また、糖鎖は多くの毒素、ウィルス及びバクテリアなどの受容体であり、また、癌のマーカーとしても注目されており、こちらの分野においても、同様に新たな医薬や医療材料を創製しようとする試みが続けられている。
生理活性物質を固定化する方法としては2通りの方法が実施されている。その一つは蛋白質の物理的吸着による固定化の方法である。この方法では、蛋白質を固定化した後に2次抗体の非特異的吸着を防止するため、吸着防止剤のコーティングが行われているが、これら吸着防止剤の非特異的吸着防止能は十分でない。また1次抗体を固定化した後に吸着防止剤をコーティングするため、固定化した蛋白質の上にコーティングされてしまい、バイオチップと2次抗体とが反応できないという問題があった。このため、1次抗体の固定化後、吸着防止剤をコーティングすることなく、生理活性物質の非特異的吸着量の少ないバイオチップが求められている。
前述の特許文献3に開示された方法では支持体上で低分子成分の硬化が進行するため、支持体がプラスチック基板の場合には反りや変形が起きる恐れがあった。また、網の目状に絡み合ったマトリックスが形成されるため、生理活性物質を固定化するための官能基の反応が抑制されたり、固定化した生理活性物質の機能発現の再現性が悪いなどの問題があった。また、マトリックス内部に入り込んだタンパク質が洗浄し難い為に非特異吸着を十分に抑制できないことがあった。

1実施形態において、本発明は、一つのエチレン性二重結合及びアルキレングリコール残基を有するエチレン系不飽和重合性モノマー(a)に由来する繰り返し単位、一つのエチレン性二重結合及び生理活性物質を固定化する官能基を有するエチレン系不飽和重合性モノマー(b)に由来する繰り返し単位、並びに、一つのエチレン性二重結合及び架橋可能な官能基を有するエチレン系不飽和重合性モノマー(c)に由来する繰り返し単位を有する共重合体(A)と、前記モノマー(a)に由来する繰り返し単位及び前記モノマー(b)に由来する繰り返し単位を有し、共重合体の少なくとも片側の末端に反応性官能基を有する共重合体(B)と、を含み、固相基体の表面を被覆するために用いられるコート剤組成物を提供する。
本発明において、「アルキレングリコール残基」とは、アルキレングリコール(HO-R-OH、ここでRはアルキレン基)の片側端末又は両端末の水酸基が他の化合物と縮合反応した後に残る「アルキレンオキシ基」(-R-O-、ここでRアルキレン基)を意味する。例えば、メチレングリコール(HO-CH2-OH)の「アルキレングリコール残基」はメチレンオキシ基(-CH2-O-)であり、エチレングリコール(HO-CH2-CH2-OH)の「アルキレングリコール残基」はエチレンオキシ基(-CH2-CH2-O-)である。
共重合体(A)及び(B)において使用されるモノマー(a)は同じ種類のものでも、異なるものでも、どちらを用いても良い。
一つのエチレン性二重結合及び前記基板と架橋可能な官能基を有するエチレン系不飽和重合性モノマー(c)から与えられる構成単位は、架橋可能な官能基を有するエチレン系不飽和重合性モノマーを重合することによって与えられることが好ましい。
1実施形態において、本発明は、表面が被覆された固相基体の製造方法を提供する。本実施形態に係る固相基体の製造方法は、(i)溶媒(有機溶剤)に上述した共重合体(A)及び(B)を0.05~10重量%濃度になるように溶解した共重合体溶液を調製し、(ii)該共重合体溶液を浸漬、吹きつけ等の公知の方法で固相基体表面に塗布した後、(iii)塗布した溶液を室温下ないしは加温下にて乾燥させる工程を備える。なお、本明細書において「乾燥させる」とは、上記の溶媒を除去することを意味する。
本発明に使用するバイオセンサー用基板(固相基体)の素材は、ガラス、プラスチック、金属その他を用いることができるが、表面処理の容易性、量産性の観点から、プラスチックが好ましく、中でも熱可塑性樹脂がより好ましい。また、固相基体の形態は、例えば板状であってもよく、膜状であってもよく、ビーズ状であってもよい。
1実施形態において、本発明は、上述した共重合体(A)と溶媒とを含む第1のコート剤組成物を、固相基体の表面に塗布する工程と、前記固相基体に塗布された前記第1のコート剤組成物から前記溶媒を除去し、前記共重合体(A)で表面が被覆された固相基体を得る工程と、上述した共重合体(B)と溶媒とを含む第2のコート剤組成物を、前記共重合体(A)で表面が被覆された固相基体の表面に塗布する工程と、前記固相基体に塗布された前記第2のコート剤組成物から前記溶媒を除去する工程と、を備える、表面が被覆された固相基体の製造方法を提供する。
1実施形態において、本発明は、それぞれ別の容器に収納された、上述した共重合体(A)、及び共重合体(B)を備え、固相基体の表面を被覆するために用いられる、コート剤キットを提供する。
