Abstract
A presente invenção é um processo de obtenção de uma formulação de resveratrol e óleo de farelo de arroz. O produto resultante na forma de óleo ou de sólido visa aumentar o potencial terapêutico do resveratrol pela ação sinergética dos componentes do óleo de arroz. O produto obtido através do processo da invenção é um principio ativo que, ao ser incorporado em composições nutracêuticas, proporciona ação antioxidante, antiinflamatória, antiviral, cardioprotetora, neuroprotetora e/ou quimiopreventiva de câncer, além de proteger contra infecções e isquemia, reduzir a obesidade e prevenir as doenças do envelhecimento.

Campo da Invenção
A presente invenção é um processo de obtenção de uma formulação de resveratrol e óleo vegetal, em especial o óleo de farelo de arroz. O produto resultante na forma de óleo ou de solido visa aumentar o potencial terapêutico do resveratrol pela ação sinergética dos componentes do óleo de arroz.
O produto obtido através do processo da invenção é um princípio ativo que, ao ser incorporado em composições nutracêuticas, proporciona ação antioxidante, antiinflamatória, antiviral, cardioprotetora, neuroprotetora e/ou quimiopreventiva de câncer, além de proteger contra infecções e isquemia, reduzir a obesidade e prevenir as doenças do envelhecimento.
Antecedentes da Invenção
O írans-resveratrol (3,5,4'-trihidroxiestilbeno) tem atividade antioxidante, antiinflamatória, antiviral, cardioprotetora, neuroprotetora, quimiopreventiva de câncer, além de proteger contra infecções e isquemia, reduzir a obesidade e prevenir o envelhecimento. O frans-resveratrol é um polifenol que pode ser encontrado principalmente na casca das uvas pretas e no vinho tinto. Quanto mais intensa for a cor do vinho ou das uvas, maior é o seu conteúdo em polifenóis. Estudos parecem indicar que o frans-resveratrol pode ajudar a diminuir os níveis de lipoproteínas de baixa densidade, também conhecidas como colesterol LDL ou colesterol "mau", e aumentar os níveis de lipoproteínas de alta densidade, colesterol HDL, ou colesterol "bom". O LDL principalmente no seu estado oxidado pode acumular-se nas paredes dos vasos sanguíneos levando à formação de placas de ateroma. Essas placas estão na origem da aterosclerose que resulta na obstrução dos vasos sanguíneos. O trans-
resveratrol favorece a produção de HDL pelo fígado, a redução da produção de LDL e impede a oxidação do LDL circulante, tendo assim um papel na redução do risco de desenvolvimento de doenças cardiovasculares, tais como o infarto do miocárdio.
Também o frans-resveratrol é amplamente conhecido como modulador da expressão e da atividade de uma classe de proteína chamada de sirtuina. Entende-se por modulador de sirtuinas os compostos que regulam para cima (ativando ou estimulando), para baixo (inibindo ou suprimindo) ou de outra forma mudam a propriedade funcional ou a atividade biológica da proteína sirtuina.
A sirtuina é um membro da família das proteínas desacetilases sirtuinas, ou preferencialmente da família Sir2, na qual se inclui a Sir2 de levedura (GenBank Accession No P53685), C. elegans Sir-2.1 (GenBank Accession No NP_501912), e humana SIRT1 (GenBank Accession No.NM_012238 e NP_036370 ou AF083106) e SIRT2 (GenBank Accession No NM_012237, NM_030593, NP_036369, NP_085096, e AF083107).
Os moduladores da proteína sirtuina, como derivados de frans- resveratrol, atuam para minimizar ou eliminar doenças ligadas ao envelhecimento - doenças crônico-degenerativas do sistema circulatório e do neurológico - como enfermidades oculares (WO 2006/127987), psicopatologias (WO 2006/138418), diabetes (WO 2006/104586), câncer (WO 2006/102557), obesidade (US 2006/11 1435).
