gegenwart von Palladium auf Kohle bei Raumtemperatur bis 40°C bei Normaldruck bis 3 bar.
Inerte Lösungsmittel sind Tetrahydrofuran, Dioxan, Dimethylformamid, Methanol, Ethanol oder Isopropanol, oder Gemische der Lösungsmittel, bevorzugt ist Ethanol oder Dimethylformamid.
Die Verbindungen der Formel (XXXI) können hergestellt werden, indem Verbindungen der

Inerte Lösungsmittel sind Tetrahydrofuran, 2-Methyl-tetrahydrofuran, 1 -Methyl-2-pyrrolidon, Dioxan, Dimethylformamid oder Acetonitril, oder Gemische der Lösungsmittel, bevorzugt ist Tetrahydrofuran, Dimethylformamid oder 1 -Methyl-2-pyrrolidon.
Die Verbindungen der Formel (XIII) können analog dem im Beispielteil beschriebenen Verfahren aus den entsprechenden Ausgangsverbindungen hergestellt werden.
Die Verbindungen der Formel (XXX) können hergestellt werden, indem Verbindungen der Formel
R9 für 9-Fluorenylmethoxycarbonyl, tert-Butoxycarbonyl oder B enzyloxyc arbonyl steht, mit Verbindungen der Formel
R1 für Wasserstoff, Fluor, Chlor, Methyl, Methoxy oder Trifluormethyl steht, R- für Fluor oder Methyl steht,
R3 für Cyclohexyl, über ein Kohlenstoffatom gebundenes Pyrrolidinyl oder über ein Kohlenstoffatom gebundenes Piperidinyl steht, wobei Cyclohexyl substituiert ist mit einem Substituenten ausgewählt aus der Gruppe bestehend aus Hydroxy, Amino und Ci-C3-Alkylamino,
und wobei Pyrrolidinyl und Piperidinyl substituiert sein können mit 1 bis 2 Substituenten unabhängig voneinander ausgewählt aus der Gruppe bestehend aus Oxo und Methyl, und R10 für -B(OH)2, einen Boronsäure -Ester, bevorzugt Boronsäurepinakolester, oder -BF-, ' steht, unter Suzuki-Kupplungsbedingungen umgesetzt werden.
Die Umsetzung erfolgt in inerten Lösungsmitteln, in Gegenwart eines Katalysators, in Gegenwart eines Zusatzreagenzes, bevorzugt in einem Temperaturbereich von 50°C bis 150°C bei Normaldruck bis 3 bar.
Katalysatoren sind Dichlorbis(triphenylphosphin)palladium, Tetrakistriphenylphosphin- palladium(O), Palladium(II)acetat/Triscyclohexylphosphin, Tris(dibenzylidenaceton)dipalladium, Bis-(diphenylphosphanferrocenyl)-palladium-(II)-chlorid, 1 ,3 -Bis(2,6-diisopropylphenyl)imidazol- 2-yliden( 1 ,4-napthtochinon)palladiumdimer, Allyl(chlor)-( 1 ,3 -dimesityl- 1 ,3 -dihydro-2H-imidazol- 2-yliden)palladium, Palladium(II)acetat/'Dicyclohexyl-(2',4',6'-triisopropyl-biphenyl-2-yl)-phosphin oder [1,1 -Bis-(diphenylphosphino)-ferrocen] -palladium(II)chlorid-Monodichlormethan-addukt, bevorzugt sind Tetrakistriphenylphosphin-palladium(O) und [1,1 -Bis-(diphenylphosphino)- ferrocen]-palladium(II)chlorid.
Zusatzreagenzien sind Natriumacetat, Kaliumacetat, Cäsiumcarbonat, Kaliumc arbonat, Natrium- c arbonat oder Kaliumphosphat, bevorzugt ist Natriumacetat, Kaliumacetat oder Natriumkarbonat.
Inerte Lösungsmittel sind Dioxan, Tetrahydrofuran, 2-Methyl-tetrahydrofuran, 1,2- Dimethoxyethan, Dimethylformamid, Dimethylacetamid, 1 -Methyl-2-pyrrolidon oder Acetonitril, bevorzugt ist Dioxan oder Tetrahydrofuran.
Die Verbindungen der Formeln (X) und (XXIX) können analog dem im Beispielteil beschriebenen Verfahren aus den entsprechenden Ausgangsverbindungen hergestellt werden.
Gegenstand der Erfindung ist weiterhin ein Verfahren zur Herstellung der Verbindung (Ia), oder ihrer Salze, ihrer Solvate oder der Solvate ihrer Salze, dadurch gekennzeichnet, dass von Verbindungen der Formel
R s für Wasserstoff, Fluor, Chlor, Methyl, Methoxy oder Trifluormethyl steht, R2 für Fluor oder Methyl steht,
R3 für Cyclohexyl, über ein Kohlenstoffatom gebundenes Pyrrolidinyl oder über ein Kohlenstoffatom gebundenes Piperidinyl steht, wobei Cyclohexyl substituiert ist mit einem Substituenten ausgewählt aus der Gruppe bestehend aus Hydroxy, Amino und C i -C3- Alkylamino, und wobei Pyrrolidinyl und Piperidinyl substituiert sein können mit 1 bis 2 Substituenten unabhängig voneinander ausgewählt aus der Gruppe bestehend aus Oxo und Methyl, und
R13 für 9-Fluorenylmethoxycarbonyl, tert-Butoxycarbonyl oder Benzyloxycarbonyl steht, die Schutzgruppe R13 abgespalten wird.
Bevorzugt sind Verbindungen der Formel (XXXV), in welcher R13 für tert-Butoxycarbonyl steht.
Im Falle von tert-Butoxy c arbonyl erfolgt die Umsetzung in inerten Lösungsmitteln in Gegenwart einer Säure in einem Temperaturbereich von Raumtemperatur bis 60°C bei Normaldruck.
Inerte Lösungsmittel sind Dichlormethan, Tetrahydrofuran oder Dioxan, bevorzugt ist Dioxan.
Säure ist Chlorwasserstoff in Dioxan.
Die Verbindungen der Formel (XXXV ) können hergestellt werden, indem Verbindungen der
Formel

