ビフィズス菌のヒストン様プロモーター(Huプロモーター)にて分泌型Trastuzumab scFv(抗HER2低分子化一本鎖抗体[scFv])を発現させるプラスミドを構築した。最初にシグナルペプチド1(SP1)にTrastuzumab scFvを連結したプラスミドpHuSP1-Trastuzumab scFvを作製後、シグナルペプチド部分を入れ換えてpHuSPx-Trastuzumab scFvを作製した。作製の概要を図1に示す。詳細は以下のとおりである。
Trastuzumab scFvインサートを以下のように調製した。まず、Trastuzumab full-body抗体のアミノ酸配列を、RCSB Protein data bank(PDB)(http://www.pdb.org/pdb/home/home.do)からダウンロード(PDB 1N8Z)した。VH、VLおよびリンカー配列のDNAは、PDBデータベースに登録されたcDNA配列をもとに決定し(図2)(配列番号5~11)、このTrastuzumab scFvコード配列(末端にHis-tag配列を含む)をpOZ1ベクターに挿入したプラスミドpOZ Trastuzumab scFv-Hisを鋳型としてPCRを行った。プライマーとしてTrastuzumab scFv_ins_F3プライマー(配列番号14)及びTrastuzumab scFv_ins_R2プライマー(配列番号15)(表1、各プライマーの5’側の15塩基は、以下に示すベクターと相同な配列を有する。)を使用し、Trastuzumab scFvコード領域を増幅して777bpのインサートPCR産物を得た。2.0%アガロースゲル(1×TEB buffer、エチジウムブロマイド入り)を使用してインサートPCR産物をDNA濃度マーカーとともに電気泳動し、濃度を見積もった。
ベクターを以下のように調製した。プラスミドpSP1B-9(GFPuv遺伝子、大腸菌の複製起点、ビフィズス菌の複製起点、スペクチノマイシン耐性遺伝子を含む。(図3)(配列番号16)を、鋳型としてPCRを行った。プライマーとしてSP1_Vec_F3プライマー(配列番号12)及びSP1_Vec_R2プライマー(配列番号13)(表1)を使用し、GFPuv遺伝子を除いた領域を増幅して3983bpのベクターPCR産物を得た。(PrimeSTAR;登録商標 HS Premix、タカラバイオ社製)。0.8%アガロースゲル(1×TEB buffer、エチジウムブロマイド入り)を使用してベクターPCR産物をDNA濃度マーカーとともに電気泳動し、濃度を見積もった。
In-Fusion(登録商標)HD Cloning Kit with Cloning Enhancer(Clontech社製)を用いてインサートPCR産物とベクターPCR産物の連結を行った。まずClontech社のウェブサイトIn-Fusion(登録商標) Molar Ratio Calculator (http://bioinfo.clontech.com/infusion /molarRatio.do)にて必要なインサート量及びベクター量を算出した。5×In-Fusion HD Enzymes premix 2μL、Cloning Enhancer 1μL、インサート及びベクターの必要量を混合し、滅菌水で反応系の総量を10μLとした。37℃で15分間反応後、50℃で15分間反応し、4℃に静置した。
In-Fusion反応液2μLを用いて大腸菌TOP10 chemically Competent Cell(Invitrogen社製)の形質転換を行い、LB(75μg/mL、スペクチノマイシン含有)プレートに塗布後37℃で一晩培養した。形質転換条件は、製品説明書に従った。形質転換した大腸菌コロニーをLB(75μg/mL、スペクチノマイシン含有)液体培地にて37℃で一晩培養し、これよりプラスミドを抽出した(QIAprep Spin Miniprep Kit,キアゲン社製)。このプラスミドの全塩基配列が設計通りになっていることを確認し、プラスミドpHuSP1-Trastuzumab scFvとした。
各シグナルペプチドを含むプラスミドを鋳型としてPCRを行い、インサートを調製した。表2に示したプライマー(各プライマーの5’側の15塩基は以下に示したベクターと相同な配列を有する)及び鋳型を用いてPCRを行い、インサートPCR産物を得た。2.0%アガロースゲルを使用してインサートPCR産物をDNA濃度マーカーとともに電気泳動し、濃度を見積もった。
プラスミドpHuSP1-Trastuzumab scFvを鋳型としてPCRを行い、ベクターを調製した。プライマーとしてHu-Trastuzumab_vec_F1プライマー(配列番号17)及びHu-vec_R1プライマー(配列番号18)(表2)を使用し、シグナルペプチド部分(SP1)を除いた領域を増幅して4589bpのベクターPCR産物を得た(図4,Trastuzumab scFv遺伝子、大腸菌の複製起点、ビフィズス菌の複製起点、スペクチノマイシン耐性遺伝子を含む)。ベクターPCR産物を0.8%アガロースゲルにてDNA濃度マーカーとともに電気泳動し、濃度を見積もった。
上記と同様にIn-Fusion(登録商標)HD Cloning Kit with Cloning Enhancer(Clontech社製)を用い、インサートPCR産物とベクターPCR産物の連結を行った。
In-Fusion反応液1μLを用いて大腸菌TOP10 chemically Competent Cell(Invitrogen社)の形質転換を行い、LB(75μg/mL、スペクチノマイシン含有)プレートに塗布後37℃で一晩培養した。形質転換条件は、製品説明書に従った。形質転換した大腸菌コロニーをLB(75μg/mL、スペクチノマイシン含有)液体培地にて37℃で一晩培養し、これよりプラスミドを抽出した(QIAprep Spin Miniprep Kit,キアゲン社製)。このプラスミドのHuプロモーター付近からターミネーター付近までの塩基配列が設計通りになっていることを確認した。表3に作製したプラスミドを示す。
