眼血管新生に起因する眼科疾患の別の好ましい態様は、滲出型加齢黄斑変性症である。

一般式(1)の化合物の医薬上許容される塩としては、塩酸塩、臭化水素酸塩、カリウム塩等が挙げられ、好ましい例として、臭化水素酸塩が挙げられる。
20.76kgの2-{[(2R)-4-(3,4-ジクロロベンジル)モルホリン-2-イル]メチル}-1H-イソインドール-1,3-(2H)-ジオンを158.4kgのエタノールに溶解させ、4.82kgのヒドラジンを6分間かけて滴下し、(2S)-2-アミノメチル-4-(3,4-ジクロロベンジル)モルホリンが得られた。この(2S)-2-アミノメチル-4-(3,4-ジクロロベンジル)モルホリンに108kgのトルエンを加えた後、全量60Lになるまで濃縮し、そこに246.2kgのエタノールを加えた。ここに、4.62kgのシュウ酸を32.8kgのエタノールに溶解させた溶液を1時間8分かけて滴下し、15.14kgの(2S)-2-アミノメチル-4-(3,4-ジクロロベンジル)モルホリン・シュウ酸塩を得た。
8.18kgの4-エトキシカルボニルメチル-2-メルカプトチアゾールを22.80kgのt-ブタノールと25.72kgの水の混合溶媒に溶解し、13.2kgの8N水酸化ナトリウムを15分間かけて滴下し、その後1時間撹拌した。ここに上記合成例1で得られた(2S)-N-{[4-(3,4-ジクロロベンジル)モルホリン-2-イル]メチル}クロロアセトアミドのt-ブチルメチルエーテル溶液の全量を加え、(2S)-[4-(カルボキシメチル)チアゾール-2-イルチオ]-N-{[4-(3,4-ジクロロベンジル)モルホリン-2-イル]メチル}アセトアミドが得られた。この(2S)-[4-(カルボキシメチル)チアゾール-2-イルチオ]-N-{[4-(3,4-ジクロロベンジル)モルホリン-2-イル]メチル}アセトアミドに68.4kgのt-ブタノールを加えた後、全量44Lになるまで濃縮し、これに81.0kgのアセトニトリルを加えた。ここに6.8kgの臭化水素酸を33分間かけて滴下し撹拌後、7.35kgのt-ブタノールを5分かけて添加したところ、添加後15分で(2S)-[4-(カルボキシメチル)チアゾール-2-イルチオ]-N-{[4-(3,4-ジクロロベンジル)モルホリン-2-イル]メチル}アセトアミド 臭化水素酸塩の結晶の析出が始まり、最終的に17.16kgの結晶が回収された。
レーザー照射によって実験的に脈絡膜血管新生(以下「CNV」という)を誘導するマウスモデル(CNVモデル)を用いて、CNVの面積に対する有効性を評価した。CNVモデルはレーザー照射による炎症の惹起でマクロファージが遊走される。その結果としてVEGFが産生され、新生血管が発生することから、CNVモデルはVEGFを介する眼血管新生を伴う疾患に対する有効性を示す実験モデルである。
(2S)-[4-(カルボキシメチル)チアゾール-2-イルチオ]-N-{[4-(3,4-ジクロロベンジル)モルホリン-2-イル]メチル}アセトアミド 臭化水素酸塩を被験化合物として以下の実験を行った。
雄性のC57BL/6Jマウス(7週齢)の片眼にレーザーを照射(6スポット照射)して実験的にCNVを誘導した。マウスは塩酸ケタミン/塩酸キシラジン(1:1、v/v)混合溶液を大腿筋肉内に投与(1mL/kg)し、全身麻酔を施した後、散瞳点眼剤で散瞳させ、網膜の太い毛細血管を避けて6箇所にレーザー照射(波長532 nm、スポットサイズ50 μm、照射時間0.1秒、レーザー出力120 mW)を行った。コントロール抗体および抗マウスVEGF抗体(R&D Systems社より購入)は、レーザー照射直後に1回だけ硝子体内に(2μL/眼)投与した。被験化合物は直近の体重をもとに、調製した被験液を0.1 mL/10 g(媒体:0.5%ヒドロキシプロピルメチルセルロース)となるように、ゾンデを用いて経口投与した。なお、被験液はレーザー照射後に試験動物の麻酔が十分に覚めた事を確認して、レーザー誘発日のみ1日1回経口投与し、レーザー照射の翌日から最終日の前日までは、1日2回反復経口投与した。例数は14匹/群とし、表1に記載の通り、1群あたり10例以上の例数を確保した。
レーザー照射7日後のCNV面積を図1に示す。抗マウスVEGF抗体の硝子体内投与群では、コントロール抗体の硝子体内投与群と比較して、有意なCNV抑制作用が認められた。また、被験化合物の経口投与群では、媒体のみの経口投与群と比較して、有意なCNV抑制作用が認められた。これらの結果から、レーザー照射により実験的に誘導したCNVには、抗マウスVEGF抗体が有意に抑制したことから、VEGFが関与していることが示された。また、本モデルのCNVに反復経口投与で被験化合物が有意な抑制作用を示したことから、被験化合物に代表されるモルホリン化合物がCNVを阻害することを示している。
(2S)-[4-(カルボキシメチル)チアゾール-2-イルチオ]-N-{[4-(3,4-ジクロロベンジル)モルホリン-2-イル]メチル}アセトアミド 臭化水素酸塩を被験化合物として、ヒトCCR3のリガンド(例えば、ヒトCCL11、ヒトCCL24およびヒトCCL26など)によって誘発されるヒト血管内皮細胞の遊走に対する阻害作用を評価する。
血管新生が起こる過程において血管内皮細胞の遊走が起こると考えられており、血管内皮細胞の遊走を阻害する化合物は血管新生に起因する疾患、例えば眼血管新生に起因する眼科疾患の治療剤として有用であると考えられる。
