本発明の方法は、カリウムイオン存在下においてもG-quadruplexを特異的に検出することを目的とする。 本発明の方法は、カリウムイオン存在下において、チオフラビンTが、G-quadruplexと反応させたときに強い蛍光を発する現象を利用し、標的DNAがG-quadruplexを形成しているかどうかを判定する。本発明の方法においては、カリウムイオンおよび標的DNAを含有する第1試料溶液をG-quadruplexが形成される条件に維持した後、チオフラビンTを添加して第1蛍光強度値を測定する。標的DNAを含有する第2試料溶液をG-quadruplexが不安定化される条件に維持し、チオフラビンTを添加して第2蛍光強度値を測定し、これらの値の差が0以上であれば、標的DNAはG-quadruplexを形成できると判定される。
Description
本発明はDNAの高次構造の検出方法に関する。
ヒトのテロメアDNAは、5’-TTAGGG-3’(配列番号:1)と5’-CCCTAA-3’ (配列番号:2、配列番号1の相補鎖)からなる二本鎖DNAが繰り返された配列と、その最末端に5’-TTAGGG-3’(配列番号:1)のみが繰り返された一本鎖DNAを含む。この5’-TTAGGG-3’(配列番号:1)が繰り返された配列は、G-quadruplexと呼ばれる四重鎖DNA構造を形成できる。G-quadruplexは、4つのグアニンがG-quartetと呼ばれる構造を形成し、それがπ-πスタッキング相互作用により積み重なった構造である(図1)。このG-quadruplexは細胞の癌化や寿命に関連していると考えられているため、近年精力的に研究されている。
また最近行われたコンピュータによるゲノムワイド解析は、G-quadruplexを形成できると考えられる配列が、ゲノムDNA中にテロメアDNA以外にも多数存在することを示した。それらの多くは、c-kit、c-myc、H-ras、K-ras遺伝子を含む癌原因遺伝子のプロモーター領域に存在する。したがって、これらG-quadruplexを形成できると考えられる配列についても研究が行われている。以上の事実は、G-quadruplexが細胞の活動において重要な役割を果たしている可能性を示唆している。
以上の背景のもと、G-quadruplexを形成できると考えられるDNAが、本当にG-quadruplexを形成できるか否かを簡便に解析する技術が必要とされている。特に、細胞内のカリウムイオン濃度はおよそ100-150mMであるため、このカリウムイオン濃度条件下でG-quadruplexの形成を解析できる技術が必要である。そのために、一本鎖DNAや二本鎖DNAと比べて、G-quadruplexと反応した際に格段に強い蛍光を発する化合物(以下、G-quadruplexプローブと呼ぶ)の探索が行われてきた。言い換えれば、G-quadruplexプローブは、一本鎖DNAや二本鎖DNAと反応してもほとんど蛍光を発しないが、G-quadruplexと反応すると強い蛍光を発する性質を持っていなければいけない。
近年、最も精力的に研究されているG-quadruplexプローブの一つは、ベンゾチアゾール誘導体である。ベンゾチアゾール誘導体の研究が盛んな理由は、ベンゾチアゾール誘導体は水溶性が高く、蛍光強度変化が非常に大きいためである。たとえば、非特許文献1ではCyan40(化1)およびCyan2(化2)を用いたG-quadruplex検出技術が報告されている。この報告では、カリウムイオンが存在しない条件下で、Cyan2とG-quadruplexを反応させた場合、二本鎖DNA反応させた場合と比べて、優位に強い蛍光が発せられることが示されている。しかし、100mMカリウムイオン存在下では、Cyan2とG-quadruplexを反応させてもほとんど蛍光が検出されないことも示されている。したがって、Cyan2は、カリウムイオン存在下におけるG-quadruplexの検出には使えない。また非特許文献2では、チアゾールオレンジ(化3)(以下、TOと呼ぶ)を用いたG-quadruplex検出技術が報告されている。この報告の中で、TOは、100mMカリウムイオン存在下で、G-quadruplexと反応させると強い蛍光を発することが示されている。しかし、同じ条件下でTOを二本鎖DNAと反応させた場合、TOは、G-quadruplexと反応させた場合よりも、より強い蛍光を発することも示されている。したがって、100mMカリウムイオン存在下で、TOを用いてG-quadruplexを特異的に検出することはできない。
