マイクロパターン化温度応答性表面を作製するオリジナルの手順は、以前に報告されている(Biomacromolecules 12, 1414-1418 (2011))。具体的には、PIPAAmブラシを表面開始可逆的付加開裂連鎖移動(RAFT)重合法(Biomacromolecules 11, 1991-1999 (2010))によってガラス基材上に形成し、次にポジ型フォトレジスト(OFPR‐800 LB、34cp)(東京応化工業)を、PIPAAmブラシ表面上に、スピンコーターACT‐300D(ACTIVE)で8,000rpmにて30秒間スピンコートした。フォトマスク(ストライプ幅:50μm/50μm)を用いて紫外線を照射した後、照射領域でフォトレジストを現像液(2.38%テトラメチルアンモニウムヒドロキシド溶液)(NMD‐3)(東京応化工業)を用いて選択的に取り除き、さらなる重合を防ぐために、PIPAAmブラシの露わになった反応性末端DTB基を、不活性なマレイミド基に変換した。残留フォトレジストをアセトンで取り除いた後、親水性ポリマーPAcMoを、DTBを末端にもつPIPAAmブラシ領域上に、空間選択的に重合して、PAcMo‐b‐PIPAAmブロックポリマーブラシおよびPIPAAmブラシ領域(50μm/50μmストライプ)から成るマイクロパターンを生じた(図1a左上)。対照として、表面にPAcMoのマイクロパターンを有しない、PIPAAmポリマーがグラフトされたガラス基材を用意した(図1a左下)。
ヒト骨格筋の筋芽細胞を、組織培養ポリスチレン(TCPS)ディッシュ上で5%CO2、37℃にて骨格筋の筋芽細胞の生育培地(SkGM‐2:Lonza)中で培養した。上記で作製した、パターン化または非パターン化PIPAAmグラフトガラス基材(20x20mm)を、TCPSディッシュ(直径35mm)に置き、筋芽細胞(継代:<5)を、該ポリマーブラシ表面上に5×104細胞/cm2の密度で播種した。コンフルエントに達するまで、接着細胞を5%CO2、37℃にて培養し、顕微鏡ECLIPSE TE2000‐U(ニコン、東京、日本)を用いて位相差顕微鏡での観察を行った。
筋芽細胞を分化誘導培地(2%ウマ血清を補充したDMEM/F12 1:1)で5日間培養することによって筋管形成が達成された。細胞培養用の全試薬は、ロンザ(Lonza)(Walkersville、メリーランド州、米国)から入手した。
ゼラチンゲルをPIPAAmグラフト表面上に作られた筋芽細胞シート上に置き、それらを共に30分間28℃でインキュベーションして、ゼラチンゲルを筋芽細胞シートに接着させた。次に、それらを30分間20℃でインキュベーションして、基材表面から筋芽細胞シートを剥離させた。ゼラチンゲルとともに細胞シートを新しいTCPSディッシュ上に移動し、ディッシュ中で37℃にて培地中でさらにインキュベーションし、ゼラチンゲルを溶かした。
細胞シートの積層については、採取した第2の細胞シートをもう一方の細胞シート上に移動した。2層をなした細胞シートを28℃にて30分間インキュベーションし、2枚の細胞シートを互いに接着させ、次に培養温度(20℃)を下げることによって、それらのシートを一緒に採取した。新しいTCPSディッシュまたはガラス基材上に移動した後、さらにそれらを28℃で30分間インキュベーションし、37℃にて30分間インキュベーションすることによってゼラチンゲルを溶かした。
コンフルエントな細胞を、リン酸緩衝食塩水(PBS)中の4%パラホルムアルデヒドで15分間37℃にて固定し、リン酸緩衝食塩水中の0.5%トリトンX‐100で5分間、室温(RT)にて透過処理をした。固定した細胞を、リン酸緩衝食塩水中の2%ウシ血清アルブミン(BSA)で30分間、RTにてブロックした。リン酸緩衝食塩水で3回洗浄した後、細胞を、アレクサフルオル(AlexaFluor)568結合ファロイジン(インビトロジェン社、カールズバッド、カリフォルニア州、米国)(BSA‐リン酸緩衝食塩水中で1:200の希釈)で1時間、RTにてインキュベーションした。MHC染色については、フルオレセイン結合マウス抗ヒトMHC抗体(1:50)(R&Dシステム、ミネアポリス、ミネソタ州、米国)で、1時間、RTにて細胞を処理した。フィブロネクチンを観察するために、細胞シートを、抗フィブロネクチン抗体(Abcam、ケンブリッジ、マサチューセッツ州、米国)で処理し、次にアレクサフルオル(AlexaFluor)488(インビトロジェン社)で標識された二次抗体で処理した。共焦点レーザー走査型顕微鏡(LSM510;オーバーコッヘン(Oberkochen)、カールツァイスオーバーコッヘン(Carl Zeiss Oberkochen)、ドイツ)によって、蛍光染色した細胞の画像を取得した。
筋芽細胞の配向性を制御するために、親水性のポリ(N‐アクリロイルモルホリン)(PAcMo)を、従来のフォトリソグラフィー技術を使用して温度応答性PIPAAmブラシ表面上に空間選択的にグラフトした。結果として、2つの異なるポリマー領域のストライプパターン(50μm)が、その表面上に形成された(図1a)。
また、この自己組織化挙動は、複数の細胞シートから成るよく整列された筋管構築物を作るのに有用であった。2枚のランダム(Random)シート上に積層したとき、1枚の整列(Aligned)シートは、下部の筋芽細胞配向性を変え、最終的には3重の層をなした細胞シートにおいてすべての筋管がよく同一方向に整列された(図5b)。すなわち、1枚の整列筋芽細胞シートがあれば、複数の細胞シートから整列された筋管構築物を作るのに十分であった。
