本発明者らは、その様な作用をもつ食品成分の中で、異なる作用メカニズムを有することが知られているローズヒップ抽出物とカフェインに着目した。ローズヒップ抽出物は、体重低下作用や体脂肪率低下作用、肝臓中の中性脂質減少作用があると報告されている(特許文献1)。さらに、その作用をもたらす主要成分はチリロサイドであり、呼吸商の低下や脂質燃焼に関与するPPARα遺伝子の発現量を上昇させることが報告されている(非特許文献1、2)。一方、カフェインは、血中カテコールアミンレベルの増加作用およびホスホジエステラーゼ活性の阻害作用を介して脂質代謝を促進し(非特許文献3)、肝臓の脂肪合成を抑制し、さらに、安静時代謝率やエネルギー消費量を増加することにより(非特許文献4)、体脂肪量や体重を減少させることが知られている。
このように、各成分が単独で脂質代謝に作用があることは公知である。しかし、これらの成分を組み合わせた報告はない。

肥満を予防または解消するため、摂取カロリーを制限する食事療法、運動療法、食欲抑制剤による薬物療法などが行われている。しかし、食事療法は過度な制限食であることが多く、煩雑なカロリー計算と強固な意志を必要とし、個人で長期間管理するには困難が伴う。また、運動療法は精神的にも肉体的にも苦痛を伴い、多忙な現代社会においては長期間継続するのは非常に困難である。運動エネルギー源として体脂肪をできるだけ多く消費することは有効であるが、肥満傾向にあるものは脂肪代謝が緩慢であり、運動によっても体脂肪を減少させることが難しい。したがって、肝臓中での脂肪合成を抑制し、脂肪細胞中に蓄積する脂肪の分解を促進し、また、脂肪を効果的に燃焼することにより、肥満の予防や改善に寄与し得る組成物およびそれを含む飲食品、医薬品の開発が望まれている。
本発明は、上記問題点に鑑みてなされたものであり、その目的はメタボリックシンドロームの予防、改善または治療用組成物の提供、および上記組成物を含有する飲食品、医薬品または飼料を提供すること等にある。
本発明者らは、体脂肪を効果的に減少させる組成物について鋭意研究を行ったところ、ローズヒップ抽出物とカフェインとを含有する組成物が、驚くべきことにそれぞれを単独で使用した場合よりも強い脂肪低減作用のあることを見出し、さらに検討を重ねて本発明を完成するに至った。
すなわち、本発明は以下の発明を包含する。

本発明の組成物を用いることによりメタボリックシンドロームの予防、改善または治療することができ、より詳しくは、脂肪の低減、体重の減少、肥満の予防、改善または治療に加え、内臓脂肪蓄積が原因とされる糖尿病、高中性脂肪血症、高コレステロール血症や動脈硬化症などの予防、改善または治療することができる。
本発明は、以下の(a)および(b)を含有し、好ましくは(a)と(b)との質量比が1000:1~1:0.1であるメタボリックシンドロームの予防、改善または治療用組成物を提供する。

(ローズヒップ抽出物)
ローズヒップ抽出物は、ローズヒップを溶媒で抽出して得られる抽出液、その希釈液、濃縮液または後述するそれらの種々の形態の乾燥物をいう。
ローズヒップ抽出物は、ローズヒップを溶媒で抽出して得られる抽出液、その希釈液、濃縮液または後述するそれらの種々の形態の乾燥物をいう。
ローズヒップは、バラ科植物ローザ(Rosa)属に属するローザ・カニーナ(R. canina)、ローザ・センチフォリア(R. centifolia)、ローザ・ルゴーサ(R. rugosa)などの花、茎、鱗茎、根、種子(偽果)、種皮、果皮、果肉等の乾燥物、破断物、破砕物、細断物、あるいはこれらを粉末化した乾燥粉末、乾燥物を粉砕後成形したものなどをいう。中でも、ローズヒップの種子(偽果)を含む果実を用いることが好ましく、ローズヒップの種子(偽果)を用いることがより好ましい。