Entretanto, o frans-resveratrol tem baixa biodisponibilidade, quando é administrada oralmente essa molécula é rapidamente metabolizada para formar vários derivados que são prontamente eliminados no corpo, isso dificulta sua absorção e é um problema tecnológico a ser resolvido. Uma estratégia para aumentar absorção do resveratrol é misturar com compostos químicos que competem ou inibem com essas rotas metabólicas derivatização. Um produto comercial que apresenta esse efeito sinérgico é o Longevinex (www.longevinex.com) que é uma formulação de trans-resveratrol, extrato de uva e quercetina.
A presente invenção é o desenvolvimento de um processo e produto de uma e mistura de trans-resveratrol com óleo de farelo de arroz. Esse óleo contém altos níveis de tocoferóis, tocotrienóis e fitoesterois e principalmente rico em gama-orizanol.
O gama-orizanol tem ação antioxidante mais efetiva que os saudáveis tocoferóis, por ser mais resistente ao calor. Dentre suas múltiplas ações, destacam-se: efeitos no crescimento; combate a doenças cefálicas e cervicais; minimização dos sintomas da menopausa; combate à anemia; tratamento de úlceras de estresse e coadjuvante no tratamento de doenças circulatórias. As propriedades do orizanol permitem o amplo uso, seja como medicamento, em composição de cosméticos, como agente antienvelhecimento da pele e até como fármaco para redução do colesterol do tipo LDL.
Na recente literatura temos vários exemplos da potencialização do trans- resveratrol em conjuntos com outras moléculas, como por exemplo, junto com gama-tocotrienol suprime proliferação de células de câncer de mana (Hsieh, TC, et al. Int J OncoL 2008 Oct;33(4):851-9), com roscovitina inibe progressão de células leucêmicas tipo HL-60 (Komina, O, et al. Biochem Pharmacol. 2008 Dec 1 ;76(11): 1554-62), com os tocoferóis regeneram a atividade do resveratrol (Fang, JG, eT al J. Agric. Food Chem., 2008, 56 (23), 11458-11463) e com 4- hidroxinonenal aumenta a capacidade de desintoxicante ( Zhang, H. et al Arch Biochem Biophys, 2009 Jan 1 ;481 (1):110-5).
A literatura patentária contempla alguns documentos relacionados a processos de írans-resveratrol e óleo de farelo arroz. Embora nenhum dos documentos encontrados antecipe ou sequer sugira, ainda que indiretamente, o conceito inventivo da presente invenção, são aqui citados alguns documentos utilizados como referência.
Os pedidos de patentes, US 2003/6638545, US 2003/6642277, US 2003/0133945, US 2008/0070991 , WO 2000/064282, WO 2004/000043 e WO 2009/003798 o frans-resveratrol como parte da formulações de um alimento ou suplemento alimentar no estado sólido, semi-solido e líquido que são obtidas através de uma mistura física com outros substâncias.
Outros pedidos de patentes referem-se a obtenção de micro-emulsão ou emulsão de óleo com trans-resveratrol. O documento US 2003/6660286 descreve uma emulsão com biosurfactantes e alfa-tocoferol. O US 2005/0196347 relata uma emulsão para spray a base de óleo/água. O documento US 2006/011051 descreve uma composição com ácidos graxos poliinsaturados e US 2008/0085357 consiste a formação de micro-emulsão através da eliminação a vácuo do solvente para alcançar a concentração micelar critica.
Sumário da Invenção
Em um primeiro aspecto a presente invenção proporciona uma composição capaz de evitar a eliminação rápida do trans-resveratrol, aumentar a sua absorção e consequentemente aumentar o potencial terapêutico do trans-resveratrol.
É, portanto um objeto da presente invenção uma composição compreendendo:
a) óleo vegetal;
b) resveratrol; e
c) um veículo adequado.
Em uma realização preferencial, o óleo vegetal é o óleo de farelo de arroz e a composição está na forma de pó.