他方、3D直角指向筋管形成もまた、2枚の整列筋芽細胞シートの積層によって創出可能であった。初めに分化誘導培地中で培養して筋管を形成させ、次に整列筋管構築物を、細胞シート積層プロセスを使用して直角に積層した。筋管配向性は設計通り維持されることから、整列細胞シートを積層することによって、垂直に異なる配向性をも作ることができた(図5c)。
パターン化PIPAAmグラフトガラス基材(20x20mm)を、TCPSディッシュ(直径35mm)に置き、ヒト骨格筋の筋芽細胞(継代:<5)を、該ポリマーブラシ表面上に5×104細胞/cm2の密度で播種した。コンフルエントに達するまで、生育培地(SkGM‐2:Lonza)中で5%CO2、37℃にて培養し、配向制御型の筋芽細胞シートを作製した。次に、あらかじめCellTrackerで染色したヒト臍帯静脈由来の血管内皮細胞(HUVEC)を5x104 cells/cm2の密度で配向制御型の筋芽細胞シート上に播種した。3時間程度~1日培養してHUVECが筋芽細胞シート上に十分に接着したことを確認した後、別の配向制御型筋芽細胞シートをゼラチンゲルスタンプにより回収し、HUVECを2枚の細胞シートで挟むように積層し、血管内皮細胞増殖培地で培養した。別の配向制御型筋芽細胞シートを積層する前の蛍光像を図7左上に、積層後の蛍光像を図7左下に示す。
対照として、ランダムな配向を有する筋芽細胞シートの上にHUVECの層を形成させ(図7左下)、その上に、別のランダムな配向を有する筋芽細胞シートを作製した(図7右下)。
その結果、HUVECの細胞層を配向制御型の筋芽細胞シートでサンドイッチした場合、筋芽細胞と同一の方向に、HUVECによる血管のネットワーク構造が形成されることが分かった。
パターン化PIPAAmグラフトガラス基材(20x20mm)を、TCPSディッシュ(直径35mm)に置き、ヒト骨格筋の筋芽細胞(継代:<5)を、該ポリマーブラシ表面上に5×104細胞/cm2の密度で播種した。コンフルエントに達するまで、生育培地(SkGM‐2:Lonza)中で5%CO2、37℃にて培養し、配向制御型の筋芽細胞シートを作製した。次に、ヒトiPS細胞から分化させた神経細胞(iCell Neurons, Cellular Dynamics international社製)を1x103~2x104 cells/cm2の密度で配向制御型の筋芽細胞シート上に播種した。24時間培養して神経細胞が筋芽細胞シート上に十分に接着したことを確認した後、別の配向制御型筋芽細胞シートをゼラチンゲルスタンプにより回収し、神経細胞を2枚の細胞シートで挟むように積層し、神経細胞増殖培地で5日間培養した。その後、細胞をパラホルムアルデヒドで固定し、神経細胞マーカーのニューロフィラメントを赤に、筋芽細胞マーカーのデスミンを緑に、それぞれ抗体を用いて染色した。結果を図9左に示す。
対照として、ランダムな配向を有する筋芽細胞シートの上に神経細胞の層を形成させ、その上に、別のランダムな配向を有する筋芽細胞シートを作製した(図9右)。
その結果、神経細胞の層を配向制御型の筋芽細胞シートでサンドイッチした場合、筋芽細胞と同一の方向に、神経のネットワーク構造が形成されることが分かった。
Claims (8)
- 筋芽細胞を含む細胞シートを複数積層させてなる、細胞シート積層体であって、各細胞シートは制御された配向性の筋芽細胞を含む細胞群を含有することを特徴とする、細胞シート積層体。
- 各細胞シートにおける筋芽細胞を含む細胞群の配向が互いに同一である領域を有する、請求項1に記載の細胞シート積層体。
- 前記細胞シート積層体は、筋芽細胞を含む細胞群を含有する細胞シート間に、筋芽細胞を含む細胞群と配向が同一の血管内皮細胞又は神経細胞を含む、請求項2に記載の細胞シート積層体。
- 各筋芽細胞を含む細胞群を含有する細胞シートが、温度応答性ポリマー被覆基材上で筋芽細胞を培養し、基材から剥離することにより得られたものである、請求項1~3のいずれか一項に記載の細胞シート積層体。
- 各細胞シートにおける、筋芽細胞の割合が50%以上である、請求項1~4のいずれか一項に記載の細胞シート積層体。
- 制御された配向性を有する第1の筋芽細胞を含む細胞群を含有する細胞シートに、ランダムな配向性を有する第2の筋芽細胞を含む細胞群を含有する細胞シートを重ねる工程を含む、各細胞シートにおける筋芽細胞を含む細胞群の配向が互いに同一である領域を有する、細胞シート積層体の製造方法。
- 制御された配向性を有する第1の筋芽細胞を含む細胞群を含有する細胞シート上に、血管内皮細胞又は神経細胞の層を形成させ、その上に第2の筋芽細胞を含む細胞シートを重ねる工程を含む、各細胞シートにおける筋芽細胞を含む細胞群と血管内皮細胞又は神経細胞の配向が互いに同一である領域を有する、筋芽細胞および血管内皮細胞又は神経細胞を含む細胞シート積層体の製造方法。
- 制御された配向性を有する第1の筋芽細胞を含む細胞群を含有する細胞シートの下に、第1の筋芽細胞を含む細胞シートとは異なる方向の配向性を有する第2の筋芽細胞を含む細胞群を含有する細胞シートを配置する工程を含む、各細胞シートにおける筋芽細胞を含む細胞群の配向が互いに同一である領域を有する、細胞シート積層体の製造方法。
