(i)アドレノメデュリンのアミノ酸配列からなるペプチド、
(ii)アドレノメデュリンのアミノ酸配列からなり、且つ該アミノ酸配列中の2個のシステイン残基がジスルフィド結合を形成しているペプチド、
(iii)(ii)のペプチドにおいて、前記ジスルフィド結合が、エチレン基によって置換されており、且つアドレノメデュリン活性を有するペプチド、
(iv)(i)~(iii)のいずれかのペプチドにおいて、1~15個のアミノ酸が欠失、置換若しくは付加されており、且つアドレノメデュリン活性を有するペプチド、
(v)(i)~(iv)のいずれかのペプチドにおいて、C末端がアミド化されているペプチド、並びに

本発明は、式(I):
A-Ln-B (I)
で表される化合物若しくはその塩、又はそれらの水和物に関する。本明細書において、式(I)で表される化合物を、「アドレノメデュリン誘導体」と記載する場合がある。
(2)腎臓・水電解質系:利尿作用、ナトリウム利尿作用、抗利尿ホルモン抑制作用、アルドステロン低下作用、腎保護作用(例えば、高血圧又は虚血再灌流障害における心筋保護作用)、飲水行動抑制作用、及び食塩要求抑制作用。
(3)脳・神経系:神経保護・脳障害抑制作用、抗炎症作用、アポトーシス抑制作用(例えば、虚血再灌流障害又は炎症におけるアポトーシス抑制作用)、自動調節能維持作用、及び酸化ストレス抑制作用。
(4)泌尿生殖器:勃起改善作用、血流改善作用、及び着床促進作用。
(5)消化器系:抗潰瘍作用、組織修復作用、粘膜新生作用、血流改善作用、抗炎症作用、及び肝機能改善作用。
(6)整形外科系:骨芽細胞刺激作用、及び関節炎改善作用。
(7)内分泌代謝系:脂肪細胞分化作用、脂肪分解制御作用、インスリン感受性改善作用、及びインスリン分泌制御作用。
(8)その他:循環改善作用、抗炎症作用、サイトカイン制御作用、臓器保護作用、酸化ストレス抑制作用、組織修復作用(例えば、抗褥瘡作用)、敗血症性ショックの改善作用、多臓器不全の抑制作用、及び自己免疫疾患の抑制作用。

Bが、アドレノメデュリン又はアドレノメデュリン活性を有するその修飾体から誘導されるペプチド部分であり、
但し、ペプチド部分Bが、そのN末端アミノ基を介して修飾基A又は連結基Lと結合されている。
本発明の式(I)で表される化合物は、生体内において、親分子であるアドレノメデュリンと実質的に略同等の生物活性(すなわちアドレノメデュリン活性)を、持続的に発現することができる。それ故、本発明は、本発明の式(I)で表される化合物を有効成分として含有する医薬に関する。
(2)腎臓・水電解質系疾患:腎不全、及び腎炎。
(3)脳・神経疾患:脳梗塞、及び脳炎。
(4)泌尿生殖器疾患:勃起不全(ED)。
(5)消化器疾患:炎症性腸疾患、潰瘍性疾患、腸管ベーチェット、及び肝不全。
(6)整形外科疾患:関節炎。
(7)内分泌代謝疾患:糖尿病及び糖尿病による臓器障害。
(8)その他:敗血症性ショック、自己免疫疾患、多臓器不全、及び褥瘡。
本発明はまた、本発明の式(I)で表される化合物の製造方法に関する。
本発明の方法は、アドレノメデュリン又はその修飾体から誘導されるペプチド部分Bの前駆体、パルミトイル基及びポリエチレングリコール基からなる群より選択される修飾基Aの前駆体及び2価基Lnの前駆体の少なくともいずれかを準備する工程を含んでもよい。
本発明の方法は、アドレノメデュリン又はその修飾体から誘導されるペプチド部分Bの前駆体と、パルミトイル基及びポリエチレングリコール基からなる群より選択される修飾基Aの前駆体と、2価基Lnの前駆体とを連結させて、式(I)で表される化合物を得る、連結工程を含むことが必要である。
下記のスキームに従い、アドレノメデュリン(AM)から誘導される基Bの前駆体を合成した。ヒトアドレノメデュリンの1~52アミノ酸残基に対応するペプチド(h.AM(1-52))を、6個のセグメント(Seg-1~Seg-6)に分割した。各セグメントを合成した後、それぞれのセグメントを縮合して、基Bの前駆体ペプチドを合成した。その後、基Bの前駆体ペプチドに、パルミトイル基(Pal)又はポリエチレングリコール基(PEG)を連結して、アドレノメデュリン誘導体を合成した。
Boc-Gln-Gly-OPac
