Abstract
本発明は、IgGのFab領域に対して優れた選択的吸着能を示す一方で、比較的高いpHの酸性溶出液により当該Fab領域含有タンパク質を放出可能な改変型Fab領域結合性ペプチドを提供することを目的とする。また、本発明は、当該改変型Fab領域結合性ペプチドをリガンドとして有するアフィニティー分離マトリックス、当該アフィニティー分離マトリックスを用いるFab領域含有タンパク質の製造方法、当該改変型Fab領域結合性ペプチドをコードするDNA、当該DNAを含むベクター、および当該ベクターにより形質転換された形質転換体を提供することも目的とする。特定の位置にアミノ酸置換変異を有するプロテインG変異体を利用することによって、上記課題を解決する。
Description
本発明は、IgGのFab領域に結合し、且つ、よりマイルドな酸性条件でFab領域と解離する改変型Fab領域結合性ペプチド、当該改変型Fab領域結合性ペプチドをリガンドとして有するアフィニティー分離マトリックス、当該アフィニティー分離マトリックスを用いるFab領域含有タンパク質の製造方法、当該改変型Fab領域結合性ペプチドをコードするDNA、当該DNAを含むベクター、および当該ベクターにより形質転換された形質転換体に関するものである。
タンパク質の重要な機能の一つとして、特定の分子に特異的に結合する機能が挙げられる。この機能は、生体内における免疫反応やシグナル伝達に重要な役割を果たす。この機能を有用物質の分離精製に利用する技術開発も盛んになされている。実際に産業的に利用されている一例として、抗体医薬を動物細胞培養物から一度に高い純度で精製(キャプチャリング)するために利用される、プロテインAアフィニティー分離マトリックス(以下、プロテインAを「SpA」と省略する場合がある)が挙げられる(非特許文献1,2)。
抗体医薬として開発されているのは基本的にモノクローナル抗体であり、組換え培養細胞技術などを用いて大量に生産されている。「モノクローナル抗体」とは、単一の抗体産生細胞に由来するクローンから得られた抗体を指す。現在上市されている抗体医薬のほとんどは、分子構造的には免疫グロブリンG(IgG)サブクラスである。また、免疫グロブリンを断片化した分子構造を有する抗体誘導体(断片抗体)からなる抗体医薬も盛んに臨床開発されており、免疫グロブリンのFab断片からなる抗体医薬が複数上市された(非特許文献3)。
抗体医薬製造工程における初期精製工程には、先述のSpAアフィニティー分離マトリックスが利用されている。しかし、SpAは基本的にIgGのFc領域に特異的に結合するタンパク質である。よって、Fc領域を含まない断片抗体は、SpAアフィニティー分離マトリックスを利用したキャプチャリングができない。従って、抗体医薬精製プロセスのプラットフォーム化の観点から、IgGのFc領域を含まない断片抗体をキャプチャリング可能なアフィニティー分離マトリックスに対する産業的なニーズは高い。
IgGのFc領域以外に結合するタンパク質はすでに複数知られている(非特許文献4)。しかし、そのようなタンパク質をリガンドとしたアフィニティー分離マトリックスが、SpAアフィニティー分離マトリックスと同様に、抗体医薬の精製に標準的に産業利用されているという事実はない。
例えば、グループGの連鎖球菌(Streptococcus sp.)より見出されたプロテインGと呼ばれるタンパク質(以下、プロテインGを「SpG」と略記する場合がある)は、IgGに結合する性質を有し、このSpGをリガンドとして固定化したSpGアフィニティー分離マトリックス製品もある(GEヘルスケア社製,製品名「Protein-G Sepharose 4 Fast Flow」,特許文献1)。SpGはIgGのFc領域に強く結合するが、Fab領域にも弱いながら結合することが分かっている(非特許文献4,5)。しかし、SpGのFab領域への結合力は弱いので、SpGアフィニティー分離マトリックス製品は、Fc領域を含まずFab領域のみを含む断片抗体の保持性能は低いといえる。そこで、SpGに変異を導入することでFab領域への結合力を向上する取組みがなされている(特許文献2,非特許文献6)。
一方、このようなアフィニティー分離マトリックス製品に使われるリガンドは、相手分子への結合が強い必要があるだけでなく、該マトリックスに吸着した相手分子が回収できるよう適切な条件で結合が解離する必要がある。SpGのようなタンパク質とIgG類との結合は酸性条件下で解離する。SpGとFabとの相互作用は、pHが低い方が確実に解離するが、pHが低いとFabに化学修飾や切断などといったダメージがあるため、なるべく中性に近いpHで解離することが好ましい。例えば、ヒスチジン(His)の側鎖のpKaはpH6付近であるので、SpGに対してHisに置換する変異を導入し、Fcと解離するpHがやや中性側にシフトした例(特許文献3,非特許文献7)や、Fabとの解離するpHがやや中性側にシフトした例(非特許文献6)がある。但し、アフィニティー分離マトリックスを用いたクロマト操作では、HisのpKaより中性側のpH5程度で中間洗浄することもあるため、変異する部位および変異するアミノ酸の種類に関する研究は今後も必要と考えられる。

上述したように、免疫グロブリンまたはその断片を精製するためのアフィニティー分離マトリックスは種々開発されている。アフィニティー分離マトリックスにより免疫グロブリンまたはその断片を精製するには、一般的に、中性の溶液とアフィニティー分離マトリックスとを接触させて免疫グロブリンまたはその断片を選択的に吸着させ、洗浄して不純物を除去し、次いで酸性の溶出液により吸着された免疫グロブリンまたはその断片を溶出する。この際、純度の高い目的化合物を得るには、選択的吸着力の高いアフィニティー分離マトリックスを用い、洗浄を十分に行う必要がある。そして、目的化合物を高い回収率で得るために、選択的吸着を確実に解離できる酸性pHで溶出する必要がある。しかし、強い酸性条件下では、目的化合物である免疫グロブリンまたはその断片がダメージを受けるおそれがあった。
本発明者は、過去の検討より、野生型のプロテインG(SpG)のFab結合力が、Fc結合力に比較すると弱く、アフィニティー分離マトリックスのリガンドとしてFab結合力を強化する必要があることを見出していた(特許文献2)。しかし、変異によってSpGのFab結合力を強化することで、変異導入前に比較して、Fabが解離するpHがより強い酸性側にシフトする、即ち、酸解離pH値が低下する場合があることも判明した。
そこで本発明は、IgGのFab領域に対して優れた選択的吸着能を示す一方で、比較的高いpHの酸性溶出液により当該Fab領域含有タンパク質を放出可能な改変型Fab領域結合性ペプチドを提供することを目的とする。また、本発明は、当該改変型Fab領域結合性ペプチドをリガンドとして有するアフィニティー分離マトリックス、当該アフィニティー分離マトリックスを用いるFab領域含有タンパク質の製造方法、当該改変型Fab領域結合性ペプチドをコードするDNA、当該DNAを含むベクター、および当該ベクターにより形質転換された形質転換体を提供することも目的とする。
本発明者は、上記課題を解決するために、SpGのIgG結合ドメインの変異体を分子設計し、タンパク質工学的手法および遺伝子工学的手法を用いて該変異体を形質転換細胞から取得し、取得した該変異体の特性を比較検討する検討を行った。その結果、SpGのIgG結合性ドメインに特定の変異を導入することにより上記課題を解決できることを見出して、本発明を完成した。
具体的には、本発明者は、以下のような分子計算化学の手法を用いて、Fab結合力を大きく損なうことなく、酸解離pHに影響を与え得る部位を特定した。
即ち、SpGのIgG結合性ドメインとIgGのFab領域の複合体の結晶構造(Derrick J.P.,Wigley D.B.著,J.Mol.Biol.,1994年,243巻,906-918頁)の構造座標をPDB(Protein Data Bank,http://www.rcsb.org/pdb/home/home.do,当該座標のPDBコード:1IGC)より入手し、分子モデリングの雛形座標として選択した。次に、Swiss-PDBViewer(Guex N.ら,Electrophoresis,1997年,18巻,2714-2723頁)などの三次元グラフィックス・プログラムを用いて、当該結晶構造中のIgG結合性ドメインがSpGのβ1ドメインのアミノ酸配列(配列番号1)に適合するよう分子モデリングを行い、比較対象用のモデリング構造とした。次に、SpGのIgG結合性ドメインに変異を導入した様々なアミノ酸配列に関しても同様の作業を行った。タンパク質の安定性については、構造エネルギーによって議論することが可能である(A.R.リーチ著,「分子モデリング概説」,2004年,第11章)。分子モデリング構造(主鎖のフレームワーク)を利用すれば、分子力場法を利用した分子シミュレーション計算(エネルギー極小化計算)によって、SpGのIgG結合性ドメインとIgGのFab領域の複合体の構造エネルギーが計算できる。従って、変異導入前後の各々のモデリング構造の構造エネルギーを比較することで、タンパク質複合体の安定性を指標に、変異が与える影響を評価することができる。具体的には、プログラムShrike(特開2001-184381号公報)を用い、溶媒の誘電率に係るパラメータを変えたいくつかのエネルギー極小化計算(力場パラメータ:CHARMM,主鎖α炭素位置束縛:0.01 kcal/A)の計算結果を比較することによって評価した。
その結果、SpGのIgG結合ドメインの第17位および第36位が変異部位として適切であることを見出した。第17位は、FabのCH1との相互作用界面からは離れたところに位置する。驚くべきことに、第17位への変異は、溶媒条件によって、Fab結合力を極端に損なわない程度に、タンパク質複合体の安定性を変える可能性が示唆された。また、第36位は、FabのCLとの相互作用界面に位置する。その相互作用界面面積はCH1との相互作用界面に比較するとかなり狭いにもかかわらず、驚くべきことに、第36位への変異は、溶媒条件によって、Fab結合力を極端に損なわない程度にタンパク質複合体の安定性を変える可能性が示唆された。