形質転換した大腸菌から抽出したプラスミドDNA(表3)を1.5~3μL用いて、ビフィズス菌Bifidobacterium longum 105Aの形質転換をエレクトロポレーションシステム(ジーンパルサーII、バイオ・ラッドラボラトリーズ社製)により行った。電気ショック後は、キュベットにIMR液体培地800μLとビタミンC添加液50μLの混合液を即時に加え、これを滅菌済みの2mLマイクロチューブに回収した。各チューブについて同様の操作を行い、これら2mLチューブの蓋をゆるめてアネロパックとともに密閉容器に入れ、37℃に設定したインキュベータに入れ、3時間保温した。保温後の各菌懸濁液をよく混ぜた後、その100μLを量り取り、IMR寒天培地(75μg/mL SPCMを含有)1枚にそれぞれ塗抹した。これらの寒天培地を脱酸素・炭酸ガス発生剤(アネロパック・ケンキ、三菱ガス化学社製)とともに密閉容器に入れ、37℃に設定したインキュベータにて2~3日間培養した。生育したプレート上のコロニーをディスポスティックでピックアップし、BL-bS寒天培地(75μg/mL SPCMを含有)に画線し、脱酸素・炭酸ガス発生剤(アネロパック・ケンキ、三菱ガス化学社製)とともに密閉容器に入れ、37℃に設定したインキュベータにて1日間培養し、画線培養物とした。
上記で得た組換えビフィズス菌(Bifidobacterium longum 105A/pHuSPx-Trastuzumab scFv(x=2~10,12~16,19,21~27)の画線培養物を、APS-2S-2.5SE(75μg/mL、スペクチノマイシン)液体培地に植菌し、37℃で24時間嫌気培養した(活性化培養液)。次に、DMEM(低グルコース、ピルビン酸、HEPES)細胞培養用培地(ライフテクノロジーズ社製、cat #12320-032):APS-2S-2.5SE=9:1にスペクチノマイシンを75μg/mLになるよう添加した培地に、活性化培養液0.5%を植菌した。これを37℃で15時間嫌気培養した。この培養液を用いて、培養上清及び細胞内タンパク質を以下のように調製した。培養液を遠心分離後、培養上清を回収した。この培養上清中のタンパク質をトリクロロ酢酸(TCA)により沈殿させ、アセトンにて洗浄後、電気泳動用バッファーに溶解し、培養上清中のタンパク質を濃縮した。別に、細胞内タンパク質を以下のように抽出した。培養液1mLを4mLのPBSと混合し、12,000rpm、5min、4℃にて遠心分離後、上清を除去し、この沈殿に5mLのPBSを加え懸濁、遠心分離し上清を除去する操作を2回行なった。洗浄後の菌体にPBSを加え全量を1mLとした後、超音波処理により菌体を破砕した。これを遠心分離後、上清を回収し、これを細胞内抽出物とした。上記培養上清濃縮物(培養液1mL相当)、細胞内タンパク抽出物培養液1mL相当)をミニプロティアンTGXゲル4-20%(バイオラッド社製)にて電気泳動した。これをPVDF膜(Invitrogen社製、iBlot Transfer Stacks)にiBlot Transfer Device(Invitrogen社製)を用いて転写した。ブロッティング終了後の膜をブロッキングした後、一次抗体にAnti-His antibody(GE Healthcare社製)、二次抗体にanti-mouse Ab-HRP(GE Healthcare社製)を使用して反応を行ない、Western Lightning Ultra(Perkin Elmer社製)にて発光させた。これをイメージングアナライザー(Fluor S Max,バイオラッド社製)にて解析した。その結果、3種の菌(B. longum 105A/pHuSP3L22-Trastuzumab scFv、B. longum 105A/pHuSP27L0-Trastuzumab scFv、B. longum 105A/pHuSP27L6-Trastuzumab scFv)で分泌が認められた(図5)。ウェスタンブロッティングの結果のうち、B. longum 105A/pHuSP27L6-Trastuzumab scFvとB. longum 105A/pHuSP3L22-Trastuzumab scFvの結果を図6に示す。Trastuzumab scFvのサイズ付近(約25kDa)にバンドを確認することができた。
分泌型Trastuzumab scFvによる組換えビフィズス菌(Bifidobacterium longum 105A)の画線培養物を、APS-2S-2.5SE(75μg/mL、スペクチノマイシン)液体培地に植菌し、37℃で24時間嫌気培養した(活性化培養液)。次に、DMEM(低グルコース、ピルビン酸、HEPES)細胞培養用培地(ライフテクノロジーズ社製、cat #12320-032):APS-2S-2.5SE=9:1にスペクチノマイシンを75μg/mLになるよう添加した培地に、活性化培養液0.5%を植菌した。これを37℃で16時間嫌気培養した。培養液を遠心分離後、培養上清に、硫酸アンモニウム(酵素精製用、和光純薬工業社製)を80%飽和溶液になるよう添加し、4℃にて一晩撹拌した。これを遠心分離後、沈殿物に1×PBSバッファー(pH7.4)を加え、沈殿を溶解した。この溶液を、ヒスチジンタグタンパク質の精製キット(TALON Metal Affinity Resin、タカラバイオ社製)を用いて精製した。精製溶液を限外濾過(アミコンウルトラ 10K、メルクミリポア社製)にて濃縮した。濃縮タンパク質溶液に、等量の2×SDS sample bufferを混合し、95℃にて3分間加熱し、これをSDS-PAGEの試料とした。上記試料を、ミニプロティアンTGXゲル(4-20%、BIO-RAD社製)を用いて1×SDSバッファーにて電気泳動した。電気泳動終了後のゲルを水にて洗浄後、染色液(SimplyBlueTM SafeStain)にて染色し、水にて脱色した。結果を図7に示す。リンカーのないSP27L0では、Trastuzumab scFvのサイズ付近(約25kDa)のバンドを確認することができなかったが、(図中上段矢印)、リンカーを組み込んだSP27L6、SP3L22では、予想されるTrastuzumab scFvのサイズのバンドが確認された(図7)。