Boc-Gly-OPac(88.0 g)を、トリフルオロ酢酸に溶解した。得られた溶液を、冷却下で10分間、その後、室温で50分間攪拌した。反応溶液から、トリフルオロ酢酸を減圧留去し、塩化水素のジオキサン溶液(5.5 mol/L)(65.5 mL)を添加した。得られた混合物に、ジエチルエーテルを加えた。析出した沈殿を濾取し、ジエチルエーテルで洗浄した。得られた塩酸塩を、N,N-ジメチルホルムアミドに溶解した。得られた溶液に、Boc-Gln-OH(81.3 g)及び1-ヒドロキシベンゾトリアゾール(48.7 g)を添加した。この混合物に、冷却下で、1-エチル-3-(3-ジメチルアミノプロピル)カルボジイミド(65.9 mL)を滴下した。反応溶液を、室温で3時間攪拌した。N,N-ジメチルホルムアミドを減圧留去した。酢酸エチルを加えて、得られた溶液を希釈した。この溶液を、5%炭酸水素ナトリウム水溶液、0.5 N塩酸水及び飽和食塩水にて洗浄した。有機層を、MgSO4にて乾燥した後、酢酸エチルを減圧留去した。残渣にジエチルエーテルを加えた。析出した沈殿を濾取し、ジエチルエーテルで洗浄した。得られた結晶を、クロロホルム/メタノール混合液に溶解した。溶媒を減圧留去した。ジエチルエーテルを加えた。析出した沈殿を濾取し、ジエチルエーテルで洗浄した。前記操作により、56 gの目的物を得た。
Boc-Gln-Gly-OPac(54.8 g)を、トリフルオロ酢酸に溶解した。得られた溶液を、冷却下で10分間、その後、室温で50分間攪拌した。反応溶液から、トリフルオロ酢酸を減圧留去し、塩化水素のジオキサン溶液(5.5 mol/L)(28 mL)を添加した。得られた混合物に、ジエチルエーテルを加えた。析出した沈殿を濾取し、ジエチルエーテルで洗浄した。得られた塩酸塩を、N,N-ジメチルホルムアミドに溶解した。得られた溶液に、Boc-Phe-OH(35.2 g)及び1-ヒドロキシベンゾトリアゾール(19.3 g)を添加した。この混合物に、冷却下で、1-エチル-3-(3-ジメチルアミノプロピル)カルボジイミド(26.2 mL)を滴下した。反応溶液を、室温で4時間攪拌した。反応溶液に、水を加えた。析出した沈殿を濾取し、水及びジエチルエーテルで洗浄した。得られた結晶を、クロロホルム/メタノール混合液に溶解した。溶媒を減圧留去した。ジエチルエーテルを加えた。析出した沈殿を濾取し、ジエチルエーテルで洗浄した。前記操作により、68.7 gの目的物を得た。
Boc-Phe-Gln-Gly-OPac(67.9 g)を、トリフルオロ酢酸に溶解した。得られた溶液を、冷却下で10分間、その後、室温で50分間攪拌した。反応溶液から、トリフルオロ酢酸を減圧留去し、塩化水素のジオキサン溶液(5.5 mol/L)(26 mL)を添加した。得られた混合物に、ジエチルエーテルを加えた。析出した沈殿を濾取し、ジエチルエーテルで洗浄した。得られた塩酸塩を、N,N-ジメチルホルムアミドに溶解した。得られた溶液に、Boc-Asn-OH(30.5 g)及び1-ヒドロキシベンゾトリアゾール(19.4 g)を添加した。この混合物に、冷却下で、1-エチル-3-(3-ジメチルアミノプロピル)カルボジイミド(21.8 mL)を滴下した。反応溶液を、室温で終夜攪拌した。反応溶液に、水を加えた。析出した沈殿を濾取し、水及びジエチルエーテルで洗浄した。得られた結晶を、N,N-ジメチルホルムアミドに溶解した。溶媒を減圧留去した。ジエチルエーテルを加えた。析出した沈殿を濾取し、ジエチルエーテルで洗浄した。前記操作により、81 gの目的物を得た。
Boc-Asn-Phe-Gln-Gly-OPac(81 g)を、トリフルオロ酢酸に溶解した。得られた溶液を、冷却下で10分間、その後、室温で50分間攪拌した。反応溶液から、トリフルオロ酢酸を減圧留去し、塩化水素のジオキサン溶液(5.5 mol/L)(26 mL)を添加した。得られた混合物に、ジエチルエーテルを加えた。析出した沈殿を濾取し、ジエチルエーテルで洗浄した。得られた塩酸塩を、N,N-ジメチルホルムアミド/1,3-ジメチル-2-イミダゾリジノン混合液に溶解した。得られた溶液に、Boc-Asn-OH(28.9 g)及び1-ヒドロキシベンゾトリアゾール(19.2 g)を添加した。この混合物に、冷却下で、1-エチル-3-(3-ジメチルアミノプロピル)カルボジイミド(26.0 mL)を滴下した。反応溶液を、室温で終夜攪拌した。反応溶液に、水を加えた。析出した沈殿を濾取し、水、アセトニトリル及びジエチルエーテルで洗浄した。前記操作により、74 gの目的物を得た。
Boc-Asn-Asn-Phe-Gln-Gly-OPac(50.0 g)を、トリフルオロ酢酸に溶解した。得られた溶液を、冷却下で10分間、その後、室温で50分間攪拌した。反応溶液から、トリフルオロ酢酸を減圧留去し、塩化水素のジオキサン溶液(5.5 mol/L)(13.7 mL)を添加した。得られた混合物に、ジエチルエーテルを加えた。析出した沈殿を濾取し、ジエチルエーテルで洗浄した。得られた塩酸塩を、N,N-ジメチルホルムアミド/1-メチル-2-ピロリドン/1,3-ジメチル-2-イミダゾリジノン混合液に溶解した。得られた溶液に、Boc-Met-OH(16.4 g)及び1-ヒドロキシベンゾトリアゾール(9.4 g)を添加した。この混合物に、冷却下で、1-エチル-3-(3-ジメチルアミノプロピル)カルボジイミド(12.6 mL)を滴下した。反応溶液を、室温で終夜攪拌した。反応溶液に、水を加えた。析出した沈殿を濾取し、水、アセトニトリル及びジエチルエーテルで洗浄した。前記操作により、54 gの目的物を得た。

