Description
多糖類、特にヒアルロン酸は、生体適合性に優れるため、点眼液、関節痛緩和剤、腹膜癒着防止材料などとして、広く用いられている。非特許文献1は、該ヒアルロン酸を用いたハイドロゲルについて、詳細に開示する。非特許文献1は、特に、共有結合を形成してハイドロゲルを調製する方法を網羅的に開示する。しかしながら、該共有結合を形成する方法は、生体分子との非特異的反応による細胞毒性が生じうる問題から、生体内での利用が制限されてくる。
また、本発明の目的は、上記目的以外に、上記目的に加えて、該ハイドロゲルを用いた材料、特に生分解性に優れた生体適合性材料を提供することにある。
さらに、本発明の目的は、上記目的以外に、上記目的に加えて、該ハイドロゲルの製造方法を提供することにある。

2’)-2) 1-ブロモシクロオクテン基及びエステル基を有する物質を、溶媒中、脱臭素アルキン化して、シクロオクチン基及びエステル基を有する物質を得る工程;及び
2’)-3) シクロオクチン基及びエステル基を有する物質を加水分解反応に付す工程;
を有することにより得られるのがよい。
1’)カルボキシメチルデキストランを準備する工程;

2’)-2) 1-ブロモシクロオクテン基及びエステル基を有する物質を、溶媒中、脱臭素アルキン化して、シクロオクチン基及びエステル基を有する物質を得る工程;及び
2’)-3) シクロオクチン基及びエステル基を有する物質を加水分解反応に付す工程;
を有することにより得られるのがよい。
また、本発明により、上記効果以外に、又は、上記効果に加えて、該ハイドロゲルを有する材料、特に生体適合性材料を提供することができる。ここで、本発明の生体適合性材料は、従来のポリエチレングリコール等によるハイドロゲルと比較して、優れた生分解性を有する生体適合性材料を提供することができる点で有益である。
さらに、本発明により、上記効果以外に、又は、上記効果に加えて、該ハイドロゲルの製造方法を提供することができる。
また、本発明により、上記効果以外に、又は、上記効果に加えて、該ハイドロゲルの原料となる薬剤及びその製造方法を提供することができる。
本願は、ハイドロゲル、該ハイドロゲルの製造方法、ハイドロゲルの原料である特定のヒアルロン酸又は特定のカルボキシメチルデキストラン、及び該原料の製造方法を提供する。以下、順に説明する。
本願は、a)ヒアルロン酸、カルボキシメチルデキストラン、セルロース誘導体(例えばカルボキシメチルセルロース)及びキトサンからなる群から選ばれる第1のポリマー部位;
b)上記a)と同種であっても異種であってもよいが、上記a)とは異なる分子からなる、ヒアルロン酸、カルボキシメチルデキストラン、セルロース誘導体(例えばカルボキシメチルセルロース)及びキトサンからなる群から選ばれる第2のポリマー部位;及び
c)トリアゾール環基又はその誘導体基;
を有し、
c)トリアゾール環基又はその誘導体基を介して、a)第1のポリマー部位とb)第2のポリマー部位とが架橋する構造を有するハイドロゲルを提供する。
即ち、図1は、本願のハイドロゲルの構造1を模式的に示す図である。
ハイドロゲルの構造1は、第1のポリマー部位2;及び第2のポリマー部位3;並びにトリアゾール環基又はその誘導体基4を有する。なお、図1において、図示を簡素化するために、4として、トリアゾール環基のみを記載する。
第1のポリマー部位2;及び第2のポリマー部位3は、トリアゾール環基4を介して架橋される。なお、第1のポリマー部位2とトリアゾール環基4との間にスペーサー基X1を有してなる。また、第2のポリマー部位とトリアゾール環基4との間にスペーサー基X2を有してなる。なお、X1及びX2については後述する。
第2のポリマー部位は、第1のポリマー部位と同種であっても異種であってもよいが、第1のポリマー部位とは異なる分子からなる。第2のポリマー部位は、ヒアルロン酸、カルボキシメチルデキストラン、セルロース誘導体(例えばカルボキシメチルセルロース)及びキトサンからなる群から選ばれるのがよく、好ましくはヒアルロン酸、カルボキシメチルデキストラン、又はキトサンであるのがよい。
第1及び第2のポリマー部位は、上記の群から選ばれ、それらはOH基及び/又は-NH2基を有する。トリアゾール環基又はその誘導体基は、該第1のポリマー部位の全-OH基又は全-NH2基を100mol%とした場合、5~60mol%、好ましくは10~50mol%、より好ましくは20~30mol%で存在するのがよい。また、トリアゾール環基又はその誘導体基は、第2のポリマー部位の全-OH基又は全-NH2基を100mol%とした場合、5~60mol%、好ましくは10~50mol%、より好ましくは20~30mol%で存在するのがよい。
また、第1のポリマー部位は、上述の群から選ばれ、それらは-OH基及び/又は-NH2基を有する。X1は、該OH基を置換して結合し、-O-X1-を形成するのがよく、好ましくは-O-CO-X11-、-CO-NH-X11-、-CO-NH-NH-X11-、-S-S-X11-、-CO-X11-(X11は、上記X1と同様にアルキレン基又は多糖類由来の基を示す)、好ましくは-O-CO-X11-、-CO-NH-X11-を形成するのがよい。
なお、-OH基は、-COOH基、-SH基などに適宜、置換され、さらにそれらが置換して、上記の基を形成してもよい。
また、第2のポリマー部位は、上述の群から選ばれ、それらは-OH基及び/又は-NH2基を有する。X2は、該OH基を置換して結合し、-O-X2-を形成するのがよく、好ましくは-O-CO-X21-、-CO-NH-X21-、-CO-NH-NH-X21-、-S-S-X21-、-CO-X21-(X21は、上記X2と同様にアルキレン基又は多糖類由来の基を示す)、好ましくは-O-CO-X21-、-CO-NH-X21-を形成するのがよい。
なお、-OH基は、-COOH基、-SH基などに適宜、置換され、さらにそれらが置換して、上記の基を形成してもよい。
即ち、
A) a)アジド基を有するヒアルロン酸、アジド基を有するカルボキシメチルデキストラン、又はアジド基を有するキトサンを含む第1の液;を準備する工程;
B) b)シクロオクチン基又はシクロオクチン基誘導体を有するヒアルロン酸、シクロオクチン基又はシクロオクチン基誘導体を有するカルボキシメチルデキストラン、もしくはシクロオクチン基又はシクロオクチン基誘導体を有するキトサンを含む第2の液;を準備する工程;及び