É um adicional objeto da presente invenção um processo de produção de uma composição compreendendo óleo vegetal e resveratrol compreendendo as etapas de:
a) dissolução do resveratrol em álcool;
b) adição do óleo vegetal à solução de resveratrol da etapa a) sob agitação e banho de ultrassom.
Em uma realização preferencial, o processo de produção de uma composição em pó compreende adicionalmente as etapas de:
c) adição da solução obtida em b) a uma suspensão de ciclodextrina em água, sob agitação;
d) adição de um agente adsorvente para remoção de água.
É um adicional objeto da presente invenção uma composição nutracêutica compreendendo a composição que compreende óleo vegetal e resveratrol.
Estes e outros objetos da presente invenção serão valorizados a partir da descrição detalhada a seguir.

Descrição Detalhada da Invenção
Os exemplos a seguir ilustram, mas não limitam, as formas preferenciais de concretizar a invenção.
Composição
A composição da presente invenção é uma composição que compreende:
a) óleo vegetal;
b) resveratrol; e
c) um veículo adequado.
Em especial, a composição apresenta atividade antioxidante, antiinflamatória, antiviral, cardioprotetora, neuroprotetora e/ou quimiopreventiva de câncer, além de proteger contra infecções e isquemia, reduzir a obesidade e prevenir as doenças do envelhecimento.
Óleo Vegetal
Os óleos vegetais da presente invenção são óleos vegetais dotados de substâncias anti-oxidantes e/ou capazes de proporcionar um sinergismo em conjunto com o resveratrol.
Exemplos não limitantes de tais óleos incluem óleo de farelo de arroz, óleo de milho, óleo de soja, óleo de algodão. Preferencialmente utiliza-se óleo de farelo de arroz. O óleo de farelo de arroz foi obtido da empresa A HELMUT
TESSMANN ÓLEOS VEGETAIS (PI 0604880-3A) com a seguinte composição: gama-orizanol 1%; ácido oleico 40%; ácido linoléico 32%; ácido linoléico 1 ,5 %; vitamina E (tocoferol) 1.200 mg/Kg.
Resveratrol
Para efeitos da presente invenção a expressão resveratrol engloba o resveratrol, em especial o trans-resveratrol bem como a derivados do resveratrol, como seus derivados metilados e/ou acetilados.
Os versados na arte identificarão que os derivados metilados incluem, sem limitações trans-3,5-dimetoxi-4'-hidroxi-estilbeno, trans-3,5,4'-trimetoxi- estilbeno e trans-3,5-hidroxi-4'-metoxi-estilbeno.

Outros derivados como os derivados acetilados, podem ser utilizados nas composições da presente invenção. Exemplos destes derivados incluem, sem limitações trans-3,5-diacetili-4'-hidroxi-estilbeno, trans-3,5,4'-triacetil- estilbeno e trans-3,5-hidroxi-4'-acetil-estiIbeno.

O írans-resveratrol utilizado foi o obtido pelo processo de purificação descrito no documento PI0700152-5 ou o adquirido da empresa Sigma-Aldrich. A beta-ciclodextrina (CAVAMAX W& Pharma) foi adquirida da empresa ISP Technologies, Inc.
Os versados na arte também identificarão que o processo de aumento da bioatividade da invenção pode ser aplicado ã derivados do resveratrol, como seus derivados metilados e/ou acetilados.
Processo de Produção de Composição
A composição da presente invenção é uma composição que pode ser obtida pelo processo que compreende as etapas de:
a) dissolução do resveratrol em álcool;
b) adição do óleo vegetal à solução de resveratrol da etapa a) sob agitação e banho de ultrassom.
O resveratrol dissolvido em álcool deve apresentar uma concentração final dentro da faixa de 0,2-1 ,5 g/mL de álcool, sendo preferencial a concentração de 0,75 g/mL, e o banho de ultrassom dura o tempo suficiente para proporcionar a dissolução, geralmente em torno de 30 minutos.