Boc-Gln-Ser(Bzl)-Met-Asn-Asn-Phe-Gln-Gly-OPac(16.2 g)を、トリフルオロ酢酸に溶解した。得られた溶液を、冷却下で10分間、その後、室温で50分間攪拌した。反応溶液から、トリフルオロ酢酸を減圧留去し、塩化水素のジオキサン溶液(5.5 mol/L)(2.9 mL)を添加した。得られた混合物に、ジエチルエーテルを加えた。析出した沈殿を濾取し、ジエチルエーテルで洗浄した。得られた塩酸塩を、ジメチルスルホキシド/1-メチル-2-ピロリドン混合液に溶解した。得られた溶液に、Boc-Arg(Tos)-OH(7.4 g)及び1-ヒドロキシベンゾトリアゾール(2.1 g)を添加した。この混合物に、冷却下で、1-エチル-3-(3-ジメチルアミノプロピル)カルボジイミド(2.9 mL)を滴下した。反応溶液を、室温で終夜攪拌した。反応溶液に、水を加えた。析出した沈殿を濾取し、水、アセトニトリル及びジエチルエーテルで洗浄した。前記操作により、12.5 gの目的物を得た。
Boc-Arg(Tos)-Gln-Ser(Bzl)-Met-Asn-Asn-Phe-Gln-Gly-OPac(12.5 g)を、トリフルオロ酢酸に溶解した。得られた溶液を、冷却下で10分間、その後、室温で50分間攪拌した。反応溶液から、トリフルオロ酢酸を減圧留去し、塩化水素のジオキサン溶液(5.5 mol/L)(1.7 mL)を添加した。得られた混合物に、ジエチルエーテルを加えた。析出した沈殿を濾取し、ジエチルエーテルで洗浄した。得られた塩酸塩を、ジメチルスルホキシド/1-メチル-2-ピロリドン混合液に溶解した。得られた溶液に、Boc-Tyr(BrZ)-OH(4.4 g)及び1-ヒドロキシベンゾトリアゾール(1.5 g)を添加した。この混合物に、冷却下で、1-エチル-3-(3-ジメチルアミノプロピル)カルボジイミド(1.7 mL)を滴下した。反応溶液を、室温で終夜攪拌した。反応溶液に、アセトニトリルを加えた。析出した沈殿を濾取し、アセトニトリル及びジエチルエーテルで洗浄した。前記操作により、11 gの目的物を得た。
Boc-Arg(Tos)-Gln-Ser(Bzl)-Met-Asn-Asn-Phe-Gln-Gly-OPac(6.0 g)を、ジメチルスルホキシドに溶解した。得られた溶液に、30℃でギ酸アンモニウム水溶液(10 mmol/L)(15.6 mL)、酢酸(18.7 mL)及び亜鉛末(10.2 g)を添加した。1時間後、反応溶液から亜鉛末を濾過して除き、ジメチルスルホキシドで洗浄した。反応溶液に、水を加えた。析出した沈殿を濾取し、水、アセトニトリル及びジエチルエーテルで洗浄した。前記操作により、5.5 gの目的物を得た。
Boc-Phe-Gly-OPac
Boc-Gly-OPac(44 g)を、トリフルオロ酢酸に溶解した。得られた溶液を、冷却下で10分間、その後、室温で50分間攪拌した。反応溶液から、トリフルオロ酢酸を減圧留去し、塩化水素のジオキサン溶液(5.5 mol/L)(33 mL)を添加した。得られた混合物に、ジエチルエーテルを加えた。析出した沈殿を濾取し、ジエチルエーテルで洗浄した。得られた塩酸塩を、塩化メチレンに溶解した。得られた溶液に、Boc-Phe-OH(36.6 g)を添加した。この混合物に、冷却下で、1-エチル-3-(3-ジメチルアミノプロピル)カルボジイミド(27.5 mL)を滴下した。反応溶液を、室温で終夜攪拌した。反応溶液を、0.5 N塩酸水及び飽和食塩水にて洗浄した。有機層を、MgSO4にて乾燥した後、塩化メチレンを減圧留去した。残渣にジエチルエーテルを加えた。析出した沈殿を濾取し、ジエチルエーテルで洗浄した。得られた結晶を、クロロホルムに溶解した。溶媒を減圧留去した。ジエチルエーテルを加えた。析出した沈殿を濾取し、ジエチルエーテルで洗浄した。前記操作により、38 gの目的物を得た。