C’) a)第1の液とb)第2の液とを、生体内の癒着を防止する領域に注入し且つa)第1の液とb)第2の液とを混合する工程;
を有することにより、ハイドロゲルを形成し、該ハイドロゲルにより癒着を防止することができる。
即ち、 A) a)アジド基を有するヒアルロン酸、アジド基を有するカルボキシメチルデキストラン、又はアジド基を有するキトサンを含む第1の液;を準備する工程;
B) b)シクロオクチン基又はシクロオクチン基誘導体を有するヒアルロン酸、シクロオクチン基又はシクロオクチン基誘導体を有するカルボキシメチルデキストラン、もしくはシクロオクチン基又はシクロオクチン基誘導体を有するキトサンを含む第2の液;を準備する工程;及び
C”) a)第1の液とb)第2の液とを、生体内でのターゲット細胞の近傍に注入し且つa)第1の液とb)第2の液とを混合する工程;
を有することにより、ハイドロゲルを形成し、該ハイドロゲルにより、ターゲット細胞を封入することができる。
上記ハイドロゲルは、次のような方法で製造することができる。
即ち、A) a)ヒアルロン酸、カルボキシメチルデキストラン、セルロース誘導体(例えばカルボキシメチルセルロース)及びキトサンからなる群から選ばれる第1のポリマーであって、且つ該第1のポリマーがアジド基を有する、第1のポリマーを含む液を準備する工程;
B) b)上記a)と同種であっても異種であってもよいが、上記a)とは異なる分子からなる、ヒアルロン酸、カルボキシメチルデキストラン、セルロース誘導体(例えばカルボキシメチルセルロース)及びキトサンからなる群から選ばれる第2のポリマーであって、且つ該第2のポリマーがシクロオクチン基又はシクロオクチン基誘導体を有する、第2のポリマーを含む液を準備する工程;及び
C) a)第1の液とb)第2の液とを混合する工程;
を有することにより、アジド基とシクロオクチン基又はシクロオクチン基誘導体とのクリック反応により、トリアゾール環又はその誘導体が形成され、該トリアゾール環又はその誘導体を介して、第1のポリマーと第2のポリマーとが架橋する構造を有するハイドロゲルを形成して、ハイドロゲルを得ることができる。ここで、工程C)は、上述のとおり、生体内で行われることができる。
第1のポリマーは、ヒアルロン酸、カルボキシメチルデキストラン、セルロース誘導体(例えばカルボキシメチルセルロース)及びキトサンからなる群から選ばれ、且つアジド基を有する。
また、第2のポリマーは、ヒアルロン酸、カルボキシメチルデキストラン、セルロース誘導体(例えばカルボキシメチルセルロース)及びキトサンからなる群から選ばれ、且つアジド基を有する。
第1及び第2のポリマー自体の調製については、後述する。
また、第2のポリマーを含む液も、第1のポリマーを含む液と同様に、得られた第1のポリマーを、リン酸緩衝生理食塩水(PBS)、トリス緩衝液、クエン酸緩衝液、炭酸-重炭酸緩衝液、ハンクス液、生理食塩水、ダルベッコ変法イーグル培地(DMEM)やロズウエルパークメモリアルインスティテュート1640(RPMI 1640)細胞培養培地などの溶媒(第2のポリマーを含む液に用いる溶媒は、第1のポリマーを含む液に用いるそれと同じであっても異なってもよい)に溶解することにより得ることができる。
混合は、種々の方法により行うことができる。混合体を所望の位置に配置し、その位置でゲルを形成する場合、C)混合工程は、ダブルシリンジを用いるのが好ましい。
図2は、ダブルシリンジ11を模式的に説明する図である。
ダブルシリンジ11は、シリンジ12とシリンジ13とを隣接して配置し、且つその射出部を連結してシリンジ12内の液とシリンジ13内の液とを混合して射出する混合射出部14を有する。なお、図2は、ダブルシリンジの一例であって、2液を混合し且つ所望の位置に該混合液を射出する機能を有する限り、その他の構成を有してもよい。
X1は、好ましくはエーテル基、エステル基、アミド基、ヒドラジド基、ジスルフィド基、及びカルボニル基からなる群から選択される1以上の置換基を有するアルキレン基又は多糖類由来の基であるのがよく、より好ましくはエステル基、エーテル基、またはアミド基を有する、アルキレン基又は多糖類由来の基であるのがよい。
また、第1のポリマーは、上述の群から選ばれ、それらは-OH基及び/又は-NH2基を有する。X1は、該OH基を置換して結合し、-O-X1-を形成するのがよく、好ましくは-O-CO-X11-、-CO-NH-X11-、-CO-NH-NH-X11-、-S-S-X11-、-CO-X11-(X11は、上記X1と同様にアルキレン基又は多糖類由来の基を示す)、好ましくは-O-CO-X11-、-CO-NH-X11-を形成するのがよい。特に、-X1-が、-CO-(CH2)3-、又は-CO-NH-(CH2CH2O)m-NH-CO-CH2-O-であるのがよい。

本願は、ハイドロゲルの原料である特定のヒアルロン酸の製造方法を提供する。
<<アジド基を有するヒアルロン酸の製造方法>>
上述のアジド基を有するヒアルロン酸は、次の方法により製造することができる。
即ち、1)ヒアルロン酸を準備する工程;
2)アジド基及びアジド基以外の官能基(例えば、カルボキシル基、アミノ基、チオール基、ヒドラジド基、アルデヒド基、又は水酸基などを挙げることができるがこれらに限定されない。好ましくはカルボキシル基、アミノ基又はヒドラジド基、より好ましくはカルボキシル基又はアミノ基であるのがよい)を有する物質を準備する工程;
3) 1)ヒアルロン酸を、水にも有機溶媒にも可溶な長鎖アルキルアンモニウムカチオンを持つ塩(例えば、テトラブチルアンモニウム塩、トリオクチルメチルアンモニウム塩、ベンジルジメチルオクタデシルアンモニウム塩など、好ましくはテトラブチルアンモニウム塩、トリオクチルメチルアンモニウム塩、より好ましくはテトラブチルアンモニウム塩を挙げることができるがこれらに限定されない)でイオン交換し有機溶媒に可溶化させ、可溶化体の溶液を調製する工程;
4) 2)物質の有機溶媒の溶液を得る工程;及び
5) 4)溶液と3)可溶化体の溶液とを混合し、且つカルボキシル化(例えば、カルボジイミド化、イミダゾール化などを挙げることができるがこれらに限定されない。好ましくはイミダゾール化であるのがよい)反応により官能基とヒアルロン酸のOH基とを反応させる工程;
