- the control unit performs the MS / MS analysis.
- the operation of the first mass separation unit and the second mass separation unit is controlled to satisfy the relationship of Prod .
- the valence determination unit can determine the valence based on the interval between the peaks corresponding to the isotope ions. Specifically, for example, if the interval between isotope ion peaks of a certain precursor ion on the mass spectrum is about 1 Da, the valence is 1, and if the interval is about 0.5 Da, the valence is 2, and the interval is about The valence can be determined by obtaining the reciprocal of the interval between isotope peaks, such as 3 for 0.33 Da, such as 3.
- the selection criterion by the third input unit selects one of the valences determined by the valence determination unit when there are a plurality of types.
- the selection criterion by the third input unit is to select a plurality of types of valences, and the precursor ion valence determining unit determines the valence of the precursor ions.
- the control unit performs the MS / MS analysis while sequentially changing the valence of the precursor ions, so that the first mass separation unit and the second mass separation unit It can be set as the structure which controls operation
- the neutral loss is the same for all multivalent ions derived from the same compound. All of a plurality of product ions that are can be obtained.
- the first to third input units may input mass and valence numerically, but not the numerical input but the precursor. It is assumed that the composition formula and valence of the fragment desorbed from the ion, or the ionic formula is input, and further includes a calculation unit that calculates the mass and valence of the fragment based on the information input from the input unit. Also good.
- the first to third input units select a name of a fragment to be desorbed from the precursor ion from a plurality of names registered in advance, and are associated with the name selected from the input unit. It is good also as a structure further provided with the acquisition part which acquires the mass and valence of a piece. With such a configuration, the operability is improved as compared with the case of numerical input.
- ions generated from the target component to be analyzed are multivalent. Even if it is an ion, the mass-to-charge ratio of the precursor ion and the mass-to-charge of the product ion so that the mass of the fragment desorbed from the precursor ion becomes a certain constant value, as in the general neutral loss scan method. It is possible to scan in conjunction with the ratio. Thereby, it becomes easy to search for a substance having a specific chemical structure.

- a pre-quadrupole mass filter 13 consisting of two rod electrodes and selectively passing ions having a specific mass-to-charge ratio
- a quadrupole ion guide 15 consisting of four rod electrodes inside are cleaved.
- the former quadrupole mass filter 13 Similar to the collision cell 14, the former quadrupole mass filter 13, the latter quadrupole mass filter 16 that selectively consists of four rod electrodes and has a specific mass-to-charge ratio, and detects ions.
- a detector 17 that outputs a detection signal corresponding to the amount of ions is disposed.
- a voltage obtained by combining a DC voltage and a high-frequency voltage is applied from the Q1 power supply unit 25 to the first-stage quadrupole mass filter 13, and a DC voltage and a high-frequency voltage are combined from the Q3 power supply unit 26 to the second-stage quadrupole mass filter 16. Applied voltage.
- an appropriate voltage is applied to other parts such as the quadrupole ion guide 15, but the description is omitted because it is not directly related to the present invention.
- the detection signal (ion intensity signal) output from the detector 17 is input to the data processing unit 18 and converted into digital data, and then data processing such as mass spectrum creation is executed.
- An input unit 20 and a display unit 21 operated by an analyst (user) are connected to the control unit 19 that controls the entire apparatus.
- the quadrupole drive control unit 22 includes, as functional blocks, a valence calculation unit 221, a precursor ion m / z setting unit 222, a passing product ion m / z calculation unit 223, and a quadrupole drive voltage calculation unit 224.
- the Q1 power supply unit 25 and the Q3 power supply unit 26 are controlled.
- the input unit 20 is a general input unit such as a keyboard, but includes a mass input unit 201 and a valence input unit 202 as functional blocks.
- At least a part of the data processing unit 18, the control unit 19, and the quadrupole drive control unit 22 uses a general-purpose personal computer as hardware resources and operates dedicated control / processing software installed in the computer. By doing so, it can be set as the structure which embodies the function of these each part. That is, the triple quadrupole mass spectrometer of the present embodiment has the same hardware as the conventional apparatus, and the software for operating the apparatus and processing the data obtained by the analysis is the conventional one. It can be realized by changing from a thing.