その結果として完成した本発明を以下に示す。
[1] 下記(1)~(3)の何れかの改変型Fab領域結合性ペプチド。
(1) 配列番号1~5のいずれかのアミノ酸配列において、第17位および第36位からなる群より選択される1つ以上のアミノ酸残基が置換されているアミノ酸配列を有し、且つ、その酸解離pHが置換導入前に比べて中性側にシフトしているFab領域結合性ペプチド;
(2) 上記(1)に規定されるアミノ酸配列において、上記第17位および第36位を除く領域中で1個または数個のアミノ酸がさらに欠失、置換および/または付加されたアミノ酸配列を有するFab領域結合性ペプチドであり、且つ、その酸解離pHが変異導入前に比べて中性側にシフトしているFab領域結合性ペプチド;
(3) 上記(1)に規定されるアミノ酸配列に対して80%以上の配列同一性を有するアミノ酸配列を有し、且つ、その酸解離pHが変異導入前に比べて中性側にシフトしているFab領域結合性ペプチド(但し、上記(1)に規定されるアミノ酸配列における第17位および第36位からなる群より選択される1以上の置換アミノ酸残基は、(3)においてさらに変異しないものとする)。
(2) 上記(1)に規定されるアミノ酸配列において、上記第17位および第36位を除く領域中で1個または数個のアミノ酸がさらに欠失、置換および/または付加されたアミノ酸配列を有するFab領域結合性ペプチドであり、且つ、その酸解離pHが変異導入前に比べて中性側にシフトしているFab領域結合性ペプチド;
(3) 上記(1)に規定されるアミノ酸配列に対して80%以上の配列同一性を有するアミノ酸配列を有し、且つ、その酸解離pHが変異導入前に比べて中性側にシフトしているFab領域結合性ペプチド(但し、上記(1)に規定されるアミノ酸配列における第17位および第36位からなる群より選択される1以上の置換アミノ酸残基は、(3)においてさらに変異しないものとする)。
[2] 上記(1)に規定されるアミノ酸配列において、第17位のアミノ酸残基が置換されている上記[1]に記載の改変型Fab領域結合性ペプチド。
[3] 上記(1)に規定されるアミノ酸配列において、第17位がAla、Val、Ser、LeuまたはIleに置換されている上記[1]または[2]に記載の改変型Fab領域結合性ペプチド。
[4] 上記(1)に規定されるアミノ酸配列において、第36位がAla、Val、ThrまたはGluに置換されている上記[1]~[3]のいずれかに記載の改変型Fab領域結合性ペプチド。
[5] 上記(2)に規定されるアミノ酸配列において、上記変異の部位が、第6位、第7位、第10位、第13位、第15位、第18位、第19位、第21位、第24位、第28位、第29位、第30位、第31位、第33位、第35位、第37位、第39位、第40位、第42位および第47位からなる群より選択される1以上の部位である上記[1]~[4]のいずれかに記載の改変型Fab領域結合性ペプチド。
[6] 上記(2)に規定されるアミノ酸配列において、上記変異の部位がN末端および/またはC末端である上記[1]~[5]のいずれかに記載の改変型Fab領域結合性ペプチド。

本発明によって得られた改変型Fab領域結合性ペプチドを不溶性担体に固定化したアフィニティー分離マトリックスは、Fab領域含有タンパク質に対して優れた選択的吸着能を示す。その一方で、優れた酸解離特性という互いに相反する特性を示す。ここでの「優れた酸解離特性」とは、より中性側の酸性溶出条件でFab領域含有タンパク質が解離され、且つ、Fab領域含有タンパク質を酸性条件下で溶出させたときの溶出ピーク・プロファイルがよりシャープであるといった特性を指す。クロマトグラフィーの溶出ピークがよりシャープになることで、より少ない容量の溶出液でより高濃度の抗体含有溶出液を回収可能とすることができる。
本発明に係る改変型Fab領域結合性ペプチドは、下記(1)~(3)の何れかのものである。
改変型Fab領域結合性ペプチド(1): 配列番号1~5のいずれかのアミノ酸配列において、第17位および第36位からなる群より選択される1つ以上のアミノ酸残基が置換されているアミノ酸配列を有し、且つ、その酸解離pHが置換導入前に比べて中性側にシフトしているFab領域結合性ペプチド;
改変型Fab領域結合性ペプチド(2): 上記(1)に規定されるアミノ酸配列において、上記第17位および第36位を除く領域中で1または数個のアミノ酸がさらに欠失、置換および/または付加されたアミノ酸配列を有するFab領域結合性ペプチドであり、且つ、その酸解離pHが変異導入前に比べて中性側にシフトしているFab領域結合性ペプチド;
改変型Fab領域結合性ペプチド(3): 上記(1)に規定されるアミノ酸配列に対して80%以上の配列同一性を有するアミノ酸配列を有し、且つ、その酸解離pHが、変異導入前に比べて中性側にシフトしているFab領域結合性ペプチド(但し、上記(1)に規定されるアミノ酸配列における第17位および第36位からなる群より選択される1以上の置換アミノ酸残基は、(3)においてさらに変異しないものとする)。

改変型Fab領域結合性ペプチド(3): 上記(1)に規定されるアミノ酸配列に対して80%以上の配列同一性を有するアミノ酸配列を有し、且つ、その酸解離pHが、変異導入前に比べて中性側にシフトしているFab領域結合性ペプチド(但し、上記(1)に規定されるアミノ酸配列における第17位および第36位からなる群より選択される1以上の置換アミノ酸残基は、(3)においてさらに変異しないものとする)。
「免疫グロブリン(IgG)」は、リンパ球のB細胞が産生する糖タンパク質であり、特定のタンパク質などの分子を認識して結合する働きを持つ。免疫グロブリンは、抗原と呼ばれる特定の分子に特異的に結合する機能と、他の生体分子や細胞と協同して抗原を有する因子を無毒化・除去する機能を有する。免疫グロブリンは、一般的に「抗体」と呼ばれるが、それはこのような機能に着目した名称である。全ての免疫グロブリンは、基本的には同じ分子構造からなり、それぞれ2本ずつの軽鎖と重鎖のポリペプチドから構成されている4本鎖“Y”字型構造を基本構造としている。軽鎖(L鎖)には、λ鎖とκ鎖の2種類があり、すべての免疫グロブリンはこのどちらかを持つ。重鎖(H鎖)には、γ鎖、μ鎖、α鎖、δ鎖、ε鎖という構造の異なる5種類があり、この重鎖の違いによって免疫グロブリンの種類(アイソタイプ)が変わる。免疫グロブリンG(IgG)は、単量体型の免疫グロブリンで、2本の重鎖(γ鎖)と2本の軽鎖から構成され、2箇所の抗原結合部位を持っている。
免疫グロブリンの“Y”字の下半分の縦棒部分にあたる場所をFc領域と呼び、上半分の“V”字の部分をFab領域と呼ぶ。Fc領域は抗体が抗原に結合した後の反応を惹起するエフェクター機能を有し、Fab領域は抗原と結合する機能を有する。重鎖のFab領域とFc領域はヒンジ部でつながっており、パパイヤに含まれるタンパク分解酵素パパインは、このヒンジ部を分解して2つのFab領域と1つのFc領域に切断する。Fab領域のうち“Y”字の先端に近い部分のドメインは、多様な抗原に結合できるように、アミノ酸配列に多彩な変化が見られるため、可変領域(V領域)と呼ばれている。軽鎖の可変領域をVL領域、重鎖の可変領域をVH領域と呼ぶ。V領域以外のFab領域とFc領域は、比較的変化の少ない領域であり、定常領域(C領域)と呼ばれる。軽鎖の定常領域をCL領域と呼び、重鎖の定常領域をCH領域と呼ぶが、CH領域はさらにCH1~CH3の3つに分けられる。重鎖のFab領域はVH領域とCH1からなり、重鎖のFc領域はCH2とCH3からなる。ヒンジ部はCH1とCH2の間に位置する。SpG-βのIgGへの結合は、より詳細には、IgGのCH1領域(CH1γ)とCL領域への結合であり、特にCH1への結合が主要である(非特許文献5)。
本発明に係る改変型Fab領域結合性ペプチドは、免疫グロブリンのFab領域に結合する。本発明ペプチドが結合すべきFab領域含有タンパク質は、Fab領域を含むものであれば特に制限されず、Fab領域とFc領域を不足なく含有する免疫グロブリン分子であってもよいし、免疫グロブリン分子の誘導体であってもよい。本発明に係る改変型Fab領域結合性ペプチドが結合する免疫グロブリン分子誘導体は、Fab領域を有する誘導体であれば特に制限されない。例えば、免疫グロブリンGのFab領域のみに断片化されたFabフラグメント、ヒト免疫グロブリンGの一部のドメインを他生物種の免疫グロブリンGのドメインに置き換えて融合させたキメラ型免疫グロブリンG、Fc領域の糖鎖に分子改変を加えた免疫グロブリンG、薬剤を共有結合したFab断片などを挙げることができる。
本発明において「ペプチド」とは、ポリペプチド構造を有するあらゆる分子を含むものであって、いわゆるタンパク質のみならず、断片化されたものや、ペプチド結合によって他のペプチドが連結されたものも包含されるものとする。「ドメイン」とは、タンパク質の高次構造上の単位であり、数十から数百のアミノ酸残基配列から構成され、なんらかの物理化学的または生物化学的な機能を発現するに十分なタンパク質の単位をいう。タンパク質やペプチドの「変異体」は、野生型のタンパク質やペプチドの配列に対し、アミノ酸レベルで、少なくとも1つ以上の置換、付加または欠失が導入されたタンパク質またはペプチドをいう。アミノ酸を置換する変異の表記について、置換位置の番号の前に、野生型または非変異型のアミノ酸を付し、置換位置の番号の後に、変異したアミノ酸を付して表記する。例えば、17位のThrをValに置換する変異は、T17Vと記載する。
「プロテインG(SpG)」は、グループGの連鎖球菌(Streptococcus sp.)の細胞壁に由来するタンパク質である。SpGは、ほとんどの哺乳類の免疫グロブリンG(IgG)と結合する能力を有しており、IgGのFc領域に強く結合し、IgGのFab領域にも弱く結合する。
SpGのIgG結合性を示す機能ドメインは、βドメイン(SpG-β)と呼ばれる。なお、β(B)ドメインと呼ぶ場合と、Cドメインと呼ぶ場合の2通りがあり(Akerstrom et al.,J.Biol.Chem.,1987,28,13388-,Fig.5参照)、本明細書では、Fahnestockらの定義に従ってβドメインと呼ぶ(Fahnestock et al.,J.Bacteriol.,1986167,870-)。