を有することにより、アジド基を有するヒアルロン酸を得ることができる。
特に、該物質の官能基がカルボン酸基である場合、次のように調製することができる。
即ち、2)-1)分子量1万以下、好ましくは5千以下、より好ましくは3千以下であり、一端がアミノ基、他端がカルボン酸基を有する化合物を準備する工程;及び
2)-2) 該化合物を、溶媒中、触媒存在下で、アジド化物(例えば、イミダゾール-1-スルホニルアジド、又はトリフリックアジドなどを挙げることができるがこれらに限定されない。好ましくはイミダゾール-1-スルホニルアジドであるのがよい)と反応させる工程;
を有することにより得られることができる。
工程2)-2)の溶媒として、メタノール、エタノール、イソプロピルアルコール、クロロホルム、塩化メチレンなどを挙げることができるがこれらに限定されない。工程2)-2)の溶媒として、好ましくはメタノール又はエタノール、より好ましくはメタノールであるのがよい。
また、触媒として、硫酸銅などを挙げることができるがこれに限定されない。
さらに、アジド化物として、例えば、イミダゾール-1-スルホニルアジド、又はトリフリックアジドなどを挙げることができるがこれらに限定されない。好ましくはイミダゾール-1-スルホニルアジドであるのがよい。
ここで、工程2)-2)は、用いる化合物、用いる溶媒、用いる触媒、及び用いるアジド化物などに依存するが、0℃~4℃の条件で行うのがよい。
水にも有機溶媒にも可溶な長鎖アルキルアンモニウムカチオンを持つ塩として、例えば、テトラブチルアンモニウム塩、トリオクチルメチルアンモニウム塩、ベンジルジメチルオクタデシルアンモニウム塩など、好ましくはテトラブチルアンモニウム塩、トリオクチルメチルアンモニウム塩、より好ましくはテトラブチルアンモニウム塩を挙げることができるがこれらに限定されない。
また、可溶化する際に用いる有機溶媒として、例えば、メタノール、エタノール、イソプロピルアルコール、ジエチルエーテル、ジメチルスルホキシド、ジメチルアセトアミド、テトラヒドロフラン、ジメチルフォルムアミド、ジオキサン、N-メチルピロリドン、トルエン、及びシクロヘキサンなどを挙げることができるがこれらに限定されない。好ましくはエタノール、アセトン、テトラヒドロフラン、又はジメチルフォルムアミド、より好ましくはエタノール、アセトン、又はテトラヒドロフランであるのがよい。
ここで、工程3)は、用いる水にも有機溶媒にも可溶な長鎖アルキルアンモニウムカチオンを持つ塩、及び用いる有機溶媒などに依存するが、温度20~40℃、pH4~12の条件で行うのがよい。
カルボキシル化反応は、例えば、カルボジイミド化反応、イミダゾール化反応などを挙げることができるがこれらに限定されない。好ましくはイミダゾール化反応であるのがよい。
工程5)は、工程4)で得られた溶液、工程3)で得られた可溶化体の溶液、カルボキシル化反応などに依存するが、温度20~40℃、pH4~12の条件で行うのがよい。
上述のシクロオクチン基又はシクロオクチン誘導体基を有するヒアルロン酸は、次の方法により製造することができる。
即ち、1)ヒアルロン酸を準備する工程;
2’)シクロオクチン基及びシクロオクチン基以外の官能基(例えば、カルボキシル基、アミノ基、チオール基、ヒドラジド基、アルデヒド基、又は水酸基などを挙げることができるがこれらに限定されない。好ましくはカルボキシル基、アミノ基又はヒドラジド基、より好ましくはカルボキシル基又はアミノ基であるのがよい)を有する物質を準備する工程;
3) 1)ヒアルロン酸を、水にも有機溶媒にも可溶な長鎖アルキルアンモニウムカチオンを持つ塩(例えば、テトラブチルアンモニウム塩、トリオクチルメチルアンモニウム塩、ベンジルジメチルオクタデシルアンモニウム塩など、好ましくはテトラブチルアンモニウム塩、トリオクチルメチルアンモニウム塩、より好ましくはテトラブチルアンモニウム塩を挙げることができるがこれらに限定されない)でイオン交換し有機溶媒に可溶化させ、可溶化体の溶液を調製する工程;
4) 2)物質の有機溶媒の溶液を得る工程;及び
5) 4)溶液と3)可溶化体の溶液とを混合し、且つカルボキシル化反応により官能基とヒアルロン酸のOH基とを反応させる工程;
を有することにより、シクロオクチン基又はシクロオクチン誘導体基を有するヒアルロン酸を得ることができる。
工程2’)は、工程1)とは別に、シクロオクチン基及びシクロオクチン基以外の官能基を有する物質を準備する工程である。該物質は、シクロオクチン基及びシクロオクチン基を有するヒアルロン酸において、上述のスペーサー基X4及びシクロオクチン基又はシクロオクチン基誘導体となる箇所である。
2’)-1) 水酸基及びエステル基を有する物質(例えば、グリコール酸メチル、乳酸メチル、tert-ブチルー4―ヒドロキシブチレートなどを挙げることができるがこれらに限定されない。好ましくはグリコール酸メチル又は乳酸メチル、より好ましくはグリコール酸メチルであるのがよい)と、シクロへプテンのブロモホルム付加体とを、有機溶媒中、触媒(例えば、トリフルオロメタンスルホン酸銀を挙げることができるがこれらに限定されない)存在下で反応させて、1-ブロモシクロオクテン基及びエステル基を有する物質を得る工程;
2’)-2) 1-ブロモシクロオクテン基及びエステル基を有する物質を、溶媒中、脱臭素アルキン化して、シクロオクチン基及びエステル基を有する物質を得る工程;及び
2’)-3) シクロオクチン基及びエステル基を有する物質を加水分解反応に付す工程;
を有することにより得ることができる。
水酸基及びエステル基を有する物質として、例えば、グリコール酸メチル、乳酸メチル、tert-ブチルー4―ヒドロキシブチレートなどを挙げることができるがこれらに限定されない。好ましくはグリコール酸メチル又は乳酸メチル、より好ましくはグリコール酸メチルであるのがよい。
有機溶媒として、用いる水酸基及びエステル基を有する物質、用いる触媒などに依存するが、トルエン、ベンゼン、シクロヘキサン、又はノルマルヘキサンなどを挙げることができるがこれらに限定されない。
触媒として、用いる水酸基及びエステル基を有する物質、用いる有機溶媒などに依存するが、例えば、トリフルオロメタンスルホン酸銀を挙げることができるがこれらに限定されない。