Boc-Arg(Tos)-Ser(Bzl)-Phe-Gly-OPac(15.7 g)を、トリフルオロ酢酸に溶解した。得られた溶液を、冷却下で10分間、その後、室温で50分間攪拌した。反応溶液から、トリフルオロ酢酸を減圧留去し、塩化水素のジオキサン溶液(5.5 mol/L)(4 mL)を添加した。得られた混合物に、ジエチルエーテルを加えた。析出した沈殿を濾取し、ジエチルエーテルで洗浄した。得られた塩酸塩を、N,N-ジメチルホルムアミドに溶解した。得られた溶液に、Boc-Leu-OH・H2O(4.6 g)及び1-ヒドロキシベンゾトリアゾール(2.6 g)を添加した。この混合物に、冷却下で、1-エチル-3-(3-ジメチルアミノプロピル)カルボジイミド(3.6 mL)を滴下した。反応溶液を、室温で4時間攪拌した。反応溶液に、水を加えた。析出した沈殿を濾取し、水で洗浄した。得られた結晶を、クロロホルム/メタノール混合液に溶解した。溶媒を減圧留去した。ジエチルエーテルを加えた。析出した沈殿を濾取し、ジエチルエーテルで洗浄した。前記操作により、17.6 gの目的物を得た。
Boc-Leu-Arg(Tos)-Ser(Bzl)-Phe-Gly-OPac(17.6 g)を、酢酸に溶解した。得られた溶液に、40℃で亜鉛末(55.2 g)を添加した。2時間後、反応溶液から亜鉛末を濾過して除き、酢酸で洗浄した。酢酸を減圧留去した後、酢酸エチルにて溶液を希釈した。得られた溶液を、0.1 N塩酸水及び飽和食塩水にて洗浄した。有機層を、MgSO4にて乾燥した後、酢酸エチルを減圧留去した。残渣にジエチルエーテルを加えた。析出した沈殿を濾取し、ジエチルエーテルで洗浄した。得られた結晶を、クロロホルムに溶解した。溶媒を減圧留去した。ジエチルエーテルを加えた。析出した沈殿を濾取し、ジエチルエーテルで洗浄した。前記操作により、13.9 gの目的物を得た。
Boc-Phe-Gly-OPac(8.8 g)を、トリフルオロ酢酸に溶解した。得られた溶液を、冷却下で10分間、その後、室温で50分間攪拌した。反応溶液から、トリフルオロ酢酸を減圧留去し、塩化水素のジオキサン溶液(5.5 mol/L)(4.0 mL)を添加した。得られた混合物に、ジエチルエーテルを加えた。析出した沈殿を濾取し、ジエチルエーテルで洗浄した。得られた塩酸塩を、N,N-ジメチルホルムアミドに溶解した。得られた溶液に、Boc-Arg(Tos)-OH(10.3 g)及び1-ヒドロキシベンゾトリアゾール(2.8 g)を添加した。この混合物に、冷却下で、1-エチル-3-(3-ジメチルアミノプロピル)カルボジイミド(3.8 mL)を滴下した。反応溶液を、室温で終夜攪拌した。酢酸エチルにて溶液を希釈した。得られた溶液を、5%炭酸水素ナトリウム水溶液、0.5 N塩酸水及び飽和食塩水にて洗浄した。有機層を、MgSO4にて乾燥した後、酢酸エチルを減圧留去した。残渣にジエチルエーテルを加えた。析出した沈殿を濾取し、ジエチルエーテルで洗浄した。得られた結晶を、酢酸エチルに溶解した。溶媒を減圧留去した。ジエチルエーテルを加えた。析出した沈殿を濾取し、ジエチルエーテルで洗浄した。前記操作により、14.7 gの目的物を得た。
Boc-Arg(Tos)-Phe-Gly-OPac(14.5 g)を、トリフルオロ酢酸に溶解した。得られた溶液を、冷却下で10分間、その後、室温で50分間攪拌した。反応溶液から、トリフルオロ酢酸を減圧留去し、塩化水素のジオキサン溶液(5.5 mol/L)(3.9 mL)を添加した。得られた混合物に、ジエチルエーテルを加えた。析出した沈殿を濾取し、ジエチルエーテルで洗浄した。得られた塩酸塩を、N,N-ジメチルホルムアミドに溶解した。得られた溶液に、Boc-Cys(4-MeBzl)-OH(6.9 g)及び1-ヒドロキシベンゾトリアゾール(2.8 g)を添加した。この混合物に、冷却下で、1-エチル-3-(3-ジメチルアミノプロピル)カルボジイミド(3.8 mL)を滴下した。反応溶液を、室温で3時間攪拌した。反応溶液に、水を加えた。析出した沈殿を濾取し、水及びn-ヘキサンで洗浄した。得られた結晶を、クロロホルム/メタノール混合液に溶解した。溶媒を減圧留去した。ジエチルエーテルを加えた。析出した沈殿を濾取し、ジエチルエーテルで洗浄した。前記操作により、17.2 gの目的物を得た。
Boc-Cys(4-MeBzl)-Arg(Tos)-Phe-Gly-OPac(9.1 g)を、トリフルオロ酢酸に溶解した。得られた溶液を、冷却下で10分間、その後、室温で50分間攪拌した。反応溶液から、トリフルオロ酢酸を減圧留去し、塩化水素のジオキサン溶液(5.5 mol/L)(1.9 mL)を添加した。得られた混合物に、ジエチルエーテルを加えた。析出した沈殿を濾取し、ジエチルエーテルで洗浄した。得られた塩酸塩を、N,N-ジメチルホルムアミドに溶解した。得られた溶液に、Boc-Leu-Arg(Tos)-Ser(Bzl)-Phe-Gly-OH(9.2 g)及び1-ヒドロキシベンゾトリアゾール(1.4 g)を添加した。この混合物に、冷却下で、1-エチル-3-(3-ジメチルアミノプロピル)カルボジイミド(1.8 mL)を滴下した。反応溶液を、室温で終夜攪拌した。反応溶液に、水を加えた。析出した沈殿を濾取し、水及びジエチルエーテルで洗浄した。得られた結晶を、N,N-ジメチルホルムアミドに溶解した。溶媒を減圧留去した。アセトニトリルを加えた。析出した沈殿を濾取し、アセトニトリル及びジエチルエーテルで洗浄した。前記操作により、16.2 gの目的物を得た。

