本発明は、従来の薬物放出性試験方法が有していた問題を解決しようとするものであり、人体、実験動物、培養細胞、血清または血漿等のような、生体または生体由来物質を使用せず、in vitroの系で、薬物を内封したリポソーム製剤の薬物放出性を測定する、簡便、正確かつ再現性に優れ、しかもIn Vivo/In Vitro相関(IVIVC)を達成できる、リポソームからの薬物放出手段およびリポソーム製剤の薬物放出性評価方法を提供することを目的とするものである。
(1)リポソームのリン脂質膜をイオン化しない状態で透過可能である;および
(2)リポソーム内水相において、脱プロトン化剤はブレンステッド塩基として働き薬物を脱プロトン化し、プロトン化剤はブレンステッド酸として働き薬物をプロトン化する。
薬物(「A」と表記する。)を封入したリポソームを、脱プロトン化試薬(「B」と表記する。)を含む溶液中に存在させ、該脱プロトン化試薬を、非イオン化状態でリポソーム脂質膜を透過して外水相から内水相に移動させる。
A:薬物(非イオン化状態);

薬物(「HC」と表記する。)を封入したリポソームを、プロトン化試薬(「HD」と表記する。)を含む溶液中に存在させ、該プロトン化試薬を、非イオン化状態でリポソーム脂質膜を透過して外水相から内水相に移動させる。
プロトン化試薬(HD)は、アニオン化した状態で存在する薬物(C-)に水素イオン(プロトン)を供与して脱プロトン化され(D-)、その一方で薬物(C-)はプロトン化試薬から水素イオン(プロトン)を受容してプロトン化される(HC)。

<リポソーム-膜構造>
本発明において、リポソームとは、リン脂質二重膜で形成される閉鎖小胞を意味するが、そのようなリポソームを含んでなる懸濁液であるリポソーム製剤をも意味する場合がある。
本発明のリポソームの粒径は、EPR効果を利用できる範囲で設定されることが好ましい。より詳細には、リポソームの粒径は、200nm以下とすることが好ましく、50~200nmとすることがより好ましい。ただし、EPR効果を利用することを求めない場合は、この限りではない。
本発明のリポソームのリン脂質膜を構成する主膜材であるリン脂質は、当業者が知るリン脂質を単一種または複数種組合せで使用することができるが、その相転移点が、生体内温度(35~37℃)より高いことが好ましく、40℃以上であることがより好ましい。
本発明のリポソームのリン脂質膜は、リポソームを安定的に形成できるものであれば、リン脂質以外の膜成分を含むこともできる。
本発明のリポソームは、その表面のリン脂質膜が修飾されてもよい。
本発明の薬物内封リポソームは、好ましくはリモートローディング法を用いて製造される。リモートローディング法の方法については特に限定されないが、クエン酸緩衝液や硫酸アンモニウムを用いた方法が例示される。
本発明の薬物内封リポソームに保持される薬物は、リモートローディング法によってリポソーム内に封入し、保持可能であればよく特に限定されないが、両親媒性化合物が好ましく、両親媒性弱塩基性化合物がより好ましい。
両親媒性弱塩基性化合物をリポソーム内に封入するために用いられるリポソームの内部水相は、対イオンの選択が重要である。
本発明において、外水相溶液はシフト試薬と混合する前には、シフト試薬を含まない。例えば、脱プロトン化剤およびその共役酸ならびにプロトン化剤およびその共役塩基等に例示されるシフト試薬に該当する物質を含まない。
本発明でいうシフト試薬は、それを用いることによって、薬物を封入したリポソームの内水相と外水相との間に、薬物封入時に形成されたイオン勾配とは正反対のイオン勾配を形成し、薬物を保持するために形成されているイオン勾配を弱め、内水相の化学平衡の移動(シフト)を引き起こすことによって、内水相に溶解して保持されている薬物の保持力を弱め、リポソーム内部から薬物を放出しやすい環境を作り出すと考えられる物質である。例えば、脱プロトン化試薬またはプロトン化試薬が例示される。
本発明の薬物放出手段および薬物放出性評価方法においては、リポソーム製剤とシフト試薬を添加した溶液との混合系は、所定の温度で、所定の時間加温され放出を促進することができる。
本発明の薬物放出手段および薬物放出性評価方法においては、リポソーム製剤とシフト試薬を添加した溶液との混合系は、所定の温度で、所定の時間加温され放出を促進することができる。
本発明の薬物放出性評価方法においては、放出された薬物をリポソームと分離することが好ましい。
本発明の薬物放出試験において使用する容器または機器は、マイクロチューブから既存の溶出試験器を使用することが可能であり、特に限定されない。
塩化ナトリウム5.84gと、リン酸二水素ナトリウム二水和物15.60gとを、水900mLに溶かした。次に、その溶液にリン酸緩衝液を加えてpH3.0に調整し、さらに水を加えて全容量を1000mLとした。