工程2’)-1)は、用いる水酸基及びエステル基を有する物質、用いる有機溶媒、用いる触媒などに依存するが、温度20~40℃、pH4~12の条件で行うのがよい。
溶媒として、トルエン、ベンゼン、シクロヘキサン、又はノルマルヘキサンなどを挙げることができるがこれらに限定されない。
工程2’)-2)は、温度20~40℃、pH4~12の条件で行うのがよい。
工程2’)-3)は、温度20~40℃、pH4~12の条件で行うのがよい。
本願は、ハイドロゲルの原料である特定のカルボキシメチルデキストランの製造方法を提供する。
なお、アジド基を有するカルボキシメチルデキストラン及びシクロオクチン基又はシクロオクチン誘導体基を有するカルボキシメチルデキストランの製造方法は、それぞれ、アジド基を有するヒアルロン酸及びシクロオクチン基又はシクロオクチン誘導体基を有するヒアルロン酸の製造方法において、ヒアルロン酸の代わりにカルボキシメチルデキストランを用いることによって、提供することができる。
<<イミダゾール-1-スルホニルアジドの合成>>
飽和炭酸水素ナトリウム水溶液100mLを、水相除去後の漏斗に加えて有機相を洗浄した。この操作を3回繰り返した。
有機相を回収し、硫酸マグネシウムを加えて室温で一晩乾燥し、その後、硫酸マグネシウムを除去し、エバポレーターによって溶媒を留去した。さらに、数日間、真空乾燥させ、無色透明の題記目的物(収量5.14g(29.7mmol)、収率60.3%)を得た。
ヒアルロン酸のAGU(Anhydrous Glucose Units:繰返単位):カルボニルイミダゾール(CDI):γ-アジド酪酸の量論比が1:2:2となるように、それぞれを用いた。
ナスフラスコにヒアルロン酸(Mw=80,000Da)を加え、DMSOに溶解した。
別途、ナスフラスコにγ-アジド酪酸及びCDIを加え、ジメチルスルホキシド(DMSO)に溶解してγ-アジド酪酸のカルボキシル基を活性化させ、1時間反応させた。
反応物を、ヒアルロン酸のDMSO溶液に滴下した。なお、滴下10分前から60℃に加熱し、16~20時間後、加熱、撹拌を終了し、室温で静置した。静置後一時間後に、純水3L中で透析し、1時間ごとに水を取り換えた。12時間に1回透析液を交換して3日間透析を続けた後、ファルコンチューブに回収し、凍結乾燥し、アジド基を有するヒアルロン酸A-1を得た。1H-NMRにより、用いたヒアルロン酸(Mw=80,000Da)由来のプロトンと側鎖であるアジド基側に由来するプロトンとの存在比から、用いたヒアルロン酸(Mw=80,000Da)の全-OH基を100mol%とした場合、アジド基は、10.8mol%で存在することを確認した。
実施例1のヒアルロン酸(Mw=80,000Da)の代わりに、ヒアルロン酸(Mw=800,000Da)を用いた以外、実施例1と同様の方法により、アジド基を有するヒアルロン酸A-2を得た。実施例1と同様に、1H-NMRにより、アジド基は、12.6mol%で存在することを確認した。
実施例1のヒアルロン酸(Mw=80,000Da)の代わりに、カルボキシメチルデキストラン(Mw=40,000Da)を用いて、AGU:CDI:γ-アジド酪酸の量論比が1:3:3とした以外、実施例1と同様の方法により、アジド基を有するカルボキシメチルデキストランA-3を得た。実施例1と同様に、1H-NMRにより、アジド基は、6.9mol%で存在することを確認した。
実施例3のカルボキシメチルデキストラン(Mw=40,000Da)の代わりに、カルボキシメチルデキストラン(Mw=160,000Da)を用いた以外、実施例3と同様の方法により、アジド基を有するカルボキシメチルデキストランA-4を得た。実施例1と同様に、1H-NMRにより、アジド基は、22.2mol%で存在することを確認した。
実施例3のカルボキシメチルデキストラン(Mw=40,000Da)の代わりに、カルボキシメチルデキストラン(Mw=580,000Da)を用いた以外、実施例3と同様の方法により、アジド基を有するカルボキシメチルデキストランA-5を得た。実施例1と同様に、1H-NMRにより、アジド基は、27.5mol%で存在することを確認した。
100mLビーカー中で、キトサン0.2696g(1.01mmol per unit)を0.1NHCl 30mLに溶解させた。5分程度で完全に溶解した。20mLナスフラスコ中にアジド酪酸 0.3848g(2.98mmol、2.9eq)を加え、DMSO 3mLに溶解させた。これにHOBt 0.4061g(3.01mmol、3.0eq)を加え、カルボニル基を活性化させた。当該溶液を、上記キトサン溶液に滴下し、さらに、EDC 1.1522g(6.01mmol、6.0eq)を加えた。18時間後撹拌を止め、純水及び炭酸水素ナトリウム中で透析を行った。4日後透析を終了し、凍結乾燥し、目的物であるキトサンアジドを回収した。実施例1と同様に、1H-NMRにより、アジド基の存在比を確認した。
<<シクロへプテンへのブロモホルム付加>>
上記で得られた物質を、E2脱離により、HBrが引き抜かれてアルキンとした後、末端メチルエステルを加水分解し、カルボン酸とした。
ヒアルロン酸のAGU:カルボニルイミダゾール(CDI):上記で得られたシクロオクチン基を有するカルボン酸の量論比が1:2:2となるように、それぞれを用いた。
ナスフラスコにヒアルロン酸(Mw=80,000Da)を加え、DMSOに溶解した。
別途、ナスフラスコに上記で得られたシクロオクチン基を有するカルボン酸及びCDIを加え、ジメチルスルホキシド(DMSO)に溶解して、上記で得られたシクロオクチン基を有するカルボン酸のカルボキシル基を活性化させ、1時間反応させた。
反応物を、ヒアルロン酸のDMSO溶液に滴下した。なお、滴下10分前から60℃に加熱し、16~20時間後、加熱、撹拌を終了し、室温で静置した。静置後一時間後に、純水3L中で透析し、1時間ごとに水を取り換えた。3日間、透析を続けた後、ファルコンチューブに回収し、凍結乾燥し、シクロオクチン基を有するヒアルロン酸B-1を得た。
1H-NMRにより、用いたヒアルロン酸(Mw=80,000Da)由来のプロトンと側鎖であるシクロオクチン基側に由来するプロトンとの存在比から、用いたヒアルロン酸(Mw=80,000Da)の全-OH基を100mol%とした場合、シクロオクチン基は、25.3mol%で存在することを確認した。