上記改変型Fab領域結合性ペプチド(1)に規定されるアミノ酸配列は、基本的にSpG-βとしての構造・機能を有するアミノ酸配列である。上記改変型Fab領域結合性ペプチド(1)としては、Fab結合力が向上したSpG-β変異体が好ましく、例えば、配列番号1~5のいずれかで示されるアミノ酸配列を有するSpG-β変異体のさらなる変異体が好ましい。
本発明における好適な形態の1つとして、上記(1)に規定される配列番号1~5のアミノ酸配列のうち、配列番号2で示される、Fab結合力が向上したSpG-β変異体のアミノ酸配列が好ましい。また、野生型SpG-βに本発明を適用することも可能であり、本発明における好適な形態の1つとして、上記(1)に規定される配列番号1~5のアミノ酸配列のうち、グループGの連鎖球菌のGX7809株由来のβ1ドメイン(SpG-β1,配列番号3)またはβ2ドメイン(SpG-β2,配列番号4)のアミノ酸配列であることが好ましい。本発明における好適な形態の1つとして、熱安定性に優れた変異型SpG-βとして、例えば、公知特許文献(特開2003-88381号公報)に記載のアミノ酸配列(配列番号5)が挙げられる。なお、当該アミノ酸配列は、特開2003-88381号公報の配列番号8のアミノ酸配列(C36ペプチド)に該当する。なお、配列番号1~5に関しては、実験上の理由からN末端/C末端のアミノ酸に改変を加えている場合がある。
本発明は、配列番号1~5のいずれかのアミノ酸配列に特定のアミノ酸置換変異を導入することによって、Fab領域を含むタンパク質の酸解離pHを、変異導入前に比べて中性側にシフトさせることを特徴とする技術である。本発明のFab領域結合性ペプチド(1)において、特定のアミノ酸置換変異とは、第17位および第36位からなる群より選択される1つ以上のアミノ酸残基を他のアミノ酸に置換する変異のことであり、変異導入前(置換導入前)のアミノ酸配列とは、配列番号1のアミノ酸配列である。例えば、上記(1)に記載の置換がT17Aのみであった場合、このペプチドのアミノ酸配列にA17T(復帰変異)を導入したアミノ酸配列が、置換導入前のアミノ酸配列である。
本発明における改変型Fab領域結合性ペプチド(1)は、後記の実施例において確かめられている通り、より高pHの酸性溶出条件でFab領域含有タンパク質を解離し、且つ、Fab領域含有タンパク質を酸性条件下で溶出させたときの溶出ピークがよりシャープである。
本発明における改変型Fab領域結合性ペプチド(1)の置換導入前のアミノ酸配列としては、配列番号1~5のいずれかのアミノ酸配列が好ましく、配列番号1または配列番号2のアミノ酸配列がより好ましく、配列番号1のアミノ酸配列が最も好ましいが、これらに限定はされない。
本発明における改変型Fab領域結合性ペプチド(1)は、上述の第17位および第36位からなる群より選択される1つ以上のアミノ酸残基が置換されたアミノ酸配列を有する。置換される位置としては、第17位がより好ましい。例えば、配列番号1~5のアミノ酸配列において、第17位はThr、第36位はAspである。
本発明においてペプチドが「(特定の)アミノ酸配列を有する」とは、そのペプチドのアミノ酸配列が特定されたアミノ酸配列を含んでいればよく、且つ、そのペプチドの機能が維持されていることを意味する。ペプチドにおいて特定されたアミノ酸配列以外の配列としては、ヒスチジンタグや固定化のためのリンカー配列の他、-S-S-結合などの架橋構造などが挙げられる。
例えばN末端にペプチドが付加されたような場合であっても、配列同一性を基準に、配列番号1~5のアミノ酸配列の第17位および第36位に相当する位置を同定することは、当業者であれば容易に可能である。例えば、遺伝情報処理ソフトウエアであるGENETYX(https://www.genetyx.co.jp/)のアラインメント機能を使って位置を特定することが可能である。
変異後のアミノ酸、即ち配列番号1~5のアミノ酸配列における第17位のThrおよび/または第36位のAspに置き換わるアミノ酸の種類は、非タンパク質構成アミノ酸や非天然アミノ酸への置換を含め、特に限定されるものではないが、遺伝子工学的生産の観点から、天然型アミノ酸を好適に用いることができる。さらに、天然型アミノ酸は、中性アミノ酸;AspとGluの酸性アミノ酸;Lys、Arg、Hisの塩基性アミノ酸に分類される。中性アミノ酸は、脂肪族アミノ酸;Proのイミノ酸;Phe、Tyr、Trpの芳香族アミノ酸に分類される。脂肪族アミノ酸は、さらに、Gly;Ala;Val、Leu、Ileの分枝アミノ酸;Ser、Thrのヒドロキシアミノ酸;Cys、Metの含硫アミノ酸;Asn、Glnの酸アミドアミノ酸に分類される。また、Tyrはフェノール性水酸基を有することから、芳香族アミノ酸のみでなくヒドロキシアミノ酸に分類してもよい。さらに、別の観点からは、天然アミノ酸を、Gly、Ala、Val、Leu、Ile、Trp、Cys、Met、Pro、Pheの疎水性の高い非極性アミノ酸;Asn、Gln、Ser、Thr、Tyrの中性の極性アミノ酸;Asp、Gluの酸性の極性アミノ酸;Lys、Arg、Hisの塩基性の極性アミノ酸に分類することもできる。また、酸性条件でプロトネーションする官能基を側鎖に有するアミノ酸であるHis、AspおよびGluを好適に用いることもできる。
第17位に置換するアミノ酸としては、Ala、Val、Ser、LeuまたはIleが好ましく、AlaまたはValがより好ましく、Valがさらにより好ましい。第36位で置換するアミノ酸としては、Ala、Val、ThrまたはGluが好ましく、Ala、ValまたはThrがより好ましく、Valがよりさらに好ましい。また、別の観点からは、第17位と第36位の両位置において、βシート構造を不安定化する可能性のあるGlyは好ましくない。
本発明における改変型Fab領域結合性ペプチド(2)は、上記(1)のアミノ酸配列において、上記第17位および第36位を除く領域中で、1個または数個のアミノ酸残基が、欠失、置換および付加から選択される1以上の変異を付与されたアミノ酸配列を有し、且つ、その酸解離pHが当該変異の導入前に比べて中性側にシフトしているFab領域結合性ペプチドである。
本発明に係る改変型Fab領域結合性ペプチド(2)に関し、「1個から数個」の範囲とは、欠失などを有する改変型Fab領域結合性ペプチドが免疫グロブリンのFab領域への高い結合力を有する限り特に限定されるものではない。上記「1個から数個」の範囲は、例えば30個以下とすることができ、好ましくは20個以下、より好ましくは10個以下、さらに好ましくは7個以下、一層好ましくは5個以下、特に好ましくは3個以下、1個以上または2個以下、1個程度であることができる。
1個または数個の変異位置は、第17位または第36位以外であれば、どの部位でも問題ない。例えば、配列番号1~3の間でアミノ酸の種類が異なる部位である、第10位、第13位、第15位、第18位、第19位、第21位、第28位、第30位、第31位、第33位および第39位が変異位置として好ましい。また、配列番号3~5の間でアミノ酸の種類が異なる部位である、第6位、第7位、第19位、第24位、第28位、第29位、第31位、第35位、第40位、第42位および第47位も変異位置として好ましい。
さらに、アミノ酸残基の欠失、置換および/または付加の位置としては、例えば、N末端および/またはC末端を挙げることができる。これら部位は、特に欠失および/または付加の部位として好ましい。付加するアミノ酸配列の実施形態としては、該ペプチドをマトリックスに固定化する際に有用な、LysやCysを含むアミノ酸配列をC末端側に付加することが挙げられる。
なお、上記の欠失や付加が加わってアミノ酸数が変化した場合であっても、変異導入前におけるアミノ酸残基の位置に対応する変異導入後におけるアミノ酸残基の位置は、変異導入前後のアミノ酸配列のアライメント解析を行えば容易に検索可能である。このようなアライメント解析の手法は、改変型Fab領域結合性ペプチド(1)に関して説明したとおり、当業者に広く知られている。
本発明に係る改変型Fab領域結合性ペプチド(3)に関し、配列同一性としては、80%以上が好ましく、85%以上がより好ましく、90%以上がさらに好ましく、95%以上がさらに好ましく、98%以上がさらに好ましい。上記配列同一性は、改変型Fab領域結合性ペプチド(1)に関して説明したとおり、アミノ酸配列多重アラインメント用プログラムであるClustal(http://www.clustal.org/omega/)などを使って測定することができる。
但し、上記(1)に規定されるアミノ酸配列における第17位および第36位からなる群より選択される1以上の置換アミノ酸残基は、(3)においてさらに変異しないものとする。変異導入前のアミノ酸配列のアミノ酸数が異なる場合においても、配列同一性が80%以上である場合に、配列番号1~5の第17位または第36位に相当する位置を同定することは、当業者であれば容易に可能である。
本発明に係るFab領域結合性ペプチド(1)~(3)は、Fab領域含有タンパク質に対する酸解離pHが、置換導入前または変異導入前に比べて中性側にシフトすることを特徴とする。一般的に、強酸条件下や強塩基条件下では、分子表面電荷の変化や変性による立体構造の変化が要因となって、ペプチド間の結合は失われる。「酸解離pH」とは、酸性側(値が7より小さいpH)でその特異的結合が失われ解離するpHのことである。酸解離pHは、本発明に係るFab領域結合性ペプチド(1)~(3)をリガンドとするアフィニティー分離マトリックスに吸着したFab領域含有タンパク質を分離(溶出)することが可能な、最も高いpH値ということができる。よって、本明細書においては、酸解離pHと酸溶出pHは、基本的には同義である。酸解離pHが「置換導入前または変異導入前に比べて中性側にシフトする」というのは、結合の解離に必要なpHの数値が、置換または変異の導入前の方が小さく、置換または変異の導入後の方が大きいということを意味する。即ち、置換または変異の導入後の方が、より弱い酸性溶液で結合が解離するということを意味する。