抽出に用いる溶媒は、極性溶媒であってもよいし、非極性溶媒であってもよい。抽出に用いる溶媒は、例えば、水;メタノール、エタノール、1-プロパノール、2-プロパノール、1-ブタノール、2-ブタノール等のアルコール類;ジメチルエーテル、ジエチルエーテル、ジグリム、テトラヒドロフラン等のエーテル類;酢酸メチル、酢酸エチル等のエステル類;アセトン、メチルエチルケトン等のケトン類;アセトニトニル、プロピオニトリル等のニトリル類;ヘプタン、ヘキサン、シクロヘキサンなどの炭化水素類;ベンゼン、トルエン等の芳香族炭化水素類;および、塩化メチレン、クロロホルム等のハロゲン化脂肪族炭化水素類等が挙げられる。これらの溶媒は、単独で用いてもよいし、2種以上を組み合わせて用いてもよい。
抽出に用いる溶媒は、極性溶媒を用いることが好ましく、アルコール類、エステル類、水を単独で又は2種以上を混合して用いることがより好ましく、エタノール、1-プロパノール、2-プロパノール、1-ブタノール、2-ブタノール等の炭素数1~6の低級アルコール、酢酸エチル及び水を単独で又は2種以上を混合して用いることがさらに好ましく、エタノール及び水の混合溶液を用いることが特に好ましい。
抽出に用いる溶媒として炭素数1~6の低級エタノールと水の混合溶液を用いる場合、その混合比は特に限定されないが、例えば容量比で99.9:0.1~0.1:99.9とすることができ、95:5~5:95とすることが好ましく、80:20~20:80とすることがより好ましく、80:20~51:49とすることが特に好ましい。
抽出に用いる溶媒は、極性溶媒を用いることが好ましく、アルコール類、エステル類、水を単独で又は2種以上を混合して用いることがより好ましく、エタノール、1-プロパノール、2-プロパノール、1-ブタノール、2-ブタノール等の炭素数1~6の低級アルコール、酢酸エチル及び水を単独で又は2種以上を混合して用いることがさらに好ましく、エタノール及び水の混合溶液を用いることが特に好ましい。
抽出に用いる溶媒として炭素数1~6の低級エタノールと水の混合溶液を用いる場合、その混合比は特に限定されないが、例えば容量比で99.9:0.1~0.1:99.9とすることができ、95:5~5:95とすることが好ましく、80:20~20:80とすることがより好ましく、80:20~51:49とすることが特に好ましい。
ローズヒップの抽出方法としては特に制限はないが、抽出物の安全性の確保、製造コスト及び栄養成分の損失の低減のため、緩やかな条件で行うことが好ましい。ローズヒップの抽出は、例えば、ローズヒップの種子を含む果実を粉砕、破砕または裁断し、これに5~20倍量の溶媒を加え、0℃~溶媒の還流温度の範囲で30分~48時間、振盪、撹拌あるいは還流などの条件下で行うことができる。抽出後、濾過、遠心分離などの分離操作を行い、不溶物を除去して、抽出物を得ることができる。除去した不溶物について上記抽出操作を繰り返し、得られた抽出物に加えてもよい。
ローズヒップ抽出物はそのまま用いてもよく、必要に応じて希釈または濃縮して用いてもよい。また、液状物、濃縮物、ペースト状でのいずれの状態で用いてもよく、これらを乾燥した乾燥物として用いてもよい。
ローズヒップ抽出物の乾燥は、上記液状の、あるいは濃縮された、またはペースト状の抽出物をそのまま乾燥して粉末あるいは固形物としてもよい。乾燥は、噴霧乾燥、凍結乾燥、減圧乾燥、流動乾燥等の当業者が通常用いる方法により行うことができる。
ローズヒップ抽出物の乾燥は、上記液状の、あるいは濃縮された、またはペースト状の抽出物をそのまま乾燥して粉末あるいは固形物としてもよい。乾燥は、噴霧乾燥、凍結乾燥、減圧乾燥、流動乾燥等の当業者が通常用いる方法により行うことができる。

チリロサイド(tiliroside)は、ポリフェノールの一種である。チリロサイドは、体内において、β酸化を促す酵素を活性化する作用を有するため、体脂肪(特に内臓に蓄積された脂肪)の代謝を促進する。また、チリロサイドは呼吸商の低下や脂質燃焼に関与するPPARα遺伝子の発現量を上昇させるため、体脂肪の代謝を増大させる。さらに、チリロサイドは優れた耐糖能改善効果も発揮する。したがって、肥満等に起因する各種疾患、メタボリックシンドローム等の予防、改善および治療に優れた効果を発揮する。
