Abstract
NFATシグナル阻害剤等の提供。 一般式(式(I)中、R1及びR2は、各々同じであっても異なっていてもよく、水素原子又は式(II)で示される基(一般式(II)中、R3は炭素数1~20の直鎖又は分枝アルキル基又はアルケニル基を示す)である)で示されるクマリン誘導体及び薬理学的に許容されるそれらの塩からなる群より選択される少なくとも1種の化合物を有効成分とするNFATシグナル阻害剤等。
Description
本発明は、NFAT(nuclear factor of activated T cells)シグナル阻害剤及び育毛剤に関する。
Nuclear factor of activated T cells(以下、NFATと略称する)は、T細胞活性化に重要なInterleukin-2(IL-2)の転写を活性化する因子として発見され、免疫抑制剤であるサイクロスポリンA(以下、CsAと略称する)やタクロリムス(以下、FK506と略称する)の標的であるセリン/スレオニン脱リン酸化酵素カルシニューリンによりその転写活性調節が行われていることが報告されている(図5参照)。すなわちCsAやFK506はNFATシグナルを阻害することによりT細胞活性化を抑制する。CsAやFK506は移植免疫抑制剤としてのみならず、免疫系の関与することが知られている関節リウマチ、乾癬、アトピー性皮膚炎の治療薬としても認可されている。このようなNFATがNFAT結合配列(図2においては“NFAT site”)に結合することによって、当該NFAT結合配列より下流の遺伝子の転写が促進される系を『NFATシグナル』と称する。
一方、NFATシグナルを阻害することによって育毛(発毛を含む)効果を期待できると報告されている(非特許文献1)。また、NFATシグナルを阻害するCsAの誘導体が育毛剤として使用可能であることも報告されている(特許文献1及び2)。
ここで、従来、育毛剤として、男性型脱毛症、円形脱毛症などの脱毛症の治療には、経験的に、血行促進剤、免疫抑制剤、代謝促進剤、ビタミン剤、抗男性ホルモン剤等の薬剤が用いられている。しかしながら、これらの薬剤では、症状や体質によっては効果が異なる場合が多く、その効果も未だ満足出来るものではない。また、多量に使用すると適応部位に不快な刺激臭感覚を与えたり、継続使用により皮膚炎が発生するといった場合がある。男性型脱毛症、円形脱毛症などの脱毛症の多くは、未だその発症機序の詳細が不明であり、適した育毛剤を探索しているのが現状である。
ここで、従来、育毛剤として、男性型脱毛症、円形脱毛症などの脱毛症の治療には、経験的に、血行促進剤、免疫抑制剤、代謝促進剤、ビタミン剤、抗男性ホルモン剤等の薬剤が用いられている。しかしながら、これらの薬剤では、症状や体質によっては効果が異なる場合が多く、その効果も未だ満足出来るものではない。また、多量に使用すると適応部位に不快な刺激臭感覚を与えたり、継続使用により皮膚炎が発生するといった場合がある。男性型脱毛症、円形脱毛症などの脱毛症の多くは、未だその発症機序の詳細が不明であり、適した育毛剤を探索しているのが現状である。
また、NFATは、育毛作用や免疫系への作用に留まらず、心筋・骨格筋の分化調節による筋組織形成、脳における神経ネットワークの形成、骨芽細胞分化調節による骨代謝など、多くの臓器で発現し、重要な役割を果たす"多機能転写因子"として捉えられるようになってきた。
NFATシグナルが生体において及ぼす役割としては上述したとおりであるが、NFATシグナルを阻害することで、免疫抑制作用(非特許文献2)、乾癬治療(非特許文献3)、アトピー性皮膚炎治療(非特許文献4)、(心)筋肥大抑制(非特許文献5)、抗リウマチ薬としての可能性(非特許文献6)及び破骨細胞分化抑制作用(非特許文献7)などが期待できると報告されている。
また、前述のようにNFATは免疫性サイトカインIL-2を産生する経路を介していることから、NFATシグナル阻害剤は自己免疫疾患を含む免疫性サイトカインが関与していると考えられる疾患の治療または予防に有用である。
かかる対象疾患としては、例えば各種のガン、各種白血病、各種肝炎、各種感染症、全身性エリテマトーデス、炎症性腸疾患(潰瘍性大腸炎、クローン病)、多発性硬化症、インスリン依存性糖尿病、消化性潰瘍、敗血症ショック、結核、不妊症、動脈硬化、ベーチェット病、喘息、腎炎、急性バクテリア髄膜炎、急性心筋梗塞、急性膵炎、急性ウイルス脳炎、成人呼吸促迫症候群、バクテリア肺炎、慢性膵炎、末梢血管疾患、敗血症、間質性肝疾患、時局性回腸炎、多発性硬化症などが挙げられる。
また、前述のようにNFATは免疫性サイトカインIL-2を産生する経路を介していることから、NFATシグナル阻害剤は自己免疫疾患を含む免疫性サイトカインが関与していると考えられる疾患の治療または予防に有用である。

したがって、新規にNFATシグナル阻害剤が同定されれば、免疫抑制剤、乾癬治療剤、アトピー性皮膚炎治療剤、(心)筋肥大抑制剤、抗リウマチ薬、骨代謝疾患治療剤及び、上記に列記したような新規な医薬用途が期待される。また、新規にNFATシグナル阻害剤が同定されれば、育毛剤、発毛促進剤としての医薬部外品や化粧品用途が期待される。
ところで、アンジェリカ属に属する植物については、その抽出物に育毛、発毛或いは養毛効果が認められるという報告もある(特許文献3~6)。具体的に特許文献3には、カラトウキ(Angelica sinensis)抽出物を含む養毛・育毛剤が開示されている。特許文献4には、Angelica glaucaの精油を含有する養毛剤が開示されている。特許文献5には、ビャクシ(Angelica dahurica Benth. Et Hook.)の抽出物を含有する発毛・育毛促進料が開示されている。特許文献6にはアシタバ(Angelica Keiskei Koidz)の抽出物を含む育毛養毛等美髪料が開示されている。
また、アンジェリカ植物に含まれる成分については、非特許文献8、9を参照することができ、アメリカンアンジェリカの抽出物にインペラトリン、キサントトキシン等のリニア型フラノクマリンや、アンジェリシンなどのアンギュラー型フラノクマリンが含まれていることが示されている。また非特許文献10にはインペラトリン等のリニア型フラノクマリンがNFATシグナルの阻害作用を有するとの記載があるが、一方、本発明に係るアンギュラー型フラノクマリンについては、これまでにNFATシグナルとの関連や育毛、発毛或いは養毛効果との関連については知られていない。
なお、現在のところ、上記以外にNFATシグナル阻害活性を有する化合物としては、種々の食用植物に含まれるアピゲニン(非特許文献11)及びロスマリン酸(非特許文献12)が知られており、またNFATシグナル亢進活性を有する化合物としてゲニステイン(非特許文献13)が知られている。
なお、現在のところ、上記以外にNFATシグナル阻害活性を有する化合物としては、種々の食用植物に含まれるアピゲニン(非特許文献11)及びロスマリン酸(非特許文献12)が知られており、またNFATシグナル亢進活性を有する化合物としてゲニステイン(非特許文献13)が知られている。