Atisführangsbeispiele Beispiel 1 (25)-2-[(teri-Butoxycarbonyl)amino]-3-{4'-[(ira«Ä-4-{[teri-butyl(dimethyl)silyl]oxy}cyclohexyl)- carbamoyl]-2'-methylbiphenyl-4-yl}propansäure
4 -Brom-A^ ter ί-butoxy carbonyl) -L -phenylal anin (90.0 g, 261.5 mmol) sowie AL(/ra«5-4-{[/er/- Butyl(dimethyl)silyl]oxy} cyclohexyl)-3-methyl-4-(4,4,5,5 etramethyl-l,3,2-dioxaborolan-2- yl)benzamid (130.0 g, 274.5 mmol) wurden in 1440 ml Dioxan vorgelegt und die Mischung durch Einleiten von Argon inertisiert. Anschließend wurden 392 ml wässrige Natriumcarbonatlösung (2M) zugefügt und die Mischung durch Einleiten von Argon inertisiert. Die Mischung wurde auf 85°C erwärmt, dann l, -Bis(diphenylphosphino)ferrocenpalladium(II)chlorid-Dichlormethan- komplex (4.27 g, 5.23 mmol) zugefügt und die Mischung bei 85°C für insgesamt 5 h nachgerührt. Die auf Raumtemperatur gebrachte Reaktionsmischung wurde auf ein Gemisch aus 2000 ml Essigsäureethylester und 9500 ml 10%iger wässriger Natriumchloridlösung gegossen und durch portionsweise Zugabe von Zitronensäure auf pH 3 gebracht. Die organische Phase wurde abgetrennt und die wässrige Phase mit 2000 ml Essigsäureethylester extrahiert. Die gesammelten organischen Phasen wurden über Natriumsulfat getrocknet, durch Kieselgel filtriert und im Vakuum eingeengt. Das so erhaltene Rohprodukt wurde anschließend mittels Kieselgelchromatographie getrennt (Dichlormethan/'Aceton-Gradient). Man erhielt insgesamt 90.0 g der Titelverbindung, die ohne weitere Reinigung umgesetzt wurden.

Alternatives Verfahren: Durchführung unter Stickstoff ! 92.861 g (196.1 mmol) ~N-(trans-4-{[tert- Butyl(dimethyl)silyl]oxy} cyclohexyl)-3-methyl-4-(4,4,5,5-tetramethyl-l,3,2-dioxaborolan-2-yl)- benzamid und 75 g (217.89 mmol) 4-Brom-/V-(ier/-butoxycarbonyl)-L-phenylalanin wurden in 1200 ml Dioxan vorgelegt und bei Raumtemperatur wurden 327 ml einer 2M wässrigen Natriumcarbonat-Lösung zugegeben. Anschließend wurde auf 85°C erhitzt und dann 1.779 g (2.179 mmol) 1 , Γ -Bis(diphenylphosphino)ferrocenpalladium(II)chlorid-Dichlormethankomplex zugegeben. Es wurde über Nacht bei 85°C gerührt. Nach Abkühlen auf Raumtemperatur wurden 1000 ml Methyl-tert.-butylether zugegeben. Durch langsames Zutropfen einer 30%igen wässrigen Zitronensäure -Lösung wurde auf pH 3.29 eingestellt. Anschließend wurde 10 min. nachgerührt und dann die Phasen getrennt, dann mit 75 g Aktivkohle versetzt und weitere 10 min. nachgerührt. Die Aktivkohle wurde abfiltriert (über Kieselgur) und mit 150 ml Methyl-tert.-butylether nachgewaschen. Die organische Phase wurde zur Trockene eingeengt. Man erhielt 124.52 g (93.55% d.Th) der Titelverbindung. Das Rohprodukt wurde direkt in die Folgestufe eingesetzt.