チリロサイドの含有量は特に限定されないが、乾燥質量換算で0.01質量%以上、より好ましくは0.02質量%以上、さらに好ましくは0.04質量%以上含有する。
なお、本発明において「内臓脂肪」とは、腹腔内にある肝臓、腎臓、膵臓、腸などの内臓、これらの内臓の周辺(例えば、肝臓周囲、腸間膜周辺など)に蓄積された脂肪をいい、副睾丸脂肪を含む。
(キサンチン誘導体)
本発明に用いるキサンチン誘導体は特に限定されず、例えば下記式(2)

で表わされる化合物またはその塩が挙げられる。
キサンチン誘導体としては、例えば、キサンチン、アミノフィリン、テオフィリン、コリンテオフィリン、カフェイン、テオブロミン、1,7-ジメチルキサンチン、オクストリフィン、ジプロフィリン、プロキシフィリン等が挙げられる。中でもカフェイン、テオフィリン、テオブロミンが好ましく、カフェインがより好ましい。
成分(b)は、上記例示したキサンチン誘導体のいずれか一種のみを使用してもよく、二種以上を組み合わせて使用してもよい。これらのキサンチン誘導体は、公知の方法により合成したものを使用してもよく、茶葉、コーヒー豆等から抽出したものを使用してもよい。
成分(b)は、上記例示したキサンチン誘導体のいずれか一種のみを使用してもよく、二種以上を組み合わせて使用してもよい。これらのキサンチン誘導体は、公知の方法により合成したものを使用してもよく、茶葉、コーヒー豆等から抽出したものを使用してもよい。
(その他の成分)
本発明の組成物は、任意成分として中性脂肪の合成を抑制する物質(例えば、脂肪合成酵素阻害活性を有するサラシア、緑茶など)、脂肪の吸収阻害効果を有する物質(例えば、消化管リパーゼ活性を阻害するローズマリー、セイジ、ショウガ、マンゴージンジャー、キュウリなど)、脂肪細胞から血中への脂肪の遊離を促進する物質(例えば、シトラス、ハイビスカス、ニガウリ、共役リノール酸など)、アルコール吸収阻害効果を有する物質(例えば、ローレル、セイヨウトチノキ種子、タラノメなど)などを含んでもよい。これらの物質は、原料物質をそのまま用いてもよいし、濃縮、抽出、乾燥等して用いてもよい。
また、本発明の組成物は、さらにカルニチンを含んでいてもよい。
本発明の組成物は、上記物質を含有することにより、さらなる脂肪低減作用、ダイエット作用を発揮することができる。
本発明の組成物は、任意成分として中性脂肪の合成を抑制する物質(例えば、脂肪合成酵素阻害活性を有するサラシア、緑茶など)、脂肪の吸収阻害効果を有する物質(例えば、消化管リパーゼ活性を阻害するローズマリー、セイジ、ショウガ、マンゴージンジャー、キュウリなど)、脂肪細胞から血中への脂肪の遊離を促進する物質(例えば、シトラス、ハイビスカス、ニガウリ、共役リノール酸など)、アルコール吸収阻害効果を有する物質(例えば、ローレル、セイヨウトチノキ種子、タラノメなど)などを含んでもよい。これらの物質は、原料物質をそのまま用いてもよいし、濃縮、抽出、乾燥等して用いてもよい。
また、本発明の組成物は、さらにカルニチンを含んでいてもよい。
本発明の組成物は、上記物質を含有することにより、さらなる脂肪低減作用、ダイエット作用を発揮することができる。
本発明の組成物は、医薬品、飲食品、飼料、食品添加剤、飼料添加剤等として用いることができる。なかでも、医薬品、飲食品または飼料に用いることが好ましい。
本発明の組成物を含有してなる医薬品は、経口または非経口のいずれかの経路で哺乳動物に投与することができる。経口剤としては、顆粒剤、散剤、錠剤(糖衣錠を含む)、丸剤、カプセル剤、シロップ剤、乳剤、懸濁剤などが挙げられる。非経口剤としては、注射剤(例えば、皮下注射剤、静脈内注射剤、筋肉内注射剤、腹腔内注射剤等)、点滴剤、外用剤(例えば、経鼻投与製剤、経皮製剤、軟膏剤等)、坐剤(例えば、直腸坐剤、膣坐剤等)などが挙げられる。これらの製剤は、当該分野で通常行われている手法により、薬学上許容される担体を用いて製剤化することができる。薬学上許容される担体としては、賦形剤、結合剤、希釈剤、添加剤、香料、緩衝剤、増粘剤、着色剤、安定剤、乳化剤、分散剤、懸濁化剤、防腐剤等が挙げられ、具体的には、炭酸マグネシウム、ステアリン酸マグネシウム、タルク、砂糖、ラクトース、ペクチン、デキストリン、澱粉、ゼラチン、トラガント、メチルセルロース、ナトリウムカルボキシメチルセルロース、低融点ワックス、カカオバター等を用いることができる。