組換えビフィズス菌B. longum 105A/pHuSP27L6-scFvを培養し、培養上清よりヒスチジンタグ融合タンパク質精製キット(TALON Metal Affinity Resin,タカラバイオ社製)を用いて精製を行った。精製タンパク質を4-20%ポリアクリルアミドゲル(ミニプロティアンTGXゲル、Bio-Rad社製)にて電気泳動後、Simply Blue Stain(Invitrogen社製)で染色してバンドを切出した。染色後のゲルからバンドを切り出し、1×1mm3サイズのゲルキューブにし、ゲルを脱色液(30%アセトニトリル(和光純薬社製)、25mM重炭酸アンモニウム(SIGMA社製)を含む水溶液)で脱色後、10mM DTT((±)-ジチオトレイトール、和光純薬社製)で還元(56℃、45min.)、室温まで空冷後55mM ICH2CONH2(ヨードアセトアミド、和光純薬社製)でアルキル化(室温、遮光、30min.)した。アルキル化後、12.5ng/mLトリプシン(Promega社製)でゲル内消化(37℃、16h)し、消化ペプチド断片をゲル内より抽出し濃縮した。(Reference: Shevchenko A. et al. Anal. Chem. 68, 850-858, 1996.)得られたペプチド断片を用いてLC-MS/MS(Waters nano ACQUITY UPLC, Xevo QTOF社製)で分析及び解析し、同定されたペプチドを、Trastuzumab scFvアミノ酸配列と照合した(図8)。LC-MS/MS分析によりTrastuzumab scFvアミノ酸配列と一致したペプチド断片を図9に示す。
上述と同様にPDBデータベースから得られた、TrastuzumabcDNA配列をもとに、人工的に遺伝子合成されたDNAを、pUC119プラスミドへ組み入れてベクターを作製し、Trastuzumab scFv発現用ベクターとした。Rosetta2 E. coli(Merck Millipore,Darmstadt,Germany)にtrastuzumab-scFv-His plasmidを遺伝子導入し、アンピシリンとクロラムフェニコールを含むagarでコロニーを選別した。コロニーは、2%グルコースを含むアンピシリン含有LB培地10mLにて37℃で増殖させた。その後IPTGを含む200mLのLB培地に移し、25℃で20時間培養を継続した。大腸菌を回収し、ベンゾヌクレアーゼを含むBugBuster Protein Extraction Reagent(Novagen社製)にて細胞ライセ―トを抽出し、その後10000×gで5分間遠心し、抗体タンパクを含む上清を回収した。Hisタグの付いた抗体タンパクは、HisTrap HP Ni Sepharose columnおよびSephadex G25 gel filtrationにて精製された。精製後、Tris-Glycine gelにてSDS-PAGEを施行し、30kDaに一致したTrastuzumab scFvのバンドが確認された(図10)。精製されたscFv抗体は、フローサイトメトリ―やビアコアでの表面プラズモン共鳴(SPR)アッセイに使用された。
Trastuzumab scFvは大腸菌Rozetta2株を用いて作製したものを使用し、HER2細胞外ドメインタンパク(HER2.ex.)及びTrastuzumab full-body抗体は昆虫由来HF細胞株を用いて作製したものを使用した。SPR解析はBiacore X100を用いて行った。測定に使用したすべての試薬やセンサーチップはGEヘルスケアから購入した。HER2.ex.のCM5センサーチップ上への固定にはpH5.0の10mM Acetate溶液を、Trastuzumab full-body抗体の固定にはpH5.5の溶液を用いた。測定は0.05%又は0.005%Tween-PBS(pH7.4)の緩衝液を用い、流速30μL/mL、25℃で行った。センサーチップ上で、固定化したHER2.ex.に対し、添加したTrastuzumab scFvとの相互作用を観察した後、結合したTrastuzumab scFvを再生溶液で完全に除去し、続いて、濃度の異なるTrastuzumab scFvについても同様の測定サイクルを繰り返し、得られた結果をまとめて解析するマルチサイクル法と、1回の測定サイクル中に5通りの濃度のTrastuzumab scFvを順次添加するだけで、解析に必要な情報を得ることができ、HER2.ex.に結合したTrastuzumab scFvを除去する必要のないシングルサイクル法の、ふたつの方法でTrastuzumab scFvの抗原に対する結合親和性を求めた。結合、解離に関する動態定数の計算はBiacore X100 evaluation softwareを用いて算出した。解析結果を図11に示す。
上記と同様に、Trastuzumab full-body抗体を固定側、流路側にHER2.ex.を用いて、HER2抗原との結合親和性をBiacore X100によるシングルサイクル法にて解析し、作製したTrastuzumab scFvにおける結合親和性との比較を行った。便宜上、Trastuzumab scFvでの解析は、固定側にHER2.ex.、流路側にTrastuzumab scFvを用いたため、流路と固定の関係がTrastuzumab full-body抗体での解析とは逆転した形となった(図12)。結果、作製したTrastuzumab scFvは、Trastuzumab full-body抗体と比べて同等の結合親和性があることが分かった。また、反応速度定数のka値(大きいほど抗原との親和性が高い)及びkd値(小さいほど抗原との結合が強い)の値から、作製したTrastuzumab scFvは、Trastuzumab full-body抗体と比べて抗原との親和性は高いが、抗原との結合が弱い性質であることが示された。
