【배경기술】
전반사 형광 현미경 (total internal, reflection fluorescence microscope:
TIRF 현미경)은, 입사광의 전반사로부터 발생하는 소실파를 이용하여 형광 물질로 염색돤 시료의 수직 방향으로의 국소 영역에 대한 형광 신호를 검출하고 이를 영상화한 정보를 얻을 수 있다. 전반사 형광 현미경은, 세포 생물학, 분자 생물학, 의학 분야의 연구에서 많이 사용되며 특히 세포의 표면에서 일어나는 다양한 단백질 반응이나 약물에 의한세포표면 변화 연구에 직접적으로 이용되고 있다. . ' 기존의 전반사 형광 현미경은, 시료와 기판 사이의 계면에서 입사광를 : 전반사시켜 발생하는, 깊이 방향으로 국소화된 소실파로 시료에 염색된 형광 분자를 여기 (excitation) 시키고 여기된 형광 ^자에서 방출된 형광 신호를 검출하고 염상화하는 기본적인 구성을 가지고 있다. 그러나 아베 (Abbe)의 회절 방정식에 의해 계산될 수ᅳ있는 수평 방향으로의 분해능 한계보다 작은 분자 또는 분자의 이동. 경로 등을 기존의 전반사 형광 현미경으로는 검출하기가 블가능하거나 어렵다. 따라서 깊이. 방향으로의 높은 분해능뿐만 아니라, 수평 방향으로의 높은 분해능을 갖는 전반사 형광 현미경이 필요하다.
【발명의 상세한설명】 【기 ^작 과제]
- 본 발명의 실시.예는 깊이 방향으로의 높은 분해능뿐만 아나라, 수평 방향으로뚝 아베 (Abbe)와 회절 방정식에 의해 계산된 분해능 한계 이상의 높은 분해능을 제공할 수 밌는 고해상도의 전반사 형광 · 현미경을 제공한다.
본 발명의 다른 실시 예는 깊이 방향으로의 높은 분해능뿐만 아니라, 수평 방향으로도 아베 (Abbe)의 회 절 방정식에 의해 계산된 분해능 . 한계 이상의 높은 분해능을 제농할 수 있는 고해상도의 전반사 형광 현미경에 사용되는 전반사 형광 현미경용 검출 모들을 제공한다.

본 발명와실시예에 따른 전반사 형광 영상'장치는 수평 방향으로의 높은 분해능을 얻기 위해 표면 플라즈몬 공명 현상이 일어날수 있는 광학 배치 구조를 가지며, 이 광학 배치 구조는 전반사가 일어나는 조건의 입사광에 의해 발생하는 소실파를 수직 방향 및 수평 방향의 국소 영역에서 발생시킨다. 이 광학 배치 구조는 유리 기판과 같은 투명 기판과, 그 위에 형성된 금속 박막,ᅳ그리고 금속 박막.위에 불규칙하게 배열되거나 또는 규칙적으로 일정한 주기를 갖고 배열된 금속 나노구조 (예컨대 나노선, 나노섬,..나노홀, 나노기등 등)를 포함함다.