経口用の固形剤(例えば、錠剤、丸剤、カプセル剤、散剤、顆粒剤等)は、有効成分を賦形剤(例えば、ラクトース、マンニトール、グルコース、微結晶セルロース、デンプン等)、結合剤(例えば、ヒドロキシプロピルセルロース、ポリビニルピロリドン、メタケイ酸アルミン酸マグネシウム等)、崩壊剤(例えば、繊維素グリコール酸カルシウム等)、滑沢剤(例えば、ステアリン酸マグネシウム等)、安定剤、溶解補助剤(例えば、グルタミン酸、アスパラギン酸等)等と混合し、常法に従って製剤化することができる。必要に応じて、コーティング剤(例えば、白糖、ゼラチン、ヒドロキシプロピルセルロース、ヒドロキシプロピルメチルセルロースフタレート等)で被覆していてもよいし、また2以上の層で被覆していてもよい。
経口用の液剤(例えば、水剤、懸濁剤、乳剤、シロップ剤、エリキシル剤等)は、有効成分を一般的に用いられる希釈剤(例えば、精製水、エタノールまたはそれらの混液等)に溶解、懸濁または乳化して製剤化される。さらにこの液剤は、湿潤剤、懸濁化剤、乳化剤、甘味剤、風味剤、芳香剤、保存剤、緩衝剤等を含有していてもよい。
注射剤は、溶液、懸濁液、乳濁液、および用時溶剤に溶解または懸濁して用いる固形の注射剤を包含する。注射剤は、有効成分を溶剤に溶解、懸濁または乳化して製剤化される。溶剤として、例えば注射用蒸留水、生理食塩水、植物油、プロピレングリコール、ポリエチレングリコール、エタノールのようなアルコール類等およびそれらの組み合わせが用いられる。さらにこの注射剤は、安定剤、溶解補助剤(例えば、グルタミン酸、アスパラギン酸、ポリソルベート80(登録商標)等)、懸濁化剤、乳化剤、無痛化剤、緩衝剤、保存剤等を含んでいてもよい。これらは最終工程において滅菌するか無菌操作法によって製造される。また無菌の固形剤、例えば凍結乾燥品を製造し、その使用前に無菌化または無菌の注射用蒸留水または他の溶剤に溶解して使用することもできる。
本発明の組成物を含有してなる飲食品は、一般的に飲食品に用いられる食品添加剤、例えば甘味料、着色料、保存料、増粘安定剤、酸化防止剤、発色料、漂白料、防かび剤、ガムベース、苦味料、酵素、光沢剤、酸味料、調味料、乳化剤、強化剤、製造用剤、香料、香辛料抽出物等が添加されてもよい。なお、飲食品には、健康食品、機能性食品、特定保健用食品、乳児用食品、幼児用食品、妊産婦用食品、病者用食品が含まれる。
本発明に好適な飲食品は特に限定されない。具体的には、例えば、いわゆる栄養補助食品(サプリメント)としての錠剤、顆粒剤、散剤、ドリンク剤等を挙げることができる。これ以外には、例えば茶飲料、清涼飲料、炭酸飲料、栄養飲料、果実飲料、乳酸飲料等の飲料、そば、うどん、中華麺、即席麺等の麺類、飴、キャンディー、ガム、チョコレート、スナック菓子、ビスケット、ゼリー、ジャム、クリーム、焼き菓子、パン等の菓子およびパン類、かまぼこ、ハム、ソーセージ等の水産・畜産加工食品、加工乳、発酵乳等の乳製品、サラダ油、てんぷら油、マーガリン、マヨネーズ、ショートニング、ホイップクリーム、ドレッシング等の油脂および油脂加工食品、ソース、たれ等の調味料、カレー、シチュー、丼、お粥、雑炊等のレトルトパウチ食品、アイスクリーム、シャーベット、かき氷等の冷菓などを挙げることができる。
本発明の組成物を飲食品の形態に調製した場合、当該飲食品には、本発明の組成物の作用に鑑みて、脂質代謝を改善する旨の表示、基礎代謝を促進する旨の表示、体重を減少させる旨の表示、内臓脂肪または皮下脂肪を減少させる旨の表示、ダイエット効果を有する旨の表示、肥満の予防、治療もしくは症状の改善に用いることができる旨の表示、および/またはメタボリックシンドロームの予防、治療もしくは症状の改善に用いることができる旨の表示をすることができる。