これを停止液(本明細書において「放出停止液」ともいう。)とした。
なお、上記調製方法に従って調製した停止液の浸透圧は300mOsmであった。
リン脂質定量キット(リン脂質Cテストワコー、和光純薬工業株式会社)を用いてリポソーム分散液中でのリン脂質(HSPC、DPPC,DMPC,DSPC)濃度(単位:mg/mL)を測定した。
リポソーム分散液20μLを生理食塩水で3mLに希釈し、ゼータサイザー(Zetasizer) 3000 HS(マルバーン(Malvern)社製)を用いて、フォトン相関分光法によって平均粒子径(単位:nm)を測定した。
(1)ビンクリスチン(VCR)定量方法
後記する調製例の記載に従って調製したVCR含有リポソーム100μLを2mLのメタノール中に分散し、試料溶液とした。
これとは別に、濃度の異なるVCR水溶液を100μLとり、2mLのメタノール中に分散させ、検量線用標準溶液を調製した。
これらの液について、下記測定条件に従いHPLC法により測定を実施した。
VCR濃度は検量線式より算出した。

後記する調製例の記載に従って調製したVCR含有リポソームを生理食塩水で10倍希釈した後、超遠心分離によりリポソームに含有されていない薬物を分離し、試料溶液とする。別に、濃度の異なるVCR水溶液を100μLとり、水900μLを加えて、検量線用標準溶液を調製した。試料溶液および標準溶液につき、VCR定量に示したHPLC条件に従い、定量を行った。
試料を停止液で10倍希釈した後、超遠心分離によりリポソームに含有されていない薬物を分離し、試料溶液とする。別に、濃度の異なるVCR水溶液を100μLとり、水900μLを加えた。さらに、この液を水で10倍希釈し、検量線用標準溶液とした。試料溶液および標準溶液につき、VCR定量に示したHPLC条件に従い、定量を行ったが、本定量方法に限り、注入量を50μLとした。
後記する調製例の記載に従って調製したCFX含有リポソーム100μLを2mLのメタノール中に分散し、試料溶液とする。別に、濃度の異なるCFX水溶液を100μLとり、2mLのメタノール中に分散させ、検量線用標準溶液を調製した。これらの液について、下記測定条件に従いHPLC法により測定を実施した。CFX濃度は検量線式より算出した。

試料を停止液で10倍希釈した後、超遠心分離によりリポソームに含有されていない薬物を分離し、試料溶液とする。別に、濃度の異なるVCR水溶液を100μLとり、水900μLを加えた。さらに、この液を水で10倍希釈し、検量線用標準溶液とした。試料溶液および標準溶液につき、CFX定量に示したHPLC条件に従い、定量を行った。
後記する調製例の記載に従って調製したDXR含有リポソーム0.5mLを20mLのメタノール中に分散した。この溶液1mLにpH3.0の0.1mol/Lリン酸ニ水素ナトリウム溶液1mLを加え混合し、試料溶液とした。別に、濃度の異なるDXR溶液1mLに、pH3.0の0.1mol/Lリン酸ニ水素ナトリウム溶液1mLを加え混合し、検量線用標準溶液を調製した。試料溶液および標準溶液について、下記測定条件に従いHPLC法により測定を実施した。DXR濃度は検量線式より算出した。

後記する調製例の記載に従って調製したDXR含有リポソームを生理食塩水で10倍希釈した後、超遠心分離によりリポソームに含有されていない薬物を分離した。超遠心後サンプルの上清50μLを蛍光分析用メタノール2mLに分散し試料溶液とした。別に、濃度の異なるDXR水溶液50μLを蛍光分析用メタノール2mLに分散し、検量線用標準溶液を調製した。試料溶液および標準溶液について、励起波長480nmならびに蛍光波長580nmでの蛍光強度を分光蛍光光度計にて定量した。
試料を停止液で10倍希釈した後、超遠心分離によりリポソームに含有されていない薬物を分離した。超遠心後サンプルの上清1mLにpH3.0の0.1mol/Lリン酸ニ水素ナトリウム溶液1mLを加え混合し、試料溶液とした。別に、濃度の異なるDXR溶液1mLに、pH3.0の0.1mol/Lリン酸ニ水素ナトリウム溶液1mLを加え混合し、検量線用標準溶液を調製した。試料溶液および標準溶液について、DXR定量に示したHPLC条件に従い、定量を行った。

1.VCR含有リポソームの製造
以下の工程に従い、VCR含有リポソームを製造した。
HSPC 0.71gおよびChol. 0.29gを、それぞれ量りとった。これらを無水エタノール1mLと混合し、70℃の恒温層で加温、溶解して、脂質のエタノール溶液を得た。このエタノール溶液に濃度250mMのクエン酸水溶液およびスクロースを添加することにより、このエタノール溶液の浸透圧を500mOsmとした。次に、このエタノール溶液に、pH2に調整した液9mLを添加し、さらに加温して、脂質分散体を得た。