Methyl-3 -[5-(4- { [(25)-2-[(/er/-butoxycarbonyl)amino] -3 -{4'-[(trans-4- { [/ert-butyl(dimethyl)- silyl]oxy} cyclohexyl)carbamoyl]-2'-methylbiphen^
triazol-3 -yl] -2,2,3 ,3 -tetrafluorpropanoat
(25)-2-[(ier^Butoxycarbonyl)amino]-3-{4'-[(irawÄ-4-{[ieri-butyl(dimethyl)silyl]oxy}cyclohexyl)- carbamoyl]-2'-methylbiphenyl-4-yl}propansäure (97%ig, 111.3 g, 176.8 mmol), (l-Cyano-2- ethoxy-2-oxoethylidenaminooxy)dimethylamm^
(79.5 g, 185.6 mmol) und 2,2,6, 6-Tetramethylpiperidin (26.2 g, 185.6 mmol) wurden in 1900 ml 1- Methyl-2-pyrrolidon vorgelegt und die Mischung durch Einleiten von Argon inertisiert. Anschließend wurde Methyl-3 -[5-(4-aminophenyl)- 1 //- 1 ,2,4-triazol-3 -yl] -2,2,3 ,3 -tetrafluorpropanoat (45.0 g, 141.4 mmol) zugefügt und die Re aktionsmis chung über Nacht bei Raumtemperatur nachgerührt. Danach wurde die Mischung auf 1650 ml Wasser gegeben und mit Ammoniumchlorid auf H 5.0 eingestellt. Der entstandene Feststoff wurde abfiltriert, in Essigsäureethylester aufgenommen und mit wässriger Natriumchloridlösung mehrfach gewaschen. Die organische Phase wurde getrocknet, im Vakuum eingeengt und mittels Kieselgelchromatographie gereinigt (Dichlormethan/Acton 10: 1).

Methyl-3 -[5-(4- { [(2S)-2-[(ieri-butoxycarbonyl)amino] -3 -{4'-[(tran$-4- { [ieri-butyl(dimethyl)- silyl]oxy} cyclohexyl)carbamoyl]-2'-methylbiphenyl-4-yl}propanoyl]amino}phenyl)-4jf -l,2,4- triazol-3 -yl] -2,2,3 ,3 -tetrafluorpropanoat (172.0 g, 188.8 mmol) wurden in 2000 ml Dioxan gelöst, anschließend mit 800 ml Chlorwasserstoff in Dioxan (4M) versetzt und die Mischung für 4.5 h bei Raumtemperatur nachgerührt. Der entstandene Feststoff wurde abfiltriert und der Filterrückstand zweimal mit je 500 ml Diethylether gewaschen. Der noch feuchte Rückstand wurde im Hochvakuum getrocknet. Man erhielt 141.9 g (93% d. Th.) der Titelverbindung.

amino } phenyl) -AH- 1 ,2,4-triazol-3 -yl] -2,2,3 ,3 -tetrafluo ropanoat
/ra«s-4-{[(feri-Butoxycarbonyl)amino]methyl}cyclohexancarbonsäure (2.41 g, 9.36 mmol), (1- Cyano-2-ethoxy-2-oxoethylidenaminooxy)dimethylamino-morpholino-carbenium-hexafluoro- phosphat (4.21 g, 9.83 mmol) und 2,2,6,6-Tetramethylpiperidin (5.29 g, 37.44 mmol) wurden in 79 ml 1 -Methyl-2-pyrrolidon vorgelegt und die Mischung durch Einleiten von Argon inertisiert. Anschließend wurde Methyl-3-[5-(4-{[(25)-2-amino-3- {4'-[(ira«s-4-hydroxy-cyclohexyi)- carbamoyl]-2'-methylbiphenyl-4-yl}propanoyl]amino}phenyl)-4j /-l,2,4-triazol-3-yl]-2,2,3,3- tetrafluo ropanoat-Hydrochlorid (94%ig, 7.70 g, 9.36 mmol) zugefügt und die Reaktionsmischung 2 h bei Raumtemperatur nachgerührt. Anschließend wurden erneut 0.1 eq (1- Cyano-2-ethoxy-2-oxoethylidenaminooxy)dimethylamino-mo holino-carbenium-hexafluoro- phosphat und 0.1 eq ira«s-4-{[(ieri-Butoxycarbonyl)amino]methyl}cyclohexancarbonsäure zugefügt und die Re aktionsmis chung über Nacht bei Raumtemperatur nachgerührt. Danach wurde mit Essigsäureethylester verdünnt und die organische Phase je einmal mit 10%iger wässriger Zitronensäure und Wasser gewaschen. Die organische Phase wurde eingeengt, der Rückstand mit Diethylether verrührt, abfiltriert und im Hochvakuum getrocknet. Man erhielt 8.30 g (86% d. Th.) der Titelverbindung.

