 養殖試験は、試験区としてスッポン用養殖池2面を用いた。また、養殖試験中の水温は、スッポンが最も生育効率がよいとされる約30℃となるように温度を調節し加温養殖を行った。評価対象のスッポンとして、佐賀産、糸田産の養殖スッポンを養殖した。また、餌を与えた環境のスッポンは30g~出荷サイズ(約1kg)までのスッポンであり、従来の餌により養殖されている環境のスッポンに与えていた餌を、実施例1の餌に切り替えて養殖を行った。なお、スッポン用養殖池は、稚亀(約20~30g頃)からおよそ300gまでは40~50匹/m2となるように養殖し、およそ300g~800g(出荷サイズ)までは8~10匹/m2となるように養殖するように使い分けた。また、養殖池はため池式のものであり、適宜、水質を維持するために水の入れ替えを行った。