Boc-Leu-Ala-OPac

Boc-Tyr(BrZ)-Gln-OPac

Boc-His-Gln-Ile-Tyr(BrZ)-Gly-OPac (15.0 g)を、塩化メチレン/2,2,2-トリフルオロエタノール/酢酸混合液に溶解した。得られた溶液に、40℃で亜鉛末(43.9 g)を添加した。30分後、反応溶液から亜鉛末を濾過して除き、塩化メチレン/2,2,2-トリフルオロエタノール混合液で洗浄した。溶媒を減圧留去し、残渣に水を加えた。析出した沈殿を濾取し、水、塩酸水、水、ジエチルエーテル、アセトニトリル及びジエチルエーテルで洗浄した。得られた結晶を、N,N-ジメチルホルムアミドに溶解した。溶媒を減圧留去した。ジエチルエーテルを加えた。析出した沈殿を濾取し、ジエチルエーテルで洗浄した。前記操作により、9.5 gの目的物を得た。
Boc-Ala-Pro-OPac
Boc-Ala-OH(95 g)、Pro-OBzl・HCl(127 g)及び1-ヒドロキシベンゾトリアゾール(7 g)を塩化メチレンに添加した。得られた溶液に、冷却下で、1-エチル-3-(3-ジメチルアミノプロピル)カルボジイミド(96 mL)を滴下した。反応溶液を、室温で終夜攪拌した。塩化メチレンを減圧留去し、残渣を酢酸エチルにて希釈した。有機層を、5%炭酸水素ナトリウム水溶液、0.5 N塩酸水及び飽和食塩水にて洗浄した。酢酸エチルを減圧留去し、残渣にn-ヘキサンを加えた。析出した沈殿を濾取し、n-ヘキサンで洗浄した。前記操作により、158 gの結晶を得た。27.1 gの前記結晶をメタノールに溶解した。得られた溶液に、5% Pd-C(5 g)を添加した。反応溶液に水素ガスを添加した。反応終了後、5% Pd-Cを濾過して除いた。メタノールを減圧留去した。ジエチルエーテルを加えた。析出した沈殿を濾取し、ジエチルエーテルで洗浄した。前記操作により、19.7 gの結晶を得た。19.7 gの前記結晶をN,N-ジメチルホルムアミドに溶解した。得られた溶液に、フェナシルブロマイド(14.4 g)及びトリエチルアミン(10.1 mL)を添加した。反応溶液を、1時間撹拌した。反応溶液を、酢酸エチルにて希釈した。得られた溶液を、5%炭酸水素ナトリウム水溶液、0.5 N塩酸水及び飽和食塩水にて洗浄した。有機層を、MgSO4にて乾燥した後、酢酸エチルを減圧留去した。n-ヘキサンを加えた。析出した沈殿を濾取し、n-ヘキサンで洗浄した。前記操作により、25.2 gの目的物を得た。

Pal-h.AM(1-52) トリフルオロ酢酸塩

Pal-h.AM(1-52)酢酸塩
Pal-h.AM(1-52) トリフルオロ酢酸塩(38 mg)を、5%酢酸水に溶解した。得られた溶液を、ムロマック 1×2(酢酸型)カラム(3 mL)に通液し、5%酢酸水で流出させた。流出液を凍結乾燥した。目的物は、凍結乾燥の後に、32 mgの白色粉末として得られた。
Pal-h.AM(1-52)酢酸塩 バイアル詰め
Pal-h.AM(1-52) 酢酸塩(16.5 mg)を、ミリQ水(18.3 mL)に溶解した。得られた溶液を、0.30 mLずつバイアル瓶に分注し、凍結乾燥した。得られた凍結乾燥物のうち、3本を、6 M塩酸水にて加水分解した。得られた加水分解物を、アミノ酸分析した。前記アミノ酸分析により、内容量を定量した。 収量:38 nmol×57本。

PEG5000-h.AM(1-52) トリフルオロ酢酸塩
PEG5000-Tyr-Arg-Gln-Ser-Met-Asn-Asn-Phe-Gln-Gly-Leu-Arg-Ser-Phe-Gly-Cys-Arg-Phe-Gly-Thr-Cys-Thr-Val-Gln-Lys(Boc)-Leu-Ala-His-Gln-Ile-Tyr-Gln-Phe-Thr-Asp-Lys(Boc)-Asp-Lys(Boc)-Asp-Asn-Val-Ala-Pro-Arg-Ser-Lys(Boc)-Ile-Ser-Pro-Gln-Gly-Tyr-NH2 (Cys16-Cys21ジスルフィド架橋体)(34 mg)に、氷冷下で、95%トリフルオロ酢酸水(5 mL)を加えて、粗ペプチドを溶解した。得られた混合物を、氷冷下で40分間反応させた。反応溶液から、溶媒を減圧留去した。残渣をミリQ水に溶解した。得られた溶液から、目的物を逆相HPLCにて精製した。目的物は、凍結乾燥の後に、30 mgの白色粉末として得られた。