- a voltage obtained by synthesizing a DC voltage and a high frequency voltage is applied to the front-stage quadrupole mass filter 13 from the Q1 power supply unit 25, and among the various ions derived from the target component, ions having a specific mass-to-charge ratio corresponding to the applied voltage Only passes through the filter 13 as precursor ions.
- the ion source 11 is an electrospray ion (ESI) source, for example, depending on the compound (for example, a high molecular compound such as protein), multivalent ions are likely to be generated, and the range of the valence is considerably wide. .
- ESI electrospray ion
- production of a polyvalent ion is assumed, or when generation
- the analyst inputs the mass value m Loss of the fragment desorbed from the precursor ion during the dissociation operation by the mass input unit 201.
- the mass value of the specific chemical structure may be input. This is the same as the neutral loss setting in the conventional neutral loss scan method.
- the analyst inputs at least two values of the valence z Loss of the desorbed fragment, the valence z Prec of the precursor ion, and the valence z Prod of the product ion through the valence input unit 202.
- the valence of the precursor ion and the product ion is 1, there is no procedure for setting these valences.
- other analysis conditions for example, the range of the mass-to-charge ratio of the precursor ion to be scanned (or the range of the mass-to-charge ratio of the precursor ion to be scanned) are input in the case of the neutral loss scan.
- the precursor ion m / z setting unit 222 sets the mass-to-charge ratio M Prec of ions that pass through the preceding quadrupole mass filter 13. For example, in the case of the neutral loss scan, if the range of the mass-to-charge ratio of the precursor ion to be scanned is specified as described above, the mass-to-charge ratio M Prec of the precursor ion is gradually increased from the lower limit value of the range according to this specification. Increase to. When a specific precursor ion is designated, the precursor ion m / z setting unit 222 outputs a constant mass-to-charge ratio M Prec according to the designation.
- specific product ions (or charged fragments) generated by cleavage of multiply charged ions can be selectively detected instead of the neutral loss scan. That is, when the product ions generated from the ions derived from the target component and the desorbed charged fragments can be assumed, the one that can easily pass through the collision cell 14 and the subsequent quadrupole mass filter 16 stably and with high efficiency.
- the analyst inputs parameters in advance so that the other can be detected. Thereby, information about the target component (for example, the content of the target component) can be obtained with high sensitivity.
- composition formula of the fragment to be detached and its valence, or the ionic formula May be selected by the analyst from a number of options prepared in advance, and the mass and valence may be calculated from the composition formula or ion formula inside the quadrupole drive controller 22.
- the analyst may simply select the name of the desorbed fragment from a number of options prepared in advance.
- FIG. 2 is a block diagram of the main part of a triple quadrupole mass spectrometer according to the second embodiment of the present invention. Components identical or corresponding to those of the triple quadrupole mass spectrometer of the first embodiment shown in FIG.
- the input unit 20 selects a valence of a precursor ion in addition to the mass input unit 201 and the valence input unit 202 as a functional block.
- a reference input unit 203 for inputting selection criteria is included.
- the quadrupole drive control unit 22 includes a precursor ion valence determination unit 225 that determines the valence of a precursor ion based on the mass spectrum created by the mass spectrum creation unit 181 included in the data processing unit 18. The method for determining the valence and the above selection criteria will be described in detail later.
- the analyst inputs the mass value m Loss of the fragment desorbed from the precursor ion during the dissociation operation by the mass input unit 201. This is the same as in the first embodiment. Further, the analyst inputs one of the valence z Loss of the desorbed fragment and the valence z Prod of the product ion by the valence input unit 202, and the valence of the precursor ion by the reference input unit 203. Enter selection criteria for selection.
- the selection criterion is a condition for determining the valence of an ion selected as a precursor ion among various multivalent ions having different valences.
- the selection criterion can be any of the following. (1) One arbitrary valence having a valence of 2 or more is set. In this case, multivalent ions having one valence that meet the set selection criteria are selected as precursor ions. (2) An arbitrary plurality of valences among two or more valences are set.
- multivalent ions having a plurality of valences that meet the set selection criteria are sequentially selected as precursor ions.
