Abstract
本発明は、粘膜に存在する免疫組織に効率的に抗原を送達し、粘膜免疫システムおよび全身系免疫システムのいずれをも誘導できる粘膜ワクチンを提供する。本発明の粘膜ワクチンは、ウエルシュ菌エンテロトキシンのC末端断片と抗原との融合体を含み、鼻粘膜、消化器粘膜、呼吸器粘膜、口腔粘膜、膣粘膜、眼粘膜等の粘膜面に投与することにより、粘膜免疫および全身免疫のいずれをも賦活化させることができる。抗原は、感染性病原体由来、癌細胞由来、またはアレルゲン由来であることが好ましい。
Description
本発明は、粘膜ワクチンに関するものであり、より詳細には、抗原を効率よく粘膜面の免疫細胞に送達することを可能とした粘膜ワクチンに関するものである。
現在世界では毎年6000万人が亡くなっており、感染症に起因する死者は2000万人を数える。昨今のボーダレス化と相俟って、地球規模でエイズ、インフルエンザ、重症呼吸器不全症等感染症の伝播が加速しつつあり、エイズウイルス、インフルエンザウイルス、コロナウイルス等の感染性病原体に対する感染予防対策法の確立が焦眉の急となっている。これら感染性病原体の多くは粘膜面を侵入門戸としていることから、粘膜面に効果的な免疫防御機構を構築することが感染予防、感染制御の観点から重要である。また、この粘膜系免疫機構をすり抜け血中等の体内に侵入してしまった病原体に対して全身系の特異的体液性免疫機構を誘導し、侵入した病原体を速やかに除去し組織細胞等への感染を未然に防ぐことも必須となる。さらに、病原体が感染してしまった病態細胞を効率よく排除し治療効果を期待するには全身系の体液性・細胞性免疫機構の構築が必要となる。
感染症対策としてワクチン接種が世界的に行われているものの、現在の主流である注射によるワクチン接種では脾臓等の全身系体液性免疫の惹起に限局されてしまい、粘膜面における免疫賦活化作用を持たないために治療効果は期待できるが感染予防効果は期待できない。このように、現在のところ、感染性病原体に起因する難治性疾患に対して有効な予防法は皆無に等しく、これら病原体の初発感染部位である粘膜面の免疫系と全身系の体液性・細胞性免疫系をともに活性化するワクチン開発が待望されている。
粘膜ワクチンは、「吸う、飲む」といった方法により経粘膜的に抗原を投与するワクチンであり、投与部分の粘膜面のみならず、遠隔の粘膜面、全身系免疫組織にも抗原特異的な免疫応答を誘導できるため、初発感染防御に働く粘膜系免疫と万一感染が成立した際の排除機構として働く全身系の体液性・細胞性免疫という二段、三段構えの防御機構を構築し得ることから、唯一の理想的な感染症防御法であると言える。また、粘膜ワクチンは、注射を必要としないことから、医療従事者を必要とせず、医療廃棄物も発生しないので、保険医療や環境問題という観点や、衛生状態の悪い地域での感染防御の観点からも、次世代ワクチンとして大いに期待されている。
しかし、粘膜ワクチンの実用化に向けては多くの克服すべき問題があり、極一部の例(非特許文献1参照)を除いて粘膜ワクチンの開発は遅々として進展していない。最も困難な問題としては、腸管や呼吸器は、元来、外来異物を分解、排除するための機能が発達しているため、ワクチン抗原を投与しても効果的に抗原特異的免疫応答が得られないことが挙げられる。この問題を克服するためには、粘膜面に存在する免疫組織に効率的に抗原を送達するシステムの開発が不可欠である。
非特許文献2には、ポリ乳酸マイクロスフェアなど、臨床で既に薬物DDSに用いられている粒子担体のいくつかは、消化管における消化、分解から抗原を防御することで粘膜ワクチン担体としても有効であることが記載されている。また、非特許文献3には、コメがすぐれた経口ワクチン担体となり得ることが記載されている。さらに、非特許文献4には、センダイウイルス由来の膜タンパク質を利用した膜融合リポソームを用いて抗原を経鼻投与すると鼻咽頭関連リンパ組織(NALT)のM細胞に抗原を送達するとともに、その下層に存在すると思われる抗原提示細胞へ非常に効率よく抗原送達していることが記載されている。このように、粘膜面に存在する免疫組織に効率的に抗原を送達する種々のシステムが開発中であるが、未だ実用化には至っていない。

本発明は、粘膜に存在する免疫組織に効率的に抗原を送達し、粘膜免疫システムおよび全身系免疫システムのいずれをも誘導できる粘膜ワクチンを提供することを目的とする。
本発明は、上記課題を解決するために、以下の発明を包含する。
[1]ウエルシュ菌エンテロトキシンのC末端断片と抗原との融合体を含むことを特徴とする粘膜ワクチン。
[2]ウエルシュ菌エンテロトキシンのC末端断片が、毒性を発現しないことを特徴とする上記[1]に記載の粘膜ワクチン。
[3]ウエルシュ菌エンテロトキシンのC末端断片が、以下の(A)または(B)のタンパク質であることを特徴とする上記[1]または上記[2]に記載の粘膜ワクチン。
(A)配列番号1で表わされるアミノ酸配列の部分配列からなり、少なくとも第304位~第319位のアミノ酸を含むタンパク質

本発明によれば、粘膜に存在する免疫組織に効率的に抗原を送達し、粘膜免疫システムおよび全身系免疫システムのいずれをも誘導できる粘膜ワクチンを提供することができる。また、本発明の粘膜ワクチンは、Th1系およびTh2系の両方の免疫系を活性化でき、予防的ワクチン効果および治療的ワクチン効果の両方の効果を奏することができる。
本発明の粘膜ワクチンは、ウエルシュ菌エンテロトキシンのC末端断片と抗原との融合体を含むものである。本明細書において「粘膜ワクチン」とは経粘膜的に抗原を投与するワクチンを意味する。