酸解離pHの数値は、Fab領域含有タンパク質の種類、酸性溶液のバッファー成分の種類や濃度などによっても変わり、アフィニティー分離マトリックスの場合には、水不溶性担体の種類・構造やリガンド固定化用リンカーの種類・構造によっても変わるので、一義的に数値の範囲で示すのは難しい。一般的には、変異を導入する前の酸解離pHは2.0以上3.5以下程度であるのが、本発明に係る変異を導入することによって、pH3.0以上4.5以下程度にシフトするが、これには限定されない。この条件下で溶出すると、抗体へのダメージが小さい(Ghose S.ら,Biotechnology and bioengineering,2005年,92巻,6号)。本発明に係る変異を導入することによって、酸解離pHは、pH値にして0.1以上中性側にシフトすることが好ましく、pH値にして0.2以上中性側にシフトすることがより好ましく、pH値にして0.3以上中性側にシフトすることがさらにより好ましく、pH値にして0.4以上中性側にシフトすることがさらにより好ましく、pH値にして0.5以上中性側にシフトすることがさらにより好ましい。
変異導入前後の酸解離pHを評価する方法は、生体分子間相互作用を検出できる手法であれば特に限定はされないが、例えば、表面プラズモン共鳴原理を用いたBiacoreシステム(GEヘルスケア社)などのバイオセンサーによって試験することができる。酸解離pHの測定方法としては、例えば、結合する一方のペプチドまたはタンパク質をセンサーチップに固定化し、所望のpHに調整したもう一方のタンパク質溶液またはペプチド溶液を流路に添加し、結合シグナルの有無を検出し、結合シグナルが検出できなくなるpHを評価すればよいが、これに限定されない。別の方法としては、ペプチド溶液またはタンパク質溶液を添加した後に、所望のpHの緩衝液を添加し、添加した前後でのチップ上で解離せずに残った結合シグナルの変化を評価してもよい。測定条件としては、温度は20~40℃の一定温度にし、結合状態を見るときのpHはpH5~8の中性条件にすることが好ましい。緩衝液の成分としては、中性の場合には、リン酸、トリス、ビストリスなどが挙げられ、酸性の場合には、酢酸、クエン酸、グリシンなどが挙げられるが、これらに限定されない。緩衝液のNaCl濃度は、特に限定はされないが、0~0.15M程度が好ましく、特に解離状態を見るときは0Mが好ましい。この評価においては、変異導入前後の違いを比較するために、変異以外の条件を全て揃えることが重要である。例えば、Biacoreシステムを用いた評価では、Fab領域結合性ペプチドを、チップに固定化する側(固相)としてもよいし、流路添加する側(液相)としても、どちらでも評価は可能である。例えば、Fab領域結合性ペプチドをチップに固定化する量を揃えるのが難しい場合には、流路添加する際の濃度を揃えてFab領域結合性ペプチドを流路添加する方が好ましい。
酸解離pHを評価する方法として、アフィニティー分離マトリックスを用いて、クロマトグラフィー・プロファイルを評価する方法も好ましい。例えば、結合する一方のペプチドまたはタンパク質をクロマトグラフィー用カラムに固定化し、そのカラムをクロマトグラフィー装置に接続した後で、もう一方のタンパク質溶液またはペプチド溶液をそのカラムに添加し、pHを徐々に下げる直線勾配をかけて吸着したタンパク質またはペプチドが溶出したピークトップのpHを評価すればよいが、これに限定されない。測定時の温度、緩衝液、塩濃度などは上述の条件に従うのが好ましいが、特に限定されない。例えば、鮮明なpH直線勾配を得る為に、結合用緩衝液(緩衝液A)と解離用緩衝液(緩衝液B)の成分はpH以外同一とすることが好ましい。この評価においても、変異導入前後の違いを比較するために、変異以外の条件を全て揃えることが重要である。この評価系においても、Fab領域結合性ペプチドを、カラムに固定化する側(固相)としてもよいし、添加して溶出する側(液相)としても、どちらでも評価は可能である。例えば、Fab領域結合性ペプチドをカラムに固定化する量を揃えるのが難しい場合には、カラムに添加する際の濃度を揃えてFab領域結合性ペプチドを添加し、得られたクロマトグラフィー・プロファイルを重ねて比較する方が好ましい。
プロテインGは、免疫グロブリン結合性ドメインを2個または3個タンデムに並んだ形で含むタンパク質である。本発明に係る改変型Fab領域結合性ペプチドも、実施形態の1つとして、単量体または単ドメインである当該改変型Fab領域結合性ペプチドが2個以上、好ましくは3個以上、より好ましくは4個以上、より好ましくは5個以上連結された複数ドメインの多量体であってもよい。連結されるドメイン数の上限は、例えば10個、好ましくは8個、より好ましくは6個である。これらの多量体は、単一の改変型Fab領域結合性ペプチドの連結体であるホモダイマーやホモトリマー等のホモポリマーであってもよいし、複数種類の改変型Fab領域結合性ペプチドの連結体であるヘテロダイマーやヘテロトリマー等のヘテロポリマーであってもよい。
本発明に係る改変型Fab領域結合性ペプチドの連結のされ方としては、1個または複数個のアミノ酸残基で連結する方法、および、アミノ酸残基を挟まず直接連結する方法が挙げられるが、これらの方法に限定されるものではない。連結するアミノ酸残基数に特に制限は無いが、好ましくは1残基以上20残基以下であり、より好ましくは15残基以下であり、さらにより好ましくは10残基以下であり、さらにより好ましくは5残基以下であり、さらにより好ましくは2残基以下である。好ましくは、野生型SpGのβ1とβ2の間、または、β2とβ3の間を連結している配列を利用するのがよい。また、別の観点からは、単量体改変型Fab領域結合性ペプチドの3次元立体構造を不安定化しないものが好ましい。
また、本発明に係る改変型Fab領域結合性ペプチドは、改変型Fab領域結合性ペプチド(1)~(3)で規定するアミノ酸配列を有するものであればよく、他のペプチドや化合物などが結合していてもよいものとする。例えば、本発明の実施形態の1つとして、本発明により得られる改変型Fab領域結合性ペプチド、または、当該ペプチドが2個以上連結された多量体が、1つの構成成分として、機能の異なる他のペプチドと融合されていることを特徴とする融合ペプチドが挙げられる。融合ペプチドの例としては、アルブミンやGST(グルタチオンS-トランスフェラーゼ)が融合したペプチドを例として挙げることができるが、これに限定されるものではない。また、DNAアプタマーなどの核酸、抗生物質などの薬物、PEG(ポリエチレングリコール)などの高分子が融合されている場合も、本発明で得られたペプチドの有用性を利用するものであれば、本発明に包含される。
本発明に係る改変型Fab領域結合性ペプチド(1)~(3)は、アフィニティー・リガンドとしてカラムに保持可能であり、優れたFab結合力を示す。Fab領域に対する親和性は、例えば、表面プラズモン共鳴原理を用いたBiacoreシステムなどのバイオセンサーによって試験することができるが、これに限定されるものではない。
結合パラメータとしては、例えば、親和定数(KA)や解離定数(KD)を用いることができる(永田ら著,「生体物質相互作用のリアルタイム解析実験法」,シュプリンガー・フェアラーク東京,1998年,41頁)。親和定数は、結合速度定数(kon)を解離速度定数(koff)で除した値である(KA=kon/koff)。
本発明に係る改変型Fab領域結合性ペプチドとFab領域含有タンパク質との親和定数は、例えば、Biacoreシステムを利用して、センサーチップにFab領域含有タンパク質を固定化して、温度25℃、pH7.4の条件下にて、本発明に係る改変型Fab領域結合性ペプチドを流路添加する実験系で求めることができる。本発明に係る変異を導入した配列を有するペプチドについて、Fab領域含有タンパク質との親和定数(KA)が、1×105M-1以上であることがより好ましく、5×105M-1以上であることがよりさらに好ましい。しかし、親和定数は、Fab領域含有タンパク質の種類や、Fab領域結合性ペプチドのドメイン数によっても変わるので、これに限定されない。
本発明は、上記の本発明ペプチドを、免疫グロブリンやその断片、特にFab領域に親和性を有することを特徴とするアフィニティーリガンドとして利用することも、実施形態の1つとして包含する。同様に、当該リガンドを水不溶性担体に固定化したことを特徴とするアフィニティー分離マトリックスも、実施形態の1つとして包含する。ここで「アフィニティーリガンド」とは、抗原と抗体の結合に代表される特異的な分子間の親和力に基づいて、ある分子の集合から目的の分子を選択的に結合して捕集する物質や官能基を指す用語であり、本発明においては、免疫グロブリンに対して特異的に結合するペプチドを指す。本発明においては、単に「リガンド」と表記した場合も、「アフィニティーリガンド」と同義である。
本発明で用いる水不溶性担体としては、ガラスビーズ、シリカゲルなどの無機担体;有機担体;さらにはこれらの組み合わせによって得られる有機-有機、有機-無機などの複合担体などが挙げられる。有機担体としては、架橋ポリビニルアルコール、架橋ポリアクリレート、架橋ポリアクリルアミド、架橋ポリスチレンなどの合成高分子担体や、結晶性セルロース、架橋セルロース、架橋アガロース、架橋デキストランなどの多糖類担体を挙げることができる。市販品としては、多孔質セルロースゲルであるGCL2000、アリルデキストランとメチレンビスアクリルアミドを共有結合で架橋したSephacryl S-1000、アクリレート系の担体であるToyopearl、アガロース系の架橋担体であるSepharose CL4B、および、セルロース系の架橋担体であるCellufineなどを例示することができる。但し、本発明における水不溶性担体は、例示したこれらの担体のみに限定されるものではない。
また、本発明に用いる水不溶性担体は、本発明のアフィニティー分離マトリックスの使用目的および方法からみて表面積が大きいことが望ましく、適当な大きさの細孔を多数有する多孔質であることが好ましい。担体の形態としては、ビーズ状、モノリス状、繊維状、膜状(中空糸を含む)などいずれも可能であり、任意の形態を選ぶことができる。