得られた脂質分散体を、約70℃に加温したエクストルーダー(Extruder) T.10(リペックス・バイオメンブランズ(Lipex Biomembranes)社製)に二枚重ねで取り付けたフィルター(0.1μm、ポリカーボネートメンブラン;ワットマン(Whatman)社製)を通して整粒化を行い、整粒化後リポソームの懸濁液を得た。
PEG5000-DSPE水溶液(濃度約0.04g/mL)を、PEG5000-DSPEの含有量が前記の量りとった総脂質量(HSPCとChol.との和)の0.75mol%に相当する量となる量準備した。このPEG5000-DSPE水溶液を予め65℃に設定した恒温槽中で加温した。このPEG5000-DSPE水溶液と前記の整粒化後リポソームの懸濁液とを混合した。混合後、65℃に設定した恒温槽中でさらに30分間加温して、PEG修飾リポソームの懸濁液を得た。
移動相を10%スクロース/10mMヒスチジン溶液(pH6.5)で置換したゲルカラム(セファロース 4 ファストフロー(Sephalose 4 Fast Flow);アマシャムバイオサイエンス社製)を準備した。このゲルカラムを用いて、前記のPEG修飾リポソームの懸濁液の外液を10%スクロース/10mMヒスチジン溶液(pH6.5)で置換し、外液置換後リポソームの懸濁液を得た。外液置換により、リポソーム膜内外にpH勾配を形成した。
前記の外液置換後リポソームの懸濁液に、VCRとHSPCの質量比(VCR/HSPC)が0.14(w/w)となるように、VCR水溶液を加えた。これを60℃の恒温槽中で30分間加温して、薬物封入を行い、薬物封入後リポソームの懸濁液を得た。
移動相を10%スクロース/10mMヒスチジン溶液(pH6.5)で置換したゲルカラム(セファロース 4 ファストフロー)を準備した。このゲルカラムを用い、10%スクロース/10mMヒスチジン溶液(pH6.5)を移動相として使用して、前記の薬物封入後リポソームの懸濁液の外液中に残存する薬物を除去し、未封入薬物除去後リポソームの懸濁液を得た。
最後に、この懸濁液をフィルター(0.2μm)ろ過し、フィルターろ過後リポソームを得た。
表1に、調製例1に従って製造された未封入薬物除去後リポソーム(以下「LIP1」という。)のVCR濃度および脂質濃度、フィルターろ過後リポソームのVCR濃度および粒子径を示す。なお、VCR濃度の測定(VCR定量)、脂質濃度の測定および粒子径の測定は前記した方法により行った。
本実施例において、薬物を担持したリポソームからのリポソーム外液への放出に関して、シフト試薬濃度による影響を検証した。なお、本実施例において、シフト試薬として、酢酸アンモニウムを使用した。
アンモニウムイオン濃度が5、10、25、50、100mMとなるように酢酸アンモニウムを溶解し、溶液のpHを7.0に0.1M水酸化ナトリウム試液を用いて調整した。さらに、この液に塩化ナトリウムを加えて溶液浸透圧を300mOsmとし、シフト試薬溶液とした。
調製例1に示したリポソームを上記シフト試薬溶液で10倍希釈し、37℃で加温した。加温開始後、2.5、5、10、15、30分で試料を取り出した。なお、サンプルは使用するまで氷冷下で保存した。放出したVCRは、上記に述べた方法に従い定量を実施した。
図4にVCR封入リポソームからのVCR放出に関するシフト試薬溶液中のシフト試薬濃度への影響を検証した結果を示した。その結果、VCRの放出は、いずれのシフト試薬濃度を持つシフト試薬溶液においても、経時的な増加を示し、さらに、このVCR放出量はシフト試薬濃度依存的に増加することが明らかとなった。
シフト試薬濃度と放出速度定数は高い相関性(r2=0.9975)を示すことから、リポソームからの薬物の放出は、シフト試薬濃度に依存し、薬物の放出は、シフト試薬濃度を調節することにより容易に制御できることが明らかとなった。
本実施例において、薬物を担持したリポソームからのリポソーム外液への放出に関して、シフト試薬溶液のpHによる影響を検証した。なお、本実施例において、シフト試薬として、酢酸アンモニウムを使用した。
アンモニウムイオン濃度として50mMなるように酢酸アンモニウムを溶解し、溶液のpHを4.0、5.0、6.0、7.0、8.0に0.1M水酸化ナトリウム試液を用いてそれぞれ調整した。さらに、この液に塩化ナトリウムを加えて溶液浸透圧を300mOsmとし、シフト試薬溶液とした。
調製例1に示したリポソームをシフト試薬溶液で10倍希釈し、37℃で加温した。加温開始後、2.5、5、10、15、30分でサンプリングした。なお、サンプルは使用するまで氷冷下で保存した。放出したVCRは、上記に述べた方法に従い定量を実施した。