Methyl-3 -[5-(4- { [(2S)-2- { [(trans-4- { [ (tert-butoxy c arbonyl) amino] methyl } cyclohexyl)carbonyl] - amino}-3-{4'-[(iraKS-4-hydroxycyclohexyl)carbamoyl]-2'-methylbiphenyl-4-yl}propanoyl]amino}- phenyl)-4_i - 1 ,2,4-triazol-3 -yl] -2,2,3 ,3 -tetrafluo ropanoat (3.00 g, 2.86 mmol) wurde portionsweise in 30.0 ml Trifluore s sigs äure gelöst und die Lösung über Nacht bei Raumtemp eratur nachgerührt. Anschließend wurde die Mischung am Vakuum eingeengt und der Rückstand mit 50.0 ml Diethylether im Ultraschallbad verrührt. Der entstandene Niederschlag wurde abfiltriert, dreimal mit je 25 ml Diethylether gewaschen und im Hochvakuum getrocknet. Man erhielt 2.67 g (88% d. Th.) der Titelverbindung.

Methyl-3-[5-(4-{[(25)-2-({[ira«Ä-4-(aminomethyl)cyclohexyl]carbonyl} amino)-3-(2'-methyl-4'- {[irflK,s-4-(2,2,2-trifluoracetoxy)cyclohexyl]carbamoyl}biphenyl-4-yl)propanoyl]amino}phenyl)- 4i7-l,2,4-triazol-3-yl]-2,2,3,3-tetrafluorpropanoat-Trifluoracetat (80.0 g, 76.5 mmol) wurde in 2000 ml Tetrahydrofuran gelöst. Anschließend wurde eine Lösung von Lithiumhydroxid (18.3 g, 764.9 mmol) in 250 ml Wasser zugegeben und die Mischung 5 min nachgerührt. Anschließend wurde die Mischung mit Salzsäure (4M) auf pH 3.5 gebracht, wobei die Temperatur durch Kühlung bei 15-20°C gehalten wurde. Anschließend wurde das T etrahydrofuran am Vakuum entfernt, der Rückstand mit 2000 ml Wasser versetzt und 2 h nachgerührt. Der entstandene Feststoff wurde abfiltriert, mit 250 ml Wasser gewaschen und im Hochvakuum getrocknet. Man erhielt 54.5 g (86% d. Th.) der Titelverbindung.

4-Brom-iV-(ir««s-4-hydroxycyclohexyl)-3-methylbenzamid
In 1.40 1 Dimethylformamid wurden 4-Brom-3 -methylbenzoesäure (121.0 g, 562.7 mmol) sowie ira«s-4-Aminocyclohexanol (71.3 g, 618.9 mmol) vorgelegt und mit A^V-Diisopropylethylamin (294.0 ml, 1688.0 mmol) versetzt. Anschließend wurde /V-[(Dimethylamino)(3 f- [l,2,3]triazolo[4,5-b]pyridin-3-yloxy)methyliden]-ALmethyl-methanaminium-hexafluorophosphat (320.9 g, 844.0 mmol) portionsweise zugefügt wobei die Reaktionstemperatur mittels Eiskühlung unter 30°C gehalten wurden. Die Re aktionsmis chung wurde anschließend in Wasser eingerührt, der Feststoff abgesaugt und mit Wasser nachgewaschen. Der noch feuchte Filterrückstand wurde in Acetonitril verrührt, abgesaugt und mit Acetonitril nachgewaschen. Man erhielt nach Lufttrocknung 157.2 g der Titelverbindung, welche ohne weitere Reinigung weiter umgesetzt wurde.

Alternatives Verfahren: 150 g (697.5 mmol) 4-Brom-3 -methylbenzoesäure wurden in 900 ml Toluol suspendiert und 3.563 g (48.739 mmol) Dimethylformamid zugegeben. Es wurde auf 70°C erhitzt und innerhalb einer Stunde 165.97 g (1.395 mol) Thionylchlorid zugetropft (starke Gasentwicklung !). Nach beendeter Zugabe wurde auf Rückfluss erhitzt, dabei erhielt man eine klare Lösung. Die Lösung wurde im Vakuum eingeengt (40°C, 20 mbar), man gab erneut 900 ml Toluol zu und engte unter gleichen Bedingungen bis zum Öl ein, Ausbeute: quantitativ Säurechlorid. Es wurde mit dem Rohprodukt weitergearbeitet. Hierzu wurden 88.372 g (767.27 mmol) trans-Λ - Amino cy clohexanol in 1500 ml Dichlormethan und 21 1.75 g Triethylamin bei 10°C vorgelegt, das Säure chlorid, gelöst in 750 ml Dichlormethan, über 40 min. zugetropft (Innentemperatur: 10-15°C). Man ließ auf 20°C erwärmen und rührte 2 h nach. Das ausgefallene
Produkt wurde abfiltriert und zweimal mit je 400 ml Dichlormethan nachgewaschen.Es wurde über Nacht bei 40°C im Vakuum getrocknet (< 100 mbar). Ausbeute: 312 g (quantitativ) der Titelverbindung.
Beispiel 8 4-Brom-iV-(iraHS-4-{[teri-butyl(dimethyl)silyl]oxy}cyclohexyl)-3-methylbenzamid
In 3.14 1 Dimethylformamid wurde 4-Brom-AT-(/ra«Ä-4-hydroxycyclohexyl)-3-methylbenzamid (157.0 g, 502.9 mmol) vorgelegt, mit Imidazol (65.0 g, 955.4 mmol) versetzt und anschließend ter t- utyl (chlor) dimethylsil an (106.1 g, 704.0 mmol) zugefügt. Die Reaktionsmischung wurde 4 h bei Raumtemperatur nachgerührt, anschließend in Wasser eingerührt und mit Essigsäureethylester extrahiert. Die gesammelten organischen Phasen wurden mit gesättigter wässriger Natriumhydrogencarbonatlösung sowie mit Wasser gewaschen, über Natriumsulfat getrocknet, filtriert und eingeengt. Der Rückstand wurde in Petrolether verrührt, der Feststoff abgesaugt und mit Petrolether gewaschen. Man erhielt nach Lufttrocknung 180.2 g der Titelverbindung, welche ohne weitere Reinigung weiter umgesetzt wurde.