A proporção solução de resveratrol:óleo vegetal está compreendida na faixa que vai de 1 :60 a 1 :70, sendo preferencial a proporção de 1 :62,5.
Em uma realização preferencial, o processo de produção de uma composição em pó compreende adicionalmente as etapas de:
c) adição da solução obtida em b) a uma suspensão de ciclodextrina em água, sob agitação;
d) adição de um agente adsorvente para remoção de água.
A concentração da suspensão de ciclodextrina está compreendida na faixa que vai de 2,0 a 2,5 mg/mL.
O agente adsorvente, para ser eficaz, deve estar presente em uma concentração de 0,5% a 1 ,5% p/p da mistura. Em especial o agente adsorvente é celite.
Composição Nutracêutica
Entende-se por composição nutracêutica toda composição que tenha o intuito de proporcionar manutenção ou melhora das condições fisiológicas de um indivíduo, através de sua administração por uma via adequada.
A composição da presente inveção pode ser incorporada em concentrações de 0,1% até 99% p/p em composições farmacêuticas, cosméticas e suplementos alimentares.
Exemplo 1 : Obtenção do óleo de farelo de arroz com 1,2% m/v de írans-resveratrol (óleo/resv.)
Em um becker de 50 mL foram adicionados 3 g de frans-resveratrol e até 4 mL de etanol 96 %. A mistura foi colocada no banho de ultrassom por 30 min. Logo a seguir, essa mistura foi adiciona aos poucos a 250 mL de óleo de farelo de arroz. A mistura resultante foi agitada a 500 rpm por 30 min e a seguir por uma 1h no banho de ultrassom. O produto final apresenta é viscoso, de coloração marrom e com um espectro de infravermelho característico (figura 1). A quantidade de frans-resveratrol foi por cromatografia líquida de alta eficiência (CLAE). Para tal quantificação foi construída uma curva de calibração externa, com concentrações de frans-resveratrol variando entre 0,10 a 200,0 mg.L"1. A CLAE foi conduzida com eluição isocrática (fluxo de 1 ,0 mL min"1), empregando-se uma solução aquosa de acetonitrila a 25%, com pH 3,0,
ajustado com H3PO4. O cromatógrafo estava equipado com detector UV-VIS e coluna Cie de 250 mm de comprimento, 5 μιτι de diâmetro de partícula e 4,6 mm de diâmetro interno. A preparação da amostra segue as seguintes etapas: 10 g do produto final foi misturado com 400 mL metanol/acetona (7:3, v/v) e agitado (700 rpm) por 1 h a temperatura ambiente. A solução foi esfriada a - 20 for 15 h. A fase liquida e a fase solida foi separada por filtração a vácuo. O solvente foi removido pressão reduzida. O produto final foi diluído 1000 vezes em metanol e 20 μΐ_ foram injetados e detectados em 306 nm. A concentração de resveratrol foi de 10, 2 ± 0,1 mg/1mL
Exemplo 2: Preparação do óleo/resv em pó
Em um becker de 250 mL contendo 100 mL do óleo/resv foram adicionados lentamente e com agitação vigorosa (1000 rpm) 20 a 25 g de beta- ciclodextrina suspensa em até 10 mL de água durante 30 min. A mistura permaneceu sobre agitação (800 rpm) durante 1 hora. Finalmente foi adicionado até 1 % de celite. O produto final com aparência de pó. A quantidade de frans-resveratrol foi por cromatografia líquida de alta eficiência (CLAE). Para tal quantificação foi construída uma curva de calibração externa, com concentrações de frans-resveratrol variando entre 0,10 a 200,0 mg.L"1. A CLAE foi conduzida com eluição isocrática (fluxo de 1 ,0 mL min"1), empregando-se uma solução aquosa de acetonitrila a 25%, com pH 3,0, ajustado com H3PO4. O cromatógrafo estava equipado com detector UV-VIS e coluna Ci8 de 250 mm de comprimento, 5 μητι de diâmetro de partícula e 4,6 mm de diâmetro interno. A preparação da amostra segue as seguintes etapas: 10 g do produto final foi misturado com 400 mL metanol/acetona (7:3, v/v) e agitado (700 rpm) por 1 h a temperatura ambiente. A solução foi esfriada a - 20 for 15 h. A fase liquida e a fase solida foi separada por filtração a vácuo. O solvente foi removido pressão reduzida. O produto final foi diluído 1000 vezes em metanol e 20 μί foram injetados e detectados em 306 nm. A concentração de resveratrol foi de 6,2 ± 0,1 mg/1mL.