図6は、シフト試薬溶液のpHを4.0、5.0、6.0、7.0または8.0と設定したときのVCRの放出挙動の変化を示したものである。
VCRの放出挙動は、シフト試薬溶液のpHに依存することが明らかとなり、本実施例においては、pHの増加と共にVCRの放出性が促進されることが明らかとなった。
本実施例において、薬物を担持したリポソームからのリポソーム外液への放出に関して、シフト試薬溶液の浸透圧による影響を検証した。なお、本実施例において、シフト試薬として、酢酸アンモニウムを使用した。
アンモニウムイオン濃度として50mMとなるように酢酸アンモニウムを溶解し、pH7.0とした。この液に塩化ナトリウムを加えて、浸透圧を100mOsmから750mOsmに調整し、シフト試薬溶液とした。
調製例1に示したリポソームをシフト試薬溶液で10倍希釈し、37℃で加温した。加温開始後、2.5、5、10、15、30分でサンプリングした試料を取り出した。なお、サンプルは使用するまで氷冷下で保存した。放出したVCRは、上記に述べた方法に従い定量を実施した。
図7は、リポソームからのVCR放出におけるシフト試薬溶液の浸透圧の影響を示した図である。
VCRの放出率は、浸透圧に大きく影響され、シフト試薬溶液の浸透圧が低いほどVCRの放出が促進されることが明らかとなった。
1.VCR含有リポソームの製造
以下の工程に従い、VCR含有リポソームを製造した。
HSPC 0.71gおよびChol. 0.29gを、それぞれ量りとった。これらを無水エタノール1mLと混合し、70℃の恒温槽で加温、溶解して、脂質のエタノール溶液を得た。このエタノール溶液に濃度250mMのクエン酸水溶液およびスクロースを添加することにより、このエタノール溶液の浸透圧を500mOsmに調整した。次に、このエタノール溶液に、pH2.5に調整した液9mLを添加し、さらに加温して、脂質分散体を得た。
得られた脂質分散体を、約70℃に加温したエクストルーダー T.10(リペックス・バイオメンブランズ社製)に二枚重ねで取り付けたフィルター(0.1μm、ポリカーボネートメンブラン;ワットマン社製)を通して整粒化を行い、整粒化後リポソームの懸濁液を得た。
PEG5000-DSPE水溶液(濃度約0.04g/mL)を、PEG5000-DSPEの含有量が前記の量りとった総脂質量(HSPCとChol.との和)の0.75mol%に相当する量となる量準備した。このPEG5000-DSPE水溶液を予め65℃に設定した恒温槽中で加温した。このPEG5000-DSPE水溶液と前記の整粒化後リポソームの懸濁液とを混合した。混合後、65℃に設定した恒温槽中でさらに30分間加温して、PEG修飾リポソームの懸濁液を得た。
移動相を10%スクロース/10mMヒスチジン溶液(pH6.5)で置換したゲルカラム(セファロース 4 ファストフロー;アマシャムバイオサイエンス社製)を準備した。このゲルカラムを用いて、前記のPEG修飾リポソームの懸濁液の外液を10%スクロース/10mMヒスチジン溶液(pH6.5)で置換し、外液置換後リポソームの懸濁液を得た。外液置換により、リポソーム膜内外にpH勾配を形成した。
前記の外液置換後リポソームの懸濁液に、VCRとHSPCの質量比(VCR/HSPC)が0.22(w/w)となるように、VCR水溶液を加えた。これを60℃の恒温槽中で30分間加温して、薬物封入を行い、薬物封入後リポソームの懸濁液を得た。
移動相を10%スクロース/10mMヒスチジン溶液(pH6.5)で置換したゲルカラム(セファロース 4 ファストフロー)を準備した。このゲルカラムを用い、10%スクロース/10mMヒスチジン溶液(pH6.5)を移動相として使用して、前記の薬物封入後リポソームの懸濁液の外液中に残存する薬物を除去し、未封入薬物除去後リポソームの懸濁液を得た。
最後に、この懸濁液をフィルター(0.2μm)ろ過し、フィルターろ過後リポソームを得た。
表2に、調製例2に従って製造されたVCR含有リポソーム(以下「LIP2」という。)のリポソーム特性を示す。なお、リポソーム中の薬物濃度の測定(VCR定量)および粒子径の測定は前記した方法により行った。
本実施例においてシフト試薬として一級アミン(2-アミノエタノール、エチレンジアミン)、二級アミン(ジエチルアミン)を用いた時のVCR放出挙動について示す。なお、本実施例において、対照シフト試薬としてアンモニウム塩である酢酸アンモニウムを用い、リポソームは、LIP2を用いた。
シフト試薬を量りとり、リン酸緩衝液を加えてpH7.4、50mMのシフト試薬溶液を調製した。
リポソームをシフト試薬溶液を加えて10倍希釈し、37℃で加温した。加温開始後、2.5、5、10、15、30分でサンプリングした。なお、サンプルは使用するまで氷冷下で保存した。放出したVCRは、上記に述べた方法に従い定量を実施した。