Alternatives Verfahren: 150 g (480.44 mmol) 4-Brom-N-(trans-4-hydroxycyclohexyl)-3- methylbenzamid und 62.145 g (912.835 mmol) Imidazol wurden in 450 ml Dimethylformamid und 300 ml Tetrahydrofuran vorgelegt und auf 55°C Innentemperatur erhitzt (Suspension). Zu dieser Suspension tropfte man eine Lösung, bestehend aus 101.38 g (672.615 mmol) tert- Butyl(chlor)dimethylsilan in 300 ml T etrahydrofuran zu. Anschließend wurde eine Stunde bei 55°C nachgerührt. Es wurde auf 30°C abgekühlt und 30.8 g Methanol zugegeben. Man rührte 2 h bei Raumtemperatur und setzte 450 ml Wasser, sowie 150 ml gesättigte wässrige Natriumchlorid- Lösung dazu. Es wurde gut durchgerührt und anschließend die Phasen getrennt. Die organische Phase wurde mit 300 ml gesättigter wässriger Natriumchlorid-Lösung ausgeschüttelt und erneut die Phasen getrennt. Es wurde bei 700 mbar und 60°C das Lösungsmittel bis zu einem Endvolumen von 400 ml abdestilliert und anschließend in der Siedehitze 500 ml n-Heptan zugetropft. Es wurde anschließend erneut bis zu einem Endvolumen von 500 ml abdestilliert (Innentemperatur: 92°C). Es wurden 250 ml n-Heptan zugegeben und kurz zum Rückfluss erhitzt,
anschließend auf 20°C abgekühlt. Das Produkt wurde ab filtriert und zweimal mit je 75 ml n- Heptan nachgewaschen. Nach Trocknung über Nacht im Vakuum (40°C, < 100 mbar) erhielt man 170.6 g (83.3% d. Th.) der Titelverbindung.
Beispiel 9 iV-(iraK5-4-{[tert-Butyl(dimethyl)silyl]oxy}cyclohexyl)-3-methyl-4-(4,4,5,5-tetramethyl-l ,3,2- dioxaborolan-2-yl)benzamid
In 2.02 1 Dioxan wurde 4-Brom- V-(/ra«Ä-4-{[feri-butyl(dimethyl)silyl]oxy}cyclohexyl)-3- methylbenzamid (202.0 g, 473.7 mmol) unter Argon vorgelegt und mit Kaliumacetat (139.5 g, 1420.9 mmol) sowie Bis(pinacolato)diboron (144.3 g, 568.4 mmol) versetzt. Anschließend wurde l, -Bis(diphenylphosphino)ferrocenpalladium(II)chlorid-Dichlormethankomplex (1 1.6 g, 14.2 mmol) zugefügt und die Mischung bei 90°C bis zum vollständigen Umsatz gerührt ( l .C -MS Reaktionskontrolle). Anschließend wurde die Re aktionsmi schung bei Raumtemperatur mit Essigsäureethylester verdünnt, durch Kieselgur filtriert und der Filterrückstand mit Essigsäureethylester nachgewaschen. Das gesammelte Filtrat wurde mit Wasser sowie gesättigter wässriger Natriumchloridlösung gewaschen, über Magnesiumsulfat getrocknet, filtriert und eingeengt. Der Rückstand wurde in 1.8 1 Dichlormethan gelöst und mit 300 g Kieselgel (0.04-0.06 μπι) versetzt. Die Mischung wurde abgesaugt und der Rückstand mehrmals mit Dichlormethan gewaschen. Das gesammelte Filtrat wurde eingeengt und der Rückstand aus 1.0 1 Acetonitril umkristallisiert. Man erhielt so 162.3 g der Titelverbindung, welche ohne weitere Reinigung umgesetzt wurde.