本発明は、以下の1)~9)に係るものである。

2)上記式(I)中R1が、アンゲロイル基、イソバレロイル基及びセネシオイル基からなる群から選ばれる少なくとも1種の官能基であり、R2はアンゲロイル基、イソバレロイル基、2メチルブチル基及びセネシオイル基からなる群から選ばれる少なくとも1種の官能基である上記記載のNFATシグナル阻害剤。
3)上記一般式(1)で表されるクマリン誘導体が、アルカンジェリシン、3’-angeloyloxy-4’-isovaleryloxy-2’,3’-dihydrooroselol、3’-angeloyloxy-4’-senecioyloxy-2’,3’-dihydrooroselol及び3’-hydroxy-4’-angeloyloxy-2’,3’-dihydrooroselolからなる群より選択される少なくとも1種の化合物である上記記載のNFATシグナル阻害剤。
4)上記一般式(1)で表されるクマリン誘導体及び薬理学的に許容されるそれらの塩からなる群より選択される少なくとも1種の化合物を有効成分とする育毛剤。
5)次の一般式(III)で示される化合物。
6)NFATシグナル阻害剤を製造するための、上記一般式(1)で表されるクマリン誘導体及び薬理学的に許容されるそれらの塩からなる群より選択される少なくとも1種の化合物の使用。
7)育毛剤を製造するための、上記一般式(1)で表されるクマリン誘導体及び薬理学的に許容されるそれらの塩からなる群より選択される少なくとも1種の化合物の使用。
8)上記一般式(1)で表されるクマリン誘導体及び薬理学的に許容されるそれらの塩からなる群より選択される少なくとも1種の化合物を投与するNFATシグナル阻害方法。
9)上記一般式(1)で表されるクマリン誘導体及び薬理学的に許容されるそれらの塩からなる群より選択される少なくとも1種の化合物を投与する育毛方法。
本発明は、NFATシグナル阻害作用、又は育毛作用を有する、医薬品、外用剤、化粧品等を提供することに関する。
本願発明者は、NFATシグナル阻害作用又は育毛作用を有する物質について鋭意研究を重ねた結果、上記式(I)で表される化合物、特にアメリカンアンジェリカに含まれていた既知化合物及び上記式(III)の新規化合物に優れたNFATシグナル阻害作用及び育毛作用があることから、上記式(I)で表される化合物がNFATによる転写促進活性の亢進に起因する疾患の予防、改善或いは治療、又は育毛・養毛、発毛促進等の効果を発揮する、医薬品、外用剤、化粧品等として有用であることを見出した。
本発明のNFATシグナル阻害剤及び育毛剤(以下、「NFATシグナル阻害剤等」とも云う)によれば、NFATによる転写促進活性の亢進に起因する症状や疾患の予防、改善或いは治療又は育毛・養毛が可能となる。
本発明に係わるNFATシグナル阻害剤等に使用する活性成分は、アメリカンアンジェリカ植物又は当該植物の抽出物などから得ることができる。ここで、アメリカンアンジェリカとは、学名をAngelica atropurpureaと称し、セリ科に分類される植物である。
ここで、「活性成分」とは、次の一般式(I)で示されるクマリン誘導体若しくは薬理学的に許容されるその塩である。
一般式(I)に示すように、上記活性成分は、クマリン誘導体の中でもアンギュラー型クマリン誘導体である。後記実施例に示すように、アメリカンアンジェリカを分画し、活性成分を単離する工程において、インペラトリンやキサントトキシン等のリニア型フラノクマリン誘導体を含む画分にはNFATシグナル阻害活性が全く或いは殆ど認められなかったのに対し、本発明に係るアンギュラー型フラノクマリン誘導体を含む画分、更に単離した成分A、成分B及び成分Cには強いNFATシグナル阻害活性が認められたことから、本発明に係るアンギュラー型フラノクマリン誘導体は、既知のNFATシグナル阻害剤であるインペラトリン等のリニア型フラノクマリン誘導体に比し、優れたNFATシグナル阻害活性作用を有するものとして見出されたものである。
ここで、「活性成分」とは、次の一般式(I)で示されるクマリン誘導体若しくは薬理学的に許容されるその塩である。

