Description
プルロニックハイドロゲルシート(第1の基材)の製造
ハイドロゲルシートを製造するため、ゼラチンモールドを準備し、モールドの底は、マイクロパターンを形成した。約25℃の雰囲気で、前記ゼラチンモールド内に約10wt%のプルロニックf-127(Sigma)水溶液を入れた。そして、製造されるハイドロゲルシートの上面を平らにするためにニトロセルロース膜で覆い、これを固定するためのプラスチックメッシュ及びガラス板を積層した。その後、温度を約37℃に上昇させてモールド内のプルロニックf-127(Sigma)水溶液のみを硬化させてハイドロゲル化した。以後、約37℃のwashing buffer(Saline、PBS、ddH2O)を用いてゼラチンモールドを溶かす方法で除去し、プルロニックハイドロゲルシートを製造した。
ハイドロゲルシートを製造するため、ゼラチンモールドを準備し、モールドの底は、マイクロパターンを形成した。約25℃の雰囲気で、pH6-7のMESバッファーに約2wt%の含量でアルジネートが含まれたアルジネート溶液を前記ゼラチンモールド内に入れた。そして、製造されるハイドロゲルシートの上面を平らにするため、ニトロセルロース膜で覆い、これを固定するためのプラスチックメッシュ及びガラス板を積層した。その後、アルジネート架橋水溶液を適加してモールド内のアルジネート溶液をハイドロゲル化した。以後、約37℃のwashing buffer(Saline、PBS、ddH2O)を用いてゼラチンモールドを溶かす方法で除去してアルジネートハイドロゲルシートを製造した。
約37℃の雰囲気で、前記製造されたプルロニックハイドロゲルシートのパターンが形成された面上にC2C12骨格筋細胞を培養して細胞層を形成し、アルジネートハイドロゲルシートの一面上にC2C12骨格筋細胞を培養して細胞層を形成した。その後、細胞層が接するようにした後、温度を約10℃に下げてプルロニックハイドロゲルシートを溶かして除去し、2-スタックの細胞シートを製造した。その後、細胞層の活性のために再び温度を約37℃に上昇させた。
Claims (10)
- (a)0℃~30℃のいずれかの地点の温度で溶解点を有するか、疎水性から親水性に変換される第1の基材上に第1の細胞層を形成する段階と、
(b)酵素によって分解される第2の基材上に第2の細胞層を形成する段階と、
(c)前記第1の細胞層と前記第2の細胞層を接触させる段階と、
(d)前記第1の基材の溶解点以下の温度または前記第1の基材が親水性に変換される温度を提供し、前記第1の基材を選択的に除去する段階と、
(d’)露出した前記第1の細胞層と追加の第1の基材上に備えられた追加の第1の細胞層を接触させた後、前記追加の第1の基材の溶解点以下の温度または前記追加の第1の基材が親水性に変換される温度を提供し、前記追加の第1の基材を選択的に除去する段階と、
(e)酵素を含む溶液に前記第2の基材を接触させて前記第2の基材を選択的に除去する段階と、を含み、
(d)段階、(d’)段階及び(e)段階が順次行われ、
(d’)段階は、2回以上繰り返して行われ、前記第1の細胞層上に複数の追加の第1の細胞層を形成する、多層細胞シートの製造方法。 - (d)段階は、前記第1の基材の溶解点以下の温度または前記第1の基材が親水性に変換される温度を持つ溶液に前記第1の基材を接触させるか、または周辺温度を前記第1の基材の溶解点以下の温度または前記第1の基材が親水性に変換される温度に下げることを特徴とする、請求項1に記載の多層細胞シートの製造方法。
- 前記第1の細胞層及び前記第2の細胞層は、それぞれ前記第1の基材及び前記第2の基材上に培養されて形成されることを特徴とする、請求項1に記載の多層細胞シートの製造方法。
- (a)段階及び(c)段階は、35℃~40℃の雰囲気で行われることを特徴とする、 請求項1に記載の多層細胞シートの製造方法。
- 前記第1の基材は、ポリホスファゼン系ハイドロゲル、プルロニック系ハイドロゲル及びポリ(N-イソプロピルアクリルアミド)のうち少なくとも1種を含み、
前記プルロニック系ハイドロゲルは、ポリエチレンオキシド(PEO)-ポリプロピレンオキシド(PPO)-ポリエチレンオキシド(PEO)の構造(PEO-PPO-PEO)を有する高分子を利用する材料である、請求項1に記載の多層細胞シートの製造方法。 - 前記第1の基材は、ポリ(N-イソプロピルアクリルアミド)で表面処理されたことを特徴とする、請求項1に記載の多層細胞シートの製造方法。
- 前記第2の基材は、カルボキシメチルセルロースまたはアルジネートを含むハイドロゲル基材であることを特徴とする、請求項1に記載の多層細胞シートの製造方法。
- 前記カルボキシメチルセルロースまたはアルジネートは、チラミンでコンジュゲーションされたことを特徴とする、請求項7に記載の多層細胞シートの製造方法。
- 前記酵素は、カルボキシメチルセルロース分解酵素またはアルジネート分解酵素であることを特徴とする、請求項1に記載の多層細胞シートの製造方法。
- 前記第1の細胞層及び前記第2の細胞層は、それぞれ間葉系幹細胞(MSCs;mesenchymal stem cells)、筋組織前駆細胞(myocyte precursor cells)、筋細胞(myocytes)、繊維芽細胞(fibroblasts)、軟骨細胞(chondrocytes)、内皮細胞(endothelial cells)、上皮細胞(epithelial cells)、胚性幹細胞(ESCs;embryonic stem cells)、造血幹細胞(hematopoietic stem cells)、付着依存性細胞前駆体(anchorage-dependent cell precursors)、誘導された全分化能幹細胞(iPSCs;induced pluripotent stem cells)、及び心筋細胞(cardiomyocytes)からなる群から選ばれる細胞を含むことを特徴とする、請求項1に記載の多層細胞シートの製造方法。