Alternatives Verfahren: Durchführung unter Stickstoff ! 241.90 g (567 mmol) 4-Brom-/V-(iraw«-4- {[ieri-butyl(dimethyl)-silyl]oxy} cyclohexyl)-3-methyl-benzamid, 167 g (1.702 mol) Kaliumacetat, 172.864 g (681 mmol) Bis(pinacolato)diboron und 9.264 g (1 1 mmol) Ι,Γ- Bis(diphenylphosphino)-ferrocenpalladium(II)chlorid-Dichlormethankomplex wurden in 725.7 ml Dioxan suspendiert und 2 h bei 90°C gerührt. Nach Abkühlen auf 20°C wurden 725.7 ml Essigsäureethylester und 48.38 g Aktivkohle zugegeben und über Nacht gerührt. Die Aktivkohle und ausgefallenen Salze wurden abfiltriert (über eine Schicht Kieselgur) und der Rückstand
fünfmal mit je 200 ml Essigsäureethylester nachgewaschen. Die organische Lösung wurde mit 484 ml Wasser, 242 ml gesättigter wässriger Natriumchloridlösung gewaschen. Die organische Phase wurde erneut zweimal mit je 363 ml gesättigter wässriger Natriumchloridlösung gewaschen. Die organische Phase wurde weitgehend eingeengt und der Rückstand zweimal in 968 ml Acetonitril aufgenommen und zur Trockene eingedampft. Der Rückstand wurde in 968 ml Acetonitril aufgenommen, zum Rückfluss erhitzt und auf Raumtemperatur abgekühlt. Nach Rühren über Nacht, wurde das Produkt abfiltriert und zweimal mit je 300 ml kaltem Acetonitril gewaschen. Es wurden 222.3 g (82.8% d. Th.) der Titelverbindung erhalten.
Beispiel 10 4-Nitrobenzolcarboximidamid-Hydrochlorid
100 g (675.119 mmol) 4-Nitrobenzonitril wurden in 1000 ml Methanol vorgelegt und 6.1 ml einer Natriummethanolat-Lösung (30%ig in Methanol) zugegeben. Es wurde über Nacht bei 20°C gerührt. Anschließend wurden 39.724 g (742.63 mmol) Ammoniumchlorid zugegeben und 2 h auf 65°C erwärmt. Es wurde im Vakuum zur Trockene eingeengt und der Rückstand in 1000 ml Aceton aufgenommen und 30 min. auf 45°C erhitzt. Dann wurde auf 20°C abgekühlt und 2 h bei dieser Temperatur gerührt. Das Produkt wurde abfiltriert und zweimal mit je 100 ml Aceton gewaschen. Nach Trocknung im Vakuum (40°C, 30 mbar) erhielt man 120.6 g (88.6% d. Th.) der Titelverbindung. Beispiel 1 1
198 g (0.982 mol) 4-Nitrobenzolcarboximidamid-Hydrochlorid wurden in 792 ml Methanol bei 0°C suspendiert und 165.01 g (1.277 mol) Ν,Ν-Diisopropylethylamin zugegeben. Anschließend wurden 178.8 g (1.964 mol) Hydrazinhydrat (55%ige wässrige Lösung) bei 0°C zu getropft und eine Stunde bei dieser Temperatur nachgerührt. Dann wurde eine Stunde bei 20°C nachgerührt, erneut auf 0°C abgekühlt und das Produkt abfiltriert. Es wurde zweimal mit je 75 ml Methanol nachgewaschen. Nach Trocknung im Vakuum (30°C, < 100 mbar) erhielt man 128.9 g (72.85% d. Th.) der Titelverbindung.

Beispiel 12
2,2,3, 3-Tetrafluor-3-[5-(4-nitrophenyl)-liL/-l,2,4-triazol-3-yl]propansäure
Eine Lösung aus 1700 mg (9.4 mmol) 4-Nitrobenzolcarboximidohydrazid in 30 ml Dichlormethan wurde mit 4870 mg (28 mmol) 3,3,4,4-Tetrafluordihydrofuran-2,5-dion 2 min bei Raumtemperatur gerührt. Nun wurden 30 ml Acetonitril zugegeben und 16 h bei Raumtemp eratur gerührt. Es wurde trockenes Molsieb (4Ä) zugesetzt und weitere 24 h bei Raumtemperatur gerührt. Das Molsieb wurde abfiltriert und das Filtrat eingeengt. Der Rückstand wurde mittels präparativer HPLC getrennt (Laufmittel: Gradient von Acetonitril/W asser mit 0.1% Trifluoressigsäure). Die produkthaltigen Fraktionen wurden vereinigt und am Rotationsverdampfer eingeengt. Der Rückstand wurde am Hochvakuum getrocknet. 2434 mg (77% d. Th.) der Titelverbindung wurden erhalten.