上記一般式(I)中、R1及びR2は、各々同じであっても異なっていてもよく、水素原子又は一般式(II):
で示される基(上記一般式(II)中、R3は炭素数1~20の直鎖又は分枝のアルキル基又はアルケニル基を示す)である。
すなわち、上記活性成分として使用できるクマリン誘導体は、上述のように規定したR1、R2及びR3の範囲で種々の化合物を含むことになる。
特に、上記R1としては、アンゲロイル基、イソバレロイル基及びセネシオイル基からなる群から選ばれる官能基であることが好ましい。また、上記R2はアンゲロイル基、イソバレロイル基、2メチルブチル基及びセネシオイル基からなる群から選ばれる官能基であることが好ましい。
さらに、上記R2がアンゲロイル基、イソバレロイル基、2メチルブチル基又はセネシオイル基であるクマリン誘導体を、脱アシル化することによってクマリン骨格の3’位をヒドロキシル基(上記R2が水素原子の場合に相当)とすることができる。
特に、上記R1としては、アンゲロイル基、イソバレロイル基及びセネシオイル基からなる群から選ばれる官能基であることが好ましい。また、上記R2はアンゲロイル基、イソバレロイル基、2メチルブチル基及びセネシオイル基からなる群から選ばれる官能基であることが好ましい。
さらに、上記R2がアンゲロイル基、イソバレロイル基、2メチルブチル基又はセネシオイル基であるクマリン誘導体を、脱アシル化することによってクマリン骨格の3’位をヒドロキシル基(上記R2が水素原子の場合に相当)とすることができる。
なかでも、上記活性成分としては、R1がアンゲロイル基であり、R2がアンゲロイル基であるクマリン誘導体;R1がイソバレロイル基であり、R2がアンゲロイル基であるクマリン誘導体、及びR1がセネシオイル基であり、R2がアンゲロイル基であるクマリン誘導体が好ましい。また、上記活性成分としては、これらクマリン誘導体を脱アシル化して得られるアンギュラー型フロクマリン誘導体も好ましい。
特に、R1がアンゲロイル基であり、R2がアンゲロイル基であるクマリン誘導体は、アルカンジェリシンとして抗腫瘍、抗炎症作用が知られた化合物であり、また、R1がイソバレロイル基であり、R2がアンゲロイル基であるクマリン誘導体、例えば3’-angeloyloxy-4’-isovaleryloxy-2’,3’-dihydrooroselolは、公知化合物であるが、これらは、後記実施例に示すように、本発明において、育毛活性及びNFATシグナル阻害活性を有することが明らかになったものである。
さらに、R1がセネシオイル基であり、R2がアンゲロイル基であるクマリン誘導体、例えば3’-angeloyloxy-4’-senecioyloxy-2’,3’-dihydrooroselolは、後記実施例に示すように、本発明において、NFATシグナル阻害活性を有する新規化合物として見出されたものである。
さらに、R1がセネシオイル基であり、R2がアンゲロイル基であるクマリン誘導体、例えば3’-angeloyloxy-4’-senecioyloxy-2’,3’-dihydrooroselolは、後記実施例に示すように、本発明において、NFATシグナル阻害活性を有する新規化合物として見出されたものである。
また、上記化学式(I)で示される公知化合物としては、例えば、アサマンチン(Athamantin)、シニフォリンB(Cniforin B)、エデュリシンII(Edulisin II)及びエデュリシンV(Edulisin V)等が挙げられる。
なお、上記活性成分として、これら公知化合物及び新規化合物から選ばれる1種又は2種以上のものを使用してもよい。

ここで、本発明のクマリン誘導体には、上述のように、公知の化学合成法により得られるものや、これを活性の主体成分として含有する植物抽出物若しくはその精製物や単離物も包含される。
なお、必要に応じて、上記植物等抽出物を、活性炭処理、液体クロマトグラフィー、液々分配、ゲルろ過、精密蒸留等の分離精製技術に供し、当該抽出物から、不活性な夾雑物等を除去し、目的の化合物を精製及び単離してもよい。
一例として、活性成分であるクマリン誘導体、特に上述したアルカンジェリシン、3’-angeloyloxy-4’-isovaleryloxy-2’,3’-dihydrooroselol及び3’-angeloyloxy-4’-senecioyloxy-2’,3’-dihydrooroselolについては、セリ科植物(好ましくは、アメリカンアンジェリカ植物)又はその抽出物から、上記クマリン誘導体、特にこれら化合物から選ばれる1種又は2種以上のものを活性の主体成分として含有する画分(以下、「活性画分」とも云う)を得ること又はこれらから目的の化合物を単離することができる。また、これらの中間体を植物から抽出し、次いで化学合成により目的物を得ても良い。
ここで、上記植物の全草、葉、茎、花、果実、種子、根茎又は根等を、そのまま、破砕、粉砕、搾取して用いるか、又はこれら処理されたものを乾燥若しくは粉末化して用いることができる。用いる部位としては、根を、使用するのが好ましい。
上記植物の抽出物とは、当該植物を一定温度(低温、常温又は加温)下にて抽出すること;又はソックスレー抽出器等の抽出器具を用いて抽出すること等の抽出手段により得られる各種溶剤抽出液、その希釈液、その濃縮液又はその粉末を意味するものである。抽出手段は、具体的には、固液抽出、液液抽出、浸漬、煎出、浸出、還流抽出、超音波抽出、マイクロ波抽出、攪拌等の手段を用いることができる。
上記植物抽出物を得るために用いられる抽出溶剤としては、極性溶剤又は非極性溶剤のいずれをも使用することができる。抽出溶剤としては、例えば水;メタノール、エタノール、プロパノール、ブタノール等のアルコール類;プロピレングリコール、ブチレングリコール等の多価アルコール類;アセトン、メチルエチルケトン等のケトン類;酢酸メチル、酢酸エチル等のエステル類;テトラヒドロフラン、ジエチルエーテル等の鎖状及び環状エーテル類;ポリエチレングリコール等のポリエーテル類;スクワラン、ヘキサン、シクロヘキサン、石油エーテル等の炭化水素類;トルエン等の芳香族炭化水素類;ジクロロメタン、クロロホルム、ジクロロエタン等のハロゲン化炭化水素類;及び二酸化炭素等が挙げられる。或いは、上記溶剤の2種以上を組み合わせた混合物を、抽出溶剤として用いることができる。
好ましくは、アルコール類(好ましくは、炭素数1~4のアルコール類)、アルコール類・水混合液であり、このときのアルコール類の濃度は70~100容量%(好ましくは80~100容量%)であるのが好ましい。