実施例6のヒアルロン酸(Mw=80,000Da)の代わりに、ヒアルロン酸(Mw=800,000Da)を用いた以外、実施例6と同様の方法により、シクロオクチン基を有するヒアルロン酸B-2を得た。実施例6と同様に、1H-NMRにより、シクロオクチン基は、14.6mol%で存在することを確認した。
実施例6のヒアルロン酸(Mw=80,000Da)の代わりに、カルボキシメチルデキストラン(Mw=40,000Da)を用いて、AGU:CDI:γ-アジド酪酸の量論比が1:3:3とした以外、実施例6と同様の方法により、シクロオクチン基を有するカルボキシメチルデキストランB-3を得た。実施例6と同様に、1H-NMRにより、シクロオクチン基は、56.6mol%で存在することを確認した。
実施例8のカルボキシメチルデキストラン(Mw=40,000Da)の代わりに、カルボキシメチルデキストラン(Mw=160,000Da)を用いた以外、実施例8と同様の方法により、シクロオクチン基を有するカルボキシメチルデキストランB-4を得た。実施例6と同様に、1H-NMRにより、シクロオクチン基は、12.4mol%で存在することを確認した。
実施例8のカルボキシメチルデキストラン(Mw=40,000Da)の代わりに、カルボキシメチルデキストラン(Mw=580,000Da)を用いた以外、実施例8と同様の方法により、シクロオクチン基を有するカルボキシメチルデキストランB-5を得た。実施例6と同様に、1H-NMRにより、シクロオクチン基は、25.3mol%で存在することを確認した。
実施例8のカルボキシメチルデキストラン(Mw=40,000Da)の代わりに、キトサンを用いた以外、実施例8と同様の方法により、シクロオクチン基を有するキトサンB-6を得た。実施例6と同様に、1H-NMRにより、シクロオクチン基の存在比を確認した。
<<ポリマー溶液の調製>>
アジド基を有するヒアルロン酸A-1をリン酸緩衝生理食塩水(PBS)に溶解し、濃度がそれぞれ、5.0、3.0、2.0、1.5、1.0wt%となるように、アジド基を有するヒアルロン酸A-1のPBS溶液X-1~X-5を調製した。
また、シクロオクチン基を有するヒアルロン酸B-1も同様に、濃度がそれぞれ5.0、3.0、2.0、1.5、1.0wt%となるPBS溶液Y-1~Y-5を調製した。
35mmディッシュ(IWAKI製)上に、PBS溶液X-1 100μLを滴下した後、PBS溶液Y-1 100μLを滴下し、滴下後からゲル化までの時間を測定したところ、8分19秒でハイドロゲルZ-1を得た。
同様に、PBS溶液X-2~X-4とPBS溶液Y-2~Y-4とからハイドロゲルZ-2~Z-4を得た。なお、PBS溶液X-5とPBS溶液Y-5との組合せでは、ゲル化が確認できなかった。これらの結果を表1に示す。
<<ポリマー溶液の調製>>
実施例11において、アジド基を有するヒアルロン酸A-1の代わりにアジド基を有するヒアルロン酸A-2を用い、且つシクロオクチン基を有するヒアルロン酸B-1の代わりにシクロオクチン基を有するヒアルロン酸B-2を用いて、それぞれ、アジド基を有するヒアルロン酸A-2のPBS溶液X-6~X-10及びシクロオクチン基を有するヒアルロン酸B-2のPBS溶液Y-6~Y-10を調製した。
実施例11と同様の方法により、PBS溶液X-6~X-10とPBS溶液Y-6~Y-10とからゲルZ-6~Z-10を調製した。
それらの結果を表2に示す。
また、ハイドロゲルの膨潤度は初期重量を100%として、ヒアルロン酸の100%から400%の間で体積が膨張し、PBS中では250時間以上かけて分解し、10%ウシ血清を含むDMEM培地中では70時間程度で分解消失した。
アジド基を有するカルボキシメチルデキストランA-4及びシクロオクチン基を有するカルボキシメチルデキストランB-4を用いて、ハイドロゲルを調製した。
具体的には、実施例11において、アジド基を有するヒアルロン酸A-1の代わりにアジド基を有するカルボキシメチルデキストランA-4を用い、且つシクロオクチン基を有するヒアルロン酸B-1の代わりにシクロオクチン基を有するカルボキシメチルデキストランB-4を用いて、それぞれ、アジド基を有するカルボキシメチルデキストランA-4のPBS溶液X-11(濃度:5.0wt%)及びシクロオクチン基を有するカルボキシメチルデキストランB-4のPBS溶液Y-11(濃度:5.0wt%)を調製した。
それらの溶液X-11及びY-11をそれぞれ、図2に示すダブルシリンジの各々のシリンジに充填し、35mmディッシュ(IWAKI製)上に滴下したところ、ハイドロゲルZ-11を得た。
具体的には、実施例11において、アジド基を有するヒアルロン酸A-1の代わりにアジド基を有するキトサンA-6を用い、且つシクロオクチン基を有するヒアルロン酸B-1の代わりにシクロオクチン基を有するキトサンB-6を用いて、それぞれ、アジド基を有するキトサンA-6のPBS溶液X-11(濃度:5.0wt%)及びシクロオクチン基を有するキトサンB-6のPBS溶液Y-11(濃度:5.0wt%)を調製した。
それらの溶液X-11及びY-11をそれぞれ、図2に示すダブルシリンジの各々のシリンジに充填し、35mmディッシュ(IWAKI製)上に滴下したところ、ハイドロゲルZ-12を得た。
LIVE/DEAD Assay(ロンザ社)により、ハイドロゲルの細胞毒性について評価した。なお、細胞:NIH 3T3 NIH Swissマウス胎仔由来の線維芽細胞;培地:D-MEM(調整済み);を用いた。
<<ハイドロゲル調製用ポリマー溶液の調製>>
実施例1で得たヒアルロン酸A-1をUV滅菌し、PBSに溶解し、ヒアルロン酸A-1のPBS溶液X’-1(濃度5.0wt%)を得た。
同様に、実施例6で得たヒアルロン酸B-1をUV滅菌し、PBSに溶解し、ヒアルロン酸B-1のPBS溶液Y’-1(濃度5.0wt%)を得た。
細胞:NIH3T3を培養した150cm3フラスコにPBS10mLを加え、洗浄・吸引し、浮遊細胞や死細胞を除去した。また、同フラスコにトリプシン5mLを加え、5分間37℃恒温槽でインキュベートし、細胞を剥離させた。
同フラスコに培地20mLを加え、剥離した細胞をトリプシンごと50mLファルコンチューブに回収した。一部を取り分け、トリパンブルーで染色して細胞数をカウントした。