ヒト乳がん細胞株は、HER2陽性株(SKBR-3)、陰性株(MDA-MD231、468)を使用した。FACS用の抗His抗体は、PE標識抗His抗体(CAT. 130-092-691、Miltenyi社製)を使用した。またTrastuzumab scFvの対照コントロールとして、human CMVpp65 scFvを使用した。試薬には、Ab bufferとしてPBS(-)+0.1% BSA+0.1% sodium azide、FACS bufferとしてPBS(-)+1% FBS+0.1% sodium azide+2mM EDTA、fixation bufferとしてFACS buffer+0.5% PFA(paraform aldehyde)を使用し、Trastuzumab scFv及びhuman CMVpp65 scFvは10μg/mLに調整した。2次抗体にはPE-anti-His抗体を用いた。
1~2×105/wellの細胞をFACS bufferに懸濁し、96well round-bottom plateに撒き、Plateを1400rpmにて2分間遠心後、上清を吸引した。10μLのAb bufferを添加し懸濁後、10μLのTrastuzumab scFv又は、対照用のhuman CMVpp65 scFv(10μg/mL)を添加した。4℃にて15分間のインキュベーションの後、150μLのFACS bufferにて2回洗浄した。1400rpmにて4分間遠心した後、上清を吸引して除去した。10μLのAb bufferを添加した後、10μLのPE-anti-His抗体(希釈なし)を加え、4℃にて15分間インキュベーションを行った。150μLのFACS bufferにて2回洗浄した後、1400rpmにて2分間遠心を行い、上清を吸引した。400μLのfixation bufferを加え、ピペッティングにてFACS tubeに移し、24時間以内にFlow Cytometry(FACSCanto、BD Biosciences社製)による測定を行った。結果を図13に示す。full-body抗体にはやや劣るものの、Trastuzumab scFvは、十分に陰性コントロール細胞との分離が良い抗体であることが示された(図13中央と右)。また、SKBR-3細胞の発現するHER2抗原を検出できない、コントロールの抗サイトメガロウイルス抗原scFvとの比較において、Trastuzumab scFvが抗原特異的に結合していることが示された(図13左)。
HER2抗原を高発現するヒト乳腺癌細胞株MDA-MB-361(5×106細胞/個体[マトリゲル含有])を、免疫学的にT細胞機能が欠如したBALB/cA-nu/nuマウス(メス、7週齢、日本クレア社製)の乳腺上皮に移植した。腫瘍体積が約290mm3に達したMDA-MB-361担癌マウス(1匹)に蛍光色素Cy5.5を標識したTrastuzumab scFvの3.75mg/kg相当量を腫瘍内に1回投与(100μL)した。Cy5.5標識Trastuzumab scFvの腫瘍内投与における滞留性を確認するためにイソフルラン吸入麻酔下にて、in vivoイメージング装置eXplore Optix(GEヘルスケア社製)を使用して1週間測定した(図14)。1週目では、安楽死処置を行った後、腫瘍、肝臓、脾臓、肺、及び小腸を摘出してイメージング能を測定した。結果、Cy5.5標識Trastuzumab scFvを腫瘍内投与してから、7日目後においても検出可能であることが示された。
上記と同様に、4×106細胞/個体のMDA-MB-361を乳腺上皮に移植した、担癌ヌードマウスの系を用い、Cy5.5標識Trastuzumab full-body抗体の腫瘍内投与における滞留性を確認するため、移植後56日目の担癌マウス(400mm3)に、Cy5.5標識Trastuzumab full-body抗体3.75mg/kg相当量を腫瘍内投与した。腫瘍内滞留性はイソフルラン吸入麻酔下にeXplore Optixを使用して投与後5日目まで測定した(図15)。腫瘍内投与から5日後においても、マウス生体内での蛍光標識抗体を観察できたことから、Cy5.5標識Trastuzumab scFv同様に長期間の検出が可能であることが示された。
HER2抗原を高発現するヒト乳腺癌細胞株MDA-MB-361(5×106細胞/個体[マトリゲル含有])を、上述のBALB/cA-nu/nuマウス(n=4~5/群)乳腺上皮に移植した。平均腫瘍体積が250mm3に達したMDA-MB-361担癌マウスに、Trastuzumab scFvの3.75mg/kg相当量を1日おきに5回、腫瘍内投与(50μL)した。Trastuzumab scFv投与開始から経日的に腫瘍体積を測定して、無処理群(コントロール群)との抗腫瘍活性を比較した。3~4日ごとに計測した腫瘍体積を指標とした結果から、明確な腫瘍細胞の増殖抑制効果が示された(図16)。
Trastuzumab scFv遺伝子の塩基配列をビフィズス菌のコドンに最適化した(opt-Trastuzumab scFv遺伝子:配列番号92)。更に、opt-Trastuzumab scFv遺伝子を組み込んだ分泌プラスミド(pHuSP7L20-opt-Trastuzumab scFv:配列番号94)を作製した。
opt-Trastuzumab scFv遺伝子は、大腸菌用プラスミドpUC57にサブクローニングされたプラスミドpUC57-opt-Trastuzumab scFvとして、人工遺伝子合成(ジェンスクリプトジャパン社製)した。第一段階として、既存のTrastuzumab scFv 発現・分泌プラスミドであるpHuSP27L0-Trastuzumab scFv(配列番号93)のTrastuzumab scFv遺伝子をopt-Trastuzumab scFv遺伝子と交換し、プラスミドpHuSP27L0-opt-Trastuzumab scFvを作製した。第二段階として、シグナルペプチド+リンカー配列であるSP27L0をSP7L20に交換し、pHuSP7L20-opt-Trastuzumab scFv(配列番号94)を作製した。第三段階として、pHuSP7L20-opt-Trastuzumab scFvのHuプロモーターをP30プロモーターに交換し、pP30SP7L20-opt-Trastuzumab scFv(配列番号95)を作製した。詳細は以下のとおりである。