抽出溶剤の使用量は、植物1質量部(乾燥物換算)に対して、1~50質量部であるのが好ましく、5~40質量部であるのが好ましい。抽出温度は、0~100℃であるのが好ましく、より4~80℃、更に10~40℃であるのが好ましい。抽出時間は、1分~50日間であるのが好ましく、より1時間~50日間、更に1~10日間であるのが好ましい。
更に、上記植物又はその抽出物から、液体クロマトグラフィーや液々分配等の分離技術に供し、活性成分である上述したアルカンジェリシン、3’-angeloyloxy-4’-isovaleryloxy-2’,3’-dihydrooroselol及び3’-angeloyloxy-4’-senecioyloxy-2’,3’-dihydrooroselolを含有する活性画分を得てもよいし、これらを単離してもよい。
具体的には、当該抽出物を、オクタデシル化シリカゲル、オクチル化シリカゲル、トリメチルシリル化シリカゲル等の逆相樹脂カラムクロマトに付し、アセトニトリル-0.05~0.2%TFA系溶媒で溶出し、活性成分を含有する活性画分を得てもよい。
また、アルカンジェリシンを脱アシル化することによって3’-hydroxy-4’-angeloyloxy-2’,3’-dihydrooroselolが得られる。
具体的には、当該抽出物を、オクタデシル化シリカゲル、オクチル化シリカゲル、トリメチルシリル化シリカゲル等の逆相樹脂カラムクロマトに付し、アセトニトリル-0.05~0.2%TFA系溶媒で溶出し、活性成分を含有する活性画分を得てもよい。
また、アルカンジェリシンを脱アシル化することによって3’-hydroxy-4’-angeloyloxy-2’,3’-dihydrooroselolが得られる。
ただし、本発明においては、上記化学式(I)で示される化合物であれば、従来公知の化合物を活性成分として使用することができる。上記化学式(I)で示される公知化合物としては、例えば、アサマンチン(Athamantin)、シニフォリンB(Cniforin B)、エデュリシンII(Edulisin II)及びエデュリシンV(Edulisin V)を挙げることができ、活性成分として使用することができる。
また、本発明のクマリン誘導体は、塩にすることにより水溶性を向上させ、生理学的有効性を増大させることができる。これらの塩としては、薬学的に許容される塩であればよい。このような塩形成用の塩基物質としては、例えば、水酸化リチウム、水酸化ナトリウム、水酸化カリウム等のアルカリ金属の水酸化物;水酸化マグネシウム、水酸化カルシウム等のアルカリ土類金属の水酸化物;水酸化アンモニウム等の無機塩基、アルギニン、リジン、ヒスチジン、オルニチン等の塩基性アミノ酸;モノエタノールアミン、ジエタノールアミン、トリエタノールアミン等の有機塩基が用いられるが、特にアルカリ金属又はアルカリ土類金属の水酸化物が好ましい。本発明においては、これらの塩を調製してから、その他の成分からなる組成物中に添加したものでもよいし、クロロゲン酸類等と塩形成成分とを別々に該組成物中に添加して、この中で塩を形成せしめたものでもよい。
本発明のクマリン誘導体又はその塩は、後記実施例に示すとおり、NFATシグナル阻害作用、及び育毛作用を有する。
ここで、NFATシグナル阻害作用とは、NFATに対して直接的又は間接的に作用してNFATによる転写促進活性を低減させることを意味する。
また、上述したNFATシグナル阻害作用(活性)は、NFATシグナル阻害率を指標として評価することができる。

このNFATシグナル阻害作用(活性)を測定する方法としては、特に限定されないが、例えば公知のNFAT結合配列と当該NFAT結合配列の下流にレポーター遺伝子と有するプラスミドを導入した宿主を用いるレポーターアッセイを挙げることができる。
因みに、NFATの活性はカルシウムイオンに依存するため、レポーターアッセイはカルシウムイオンを宿主に流入させる条件下で行う。レポーター遺伝子としては、特に限定されず、従来、生化学実験の分野で使用されている如何なるレポーター遺伝子も使用することができる。例えばレポーター遺伝子としては、ルシフェラーゼ遺伝子、βグルクロニダーゼ遺伝子(GUS遺伝子)及びグリーンフルオレセントプロテイン遺伝子(GFP遺伝子)等を挙げることができる。
因みに、NFATの活性はカルシウムイオンに依存するため、レポーターアッセイはカルシウムイオンを宿主に流入させる条件下で行う。レポーター遺伝子としては、特に限定されず、従来、生化学実験の分野で使用されている如何なるレポーター遺伝子も使用することができる。例えばレポーター遺伝子としては、ルシフェラーゼ遺伝子、βグルクロニダーゼ遺伝子(GUS遺伝子)及びグリーンフルオレセントプロテイン遺伝子(GFP遺伝子)等を挙げることができる。
NFAT結合配列とは、NFATが結合する配列からなるオリゴヌクレオチド、例えば、配列番号1に示すGGAGGAAAAACTGTTTCATACAGAAGGCGT(pNFAT-Luc、Stratagene)といった塩基配列を例示することができる。なお、上述したプラスミドにおいては、このようなNFAT結合配列を1セットとして複数セットを連結して導入してもよい。複数のNFAT結合配列を連結して使用することによって、NFATによる転写促進活性をより高感度に測定することができる。なお、NFATシグナル阻害活性は、上述したレポーターアッセイから算出したNFATシグナル阻害率として評価することができる。
また、育毛作用(活性)とは、毛包に作用し、毛の伸長の促進、太さの増大、毛周期の休止期から成長期への移行促進、成長期から退行期への移行阻害などを行なうことにより毛の量を増加させることを意味する。従って、育毛には、発毛、養毛及び脱毛予防の概念が包含されるものとする。
育毛作用(活性)は、毛伸長促進率を指標として評価することができる。ここで、毛伸長促進率とは、要するに、クマリン誘導体を作用させないときの毛伸長率に対して、クマリン誘導体を作用させたときの当該毛伸長率の促進率を意味している。
この育毛作用(活性)を測定する方法としては、特に限定されないが、例えば単離毛包を器官培養し、培養期間中の毛の伸長量を測定する方法が挙げられる。
育毛作用(活性)は、毛伸長促進率を指標として評価することができる。ここで、毛伸長促進率とは、要するに、クマリン誘導体を作用させないときの毛伸長率に対して、クマリン誘導体を作用させたときの当該毛伸長率の促進率を意味している。
この育毛作用(活性)を測定する方法としては、特に限定されないが、例えば単離毛包を器官培養し、培養期間中の毛の伸長量を測定する方法が挙げられる。
上述した活性成分は、NFATによる転写促進(正に制御される転写)活性を低減させることから、細胞や組織におけるNFATシグナルを阻害することができる。具体的には、本発明のクマリン誘導体又はその塩を、対象とする細胞や組織に接触させることによって、当該細胞や組織におけるNFATシグナルを阻害することができる。これにより、当該細胞や組織においては、NFATシグナルによって生ずる種々の遺伝子発現を転写レベルで抑制することができる。
また、上述した活性成分は、育毛・発毛を促進し、種々の脱毛症に有効である。