ファルコンチューブに1000rpm、3分間、遠心分離を施し、細胞をチューブ底に集めた。
上澄み液をアスピレーターで除去し、PBSを加え、細胞分布が均一となるように、細胞濃度が5×106個/mLとなる細胞懸濁液を調製した。
得られたPBS溶液X”-1及びY”-1をダブルシリンジ(バクスター社製)の各々に充填し、35mmガラスボトムディッシュ(マツナミ)上に混合しながら滴下した。2時間程室温で静置し、ハイドロゲルZ-14を得た。
その後、ディッシュに新しい培地4mLを加え、37℃恒温槽でインキュベートした。2日後、アスピレーターで培地を吸引し、ディッシュ上に残ったゲルをPBSに浸してゲルZ-14中に浸透した培地を除去した。ハイドロゲルZ-14から培地が完全に除去されたのを確認し、CalceinAM及びEthD-1を各々100μL添加した。30分程静置し、共焦点顕微鏡を用いてゲルZ-14内部の蛍光画像を取得した。それぞれの色素で染色された細胞数の比から、ゲルZ-14に封入された細胞の生存率を算出した。具体的には、画質改善、ノイズ除去、オブジェクト分離、手動補正等を行い、一定の蛍光強度、蛍光半径のものを拾ってカウントした。ここで、生細胞を示す緑色蛍光強度と、死細胞を示す赤色蛍光強度の比を生存率とし、両者で蛍光が重なる細胞を死細胞として敢えてカウントしたとしても、該生存率は、一日目生存率が68%、二日目生存率が64%であった。この結果から、ハイドロゲルZ-14は、生体適合性を有することが確認された。
実際に生きたラットに、ヒアルロン酸ベースのポリマー溶液を皮下注射してin situで生体内にハイドロゲルを形成し、周辺組織や代謝系臓器にどのような影響が表れるかを組織的に評価した。
<<ハイドロゲル調製用ポリマー溶液の調製>>
実施例1で得たヒアルロン酸A-1をUV照射によって3時間滅菌処理を行い、PBSに溶解し、ヒアルロン酸A-1のPBS溶液X’-3(濃度2.0wt%)を得た。
同様に、実施例6で得たヒアルロン酸B-1をUV滅菌し、PBSに溶解し、ヒアルロン酸B-1のPBS溶液Y’-3(濃度2.0wt%)を得た。
得られたPBS溶液X’-3及びY’-3を、ダブルシリンジの各々に充填し、9週齢のSDラット♂の頚部に、各々のポリマー溶液を、ダブルシリンジを用いて混合しながら、合計1mLを注入した。注入後、ラットに急性毒性症状は観察されず、一週間の健康状態も良好であった。
一週間後にマウスを安楽死させ、表皮切開および腹腔開腹により各臓器を観察した。ゲル周辺組織、肝臓、脾臓、腎臓を切除し、ホルマリンに浸けて保存し、後日、各臓器の組織断片を作製し、ヘマトキシン・エオシンにより染色(HE染色)した。作製した標本を光学顕微鏡で観察し、組織的評価を行った。得られた真皮組織の染色図を図3に示す。
真皮組織中にゲルの形成を確認した。したがって、ハイドロゲルがin vivo調製され、且つ最大で一週間も残存していたことがわかった。
図3より、ポリマー溶液の注入時に出血した際の赤血球が多少混入しており、ゲル周辺には広範囲の炎症が確認されたが、HE染色後の組織評価において、炎症の度合いは従来の生体適合性を有するバイオマテリアル移植時に生じる異物反応とそれほど変わらず、組織内部では炎症は見られなかった。したがって、本件発明に係るゲルは、高い生体適合性を有し、新たなin situ架橋ゲルとしての可能性が示唆された。
<<<肝臓>>>
肝臓表面に変色が確認された。原因は明らかではないが、HE染色後の組織評価の結果、変色は表面のみで、内部の肝組織に目立った影響は見られなかった。肝表面付近の類洞が変形しており、何らかの影響により肝細胞が収縮した可能性が高い。本実験でポリマー溶液を注入したのは頚部皮下であり、分解物は血中あるいはリンパ中に移行して臓器内部から浸潤していくと考えられるが、何故表面部分に影響が現れたのかは不明である。
脾臓は異物を食作用により分解するマクロファージや、B細胞・T細胞などの免疫抗体を産生する器官である。腎臓は血中の溶質のうち分子量50,000以下のものを糸球体濾過により尿に移行させ、体外に排出させる器官である。これらは肝臓と共に、材料を投与した際にその影響が最も強く現れる器官であるため、それぞれ回収してHE染色を行い、組織的に評価した。その結果、肝表面の変色以外、脾臓、腎臓には特に目立った影響は見られず、病理組織学的に正常に保たれていることが明らかとなった。
以上の結果より、PBS溶液X’-3及びY’-3並びにそれによって形成されたハイドロゲルは、生体内に入れても急性毒性は無いことがわかった。また、PBS溶液は、in situ架橋性があることが示された。
Claims (54)
- a)ヒアルロン酸、カルボキシメチルデキストラン、セルロース誘導体及びキトサンからなる群から選ばれる第1のポリマー部位;
b)上記a)と同種であっても異種であってもよいが、上記a)とは異なる分子からなる、ヒアルロン酸、カルボキシメチルデキストラン、セルロース誘導体及びキトサンからなる群から選ばれる第2のポリマー部位;及び
c)トリアゾール環基又はその誘導体基;
を有するハイドロゲルであって、
該ハイドロゲルは、前記c)トリアゾール環基又はその誘導体基を介して、前記a)第1のポリマー部位と前記b)第2のポリマー部位とが架橋する構造を有する、上記ハイドロゲル。 - 前記a)第1のポリマー部位と前記c)トリアゾール環基又はその誘導体基との間にスペーサー基X1を有し、ここで、当該X1は、エーテル基、エステル基、アミド基、ヒドラジド基、ジスルフィド基、及びカルボニル基からなる群から選択される1以上の置換基を有するアルキレン基である、請求項1記載のハイドロゲル。
- 前記b)第2のポリマー部位と前記c)トリアゾール環基又はその誘導体基との間にスペーサー基X2を有し、ここで、当該X2は、エーテル基、エステル基、アミド基、ヒドラジド基、ジスルフィド基、及びカルボニル基からなる群から選択される1以上の置換基を有するアルキレン基である、請求項1又は2記載のハイドロゲル。
- 前記X1は、前記a)第1のポリマー部位の-OH基を置換して結合し、-O-X1-を形成する請求項2又は3に記載のハイドロゲル。
- 前記X2は、前記b)第2のポリマー部位の-OH基を置換して結合し、-O-X2-を形成する請求項3又は4に記載のハイドロゲル。