pHuSP27L0-Trastuzumab scFvのTrastuzumab scFv遺伝子部分をopt-Trastuzumab scFv遺伝子に入れ換える手順を以下に示す。まず、opt-Trastuzumab scFvインサートを以下のように調製した。opt-Trastuzumab scFvコード配列(末端にHis-tag配列を含む)を有するプラスミドpUC57-opt-Trastuzumab scFvを鋳型としてPCRを行った。プライマーとして、opt-Trastuzumab scFv_ins_INF_F1プライマー(配列番号96)、及びopt-Trastuzumab scFv_ins_INF_R1プライマー(配列番号97)(各プライマーの5’側の15塩基は、以下に示すベクターと相同な配列を有する)を使用し、opt-Trastuzumab scFvコード領域を増幅して777bpのインサートPCR産物を得た。2.0%アガロースゲル(1×TEB buffer、エチジウムブロマイド入り)を使用してインサートPCR産物をDNA濃度マーカーとともに電気泳動し、濃度を見積もった。
ベクターを以下のように調製した。pHuSP27L0-Trastuzumab scFv(Trastuzumab scFv遺伝子、大腸菌の複製起点、ビフィズス菌の複製起点、スペクチノマイシン耐性遺伝子を含む)を鋳型としてPCRを行った。プライマーとしてSP1_Vec_F1プライマー(配列番号98)及びd0018_0aa_Vec_R3プライマー(配列番号99)を使用し、Trastuzumab scFv遺伝子を除いた領域を増幅して約4kbのベクターPCR産物を得た。(PrimeSTAR;登録商標 HS Premix、タカラバイオ社製)。0.8%アガロースゲル(1×TEB buffer、エチジウムブロマイド入り)を使用してベクターPCR産物をDNA濃度マーカーとともに電気泳動し、濃度を見積もった。
In-Fusion(登録商標)HD Cloning Kit with Cloning Enhancer(Clontech社製)を用いてインサートPCR産物とベクターPCR産物の連結を行った。まずClontech社のウェブサイトIn-Fusion(登録商標) Molar Ratio Calculator (http://bioinfo.clontech.com/infusion /molarRatio.do)にて必要なインサート量及びベクター量を算出した。5×In-Fusion HD Enzymes premix 2μL、Cloning Enhancer 1μL、インサート及びベクターの必要量を混合し、滅菌水で反応系の総量を10μLとした。37℃で15分間反応後、50℃で15分間反応し、4℃に静置した。
In-Fusion反応液2μLを用いて大腸菌HST16CR Competent Cells(タカラバイオ社製)の形質転換を行い、LB(75μg/mL、スペクチノマイシン含有)プレートに塗布後、37℃で一晩培養した。形質転換条件は、製品説明書に従った。形質転換した大腸菌コロニーをLB(75μg/mL、スペクチノマイシン含有)液体培地にて37℃で一晩培養した後、プラスミドを抽出した(QIAprep Spin Miniprep Kit、キアゲン社製)。このプラスミドの全塩基配列が設計通りになっていることを確認し、プラスミドpHuSP27L0-opt-Trastuzumab scFvとした。
pHuSP27L0-opt-Trastuzumab scFv のシグナルペプチド+リンカー配列(SP27L0)をSP7L20に入れ換える手順を以下に示す。まずBifidobacterium longum 105AのゲノムDNAを鋳型としてPCRを行い、シグナルペプチドインサートを以下のように調製した。SP7L20-opt-Trastuzumab scFv_ins_INF_F1プライマー(配列番号100)及びSP7L20-opt-Trastuzumab scFv_ins_INF_R1プライマー(配列番号101)(各プライマーの5’側の15塩基は、以下に示すベクターと相同な配列を有する)を用いてPCRを行い、189bpのインサートPCR産物(SP7L20)を得た。2.0%アガロースゲルを使用してインサートPCR産物をDNA濃度マーカーとともに電気泳動し、濃度を見積もった。
プラスミドpHuSP27L0-opt-Trastuzumab scFvを鋳型としてPCRを行い、ベクターを調製した。プライマーとしてHu-opt-Trastuzumab_vec_F1プライマー(配列番号102)及びHu-Vec_R1プライマー(配列番号18)を使用し、シグナルペプチド+リンカー部分を除いた領域を増幅して約4.5kbのベクターPCR産物を得た。ベクターPCR産物を0.8%アガロースゲルにてDNA濃度マーカーとともに電気泳動し、濃度を見積もった。
上記と同様にIn-Fusion(登録商標)HD Cloning Kit with Cloning Enhancer(Clontech社製)を用い、インサートPCR産物とベクターPCR産物の連結を行った。
上記と同様にIn-Fusion反応液を用いた大腸菌HST16CR Competent Cellsの形質転換、培養、及びプラスミド抽出を行い、このプラスミドのopt-Trastuzumab scFv発現カセット(Huプロモーターからターミネーターまで)の塩基配列が設計通りになっていることを確認した。