図8は、リポソームからのVCR放出におけるシフト試薬の影響を示した図である。
アミノ化合物を用いることによりその放出挙動は異なるが、VCRを放出させることができることが明らかとなり、アンモニウム塩以外でもアミノ化合物を本放出試験におけるシフト試薬として選択可能であることが示された。
実施例1から4に示すように、リポソームからの薬物の放出は、シフト試薬溶液の溶液特性により影響されることが明らかになった。つまり、リポソームからの薬物の放出には、外部環境により大きく異なることが示唆される。このような観点から、in vivoを想定した薬物放出性を評価する場合には、生体に近い溶液特性に調整することが望ましいことが示唆された。
1.VCR含有リポソームの製造
以下の工程に従い、膜物性の異なるVCR含有リポソームを製造した。
PC1(HSPC、DSPC、DPPCまたはDMPC)およびChol.を、表3に示す秤量値(単位:g)のとおり、それぞれ量りとった。これらを無水エタノール1mLと混合し、70℃の恒温層で加温、溶解して、脂質のエタノール溶液を得た。このエタノール溶液に濃度250mMのクエン酸水溶液を添加して、エタノール溶液のpHを2.5に調整した。塩化ナトリウムを添加することにより、このエタノール溶液の浸透圧を500mOsmに調整した。さらに、70℃に設定した恒温槽中で、このエタノール溶液を加温して、脂質分散体を得た。
得られた脂質分散体を、約70℃に加温したエクストルーダー T.10(リペックス・バイオメンブランズ社製)に二枚重ねで取り付けたフィルター(0.1μm、ポリカーボネートメンブラン;ワットマン社製)を通して整粒化を行い、整粒化後リポソームの懸濁液を得た。
PEG5000-DSPE水溶液(濃度約0.04g/mL)を、PEG5000-DSPEの含有量が前記の量りとった総脂質量(PC1とChol.との和)の0.75mol%に相当する量となる量準備した。このPEG5000-DSPE水溶液を予め65℃に設定した恒温槽中で加温した。このPEG5000-DSPE水溶液と前記の整粒化後リポソームの懸濁液とを混合した。混合後、65℃に設定した恒温槽中でさらに30分間加温して、PEG修飾リポソームの懸濁液を得た。
移動相を10%スクロース/10mMヒスチジン溶液(pH6.5)で置換したゲルカラム(セファロース 4 ファストフロー;アマシャムバイオサイエンス社製)を準備した。このゲルカラムを用いて、前記のPEG修飾リポソームの懸濁液の外液を10%スクロース/10mMヒスチジン溶液(pH6.5)で置換し、外液置換後リポソームの懸濁液を得た。外液置換により、リポソーム膜内外にpH勾配を形成した。
前記の外液置換後リポソームに含まれるリン脂質を、リン脂質測定キットを用いて定量した。前記の外液置換後リポソームの懸濁液に、リン脂質定量結果から算出された総脂質濃度に対するVCR/PC1の値が0.10mol/molとなるように、VCR水溶液を加えた。これを60℃の恒温槽中で30分間加温して、薬物封入を行い、薬物封入後リポソームの懸濁液を得た。
移動相を10%スクロース/10mMヒスチジン溶液(pH6.5)で置換したゲルカラム(セファロース 4 ファストフロー)を準備した。このゲルカラムを用い、10%スクロース/10mMヒスチジン溶液(pH6.5)を移動相として使用して、前記の薬物封入後リポソームの懸濁液の外液中に残存する薬物を除去し、未封入薬物除去後リポソームの懸濁液を得た。
最後に、この懸濁液をフィルター(0.2μm)ろ過し、フィルターろ過後リポソームを得た。
表4に、調製例3に従って製造された膜物性の異なるVCR含有リポソーム(以下、「LIP3」、「LIP4」、「LIP5」、「LIP6」または「LIP7」という)のリポソーム特性を示す。
本実施例において、リポソームはLIP3から7のリポソームを用いて検討を行った。
pH7.4のリン酸緩衝液にアンモニウムイオン濃度として50mMとなるように酢酸アンモニウムを溶解した。さらに、この液に塩化ナトリウムを加えて300mOsmの浸透圧に調整した。この液をシフト試薬溶液として使用した。
リポソームをシフト試薬溶液で10倍希釈し、37℃で加温した。加温開始後、2.5、5、10、15、30分でサンプリングした。なお、サンプルは使用するまで氷冷下で保存した。放出したVCRは、上記に述べた方法に従い定量を実施した。
図9は、異なるリン脂質とコレステロールの組成比をもつリポソームからの薬物放出性を示したものである。