- 前記X1は、前記a)第1のポリマー部位の-NH2基を置換して結合し、-NH-X1-を形成する請求項2又は3に記載のハイドロゲル。
- 前記X2は、前記b)第2のポリマー部位の-NH2基を置換して結合し、-NH-X2-を形成する請求項3又は6に記載のハイドロゲル。
- 前記-X1-が、-CO-(CH2)3-である請求項2~7のいずれか1項記載のハイドロゲル。
- 前記-X1-が、-CO-NH-(CH2CH2O)m-NH-CO-(CH2)3-(ここで、mは1以上の整数を示す)である請求項2~7のいずれか1項記載のハイドロゲル。
- 前記-X2-が、-CO-CH2-O-である請求項3~9のいずれか1項記載のハイドロゲル。
- 前記-X2-が、-CO-NH-(CH2CH2O)m-NH-CO-CH2-O-(ここで、mは1以上の整数を示す)である請求項3~9のいずれか1項記載のハイドロゲル。
- 前記c)トリアゾール環基又はその誘導体基は、前記a)第1のポリマー部位の全-OH基又は全-NH2基を100mol%とした場合、5~60mol%で存在する請求項1~11のいずれか1項記載のハイドロゲル。
- 前記c)トリアゾール環基又はその誘導体基は、前記b)第2のポリマー部位の全-OH基又は全-NH2基を100mol%とした場合、5~60mol%で存在する請求項1~12のいずれか1項記載のハイドロゲル。
- 前記a)第1のポリマー部位;及び前記b)第2のポリマー部位が、ヒアルロン酸である請求項1~13のいずれか1項記載のハイドロゲル。
- 前記a)第1のポリマー部位;及び前記b)第2のポリマー部位が、カルボキシメチルデキストランである請求項1~13のいずれか1項記載のハイドロゲル。
- 前記a)第1のポリマー部位;及び前記b)第2のポリマー部位が、キトサンである請求項1~13のいずれか1項記載のハイドロゲル。
- 請求項1~16のいずれか1項記載のハイドロゲルを有する生体適合性材料。
- 前記生体適合性材料が癒着防止材料である請求項17記載の材料。
- 前記生体適合性材料がドラッグデリバリシステム担体である請求項17記載の材料。
- 前記生体適合性材料が細胞封入用材料である請求項17記載の材料。
- 請求項1~16のいずれか1項記載のハイドロゲルを作成するためのキット。
- A) a)ヒアルロン酸、カルボキシメチルデキストラン、セルロース誘導体及びキトサンからなる群から選ばれる第1のポリマーであって、且つ該第1のポリマーがアジド基を有する、第1のポリマーを含む液を準備する工程;
B) b)上記a)と同種であっても異種であってもよいが、上記a)とは異なる分子からなる、ヒアルロン酸、カルボキシメチルデキストラン、セルロース誘導体及びキトサンからなる群から選ばれる第2のポリマーであって、且つ該第2のポリマーがシクロオクチン基又はシクロオクチン基誘導体を有する、第2のポリマーを含む液を準備する工程;及び
C) 前記a)第1の液と前記b)第2の液とを混合する工程;
を有することにより、前記アジド基とシクロオクチン基又はシクロオクチン基誘導体とのクリック反応により、トリアゾール環又はその誘導体が形成され、該トリアゾール環又はその誘導体を介して、前記第1のポリマーと前記第2のポリマーとを架橋する構造を有するハイドロゲルを形成する、ハイドロゲルの製造方法。 - 工程C)が生体内で行われることを特徴とする、請求項22に記載の製造方法。
- 請求項22又は23記載の製造方法によりハイドロゲルを作成するためのキット。
- -OH基の少なくとも一部が-O-X3-N3基に置換されたヒアルロン酸であって、ここで、当該X3は、エーテル基、エステル基、アミド基、ヒドラジド基、ジスルフィド基、及びカルボニル基からなる群から選択される1以上の置換基を有するアルキレン基である、当該ヒアルロン酸。
- 前記-X3-が、-CO-(CH2)3-である請求項25記載のヒアルロン酸。
- 前記-X3-が、-CO-NH-(CH2CH2O)m-NH-CO-(CH2)3-(ここで、mは1以上の整数を示す)である請求項25記載のヒアルロン酸。
- -OH基の少なくとも一部が-O-X4-(シクロオクチン又はシクロオクチン誘導体)基に置換されたヒアルロン酸であって、ここで、当該X4は、エーテル基、エステル基、アミド基、ヒドラジド基、ジスルフィド基、及びカルボニル基からなる群から選択される1以上の置換基を有するアルキレン基である、当該ヒアルロン酸。
- 前記-X4-が、-CO-CH2-O-である請求項29項記載のヒアルロン酸。
- 前記-X4-が、-CO-NH-(CH2CH2O)m-NH-CO-CH2-O-(ここで、mは1以上の整数を示す)である請求項27記載のヒアルロン酸。
- 下記式(2)(式(2)中、Rのうち少なくとも1つは式(2)-1で表す基であり、それ以外はHを示し、n’は100~20,000の整数を示す)を有する請求項29又は30記載のヒアルロン酸。
- -OH基の少なくとも一部が-O-X3-N3基に置換されたカルボキシメチルデキストランであって、ここで、当該X3は、エーテル基、エステル基、アミド基、ヒドラジド基、ジスルフィド基、及びカルボニル基からなる群から選択される1以上の置換基を有するアルキレン基である、当該カルボキシメチルデキストラン。
- 前記-X3-が、-CO-(CH2)3-である請求項33記載のカルボキシメチルデキストラン。
- 前記-X3-が、-CO-NH-(CH2CH2O)m-NH-CO-(CH2)3-(ここで、mは1以上の整数を示す)である請求項33記載のカルボキシメチルデキストラン。
- -OH基の少なくとも一部が-O-X4-(シクロオクチン又はシクロオクチン誘導体)基に置換されたカルボキシメチルデキストランであって、ここで、当該X4は、エーテル基、エステル基、アミド基、ヒドラジド基、ジスルフィド基、及びカルボニル基からなる群から選択される1以上の置換基を有するアルキレン基である、当該カルボキシメチルデキストラン。
- 前記-X4-が、-CO-CH2-O-である請求項37項記載のカルボキシメチルデキストラン。
- 前記-X4-が、-CO-NH-(CH2CH2O)m-NH-CO-CH2-O-(ここで、mは1以上の整数を示す)である請求項37記載のカルボキシメチルデキストラン。