以上のように、近年、ベンゾチアゾール誘導体をG-quadruplexプローブとして用い、G-quadruplexを特異的に検出する技術の開発行われている。しかし、細胞内条件と同様にカリウムイオンが存在する条件下で特異的にG-quadruplexを検出できる技術は知られていなかった。

カリウムイオン存在下で、G-quadruplexを形成できると考えられるDNAが、本当にG-quadruplexを形成できるか否かを簡便に解析する技術は非常に有用である。そのため、ベンゾチアゾール誘導体を用いたG-quadruplex検出技術の研究が精力的になされているが、カリウムイオン存在下で特異的にG-quadruplexを検出できる技術は開発されていなかった。そこで本発明者は鋭意検討した結果、カリウムイオン存在下において、チオフラビンT(化4)が、G-quadruplexと反応させたときに強い蛍光を発することを見出した。この蛍光強度は、チオフラビンTを二本鎖DNAや一本鎖DNAと反応させた場合と比べ、優位に大きい。したがって、本発明によりカリウムイオン存在下においてもG-quadruplexを特異的に検出できることを見出した。
上記課題を解決する本発明は、
標的DNAがG-quadruplexをカリウムイオン存在下で形成しているかどうかを判定する方法であって、以下の工程を具備する方法である:
カリウムイオン、チオフラビンT、および前記標的DNAを含有する第1試料溶液をG-quadruplex形成反応条件下で維持する工程、
ラムダ1の波長における第1蛍光強度値Aを測定する工程、ここで
前記第1蛍光強度値Aは、前記第1試料溶液に含有されるチオフラビンTに由来し、
ラムダ1は、465ナノメートル以上505ナノメートル以下であり、
チオフラビンTおよび前記標的DNAを含有する第2試料溶液を、前記G-quadruplexの構造が不安定化する条件下に維持する工程、
前記ラムダ1の波長における第2蛍光強度値Bを測定する工程、ここで
前記第2蛍光強度値Bは、前記第2試料溶液に含有されるチオフラビンTに由来し、および
以下の不等式が充足されれば、前記標的DNAがカリウムイオン存在下で前記G-quadruplexを形成していると判定される工程:

標的DNAがG-quadruplexをカリウムイオン存在下で形成しているかどうかを判定する方法であって、以下の工程を具備する方法である:
カリウムイオン、チオフラビンT、および前記標的DNAを含有する第1試料溶液をG-quadruplex形成反応条件下で維持する工程、
ラムダ1の波長における第1蛍光強度値Aを測定する工程、ここで

本発明の方法では、前記第1蛍光強度値-前記第2蛍光強度値≦0の不等式が充足されれば、前記標的DNAがカリウムイオン存在下で前記G-quadruplexを形成していないと判定されることが好ましい。
本発明の方法では、前記G-quadruplexの構造が不安定化する条件は、リチウム存在下の条件であることが好ましい。
本発明の方法では、前記ラムダ1が485ナノメートルであることが好ましい。
本発明の上記目的、他の目的、特徴および利点は、添付図面参照の下、以下の好適な実施態様の詳細な説明から明らかにされる。
本発明により、カリウムイオン存在下において、G-quadruplexを特異的かつ定量的に検出する方法、G-quadruplexを形成できるDNAを特異的かつ定量的に検出する方法が提供される。