リガンドの固定化方法については、例えば、リガンドに存在するアミノ基、カルボキシ基またはチオール基を利用した従来のカップリング法で担体に結合してよい。カップリング法としては、臭化シアン、エピクロロヒドリン、ジグリシジルエーテル、トシルクロライド、トレシルクロライド、ヒドラジンまたは過ヨウ素酸ナトリウムなどと担体とを反応させて担体を活性化するか、或いは担体表面に反応性官能基を導入し、リガンドとして固定化する化合物とカップリング反応を行い固定化する方法、また、担体とリガンドとして固定化する化合物が存在する系にカルボジイミドのような縮合試薬、または、グルタルアルデヒドのように分子中に複数の官能基を持つ試薬を加えて縮合、架橋することによる固定化方法が挙げられる。
また、リガンドと担体の間に複数の原子からなるスペーサー分子を導入してもよいし、担体にリガンドを直接固定化してもよい。従って、固定化のために、本発明に係る改変型Fab領域結合性ペプチドを化学修飾してもよいし、固定化に有用なアミノ酸残基を加えてもよい。固定化に有用なアミノ酸としては、側鎖に固定化の化学反応に有用な官能基を有しているアミノ酸が挙げられ、例えば、側鎖にアミノ基を含むLysや、側鎖にチオール基を含むCysが挙げられる。本発明の本質は、本発明においてペプチドに付与したFab領域結合性が、当該ペプチドをリガンドとして固定化したマトリックスにおいても同様に付与されることにあり、固定化のためにいかように修飾・改変しても、本発明の範囲に含まれる。
本発明のアフィニティー分離マトリックスを利用して、免疫グロブリンのFab領域を含むタンパク質をアフィニティーカラム・クロマトグラフィー精製法により分離精製することが可能となる。これらの免疫グロブリンGのFab領域を含むタンパク質の精製法は、免疫グロブリンのアフィニティーカラム・クロマトグラフィー精製法、例えばSpAアフィニティー分離マトリックスを利用した精製法に準じる手順により達成することができる(非特許文献1)。即ち、免疫グロブリンGのFab領域を含むタンパク質を含有する緩衝液を調製(pHは中性付近)した後、当該溶液を本発明のアフィニティー分離マトリックスを充填したアフィニティーカラムに通過させるなどして接触させ、免疫グロブリンGのFab領域を含むタンパク質を吸着させる。次いで、アフィニティーカラムに純粋な緩衝液を適量通過させ、カラム内部を洗浄する。この時点では所望の免疫グロブリンGのFab領域を含むタンパク質は、カラム内の本発明に係るアフィニティー分離マトリックスに吸着されている。そして、本発明ペプチドをリガンドとして固定化したアフィニティー分離マトリックスは、このサンプル添加の工程からマトリックス洗浄の工程において、目的とする免疫グロブリンGのFab領域を含むタンパク質を吸着保持する性能に優れる。次いで、適切なpHに調整した酸性緩衝液をカラムに通液し、所望の免疫グロブリンGのFab領域を含むタンパク質を溶出することにより、高純度な精製が達成される。溶出に用いる酸性緩衝液には、マトリックスからの解離を促進する物質を添加してもよい。
特に、本発明のアフィニティー分離マトリックスでは、より中性側に近いpHの酸性緩衝液を溶出液として用いることが可能である。用いる酸性緩衝液としては、pH値がpH3.0以上であることが好ましく、pH3.1以上であることがより好ましく、pH3.2以上であることがさらにより好ましく、pH3.3以上であることがさらにより好ましく、pH3.4以上であることがさらにより好ましく、pH3.5以上であることがさらにより好ましく、pH3.6以上であることがさらにより好ましく、pH3.7以上であることがさらにより好ましく、pH3.8以上であることがさらにより好ましい。用いる酸性緩衝液の上限は特に無いが、アフィニティークロマトグラフィーにおける中間洗浄でpH5.0程度で洗浄する場合や、精製後にウイルス除去のためにpH3.8程度でインキュベーションする場合においては、用いる酸性緩衝液はpH4.5以下であることが好ましく、pH4.0以下であることがより好ましい。
本発明のアフィニティー分離マトリックスは、リガンド化合物や担体の基材が完全に機能を損なわない程度の、適当な強酸性または強アルカリ性の純粋な緩衝液を通過させて洗浄することにより、再利用が可能である。上記の再生用緩衝液には、適当な変性剤や有機溶剤を配合してもよい。
本発明のアフィニティー分離マトリックスは、リガンド化合物や担体の基材が完全に機能を損なわない程度の、適当な強酸性または強アルカリ性の純粋な緩衝液を通過させて洗浄することにより、再利用が可能である。当該洗浄液としては、適当な変性剤や有機溶剤を含む溶液も使用できる。
本発明は、本発明に係る改変型Fab領域結合性ペプチドをコードするDNAにも関する。本発明ペプチドをコードするDNAは、その塩基配列を翻訳したアミノ酸配列が本発明ペプチドを構成するものであればいかなるものでもよい。そのような塩基配列は、通常用いられる公知の方法、例えば、ポリメラーゼ・チェーン・リアクション(以下、「PCR」と略記する)法を利用して取得できる。また、公知の化学合成法で合成することも可能であり、さらに、DNAライブラリーから得ることもできる。当該塩基配列は、コドンが縮重コドンで置換されていてもよく、翻訳されたときに同一のアミノ酸をコードしている限り、本来の塩基配列と同一である必要性は無い。当該塩基配列を一つ又はそれ以上有する組換えDNA、または、当該組換えDNAを含むプラスミドおよびファージなどのベクター、さらには、当該DNAを有するベクターにより形質転換された形質転換体、当該DNAを導入した遺伝子改変生物、および、当該DNAを転写の鋳型DNAとする無細胞タンパク質合成系を得ることができる。
また、本発明に係る改変型Fab領域結合性ペプチドは、タンパク質発現を補助する作用または精製を容易にするという利点がある公知のタンパク質との融合ペプチドとして取得することができる。即ち、本発明に係る改変型Fab領域結合性ペプチドを含む融合ペプチドをコードする組換えDNAを少なくとも一つ含有する微生物または細胞を得ることができる。上記タンパク質の例としては、マルトース結合タンパク質(MBP)、グルタチオン-S-トランスフェラーゼ(GST)等が挙げられるが、それらのタンパク質に限定されるものではない。
本発明のペプチドをコードするDNAを改変するための部位特異的な変異の導入は、以下のように、組換えDNA技術、PCR法などを用いて行うことができる。即ち、組換えDNA技術による変異の導入は、例えば、本発明ペプチドをコードする遺伝子中において、変異導入を希望する目的の部位の両側に適当な制限酵素認識配列が存在する場合に、それら制限酵素認識配列部分を前記制限酵素で切断し、変異導入を希望する部位を含む領域を除去した後、化学合成などによって目的の部位のみに変異導入したDNA断片を挿入するカセット変異法によって行うことができる。
また、PCRによる部位特異的変異の導入は、例えば、本発明ペプチドをコードする二本鎖プラスミドを鋳型として、+鎖および-鎖に相補的な変異を含む2種の合成オリゴプライマーを用いてPCRを行うダブルプライマー法により行うことができる。また、本発明の単量体ペプチド(1つのドメイン)をコードするDNAを、意図する数だけ直列に連結することにより、多量体ペプチドをコードするDNAを作製することもできる。例えば、多量体ペプチドをコードするDNAの連結方法に関して、DNA配列に適当な制限酵素部位を導入し、制限酵素で断片化した2本鎖DNAをDNAリガーゼで連結することができる。制限酵素部位は1種類でもよいが、複数の異なる種類の制限酵素部位を導入することもできる。また、多量体ペプチドをコードするDNAにおいて、各々の単量体ペプチドをコードする塩基配列が同一の場合には、宿主にて相同組み換えを誘発する可能性があるので、連結されている単量体ペプチドをコードするDNAの塩基配列間の配列同一性が90%以下、好ましくは85%以下、より好ましくは80%以下、さらにより好ましくは75%以下であることが好ましい。なお、塩基配列の同一性も、アミノ酸配列と同様に、常法により決定することが可能である。
本発明の「発現ベクター」は、前述した本発明ペプチドまたはその部分アミノ酸配列をコードする塩基配列、およびその塩基配列に作動可能に連結された宿主で機能しうるプロモーターを含む。通常は、本発明ペプチドをコードする遺伝子を、適当なベクターに連結もしくは挿入することにより得ることができ、遺伝子を挿入するためのベクターは、宿主中で自律複製可能なものであれば特に限定されず、プラスミドDNAやファージDNAをベクターとして用いることができる。例えば、大腸菌を宿主として用いる場合には、pQE系ベクター(キアゲン社)、pET系ベクター(メルク社)およびpGEX系ベクター(GEヘルスケアバイオサイエンス社)のベクターなどが挙げられる。
本発明の形質転換体は、宿主となる細胞へ本発明の組換えベクターを導入することにより得ることができる。宿主への組換え体DNAの導入方法としては、例えばカルシウムイオンを用いる方法、エレクトロポレーション法、スフェロプラスト法、酢酸リチウム法、アグロバクテリウム感染法、パーティクルガン法およびポリエチレングリコール法などが挙げられるが、これらに限定されるものではない。また、得られた遺伝子の機能を宿主で発現する方法としては、本発明で得られた遺伝子をゲノム(染色体)に組み込む方法なども挙げられる。宿主となる細胞については特に限定されるものではないが、安価に大量生産する上では、大腸菌、枯草菌、ブレビバチルス属、スタフィロコッカス属、ストレプトコッカス属、ストレプトマイセス属(Streptomyces)、コリネバクテリウム属(Corynebacterium)等のバクテリア(真正細菌)を好適に使用しうる。
本発明に係る改変型Fab領域結合性ペプチドは、前記した形質転換体を培地で培養し、培養体中(菌体ぺリプラズム領域中も含む)または培養液中(細胞外)に本発明のペプチドを生成蓄積させ、該培養物から所望のペプチドを採取することにより製造することができる。また、本発明ペプチドは、前記した形質転換細胞を培地で培養し、培養体中(菌体ぺリプラズム領域中も含む)または培養液中(細胞外)に、本発明ペプチドを含む融合タンパク質を生成蓄積させ、当該培養物から当該融合ペプチドを採取し、当該融合ペプチドを適切なプロテアーゼによって切断し、所望のペプチドを採取することにより製造することができる。