ウエルシュ菌(学名:Clostridium perfringens)は、クロストリジウム属に属する嫌気性桿菌である。ヒトや動物の腸内細菌叢の構成菌であり、下水、河川、海、耕地などの土壌に広く分布する。ウエルシュ菌エンテロトキシン(以下「CPE」と記す)はウエルシュ菌が産生する毒素の1つであり、感染性食中毒の原因となる。CPEは、細胞のタイトジャンクション構成タンパク質であるクローディン(Claudin)ファミリーのクローディン3、4、6、7、8および14と結合することが知られている(Fujita, K. et al., (2000) FEBS Letters 476, 258-261.)。クローディン4は、パイエル板上に高発現していることが報告されている(Tamagawa, H. et al., (2003) Laboratory Investigation 83, 1045-1053.)。クローディン3、4、6、7、8および14がバイエル板以外の粘膜組織に高発現しているか否かは不明であるが、本発明者らは粘膜への抗原の効率的な送達に各種クローディンと結合するCPEを利用することを着想し、ウエルシュ菌エンテロトキシンのC末端断片と抗原との融合体が粘膜ワクチンとして有用であることを初めて見出した。
〔ウエルシュ菌エンテロトキシンのC末端断片〕
ウエルシュ菌エンテロトキシンは配列番号1で表わされる319アミノ酸からなるタンパク質であり、これをコードする遺伝子は配列番号2で表わされる塩基配列を有する。ウエルシュ菌エンテロトキシンをコードする遺伝子(配列番号2)は、アクセッション番号:M98037として公知のデータベース(GenBankなど)に登録されている。ウエルシュ菌エンテロトキシンは、これを産生するウエルシュ菌から回収、精製することにより取得することができる。また、公知の遺伝子組換え技術によりウエルシュ菌エンテロトキシン発現ベクターを構築し、これを適当な宿主に導入して発現させた組み換えタンパク質を回収、精製して取得することもできる。
ウエルシュ菌エンテロトキシンは配列番号1で表わされる319アミノ酸からなるタンパク質であり、これをコードする遺伝子は配列番号2で表わされる塩基配列を有する。ウエルシュ菌エンテロトキシンをコードする遺伝子(配列番号2)は、アクセッション番号:M98037として公知のデータベース(GenBankなど)に登録されている。ウエルシュ菌エンテロトキシンは、これを産生するウエルシュ菌から回収、精製することにより取得することができる。また、公知の遺伝子組換え技術によりウエルシュ菌エンテロトキシン発現ベクターを構築し、これを適当な宿主に導入して発現させた組み換えタンパク質を回収、精製して取得することもできる。
ウエルシュ菌エンテロトキシンのC末端断片(以下「C-CPE」と記す)は、CPEのC末端のアミノ酸を含みN末端のアミノ酸を含まないCPEの部分タンパク質であり、クローディンとの結合能を有しているものであればよい。C-CPEをコードする遺伝子はCPE遺伝子を鋳型として適当な合成オリゴヌクレオチドプライマーを用いたpolymerase chain reaction(PCR)法によって取得することができる。得られたC-CPE遺伝子を用いて、公知の遺伝子組換え技術によりC-CPE発現ベクターを構築し、これを適当な宿主に導入して発現させた組み換えタンパク質を回収、精製してC-CPEを取得することができる。
C-CPEは、毒性を発現しないことが好ましい。毒性を発現しないとは、CPEが有する細胞毒性が消滅していることを意味する。CPEの毒性は、CPEのN末端側のアミノ酸領域に支配されていることが知られており、CPEのN末端から第44位までのアミノ酸が欠失したC-CPE(45-319)は毒性を発現するが、N末端から第52位までのアミノ酸が欠失したC-CPE(53-319)は毒性を発現しないことが報告されている(Kokai-Kun, J.F., et al. (1997) Clin. Infect. Dis. 25, Suppl. 2, 165-167.)。したがって、少なくともCPEの第1位~第52位までのアミノ酸が欠失したC-CPEとすることで、毒性を発現しないC-CPEを取得することができる。また、CPEの第52位より前の領域を有するC-CPEであっても、第45位~第52位の領域に置換、欠失、挿入、修飾等の変異を誘導することにより、毒性を発現しないC-CPEを取得することも可能である。
CPEとクローディン4との結合には、CPEのC末端の16アミノ酸(第304位~第319位)が関与していることが報告されている(Takahashi A., et al. (2005) J Control Release 108, 56-62.)。また、これら16アミノ酸のうち1アミノ酸が置換されても結合能を維持していることが報告されている(Takahashi A., et al. (2008) Biochem Pharmcol, 75, 1639-1648)。したがって、C-CPEは、配列番号1で表わされるアミノ酸配列の部分配列からなり、少なくとも第304位~第319位のアミノ酸を含むことが好ましい。また、クローディンとの結合能を有している限りにおいて、C-CPEは、アミノ酸配列に変異を有していてもよい。例えば、本発明に用いるC-CPEは、配列番号1で表わされるアミノ酸配列の部分配列からなり、少なくとも第304位~第319位のアミノ酸を含むタンパク質において、1または数個のアミノ酸が欠失、置換もしくは付加されたものであってもよい。C-CPEの好ましい改変体として、配列番号1の第309位のアスパラギン、または第313位のセリン、または第309位のアスパラギンと第313位のセリンの両方が他のアミノ酸、例えばアラニン、バリン、ロイシン、イソロイシン、トレオニン、グルタミンなどに置換されたC-CPEが挙げられる。これら改変体を用いれば、低用量の投与でも高いワクチン効果を誘導できることが発明者らにより確認されている。
ここで「1または数個のアミノ酸が欠失、置換もしくは付加された」とは、部位特異的突然変異誘発法等の公知の変異ペプチド作製法により欠失、置換もしくは付加できる程度の数(好ましくは10個以下、より好ましくは7個以下、さらに好ましくは5個以下)のアミノ酸が欠失、置換もしくは付加されることを意味する。このような変異タンパク質は、公知の変異ポリペプチド作製法により人為的に導入された変異を有するタンパク質に限定されるものではなく、天然に存在するタンパク質を単離精製したものであってもよい。