Alternatives Verfahren: 100 g (555.04 mmol) 4-Nitrobenzolcarboximidohydrazid wurden in 2000 ml Dichlormethan und 550 ml Acetonitril vorgelegt. Anschließend wurden 190.97 g ( 1.1 1 mol) 3,3,4,4-Tetrafluordihydrofuran-2,5-dion bei 20°C zugetropft. Nach beendeter Zugabe wurde 14 h zum Rückfluss erhitzt. Das Lösungsmittel wurde bis auf ein Restvolumen von 350 ml abdestilliert (300 mbar) und dann 1500 ml Wasser zugegeben. Der pH wird durch Zugabe von 2N wässriger Natronlauge (ca. 720 ml) auf pH 12 eingestellt. Es wurden 1000 ml Essigsäureethylester zugegeben und kräftig gerührt und anschließend die Phasen getrennt. Die wässrige Phase wurde einmal mit 1000 ml Essigsäureethylester nachextrahiert. Anschließend wurde die wässrige Phase durch Zugabe von 6N wässriger Salzsäure auf pH 1 gestellt, dabei fiel das Produkt aus. Es wurde über Nacht nachgerührt, dann das Produkt ab filtriert und mit 100 ml Wasser gewaschen. Es wurden 145.57 g (78.48% d. Th.) der Titelverbindung erhalten.
Beispiel 13
Methyl-2,2,3 ,3 -tetrafluor-3 - [ 5 - (4 -nitrophenyl) -IH- 1 ,2,4-triazol-3 -yljpropanoat
2,2,3,3-Tetrafluor-3-[5-(4-nitrophenyl)-l_f/-l,2,4-triazol-3-yl]propansäure (30.3 g, 90.8 mmol) wurde in Methanol (500 ml) gelöst und mit konzentrierter Schwefelsäure (3 ml) versetzt. Es wurde 22 h bei 65°C gerührt. Dann wurde konzentrierte Schwefelsäure (5 ml) zugegeben und nochmals 22 h bei 65°C gerührt. Es wurde bei Raumtemperatur Natriumhydrogencarbonat bis pH = 7 zugegeben, filtriert und im Vakuum das Lösungsmittel entfernt. Der Rückstand wurde in Pertroletlier und Diethylether verrührt und dann filtriert. Man erhielt 24.2 g (77%) d. Th.) der Titelverbindung.

Alternatives Verfahren: 145.57 g (435.593 mmol) 2 ,2 ,3 , 3 -T etrafluor-3 - [ 5 - (4 -nitrophenyl) - 1 H- l,2,4-triazol-3-yl]propansäure wurden in 1 165 ml Methanol gelöst und 82.917 g (696.95 mmol) Thionylchlorid wurden bei 0°C zugetropft. Anschließend wurde bei 20°C über Nacht nachgerührt. Man engte im Vakuum zur Trockene ein. Es wurden 145.1 g (95.67% d. Th.) der Titelverbindung erhalten.
Beispiel 14
Methyl-3 -[5-(4-aminophenyl)- IH- 1 ,2,4-triazol-3 -yl] -2,2,3 ,3 -tetrafluorpropanoat
Eine Mischung aus Methyl-2,2,3,3-tetrafluor-3-[5-(4-nitrophenyl)-l/ -i,2,4-triazol-3-yl]propanoat (4.83 g, 13.86 mmol) und Zinn(II)chlorid Dihydrat (125.1 1 g, 55.45 mmol) in Methanol (1 15 ml) wurde 1 h unter Rückfluss gerührt. Anschließend wurde die Mischung auf Eiswasser gegeben und mit festem Natriumhydrogencarbonat auf pH 8 eingestellt. Danach wurde mit Wasser verdünnt und dreimal mit Essigsäureethylester extrahiert. Die gesammelten organischen Phasen wurden je einmal mit Wasser sowie gesättigter wässriger Natriumchloridlösung gewaschen, über Natriumsulfat getrocknet und filtriert. Das Filtrat wurde eingeengt und der Rückstand im Hochvakuum getrocknet. Man erhielt 3.32 g (74% d. TL) der Titelverbindung.

Alternatives Verfahren: 1 14.94 g (330.1 mmol) Methyl-2,2,3,3-tetrafluor-3-[5-(4-nitrophenyl)-lH- l,2,4-triazol-3-yl]propanoat und 8.76 g Palladium/Kohle (20%oig, 50%o wasserfeucht) wurden in 1768 ml Tetrahydrofuran vorgelegt und 6 h bei Normaldruck hydriert. Der Katalysator wurde über Kieselgur abfiltriert, mit 295 ml Tetrahydrofuran nachgewaschen und das Filtrat zur Trockene eingeengt. Man erhielt 92.49 g (88.05% d. Th.) der Titelverbindung.