リポソームからの薬物放出性は、リン脂質とコレステロールの比が54:46と60:40の間には薬物放出性に明確な差が認められた。この結果は、コレステロール含量により膜流動性は変化することが知られていることから、膜流動性変化による薬物放出性の変化と推察される。従って、本実施例でえられた放出の差は、リポソームの流動性が大きく影響し、本発明の薬物放出性評価方法により、膜の流動性を評価できることが明らかとなった。
VCRの放出は、DMPC>DPPC>DSPCの順に高くなることが明らかとなった。その一方で、脂質の相転移温度として示される膜流動性は、DMPC>DPPC>DSPCの順に高い。リポソームからの薬物の放出は、膜流動性が高いものほど、高い放出性を示すことが明らかとなっている。従って、本実施例で得られたリポソームからの薬物放出性と相転移温度の関係は、これまでの知見と一致した。
1.DXR含有リポソームおよびVCR含有リポソームの製造
以下の工程に従い、VCR含有リポソームおよびDXR含有リポソームを製造した。
HSPC 0.71gおよびChol. 0.29gを、それぞれ量りとった。これらを無水エタノール1mLと混合し、70℃の恒温層で加温、溶解して、脂質のエタノール溶液を得た。このエタノール溶液に濃度250mMのクエン酸水溶液およびスクロースを添加することにより、このエタノール溶液の浸透圧を500mOsmとした。次に、このエタノール溶液に、pH2.5に調整した液9mLを添加し、さらに加温して、脂質分散体を得た。

DXR含有リポソームの製造においては、前記の外液置換後リポソームの懸濁液に、DXRとHSPCとの質量比(DXR/HSPC)が0.14(w/w)となるように、DXR水溶液を加えた。これを60℃の恒温槽中で30分間加温して、DXR封入を行い、DXR封入後リポソームの懸濁液を得た。
VCR含有リポソームの製造においては、前記の外液置換後リポソームの懸濁液に、VCRとHSPCの質量比(VCR/HSPC)が0.22(w/w)となるように、VCR水溶液を加えた。これを60℃の恒温槽中で30分間加温して、VCR封入を行い、VCR封入後リポソームの懸濁液を得た。
移動相を10%スクロース/10mMヒスチジン溶液(pH6.5)で置換したゲルカラム(セファロース 4 ファストフロー)を準備した。このゲルカラムを用い、10%スクロース/10mMヒスチジン溶液(pH6.5)を移動相として使用して、前記のDXR封入後リポソームの懸濁液またはVCR封入後リポソームの懸濁液の外液中に残存する薬物を除去し、未封入薬物除去後リポソームの懸濁液を得た。
最後に、この懸濁液をフィルター(0.2μm)ろ過し、フィルターろ過後リポソームを得た。
表5に、調製例4に従って製造されたDXR含有リポソーム(以下「LIP8」という。)およびVCR含有リポソーム(以下「LIP9」という。)のリポソーム特性を示す。なお、リポソーム中の薬物濃度の測定(DXR定量またはVCR定量)および粒子径の測定は前記した方法により行った。
本実施例においてリポソームはLIP8およびLIP9を使用した。
pH7.4のリン酸緩衝液にアンモニウムイオン濃度として50mMとなるように酢酸アンモニウムを溶解した。さらに、この液に塩化ナトリウムを加えて300mOsmの浸透圧に調整した。この液をシフト試薬溶液として使用した。
リポソームをシフト試薬溶液で10倍希釈し、37℃で加温した。加温開始後、2.5、5、10、15、30分でサンプリングした。なお、サンプルは使用するまで氷冷下で保存した。放出したDXRおよびVCRは、上記に記載した定量方法に従いそれぞれ測定した。
図11は、リポソームからのDXR放出またはVCR放出の挙動を示した図である。
DXR含有リポソーム(LIP8)は、薬物をほとんど放出しなかったが、VCR含有リポソーム(LIP9)は、薬物放出量を経時的に増加した。
本実施例は、DXR含有リポソームおよびVCR含有リポソームの血中体内動態を評価したものである。
リポソームは調製例4で製造したLIP8(DXR含有リポソーム)およびLIP9(VCR含有リポソーム)を使用した。
血中体内動態を評価する実験動物としては、SD系雄性ラットを用いた。
薬物投与量として0.10μmol/kgをSD系雄性ラットの尾静脈より注入した。
注入後、ラット尾静脈より経時的に採血を行った。得られた血液を遠心分離し(5000rpm、10分)、血清を得た。この血清40μLにメタノール200μLを添加した後、さらに遠心分離を行い(3500rpm、10分)、上清を回収し、試料溶液とした。
各試料溶液について、DXR定量およびVCR定量を前記の方法に従って行った。ただし、本実施例におけるDXR定量では、検出器として蛍光光度計を使用し,その測定波長は、励起波長485nm、蛍光波長590nmとし、注入量は100μLとした。また、本実施例におけるVCR定量においては、注入量を50μLとした。