従って、本発明のクマリン誘導体又はその塩は、NFATによる転写促進活性を低減させること;育毛・発毛を促進させることから、NFATシグナル阻害剤や育毛・発毛促進剤等として使用することができ、これらの製剤を製造するために使用することができる。
本発明に係るNFATシグナル阻害剤等は、NFATによる転写促進活性の亢進に起因する症状や疾患に対する治療剤、改善剤又は予防剤として使用することができる。
NFATによる転写促進活性の亢進に起因する症状や疾患としては、免疫系疾患、乾癬、アトピー性皮膚炎、心筋を含む筋肥大症、リウマチ、骨代謝疾患、各種のガン、各種白血病、各種肝炎、各種感染症、全身性エリテマトーデス、炎症性腸疾患(潰瘍性大腸炎、クローン病)、多発性硬化症、インスリン依存性糖尿病、消化性潰瘍、敗血症ショック、結核、不妊症、動脈硬化、ベーチェット病、喘息、腎炎、急性バクテリア髄膜炎、急性心筋梗塞、急性膵炎、急性ウイルス脳炎、成人呼吸促迫症候群、バクテリア肺炎、慢性膵炎、末梢血管疾患、敗血症、間質性肝疾患、時局性回腸炎、多発性硬化症等を挙げることができる。したがって、本発明に係るNFATシグナル阻害剤等は、免疫抑制剤、乾癬治療剤、(心)筋肥大抑制剤、抗リウマチ薬及び骨代謝疾患治療剤等として使用することができる。
NFATによる転写促進活性の亢進に起因する症状や疾患としては、免疫系疾患、乾癬、アトピー性皮膚炎、心筋を含む筋肥大症、リウマチ、骨代謝疾患、各種のガン、各種白血病、各種肝炎、各種感染症、全身性エリテマトーデス、炎症性腸疾患(潰瘍性大腸炎、クローン病)、多発性硬化症、インスリン依存性糖尿病、消化性潰瘍、敗血症ショック、結核、不妊症、動脈硬化、ベーチェット病、喘息、腎炎、急性バクテリア髄膜炎、急性心筋梗塞、急性膵炎、急性ウイルス脳炎、成人呼吸促迫症候群、バクテリア肺炎、慢性膵炎、末梢血管疾患、敗血症、間質性肝疾患、時局性回腸炎、多発性硬化症等を挙げることができる。したがって、本発明に係るNFATシグナル阻害剤等は、免疫抑制剤、乾癬治療剤、(心)筋肥大抑制剤、抗リウマチ薬及び骨代謝疾患治療剤等として使用することができる。
斯様に、上述した活性成分は、NFATシグナル阻害作用や育毛作用等を発揮する医薬品、外用剤、化粧品、食品、飼料等として使用できる。
医薬品として使用する場合、その剤形は、特に限定されないが、例えば散剤、顆粒剤、カプセル剤、丸剤、錠剤等の固形製剤、水剤、懸濁剤、乳剤等の液剤、軟膏剤等が挙げられる。
経口投与の医薬品として使用する場合、上述した活性成分を単独で又は、この他、経口投与剤の形態に応じて一般に用いられる、賦形剤、崩壊剤、結合剤、滑沢剤、界面活性剤、アルコール類、水、水溶性高分子、甘味料、矯味剤、酸味料等を添加し、常法に従って製造することができる。経口投与用医薬品としては、免疫抑制剤、乾癬治療剤、(心)筋肥大抑制剤及び抗リウマチ薬等が挙げられる。
経皮投与の医薬品として使用する場合、上述した活性成分を単独で又は、この他、経皮投与剤の形態に応じて一般に用いられる、植物油、動物油、合成油、脂肪酸、及び天然、合成のグリセライド等の油性基剤、滑沢剤、界面活性剤、アルコール類、増粘剤等を添加し、常法に従って製造することができる。経皮投与用医薬品としては、免疫抑制剤、乾癬治療剤、アトピー性皮膚炎治療剤、発毛促進剤等が挙げられる。
医薬品として使用する場合、その剤形は、特に限定されないが、例えば散剤、顆粒剤、カプセル剤、丸剤、錠剤等の固形製剤、水剤、懸濁剤、乳剤等の液剤、軟膏剤等が挙げられる。
経口投与の医薬品として使用する場合、上述した活性成分を単独で又は、この他、経口投与剤の形態に応じて一般に用いられる、賦形剤、崩壊剤、結合剤、滑沢剤、界面活性剤、アルコール類、水、水溶性高分子、甘味料、矯味剤、酸味料等を添加し、常法に従って製造することができる。経口投与用医薬品としては、免疫抑制剤、乾癬治療剤、(心)筋肥大抑制剤及び抗リウマチ薬等が挙げられる。
経皮投与の医薬品として使用する場合、上述した活性成分を単独で又は、この他、経皮投与剤の形態に応じて一般に用いられる、植物油、動物油、合成油、脂肪酸、及び天然、合成のグリセライド等の油性基剤、滑沢剤、界面活性剤、アルコール類、増粘剤等を添加し、常法に従って製造することができる。経皮投与用医薬品としては、免疫抑制剤、乾癬治療剤、アトピー性皮膚炎治療剤、発毛促進剤等が挙げられる。
また、外用剤及び育毛剤は、例えば皮膚用の医薬部外品として使用するものであり、使用形態に適した剤形として提供される。具体的に剤形としては、特に限定されるものではないが、例えば、軟膏、水剤、エキス剤、ローション剤、トニック剤、スプレー剤及び乳剤等が挙げられる。
当該医薬部外品には、上述した活性成分を単独で又は、この他に、助剤、安定化剤、湿潤剤、乳化剤、吸収促進剤及び界面活性剤等の薬学的に許容される担体を任意に組合せて配合することができる。また、育毛剤においては、上述した有効成分の他に、通常用いられる養毛薬効剤、例えば、毛包賦活剤、血行促進剤、抗菌剤、抗炎症剤、保湿剤、抗脂漏剤、局所刺激剤、抗男性ホルモン剤、カリウムチャンネルオープナー、抗酸化剤等を必要に応じて適宜配合し、育毛効果の向上を図ることができる。毛包賦活剤としては、フラバノノール類、N-アセチル-L-メチオニン、パントテン酸及びその誘導体、アデノシン及びその誘導体、アスパラギン酸カリウム、ペンタデカン酸グリセリド、6-ベンジアルアミノプリン、モノニトログアヤコールナトリウム等が挙げられる。血行促進剤としては、二酸化炭素、ニコチン酸アミド、ニコチン酸ベンジル、センブリエキス、ニンジンエキス、塩化カルプロニウム、ビタミンE及びその誘導体等が挙げられる。抗菌剤としては、イソプロピルメチルフェノール、塩化ベンザルコニウム、オクトピロックス、ジンクピリチオン、ヒノキチオール等が挙げられる。抗炎症剤としては、甘草エキス、グリチルリチン酸及びその誘導体、グリチルレチン酸及びその誘導体、アズレン、グアイアズレン、オウゴンエキス、カミツレエキス、クマザサエキス、シラカバエキス、ゼニアオイエキス、桃葉エキス、セイヨウノコギリソウエキス等が挙げられる。保湿剤としては、オトギリソウエキス、オーツ麦エキス、グリセリン、チューベロースポリサッカライド、冬虫夏草エキス、延命草エキス、オオムギエキス、ブドウエキス、プロピレングリコール、桔梗エキス、ヨクイニンエキス等が挙げられる。抗脂漏剤としては、イオウ、レシチン、カシュウエキス、チオキソロン等が挙げられる局所刺激剤としては、カンファー、トウガラシチンキ等が挙げられる。抗男性ホルモン剤としては、サイプロテロンアセテート、1 1α-ハイドロキシプロゲステロン、フルタマイド、3-デオキシアデノシン、酢酸クロルマジノン、エチニルエストラジオール、スピロノラクトン、エピテステロン、フィナステライド、アロエ、サンショウ、チョウジエキス、クアチャララーテエキス、オタネニンジン等が挙げられる。カリウムチャンネルオープナーとしては、ミノキシジル、クロマカリム、ジアゾキシド及びその誘導体、ピナシジル等が挙げられる。抗酸化剤としては、紅茶エキス、茶エキス、エイジツエキス、黄杞エキス、ビタミンC及びその誘導体、エリソルビン酸、没食子酸プロピル、ジブチルヒドロキシトルエン等が挙げられる。
当該医薬部外品には、上述した活性成分を単独で又は、この他に、助剤、安定化剤、湿潤剤、乳化剤、吸収促進剤及び界面活性剤等の薬学的に許容される担体を任意に組合せて配合することができる。また、育毛剤においては、上述した有効成分の他に、通常用いられる養毛薬効剤、例えば、毛包賦活剤、血行促進剤、抗菌剤、抗炎症剤、保湿剤、抗脂漏剤、局所刺激剤、抗男性ホルモン剤、カリウムチャンネルオープナー、抗酸化剤等を必要に応じて適宜配合し、育毛効果の向上を図ることができる。毛包賦活剤としては、フラバノノール類、N-アセチル-L-メチオニン、パントテン酸及びその誘導体、アデノシン及びその誘導体、アスパラギン酸カリウム、ペンタデカン酸グリセリド、6-ベンジアルアミノプリン、モノニトログアヤコールナトリウム等が挙げられる。血行促進剤としては、二酸化炭素、ニコチン酸アミド、ニコチン酸ベンジル、センブリエキス、ニンジンエキス、塩化カルプロニウム、ビタミンE及びその誘導体等が挙げられる。抗菌剤としては、イソプロピルメチルフェノール、塩化ベンザルコニウム、オクトピロックス、ジンクピリチオン、ヒノキチオール等が挙げられる。抗炎症剤としては、甘草エキス、グリチルリチン酸及びその誘導体、グリチルレチン酸及びその誘導体、アズレン、グアイアズレン、オウゴンエキス、カミツレエキス、クマザサエキス、シラカバエキス、ゼニアオイエキス、桃葉エキス、セイヨウノコギリソウエキス等が挙げられる。保湿剤としては、オトギリソウエキス、オーツ麦エキス、グリセリン、チューベロースポリサッカライド、冬虫夏草エキス、延命草エキス、オオムギエキス、ブドウエキス、プロピレングリコール、桔梗エキス、ヨクイニンエキス等が挙げられる。抗脂漏剤としては、イオウ、レシチン、カシュウエキス、チオキソロン等が挙げられる局所刺激剤としては、カンファー、トウガラシチンキ等が挙げられる。抗男性ホルモン剤としては、サイプロテロンアセテート、1 1α-ハイドロキシプロゲステロン、フルタマイド、3-デオキシアデノシン、酢酸クロルマジノン、エチニルエストラジオール、スピロノラクトン、エピテステロン、フィナステライド、アロエ、サンショウ、チョウジエキス、クアチャララーテエキス、オタネニンジン等が挙げられる。カリウムチャンネルオープナーとしては、ミノキシジル、クロマカリム、ジアゾキシド及びその誘導体、ピナシジル等が挙げられる。抗酸化剤としては、紅茶エキス、茶エキス、エイジツエキス、黄杞エキス、ビタミンC及びその誘導体、エリソルビン酸、没食子酸プロピル、ジブチルヒドロキシトルエン等が挙げられる。
化粧料として使用する場合には、剤形としては、特に限定されるものではないが、例えば、油中水型又は水中油型の乳化化粧料、クリーム、ローション、ジェル、フォーム、エッセンス、ファンデーション、パック、スティック及びパウダー等が挙げられる。
当該化粧料には、上述した活性成分を単独で又は、この他に、化粧料成分として一般に使用されている油分、界面活性剤、紫外線吸収剤、アルコール類、キレート剤、pH調整剤、防腐剤、増粘剤、色素類、香料及び各種皮膚栄養剤等を任意に組合せて配合することができる。
具体的には、皮膚化粧料に配合される薬効成分、例えば微粒子酸化亜鉛、酸化チタン、パーソールMCX、パーソール1789等の紫外線吸収剤、アスコルビン酸等のビタミン類、ヒアルロン酸ナトリウム、ワセリン、グリセリン、尿素等の保湿剤、ホルモン剤、及びコウジ酸、アルブチン、プラセンタエキス、ルシノール等の他の美白成分、ステロイド剤、アラキドン酸代謝物やヒスタミン等に代表される化学伝達物質産生・放出抑制剤(インドメタシン、イブプロフェン)、レセプター拮抗剤等の抗炎症剤、抗男性ホルモン剤、ビタミンA酸、ローヤルゼリーエキス、ローヤルゼリー酸等の皮脂分泌抑制剤、ニコチン酸トコフェロール、アルプロスタジル、塩酸イソクスプリン、塩酸トラゾリン等の抹消血管拡張剤及び末梢血管拡張作用のある炭酸ガス等、ミノキシジル、塩化カルプロニウム、トウガラシチンキ、ビタミンE誘導体、イチョウエキス、センブリエキス等の血行促進剤、ペンタデカン酸グリセリド、ニコチン酸アミド等の細胞賦活化剤、ヒノキチオール、L-メントール、イソプロピルメチルフェノール等の殺菌剤、グリチルリチン酸およびその誘導体またはその塩等の薬剤、セラミド及びセラミド類似化合物等を添加配合することができる。
当該化粧料には、上述した活性成分を単独で又は、この他に、化粧料成分として一般に使用されている油分、界面活性剤、紫外線吸収剤、アルコール類、キレート剤、pH調整剤、防腐剤、増粘剤、色素類、香料及び各種皮膚栄養剤等を任意に組合せて配合することができる。