本発明の組成物を含有してなる飼料としては、例えば、ウシ、ウマ、ブタ、ヒツジ、ウサギ、ヤギ等の家畜用飼料、ニワトリ、アヒル、ガチョウ、シチメンチョウ、ウズラ、ダチョウ、キジ等の家禽用飼料、イヌ、ネコ、ラット、マウス等のペット用飼料などが挙げられる。本発明の組成物を含有してなる飼料は、飼料中に本発明の組成物を添加することができ、一般的な飼料の製造方法を用いて加工製造することもできる。
本発明の医薬品および飲食品の投与量または摂取量は、患者または摂取者の年齢および体重、症状、投与時間、剤形、投与方法、薬剤の組み合わせ等に依存して決定できる。例えば、成人1人1日当たり、成分(a)を乾燥質量換算で0.2~100g、好ましくは0.2~20g、成分(b)を0.01~0.5g、好ましくは0.05~0.3gとなるように投与または摂取するのが好ましい。投与または摂取回数は、1日1回または複数回に分けて行うことができる。
また、成分(a)と成分(b)との質量比は、1000:1~0.1:1であればよく、500:1~0.2:1であることが好ましく、250:1~0.5:1であることがより好ましく、100:1~1:1であることが特に好ましい。なお、この範囲は1000:1~1:0.1、500:1~1:0.5又は250:1~1:1であっても良い。又、1000:1~1:1ならびに250:1~1:0.1の質量範囲も重要な範囲である。
成分(a)と成分(b)の質量比が上記範囲にあることで、本発明の組成物は優れたメタボリックシンドロームの予防、改善または治療作用を発揮することができる。
また、成分(a)と成分(b)との質量比は、1000:1~0.1:1であればよく、500:1~0.2:1であることが好ましく、250:1~0.5:1であることがより好ましく、100:1~1:1であることが特に好ましい。なお、この範囲は1000:1~1:0.1、500:1~1:0.5又は250:1~1:1であっても良い。又、1000:1~1:1ならびに250:1~1:0.1の質量範囲も重要な範囲である。
成分(a)と成分(b)の質量比が上記範囲にあることで、本発明の組成物は優れたメタボリックシンドロームの予防、改善または治療作用を発揮することができる。
以下に実施例・試験例をあげて本発明をさらに具体的に説明するが、本発明はこれらに限定されるものではない。なお、実施例において“部”表記は、重量基準である。
(実施例1.1)シームレスカプセルの製造
RHP10部、カフェイン0.5部を大豆油400部に懸濁させ、シームレスカプセルの内容液を得た。シームレスカプセルの外層の調製に用いる皮膜材料懸濁液は、ゼラチン 35部、およびD-ソルビトール10部を蒸留水200部に加え、60℃にて攪拌して懸濁させることにより得た。
上記内容液を、二重管ノズルの内側ノズルから、60℃に加熱した皮膜材料懸濁液を外側のノズルから同時に噴出させ、形成される2層構造の液滴を12℃の凝固油中に滴下した後、冷却して、湿カプセルを得た。得られた湿カプセルから凝固油を除いた後、ドラム乾燥することにより、粒径2mmのシームレスカプセルを得た。
RHP10部、カフェイン0.5部を大豆油400部に懸濁させ、シームレスカプセルの内容液を得た。シームレスカプセルの外層の調製に用いる皮膜材料懸濁液は、ゼラチン 35部、およびD-ソルビトール10部を蒸留水200部に加え、60℃にて攪拌して懸濁させることにより得た。
上記内容液を、二重管ノズルの内側ノズルから、60℃に加熱した皮膜材料懸濁液を外側のノズルから同時に噴出させ、形成される2層構造の液滴を12℃の凝固油中に滴下した後、冷却して、湿カプセルを得た。得られた湿カプセルから凝固油を除いた後、ドラム乾燥することにより、粒径2mmのシームレスカプセルを得た。
(実施例1.2)軟カプセルの製造
RHP10部、カフェイン1部を菜種油400部に懸濁させ、軟カプセルの内容液を得た。軟カプセルの皮膜に用いるゼラチン膜は、ゼラチン4部、グリセリン1部を、蒸留水2部に加えて、60℃で攪拌して懸濁させ、これをシート状に成形することにより得た。 このゼラチン膜が一対の回転円筒型金型の間に送られ、これと連動するポンプで内容液をゼラチン膜間に噴出することにより、カプセルの調製を行った。
