<실험예 1> 티로시나아제 억제 활성 실험 Experimental Example 1 Tyrosinase Inhibitory Activity Experiment
0.1M potassium phosphate buffer (pH 6.5) 220 ㎕에 농도별로 희석한 시료 20 ㎕를 첨가 후 1250 unit/ml mushroom tyrosinase 20 ul를 첨가한다. 기질 1.5 mM L-tyrosine 40 ㎕씩 첨가하여 잘 섞은 후 37℃에서 10분 반응 후 490nm에서 ELISA reader를 이용하여 흡광도를 측정하고 아래의 식에 따라 시료의 처리 농도에 따른 저해율을 구한 다음 IC50 값을 구해 아래의 [표 1]에 나타내었다. 양성대조군으로 Arbutin을 사용하였다.After adding 20 μl of diluted sample to 220 μl of 0.1M potassium phosphate buffer (pH 6.5), add 20 μl of 1250 unit / ml mushroom tyrosinase. 상기 [표 1]에서 확인되는 바와 같이, 기질인 L-tyrosine과 효소인 mushroom tyrosinase에 의해 산화되어 생성되는 멜라닌의 흡광도를 측정하여 멜라닌 생성에 중요한 역할을 하는 효소인 tyrosinase의 억제 활성 측정 결과, 실시예의 산유자 시료는 Arbutin보다는 활성이 낮지만 tyrosinase 저해 활성을 가짐을 보여준다.As shown in Table 1 above, by measuring the absorbance of melanin oxidized by the substrate L-tyrosine and the enzyme mushroom tyrosinase, measurement of the inhibitory activity of tyrosinase, an enzyme that plays an important role in melanogenesis, was carried out.
