- 제15항의 자동정제장치를 이용하여 생물학적 시료부터 타겟물질을 추출하는 방법으로서,상기 피펫(141, 142)을 이용하여 상기 멀티웰플레이트(420, 420')의 웰에 주입된 세포용해용액을 상기 생물학적 시료와 혼합하는 혼합단계(S1010, S1020, 1030);상기 자석장착부(710)를 상승시킨 상태에서 상기 히팅부(720)를 이용하여 상기 멀티웰플레이트(420, 420')의 상기 특정 단위 웰을 가열하여 상기 생물학적 시료에 열을 가하는 제1 가열단계(S1040);상기 피펫(141, 142)을 이용하여 상기 세포용해용액과 상기 생물학적 시료의 혼합물을 상기 멀티웰플레이트(420, 420')의 웰에 주입된 결합용액과 혼합하는 결합용액과의 혼합단계(S1050);상기 피펫(141, 142)을 이용하여 상기 결합용액과 혼합된 혼합물을 상기 멀티웰플레이트(420, 420')의 웰에 주입된 자성입자 수분산액과 혼합하는 자성입자 수분산액과의 혼합단계(S1060);상기 자석장착부(710)를 상승시켜 상기 자석장착부(710)로부터 상기 자성입자 수분산액과 혼합된 혼합물이 들어 있는 상기 멀티웰플레이트(420, 420')의 상기 특정 단위 웰 하부에 자기장을 인가하는 제1 자기장 인가단계(S1080);상기 자성입자 수분산액과 혼합된 혼합물 중 상기 자성입자 수분산액의 자성입자 및 상기 자성입자에 부착된 부착물이 상기 멀티웰플레이트(420, 420')의 상기 특정 단위 웰 하부의 내벽에 부착된 상태에서, 상기 피펫(141, 142)을 이용하여 상기 자성입자 수분산액과 혼합된 혼합물 중 상기 자성입자 및 상기 자성입자에 부착된 부착물을 제외한 혼합물을 제거하는 제1 제거단계(S1090);상기 피펫(141, 142)을 이용하여 상기 멀티웰플레이트(420, 420')의 웰에 주입된 세척용액을 상기 멀티웰플레이트(420, 420')의 상기 특정 단위 웰에 주입하여 상기 자성입자로부터 상기 타겟물질을 제외한 불순물을 세척 및 분리하는 특정 단위 웰로의 제3 주입 및 세척 단계(S1100);상기 세척용액과 혼합된 혼합물 중 상기 타겟물질이 부착된 상기 자성입자가 상기 멀티웰플레이트(420, 420')의 상기 특정 단위 웰 하부의 내벽에 부착된 상태에서, 상기 피펫(141, 142)을 이용하여 상기 세척용액과 혼합된 혼합물 중 상기 타겟물질이 부착된 상기 자성입자를 제외한 혼합물을 제거하는 제2 제거단계(S1120);상기 피펫(141, 142)을 이용하여 상기 멀티웰플레이트(420, 420')의 웰에 주입된 핵산 용출용액을 상기 멀티웰플레이트(420, 420')의 상기 특정 단위 웰에 주입하여 상기 자성입자로부터 상기 타겟물질을 분리시키는 특정 단위 웰로의 제4 주입 및 핵산 분리 단계(S1140);상기 자성입자로부터 분리된 상기 타겟물질이 포함된 핵산 용출용액 중 상기 자성입자가 상기 멀티웰플레이트(420, 420')의 상기 특정 단위 웰 하부의 내벽에 부착된 상태에서, 상기 피펫(141, 142)을 이용하여 상기 자성입자로부터 분리된 상기 자성입자를 제외한 타겟물질 함유용액을 획득하는 타겟물질 함유용액 획득 단계(S1160);를 포함하는 것을 특징으로 하는 생물학적 시료부터 타겟물질을 추출하는 방법.
- 제17항에 있어서,상기 세척용액은 알코올을 포함하고,상기 특정 단위 웰로의 제4 주입 및 핵산 분리 단계(S1140) 전에 상기 자석장착부(710)를 상승시켜 상기 멀티웰플레이트(420, 420')의 상기 특정 단위 웰 하부가 상기 단위 웰 인입홈(713)에 인입된 상태에서, 상기 히팅부(720)를 이용하여 상기 멀티웰플레이트(420, 420')의 상기 특정 단위 웰 하부를 가열하여 상기 자성입자에 잔류된 세척용액 중의 알코올을 제거하는 제2 가열 단계(S1130)를 포함하는 것을 특징으로 생물학적 시료로부터 타겟물질을 추출하는 방법.
- 제17항에 있어서,상기 타겟물질 함유용액 획득 단계(S1160)는 상기 피펫(141, 142)을 이용하여 상기 타겟물질 함유용액을 상기 베이스플레이트(400)에 탑재된 타겟물질 보관용 튜브(442-1) 또는 타겟물질 진단용 튜브(442-3)에 주입하는 것을 포함하는 것을 특징으로 하는 생물학적 시료로부터 타겟물질을 추출하는 방법.