以下本発明の実施の形態について、図面を参照しながら説明する。
本実施の形態では、カリウムイオン存在下で標的DNAがG-quadruplexを形成するか否かについて調べる方法について、図2を用いて説明する。
本実施の形態では、以下の工程が実施される。
(工程1)カリウムイオンと標的DNAとチオフラビンTを含む試料溶液1を調製する。カリウムイオンと標的DNAとチオフラビンTを混合する順番は問わない。
(工程2)工程1の後、この試料溶液1をG-quadruplex形成条件下に置く。
(工程3)工程2の後、この試料溶液1中のチオフラビンT由来の蛍光強度値(A)を測定する。
(工程4)標的DNAとチオフラビンTを含む試料溶液2を、G-quadruplexの構造を不安定化する条件下に置く。
(工程5)工程4の後、試料溶液2中のチオフラビンT由来の蛍光強度値(B)を測定する。蛍光強度値AとBは、同じ波長の蛍光強度値である。
(工程6)A-B>0の場合、標的DNAがカリウムイオン存在下でG-quadruplexを形成すると判断する。
(工程1)カリウムイオンと標的DNAとチオフラビンTを含む試料溶液1を調製する。カリウムイオンと標的DNAとチオフラビンTを混合する順番は問わない。
(工程2)工程1の後、この試料溶液1をG-quadruplex形成条件下に置く。
(工程3)工程2の後、この試料溶液1中のチオフラビンT由来の蛍光強度値(A)を測定する。
(工程4)標的DNAとチオフラビンTを含む試料溶液2を、G-quadruplexの構造を不安定化する条件下に置く。
(工程5)工程4の後、試料溶液2中のチオフラビンT由来の蛍光強度値(B)を測定する。蛍光強度値AとBは、同じ波長の蛍光強度値である。
(工程6)A-B>0の場合、標的DNAがカリウムイオン存在下でG-quadruplexを形成すると判断する。
上記手順では、工程1において、試料溶液1中にチオフラビンTが加えられる。しかし、必ずしも工程1において、試料溶液1中にチオフラビンTが加えられる必要はない。工程3までに、試料溶液1中にチオフラビンTが加えられればよい。また、上記手順では、工程4において、試料溶液2中にチオフラビンTが加えられる。しかし、必ずしも工程4において、試料溶液2中にチオフラビンTが加えられる必要はない。工程5までに、試料溶液2中にチオフラビンTが加えられればよい。
本発明者は、チオフラビンTをG-quadruplexと反応させた場合、485nm付近に極大蛍光波長を有する強い蛍光が観察されることを発見した。また一方で、チオフラビンTを一本鎖DNAまたは二本鎖DNAと反応させた場合には、蛍光はまったく観察されないか、観察されても極めて小さいことも発見した。言い換えれば、本発明者はチオフラビンTがG-quadruplex特異的な蛍光プローブであることを発見した。したがって、カリウムイオン存在下で標的DNAがG-quadruplexを形成する場合と形成しない場合では、それぞれ次のとおりになる。
(1)カリウムイオン存在下で標的DNAがG-quadruplexを形成する場合
上記工程1~2の後に、標的DNAはG-quadruplexを形成しているため、上記工程3ではチオフラビンT由来の強い蛍光が確認される。しかし、上記工程4の後では、標的DNAの一部または全部が、一本鎖DNAまたは二本鎖DNAを形成するため、上記工程5ではチオフラビンT由来の蛍光はまったく認められないか、認められたとしても、その蛍光強度値は、上記工程3で得られた値よりは明らかに小さい。したがって、上記工程6で得られる値は0より大きい。
上記工程1~2の後に、標的DNAはG-quadruplexを形成しているため、上記工程3ではチオフラビンT由来の強い蛍光が確認される。しかし、上記工程4の後では、標的DNAの一部または全部が、一本鎖DNAまたは二本鎖DNAを形成するため、上記工程5ではチオフラビンT由来の蛍光はまったく認められないか、認められたとしても、その蛍光強度値は、上記工程3で得られた値よりは明らかに小さい。したがって、上記工程6で得られる値は0より大きい。