광원부 (60)로서 헬륨—네온 레이저 광원을 사용한 경우를 예로 ί「면, 632 nm 파장의 헬륨 -네온 레이저광이 광원부 (60)로부터 방출되고 빔 확장기 (beam expander )(62)를 거 쳐 편광기 64)에 의해 TM 모드로 편광된다. 전반사가 일어나기 위한 입사광의' 입사각을 얻기 위해 반사 거울 (66) 등의 광학 장치가 사용될 수 있다. 전반사자 일어나기 위한 입사각을 갖는 입사광은 광학 배치 구조 (50, 20)의 프리츰 (20)으로 입사되 i 유리. 기판 등의 투명 기판 (40)을 , 거쳐 나노 구조 (불규칙 적으로 배열되거나 또는 일정한 주기로 규칙적으로 배열된' 나노선, 나노섬, 나노홀 또는 나노 기등 등 ) (33)가 상부에 집적된 금속 박막 (31)에서 전반사된다. 전반사에 의해 금속 나노구조층 (30)과 시료 사이의 —계면에서 소실파가 발생한다. 특히 , 금속 나노 구조층 (30)에 의해 표면 플라즈몬 공명 현상이 '일어나 수평 방향으로 국소화된 소실파가 형광 입자를 여기시키고, 여기된 형광 입자에서 형광이 방출된다 . ^출된 형광은 대물 렌즈 (70)와 대비 감도의 향상을 위한 밴드패스 필터 (도시하지 않음)를 통해 형광 영상 추출부 (80)에 의해 2 차원의 영상 형 태로 검출될 수 있다. 도 1 의 전반사 형광 현미경 ( 100)에서 프리즘 (20)과 투명 기판 (40)의 재질에 따라 굴절율 (refract ive index)에 차이가 있으므로, 프리즘 (20)과 투명 기판 (40)의 재질에 따라, 소실파의 수직, 수평 방향으로의 국소화에 최적화된— 금속 나노구조 (33)와 금속 박막 (31)의 조건들에 차이가 있다 . 또한 프리즘 (20)과 투명 기판 (40) 사이에 굴절율 정합 액쎄나 젤 (index matching liquid or gel)을 더 배치하여 두 재질 사이에 발생할 수 있는 공기 갭 (air gap)에 의한 굴절율 미스매칭을 방지할수 있다. 도 1의 전반사 형광 현미경 (100)에 있어서, 프리즘 (20)을 생략하고 대신에 대물 렌즈 (70)를 프리즘 (20) 위치에 배치시킬 수도 있다 예를 들어, 0.8 이상의 개구수를 갖는 대물 렌즈 (70)를 투명 기판 (40) 아래에 배치하여 (프리즘은 생략) 입사광을 전반사시킬 수도 있다. 이 경우, 형광 영상 검출부 (80)는 대물 렌즈 (70) 아래에 배치되고, 수직 및 수평 방향으로 국소화된 소실파에 의해 방출되는 형광을 투명 기판 (40) 아래에서 검출하게 ¾다. 도 2 는 본 발명의 실시예에 따른 전반사 형광 현미경에 적용될 수 있는 금속 나노구조를 포함하는 광학 배치 구조로서 , 표면 플라즈몬 공명을 발생시키기 위한 광학 배치 구조를 나타낸 도면이다. 도.2'는 전반사를 일으키는 조건의. 입사광에 대해 표면 플라 ¾몬 공명 현상이 발생되어 .소실파를 수직 방향 및 수평 방향으로 국소화시킬 수 있는 금속 나노 구조 중 하나인, 일정한 주기 (A)로 배열된 나노홀 (133a)을 개략적으로 보여준다. 일정한 주기 (A)로 배열된 각 나노홀 (133a)은 일정한 두께 (dh) 및 반지름 (rh)올 가지며, 나노홀 구조 하부의 금속 박막 (131)도일정한두께 (df)를 갖는 . 금속 나노구조층 (130), 즉 나노홀을 갖는 금속 구조 (133)와 그 하부의 금속 박막 (131)은, 동일한 금속 재료로 형성될 수 있으며, 은 (silver), 금 (gold)., 백금 (platinum) 또는 알루미늄 (aluminium) 등의 금속 재료로 형성될 수 있다. 금속 박막 (131) 하부에는 투명. 기판 (40)과 프리즘 (20)이 배치되며, 투명 기판 (40)과 프리즘 (20)의.재질은 '동일할 수' 있다. 투명 기판 (40)과 금속 박막 (131). 사이에는 예를 들어 ( 막과 같은 접착성을 향상시키기 위한 금속막이 추가로 구비될 수 있다.. 전반사 형광 현미경용 검출 모들 (150)은, 투명 기환 (40)과 그 위에 형성된 :금속 나노구조층 (130)을 포함하며 개별적 인 부품 형.태로 쎄공될 수 있다. 도 3 은 본 발^의 실시 예에 따른 전반사 형광 현미경에 적용될 수 있는 불규칙하게 배열된 나노섬을 이용한 이미징을 개략적으로 보여주는 도면이다. 대불렌즈 (70)에 의해 아베 (Abbe)의 회 절 한계 이상의 크기로 초점 이 잡인 부분 (점선 참조)에서 나노섬 (233)에 의해 국소 부분으로 여기 (excite)된 형광물질이 이미지 ' 되어 기존의 전반사 현미경 방법에서 발생뒤는 전체 부분의 형광물질에 비해 개선된 분해능올 갖게 된다/ 검출 모들 (250)은 금속 박막 (231)ᅳ 나노섬 (233) 및 투명 기판 (40)을 포함한다. 도 3 쎄서 / 도면부호 155 는 바이러스 등와 시료를 나타낸다. 도 4 는 본 발명의 실시.예에 따른 전반사 형광 현미경에 적용될 수 있는 규칙 작으로 배열된 금속 나노홀을 나타내는 SEM(Scanning Electron Microscopy). 사진이다 / 규칙 적으로 배열된 나노홀은 예를 들어 , 은 (Ag) 박막 위에서 전자범 리소그래피를 이용하여 제조될 수 있하. 도 5 는 본 발,명의 실시 예에 . 따른 전반사 형광 현미경에 적용될 수 있는 불규칙하게 배열된 금속 나노섬을 나타내는 SEM 사진이다. 불규칙하게 배열된 금속 나노섬은 예를 들어 , 은 (Ag) 박막 위에 은 (Ag)으로 된 금속층을 얇게 형성한 후 150 내지 200°C에서 가열해줌으로써 -쉽 게 형성할 수 있다. 도 6 은 본 발명의 실시예에 따른 전반사 형광 현미경에 적용될 수 있^ 불규칙하게 배열된 금속 나노홀을 나타낸 SEM 사진이다. 금속 박막 상에 형성된 금속층 (예컨대, kg 층)을 적절하 가열함으로써 도 6 에 도시된 불규칙한 나노홀 구조를 형성할수 있다.