1実施形態において、本発明は、上述した、表面が被覆された固相基体に生理活性物質を固定化させたことを特徴とするバイオセンサーを提供する。また、1実施形態において、本発明は、上述した、表面が被覆された固相基体に生理活性物質を固定化する工程を備える、バイオセンサーの製造方法を提供する。
0.01molのポリエチレングリコールモノメタクリレート(Blenmer PE-200(n=4)、日本油脂(株)製)を20mLのクロロホルムに溶解させた後、-30℃まで冷却した。-30℃に保ちながらこの溶液に、予め作成しておいた0.01molのp-ニトロフェニルクロロフォーメート(Aldrich製)と0.01molのトリエチルアミン(和光純薬(株)製)及びクロロホルム20mLの均一溶液をゆっくりと滴下した。-30℃にて1時間反応させた後、さらに2時間溶液を攪拌した。その後反応液から塩をろ過により除去し、溶媒を留去してp-ニトロフェニルオキシカルボニル-ポリエチレングリコールメタクリレート(MEONP)を得た。得られたモノマーを重クロロホルム溶媒中1H―NMRで測定し、エチレングリコール残基が4.5単位含まれていることを確認した。
[合成例(A-1)]
ポリエチレングリコールメチルエーテルメタクリレート(別名メトキシポリエチレングリコールメタクリレート)(PEGMA、数平均分子量Mn=468、新中村化学株式会社製)、MEONP、3-メタクリロキシプロピルジメチルエトキシシラン(MPDES、GELEST,INC.製)をそれぞれ順に0.90mol/L、0.05mol/L、0.05mol/L、になるように脱水エタノールに溶解させ、モノマー混合溶液を作製した。そこにさらに0.002mol/Lの2,2-アゾビスイソブチロニトリル(AIBN、和光純薬(株)製)を添加し、均一になるまで該モノマー混合溶液を撹拌した。その後、アルゴンガス雰囲気下、60℃で4時間反応させた後、反応溶液をジエチルエーテル中に滴下し、沈殿を収集した。得られた共重合体(A-1)を重クロロホルム溶媒中1H―NMRで測定し、0.7ppm付近に現れるMPDESのSiに結合したメチレンに帰属されるピーク、3.4ppm付近に現れるPEGMAの末端メトキシ基に帰属されるピーク、7.4ppm及び8.3ppm付近に現れるMEONPのベンゼン環に帰属されるピーク、それぞれの積分値より、この共重合体(A-1)の組成比を算出した。表1に結果を示した。
[合成例(B-1)]
ポリエチレングリコールメチルエーテルメタクリレート(別名メトキシポリエチレングリコールメタクリレート)(PEGMA、数平均分子量Mn=468、新中村化学株式会社製)、MEONPを2-ブタノンに溶解させ、モノマー混合溶液を作製した。総モノマー濃度は0.7mol/L、それぞれのモル比はPEGMA、MEONPの順に50:50である。そこにさらに(3-メルカプトプロピル)ジメチルメトキシシラン(以下MPDMSと記載、Aldrich社製)を0.015mol/Lになるように、及び2、2-アゾビスイソブチロニトリル(以下AIBNと記載、和光純薬工業株式会社製)を0.05mol/Lになるように添加し、均一になるまで撹拌した。その後、アルゴンガス雰囲気下、60℃で24時間反応させた後、反応溶液をヘキサンとアセトンの混合溶媒(混合比は体積比で4:1)中に滴下し、沈殿を収集した。得られた共重合体(B-1)を重クロロホルム溶媒中1H―NMRで測定し、3.4ppm付近に現れるPEGMAの末端メトキシ基に帰属されるピーク、7.6及び8.4ppm付近に現れるMEONPのベンゼン環に帰属されるピーク、0.7ppm付近に現れるMPDMSのSiに結合したメチレンに帰属されるピーク、それぞれの積分値より、この共重合体(B-1)の組成比を算出した。表1に結果を示した。
ポリエチレングリコールメチルエーテルメタクリレート(別名メトキシポリエチレングリコールメタクリレート)(PEGMA、数平均分子量Mn=468、新中村化学株式会社製)、MEONPをエタノール/2-ブタノン=9/1混合溶媒に溶解させ、モノマー混合溶液を作製した。総モノマー濃度は0.7mol/L、それぞれのモル比はPEGMA、MEONPの順に80:20である。そこにさらに(3-メルカプトプロピル)ジメチルメトキシシラン(以下MPDMSと記載、Aldrich社製)を0.015mol/Lになるように、及び2、2-アゾビスイソブチロニトリル(以下AIBNと記載、和光純薬工業株式会社製)を0.02mol/Lになるように添加し、均一になるまで撹拌した。その後、アルゴンガス雰囲気下、60℃で24時間反応させた後、反応溶液をヘキサンとアセトンの混合溶媒(混合比は体積比で4:1)中に滴下し、沈殿を収集した。得られた共重合体(B-2)を重クロロホルム溶媒中1H―NMRで測定し、3.4ppm付近に現れるPEGMAの末端メトキシ基に帰属されるピーク、7.6及び8.4ppm付近に現れるMEONPのベンゼン環に帰属されるピーク、0.7ppm付近に現れるMPDMSのSiに結合したメチレンに帰属されるピーク、それぞれの積分値より、この共重合体(B-2)の組成比を算出した。表1に結果を示した。