PEG5000-h.AM(1-52) 酢酸塩 バイアル詰め
PEG5000-h.AM(1-52) トリフルオロ酢酸塩(30 mg)を、3%酢酸水に溶解した。得られた溶液を、ムロマック 1×2(酢酸型)カラム(2 mL)に通液し、3%酢酸水で流出させた。流出液を凍結乾燥した。凍結乾燥物の全量を、ミリQ水(18.3 mL)に溶解した。得られた溶液を、0.30 mLずつバイアル瓶に分注し、凍結乾燥した。得られた凍結乾燥物のうち、3本を、6 M塩酸水にて加水分解した。得られた加水分解物を、アミノ酸分析した。前記アミノ酸分析により、内容量を定量した。 収量:37 nmol×56本。
[II-1:アドレノメデュリン誘導体による細胞内cAMP濃度上昇作用]
アドレノメデュリン(AM)の投与により、細胞内cAMPの濃度が上昇する。このため、AMの作用は、細胞内cAMPの濃度上昇を介して発現すると考えられる。そこで、AM受容体を発現させた培養細胞株(HEK293細胞株)に、Pal-h.AM(1-52)、PEG5000-h.AM(1-52)又はh.AM(1-52)を添加して、細胞内cAMPの産生量を測定した。コンフルエントのHEK293細胞(細胞数:5×104)に、0.5 mMのIBMXの存在下、10-11~10-6 mol/LのPal-h.AM(1-52)、PEG5000-h.AM(1-52)又はh.AM(1-52)を添加して、15分間インキュベートした。その後、cAMP測定用ELISAキット(GEヘルスケアー、#RPN2251)を用いて、各試験区のHEK293細胞における細胞内cAMP濃度を測定した。Pal-h.AM(1-52)、PEG5000-h.AM(1-52)又はh.AM(1-52)の濃度とcAMP濃度の上昇との用量応答曲線を図1に示す。なお、図中、縦軸は、h.AM(1-52)によるcAMP濃度上昇の最大応答値に対する、各試験区のcAMP濃度上昇の相対値(%)を示す。
麻酔下ラットの静脈内に、Pal-h.AM(1-52)、PEG5000-h.AM(1-52)又はh.AM(1-52)を単回投与して、アドレノメデュリン誘導体の血中濃度の経時変化を観察した。11~14週齢の雄性ウイスターラットを、イソフルランの吸入により麻酔導入した。気管切開の後、1.5~2.5%のイソフルラン濃度及び0.6~0.8 L/分の流量にて、吸入麻酔管理を行った。前記ラットから、右頸静脈を単離し、26G相当のカテーテルチューブを挿入した。次に、前記処置後のラットから、左頸動脈を単離し、23G相当のカテーテルチューブを挿入した。生理食塩水に溶解したPal-h.AM(1-52)、PEG5000-h.AM(1-52)又はh.AM(1-52)を、右頸静脈のカテーテルチューブより投与した。頸動脈に挿入したカテーテルより、投与開始時から経時的に、300 μlの採血を行った。得られた血液検体に、直ちにEDTA-2Na(300 μg)及びアプロチニン(21 μg)を添加して、さらに3000回転、10分間の条件で遠心分離して、血漿を得た。各検体の血漿中AM濃度を、化学発光酵素免疫法にて測定した。2-コンパートメントモデルを用いて、血漿中AM濃度の測定結果から、第一相及び第二相の血中半減期を算出した。h.AM(1-52)、Pal-h.AM(1-52)又はPEG5000-h.AM(1-52)の投与開始時からの経過時間と血漿中AM濃度との関係を図2~4にそれぞれ示す。
麻酔下ラットの静脈内に、Pal-h.AM(1-52)、PEG5000-h.AM(1-52)又はh.AM(1-52)を単回投与して、該ラットの血圧の経過を観察した。11~14週齢の雄性ウイスターラットを、イソフルランの吸入により麻酔導入した。気管切開の後、1.5~2.5%のイソフルラン濃度及び0.6~0.8 L/分の流量にて、吸入麻酔管理を行った。前記ラットから、右頸静脈を単離し、26G相当のカテーテルチューブを挿入した。次に、前記処置後のラットから、左頸動脈を単離し、23G相当のカテーテルチューブを挿入した。生理食塩水に溶解したPal-h.AM(1-52)、PEG5000-h.AM(1-52)又はh.AM(1-52)を、右頸静脈のカテーテルチューブより投与した。右頸静脈のカテーテルチューブより、生理食塩水ヘパリン溶液(生理食塩水:100ml;ヘパリン:1000単位)を2.4 ml/時間で補液した。