また、上述した医薬品、医薬部外品、化粧品においては、例えばチョーク、タルク、フラー土、カオリン、デンプン、ゴム、コロイドシリカナトリウムポリアクリレート等の粉体;例えば鉱油、植物油、シリコーン油等の油又は油状物質;例えばソルビタントリオレエート、ソルビタントリステアレート、グリセロールモノオレエート、高分子シリコーン界面活性剤等の乳化剤;パラ-ヒドロキシベンゾエートエステル等の防腐剤;ブチルヒドロキシトルエン等の酸化防止剤;グリセロール、ソルビトール、2-ピロリドン-5-カルボキシレート、ジブチルフタレート、ゼラチン、ポリエチレングリコール等の湿潤剤;トリエタノールアミン又は水酸化ナトリウムのような塩基を伴う乳酸等の緩衝剤;グリセリン脂肪酸エステル、ソルビタン脂肪酸エステル、ショ糖脂肪酸エステル、アルキルグルコシド等の界面活性剤;密ろう、オゾケライトワックス、パラフィンワックス等のワックス類;増粘剤;活性増強剤;着色料;香料等、を必要に応じ適宜組合せて用いることができる。
上述した活性成分を医薬、医薬部外品又は化粧料等として使用する場合、上述した活性成分の配合量は、クマリン誘導体換算で、通常、医薬、医薬部外品又は化粧料等の全組成の0.00001~5質量%、特に0.0001~0.1質量%とすることが好ましい。また、本発明に係るNFATシグナル阻害剤を医薬として用いる場合、上述した活性成分の投与量は、通常の成人(60kg)で0.01mg~1g/1日とすることが望ましい。
以下、実施例により本発明をさらに具体的に説明する。但し、本発明はこれら実施例にその技術的範囲が限定されるものではない。
〔実施例1〕アメリカンアンジェリカからの活性成分類の単離
(1)図1に示す工程に従って、アメリカンアンジェリカ(Angelica atropurpurea(セリ科)の根茎から本発明の化合物「成分A」、「成分B」及び「成分C」を単離した。
(2)アンジェリカ(Angelica atropurpurea)の根茎(160g)を95 w/v%エタノール-水(1.6L)に、常温(10~40℃)で5日間浸漬して抽出し、95 w/v%エタノール-水抽出液を得た。この抽出液をろ過後、ロータリーエバポレーターで濃縮し、固形分14.3gを得た。この濃縮乾固物1gを中圧ODSカラムクロマトグラフィ(オクタデシルシリル化シリカゲル:内径2.6×30cm)に付し、アセトニトリル-0.1% TFA系で溶出し以下の画分(fr. A~K)を得た。
(1)図1に示す工程に従って、アメリカンアンジェリカ(Angelica atropurpurea(セリ科)の根茎から本発明の化合物「成分A」、「成分B」及び「成分C」を単離した。