RHP10部、カフェイン1部を菜種油400部に懸濁させ、軟カプセルの内容液を得た。軟カプセルの皮膜に用いるゼラチン膜は、ゼラチン4部、グリセリン1部を、蒸留水2部に加えて、60℃で攪拌して懸濁させ、これをシート状に成形することにより得た。 このゼラチン膜が一対の回転円筒型金型の間に送られ、これと連動するポンプで内容液をゼラチン膜間に噴出することにより、カプセルの調製を行った。
(実施例1.3)硬カプセルの製造
RHP10部、カフェイン2部をコーンスターチ300部に分散させて、硬カプセルの内容物を得た。硬カプセルの本体としては、市販の局方5号のカ プセルを用いた。上記内容物を常法によりカプセル本体に充填し、硬カプセルを得た。

(実施例1.4)錠剤の製造
RHP10部、カフェイン5部、コーンスターチ45部およびカルボキシメチルセルロースカルシウム20部を転動造粒機に入れ、予熱混合し、ヒドロキシプロピルセルロース1.7部を含む水溶液34部をスプレーして、造粒末を得た。ここにカルボキシメチルセルロースカルシウム100部およびタルク40部を加えて混合し、この混合末を打錠機により打錠し、錠剤を得た。
RHP10部、カフェイン5部、コーンスターチ45部およびカルボキシメチルセルロースカルシウム20部を転動造粒機に入れ、予熱混合し、ヒドロキシプロピルセルロース1.7部を含む水溶液34部をスプレーして、造粒末を得た。ここにカルボキシメチルセルロースカルシウム100部およびタルク40部を加えて混合し、この混合末を打錠機により打錠し、錠剤を得た。
(実施例1.5)顆粒剤の製造
実施例1.4と同様の操作により、乳糖、コーンスターチ、およびカルボキシメチルセルロースカルシウムを含み、ヒドロキシプロピルセルロースでコートされた造粒末を得た。この造粒末を、押出成型器を用いて顆粒を得た。

(実施例1.6)飲料の製造
RHP10部、澱粉加水分解物1部、カフェイン0.1部、香料適量を精製水100部に溶解させ、飲料を得た。

[試験方法]
マウスに高カロリー飼料を摂取させ、食餌性肥満マウスを用意した。この食餌性肥満マウスのカロリー摂取を制限し、体脂肪が分解・利用されやすい状態とし、ローズヒップ抽出物およびカフェインを含有する本発明の組成物を与えた。これにより体脂肪の分解・利用が促進し、肥満が改善されるかどうか検討した。
マウスに高カロリー飼料を摂取させ、食餌性肥満マウスを用意した。この食餌性肥満マウスのカロリー摂取を制限し、体脂肪が分解・利用されやすい状態とし、ローズヒップ抽出物およびカフェインを含有する本発明の組成物を与えた。これにより体脂肪の分解・利用が促進し、肥満が改善されるかどうか検討した。
ローズヒップ抽出物には「ローズヒップポリフェノールEX(商品名)」(チリロサイド含有量0.1質量%以上、森下仁丹社製、以下RHPと略す)を使用した。
カフェインは「茶の素(商品名)」(カフェイン純度98.5%、白鳥製薬社製)を使用した。 飼育用粉末飼料は「CE-2(商品名)」(日本クレア株式会社製)を使用した。
高カロリー飼料(4.73kcal/g)及び低カロリー飼料(3.16kcal/g)は、下記表1に示す組成のものを使用した。
その他、試験例に使用する組成物、試薬及び器具は、特記ない限り市販品を使用した。
カフェインは「茶の素(商品名)」(カフェイン純度98.5%、白鳥製薬社製)を使用した。 飼育用粉末飼料は「CE-2(商品名)」(日本クレア株式会社製)を使用した。
高カロリー飼料(4.73kcal/g)及び低カロリー飼料(3.16kcal/g)は、下記表1に示す組成のものを使用した。
その他、試験例に使用する組成物、試薬及び器具は、特記ない限り市販品を使用した。
[試験例1]
5週齢の雄性KKマウスを、水と飼育用粉末飼料で3日間飼育した後、高カロリー飼料で3週間飼育し、食餌性肥満マウスを用意した。この食餌性肥満マウスを3群(1群あたり6匹)に分け、各試験群に以下の(1)~(3)に示す飼料をそれぞれ与え、2週間飼育した。各試験群に与えた飼料は、次のとおりである。