pHuSP7L20-opt-Trastuzumab scFvのプロモーターを、B. longum 105Aのゲノムに存在する遺伝子のP30プロモーターに交換した。
まず、B. longum 105AのゲノムDNAを鋳型としてPCRを行い、シグナルペプチドインサートを以下のように調製した。P30_ins_F1プライマー(配列番号103)及びP30_SP7_ins_R1プライマー(配列番号104)(各プライマーの5’側の15塩基は、以下に示すベクターと相同な配列を有する)を用いてPCRを行い、265bpのインサートPCR産物を得た。2.0%アガロースゲルを使用してインサートPCR産物をDNA濃度マーカーとともに電気泳動し、濃度を見積もった。
プラスミドpHuSP7L20-opt-Trastuzumab scFvを鋳型としてPCRを行い、ベクターを調製した。プライマーとしてSP7_Vec_F1プライマー(配列番号105)及びpUC_ori_Vec_R2プライマー(配列番号106)を使用し、opt-Trastuzumab scFv発現ユニットのプロモーターを除いた領域を増幅して約4.4kbのベクターPCR産物を得た。ベクターPCR産物を0.8%アガロースゲルにてDNA濃度マーカーとともに電気泳動し、濃度を見積もった。
上記と同様にIn-Fusion(登録商標)HD Cloning Kit with Cloning Enhancer(Clontech社製)を用いてインサートPCR産物とベクターPCR産物の連結を行った。
前述の操作と同様にIn-Fusion反応液を用いた大腸菌HST16CR Competent Cellsの形質転換、培養及びプラスミド抽出を行い、このプラスミドのopt-Trastuzumab scFv発現カセット(P30プロモーターからターミネーターまで)の塩基配列が設計通りになっていることを確認した。
プラスミドpHuSP7L20-opt-Trastuzumab scFv、及びpP30SP7L20-opt-Trastuzumab scFvを用いてビフィズス菌の形質転換を以下のように実施した。プラスミドDNAを用いて、ビフィズス菌B. longum 105Aの形質転換をエレクトロポレーションシステム(ジーンパルサーII、バイオ・ラッドラボラトリーズ社製)により行った。電気ショック後は、キュベットにIMR液体培地800μLとビタミンC添加液50μLの混合液を即時に加え、これを滅菌済みの2mLマイクロチューブに回収した。各チューブについて同様の操作を行い、これら2mLチューブの蓋をゆるめてアネロパックとともに密閉容器に入れ、37℃に設定したインキュベータに入れ、3時間保温した。保温後の各菌懸濁液をよく混ぜた後、その100μLを量り取り、IMR寒天培地(75μg/mL SPCMを含有)1枚にそれぞれ塗抹した。これらの寒天培地を脱酸素・炭酸ガス発生剤(アネロパック・ケンキ、三菱ガス化学社製)とともに密閉容器に入れ、37℃に設定したインキュベータにて2~3日間培養した。生育したプレート上のコロニーをディスポスティックでピックアップし、BL-bS寒天培地(75μg/mL SPCMを含有)に画線し、脱酸素・炭酸ガス発生剤(アネロパック・ケンキ、三菱ガス化学社製)とともに密閉容器に入れ、37℃に設定したインキュベータにて1日間培養し、画線培養物とし、組換えビフィズス菌B. longum 105A/pHuSP7L20-opt-Trastuzumab scFv、及びB. longum 105A/pP30SP7L20-opt-Trastuzumab scFvを得た。
上記にて作製したTrastuzumab scFv分泌ビフィズス菌B. longum 105A/pHuSP7L20-opt-Trastuzumab scFvを用い、Trastuzumab scFvの精製を行った。
B. longum 105A/ pHuSP7L20-opt-Trastuzumab scFvを、APS-2S-2.5SE培地(75μg/mLスペクチノマイシン含有)に植菌し、37℃にて24時間嫌気培養を行った。この培養液をDMEM:APS-2S-2.5SE=9:1にスペクチノマイシンを75μg/mLになるよう添加した培地に0.5%添加し、37℃にて18時間嫌気培養をした。
上記培養液を遠心分離して得た培養上清を撹拌しながら、硫酸アンモニウムを80%飽和になるよう少しずつ添加し、4℃にて一晩撹拌し、塩析を行った。これを遠心分離後、沈殿を回収し、ヒスチジンタグ付きタンパク質の精製キット(TALON resin、タカラバイオ社製)にてヒスチジンタグを指標とした精製を行った。この精製溶液を限外ろ過(アミコンウルトラ-0.5、メルクミリポア社製)にて濃縮した。
上記精製単鎖抗体の一部を用いてSDS-PAGE後、クマシーブルー染色(ライフテクノロジーズ社製、SimplyBlueTM SafeStain)を行い、約9割の純度でTrastuzumab scFvが精製されていることを確認した。精製タンパク質の濃度は、Bradford法にて測定した(Coomassie Plus Protein Assay、Thermo Scientific社製)。
SDS-PAGE分析の結果を図17に示す。Trastuzumab scFvのサイズ付近(約25kDa)にバンドを確認することができた。
B. longum 105A/pHuSP7L20-opt-Trastuzumab scFvから精製したTrastuzumab scFvを用いてヒト乳癌細胞株との結合を確認した。