なお、各画分固形分を50mLの95w/v%エタノールに溶解し、それぞれODS-HPLC(内径2.1×150mm、アセトニトリル-0.1% TFA系溶離液)にて分析したところ、リニア型フラノクマリンのキサントトキシンはfr. Fおよびfr. Gに、インペラトリンはfr. Hに存在していることを確認した(図2)。各画分固形分を50mLの95w/v%エタノール溶液について、後述の方法にてNFATシグナル阻害活性を調べたところ、最も強い活性はfr. Iに認められ、一方でインペラトリンやキサントトキシンを含むfr. F、fr. G、fr. Hには活性が全く又は殆ど認められなかったことから(図3)、アメリカンアンジェリカの主たるNFATシグナル阻害活性成分は、インペラトリンやキサントトキシン以外の成分であることが分かり、この結果は前述の非特許文献10の記載からすると全く意外であった。

本発明の化合物は、分析の結果、fr. I-6(成分A)、fr. I-8(成分B)、fr. I-5(成分C)にほぼ単一化合物として得られた。
〔実施例2〕成分A、成分B及び成分Cの同定
(1)実施例1で得られた化合物について、1H NMR解析及び13C NMR解析を行った。
1H NMR(500 MHz, CDCl3)及び13C NMR(125 MHz, CDCl3)解析の結果として得られたデータを表1に示す。

