- Patent Document 1 applying the antigen-antibody reaction and mass spectrometry is disclosed. This is intended for protein quantification. First, an enzyme is added to a sample such as serum to digest a protein into a peptide. Next, a stable isotope-labeled internal standard substance is added to the antibody-magnetic bead on which the anti-peptide antibody is immobilized and the peptide as the measurement target component, and reacted to react the peptide and internal standard substance as the measurement target component with the antibody-magnetic Bind to beads. Then, the processed specimen is introduced into a device that can concentrate the component to be measured and can be detected by mass spectrometry, and the peptide is quantified.

- Patent Document 1 a pipe having an inner diameter of 50 to 300 ⁇ m is connected online via a plurality of valves in an apparatus capable of concentrating a measurement target component and detecting it by mass spectrometry.
- the measurement is performed using a high-performance liquid chromatograph / mass spectrometer (LC-ESI-MS), so the measurement time is long, and the throughput required as a general clinical laboratory equipment 200 inspection / hour (Non-patent Document 3) cannot be achieved.
- LC-ESI-MS high-performance liquid chromatograph / mass spectrometer
- Non-patent Document 1 since ionization is performed by matrix-assisted laser desorption ionization (MALDI) and analysis is performed by a mass spectrometer (MALDI-MS), the measurement time is short and the measurement throughput is small, but it is on the MALDI plate. Since pretreatment such as drying the dropped liquid takes time, the overall throughput cannot be shortened. Therefore, the measurement time is long, and it is not suitable for clinical laboratory applications where routine inspection is performed, for example, 200 inspections / hour inspection.
- MALDI-MS mass-spectrometer
- An object of the present invention is to provide an analyzer and an analysis method capable of easily quantifying even a small measurement target substance having a blood concentration of about pg / mL to ng / mL.
- the sample concentrated with the antibody-magnetic beads on the first disk cannot be directly introduced into the mass spectrometer because ionization conditions such as mixing of impurities such as salt and pH are not suitable for ionization. Therefore, in the second disk, the physiologically active substance is purified by the solid phase extraction cartridge.
- the first purification is performed on the first disk, and the sample purified on the first disk is purified again on the second disk.
- the first disk and the second disk are connected off-line, and a sample that has been purified on the second disk can also be introduced off-line into an apparatus such as a mass spectrometer. In other words, random access is possible and it is suitable for clinical laboratory use.