タンパク質のアミノ酸配列中のいくつかのアミノ酸が、このタンパク質の構造または機能に有意に影響することなく容易に改変され得ることは、当該分野において周知である。さらに、人為的に改変させるだけでなく、天然のタンパク質において、当該タンパク質の構造または機能を有意に変化させない変異体が存在することもまた周知である。
好ましい変異体は、保存性もしくは非保存性アミノ酸置換、欠失、または添加を有する。好ましくは、サイレント置換、添加、および欠失であり、特に好ましくは、保存性置換である。これらは、本発明に係るポリペプチド活性を変化させない。代表的に保存性置換と見られるのは、脂肪族アミノ酸Ala、Val、Leu、およびIleの中での1つのアミノ酸の別のアミノ酸への置換、ヒドロキシル残基SerおよびThrの交換、酸性残基AspおよびGluの交換、アミド残基AsnおよびGlnの間の置換、塩基性残基LysおよびArgの交換、ならびに芳香族残基Phe、Tyrの間の置換である。
C-CPEの大きさ(アミノ酸残基数)は特に限定されないが、C-CPEの抗原性の点から小さいことが好ましい。好ましくは140アミノ酸以下、より好ましくは100アミノ酸以下、さらに好ましくは40アミノ酸以下、特に好ましくは30アミノ酸以下である。
〔抗原〕
本発明のワクチンに含まれる抗原は特に限定されないが、感染性病原体由来、癌細胞由来、またはアレルゲン由来の抗原であることが好ましい。感染性病原体には、感染性疾患の原因となるウイルス、細菌、寄生生物、または菌類などが含まれる。具体的には、インフルエンザウイルス、エイズウイルス(HIV)、ノロウイルス、コロナウイルス、ロタウイルス、アデノウイルス、ヘルペスウイルス、風疹ウイルス、狂犬病ウイルス、腸管出血性大腸菌、コレラ菌、赤痢菌、サルモネラ菌、黄色ブドウ球菌、百日咳菌、髄膜炎菌、クリプトコッカス、アスペルギルス、熱帯熱マラリア原虫などが挙げられる。癌抗原としては、WT1(乳がん、肺がん、消化器がん、脳腫瘍、血液悪性腫瘍)、CEA(結腸直腸がん、肺がん)、HER2(乳がん、卵巣がん、肺がん、胃がん、結腸直腸がん)、MAGE(胃がん、大腸がん、肺がん)、MART-1(悪性黒色腫)、gp100(悪性黒色腫)、Tyrosinase(悪性黒色腫)などが挙げられる。アレルゲンとしては、花粉、ほこり、カビ、胞子、鱗屑、昆虫、食品などが挙げられる。好ましくは、インフルエンザウイルス、エイズウイルス(HIV)、ノロウイルス、コロナウイルス、腸管出血性大腸菌、コレラ菌である。

〔融合体〕
C-CPEと抗原との融合体は、C-CPEと抗原とを含んで一体化されているものであればよく、その形態は限定されない。また、融合体にはC-CPEと抗原以外のものが含まれていてもよい。好ましい融合体としては、例えば、C-CPEと抗原との融合タンパク質、抗原を封入したリポソームとC-CPEの結合体、抗原を担持したナノマテリアルとC-CPEの結合体などが挙げられる。
C-CPEと抗原との融合体は、C-CPEと抗原とを含んで一体化されているものであればよく、その形態は限定されない。また、融合体にはC-CPEと抗原以外のものが含まれていてもよい。好ましい融合体としては、例えば、C-CPEと抗原との融合タンパク質、抗原を封入したリポソームとC-CPEの結合体、抗原を担持したナノマテリアルとC-CPEの結合体などが挙げられる。
C-CPEと抗原との融合タンパク質は、公知の遺伝子組換え技術により製造することができる。例えば、C-CPEをコードする遺伝子と抗原とするタンパク質をコードする遺伝子とを人工的に連結した融合遺伝子(ハイブリッド遺伝子)を作製し、当該融合遺伝子を、発現ベクターのプロモーターの下流に挿入し、適当な宿主細胞に導入して発現させることにより取得することができる。融合タンパク質において、C-CPEと抗原との結合順序は限定されず、N末端が抗原でC末端側がC-CPEとしてもよく、逆にN末端がC-CPEでC末端側が抗原としてもよい。なお、抗原とするタンパク質をコードする遺伝子の塩基配列情報は、公知のデータベース(GenBank等)から取得することができる。また、公知の方法を用いて目的遺伝子をクローニングし、塩基配列解析を行うことで塩基配列情報を取得することができる。このようにして得られた塩基配列情報に基づいて、目的の抗原を有する生物から核酸を抽出し、PCR等の公知の手段を用いて遺伝子を取得することができる。
抗原を封入したリポソームとC-CPEの結合体は、抗原ペプチド、抗原タンパク質、抗原をコードする遺伝子などを封入したリポソーム膜表面にC-CPEを結合させたものであればよく、抗原、リポソームの脂質組成やサイズ、およびC-CPE結合方法は特に限定されない。リポソーム作製には、ホスファチジルコリン、コレステロール、ポリエチレングリコール結合脂質などに加え、C-CPE結合用に脂質末端カルボキシル基もしくはマレイミド基を有する脂質を使用する。リポソーム内への抗原の封入は公知の凍結融解法、逆相蒸発法、水和法などにより行うことができる。作製した抗原内封リポソームにC-CPEを加え、ペプチド縮合反応もしくはSH基反応系によりリポソームとC-CPEとの結合体を作製することができる。
抗原を担持したナノマテリアルとC-CPEの結合体としては、公知の各種末端修飾ポリマーとC-CPEのリジン、システインとの結合体を用いた自己組織化ナノマテリアルを使用することができる。なお、抗原には病原体のサブユニットペプチドやタンパク質を使用し、ポリマーの自己凝集化に伴う抗原内封反応により抗原を担持したナノマテリアルを作製することができる(Jabbari. Pharmaceutical Research,DOI:10.1007/s11095-008-9802-1)。
〔投与部位、対象〕