- An arbitrary valence having a valence of 2 or more is set as the lower limit value of the valence range. In this case, all multivalent ions having a valence equal to or higher than the lower limit set as the selection criterion are sequentially selected as precursor ions.
- a lower limit value and an upper limit value of an arbitrary valence range having a valence of 2 or more are set. In this case, all the multivalent ions having a valence of not less than the lower limit set as the selection criterion and not more than the upper limit are sequentially selected as the precursor.
- the analyst may analyze the analysis conditions other than those described above, for example, the range of the mass-to-charge ratio of the precursor ion to be scanned (or the range of the mass-to-charge ratio of the precursor ion to be scanned), etc. Also enter.
- the mass spectrum creation unit 181 creates a mass spectrum.
- the compound to be analyzed contains isotopes other than stable isotopes, multiple peaks corresponding to isotope ions with the same composition and different isotopes appear on the mass spectrum, that is, isotope peaks. .
- the interval between the isotope peaks included in one isotope peak group depends on the valence of the ion.
- the valence is 1 if the peak interval is about 1 Da, and the valence is about 0.5 Da.
- the reciprocal of the isotope peak interval indicates the valence, such that the number is 2 and the valence is 3 if the interval is about 0.33 Da. Therefore, the precursor ion valence determination unit 225 automatically determines the valence of the multiply charged ions generated based on the interval between the isotope ion peaks derived from the target component appearing on the mass spectrum.
- the precursor ion valence determination unit 225 determines the valence of the multivalent ions to be selected as the precursor ions according to the selection criteria input and set from the reference input unit 203 as described above.
- the one or more valences determined here correspond to the valence z Prec of the precursor ion input by the valence input unit 202 in the first embodiment. That is, in this second embodiment, the valence z Prec of the precursor ion is determined based on the determination result of the valence of the multivalent ion actually generated and the input selection criterion.
- MS / MS analysis is performed.
- the operation during this MS / MS analysis is the same as that of the first embodiment. That is, the precursor ion m / z setting unit 222 outputs a mass-to-charge ratio M Prec according to the designation of the analysis conditions, and the passing product ion m / z calculating unit 223 outputs the mass-to-charge ratio M Prod ,
- M Prod (M Prec ⁇ z Prec ⁇ m Loss ) / z Prod
- M Prod (M Prec ⁇ z Prec ⁇ m Loss ) / z Prod
- the mass-to-charge ratio M Prod of product ions passing through the latter-stage quadrupole mass filter 16 is obtained by calculation.
- the quadrupole drive voltage calculation unit 224 sends control signals to the Q1 power supply unit 25 and the Q3 power supply unit 26 so that voltages corresponding to the mass-to-charge ratios M Prec and M Prod of the precursor ions and product ions are generated. .
- multivalent ions derived from the target component are used as precursor ions, and target product ions generated by cleavage of the precursor ions reach the detector 17 and are detected.
- MS / MS analysis is performed using each of the plurality of ions as a precursor ion.
- the precursor ion is cleaved by the CID in the collision cell 14, but other dissociation methods may be used.
- ECD that promotes dissociation by irradiating ions with low-speed electrons
- the desorbed fragments capture and neutralize the electrons when the polyvalent ions dissociate.
- the total valence before and after dissociation by the charge of the trapped electrons (the total valence before dissociation is the valence of the precursor ion, and the total valence after dissociation is the valence and desorption of the product ion)
- the valence of the fragment (the valence is 0 if neutral) will change. Therefore, when ECD is used, the valence Z Loss of the desorbed fragment in the above description means the valence of the fragment before neutralization (before electron capture).
- ions generated by the ion source 11 emit the ion source 11 and pass through the front quadrupole mass filter 13, the collision cell 14, and the rear quadrupole mass filter 16. It takes time. In Therefore, there is no problem if the mass to charge ratio M Prod mass-to-charge ratio M Prec and product ions of the precursor ion is constant (which is at least a certain period of time to detect the ions), high-speed their mass-to-charge ratio When scanning, the time required for ions to pass may not be negligible.
- control is performed such that the mass-to-charge ratio M Prod of the product ions is scanned while being delayed by a predetermined time with respect to the scanning of the mass-to-charge ratio M Prec of the precursor ions. is there.
- the present invention is applicable even when mass scanning with such a time difference is performed.