(2)カリウムイオン存在下で標的DNAがG-quadruplexを形成しない場合
上記工程1~2の後においても、上記工程4の後においても、標的DNAはG-quadruplexを形成していないため、上記工程3と上記工程5で測定されるチオフラビンT由来の蛍光強度は実質同じである。したがって、上記工程6で得られる値は実質0である。
上記工程1~2の後においても、上記工程4の後においても、標的DNAはG-quadruplexを形成していないため、上記工程3と上記工程5で測定されるチオフラビンT由来の蛍光強度は実質同じである。したがって、上記工程6で得られる値は実質0である。
上記工程4における、G-quadruplexの構造を不安定化する条件とは、例えばリチウムイオンが存在する条件である。リチウムイオン存在下では、G-quadruplexの構造は不安定化することが知られている。また他のG-quadruplexの構造を不安定化する条件は、高温条件である。高温条件下(~100℃)では、G-quadruplexの構造は不安定化することが知られている。上記工程4における、G-quadruplexの構造を不安定化する条件は、ここに挙げた例に限られない。G-quadruplexの構造を不安定化する条件であればよい。
また、上記工程3と上記工程5で用いられる励起光の波長は、チオフラビンTの吸収帯の範囲内であればいずれの波長でもよい。しかし、チオフラビンTの極大励起波長が450nm付近であるため、上記工程3と上記工程5で用いられる励起光の波長は、450nm付近が望ましい。
[実施例]
以下に記載する実施例および比較例で用いられるDNAは、全て北海道システム・サイエンス株式会社より購入された。またチオフラビンTはシグマ・アルドリッチ(株)より購入した。またチアゾールオレンジは和光純薬工業(株)より購入された。また蛍光強度解析には、サーモフィッシャーサイエンティフィック(株)製Varioskan flashを用いた。
以下に記載する実施例および比較例で用いられるDNAは、全て北海道システム・サイエンス株式会社より購入された。またチオフラビンTはシグマ・アルドリッチ(株)より購入した。またチアゾールオレンジは和光純薬工業(株)より購入された。また蛍光強度解析には、サーモフィッシャーサイエンティフィック(株)製Varioskan flashを用いた。
(比較例1)
本比較例1では、代表的なベンゾチアゾール誘導体であるチアゾールオレンジを用いて、G-quadruplexと二本鎖DNAの検出を行った。本比較例で用いられたG-quadruplexは、5’-GGGTTAGGGTTAGGGTTAGGG-3’(配列番号:3)のヒトテロメア配列のDNAからなる(以降、この配列のDNAをG-DNA1と呼ぶ)。また本比較例で用いられた二本鎖DNAの配列は、5’-AGTTCAAGGCGCCTTGAACT-3’ (配列番号:4)であった(以降、この配列のDNAをDuplex1と呼ぶ)。これら二種類のDNAを用いて、以下のとおり実験を行った。まず表1に示す反応溶液を調製した。
本比較例1では、代表的なベンゾチアゾール誘導体であるチアゾールオレンジを用いて、G-quadruplexと二本鎖DNAの検出を行った。本比較例で用いられたG-quadruplexは、5’-GGGTTAGGGTTAGGGTTAGGG-3’(配列番号:3)のヒトテロメア配列のDNAからなる(以降、この配列のDNAをG-DNA1と呼ぶ)。また本比較例で用いられた二本鎖DNAの配列は、5’-AGTTCAAGGCGCCTTGAACT-3’ (配列番号:4)であった(以降、この配列のDNAをDuplex1と呼ぶ)。これら二種類のDNAを用いて、以下のとおり実験を行った。まず表1に示す反応溶液を調製した。