本発明の形質転換体を培地で培養する方法は、宿主の培養に用いられる通常の方法に従って行われる。得られた形質転換体の培養に用いる培地は、本発明ペプチドを高効率、高収量で生産できるものであれば特に制限は無い。具体的には、グルコース、蔗糖、グリセロール、ポリペプトン、肉エキス、酵母エキス、カザミノ酸などの炭素源や窒素源を使用することができる。その他、カリウム塩、ナトリウム塩、リン酸塩、マグネシウム塩、マンガン塩、亜鉛塩、鉄塩などの無機塩類が必要に応じて添加される。栄養要求性の宿主細胞を用いる場合は、生育に要求される栄養物質を添加すればよい。また、必要であればペニシリン、エリスロマイシン、クロラムフェニコール、ネオマイシンなどの抗生物質が添加されてもよい。
さらに、形質転換体内外に存在する宿主由来のプロテアーゼによる当該目的ペプチドの分解を抑えるために、公知の各種プロテアーゼ阻害剤、即ち、Phenylmethane sulfonyl fluoride(PMSF)、Benzamidine、4-(2-aminoethyl)-benzenesulfonyl fluoride(AEBSF)、Antipain、Chymostatin、Leupeptin、Pepstatin A、Phosphoramidon、Aprotinin、Ethylenediaminetetra acetic acid(EDTA)および/またはその他市販されているプロテアーゼ阻害剤を適当な濃度で添加してもよい。
さらに、本発明に係る改変型Fab領域結合性ペプチドを正しくフォールディングさせるために、例えば、GroEL/ES、Hsp70/DnaK、Hsp90、Hsp104/ClpBなどの分子シャペロンを利用してもよい。かかる分子シャペロンは、例えば、共発現または融合タンパク質化などの手法で、本発明のペプチドと共存させる。なお、本発明ペプチドの正しいフォールディングを目的とする場合には、正しいフォールディングを助長する添加剤を培地中に加える手法や低温にて培養するなどの手法もあるが、これらに限定されるものではない。
大腸菌を宿主として得られた形質転換細胞を培養する培地としては、LB培地(トリプトン1%,酵母エキス0.5%,NaCl1%)、または、2×YT培地(トリプトン1.6%,酵母エキス1.0%,NaCl0.5%)等が挙げられる。また、培養温度は、例えば15~42℃、好ましくは20~37℃で、通気攪拌条件で好気的に数時間~数日培養することにより、本発明ペプチドを培養細胞内(ぺリプラズム領域内を含む)または培養溶液(細胞外)に蓄積させて回収する。場合によっては、通気を遮断し嫌気的に培養してもよい。組換えペプチドが分泌生産される場合には、培養終了後に、遠心分離、ろ過などの一般的な分離方法で、培養細胞と分泌生産されたペプチドを含む上清を分離することにより生産された組換えペプチドを回収することができる。また、培養細胞内(ぺリプラズム領域内を含む)に蓄積される場合にも、例えば、培養液から遠心分離、ろ過などの方法により菌体を採取し、次いで、この菌体を超音波破砕法、フレンチプレス法などにより破砕し、および/または、界面活性剤等を添加して可溶化することにより、細胞内に蓄積生産されたペプチドを回収することができる。
本発明に係るペプチドの精製はアフィニティークロマトグラフィー、陽イオンまたは陰イオン交換クロマトグラフィー、ゲル濾過クロマトグラフィー等を単独でまたは適宜組み合わせることによって行うことができる。得られた精製物質が目的のタンパク質であることの確認は、通常の方法、例えばSDSポリアクリルアミドゲル電気泳動、N末端アミノ酸配列分析、ウエスタンブロッティング等により行うことができる。
本願は、2015年2月26日に出願された日本国特許出願第2015-36424号に基づく優先権の利益を主張するものである。2015年2月26日に出願された日本国特許出願第2015-36424号の明細書の全内容が、本願に参考のため援用される。
以下、実施例に基づいて本発明をより詳細に説明するが、本発明はこれらの実施例に限定されるものではない。
以下の実施例で取得した変異ペプチドは「ドメイン-導入した変異」の形で表記し、置換変異導入前のペプチドは、「ドメイン-Cont」の形で表記する。例えば、配列番号1で示されるFab結合力が向上したSpG-β1変異体(SpG-β1#)は「GB1#-Cont」、配列番号1のアミノ酸配列に対し第17位のThrをAlaに置換した変異(T17A)を導入したSpG-β1変異体は「GB1#-T17A」と表記する。2種類の変異を同時に導入した変異体の表記については、スラッシュを用いて併記する。例えば、変異T17AおよびD36Vを導入したSpG-β1#変異体は、「GB1#-T17A/D36V」と表記する。
また、単ドメインを複数連結したペプチドについては、ピリオドに続けて連結した数に「d」をつけて併記する。例えば、変異T17AおよびD36Vを導入したSpG-β1#変異体を2連結したペプチドは、「GB1#-T17A/D36V.2d」と表記する。さらに、例えば、水不溶性基材にペプチドを固定化するために、C末端に固定化用官能基を有するCys残基(C)を導入した場合、「d」の後ろに導入したアミノ酸の1文字表記を付与する。例えば、変異T17AおよびD36Vを導入したSpG-β1#変異体を2連結してC末端にCysを付与したSpG-β1#変異体は、「GB1#-T17A/D36V.2dC」と表記する。
実施例1:各種SpG-β1#変異体の調製
(1) 各種SpG-β1#変異体の発現プラスミド調製
各種SpG-β1#変異体発現プラスミドの調製は、GB1#-Cont.1d(比較例となる置換変異導入前のペプチド)の発現プラスミドを鋳型プラスミドとして、それに変異を導入することによって調製した。
(1) 各種SpG-β1#変異体の発現プラスミド調製
各種SpG-β1#変異体発現プラスミドの調製は、GB1#-Cont.1d(比較例となる置換変異導入前のペプチド)の発現プラスミドを鋳型プラスミドとして、それに変異を導入することによって調製した。
最初に、GB1#-Cont.1d(配列番号1)のアミノ酸配列から逆翻訳を行い、当該ペプチドをコードする塩基配列(配列番号6)を設計した。次に、図1に示す方法によって、コードDNAの調製および発現用プラスミドへの挿入を行った。GB1#-T17A.1dをコードするDNAは、同じ制限酵素サイトを有する2種の二本鎖DNA(f1とf2)を連結する形で調製し、発現ベクターのマルチクローニングサイトに組み込んだ。実際には、2種の二本鎖DNAと発現ベクターの計3種の二本鎖DNAを連結する3断片ライゲーションによって、コードDNA調製とベクター組込みを同時に実施した。2種の二本鎖DNAの調製方法は、互いに30塩基程度の相補領域を含む2種の一本鎖オリゴDNA(f1-1/f1-2、または、f2-1/f2-2)を、オーバーラップPCRによって伸長し、目的の二本鎖DNAを調製した。具体的な実験操作については、次の通りとなる。一本鎖オリゴDNAf1-1(配列番号7)/f1-2(配列番号8)を外注によって合成し(シグマジェノシス社)、ポリメラーゼとしてBlend Taq(TOYOBO社)を用い、オーバーラップPCR反応を行った。PCR反応生成物をアガロース電気泳動にかけ、目的のバンドを切り出すことで抽出した二本鎖DNAを、制限酵素BamHIとEco52I(いずれもタカラバイオ社)により切断した。同様に、一本鎖オリゴDNAf2-1(配列番号9)/f2-2(配列番号10)を外注によって合成し、オーバーラップPCR反応を経て、合成・抽出した二本鎖DNAを、制限酵素Eco52IとEcoRI(いずれもタカラバイオ社)により切断した。次に、プラスミドベクターpGEX-6P-1(GEヘルスケア・バイオサイエンス社)のマルチクローニングサイト中のBamHI/EcoRIサイトに上記2種の二本鎖DNAをサブクローニングした。サブクローニングにおけるライゲーション反応は、Ligation high(TOYOBO社)を用いて、製品に添付のプロトコルに準ずる形で実施した。
上記プラスミドベクターpGEX-6P-1を用いて、コンピテント細胞(タカラバイオ社「大腸菌HB101」)の形質転換を、本コンピテント細胞製品に付属のプロトコルに従って行った。上記プラスミドベクターpGEX-6P-1を用いれば、グルタチオン-S-トランスフェラーゼ(以下、「GST」と略記する)が融合したGB1#-Cont.1dを産生することができる。次いで、プラスミド精製キット(プロメガ社製「Wizard Plus SV Minipreps DNA Purification System」)を用い、キット付属の標準プロトコルに従って、プラスミドDNAを増幅し、抽出した。発現プラスミドのコードDNAの塩基配列確認は、DNAシークエンサー(Applied Biosystems社製「3130xl Genetic Analyzer」)を用いて行った。遺伝子解析キット(Applied Biosystems社製「BigDye Terminator v.1.1 Cycle Sequencing Kit」)と、プラスミドベクターpGEX-6P-1のシークエンシング用DNAプライマー(GEヘルスケア・バイオサイエンス社)を用いて、添付のプロトコルに従いシークエンシングPCR反応を行った。そのシークエンシング産物を、プラスミド精製キットApplied Biosystems社製「BigDye XTerminator Purification Kit」)を用いて、添付のプロトコルに従い精製し、塩基配列解析に用いた。