同じカテーテルチューブより、10 nmol/kgのPal-h.AM(1-52)、PEG5000-h.AM(1-52)又はh.AM(1-52)を、生理食塩水に溶解した形態で投与した。頸動脈に挿入したカテーテルを、圧トランスデューサーに接続した。Pal-h.AM(1-52)、PEG5000-h.AM(1-52)又はh.AM(1-52)の投与前の血圧と投与後の血圧とを、経時的に測定した。h.AM(1-52)、Pal-h.AM(1-52)又はPEG5000-h.AM(1-52)の投与開始時からの経過時間と平均血圧との関係を図5に示す。なお、図中、縦軸は、各薬剤投与時の平均血圧から、各薬剤投与前の平均血圧を差し引いた差を示す。
II-4-1:モデルの作製
C57BL/6J Jclマウス(6週齢)に対し、イソフルラン(2%)吸入麻酔下で背部皮膚を切開した。投与液を充填した浸透圧ポンプ(1002型Alzet(登録商標)ミニ浸透圧ポンプ)を皮下に埋め込み、皮膚を縫合糸で縫合した。浸透圧ポンプ埋め込み部位の背部皮下は、刈毛し消毒した。処置後のマウスに、3質量/体積%デキストラン硫酸ナトリウム(Sodium Dextran Sulfate、DSS)(和光純薬工業株式会社)を飲用水として7日間摂取させ、炎症性腸疾患モデルを作製した。DSS誘発性腸炎は、炎症性腸疾患のモデルとして広く用いられている。
(1)AM濃度
前記手順に従って採取したヘパリン加血液を、4℃、1800×g、10分間の条件で遠心分離して、血漿を得た。得られた各検体中の血漿中AM濃度を、化学発光酵素免疫法にて測定した。
投与開始後3、5、7、10及び14日目に、対象マウスの体重を測定した。
投与開始後3、5、7、10及び14日目における、対象マウスの体重及び便の性状を、以下のスコア判定の基準に基づき評価した。得られたスコア値を合計して、各検体の総スコア値を算出した。
解剖摘出した腸管を縦に開き、脂肪組織を除去した後、生理食塩液で洗浄した。次に、腸管の長さを測定した。
(1)AM濃度
前記手順で測定した血漿中AM濃度を、平均値±標準誤差で表す。対照群、並びに0.02、0.1及び0.5 nmol/kg/hr PEG5000-h.AM(1-52)投与群の血漿中AM濃度は、それぞれ、0.091±0.036、1.430±0.242、3.289±0.525及び12.96±2.432 fmol/mlであった。対照群、並びに0.02、0.1及び0.5 nmol/kg/hr PEG5000-h.AM(1-52)投与群の血漿中AM濃度を図6に示す。
前記手順で評価した総スコアを、中央値[平均値 ± 標準誤差]で表す。投与開始後3、5、7、10及び14日目における対照群の総スコアは、それぞれ0.0[0.6±0.3]、4.0[3.7±0.4]、6.0[5.4±0.5]、3.5[4.1±0.4]及び0.0[0.0±0.0]であった。投与開始後3、5、7、10及び14日目における、0.02 nmol/kg/hr PEG5000-h.AM(1-52)投与群の総スコアは、それぞれ1.0[0.8±0.2]、2.5[2.8±0.5]、5.5[5.7±0.4]、2.5[3.1±0.8]及び0.0[0.0±0.0]であった。投与開始後3、5、7、10及び14日目における、0.1 nmol/kg/hr PEG5000-h.AM(1-52)投与群の総スコアは、それぞれ0.0[0.7±0.3]、1.5[1.9±0.5]、5.0[4.1±0.4]、2.0[2.0±0.5]及び0.0[0.0±0.0]であった。投与開始後3、5、7、10及び14日目における、0.5 nmol/kg/hr PEG5000-h.AM(1-52)投与群の総スコアは、それぞれ1.0[1.3±0.3]、1.5[1.1±0.3]、3.0[3.0±0.3]、2.0[2.1±0.4]及び0.0[0.0±0.0]であった。PEG5000-h.AM(1-52)の投与開始時からの経過時間と、対照群及び各投与群の総スコアとの関係を図7に示す。図7に示すように、0.1 nmol/kg/hr PEG5000-h.AM(1-52)投与群の投与開始後10日目における総スコア値、並びに0.5 nmol/kg/hr PEG5000-h.AM(1-52)投与群の投与開始後5、7及び10日目における総スコア値は、対照群の総スコア値と比較していずれも有意な減少が認められた(Steelの多重比較検定、p<0.05又はP<0.01)。