ヒト乳癌細胞株は、HER2陽性株(SK-BR-3、及びBT-474)、及びHER2陰性株(SK-MEL-28)を使用した(全てAmerican Type Culture Collection、ATCCより購入)。免疫染色用の抗His抗体は、Alexa Fluor 488標識抗His抗体(Cat. D291-A48、医学生物学研究所社製)を使用した。また、抗HER2 full-body抗体(Cat. 427041、ニチレイバイオサイエンス社製)を添加し、細胞のHER2の発現を確認した。試薬には、wash bufferとしてPBS(-)、Ab bufferとしてPBS(-)+1.5% BSA、fixation bufferとして4% PFA(paraform aldehyde)・リン酸緩衝液(和光純薬工業社製)を使用し、Trastuzumab scFvおよび抗HER2 full-body抗体はAb bufferで5μg/mLに調製した。Trastuzumab scFvに対する2次抗体にはAnti-His-tag mAb-Alexa Fluor 488を、抗HER2 full-body抗体に対する2次抗体にはDyLight 594 goat anti-mouse IgG(Cat. 405311、BioLegend社製)を用いた。封入剤にはVECTASHIELD Mounting Medium with DAPI(Cat. H-1200、VECTOR Laboratories社製)を使用した。
6 well plate(NEST社製)に18mm×18mmのカバーガラス(松浪硝子工業社製)を置き、Poly-L-lysine(シグマアルドリッチ社製)コーティングを行い、1~2×104cells/glassで細胞を培養した。その1日後にプレートを氷上に静置して培地を吸引し、wash bufferにて3回洗浄した。細胞に100μLのAb bufferを添加し、氷上にて30分間インキュベーションを行った。その後、100μLのTrastuzumab scFv、又は陰性コントロールとしてのAb bufferを添加し、氷上にて1時間インキュベーションを行った。scFv反応後、wash bufferにて3回洗浄した。洗浄後、100μLのAnti-His-tag mAb-Alexa Fluor 488(400倍希釈)を加え、氷上にて30分間インキュベーションを行った。インキュベーション後、wash bufferにて3回洗浄し、500μLのfixation bufferを加えて氷上で10分間インキュベーションを行った。その後、wash bufferにて3回洗浄し、DAPI含有封入剤で封入し、蛍光顕微鏡(DM5000B、Leica MICROSYSTEMS社製)による観察を行った。
結果を図18に示す。HER2陽性細胞にHER2が発現し、HER2陰性細胞にはHER2が発現していないことを確認した。さらに、Trastuzumab scFvはHER2陽性細胞に特異的に結合し、細胞表面のHER2と共局在していることが示された。
B. longum 105A/pHuSP7L20-opt-Trastuzumab scFvから精製したTrastuzumab scFvを用いてヒト乳癌細胞株との結合を確認した。
ヒトHER2が陽性であるBT-474細胞及びSK-BR-3細胞、及び陰性であるSK-MEL-28細胞(全てAmerican Type Culture Collection、ATCCより購入)を使用した。これらの細胞は100mmペトリディッシュ(日本ジェネティクス社製)にて培養した。BT-474細胞はHybri-care培地、SK-BR-3細胞はMcCoy’s 5A培地、SK-MEL-28細胞はE-MEM培地を用いてそれぞれ培養した。これらの培地には全て、非働化した10%ウシ胎児血清(EQUITECH-BIO社製)、及びペニシリン/ストレプトマイシン液(コスモバイオ社製)を混合した。
上記細胞がある程度密集してきた段階で培地を除去し、PBS(-)(和光純薬工業社製)にて洗浄し、Trypsin-EDTA(Life Technologies社製)を用いてペトリディッシュから細胞を剥がし、15mLコニカルチューブ(サンプラテック社製)に移した。卓上小型遠心機(久保田製作所製)で1000rpm、5分間遠心後、上清を除去し、培地を5mL加えた。この細胞懸濁液内に含まれる細胞数を血球計算盤にて計測し、3×104cells/チューブになるように1.5mLチューブ(Eppendorf社製)に分注した。1.5mLチューブに分注した上記細胞を微量冷却遠心機(TOMY社製)にて5000rpm、4℃で1分間遠心し、遠心後上清を除去した。チューブに残った細胞のペレットを1mL PBS(-)を用いて2回洗浄後、精製した抗HER2 scFvを10μg/mLの濃度で100μL添加し、氷上にて30分間静置した。そこに蛍光標識抗His-tag抗体(Anti-His-tag Alexa Fluor 488、医学生物学研究所社製)20μLを加え、ピペッティングにてよく混合し、氷上で20分間静置した。静置後、FACSバッファー(1%BSA、0.1%NaN3を含むPBS)を500μLチューブに加えて懸濁し、微量冷却遠心機にて5000rpm、4℃で1分間遠心し、上清を除去した。この洗浄操作を再度行った後、500μL FACSバッファーを添加して細胞を懸濁し、5mLポリスチレンラウンドボトムチューブ(ベクトン・ディッキンソン社製)に細胞を移した。
解析直前にヨウ化プロピジウム溶液(5μg/mL)を5μL添加し、BD FACS cantoIIフローサイトメーター(ベクトン・ディッキンソン社製)、及びフローサイトメトリー解析ソフトウェアKaluza ver1.2(ベックマン・コールター社製)にて解析を行った。
結果を図19に示す。図19上段では、HER2陽性細胞であるBT-474細胞、及びSK-BR-3細胞にTrastuzumab scFvが結合していることが確認された。一方図19下段では、HER2陰性細胞であるSK-MEL-28細胞には、Trastuzumab scFvの結合は認められなかった。
Trastuzumab scFvの生理活性は、B. longum 105A/pP30SP7L20-opt-Trastuzumab scFvの培養上清をHisタグ精製することにより得たTrastuzumab scFv(PBS置換)を、HER2陽性細胞(BT474(乳癌)細胞)に添加し、細胞増殖抑制活性を測定した。