各種SpG-β1#変異体をコードするDNAに関しては、第17位に変異を導入する場合と第36位に変異を導入する場合とにおいて、方法を変えて調製した。第17位に変異を導入する場合には、上記GB1#-Cont.1d発現プラスミドを鋳型とし、配列番号11~12のDNAプライマーとpGEX-6P-1シークエンシング用DNAプライマー(3’側)を用いたPCR反応にて、制限酵素を用いてpGEX-6Pに挿入可能な二本鎖DNAを調製した。このPCR反応は、Blend Taq -Plus-(TOYOBO社)を用いて、添付のプロトコルに従い行った。この二本鎖DNAを制限酵素BamHIとEcoRIにて切断し、上述と同様の手法にて、pGEX-6P-1のBamHI/EcoRIサイトでライゲーションすることで、第17位に変異が導入された各種SpG-β1#変異体の発現プラスミドを調製した。第36位に変異を導入する場合には、配列番号13のDNAプライマーとpGEX-6P-1シークエンシング用DNAプライマー(3’側)を用いた同様のPCR反応にて、図1のf2に相当する二本鎖DNAを調製した。この二本鎖DNAおよびGB1#-Cont.1d発現プラスミドを制限酵素Eco52IとEcoRIで切断し、上述と同様の手法にて、ライゲーションすることで、第36位に変異が導入されたSpG-β1#変異体の発現プラスミドを調製した。形質転換、塩基配列解析に関しては、上記と同様の手法にて実施した。各々の変異体を作るときに使用したDNAプライマーの塩基配列、変異体をコードするcDNAの塩基配列、および、変異体のアミノ酸配列の配列番号を表1に示す。
(2) 各種SpG-β1#変異体の調製
上記(1)で得られた、各種SpG-β1#変異体遺伝子を導入した各形質転換細胞を、アンピシリン含有2×YT培地にて、37℃で終夜培養した。これらの培養液を、100倍量程度のアンピシリン含有2×YT培地に接種し、37℃で約1時間、その後25℃で約1時間培養した後で、終濃度0.1mMになるようイソプロピル1-チオ-β-D-ガラクシド(IPTG)を添加し、さらに25℃にて約18時間培養した。
上記(1)で得られた、各種SpG-β1#変異体遺伝子を導入した各形質転換細胞を、アンピシリン含有2×YT培地にて、37℃で終夜培養した。これらの培養液を、100倍量程度のアンピシリン含有2×YT培地に接種し、37℃で約1時間、その後25℃で約1時間培養した後で、終濃度0.1mMになるようイソプロピル1-チオ-β-D-ガラクシド(IPTG)を添加し、さらに25℃にて約18時間培養した。
培養終了後、遠心にて集菌し、PBS緩衝液5mLに再懸濁した。超音波破砕にて細胞を破砕し、遠心分離して上清画分(無細胞抽出液)と不溶性画分に分画した。pGEX-6P-1ベクターのマルチクローニングサイトに目的の遺伝子を導入すると、GSTがN末端に付与した融合ペプチドとして発現される。それぞれの画分をSDS電気泳動により分析したところ、各々の形質転換細胞培養液から調製した各種無細胞抽出液のすべてについて、分子量約25,000以上の位置にIPTGにより誘導されたと考えられるペプチドのバンドを確認した。なお、分子量はほぼ同様であるが、変異体の種類によってバンドの位置は違った。
GST融合ペプチドを含む各々の無細胞抽出液から、GSTに対して親和性のあるGSTrap FFカラム(GEヘルスケア・バイオサイエンス社)を用いたアフィニティークロマトグラフィーにて、GST融合ペプチドを粗精製した。各々の無細胞抽出液をGSTrap FFカラムに添加し、標準緩衝液(20mM NaH2PO4-Na2HPO4,150mM NaCl,pH7.4)にてカラムを洗浄し、続いて溶出用緩衝液(50mM Tris-HCl,20mMグルタチオン,pH8.0)にて目的のGST融合ペプチドを溶出した。後の実施例で、GSTを融合したままでアッセイに利用したサンプルは、この溶出液を遠心式フィルターユニットであるアミコン(メルクミリポア社)を用いて、濃縮した形で標準緩衝液に置換したペプチド溶液を用いた。
pGEX-6P-1ベクターのマルチクローニングサイトに遺伝子を導入すると、配列特異的プロテアーゼPreScission Protease(GEヘルスケア・バイオサイエンス社)でGSTを切断することが可能なアミノ酸配列が、GSTと目的ペプチドの間に導入される。PreScission Proteaseを用いて、添付プロトコルに従いGST切断反応を行った。このようにGSTを切断した形でアッセイに利用したサンプルから、Superdex 75 10/300 GLカラム(GEヘルスケア・バイオサイエンス社)を用いたゲルろ過クロマトグラフィーにて、目的のペプチドを精製した。標準緩衝液にて平衡化したSuperdex 75 10/300 GLカラムに、各々の反応溶液を添加し、目的のペプチドを、切断したGSTやPreScission Proteaseから分離精製した。なお、以上のカラムを用いたクロマトグラフィーによるペプチド精製は、全てAKTAprime plusシステム(GEヘルスケア・バイオサイエンス社)を利用して実施した。また、本実施例で得られるGST切断後の各々のペプチドは、N末端側にベクターpGEX-6P-1由来のGly-Pro-Leu-Gly-SerがN末端側に付加されたアミノ酸配列を有する。
実施例2:各種SpG-β1#変異体のaTNF-Fabへの親和性評価
(1) Fabフラグメントの調製
ヒト化モノクローナルIgG製剤を原料として、これをパパインによって、FabフラグメントとFcフラグメントに断片化し、Fabフラグメントのみを分離精製した。具体的には、抗ヒトTNFαモノクローナルIgG(一般名「インフリキシマブ」,製品名「レミケード」,田辺三菱製薬)製剤を、パパイン消化用緩衝液(0.1M AcOH-AcONa,2mM EDTA,1mMシステイン,pH5.5)に溶解し、Papain Agarose from papaya latexパパイン固定化アガロース(SIGMA社)を添加し、ローテーターで混和させながら、37℃で約8時間インキュベートした。パパイン固定化アガロースから分離した反応溶液(FabフラグメントとFcフラグメントが混在)から、MabSelect SuReカラム(GEヘルスケアバイオサイエンス社)を利用したアフィニティークロマトグラフィーにより、素通り画分でIgG-Fabを回収することで分離精製した。分取したIgG-Fab溶液を、Superdex 75 10/300 GLカラム(平衡化および分離には標準緩衝液を使用)を用いたゲルろ過クロマトグラフィーにて精製し、IgG-Fab溶液(aTNF-Fab)を得た。なお、実施例1と同様に、クロマトグラフィーによるペプチド精製は、AKTAprime plusシステムを利用して実施した。
(1) Fabフラグメントの調製
ヒト化モノクローナルIgG製剤を原料として、これをパパインによって、FabフラグメントとFcフラグメントに断片化し、Fabフラグメントのみを分離精製した。具体的には、抗ヒトTNFαモノクローナルIgG(一般名「インフリキシマブ」,製品名「レミケード」,田辺三菱製薬)製剤を、パパイン消化用緩衝液(0.1M AcOH-AcONa,2mM EDTA,1mMシステイン,pH5.5)に溶解し、Papain Agarose from papaya latexパパイン固定化アガロース(SIGMA社)を添加し、ローテーターで混和させながら、37℃で約8時間インキュベートした。パパイン固定化アガロースから分離した反応溶液(FabフラグメントとFcフラグメントが混在)から、MabSelect SuReカラム(GEヘルスケアバイオサイエンス社)を利用したアフィニティークロマトグラフィーにより、素通り画分でIgG-Fabを回収することで分離精製した。分取したIgG-Fab溶液を、Superdex 75 10/300 GLカラム(平衡化および分離には標準緩衝液を使用)を用いたゲルろ過クロマトグラフィーにて精製し、IgG-Fab溶液(aTNF-Fab)を得た。なお、実施例1と同様に、クロマトグラフィーによるペプチド精製は、AKTAprime plusシステムを利用して実施した。
(2) 各種SpG-β1#変異体のFabに対する親和性の解析
表面プラズモン共鳴を利用したバイオセンサーBiacore3000(GEヘルスケア・バイオサイエンス社)を用いて、実施例1(2)で取得した各種SpG-β1#変異体の、aTNF-Fabとの親和性を解析した。本実施例では、実施例2(1)で取得したaTNF-Fabをセンサーチップに固定化し、各種ペプチドをチップ上に流して、両者の相互作用を検出した。aTNF-FabのセンサーチップCM5への固定化は、N-ヒドロキシスクシンイミド(NHS)、および、N-エチル-N’-(3-ジメチルアミノプロピル)カルボジイミド塩酸塩(EDC)を用いたアミンカップリング法にて行い、ブロッキングにはエタノールアミンを用いた(センサーチップや固定化用試薬は、全てGEヘルスケアバイオサイエンス社製)。aTNF-Fab溶液は、固定化用緩衝液(10mM AcOH-AcONa,pH4.5)を用いて10倍程度に希釈し、Biacore 3000付属のプロトコルに従い、センサーチップへ固定した。また、チップ上の別のフローセルに対して、EDC/NHSにより活性化した後、何も固定化反応させずに、エタノールアミンでブロックした、ネガティブ・コントロールとなるリファレンスセルも用意した。各種SpG-β1#変異体は、ランニング緩衝液(20mM NaH2PO4-Na2HPO4,150mM NaCl,0.005% P-20,pH7.4)を用いて、0.01、0.1、1または10μMに濃度を調整したタンパク質溶液を流速40μL/minで1分間センサーチップに添加した。測定温度25℃にて、添加時(結合相,1分)、および添加終了後(解離相,1分)の結合反応曲線を順次観測した。各々の観測終了後に、約20mM NaOHを添加して洗浄した。得られた結合反応曲線(リファレンスセルの結合反応曲線を差し引いた結合反応曲線)に対して、システム付属ソフトBIA evaluationを用いた1:1の結合モデルによるフィッティング解析を行い、aTNF-Fabに対する親和定数(KA=kon/koff)を算出した。比較例2で測定したGB1#-Cont.1dの各種結合パラメータと共に、解析結果を表2に示す。

表2に示す結果のとおり、T17A、T17VおよびD36Vの変異を導入した変異体は、変異導入前のSpG-β1#(GB1#-Cont.1d)のaTNF-Fabに対する親和定数KAと比較して低いKA値を示した。しかし、その数値は5×105(1/M)以上であり、1/10以下の大幅な結合力低下は見られなかった。Fab結合力は、リガンドとして相手分子をカラムに保持できればよく、結合力が強過ぎると溶出には不利に働く可能性もあるので、5×105(1/M)以上であれば、リガンドとして問題なく機能すると考えられる。