本発明の粘膜ワクチンの投与部位は、粘膜であれば特に制限されない。具体的には、鼻粘膜、消化器粘膜(胃腸粘膜、腸管粘膜)、呼吸器粘膜(肺粘膜、気管粘膜)、口腔粘膜、膣粘膜、眼粘膜等が挙げられる。中でも、利便性の理由で、鼻粘膜、消化器粘膜、口腔粘膜が好ましい。
本発明の粘膜ワクチンの投与部位は、粘膜であれば特に制限されない。具体的には、鼻粘膜、消化器粘膜(胃腸粘膜、腸管粘膜)、呼吸器粘膜(肺粘膜、気管粘膜)、口腔粘膜、膣粘膜、眼粘膜等が挙げられる。中でも、利便性の理由で、鼻粘膜、消化器粘膜、口腔粘膜が好ましい。
本発明の粘膜ワクチンの投与対象としては、ヒト、サル、マウス、ラット、ウサギ、ネコ、ウシ、イヌ、ウマ、ヤギ等の哺乳動物やニワトリ等の鳥類などが挙げられる。
〔ワクチン製剤〕
本発明の粘膜ワクチンの有効成分である上記融合体の配合量については、粘膜に適用されて免疫を賦活化させる効果を奏するのに有効な量であればよく、抗原の種類、投与対象の年齢や体重、投与形態、製剤の形状や剤型により異なるが、例えば、対象がヒトの場合、1日1回~数回、好ましくは1日1回、所定の粘膜にワクチンが投与され(初回免疫)、通常2~3週間後に同様にして再投与される(追加免疫)。
本発明の粘膜ワクチンの有効成分である上記融合体の配合量については、粘膜に適用されて免疫を賦活化させる効果を奏するのに有効な量であればよく、抗原の種類、投与対象の年齢や体重、投与形態、製剤の形状や剤型により異なるが、例えば、対象がヒトの場合、1日1回~数回、好ましくは1日1回、所定の粘膜にワクチンが投与され(初回免疫)、通常2~3週間後に同様にして再投与される(追加免疫)。
本発明の粘膜ワクチンには、有効成分としての上記融合体の他に、経粘膜用医薬組成物に通常使用される担体または添加剤を適宜配合してもよい。担体または添加剤の配合割合については、医薬品分野において通常採用されている範囲に基づいて、適宜設定すればよい。配合できる担体または添加剤は特に制限されないが、例えば、水、生理食塩水、その他水性溶媒、水性または油性基剤等の各種担体;賦形剤、結合剤、pH調整剤、崩壊剤、吸収促進剤、滑沢剤、着色剤、矯味剤、香料等の各種添加剤が挙げられる。
このような添加剤として、具体的には、乳糖、白糖、マンニトール、塩化ナトリウム、ブドウ糖、炭酸カルシウム、カオリン、結晶セルロース、ケイ酸塩等の賦形剤;水、エタノール、単シロップ、ブドウ糖液、デンプン液、ゼラチン液、カルボキシメチルセルロース、カルボキシメチルセルロースNa、セラック、メチルセルロース、ヒドロキシプロピルメチルセルロース、ヒドロキシプロピルセルロース、ポリビニルピロリドン、ポリビニルアルコール、ゼラチン、デキストリン、プルラン等の結合剤;クエン酸、無水クエン酸、クエン酸ナトリウム、クエン酸ナトリウム二水和物、無水リン酸一水素ナトリウム、無水リン酸二水素ナトリウム、リン酸水素ナトリウム、リン酸二水素ナトリウム等のpH調整剤;カルメロースカルシウム、低置換度ヒドロキシプロピルセルロース、カルメロース、クロスカルメロースナトリウム、カルボキシメチルスターチナトリウム、クロスポビドン、ポリソルベート80等の崩壊剤;第4級アンモニウム塩基、ラウリル硫酸ナトリウム等の他の吸収促進剤;精製タルク、ステアリン酸塩、ポリエチレングリコール、コロイド状ケイ酸、ショ糖脂肪酸類等の滑沢剤;黄酸化鉄、黄色三二酸化鉄、三二酸化鉄、βカロテン、酸化チタン、食用色素(例えば、食用青色1号等)、銅クロロフィル、リボフラビン等の着色剤;並びにアスコルビン酸、アスパルテーム、アマチャ、塩化ナトリウム、果糖、サッカリン、粉糖等の矯味剤等が例示できる。
さらに、上記成分の他、本発明の粘膜ワクチンには、基剤として生分解性合成高分子を用いてもよい。このような生分解性合成高分子としては、例えばポリ乳酸、ポリ(乳酸-グリコール酸)共重合体、ポリヒドロキシ酪酸、ポリ(ヒドロキシ酪酸-グリコール酸)共重合体、およびこれらの混合物等が代表的なものとして挙げられる。ただし、これらに限定されるものではない。
本発明の粘膜ワクチンは、粘膜に適用できることを限度として、その形状については特に制限されず、固形状、液体状、半固形状、懸濁状、粉末状、微粒子状のいずれの形状であってもよい。
以下、実施例により本発明を詳細に説明するが、本発明はこれらに限定されるものではない。なお、実験方法において特に説明がない場合には、J. Sambrook, E. F. Fritsch & T. Maniatis (Ed.),“Molecular Cloning, A Laboratory Manual (3rd edition)”, Cold Spring Harbor Press, Cold Spring Harbor, New York (2001) や、F. M. Ausubel, R. Brent, R. E. Kingston, D. D. Moore,J.G. Seidman, J. A. Smith, K. Struhl (Ed.), “Current Protocols in Molecular Biology”, John Wiley & Sons Ltd. などの、当該技術分野における標準的なプロトコール集に記載の方法、またはそれらを改変した方法を用いた。また、市販の試薬キットや装置は、それらに添付のプロトコールや取扱説明書に従って使用した。
〔実施例1:OVA-C-CPE融合タンパク質の経鼻投与による免疫賦活化の検討〕
1.実験方法
(1-1)OVA-C-CPE発現プラスミドおよびOVA-C-CPE融合タンパク質の作製
C-CPE(配列番号1の第184位~第319位)をコードする遺伝子をpET-16bにクローニングしたpET-H10PER(J Cell Biol, 136, 1239-1247, 1997)のXhoIサイトにKpnI、SpeI、SmaI、PacIサイトを挿入したMCS-pET-H10PERプラスミドを作製した。次に、KpnI、PacI配列を付加した卵白アルブミン(OVA)遺伝子断片をPCRで増幅し、KpnIおよびPacIサイトを用いてMCS-pET-H10PERに組み込みOVA-C-CPE融合タンパク質発現プラスミドを作製した。