- 다수의 생물학적 시료로부터 타겟물질을 분리하는 자동정제장치에 있어서,수평방향 및 수직방향으로 이동 가능하게 설치되며, 유동성 물질이 흡입 및 토출되는 다수의 피펫(141, 142)을 분리 가능하도록 장착하기 위한 피펫블록(100);베이스플레이트(400)에 탑재되어 상기 피펫블록(100)의 하방에 위치하는 멀티웰플레이트(420, 420')의 특정 단위 웰에 자기장을 인가하기 위한 자기장 인가부(700);상기 베이스플레이트(400)에 탑재되며 분리된 상기 타겟물질을 수용하기 위한 타겟물질용 튜브(442-1, 442-3)를 삽착 수용시키기 위한 제1 튜브랙(440);상기 다수의 피펫(141, 142)으로부터 토출되는 상기 타겟물질로부터 발생되는 에어로졸이 상기 타겟물질용 튜브(442-1, 442-3) 외부로 확산되는 것을 방지하도록 상기 베이스플레이트(400)에 탑재되어 상기 타겟물질용 튜브(442-1, 442-3) 상단을 차단하는 오염방지기(460);를 포함하는 것을 특징으로 하는 자기장 인가부를 구비한 생물학적 시료 자동정제장치.
- 제20항에 있어서, 상기 오염방지기(460)는,상기 다수의 피펫(141, 142)에 의한 하방 압박력에 의하여 벌어져 상기 다수의 피펫(141, 142)의 하부를 통과시키기 위한 절개선(461-1)이 형성된 덮개필름(461);상기 베이스플레이트(400)에 탑재되며 상기 절개선이(461-1)이 상기 타겟물질용 튜브(442-1, 442-3)의 상방에 위치하도록 상기 덮개필름(461)을 지지하는 필름지지대;를 포함하는 것을 특징으로 하는 자기장 인가부를 구비한 생물학적 시료 자동정제장치.
- 제21항에 있어서, 상기 필름지지대는,상기 타겟물질용 튜브(442-1, 442-3)의 상부 끝단이 끼워지는 통공(463-1)이 형성되며 상기 덮개필름(461)이 안착되는 안착판(463);상기 덮개필름(461)의 둘레면과 접촉함으로써 상기 덮개필름(461)의 수평방향 이동을 억제하도록 상기 안착판(463)의 상면에 세워지는 수평이동 억제판(465);을 포함하는 것을 특징으로 하는 자기장 인가부를 구비한 생물학적 시료 자동정제장치.
- 제22항에 있어서, 상기 필름지지대는,상기 덮개필름(461)의 수직방향 이동을 억제하도록 상기 덮개필름(461)의 상면에 안착되고, 상기 덮개필름(461)의 절개선(461-1)을 노출시키는 노출공(467-1)이 형성되는 수직이동 억제판(467);을 포함하는 것을 특징으로 하는 자기장 인가부를 구비한 생물학적 시료 자동정제장치.
- 제23항에 있어서, 상기 필름지지대는,상기 제1 튜브랙(440)에 외삽되어 하단이 베이스플레이트(400)에 안착되며 상단이 상기 안착판(463)에 연결되는 안착판 지지대(469)를 포함하는 것을 특징으로 하는 자기장 인가부를 구비한 생물학적 시료 자동정제장치.
- 제22항 내지 제24항 중 어느 한 항에 있어서,상기 안착판(463)과 상기 덮개필름(461) 사이에는 상기 다수의 피펫(141, 142)에 의한 하방 압박력에 의하여 절취 가능한 은박지(462)가 탑재되는 것을 특징으로 하는 자기장 인가부를 구비한 생물학적 시료 자동정제장치.
- 제20항 내지 제24항 중 어느 한 항에 있어서,상기 타겟물질용 튜브(442-1, 442-3)는 타겟물질 보관용 튜브(442-1)와 진단용 튜브(442-3) 중 적어도 어느 하나를 포함하는 것을 특징으로 하는 자기장 인가부를 구비한 생물학적 시료 자동정제장치.
- 제20항 내지 제24항 중 어느 한 항에 있어서,상기 자기장 인가부(700)가 상승하여 상기 멀티웰플레이트(420, 420')의 특정 단위 웰 하부에 자기장을 인가하도록 상기 베이스플레이트(400) 중 상기 베이스플레이트(400)에 형성된 단위 웰 노출공(400-3)에 인접하여 탑재되며 상기 피펫블록(100)에 장착된 다수의 피펫(141, 142)으로부터 버려지는 폐액을 수용하기 위한 폐액통(450);상기 베이스플레이트(400) 중 상기 폐액통(450)을 중심으로 상기 단위 웰 노출공(400-3)과 마주보도록 상기 폐액통(450)에 인접하여 탑재되며 상기 생물학적 시료가 투입된 생물학적 시료용 튜브(472)를 삽착 수용하기 위한 제2 튜브랙(470);을 포함하는 것을 특징으로 하는 자기장 인가부를 구비한 생물학적 시료 자동정제장치.
- 제1항 내지 제7항 중 어느 한 항의 자동정제장치를 이용하여 자동으로 단백질을 발현시키고 분리하는 것을 특징으로 하는 단백질 발현 및 정제 방법.