実施例3: 各種SpG-β1#変異体のaTNF-Fabの酸解離pHの評価
(1)Fabフラグメント固定化担体の調製
カップリング目的官能基をアミノ基とする市販のリガンド固定化用カップリングカラムを利用して、実施例2で得られたaTNF-Fabを固定化したアフィニティー分離マトリックスを作製した。
(1)Fabフラグメント固定化担体の調製
カップリング目的官能基をアミノ基とする市販のリガンド固定化用カップリングカラムを利用して、実施例2で得られたaTNF-Fabを固定化したアフィニティー分離マトリックスを作製した。
水不溶性基材として、市販のプレパックカラム(GEヘルスケア・バイオサイエンス社製「Hitrap NHS activated HP」)1mLを使用した。このカラムは、架橋アガロースをベースとし、カップリング目的官能基をアミノ基とするペプチド性リガンド固定化用の活性基が導入済みなので、製品マニュアルに従ってリガンドを固定化した。氷浴で冷やした1mM HClを、流速1mL/minで2mL分流す操作を3回行い、カラム中のイソプロパノールを除去した。
その後すぐに、カップリング緩衝液(0.2M NaHCO3,0.5M NaCl,pH8.3)でaTNF-Fabを1mg/mLに希釈した溶液を同じ流速で1mL添加し、カラムの上下に栓をして25℃で30分間静置することで、取得したリガンドをカラムに固定化した。
その後開栓し、カップリング緩衝液を同じ流速で3mL流して、未反応aTNF-Fabを回収した。その後、ブロッキング用緩衝液(0.5Mエタノールアミン,0.5M NaCl,pH8.3)を2mL流す操作を3回実施し、洗浄用緩衝液(0.1M酢酸,0.5M NaCl,pH4.0)を2mL流す操作を3回実施した。ブロッキング用緩衝液と洗浄用緩衝液を流す一連の操作は交互に3回ずつ行った。最後に標準緩衝液(20mM NaH2PO4-Na2HPO4,150mM NaCl,pH7.4)を2mL流してアフィニティー分離カラムの作製を完了した。
(2) Fabフラグメント固定化担体を用いたクロマトグラフィー実験
実施例1(2)で調製した各種SpG-β1#変異体を、実施例3(1)で調製したaTNF-Fab固定化カラムを利用したアフィニティー精製クロマトグラフィーにより、各種SpG-β1#変異体の溶出ピークトップにおけるpH(酸解離pH)を解析した。具体的には、クロマトシステムとしては、AKTAprime plusシステムを利用し、イオン交換用緩衝液A(50mM クエン酸-クエン酸Na,pH5.5)にて平衡化したカラムに、約0.1mg/mLに調整した各種SpG-β1#変異体のタンパク質溶液を0.1mL添加し、pH5.5からpH2.5へのpH直線勾配にて溶出させた。より具体的には、イオン交換緩衝液Aとイオン交換緩衝液B(50mM AcOH-AcONa,1M NaCl,pH2.5)を利用しており、カラムに20カラムボリューム分の緩衝液を通液する際に、緩衝液Bの濃度を0%から100%に直線的に上げていく工程にて、溶出位置から酸解離pHを評価した。各々の溶出位置(ピークトップ)におけるpH値を、比較例3の結果も併せて表3に示す。
実施例1(2)で調製した各種SpG-β1#変異体を、実施例3(1)で調製したaTNF-Fab固定化カラムを利用したアフィニティー精製クロマトグラフィーにより、各種SpG-β1#変異体の溶出ピークトップにおけるpH(酸解離pH)を解析した。具体的には、クロマトシステムとしては、AKTAprime plusシステムを利用し、イオン交換用緩衝液A(50mM クエン酸-クエン酸Na,pH5.5)にて平衡化したカラムに、約0.1mg/mLに調整した各種SpG-β1#変異体のタンパク質溶液を0.1mL添加し、pH5.5からpH2.5へのpH直線勾配にて溶出させた。より具体的には、イオン交換緩衝液Aとイオン交換緩衝液B(50mM AcOH-AcONa,1M NaCl,pH2.5)を利用しており、カラムに20カラムボリューム分の緩衝液を通液する際に、緩衝液Bの濃度を0%から100%に直線的に上げていく工程にて、溶出位置から酸解離pHを評価した。各々の溶出位置(ピークトップ)におけるpH値を、比較例3の結果も併せて表3に示す。
また、各種SpG-β1#変異体に関し、比較例3の変異導入前のSpG-β1#との比較もできる形で重ね合わせたクロマトグラフィーのチャートを図2に示す。
表3と図2に示した結果のとおり、本発明で得られた各種SpG-β1#変異体(GB1#-T17A.1d,GB1#-T17V.1d,およびGB1#-D36V.1d)が、変異を導入していないGB1#-Cont.1dよりも早く溶出した。これは、より中性側に近いpHで各種SpG-β1#変異体が溶出されることを示している。即ち、本発明のペプチドは、Fab領域含有タンパク質と解離するpHがより中性側に改変されたことが示されたといえる。また、この実験系においては、pH5.5ではFabとの結合が担体に保持される程度に強いことを示唆する結果であり、本発明で得られたペプチドをリガンドとするアフィニティー分離マトリックスでは、この程度のpHで中間洗浄することが可能であると考えられる。
比較例1: GB1#-Cont.1d(SpG-β1#)の調製
実施例1にて調製したGB1#-Cont.1dの発現プラスミドを用いて、実施例1と同様の手法にて形質転換細胞を調製し、培養と精製を経て、GB1#-Cont.1dの溶液を調製した。
実施例1にて調製したGB1#-Cont.1dの発現プラスミドを用いて、実施例1と同様の手法にて形質転換細胞を調製し、培養と精製を経て、GB1#-Cont.1dの溶液を調製した。
比較例2: GB1#-Cont.1dのaTNF-Fabへの親和性評価
比較例1で調製したGB1#-Cont.1dに関し、実施例2(1)で調製したaTNF-Fabとの親和性を、実施例2(2)と同様の手法にて解析した。その解析結果を、上記の表2に併せて示した。
比較例1で調製したGB1#-Cont.1dに関し、実施例2(1)で調製したaTNF-Fabとの親和性を、実施例2(2)と同様の手法にて解析した。その解析結果を、上記の表2に併せて示した。
比較例3: GB1#-Cont.1dのaTNF-Fabの酸解離pH評価
比較例1で調製したGB1#-Cont.1dに関し、実施例3(1)で調製したaTNF-Fab固定化担体を用いて、実施例3(2)と同様の手法にて酸解離pHを解析した。その解析結果を、上記の表3に併せて示した。
比較例1で調製したGB1#-Cont.1dに関し、実施例3(1)で調製したaTNF-Fab固定化担体を用いて、実施例3(2)と同様の手法にて酸解離pHを解析した。その解析結果を、上記の表3に併せて示した。
Claims (12)
- 下記(1)~(3)の何れかの改変型Fab領域結合性ペプチド。
(1) 配列番号1~5のいずれかのアミノ酸配列において、第17位および第36位からなる群より選択される1つ以上のアミノ酸残基が置換されているアミノ酸配列を有し、且つ、その酸解離pHが置換導入前に比べて中性側にシフトしているFab領域結合性ペプチド;
(2) 上記(1)に規定されるアミノ酸配列において、上記第17位および第36位を除く領域中で1個または数個のアミノ酸がさらに欠失、置換および/または付加されたアミノ酸配列を有するFab領域結合性ペプチドであり、且つ、その酸解離pHが変異導入前に比べて中性側にシフトしているFab領域結合性ペプチド;
(3) 上記(1)に規定されるアミノ酸配列に対して80%以上の配列同一性を有するアミノ酸配列を有し、且つ、その酸解離pHが変異導入前に比べて中性側にシフトしているFab領域結合性ペプチド(但し、上記(1)に規定されるアミノ酸配列における第17位および第36位からなる群より選択される1以上の置換アミノ酸残基は、(3)においてさらに変異しないものとする)。 - 上記(1)に規定されるアミノ酸配列において、第17位のアミノ酸残基が置換されている請求項1に記載の改変型Fab領域結合性ペプチド。
- 上記(1)に規定されるアミノ酸配列において、第17位がAla、Val、Ser、LeuまたはIleに置換されている請求項1または2に記載の改変型Fab領域結合性ペプチド。
- 上記(1)に規定されるアミノ酸配列において、第36位がAla、Val、ThrまたはGluに置換されている請求項1~3のいずれかに記載の改変型Fab領域結合性ペプチド。
- 上記(2)に規定されるアミノ酸配列において、上記変異の部位が、第6位、第7位、第10位、第13位、第15位、第18位、第19位、第21位、第24位、第28位、第29位、第30位、第31位、第33位、第35位、第37位、第39位、第40位、第42位および第47位からなる群より選択される1以上の部位である請求項1~4のいずれかに記載の改変型Fab領域結合性ペプチド。
- 上記(2)に規定されるアミノ酸配列において、上記変異の部位がN末端および/またはC末端である請求項1~5のいずれかに記載の改変型Fab領域結合性ペプチド。
- 請求項1~6のいずれかに記載の改変型Fab領域結合性ペプチドを2個以上連結した複数ドメインを有することを特徴とする改変型Fab領域結合性ペプチド多量体。
- 請求項1~6のいずれかに記載の改変型Fab領域結合性ペプチドまたは請求項7に記載の改変型Fab領域結合性ペプチド多量体がリガンドとして水不溶性担体に固定化されたものであることを特徴とするアフィニティー分離マトリックス。
- Fab領域を含むタンパク質を製造する方法であって、
請求項8に記載のアフィニティー分離マトリックスと、Fab領域を含むタンパク質を含む液体試料とを接触させる工程と
アフィニティー分離マトリックスに結合したFab領域を含むタンパク質を、アフィニティー分離マトリックスから分離する工程を含むことを特徴とする方法。 - 請求項1~6のいずれかに記載の改変型Fab領域結合性ペプチドをコードすることを特徴とするDNA。
- 請求項10に記載のDNAを含むことを特徴とするベクター。
- 請求項11に記載のベクターにより形質転換されたものであることを特徴とする形質転換体。