Xは0、50、100、150、または500であった。また、Yは0、10、または50であった。次に、この反応溶液を90℃で2分間保温した後、0.5℃/分の降温速度で25℃まで降温させた。以上の工程後、G-DNA1はG-quadruplexを形成することがすでに知られている。同様にDuplex1は分子内二本鎖構造を形成することが知られている。その後、この反応溶液について蛍光強度解析を行った。励起光波長は501nmであった。結果を図3に示す。図3(A)はKCl濃度が0mMの際の蛍光スペクトル結果である。図3(B)は、この蛍光スペクトル結果をもとに、各DNA濃度と530nmにおける蛍光強度の関係をグラフ化したものである。同様に、図4~7はそれぞれ、50、100、150、および500mM-KCl存在下における、各DNA濃度と530nmにおける蛍光強度の関係をグラフ化したものである。以上の結果より、チアゾールオレンジはG-DNA1と反応させると蛍光を発するが、Duplex1と反応させた場合も同程度かあるいはそれ以上に蛍光を発することが分かる。すなわち、これらの結果はチアゾールオレンジのG-quadruplexへの特異性が全くないことを示している。したがって、標的DNAがG-quadruplexを形成できるか否かの解析にはチアゾールオレンジを用いることはできない。
(実施例1)
本実施例1では、比較例1と同様に、G-DNA1とDuplex1の検出を行った。ただし、TOの代わりにチオフラビンT(濃度は1μM)を用いた。また、100mM-KClの条件下においてのみ、測定対象DNAとして、5’-TTTTTTTTTTTTTTTTTTTTT-3’(配列番号:5)(以降、この配列からなるDNAをT21と呼ぶ)を用いた。T21は一本鎖DNAとして溶液中で存在することが分かっている。また、励起光波長は501nmではなく、450nmであった。それ以外については、比較例1と同じ実験を行った。
本実施例1では、比較例1と同様に、G-DNA1とDuplex1の検出を行った。ただし、TOの代わりにチオフラビンT(濃度は1μM)を用いた。また、100mM-KClの条件下においてのみ、測定対象DNAとして、5’-TTTTTTTTTTTTTTTTTTTTT-3’(配列番号:5)(以降、この配列からなるDNAをT21と呼ぶ)を用いた。T21は一本鎖DNAとして溶液中で存在することが分かっている。また、励起光波長は501nmではなく、450nmであった。それ以外については、比較例1と同じ実験を行った。
結果を図8に示す。図8(A)はKCl濃度が0mMの際の蛍光スペクトル結果を示す。図8(B)はこのスペクトル結果をもとに、各DNA濃度と485nmにおける蛍光強度の関係をプロットしたグラフを示す。同様に、図9~12はそれぞれ、50、100、150、および500mM-KCl存在下における、各DNA濃度と485nmにおける蛍光強度の関係をプロットしたグラフを示す。以上の結果より、いずれの濃度のKCl存在下においても、チオフラビンTをG-DNA1と反応させると蛍光を発することが分かる。しかし、チオフラビンTをDuplex1やT21と反応させた場合は、ほとんど蛍光を発しないことが分かる。すなわち、これらの結果はチオフラビンTのG-quadruplexへの特異性が極めて高いことを示している。したがって、チオフラビンTを用いることで、カリウムイオン存在下で標的DNAがG-quadruplexを形成でるか否かを判断できることが示された。
(実施例2)
実施例1では、チオフラビンTを用いて、G-DNA1からなるG-quadruplexの検出を行った。G-DNA1は(3+1)型のG-quadruplex構造を形成することが知られている(非特許文献3、非特許文献4)。本実施例2では、チオフラビンTを用いて、異なる構造のG-quadruplexの検出を行った。本実施例2では、5’-GGGGTTTTGGGGTTTTGGGGTTTTGGGG-3’(配列番号:6)の配列のDNAが形成するG-quadruplexを測定対象DNAとした(以降、この配列からなるDNAをG-DNA2と呼ぶ)。また、KClの濃度は100mMのみであった。それ以外は、実施例1と同じ実験を行った。この実験条件では、G-DNA2はアンチパラレル型G-quadruplexを形成することが知られている。