Claims
1. Composição caracterizada por compreender:
a) óleo vegetal;
b) resveratrol; e
c) um veículo adequado.
2. Composição, de acordo com a reivindicação 1 , caracterizada pelo óleo vegetal ser escolhido do grupo que compreende óleo de farelo de arroz, óleo de milho, óleo de soja, óleo de algodão.
3. Composição, de acordo com a reivindicação 2, caracterizada pelo óleo vegetal ser óleo de farelo de arroz.
4. Composição, de acordo com a reivindicação 3, caracterizado pelo óleo de arroz possuir a seguinte composição: gama-orizanol 1 %; ácido oleico 40%; ácido linoléico 32%; ácido linoléico 1 ,5 %; vitamina E (tocoferol) 1.200 mg/Kg.
5. Composição, de acordo com a reivindicação 1 , caracterizada pelo resveratrol ser escolhido do grupo que compreende trans-resveratrol, trans-3,5- dimetoxi-4'-hidroxi-estilbeno, trans-3,5,4'-trimetoxi-estilbeno e trans-3,5-hidroxi- 4'-metoxi-estilbeno, trans-3,5-diacetili-4'-hidroxi-estilbeno, trans-3,5,4'-triacetil- estilbeno e trans-3,5-hidroxi-4'-acetil-estilbeno e combinações dos mesmos.
6. Composição, de acordo com a reivindicação 1 , caracterizada pela proporção resveratrol:óleo vegetal está compreendida na faixa que vai de 1 :60 a 1 :70.
7. Composição, de acordo com a reivindicação 6, caracterizada pela proporção ser 1 :62,5.
8. Processo de produção de composição compreendendo óleo vegetal e resveratrol caracterizado por compreender as etapas de:
a) dissolução do resveratrol em álcool;
b) adição do óleo vegetal à solução de resveratrol da etapa a) sob agitação e banho de ultrassom; e, opcionalmente
c) adição da solução obtida em b) a uma suspensão de ciclodextrina em água, sob agitação;
d) adição de um agente adsorvente para remoção de água.
9. Processo, de acordo com a reivindicação 8, caracterizado pelo resveratrol dissolvido em álcool apresentar uma concentração final dentro da faixa de 0,2-1 ,5 g/mL de álcool,
10. Processo, de acordo com a reivindicação 9, caracterizado pelo resveratrol dissolvido em álcool apresentar uma concentração final de 0,75 g/mL.
11. Processo, de acordo com a reivindicação 8, caracterizado pela ciclodextrina estar compreendida na faixa que vai de 2,0 a 2,5 mg/mL.
12. Processo, de acordo com a reivindicação 8, caracterizado pelo agente adsorvente estar presente em uma concentração de 0,5% a 1 ,5% p/p da mistura.
13. Processo, de acordo com a reivindicação 8, caracterizado pelo o agente adsorvente ser celite.
14. Composição nutracêutica caracterizada por compreender de 0,1% a 99% p/p de uma composição compreendendo um óleo vegetal e um resveratrol.
15. Composição nutracêutica de acordo com a reivindicação 14, caracterizada por ser incorporada a composições farmacêuticas, cosméticas e/ou suplementos alimentares.