(1)対照群;低カロリー飼料
(2)0.25%RHP投与群;低カロリー飼料+RHP0.25質量%
(3)0.50%RHP投与群;低カロリー飼料+RHP0.50質量%
5週齢の雄性KKマウスを、水と飼育用粉末飼料で3日間飼育した後、高カロリー飼料で3週間飼育し、食餌性肥満マウスを用意した。この食餌性肥満マウスを3群(1群あたり6匹)に分け、各試験群に以下の(1)~(3)に示す飼料をそれぞれ与え、2週間飼育した。各試験群に与えた飼料は、次のとおりである。
(1)対照群;低カロリー飼料
(2)0.25%RHP投与群;低カロリー飼料+RHP0.25質量%
(3)0.50%RHP投与群;低カロリー飼料+RHP0.50質量%
試験期間中(低カロリー飼料の給餌開始から2週間)は、マウスの体重測定と摂餌量測定を定期的に実施した。試験終了後にマウスを安楽死させて副睾丸周囲脂肪組織を摘出し、重量を測定した。
試験最終日のマウスの体重(g)、試験期間中(12日間)のマウスの1日あたりの平均摂取カロリー(kcal/日)、体重100g当たりの副睾丸周囲脂肪(内臓脂肪)重量(g/100g 体重)を表2に示した。結果は各群の平均値±標準偏差で示した。
表2のように、副睾丸周囲脂肪重量について(1)対照群と比較し、(2)0.25%RHP投与群では脂肪低減作用が確認できなかった。また、(3)0.50%RHP投与群では、統計学的有意な脂肪低減作用はみられなかった。
[試験例2]
食餌性肥満マウスを4群(1群6匹)に分け、各試験群に以下の(4)~(7)に示す飼料をそれぞれ与えた以外は、試験例1と同様の条件で実験を行った。
(4)対照群;低カロリー飼料
(5)2.5%RHP投与群;低カロリー飼料+RHP2.5質量%
(6)0.025%カフェイン投与群;低カロリー飼料+カフェイン0.025質量%
(7)本発明品投与群(RHP:カフェイン(20:1)の混合物);低カロリー飼料+RHP0.50質量%+カフェイン0.025質量%
なお上記において混合物における混合比は重量比である。
食餌性肥満マウスを4群(1群6匹)に分け、各試験群に以下の(4)~(7)に示す飼料をそれぞれ与えた以外は、試験例1と同様の条件で実験を行った。
(4)対照群;低カロリー飼料
(5)2.5%RHP投与群;低カロリー飼料+RHP2.5質量%
(6)0.025%カフェイン投与群;低カロリー飼料+カフェイン0.025質量%
(7)本発明品投与群(RHP:カフェイン(20:1)の混合物);低カロリー飼料+RHP0.50質量%+カフェイン0.025質量%
なお上記において混合物における混合比は重量比である。
結果を表3に示した。なお表3において、Dunnettの多重比較検定を行って統計処理し、**は危険率1%未満で対照群と有意な差があることを示す。
表3のように、副睾丸周囲脂肪組織重量に関して、(4)対照群と比較して(5)2.5%RHP投与群は10.2%減少した。同様に、(6)0.025%カフェイン投与群は(4)対照群と比較して9.2%減少した。しかし、いずれも統計学的有意な脂肪低減作用はみられなかった。
一方、(7)本発明品投与群では、カフェイン0.025%とRHP0.50%を組み合わせることで、副睾丸周囲脂肪組織重量が21.9%と減少が見られ、統計学的に有意な脂肪低減作用が見られた。
これより、(7)本発明品投与群では、カフェイン0.025%を単独で摂取した場合及びRHP0.50%を単独で摂取した場合からは予測できない優れた脂肪低減作用を発揮していることがわかった。
以上の結果から、本発明の組成物は、ローズヒップ抽出物及びカフェインを両方含有することにより明確な相乗作用を奏し、内臓脂肪量を低減させることがわかった。また、本発明の組成物は内臓脂肪低減作用を有するため、本発明の組成物を含有してなる医薬品、飲食品または飼料が、メタボリックシンドロームの予防、改善または治療に有用であることが示された。