Bewertung der physiologischen Wirksamkeit
Die Eignung der erfindungsgemäßen Verbindungen der Beispiel 6, 33, 34 und 35 zur Behandlung von thromboembolischen Erkrankungen kann in folgendem Assaysystem gezeigt werden: a) Messung der F Ia-Hemmung (In vitro) Zur Bestimmung der Faktor XIa-Hemmung der erfindungsgemäßen Substanzen wird ein biochemisches Testsystem verwendet, in dem die Umsetzung eines peptidischen Faktor XI a- Substrates zur Ermittlung der enzymatischen Aktivität von humanem Faktor XIa benutzt wird. Dabei spaltet Faktor XIa von dem peptischen Faktor XIa-Substrat das C -terminale Aminomethylcoumarin (AMC) ab, dessen Fluoreszenz gemessen wird. Die Bestimmungen werden in Mikrotiterplatten durchgeführt.
Prüfsubstanzen werden in Dimethylsulfoxid aufgelöst und seriell in Dimethylsulfoxid verdünnt (3000 μΜ bis 0.0078 μΜ; resultierende Endkonzentrationen im Test: 50 μΜ bis 0.00013 μΜ). Jeweils 1 μΐ der verdünnten Substanzlösungen werden in die Vertiefungen von weißen Mikrotiterplatten der Firma Greiner (384 Vertiefungen) vorgelegt. Anschließend werden nacheinander 20 μΐ Assaypuffer (50 mmol/1 Tris-Puffer pH 7.4; 100 mmol/1 Natriumchlorid; 5 mmol/1 Calciumchlorid; 0.1% bovines Serumalbumin) und 20 μΐ Faktor XIa der Firma Kordia (0.45 nM in Assaypuffer) hinzugefügt. Nach 15 min Inkubation wird die Enzymreaktion durch Zugabe von 20 μΐ des in Assaypuffer gelösten Faktor X Ia Substrates Boc-Glu(OBzl)-Ala-Arg- AMC (10 μΜ in Assaypuffer) der Firma Bachem gestartet, für 30 min bei Raumtemperatur (22°C) inkubiert und anschließend eine Fluoreszensmessung durchgeführt (Anregung: 360 nM, Emission: 460 nM). Die gemessenen Emissionen der Testansätze mit Prüfsubstanz werden mit denen von Kontrollansätzen ohne Prüfsubstanz (ausschließlich Dimethylsulfoxid anstatt Prüfsubstanz in Dimethylsulfoxid) verglichen und aus den Konzentrations-Wirkungs-Beziehungen ICso-Werte berechnet. b) Bestimmung der antithrombotischen Wirkung (in vivo)
Arterielles Thrombose-Modell (Eisen(II)chlorid-induzierte Thrombose) in Kombination mit Ohrhlutungs/cit im Kaninchen
Die antithrombotische Aktivität der FXIa-Inhibitoren wird in einem arteriellen Thrombose-Modell getestet. Dabei wird die Thrombusbildung durch chemische Beschädigung eines Bereichs der Arteria carotis im Kaninchen ausgelöst. Simultan wird die Ohrblutungszeit bestimmt.
Männliche Kaninchen (CrhKBL (NZW)BR, Charles River) unter normaler Diät mit einem Gewicht von 2.2 - 2.5 kg Körpergewicht werden durch intramuskuläre Applikation von Xylazin und Ketamin (Rompun, Bayer, 5 mg/kg und Ketavet, Pharmacia & Upjohn GmbH, 40 mg/kg Körpergewicht) anästhesiert. Die Anästhesie wird weiterhin durch intravenöse Gabe derselben Präparate (bolus: Dauerinfusion) über die rechte Ohrvene unterstützt.
Nach Freipräparation der rechten Arteria carotis wird der Gefäßschaden dadurch erzeugt, dass ein Stück Filterpapier (10 mm x 10 mm) auf einem Streifen Parafilm® (25 mm x 12 mm) um die A. carotis gewickelt wird, ohne den Blutfluß dadurch zu beeinträchtigen. Das Filterpapier enthält 100 μΕ einer 13%igen Lösung von Eisen(II)chlorid (Sigma) in Wasser. Nach 5 min wird das Filterpapier entfernt und das Gefäß zweimal mit wässriger 0.9%iger Natriumchlorid-Lösung gespült. 30 min nach der Verletzung wird die Arteria carotis im Bereich der Schädigung herauspräpariert und eventuell vorhandenes thrombotisches Material entnommen und gewogen.
Die Prüf Substanzen werden entweder intravenös über die Vena femoralis den anästhesierten oder oral mittels Schlundsonde den wachen Tieren jeweils 5 min beziehungsweise 2 h vor Schädigung verabreicht.
Die Ohrblutungszeit wird 2 min nach der Schädigung der Arteria carotis bestimmt. Hierzu wird das linke Ohr rasiert und ein definierter Schnitt von 3 mm Länge (Klinge Art.Nummer 10-150-10, Martin, Tuttlingen, Germany) parallel zur Ohrlängsachse gesetzt. Dabei wird darauf geachtet, kein sichtbares Gefäß zu verletzen. Eventuell austretendes Blut wird in 15 S ekunden-Intervallen mit genau gewogenen Filterpapierstücken aufgenommen, ohne die Wunde direkt zu berühren. Die Blutungszeit wird berechnet als die Zeitspanne vom Setzen des Schnitts bis zu dem Zeitpunkt, an dem am Filterpapier kein Blut mehr nachweisbar ist. Das ausgetretene Blutvolumen wird nach Wiegen der Filterpapierstücke berechnet.