도 7 은 본 발명의 실시예에 적용될 수 있는 도 4 에 도시된 규칙적인 다수의 나노홀. 구조 상에서 표면 플라즈몬 공명 현상이 발생할 때 형성되는 소실파의 자기장세기를 보여주는 도면、이다. 도 7은 FDTD 에 의해 계산된 자기장' 세기 (magnetic field intensity) 결과를 나타낸 것으로 빨간색으로 표시 되는 부분이 자기장이 강하게 집적되는 부분이다. 도시된 바와 같이, 자기장이 나노홀 근방에서 강하게 국소화되어 있고, 국소화된 자기장의 수평 방향으로의 크기가 회절 한계 크기보다 작으므로, 국소,적으로 집중된 자기장에 의해 발생되는 이미지도 회절 한계보다작은 크기를 갖는 초고해상도를 구현할수 있다.
도 8 은 본 발명의 실시예에 적용될 수 있는 불규칙적인 다수의 나노섬 구조 상에서 표면 플라즈몬 공명 현상이 발생할 때 형성되는 소실파의 자기장 세기를 나타낸 '도면으로서 , 'FDTD 를 통해 계산된 결과를 보여준다. 도시된 바와 같이, 자기장이 .깅하게 국소화되어 있다. 도 7 과 달리 빨간색으로 표시 되는 부분 (자기장이 강하게 집적된 부분)이 불규칙적으로 형성되나, 자기장이 강하게 집적되는 부분은 도 7 과 마찬가지로수평 방향으로의 크기가 회절 한계 크기보다 작아서 집적되는 자기장에 의해 발생되는 .이미지 역시 그 한계보다 작은 크기를 갖는초고해상도를 구현할수 있 . 도 9 는 본 발명의 실시 예에 따른 전반사 형광 현미경을 사용하여 . 얻은 마이크로 튜불의 이미지를 나타낸다. 도 9 의 이미지는', 전반사를 일으키는 조건의 입사광에 대해 표면 플라즈몬 공명 ,현상이 발생되어 소실파를 수직 방향 및 수평 방향으로 국소화시킬 수 있는 금속 나노홀 구조를 적용하여 얻은 것 이다. 이는, 모터 단백질을 사용하예 움직 이는 마이크로 튜불의 이미지를 , 도 7 에 도시된 바와 같은 국소화된 자기장 분포를 이용하여 관찰한 것이다 . 도시된 바와 같이, 나노 홀 구조에서만 강하게 집적된 자기장은 100 내지 300 나노미터 이하의 크기를 가지고 있어 이를 통히ᅵ 보여지는 형광 이미지는 매우 높은 분해능을 가지 게 된다. 도 10 은 종래의 전반사 형광 현미경을 사용하여 얻은 , A549 폐암세포에 적용된:바이 러스의 이미지를 나타낸다. 도 10 의 이미지는, 금속 나노 구조가 없는 기존의 .광학 배치 구조 (호리즘 /유리기판 /금속층)를 사용하여 얻은 -것이다 . 이에 반하여 , 도 11 은 도 10 과 동일한 '조건에서 본 발명의 실시 예에. 따른 금속 나노섬 구조를 구비하는 전반사 형광 현미경을 사용하여 ¾은, A549 폐암세포에 적용된 바이러스의 이미지를 나타낸다. 도 11 의 이미지 획득에 사용된 금속 나노섬 구조는, 전반사를 일으키는 조건의 —입사광에 대해 표면 플라즈몬 공명 현상에 의해 소실파를 수직 방향 및 수평 방향으로 국소화시킬 수 있다. 도 10 과 도 11 에서 확인할 수 있는 바와 같이, 상술한 금속 나노구조를 적용한 광학 배치 구조를 갖는 전반사 현미경.을. 이용할 경우, 수평 방향으로의 분해능이 크게 향상된다는 것을 알 수 있다 . 도 12 는 도 10 및 도 11 의 이미지에서 특정. 수평방향 영 역에서의 형광 신호의 세기를 정량적 인 수치로 환산하여 나타낸 그래프이다'. 도 12 에 도시된 바와 같이, 나노섬 구조를 적용한 경우 수평 방향으로 국소화되어 매우 높은 형광 세기를 나타내는 피크들이 나타난다 . 따라서, 이 러 한 나노섬 구조를 적용한 전반사 형광 현미경은 향상된 분해능을 나타내고, 그 분해능의 정도가 100 내지 200 나노미터 정도로 회절 한계 이하의 크기가 될 수 있다 . 도 13 은 본 발명의 실시 예에 따른 전반사 형광 현미경용 검출 모들의 단면도이고, 도 14 는 도 13 의 검출 모들의 평면도이다. 도 13 및 14 의 실시 예에서는 금속 박막과 금속 나 ^구조 사이에 금속 격자가 배치되고, 그 금속 격자 상에 나노구조가 형성되어 있다. 도 13 ' 및 14 를 참조하면 , 전반사 형광 현미경용 검출 모들 (350)은 투명 기판 (40)과, 그 위에 형성된 금속 나노구조층 (331, 332, 333)을 포함한다. 금속 나노구조층은 금속 박막 (331), 금속 박막 (331) ' 상에 형성된 금속 격자 (332), 그리고 금속 격자 (332) 상에 형성된 불규칙하게 배열된 금속 나노섬 (333)올 포함한다. 도 14 에 도시된 바와 같이 금속 격자 (332)는 금속 박막 (331) 상에 서로 평 행하게 배열된 복수의 스트라이프 형상의 격자 구조를 가질 수 있다. 금속 격자. 상기에서는 금속 나노구조층을 이용하여 국소화된 표면 플라즈몬 공명을 발생시키고 이를 이용하여 형광 영상을 검출하는 것을 설명하였다. ' '
본 발명의 바람직한 다른 실시예에 따르면, 소정 시간 간격으로 입사광의 특성을 변경하여 전반사 형광 영상 장치의 분해능을 ;한층 더 개선할수 있다.