図12は、DXRおよびVCRの薬物血中動態を示す図である。また、表6は、図12で得られた薬物動態から薬物動態学的パラメーターを1-コンパートメントモデル(1-compartment model)に従い算出した結果を示す表である。
図12に示すように、薬物の違いによりリポソームの薬物体内動態は異なり、また、表6に示すように、薬物の滞留性は、DXR含有リポソーム(LIP8)よりもVCR含有リポソーム(LIP9)の方が低いことが明らかとなった。
1.VCR含有リポソームおよびCFX含有リポソームの製造
以下の工程に従い、VCR含有リポソームおよびCFX含有リポソームを製造した。
HSPC 7.06gおよびChol. 2.94gを、それぞれ量りとった。これらを無水エタノール10mLと混合し、70℃の恒温槽で加温、溶解して、脂質のエタノール溶液を得た。これとは別に、濃度250mMの硫酸アンモニウム水溶液90mLを準備し、予め70℃の恒温槽で加温しておいた。前記エタノール溶液に、この硫酸アンモニウム水溶液を添加し、さらに加温して、脂質分散体を得た。
得られた脂質分散体を、約70℃に加温したエクストルーダー(Extruder) T.100(リペックス・バイオメンブランズ社製)に取り付けたフィルター(ポリカーボネートメンブラン,孔径0.2μm×3回,0.1μm×10回;ワットマン社製)を順次通して整粒化を行い、整粒化後リポソ-ムを得た。
PEG5000-DSPE水溶液(濃度約37.7mg/mL)を、PEG5000-DSPEの含有量が前記の量りとった総脂質量(HSPCとChol.との和)の0.75mol%に相当する量となる量準備した。このPEG5000-DSPE水溶液を予め65℃に設定した恒温槽中で加温した。このPEG5000-DSPE水溶液と前記の整粒化後リポソームの懸濁液とを混合した。混合後、65℃に設定した恒温槽中でさらに30分間加温して、PEG修飾リポソームの懸濁液を得た。
前記のPEG修飾リポソームの懸濁液の外液を、クロスフローろ過ユニット(ビバフロー50、ビバサイエンス社製;MWCO 100,000)を用いて10%スクロース/10mMヒスチジン溶液(pH6.5)に置換し、外液置換後リポソームの懸濁液を得た。
VCR含有リポソームの製造においては、前記の外液置換後リポソームの懸濁液に、リン脂質定量キットを用いて測定した脂質濃度に基づき、VCRとHSPCとの質量比(VCR/HSPC)が0.10(w/w)となるように、VCR水溶液を加えた。これを55℃の恒温槽中で30分間加温し、VCR封入後リポソームの懸濁液を得た。
CFX含有リポソームの製造においては、前記の外液置換後リポソームの懸濁液に、リン脂質定量キットを用いて測定した脂質濃度に基づき、CFXとHSPCとの質量比(CFX/HSPC)が0.04(w/w)となるように、CFX水溶液を加えた。これを55℃の恒温槽中で30分間加温し、CFX封入後リポソームの懸濁液を得た。
クロスフローろ過ユニット(ビバフロー50、ビバサイエンス社製;MWCO 100,000)を用いて、試料量が一定となるように10%スクロース/10mMヒスチジン溶液(pH6.5)を供給しながら、前記の薬物封入後リポソームの懸濁液(VCR封入後リポソームの懸濁液またはCFX封入後リポソームの懸濁液)に含まれる未封入薬物を除去し、未封入薬物除去後リポソームの懸濁液を得た。
次に、未封入薬物除去後リポソームの薬物定量を行った。
最後に、得られた未封入薬物除去後リポソームの懸濁液をフィルター(0.2μm)ろ過し、フィルターろ過後リポソームを得た。
表7に、調製例5に従って製造されたVCR含有リポソーム(以下「LIP10」という。)およびCFX含有リポソーム(以下「LIP11」という。)のリポソーム特性を示す。
本実施例において、リポソームはLIP10およびLIP11を使用した。
酢酸アンモニウム1.9gを量り、0.2mol/Lリン酸水素ニナトリウム溶液210mL、0.2mol/Lリン酸ニ水素ナトリウム溶液40mLを加えて、さらに水を加えて500mLとし、pH6.5のシフト試薬溶液を調製した。
調製例1に示したリポソームを上記シフト試薬溶液で10倍希釈し、37℃で加温した。加温開始後、5、10、15、30分で試料を取り出した。なお、サンプルは使用するまで氷冷下で保存した。放出したVCRおよびCFXは、上記に述べた方法に従い定量を実施した。
図13にVCR含有リポソーム(LIP10)およびCFX含有リポソーム(LIP11)の薬物放出率の経時的変化を示す。
同一の膜を用いても内封する薬物が異なることによりその放出性は大きく異なることが図13からわかる。これは、膜に対する親和性に由来するものであると考えられる。
1.DXR含有リポソームの製造
以下の工程に従い、リン脂質がHSPCまたはDMPCのDXR含有リポソームを製造した。