前記手順で測定した腸管の長さを、平均値 ± 標準誤差で表す。対照群、並びに0.02、0.1及び0.5 nmol/kg/hr PEG5000-h.AM(1-52)投与群の腸管の長さは、それぞれ65.0±1.3、69.4±1.5、71.4±0.9及び72.4±0.9 mmであった。対照群、並びに0.02、0.1及び0.5 nmol/kg/hr PEG5000-h.AM(1-52)投与群の腸管の長さを図8に示す。図8に示すように、PEG5000-h.AM(1-52)投与群の腸管の長さは、全ての濃度処理群において、対照群の腸管の長さと比較していずれも有意な差が認められた(Dunnettの多重比較検定、P<0.05又はP<0.01)。
Claims (7)
- 式(I):
A-Ln-B (I)
[式中、
Aは、パルミトイル基及びポリエチレングリコール基からなる群より選択される修飾基であり、
Lは、2価の連結基であり、
nは、0又は1の整数であり、
Bは、アドレノメデュリン又はアドレノメデュリン活性を有するその修飾体から誘導されるペプチド部分であり、
但し、ペプチド部分Bは、そのN末端アミノ基を介して修飾基A又は連結基Lと結合されている。]
で表される化合物若しくはその塩、又はそれらの水和物。 - 前記アドレノメデュリン又はアドレノメデュリン活性を有するその修飾体が、下記:
(i)アドレノメデュリンのアミノ酸配列からなるペプチド、
(ii)アドレノメデュリンのアミノ酸配列からなり、且つ該アミノ酸配列中の2個のシステイン残基がジスルフィド結合を形成しているペプチド、
(iii)(ii)のペプチドにおいて、前記ジスルフィド結合が、エチレン基によって置換されており、且つアドレノメデュリン活性を有するペプチド、
(iv)(i)~(iii)のいずれかのペプチドにおいて、1~15個のアミノ酸が欠失、置換若しくは付加されており、且つアドレノメデュリン活性を有するペプチド、
(v)(i)~(iv)のいずれかのペプチドにおいて、C末端がアミド化されているペプチド、並びに
(vi)(i)~(iv)のいずれかのペプチドにおいて、C末端にグリシン残基が付加されているペプチド
からなる群より選択されるペプチドである、請求項1に記載の化合物。 - 前記アドレノメデュリン又はその修飾体が、下記:
(a)配列番号1のアミノ酸配列からなるペプチド、又は配列番号1のアミノ酸配列からなり、且つ16位のシステイン残基と21位のシステイン残基とがジスルフィド結合を形成しているペプチド;
(b)配列番号3のアミノ酸配列からなるペプチド、又は配列番号3のアミノ酸配列からなり、且つ16位のシステイン残基と21位のシステイン残基とがジスルフィド結合を形成しているペプチド;
(c)配列番号5のアミノ酸配列からなるペプチド、又は配列番号5のアミノ酸配列からなり、且つ16位のシステイン残基と21位のシステイン残基とがジスルフィド結合を形成しているペプチド;
(d)配列番号7のアミノ酸配列からなるペプチド、又は配列番号7のアミノ酸配列からなり、且つ16位のシステイン残基と21位のシステイン残基とがジスルフィド結合を形成しているペプチド;
(e)配列番号9のアミノ酸配列からなるペプチド、又は配列番号9のアミノ酸配列からなり、且つ14位のシステイン残基と19位のシステイン残基とがジスルフィド結合を形成しているペプチド;
(f)配列番号11のアミノ酸配列からなるペプチド、又は配列番号11のアミノ酸配列からなり、且つ14位のシステイン残基と19位のシステイン残基とがジスルフィド結合を形成しているペプチド;
(g)前記(a)~(f)のいずれかのペプチドにおいて、前記ジスルフィド結合が、エチレン基によって置換されており、且つアドレノメデュリン活性を有するペプチド;
(h)前記(a)~(g)のいずれかのペプチドにおいて、1~15個のアミノ酸残基が欠失、置換若しくは付加されており、且つアドレノメデュリン活性を有するペプチド;
(i)前記(a)~(h)のいずれかのペプチドにおいて、C末端がアミド化されているペプチド;並びに
(j)前記(a)~(h)のいずれかのペプチドにおいて、C末端にグリシン残基が付加されているペプチド;
からなる群より選択されるペプチドである、請求項1又は2に記載の化合物。 - アドレノメデュリン又はその修飾体から誘導されるペプチド部分Bの前駆体と、パルミトイル基及びポリエチレングリコール基からなる群より選択される修飾基Aの前駆体と、2価基Lnの前駆体とを連結させて、式(I)で表される化合物を得る、連結工程を含む、請求項1~3のいずれか1項に記載の化合物の製造方法。
- 請求項1~3のいずれか1項に記載の化合物を有効成分として含有する医薬。
- 循環器疾患、炎症性疾患又は末梢血管疾患の予防又は治療に使用するための、請求項5に記載の医薬。
- 請求項1~3のいずれか1項に記載の化合物を有効成分として含有する、循環器疾患、炎症性疾患又は末梢血管疾患の予防又は治療剤。