일반적으로, 형광 영상. 추출부 (80)는 아베. (Abbe)의 회절 방정식에 의해 계산될 수 있는. 수평 방향으로의 분해능이 미리 설정되어 있으며., 도 18 에 도시된 바와 같은 분해능 조건 하에서 금속 박막 (1800) .상에. 여기된 형광체가 다수 존재하는 경우, 형광 영상 추출부 (80)는 서로 인접한 형광체 (1802-1 ,1802-· 2)를 구분할 수 없는 문제점이 있다 (도 21a 참조〉.
도 19 는 본 발명의 다른 실사예께 따른 입사각이 서로 다른 입사광이 제공되는 경우의 국소표면 플라즈몬 공명을 도시한도면이다.
도 19 는 복수의 나노구조가 나^홈으로 이루어진 것을 도시한 것으로서, 도 19 와 같아 금속 나노구조층 (30)이 복수의 나노구조로 이루어지는 경우 도 1.8 과 달리 국소 영역에 표면 플라스몬 공명을 발생시킬 수 있으며, 국소 영역에 위치한 형광체만을선택적으로 여기시킬 수 있다.
본 발명의 바람직한 다른 실시예에 따르면, 도.19 의 (a) 및 (b 에 도시된 바와 입사 입사 방향을 달리함으로써 나노구조의 서로 다른 국소 영역 (1900, 1902)에 위치한형광체 (1904, 1906)를 선택적으로 여기시킬 수 있다 (도 21b 참조) .