- -NH2基が-NH-X3-N3基に置換されたキトサンであって、ここで、当該X3は、エーテル基、エステル基、アミド基、ヒドラジド基、ジスルフィド基、及びカルボニル基からなる群から選択される1以上の置換基を有するアルキレン基である、当該キトサン。
- 前記-X3-が、-CO-(CH2)3-である請求項41記載のキトサン。
- 前記-X3-が、-CO-NH-(CH2CH2O)m-NH-CO-(CH2)3-(ここで、mは1以上の整数を示す)である請求項41記載のキトサン。
- -NH2基が-NH-X4-(シクロオクチン又はシクロオクチン誘導体)基に置換されたキトサンであって、ここで、当該X4は、エーテル基、エステル基、アミド基、ヒドラジド基、ジスルフィド基、及びカルボニル基からなる群から選択される1以上の置換基を有するアルキレン基である、当該キトサン。
- 前記-X4-が、-CO-CH2-O-である請求項44項記載のキトサン。
- 前記-X4-が、-CO-NH-(CH2CH2O)m-NH-CO-CH2-O-(ここで、mは1以上の整数を示す)である請求項44記載のキトサン。
- -OH基の少なくとも一部が-O-X3-N3基(式中、X3は、単結合、又は分子量1万以下の基を表す)に置換されたヒアルロン酸の製造方法であって、
1)ヒアルロン酸を準備する工程;
2)アジド基及びアジド基以外の官能基を有する物質を準備する工程;
3)前記1)ヒアルロン酸を水にも有機溶媒にも可溶な長鎖アルキルアンモニウムカチオンを持つ塩でイオン交換し有機溶媒に可溶化させ、可溶化体の溶液を調製する工程;
4) 前記2)物質の有機溶媒の溶液を得る工程;及び
5) 前記4)溶液と前記3)可溶化体の溶液とを混合し、且つカルボキシル化反応により前記官能基と前記ヒアルロン酸のOH基とを反応させる工程;
を有することにより、上記ヒアルロン酸を得る、上記方法。 - 前記2)物質の官能基がカルボン酸基であり、
該物質を、
2)-1)分子量1万以下であり、一端がアミノ基、他端がカルボン酸基を有する化合物を準備する工程;及び
2)-2)前記化合物を、溶媒中、触媒存在下で、アジド化物と反応させる工程;
を有することにより得られる請求項47記載の方法。 - -OH基の少なくとも一部が-O-X4-(シクロオクチン又はシクロオクチン誘導体)基(式中、X4は、単結合、又は分子量1万以下の基を表す)に置換されたヒアルロン酸の製造方法であって、
1)ヒアルロン酸を準備する工程;
2’)シクロオクチン基及びシクロオクチン基以外の官能基を有する物質を準備する工程;
3)前記1)ヒアルロン酸を水にも有機溶媒にも可溶な長鎖アルキルアンモニウムカチオンを持つ塩でイオン交換し有機溶媒に可溶化させ、可溶化体の溶液を調製する工程;
4) 前記2)物質の有機溶媒の溶液を得る工程;及び
5) 前記4)溶液と前記3)可溶化体の溶液とを混合し、且つカルボキシル化反応により前記官能基と前記ヒアルロン酸のOH基とを反応させる工程;
を有することにより、上記ヒアルロン酸を得る、上記方法。 - 前記2’)物質の官能基がカルボン酸基であり、
該物質を、
2’)-1) 水酸基及びエステル基を有する物質と、シクロへプテンのブロモホルム付加体とを、有機溶媒中、触媒存在下で反応させて、1-ブロモシクロオクテン基及びエステル基を有する物質を得る工程;
2’)-2) 1-ブロモシクロオクテン基及びエステル基を有する物質を、溶媒中、脱臭素アルキン化して、シクロオクチン基及びエステル基を有する物質を得る工程;及び
2’)-3) シクロオクチン基及びエステル基を有する物質を加水分解反応に付す工程;
を有することにより得られる請求項49記載の方法。 - -OH基の少なくとも一部が-O-X3-N3基(式中、X3は、単結合、又は分子量1万以下の基を表す)に置換されたカルボキシメチルデキストランの製造方法であって、
1’)カルボキシメチルデキストランを準備する工程;
2)アジド基及びアジド基以外の官能基を有する物質を準備する工程;
3’)前記1’)カルボキシメチルデキストランを水にも有機溶媒にも可溶な長鎖アルキルアンモニウムカチオンを持つ塩でイオン交換し有機溶媒に可溶化させ、可溶化体の溶液を調製する工程;
4) 前記2)物質の有機溶媒の溶液を得る工程;及び
5’) 前記4)溶液と前記3)可溶化体の溶液とを混合し、且つカルボキシル化反応により前記官能基と前記カルボキシメチルデキストランのOH基とを反応させる工程;
を有することにより、上記カルボキシメチルデキストランを得る、上記方法。 - 前記2)物質の官能基がカルボン酸基であり、
該物質を、
2)-1)分子量1万以下であり、一端がアミノ基、他端がカルボン酸基を有する化合物を準備する工程;及び
2)-2)前記化合物を、溶媒中、触媒存在下で、アジド化物と反応させる工程;
を有することにより得られる請求項51記載の方法。 - -OH基の少なくとも一部が-O-X4-(シクロオクチン又はシクロオクチン誘導体)基(式中、X4は、単結合、又は分子量1万以下の基を表す)に置換されたカルボキシメチルデキストランの製造方法であって、
1’)カルボキシメチルデキストランを準備する工程;
2’)シクロオクチン基及びシクロオクチン基以外の官能基を有する物質を準備する工程;
3’)前記1’)カルボキシメチルデキストランを水にも有機溶媒にも可溶な長鎖アルキルアンモニウムカチオンを持つ塩でイオン交換し有機溶媒に可溶化させ、可溶化体の溶液を調製する工程;
4) 前記2)物質の有機溶媒の溶液を得る工程;及び
5’) 前記4)溶液と前記3’)可溶化体の溶液とを混合し、且つカルボキシル化反応により前記官能基と前記カルボキシメチルデキストランのOH基とを反応させる工程;
を有することにより、上記’)カルボキシメチルデキストランを得る、上記方法。 - 前記2’)物質の官能基がカルボン酸基であり、
該物質を、
2’)-1) 水酸基及びエステル基を有する物質と、シクロへプテンのブロモホルム付加体とを、有機溶媒中、触媒存在下で反応させて、1-ブロモシクロオクテン基及びエステル基を有する物質を得る工程;
2’)-2) 1-ブロモシクロオクテン基及びエステル基を有する物質を、溶媒中、脱臭素アルキン化して、シクロオクチン基及びエステル基を有する物質を得る工程;及び
2’)-3) シクロオクチン基及びエステル基を有する物質を加水分解反応に付す工程;
を有することにより得られる請求項53記載の方法。