HSPC 0.70gおよびChol. 0.29gを、それぞれ量りとった。また、DMPC 0.67gおよびChol. 0.33gをそれぞれ量りとった。これらを無水エタノール1mLと混合し、70℃の恒温槽で加温、溶解して、脂質のエタノール溶液を得た。これとは別に、濃度250mMの硫酸アンモニウム溶液9mLを準備し、予め70℃の恒温槽で加温しておいた。前記エタノール溶液に、この硫酸アンモニウム水溶液を添加し、さらに加温して、脂質分散体を得た。
得られた脂質分散体を、約70℃に加温したエクストルーダー T.10、リペックス・バイオメンブランズ社製)に取り付けたフィルター(ポリカーボネートメンブラン,孔径0.2μm×3回,0.1μm×10回;ワットマン社製)を順次通して整粒化を行い、整粒化後リポソ-ムを得た。
PEG5000-DSPE水溶液(濃度約37.7mg/mL)を、PEG5000-DSPEの含有量が前記の量りとった総脂質量(HSPCとChol.との和)の0.75mol%に相当する量となる量準備した。このPEG5000-DSPE水溶液を予め65℃に設定した恒温槽中で加温した。このPEG5000-DSPE水溶液と前記の整粒化後リポソームの懸濁液とを混合した。混合後、65℃に設定した恒温槽中でさらに30分間加温して、PEG修飾リポソームの懸濁液を得た。
移動相を10%スクロース/10mMヒスチジン溶液(pH7.4)で置換したゲルカラム(セファロース 4 ファストフロー;アマシャムバイオサイエンス社製)を準備した。このゲルカラムを用いて、前記のPEG修飾リポソームの懸濁液の外液を10%スクロース/10mMヒスチジン溶液(pH7.4)で置換し、外液置換後リポソームの懸濁液を得た。
前記の外液置換後リポソームの懸濁液に、リン脂質定量キットを用いて測定した総脂質濃度に基づき、DXRと脂質のモル比(DXR/総脂質)が0.10(mol/mol)となるように、DXR溶液を加えた。これを60℃の恒温槽中で60分間加温し、薬物封入後リポソームの懸濁液を得た。
移動相を10%スクロース/10mMヒスチジン溶液(pH6.5)で置換したゲルカラム(セファロース 4 ファストフロー)を準備した。このゲルカラムを用い、10%スクロース/10mMヒスチジン溶液(pH6.5)を移動相として使用して、前記の薬物封入後リポソームの懸濁液の外液中に残存する薬物を除去し、未封入薬物除去後リポソームの懸濁液を得た。
最後に、この懸濁液をフィルター(0.2μm)ろ過し、フィルターろ過後リポソームを得た。
表8に、調製例6に従って製造したDXR含有リポソーム(LIP12およびLIP13)のリボソーム特性を示す。DXR定量、脂質濃度測定および粒子径は、フィルターろ過後リポソームについて、前記した方法により行った。
本実施例においては、シフト試薬としてエチレンジアミンを用いたときのDXR放出挙動について示す。なお、リポソームはLIP12およびLIP13を用いた。
シフト試薬を量りとり、リン酸緩衝液を加えてpH7.4、250mMのシフト試薬溶液を調製した。
DXR含有リポソーム(調製例6で調製したLIP12およびLIP13)を前記のシフト試薬溶液で10倍希釈し、37℃で加温した。加温開始後、0、2、4時間で試料を取り出した。なお、サンプルは使用するまで氷冷下で保存した。放出したDXRは、上記した「放出したDXRの定量方法」により定量を行った。

- 薬物を内封したリポソームを、シフト試薬を添加した溶液中に存在させ、該溶液中の該薬物の濃度を測定する、リポソーム製剤の薬物放出性評価方法。
- 前記リポソームの内水相において化学平衡の移動が引き起こされ、前記薬物が前記リポソームの外水相に放出される、請求項1に記載の方法。
- 前記薬物を内封したリポソームがリモートローディング法により薬物を内封したリポソームである、請求項1または2に記載の方法。
- 前記シフト試薬は、非イオン化状態で前記リポソームの脂質膜を透過して外水相から内水相に移動し、カチオン化して内水相に保持される薬剤を非イオン化する、請求項1~3のいずれかに記載の方法。
- 前記溶液が緩衝液である請求項1~4のいずれかに記載の方法。
- 次のステップを含む、リポソーム製剤の薬物放出性評価方法:
(1)シフト試薬を添加した溶液を調製するステップ;
(2)薬物を内封したリポソームを該溶液と混合するステップ;
(3)該薬物の該溶液中への放出を開始するステップ;
(4)該リポソームを該溶液から分離するステップ;および
(5)該溶液中に放出された薬物量を定量するステップ。 - 前記ステップ(3)において、前記リポソームを含む溶液を所定時間、所定温度で加温する、請求項6に記載の方法。
- 次のステップを(3)と(4)の間にさらに含む、請求項6または7に記載の方法。