実施例1では、チオフラビンTを用いて、G-DNA1からなるG-quadruplexの検出を行った。G-DNA1は(3+1)型のG-quadruplex構造を形成することが知られている(非特許文献3、非特許文献4)。本実施例2では、チオフラビンTを用いて、異なる構造のG-quadruplexの検出を行った。本実施例2では、5’-GGGGTTTTGGGGTTTTGGGGTTTTGGGG-3’(配列番号:6)の配列のDNAが形成するG-quadruplexを測定対象DNAとした(以降、この配列からなるDNAをG-DNA2と呼ぶ)。また、KClの濃度は100mMのみであった。それ以外は、実施例1と同じ実験を行った。この実験条件では、G-DNA2はアンチパラレル型G-quadruplexを形成することが知られている。
結果を図13に示す。図13(A)は蛍光スペクトル結果を示す。図13(B)はこの結果をもとに、G-DNA2の濃度と485nmにおける蛍光強度の関係をプロットしたグラフを示す。以上の結果より、チオフラビンTをG-DNA2と反応させると蛍光を発することが分かる。したがって、チオフラビンTを用いて、アンチパラレル型G-quadruplexを検出できることが示された。
(実施例3)
実施例1および2ではそれぞれ、チオフラビンTを用いて、(3+1)形のG-quadruplexとアンチパラレル型のG-quadruplexの検出を行った。そこで本実施例3ではチオフラビンTを用いて、実施例1、2と異なる、パラレル型のG-quadruplexの検出を行った。本実施例3では、5’-GGGTGGGTGGGTGGG-3’(配列番号:7)の配列のDNAが形成するG-quadruplexを測定対象DNAとした(以降、この配列からなるDNAをG-DNA3と呼ぶ)。また、KClの濃度は100mMのみであった。それ以外は、実施例1と同じ実験を行った。この実験条件では、G-DNA3はパラレル型G-quadruplexを形成することが知られている。
実施例1および2ではそれぞれ、チオフラビンTを用いて、(3+1)形のG-quadruplexとアンチパラレル型のG-quadruplexの検出を行った。そこで本実施例3ではチオフラビンTを用いて、実施例1、2と異なる、パラレル型のG-quadruplexの検出を行った。本実施例3では、5’-GGGTGGGTGGGTGGG-3’(配列番号:7)の配列のDNAが形成するG-quadruplexを測定対象DNAとした(以降、この配列からなるDNAをG-DNA3と呼ぶ)。また、KClの濃度は100mMのみであった。それ以外は、実施例1と同じ実験を行った。この実験条件では、G-DNA3はパラレル型G-quadruplexを形成することが知られている。
結果を図14に示す。図14(A)は蛍光スペクトル結果を示す。図14(B)はこの結果をもとに、G-DNA3の濃度と485nmにおける蛍光強度の関係をプロットしたグラフを示す。以上の結果より、チオフラビンTをG-DNA3と反応させると蛍光を発することが分かる。したがって、チオフラビンTを用いて、パラレル型G-quadruplexを検出できることが示された。
(実施例4)
G-DNA2と5’-CCCCAAAACCCCAAAACCCCAAAACCCC-’(配列番号:8、G-DNA2の相補鎖)(以降、このDNAをDuplex-Cと呼ぶ)は、リチウムイオン存在下のようなG-quadruplexの構造を不安定化する条件下では、互いに相補的に結合して二本鎖DNAを形成する。しかし、カリウムイオン存在下では、G-DNA2はG-quadruplex構造を形成し、Duplex-Cは一本鎖DNAの状態で存在する(図15)。本実施例4では、G-DNA2とDuplex-Cを混合したDNA試料が、カリウムイオン存在下ではG-quadruplexを形成できることを、カリウムイオン存在下でのチオフラビンT由来の蛍光強度と、リチウムイオン存在下でのチオフラビンT由来の蛍光強度の差によって判別できるか否かについて検討した。