飽和環状ポリオレフィン樹脂(5-メチル-2-ノルボルネンの開環重合体の水素添加物、MFR(Melt flow rate):21g/10分、水素添加率:実質的に100%、熱変形温度:123℃)を用い、射出成形により、スライドガラス形状(寸法:76mm×26mm×1mm)に加工して固相基板を作成した。酸素雰囲気下のプラズマ処理によって基板表面に酸化処理を施した。この固相基板を前述の共重合体の合成例(A-1)及び共重合体の合成例(B-1)にて得られた高分子化合物(共重合体(A-1)及び(B-1)の比率は1:1)の0.3重量%メチルエチルケトン混合溶液に浸漬することにより、基板表面に前記2種類の共重合体を含む層を導入した。この基板を100℃にて72時間加熱乾燥することにより、基板と該高分子を含む層とを化学的に結合させ、実施例1の基板を得た。

(アルデヒド基板)
飽和環状ポリオレフィン樹脂を(5-メチル-2ノルボルネンの開環重合体の水素添加物(MFR(Melt flow rate):21g/10分、水素添加率:実質的に100%、熱変形温度:123℃)を用い、射出成形によりスライドガラス形状(寸法:76mm×26mm×1mm)に加工して固相基板を作製した。酸素雰囲気下のプラズマ処理によって基板表面に酸化処理を施した。この基板を2体積%の3-アミノプロピルトリメトキシシランのエタノール溶液に浸漬した後、純水洗浄し、45℃にて2時間熱処理することによりアミノ基を導入し、さらに1体積%のグルタルアルデヒド水溶液に浸漬した後、純水洗浄することでアルデヒド基を導入し、比較例1の基板を得た。

実施例1~3及び比較例1~3の各基板について、以下のように評価を行った。
還元末端がアミノ化された糖鎖を0.3mol/Lのリン酸バッファ(pH8.5)で200μmol/Lの濃度になるように希釈した。自動スポッターを用いて、実施例及び比較例で得られた各基板にこの希釈液をスポット後、室温の環境下で1時間静置することによりアミノ化糖鎖を固定化した。
その後、実施例、比較例2及び3の各基板を0.1mol/Lのエタノールアミン(和光純薬工業(株)製、鹿特級)と0.1mol/Lのトリスバッファ(SIGMA社製)とからなる水溶液(pH9.5)に1時間浸漬することにより残りの活性エステル部を失活させた。
また、比較例1の基板には、希釈剤としてPBSバッファ(日水製薬(株)製、純水1L中に9.6gの組成培養用ダルベッコPBS(-)を溶解したバッファ)を用いて市販の吸着防止剤であるブロックエース(大日本製薬(株)製)を4倍希釈した溶液に2時間浸漬することにより吸着防止処理を施した。
ウシ血清アルブミン(以下BSAと記載、SIGMA社製)をPBSバッファで3%の濃度になるように溶解し、これにポリオキシエチレン(20)ソルビタンモノラウレート(和光純薬工業(株)製、Tween20相当、以下便宜上Tween20と記載)を1%の濃度になるように希釈した。この溶液を用いて市販血清(Gemini Bio-Products社製)を含有IgG抗体濃度が1mg/mLになるように希釈した。この血清希釈溶液を37℃にて90分、工程2で得た基板と接触させることにより血清中の抗体と糖鎖との反応を実施した。反応後、0.1%Tween20含有PBSで1回、0.001%Tween20含有PBSで3回洗浄した。
BSAをPBSバッファで3%の濃度になるように溶解し、これにTween20を0.1%の濃度になるように希釈した。この溶液を用いてビオチン標識抗IgG抗体(Thermo Scientific社製)を10μg/mLになるように希釈した。この希釈溶液を25℃にて45分、工程3で得た基板と接触させることにより血清中の抗体との反応を実施した。反応後、0.1%Tween20含有PBSで1回、0.001%Tween20含有PBSで3回洗浄した。
Cy5-ストレプトアビジン(GE Healthcare社製)を0.1%Tween20含有PBSで2μg/mLになるように希釈した。この希釈溶液を25℃にて30分、工程4で得た基板と接触させることにより蛍光標識反応を実施した。反応後、0.1%Tween20含有PBSで1回、0.001%Tween20含有PBSで3回、純水で1回洗浄し、遠心分離機を用いて基板を遠心乾燥させた。