1.実験方法
(1-1)OVA-C-CPE発現プラスミドおよびOVA-C-CPE融合タンパク質の作製
C-CPE(配列番号1の第184位~第319位)をコードする遺伝子をpET-16bにクローニングしたpET-H10PER(J Cell Biol, 136, 1239-1247, 1997)のXhoIサイトにKpnI、SpeI、SmaI、PacIサイトを挿入したMCS-pET-H10PERプラスミドを作製した。次に、KpnI、PacI配列を付加した卵白アルブミン(OVA)遺伝子断片をPCRで増幅し、KpnIおよびPacIサイトを用いてMCS-pET-H10PERに組み込みOVA-C-CPE融合タンパク質発現プラスミドを作製した。
作製したOVA-C-CPE発現プラスミドを大腸菌株BL21に形質導入し、アンピシリン含有LB培地で培養した。IPTGを添加して融合蛋白質を発現誘導させた後、遠心分離により集菌し、菌体を超音波処理で破砕し、15000rpmの遠心分離後上清を回収した。回収した大腸菌溶解液をあらかじめ6M guanidine/EDTA、MilliQ、0.1M NiSO4、bufferA(10mM Tris-HCl (pH8.0)、400mM NaCl、5mM MgCl2、0.1mM phenylmethane sulfonyl fluoride、1mM 2-mercaptoethanol、10% glycerol)により平衡化しておいたHiTrap Chelating HPカラム(GEヘルスケアバイオサイエンス)に添加し、100mM imidazole溶液で洗浄後、400mM imidazole溶液で吸着したOVA-C-CPEを溶出した。溶出液をPD-10column(GEヘルスケアバイオサイエンス)に添加して、溶媒をPBSバッファーに置換し、各種実験に使用した。
(1-2)マウスへの免疫およびサンプルの採取
BALB/c雌性マウス(7-8週齢)にOVA単独、OVAとC-CPEとの混合液、OVA-C-CPE融合蛋白質をday0、7、14の計3回、OVA量として5μg経鼻投与した。最終投与の1週間後(day21)にマウス血清、鼻腔洗浄液、糞便抽出液、膣洗浄液を回収し、回収サンプルを-20℃にて保存した。なお、鼻腔洗浄液は安楽死させたマウスの気道より鼻腔へ向け200μLのPBSを流すことにより回収したもの、糞便抽出液は糞便10mg/100μLとなるようにPBSを加え4℃で5分間強攪拌後3000×gで10分間遠心した上清を回収したもの、膣洗浄液は膣に50μLのPBSを流し込みピペティングする操作を2回繰り返すことにより回収したものである。

(1-3)抗体価の測定
96穴NUNC Immuno plateにOVA1μg/wellを固相化し、0.05%Tween-TBS(T-TBS)で3回洗浄後、4%ブロックエース(DS Pharma Biomedical)を添加し、室温にて2時間ブロッキングした。T-TBSで5回洗浄後、T-TBSにて10倍に希釈したブロックエースを用いて検体を10~100,000倍希釈(血清)、1~1,000倍希釈(粘膜サンプル)し、50μL/wellで添加した。室温にて2時間インキュベーションした後、T-TBSで5回洗浄した。HRP標識化抗IgG抗体、HRP標識化抗IgA抗体(Bethyl Laboratories)を1/10,000に希釈し、100μL/wellで添加した。室温で1時間インキュベーション後T-TBSにて5回洗浄し、TMB solution(Thermo Scientific)を100μL/well添加し20分インキュベーション後、2M硫酸を100μL/well加えた。その後、測定波長450nm、対照波長595nmにて吸光度を測定し、吸光度が0.1-1.0の間の値を基に希釈倍率を乗じて抗体価とした。
96穴NUNC Immuno plateにOVA1μg/wellを固相化し、0.05%Tween-TBS(T-TBS)で3回洗浄後、4%ブロックエース(DS Pharma Biomedical)を添加し、室温にて2時間ブロッキングした。T-TBSで5回洗浄後、T-TBSにて10倍に希釈したブロックエースを用いて検体を10~100,000倍希釈(血清)、1~1,000倍希釈(粘膜サンプル)し、50μL/wellで添加した。室温にて2時間インキュベーションした後、T-TBSで5回洗浄した。HRP標識化抗IgG抗体、HRP標識化抗IgA抗体(Bethyl Laboratories)を1/10,000に希釈し、100μL/wellで添加した。室温で1時間インキュベーション後T-TBSにて5回洗浄し、TMB solution(Thermo Scientific)を100μL/well添加し20分インキュベーション後、2M硫酸を100μL/well加えた。その後、測定波長450nm、対照波長595nmにて吸光度を測定し、吸光度が0.1-1.0の間の値を基に希釈倍率を乗じて抗体価とした。
2.結果
図1に鼻粘膜IgAの抗体価を、図2に膣粘膜IgAの抗体価を、図3に腸管粘膜IgAの抗体価を、図4に血中IgGの抗体価を、それぞれ示した。なお、図1~4において示した抗体価は平均値±標準偏差(n=5)である。
OVA単独投与群、OVAとC-CPEとの混合液投与群では、鼻粘膜面でのIgA濃度の上昇は観察されず、血中、膣粘膜、腸管粘膜面でもほとんど抗体価の上昇は観察されなかった。一方、OVA-C-CPE投与群では、鼻粘膜におけるIgA産生のみならず、血中IgG、遠隔粘膜面である膣、腸管粘膜面におけるIgA産生が観察された。この結果から、C-CPEと抗原との融合体が粘膜ワクチン技術として有用であることが明らかとなった。