이러한 '경우,' 금속 나노구조층 (30) 상에 위치한 형광체 중 일부만이 여기되기 때문에 결과적으로 형광 영상 추출부 (80)와 분해능이 향상된 .것과 동일한 결과를 얻을.수 있다.
본 실사예에 따르현., - 입사광 변경부 (1700)는 광원부 (60)에서 출사된 입사광의 특성을 변경하는 것으로서, 도 19 와 같이 금속 나노구조층 (30)에 입사 방향 (LI, L2)이 상이한 입사광을 제공한다. 상기에서는 입사광 변경부 (1700)가 입사 방향을 변경하는 것으로 도시하였으나, 이에 한정됨이 없이 입사광 변경부 (1700)는 광원부 (80)의 입사각 또는 파장을 변경할 수도 있으며, 이를 통해 .금속 나노구조층 (30)의'다양한 국소 영역에서 표면 플라스몬 공명이 발생되도록 한다.

레이저 출사부 (2000)를 통해 소정 파장의 레이저광 (입사광)이 -출사되고, 입사광은 범확장기 (2002)을 거쳐 편광기 (2004)에 의해 TMCTransverse Magnetic) 모 H로 편광된다.
본 실시예에 따르면, 편광기 (2004)에서 출력된 입사광은 입사광 변경부 (1700)를 거쳐 프리즘 (20) 상에 위치하는 금속 나노구조층 (30)으로 입사된다.
금속 나노구조층 (30)에 입사 방향이 다른 입사광을 제공하기 위해, .입사광 변경부 (1700)는 제 1 미러 (2006), 회전 미러 (2008), 제 2 미러 (2010) 및 제 3 미러 (2012)를 포함할수 있다.
여기서, 제 1 미러 (2006)는 편광기 (2004)에서 출력된 입사광을 회전 미러 (2008) 측으로반사한다.
회전 미러 (2008)는 회전을통해 제 1.미러 (2006).에서 반사된 입사광을 제 1 방향또는 제 2 방향으로 반사한다 . 회전 미러 (2008)에는 구동부와 타이머 (미도시 )가 연결되어 있을 수 있으며 , 구동부는 타이머와 연동하여 회전 미러 (2008)를 미리 설정된 시간 간격으로 회전시킨다.

상기 입사광 변경부는,
상기 광원부에서 출사된 입사광을 반사하는 제 1 미 러 ;
회 전을 통해 상기 제 1 미 러에서 반사되는 입사광을 제 1 방향 또는 제 2 방향으로 반사하는 회전 미 러 ; 상기 제 1 방향에 위치하며 상기 회전 미러에서 반사된 입사광을 상기 금속 나노구조층으로 반사하는 제 2 미러; 및
상기 제 2 방향에 위치하며 상기 회전 미러에서 반사된 입사광을 상기 금속 나노구조층으로 반사하는 제 3 미러를 포함하는 전반사 형광 영상 장치 . .

제 1항에 있어서,
상기 금속 박막 아래에 배치되는 투명 기판; 및
상기 투명 기판 아래에 배치된 프리즘을 더 포함하는 것을 톡징으로 하는 전반사 형광 영상 장치.

상기 나노구조는, 상기 금속 박막 상에 불규칙하게 배열된 다수의 나노선, 나노섬, 나노기등 또는 나노홀 중 적어도 하나를 포함하는 것을 특징으로 하는 전반사 형광 영상 장치 .

제 1항에 있어서,
상기 나노구조는, 상기 금속 박막 상에 일정한 주기를 갖고 규칙적으로 배열된 다수의 나노선, 나노섬, 나노기둥 또는 나노홀 중 적어도 하나를 포함하는 것을 특징으로 하는 전반사 형광 영상 장치.

제 1항에 있어서,
상기 금속 나노구조층은 은 (Ag), 금 (Au), 백금 (Pt) 및 알루미늄 (A1)로 이루어진 그룹 중에서 적어도 하나 선택된 금속 물질로 형성된 것을 특징으로 하는 전반사 형광 영상 장치. 【청구항 91

상기 나노구조와 상기 금속 박막은 동일한 금속 물질로 형성된 것을 특징으로 하는 전반사 형광 영상 장치 .

상기 복수의 나노구조와 상기 금속 격자는 동일한 금속 물질로 형성된 것을 특징으로 하는 전반사 형광 영상 장치 .
【청구항 14] 금속 박막과 상기 금속 박막 상에 형성된 나노구조를 구비하는 금속 나노구조층을 포함하며,
상기 금속 나노구조층에서 입사광이 전반사되어 상기 금속 나노구조층과 그 위에 배치된 시료와의 사이에서 수평 방향으로 국소화된 소실파를 일으키는 것을 특징으로 하는 검출 모듈.

상기 나노구조는, 상기 금속 박막 상에 불규칙하게 배열된 다수의 나노선, 나노섬, 나노기등 또는 나노홀 중 적어도 하나를 포함하는 것을 특징으로 하는 검출 모들.