BT474細胞をMcCoy’s5A培地(10%(v/v)FBSを含む)にて37℃、5%CO2の条件下で培養後、96穴プレートに1×104cellsずつ播種し、37℃、5%CO2の条件下で24時間培養した。培養後、培地を吸引除去し、新たなMcCoy’s5A培地(10%(v/v)FBSを含む)を98μLずつ添加した。次に、測定試料としてPBS(-)、抗Trastuzumab scFvを244ng/mL~1mg/mLに調整したものを2μLずつ添加した。このプレートを37℃にて5%CO2の条件下で5日間培養した。
上記5日間培養後の培地を吸引除去した後、新しいMcCoy’s5A培地(10%(v/v)FBSを含む)9mLにCell Counting キット-8を1mL添加したものを100μLずつ添加し、更に3時間、37℃、5%CO2の条件下で保温し、波長450nmと630nm(参照波長)での吸光度を測定することにより、上記HER2陽性細胞に対する細胞増殖抑制活性を測定した。
結果を図20に示す。B. longum 105A/pP30SP7L20-opt-Trastuzumab scFvより精製したTrastuzumab scFvは、BT474乳癌細胞に対し用量依存性の細胞増殖抑制活性を示し、Trastuzumab scFvは生理活性を有することを確認した。
ヒト胃癌細胞株NCI-N87の担癌ヌードマウスを用いて、B. longum 105A/pP30SP7L20-opt-Trastuzumab scFvの抗腫瘍効果を以下のように確認した。
ヒト胃癌細胞株NCI-N87(ATCCにて購入)を、10%FBS(EQUITECH-BIO,INC.社製)を添加したRPMI1640培地(和光純薬工業社製)で培養し、ヌードマウス(日本エスエルシー社製)に移植して担癌マウスを作製した。実験には、腫瘍体積が約200mm3となった担癌マウスを使用した。群構成は1群:無処置群(対照群)、2群:B. longum 105A/pBEshuttle株(Trastuzumab scFv非発現菌)投与群、3群:B. longum 105A/pP30SP7L20-opt-Trastuzumab scFv(Trastuzumab scFv発現菌)投与群、とした。各群につき8匹の担癌マウスを使用した。
ビフィズス菌の投与は、尾静脈から6×108cfuの用量で週2回投与を行った。またビフィズス菌投与群(2群及び3群)には10%マルトース溶液を1mLずつ1日2回の頻度で週5回投与(5投2休)を行った。試験期間を3週間として経時的に腫瘍体積の測定を行い、22日目に腫瘍を摘出してグラム染色及び免疫組織染色に使用した。
腫瘍体積の経時変化を抗腫瘍効果の結果を図21に示す。B. longum 105A/pP30SP7L20-opt-Trastuzumab scFv投与群の腫瘍体積は、試験期間を通じて他の群より低く推移した。試験終了時(22日目)ではB. longum 105A/pBEshuttleと比較して有意に腫瘍体積が減少しており、B. longum 105A/pP30SP7L20-opt-Trastuzumab scFvの抗腫瘍効果が確認された。
上記39.で摘出した腫瘍を用い、腫瘍内のビフィドバクテリウム属細菌の局在をグラム染色にて、Trastuzumab scFvの局在を抗His-tag抗体を用いた免疫組織染色で確認した。
摘出した腫瘍は、O.C.T.compound(サクラファインテック社製)で凍結包埋し、凍結ミクロトームLeica CM1900(Leica社製)を用いて薄切スライド標本を作製し、各組織染色に供した。

結果を図23に示す。ヒスチジンタグの免疫組織染色によりヒスチジンタグ陽性像(Trastuzumab scFv)が確認された(図23の矢印部分)。
Claims (15)
- 以下のa)又はb)に示されるアミノ酸配列からなるシグナルペプチドをコードするDNA。
a)配列番号1又は配列番号107に示されるアミノ酸配列;
b)配列番号1又は配列番号107に示されるアミノ酸配列において、1又は数個のアミノ酸が欠失、置換又は付加されたアミノ酸配列であって、該アミノ酸配列からなるペプチドがビフィドバクテリウム・ロンガムにおいてシグナルペプチドとして機能する、前記アミノ酸配列; - 配列番号2又は配列番号108で示される塩基配列からなる請求項1記載のDNA。
- 以下のa)又はb)に示されるアミノ酸配列からなるシグナルペプチドのC末端に、アミノ酸配列からなるリンカーが連結されたシグナルペプチド-リンカー連結体をコードするDNA。
a)配列番号1又は配列番号107に示されるアミノ酸配列;
b)配列番号1又は配列番号107に示されるアミノ酸配列において、1又は2個のアミノ酸が欠失、置換又は付加されたアミノ酸配列であって、該アミノ酸配列からなるペプチドがビフィドバクテリウム・ロンガムにおいてシグナルペプチドとして機能する、前記アミノ酸配列; - シグナルペプチド-リンカー連結体が、配列番号3又は配列番号109に示されるアミノ酸配列からなることを特徴とする請求項3記載のDNA。
- 配列番号4又は配列番号110で示される塩基配列からなる請求項4記載のDNA。
- 請求項1~5のいずれか記載のDNAの3’末端に、抗体遺伝子の5’末端が結合したDNAインサート。
- 抗体遺伝子が、抗がん作用を有する抗体の遺伝子である請求項6記載のDNAインサート。
- 抗がん作用を有する抗体がトラスツズマブである請求項7記載のDNAインサート。
- トラスツズマブが、トラスツズマブ一本鎖抗体である請求項8記載のDNAインサート。
- 請求項6~9のいずれか記載のDNAインサートが挿入されたベクター。
- 請求項10記載のベクターで形質転換された腸内細菌。
- ビフィドバクテリウム属に属する微生物である請求項11記載の腸内細菌。
- ビフィドバクテリウム属に属する微生物が、ビフィドバクテリウム・ロンガムである請求項12記載の腸内細菌。
- 請求項10~13のいずれか記載の腸内細菌を有効成分とする抗体医薬組成物。
- 抗がん剤組成物であることを特徴とする請求項14記載の抗体医薬組成物。