図1に鼻粘膜IgAの抗体価を、図2に膣粘膜IgAの抗体価を、図3に腸管粘膜IgAの抗体価を、図4に血中IgGの抗体価を、それぞれ示した。なお、図1~4において示した抗体価は平均値±標準偏差(n=5)である。
OVA単独投与群、OVAとC-CPEとの混合液投与群では、鼻粘膜面でのIgA濃度の上昇は観察されず、血中、膣粘膜、腸管粘膜面でもほとんど抗体価の上昇は観察されなかった。一方、OVA-C-CPE投与群では、鼻粘膜におけるIgA産生のみならず、血中IgG、遠隔粘膜面である膣、腸管粘膜面におけるIgA産生が観察された。この結果から、C-CPEと抗原との融合体が粘膜ワクチン技術として有用であることが明らかとなった。
〔実施例2:OVA-C-CPE融合タンパク質の経鼻投与による免疫賦活化特性の解析〕
免疫応答は、大別すると、細胞性免疫を主体とするTh1系、液性免疫を主体とするTh2系の反応に分けることができる。そこで、本実施例では、OVA-C-CPE融合タンパク質の免疫賦活化特性を解析した。

(1-2)マウスへの免疫
6週齢の雌性BALB/cマウスまたはC57BL/6マウスに週1回、計3回、ovalbumin(OVA、Sigma Aldrich Co.,)単独、OVA-C-CPE、OVA-C-CPEN309A、OVA-C-CPES313A、OVA-C-CPEN309A/S313A、もしくはOVA-C-CPE303を経鼻より投与した。なお、マウス1匹、1回当たりの投与量はOVAとして0.1、0.5、1.0、もしくは5μgとし、投与量が10~15μLとなるようにPBSに溶解した。ここで、OVA-C-CPEN309Aは、配列番号1の第309位のアスパラギンがアラニンに置換された改変C-CPEとOVAの融合タンパク質であり、OVA-C-CPES313Aは、配列番号1の第313位のセリンがアラニンに置換された改変C-CPEとOVAの融合タンパク質であり、OVA-C-CPEN309A/S313Aは、配列番号1の第309位のアスパラギンがアラニンに置換され、かつ、第313位のセリンがアラニンに置換された改変C-CPEとOVAの融合タンパク質であり、OVA-C-CPE303は、クローディン4との結合に必須と考えられる配列番号1の第304位~第319位を欠失したC-CPE(184-303)とOVAの融合タンパク質である。
6週齢の雌性BALB/cマウスまたはC57BL/6マウスに週1回、計3回、ovalbumin(OVA、Sigma Aldrich Co.,)単独、OVA-C-CPE、OVA-C-CPEN309A、OVA-C-CPES313A、OVA-C-CPEN309A/S313A、もしくはOVA-C-CPE303を経鼻より投与した。なお、マウス1匹、1回当たりの投与量はOVAとして0.1、0.5、1.0、もしくは5μgとし、投与量が10~15μLとなるようにPBSに溶解した。ここで、OVA-C-CPEN309Aは、配列番号1の第309位のアスパラギンがアラニンに置換された改変C-CPEとOVAの融合タンパク質であり、OVA-C-CPES313Aは、配列番号1の第313位のセリンがアラニンに置換された改変C-CPEとOVAの融合タンパク質であり、OVA-C-CPEN309A/S313Aは、配列番号1の第309位のアスパラギンがアラニンに置換され、かつ、第313位のセリンがアラニンに置換された改変C-CPEとOVAの融合タンパク質であり、OVA-C-CPE303は、クローディン4との結合に必須と考えられる配列番号1の第304位~第319位を欠失したC-CPE(184-303)とOVAの融合タンパク質である。
(1-3)サンプルの採取
最終投与より1週間後に、血清、鼻腔洗浄液、膣洗浄液、および糞便抽出液を回収した。血清サンプルは、麻酔したマウスから眼底採血により血液を回収し、3000×gで10分間遠心し、上清をPBSにて10倍に希釈し、-20℃で保存した。鼻腔洗浄液は、安楽死させたマウスの気道から鼻腔にむけて200μLのPBSを流し込み、その洗浄液を回収し、-20℃で保存した。膣洗浄液は、マウスの膣口に50μLのPBSを流し込み、10回ピペッティングを行い回収し、-20℃で保存した。糞便抽出液は、マウスの糞便を回収し、糞便10mgにつき100μLの割合でPBSを加え、4℃で10分間ボルテックスを行った。その後、3000×gで10分間遠心し、上清を回収し、-20℃で保存した。

(1-4)ELISA法によるOVA特異的抗体価の測定
前日にOVAを炭酸緩衝液(pH9.6,0.19M Na2CO3,1.67M NaHCO3)に溶解し、96穴NUNC Immuno plateに100μg/wellとなるように分注し、4℃、over nightで固相化した。4%ブロックエース(DS PHARMA BIOMEDICAL, Japan)を用いて、室温にて2時間ブロッキングした。T-TBS(10mM Tris-HCl [pH8.0], 0.1M NaCl, 0.05% Tween 20)にて10倍希釈したブロックエース(sample diluent)を用いて各種サンプルを適宜希釈し、50μg/wellでプレートに添加し、室温にて2時間反応させた。その後、ブロックエース(sample diluent)にてHRP detection antibody(IgG,IgG1,IgG2a,IgA,BETHYL Laboratories, Inc.,)を1/10,000に希釈し、100μg/wellとなるように加え、室温にて1時間反応させた。TMB solution(Thermo Scientific, Rockford, IL)を加え、室温で20分間反応させ、2M硫酸を加え反応を停止させた。マイクロプレートリーダーを用いて、主波長450nm、副波長595nmで吸光度を測定した。結果は吸光度をlog10の力価として表した。なお、プレートから各溶液を除去する際には、T-TBSによる洗浄操作を5回行った。

(1-5)脾臓細胞の回収