G-DNA2と5’-CCCCAAAACCCCAAAACCCCAAAACCCC-’(配列番号:8、G-DNA2の相補鎖)(以降、このDNAをDuplex-Cと呼ぶ)は、リチウムイオン存在下のようなG-quadruplexの構造を不安定化する条件下では、互いに相補的に結合して二本鎖DNAを形成する。しかし、カリウムイオン存在下では、G-DNA2はG-quadruplex構造を形成し、Duplex-Cは一本鎖DNAの状態で存在する(図15)。本実施例4では、G-DNA2とDuplex-Cを混合したDNA試料が、カリウムイオン存在下ではG-quadruplexを形成できることを、カリウムイオン存在下でのチオフラビンT由来の蛍光強度と、リチウムイオン存在下でのチオフラビンT由来の蛍光強度の差によって判別できるか否かについて検討した。
実験手順は以下のとおりであった。まず表2に示す反応溶液1を調製した。
同様に、反応溶液2を調製した。反応溶液2の組成は、反応溶液1のKClをLiClに変えた以外は、反応溶液1と同じ組成である。これらの反応溶液を90℃で2分間保温した後、0.5℃/分の降温速度で25℃まで降温させた。以上の工程後、反応溶液1中のG-DNA1の大半はG-quadruplexを形成し、Duplex-Cは一本鎖DNA状態で存在することがすでに知られている。一方、反応溶液2中では、リチウムイオンが存在しているため、G-quadruplexの構造は不安定化し、G-DNA2とDuplex-Cの大半が互いに相補的に結合し、二本鎖DNAを形成することが知られている。その後、これらの反応溶液について485nmにおける蛍光強度解析を行った。励起光波長は450nmであった。
また同様に比較実験として、G-DNA2とDuplex-Cの代わりに、5’-AGAAGAGAAAGA-3’(配列番号:9)(以降、このDNAをDuplex-AGAと呼ぶ)と5’-TCTTTCTCTTCT-3’(配列番号:10、配列番号9の相補鎖)(以降、このDNAをDuplex-TCTと呼ぶ)を用いた以外は、全く同じ実験を行った。Duplex-AGAとDuplex-TCTは互いに相補的である。またいずれもG-quadruplexを形成できない。したがって、リチウムイオン存在下でもカリウムイオン存在下でも、これらDNAは互いに相補的に結合して、二本鎖DNAを形成する。
結果を図16に示す。図16中の右側のバーは、G-DNA2とDuplex-Cを用いた場合の、反応溶液1由来の蛍光強度値から反応溶液2由来の蛍光強度値を引いた値を示している。その値はおよそ30であり、0より大きかった。一方、左側のバーは、Duplex-AGAとDuplex-TCTを用いた場合の、反応溶液1由来の蛍光強度値から反応溶液2由来の蛍光強度値を引いた値を示している。その値はおよそ0であった。以上の結果は、チオフラビンTを用いて、標的DNAがカリウムイオン存在下でG-quadruplexを形成できるか否かを判断できることを示している。
(実施例5)
G-DNA1は、カリウムイオン存在下では、そのほとんどがG-quadruplexを形成することが知られている。一方、G-DNA1は、リチウムイオン存在下では、G-quadruplexの構造が不安定化されるため、その一部は一本鎖DNA状態で存在する。本実施例5では、G-DNA1を含む試料溶液中において、チオフラビンTを用いて、カリウムイオン存在下でG-DNA2から形成されるG-quadruplexを検出できるか否かについて検討した。本実施例5では、G-DNA1が、カリウムイオン存在下ではG-quadruplexを形成できることを、カリウムイオン存在下でのチオフラビンT由来の蛍光強度と、リチウムイオン存在下でのチオフラビンT由来の蛍光強度の差によって判別できるか否かについて検討した。