一方、(7)本発明品投与群では、カフェイン0.025%とRHP0.50%を組み合わせることで、副睾丸周囲脂肪組織重量が21.9%と減少が見られ、統計学的に有意な脂肪低減作用が見られた。
これより、(7)本発明品投与群では、カフェイン0.025%を単独で摂取した場合及びRHP0.50%を単独で摂取した場合からは予測できない優れた脂肪低減作用を発揮していることがわかった。
以上の結果から、本発明の組成物は、ローズヒップ抽出物及びカフェインを両方含有することにより明確な相乗作用を奏し、内臓脂肪量を低減させることがわかった。また、本発明の組成物は内臓脂肪低減作用を有するため、本発明の組成物を含有してなる医薬品、飲食品または飼料が、メタボリックシンドロームの予防、改善または治療に有用であることが示された。
本発明の組成物はメタボリックシンドロームの予防、改善または治療効果を奏するため、産業上有用に用いることができる。
Claims (28)
- 以下の(a)および(b)を含有するメタボリックシンドロームの予防、改善または治療用組成物。
(a)ローズヒップ抽出物
(b)キサンチン誘導体の少なくとも一種 - ローズヒップ抽出物がローズヒップの種子を含む果実の極性溶媒抽出物を有効成分として含有する請求項1に記載の組成物。
- キサンチン誘導体がカフェイン、テオフィリンまたはテオブロミンである請求項1または2に記載の組成物。
- (a)と(b)との質量比が1000:1~0.1:1である請求項1~4のいずれかに記載の組成物。
- (a)と(b)との質量比が100:1~1:1である請求項1~5のいずれかに記載の組成物。
- 請求項1~6のいずれかに記載の組成物を含有してなる医薬品、飲食品または飼料。
- 以下の(a)および(b)を含有する組成物を人又は動物に投与することを特徴とするメタボリックシンドロームの予防、改善または治療方法。
(a)ローズヒップ抽出物
(b)キサンチン誘導体の少なくとも一種 - ローズヒップ抽出物がローズヒップの種子を含む果実の極性溶媒抽出物を有効成分として含有する請求項8に記載の方法。
- キサンチン誘導体がカフェイン、テオフィリンまたはテオブロミンである請求項8または9に記載の方法。
- (a)と(b)との質量比が1000:1~0.1:1である請求項8~11のいずれかに記載の方法。
- (a)と(b)との質量比が100:1~1:1である請求項8~12のいずれかに記載の方法。
- 組成物が医薬品、飲食品または飼料であることを特徴とする請求項8~13のいずれかに記載の方法。
- 以下の(a)および(b)を含有する組成物のメタボリックシンドロームの予防、改善または治療のための使用。
(a)ローズヒップ抽出物
(b)キサンチン誘導体の少なくとも一種 - ローズヒップ抽出物がローズヒップの種子を含む果実の極性溶媒抽出物を有効成分として含有する請求項15に記載の使用。
- キサンチン誘導体がカフェイン、テオフィリンまたはテオブロミンである請求項15または16に記載の使用。
- (a)と(b)との質量比が1000:1~0.1:1である請求項15~18のいずれかに記載の使用。
- (a)と(b)との質量比が100:1~1:1である請求項15~19のいずれかに記載の使用。
- 組成物が医薬品、飲食品または飼料であることを特徴とする請求項15~20の使用。
- 以下の(a)および(b)を含有する組成物のメタボリックシンドロームの予防剤、改善剤または治療剤製造のための使用。
(a)ローズヒップ抽出物
(b)キサンチン誘導体の少なくとも一種 - ローズヒップ抽出物がローズヒップの種子を含む果実の極性溶媒抽出物を有効成分として含有する請求項22に記載の組成物の使用。
- キサンチン誘導体がカフェイン、テオフィリンまたはテオブロミンである請求項22または23に記載の使用。
- (a)と(b)との質量比が1000:1~0.1:1である請求項22~25のいずれかに記載の使用。
- (a)と(b)との質量比が100:1~1:1である請求項22~26のいずれかに記載の使用。
- 組成物が医薬品、飲食品または飼料であることを特徴とする請求項22~27のいずれかに記載の使用。