最終投与より1週間後に、マウスを安楽死させ、消毒用アルコールで消毒し、無菌的に脾臓を回収した。5mLの注射筒を用いて、70μmのセルストレイナー(FALCON, Becton Dickinson, Franklin, USA)上で脾臓をホモジナイズし、50mLチューブに回収した。2,000rpmで5分間遠心し、ペレットに氷冷したACT溶液(15M NH4Cl,1mM KHCO3,1mM EDTA)を加えよく懸濁し、氷中で5分間インキュベートした。その後、さらに10%FBSを含むRPMI1640を5mL添加し、セルストレイナーを通して別の50mLチューブに移した。2,000rpmで5分間遠心し、上清を除去後、10%FBSを含むRPMI1640で再懸濁した。懸濁した脾臓細胞を96-well plate(FALCON, Becton Dickinson, Franklin, USA)に1×106cells/wellで播種し、1mg/mLのOVA溶液存在下もしくは非存在下、37℃で24時間培養し、培養上清を回収し、-80℃で保存した。
最終投与より1週間後に、マウスを安楽死させ、消毒用アルコールで消毒し、無菌的に脾臓を回収した。5mLの注射筒を用いて、70μmのセルストレイナー(FALCON, Becton Dickinson, Franklin, USA)上で脾臓をホモジナイズし、50mLチューブに回収した。2,000rpmで5分間遠心し、ペレットに氷冷したACT溶液(15M NH4Cl,1mM KHCO3,1mM EDTA)を加えよく懸濁し、氷中で5分間インキュベートした。その後、さらに10%FBSを含むRPMI1640を5mL添加し、セルストレイナーを通して別の50mLチューブに移した。2,000rpmで5分間遠心し、上清を除去後、10%FBSを含むRPMI1640で再懸濁した。懸濁した脾臓細胞を96-well plate(FALCON, Becton Dickinson, Franklin, USA)に1×106cells/wellで播種し、1mg/mLのOVA溶液存在下もしくは非存在下、37℃で24時間培養し、培養上清を回収し、-80℃で保存した。
(1-6)サイトカインELISA
回収した培養上清をサイトカインアッセイキット(R&D SYSTEMS)を用いて測定した。測定方法はキットのプロトコールに従った。

2.結果
Th1系応答の指標として血清中IgG2a抗体価および免疫マウス脾臓細胞からのインターフェロン-γ(IFN-γ)産生を解析し、Th2系応答の指標として血清中IgG1抗体価、脾臓細胞からのインターロイキン-13(IL-13)産生およびインターロイキン-17(IL-17)産生を解析した。
BALB/cマウスを用いた解析の結果を図5~8に示した。図5に示したように、OVA-C-CPE免疫により、血清中IgG1、IgG2aの抗体価が共に上昇していた。さらに、免疫マウスから採取した脾臓細胞からIFN-γの産生(図6参照)、IL-13の産生(図7参照)、IL-17の産生(図8参照)が観察された。
C57BL/6マウスを用いた解析の結果を図9~12に示した。BALB/cマウスを用いたと同様に、OVA-C-CPE免疫による血清中IgG1、IgG2aの抗体価上昇(図9参照)、免疫マウスから採取した脾臓細胞からIFN-γの産生(図10参照)、IL-13の産生(図11参照)、IL-17の産生(図12参照)が観察された。
クローディン4結合性を消失したOVA-C-CPE303ではいずれの免疫応答も活性化されていなかったことから、本発明の粘膜ワクチンはTh1系およびTh2系の免疫系を活性化すると考えられた。
Th1系応答の指標として血清中IgG2a抗体価および免疫マウス脾臓細胞からのインターフェロン-γ(IFN-γ)産生を解析し、Th2系応答の指標として血清中IgG1抗体価、脾臓細胞からのインターロイキン-13(IL-13)産生およびインターロイキン-17(IL-17)産生を解析した。
BALB/cマウスを用いた解析の結果を図5~8に示した。図5に示したように、OVA-C-CPE免疫により、血清中IgG1、IgG2aの抗体価が共に上昇していた。さらに、免疫マウスから採取した脾臓細胞からIFN-γの産生(図6参照)、IL-13の産生(図7参照)、IL-17の産生(図8参照)が観察された。
C57BL/6マウスを用いた解析の結果を図9~12に示した。BALB/cマウスを用いたと同様に、OVA-C-CPE免疫による血清中IgG1、IgG2aの抗体価上昇(図9参照)、免疫マウスから採取した脾臓細胞からIFN-γの産生(図10参照)、IL-13の産生(図11参照)、IL-17の産生(図12参照)が観察された。
クローディン4結合性を消失したOVA-C-CPE303ではいずれの免疫応答も活性化されていなかったことから、本発明の粘膜ワクチンはTh1系およびTh2系の免疫系を活性化すると考えられた。
〔実施例3:OVA-C-CPE融合タンパク質の経鼻投与による予防的ワクチン効果および治療的ワクチン効果の検討〕
本発明の粘膜ワクチンのワクチン活性を解析するために、OVA発現マウス癌細胞(EG7-OVA細胞)を用いて、癌予防効果および癌治療効果を解析した。

1.実験方法
(1-1)OVA-C-CPE融合蛋白質の予防的ワクチン効果の検討
6週齢の雌性C57BL/6マウスに週1回、計3回、OVA、OVA+C-CPE混合液、OVA-C-CPE、またはOVA-C-CPE303を経鼻より投与した。なお、マウス1匹、1回当たりの投与量はOVAとして5μgとし、投与量が10~15μLとなるようにPBSに溶解した。最終投与より1週間後に、背中にEG7-OVA細胞(1×106 cells/mouse)を皮下移植した。その後、3日毎に腫瘍径を測定し、腫瘍の大きさを算出した。なお、腫瘍の大きさは長径×短径×短径/2により算出した。