G-DNA1は、カリウムイオン存在下では、そのほとんどがG-quadruplexを形成することが知られている。一方、G-DNA1は、リチウムイオン存在下では、G-quadruplexの構造が不安定化されるため、その一部は一本鎖DNA状態で存在する。本実施例5では、G-DNA1を含む試料溶液中において、チオフラビンTを用いて、カリウムイオン存在下でG-DNA2から形成されるG-quadruplexを検出できるか否かについて検討した。本実施例5では、G-DNA1が、カリウムイオン存在下ではG-quadruplexを形成できることを、カリウムイオン存在下でのチオフラビンT由来の蛍光強度と、リチウムイオン存在下でのチオフラビンT由来の蛍光強度の差によって判別できるか否かについて検討した。
実験手順は、G-DNA2とDuplex-Cの代わりにG-DNA1を用いた以外は実施例4と全く同じであった。また、比較実験としては、G-DNA2とDuplex-Cの代わりにT21を用いた実験を行った。T21は、リチウムイオン存在下でもカリウムイオン存在下でも、一本鎖DNAの状態で存在する。
結果を図17に示す。図17中の右側のバーは、G-DNA1を用いた場合の、反応溶液1由来の蛍光強度値から反応溶液2由来の蛍光強度値を引いた値を示している。その値はおよそ6であり、0より大きかった。一方、左側のバーは、T21を用いた場合の、反応溶液1由来の蛍光強度値から反応溶液2由来の蛍光強度値を引いた値を示している。その値はおよそ0であった。以上の結果は、チオフラビンTを用いて、標的DNAがカリウムイオン存在下でG-quadruplexを形成できるか否かを判断できることを示している。
以上まとめると、本発明者は、従来のベンゾチアゾール誘導体では達成できなかったG-quadruplexの特異的検出を、チオフラビンTを用いれば達成できることを世界で初めて見出し、本発明を完成するに至った。
上記説明から、当業者にとっては、本発明の多くの改良や他の実施の形態が明らかである。したがって、上記説明は例示としてのみ解釈されるべきであり、本発明を実行する最良の態様を当業者に教示する目的で提供されたものである。本発明の精神を逸脱することなく、その構造および/または機能の詳細を実質的に変更できる。
本発明により、G-quadruplexの特異的検出方法は、バイオテクノロジー分野における分析方法として有用である。

Claims (4)
- 標的DNAがG-quadruplexをカリウムイオン存在下で形成しているかどうかを判定する方法であって、以下の工程を具備する方法:
カリウムイオン、チオフラビンT、および前記標的DNAを含有する第1試料溶液をG-quadruplex形成反応条件下で維持する工程、
ラムダ1の波長における第1蛍光強度値Aを測定する工程、ここで
前記第1蛍光強度値Aは、前記第1試料溶液に含有されるチオフラビンTに由来し、
ラムダ1は、465ナノメートル以上505ナノメートル以下であり、
チオフラビンTおよび前記標的DNAを含有する第2試料溶液を、前記G-quadruplexの構造が不安定化する条件下に維持する工程、
前記ラムダ1の波長における第2蛍光強度値Bを測定する工程、ここで
前記第2蛍光強度値Bは、前記第2試料溶液に含有されるチオフラビンTに由来し、および
以下の不等式が充足されれば、前記標的DNAがカリウムイオン存在下で前記G-quadruplexを形成していると判定される工程:
前記第1蛍光強度値A-前記第2蛍光強度値B>0。 - 請求項1に記載の方法であって、
前記第1蛍光強度値-前記第2蛍光強度値≦0の不等式が充足されれば、前記標的DNAがカリウムイオン存在下で前記G-quadruplexを形成していないと判定される、方法。 - 請求項1に記載の方法であって、
前記G-quadruplexの構造が不安定化する条件は、リチウム存在下の条件である、方法。 - 請求項1に記載の方法であって、
前記ラムダ1が485ナノメートルである、方法。