6週齢の雌性C57BL/6マウスの背中にEG7-OVA細胞(1×106 cells/mouse)を皮下移植し、腫瘍が触視できるまで観察した(5~8日)。触視できた日よりマウス1匹、1回当たり、投与量が10~15μLとなるようにPBSに溶解したOVA(OVA量として10μg)、OVA-C-CPE(OVA量として10μg)、またはOVA-C-CPE303(OVA量として10μg)を週1回、計3回経鼻より投与すると同時に、腫瘍径を測定し、腫瘍の大きさを算出した。なお、腫瘍の大きさは長径×短径×短径/2により算出した。

2.結果
図13に、予防的ワクチン効果を検討した結果を示した。図13から明らかなように、OVA-C-CPEを経鼻免疫後、腫瘍細胞を移植した場合、ほぼ完全に腫瘍の増殖は抑制された。
図14に、治療的ワクチン効果を検討した結果を示した。図14から明らかなように、担癌マウスにOVA-C-CPEを経鼻免疫した場合、顕著な抗腫瘍活性が観察された。
これらの癌予防および治療効果は、クローディン4結合性を消失したOVA-C-CPE303では観察されなかったことから、本発明の粘膜ワクチンは、癌などの疾病に対する予防および治療技術として応用できるものと考えられた。
図13に、予防的ワクチン効果を検討した結果を示した。図13から明らかなように、OVA-C-CPEを経鼻免疫後、腫瘍細胞を移植した場合、ほぼ完全に腫瘍の増殖は抑制された。
図14に、治療的ワクチン効果を検討した結果を示した。図14から明らかなように、担癌マウスにOVA-C-CPEを経鼻免疫した場合、顕著な抗腫瘍活性が観察された。
これらの癌予防および治療効果は、クローディン4結合性を消失したOVA-C-CPE303では観察されなかったことから、本発明の粘膜ワクチンは、癌などの疾病に対する予防および治療技術として応用できるものと考えられた。
〔実施例4:改変C-CPEとOVAとの融合タンパク質の経鼻投与による免疫賦活化の検討〕

図15に血中IgGの抗体価を、図16に鼻粘膜IgAの抗体価を、図17に膣粘膜IgAの抗体価を、図18に腸管粘膜IgAの抗体価を、それぞれ示した。第309位のアスパラギンをアラニンに置換したOVA-C-CPEN309A、第313位のセリンをアラニンに置換したOVA-C-CPES313Aは、いずれも低用量、中用量の投与においてOVA-C-CPE群に比して血清中IgG(図15参照)、鼻粘膜IgA(図16参照)、膣粘膜IgA(図17参照)、腸管粘膜IgA(図18参照)の抗体価を10~20倍上昇させた。また、第309位のアスパラギンと第313位のセリンの両方をアラニンに置換したOVA-C-CPEN309A/S313Aは、0.5μg投与において、OVA-C-CPE群に比して血清中IgGの抗体価を100倍以上上昇させた。以上のように、C-CPEの改変体(変異体)を作製することで、野生型OVA-C-CPEを凌駕する免疫賦活化特性を有する経鼻ワクチン技術の創出に成功した。
図15に血中IgGの抗体価を、図16に鼻粘膜IgAの抗体価を、図17に膣粘膜IgAの抗体価を、図18に腸管粘膜IgAの抗体価を、それぞれ示した。第309位のアスパラギンをアラニンに置換したOVA-C-CPEN309A、第313位のセリンをアラニンに置換したOVA-C-CPES313Aは、いずれも低用量、中用量の投与においてOVA-C-CPE群に比して血清中IgG(図15参照)、鼻粘膜IgA(図16参照)、膣粘膜IgA(図17参照)、腸管粘膜IgA(図18参照)の抗体価を10~20倍上昇させた。また、第309位のアスパラギンと第313位のセリンの両方をアラニンに置換したOVA-C-CPEN309A/S313Aは、0.5μg投与において、OVA-C-CPE群に比して血清中IgGの抗体価を100倍以上上昇させた。以上のように、C-CPEの改変体(変異体)を作製することで、野生型OVA-C-CPEを凌駕する免疫賦活化特性を有する経鼻ワクチン技術の創出に成功した。
なお本発明は上述した各実施形態および実施例に限定されるものではなく、請求項に示した範囲で種々の変更が可能であり、異なる実施形態にそれぞれ開示された技術的手段を適宜組み合わせて得られる実施形態についても本発明の技術的範囲に含まれる。また、本明細書中に記載された学術文献および特許文献の全てが、本明細書中において参考として援用される。
Claims (9)
- ウエルシュ菌エンテロトキシンのC末端断片と抗原との融合体を含むことを特徴とする粘膜ワクチン。
- ウエルシュ菌エンテロトキシンのC末端断片が、毒性を発現しないことを特徴とする請求項1に記載の粘膜ワクチン。
- ウエルシュ菌エンテロトキシンのC末端断片が、以下の(A)または(B)のタンパク質であることを特徴とする請求項1または2に記載の粘膜ワクチン。
(A)配列番号1で表わされるアミノ酸配列の部分配列からなり、少なくとも第304位~第319位のアミノ酸を含むタンパク質
(B)(A)のアミノ酸配列において、1または数個のアミノ酸が欠失、置換もしくは付加されたアミノ酸配列からなり、かつクローディンと結合するタンパク質 - ウエルシュ菌エンテロトキシンのC末端断片が、配列番号1で示されるアミノ酸配列の第1位~第52位を欠失していることを特徴とする請求項3に記載の粘膜ワクチン。
- ウエルシュ菌エンテロトキシンのC末端断片が、配列番号1で示されるアミノ酸配列の第309位のアスパラギンおよび第313位のセリンの少なくとも一方が他のアミノ酸に置換されていることを特徴とする請求項3または4に記載の粘膜ワクチン。
- ウエルシュ菌エンテロトキシンのC末端断片と抗原との融合体が、ウエルシュ菌エンテロトキシンのC末端断片と抗原との融合タンパク質である請求項1~5のいずれかに記載の粘膜ワクチン。
- 抗原が、感染性病原体由来、癌細胞由来、またはアレルゲン由来である請求項1~6のいずれかに記載の粘膜ワクチン。
- 投与部位が、鼻粘膜、消化器粘膜、呼吸器粘膜、口腔粘膜、膣粘膜、または眼粘膜である請求項1~7のいずれかに記載の粘膜ワクチン。
- 粘膜ワクチンを製造するためのウエルシュ菌エンテロトキシンのC末端断片の使用。