(2)表1に示した解析結果より、成分A、成分B及び成分Cの化合物は下記構造を有するアンギュラー型フロクマリン誘導体であることが示された。
すなわち、成分A、成分B及び成分Cは、それぞれアルカンジェリシン、3’-angeloyloxy-4’-isovaleryloxy-2’,3’-dihydrooroselol及び3’-angeloyloxy-4’-senecioyloxy-2’,3’-dihydrooroselolであることが判った。ここで、成分Cの3’-angeloyloxy-4’-senecioyloxy-2’,3’-dihydrooroselolは過去に報告例がなく、新規化合物であった。
〔実施例3〕成分A(アルカンジェリシン)の脱アシル体の調製
本例では、実施例1及び2で成分Aとして単離したアルカンジェリシンの脱アシル化を行った。まず、成分A(70mg)を7mLのアセトンに溶解後、濃アンモニア水7mLを添加し、室温で一昼夜攪拌した。反応液を濃縮後(75mg)、分取高速液体クロマトグラフィ(内径10×250mm)に付し、0.1%ギ酸-アセトニトリル系で溶出し、主生成物を分取した(17mg)。
本例では、実施例1及び2で成分Aとして単離したアルカンジェリシンの脱アシル化を行った。まず、成分A(70mg)を7mLのアセトンに溶解後、濃アンモニア水7mLを添加し、室温で一昼夜攪拌した。反応液を濃縮後(75mg)、分取高速液体クロマトグラフィ(内径10×250mm)に付し、0.1%ギ酸-アセトニトリル系で溶出し、主生成物を分取した(17mg)。
〔実施例4〕成分A(アルカンジェリシン)の脱アシル体の同定
(1)実施例3で得られた化合物について、1H NMR解析及び13C NMR解析を行った。1H NMR(500MHz, CDCl3)及び13C NMR(125MHz, CDCl3)解析の結果として得られたデータを表2に示す。

(2)表2に示した解析分析より、実施例3における生成物はアルカンジェリシンの3’位のアンゲロイル基が脱離した、下記構造を有するアンギュラー型フロクマリン誘導体であることが示された。
〔実施例5〕NFATシグナル阻害効果
本例では、実施例1で得られた成分A、成分B及び成分C並びに実施例3で調製した成分Aの脱アシル体についてのNFATシグナル阻害効果を検証した。

(1)評価システムのための材料及び方法
細胞培養
上記評価システムには、ヒト腎(HEK293)細胞をATCC(American Type Culture Collection)より購入し、使用した。HEK293細胞は、DMEM(High glucose、10% heat-inactivated FBS)中37℃、5% CO2条件下で培養した。

プラスミド、トランスフェクション
上記評価システムには、NFAT結合配列の下流にホタルルシフェラーゼが導入されたプラスミドpNFAT-Luc(Stratagene)をHEK293細胞にトランスフェクションしたものを使用した。詳細には、NFAT転写活性の評価用に4連のNFAT結合配列の下流にホタルルシフェラーゼ遺伝子が導入されたpNFAT-luc(STRATAGENE)をHEK293細胞にトランスフェクションした。また、トランスフェクション効率によるばらつきをなくすことを目的として、ホタルルシフェラーゼに由来するシグナルを補正するためにCMV promoterの下流にウミシイタケルシフェラーゼが導入されたpRL-CMV(Promega)を同時にトランスフェクションした。
上記評価システムには、NFAT結合配列の下流にホタルルシフェラーゼが導入されたプラスミドpNFAT-Luc(Stratagene)をHEK293細胞にトランスフェクションしたものを使用した。詳細には、NFAT転写活性の評価用に4連のNFAT結合配列の下流にホタルルシフェラーゼ遺伝子が導入されたpNFAT-luc(STRATAGENE)をHEK293細胞にトランスフェクションした。また、トランスフェクション効率によるばらつきをなくすことを目的として、ホタルルシフェラーゼに由来するシグナルを補正するためにCMV promoterの下流にウミシイタケルシフェラーゼが導入されたpRL-CMV(Promega)を同時にトランスフェクションした。
トランスフェクションは、LipofectAMINE 2000 reagent(Invitrogen)を用いて使用説明書に従って行った。トランスフェクションの8時間後に培地を交換し、一晩インキュベートした。その後、実施例1で得られた成分A、成分B及び成分C並びに実施例3で調製した成分Aの脱アシル体を添加し、その1時間後に1μM Ionomycinを添加した。8時間後、ルシフェラーゼレポーターアッセイを行った。
ルシフェラーゼレポーターアッセイ
ルシフェラーゼレポーターアッセイは、Dual-Glo Luciferase Assay System(Promega)を用い、使用説明書に従って行った。すなわち、培地を除去後、PBSにより2倍希釈したDual-Glo luciferase reagentを加え、攪拌した後、20分後にホタルルシフェラーゼ活性を測定した。その後、等量のDual-Glo Stop&Glo reagentを加え、攪拌した後にウミシイタケルシフェラーゼ活性を測定した。ルシフェラーゼ活性測定はMiniLumat LB 9506(EG&G BERTHOLD)を用いて行い、ルシフェラーゼによる発光量を定量的に検出した。双方ともルシフェラーゼ活性の測定時間は2秒とした。
ルシフェラーゼレポーターアッセイは、Dual-Glo Luciferase Assay System(Promega)を用い、使用説明書に従って行った。すなわち、培地を除去後、PBSにより2倍希釈したDual-Glo luciferase reagentを加え、攪拌した後、20分後にホタルルシフェラーゼ活性を測定した。その後、等量のDual-Glo Stop&Glo reagentを加え、攪拌した後にウミシイタケルシフェラーゼ活性を測定した。ルシフェラーゼ活性測定はMiniLumat LB 9506(EG&G BERTHOLD)を用いて行い、ルシフェラーゼによる発光量を定量的に検出した。双方ともルシフェラーゼ活性の測定時間は2秒とした。
NFATシグナル阻害率の算出
全てのNFAT転写活性(ホタルルシフェラーゼ活性)はトランスフェクション効率補正のために導入されたウミシイタケルシフェラーゼ活性にて除することで補正した。その後、NFATシグナル阻害率を以下の式にて算出した。
下記計算により、Ionomycin刺激によるNFATシグナル活性化をテストサンプルが何%阻害したかについて算出することができる。
NFATシグナル阻害率(%)=100-(試験サンプル及びIonomycin添加群-無刺激群)/(Ionomycinのみ添加群-無刺激群)×100
全てのNFAT転写活性(ホタルルシフェラーゼ活性)はトランスフェクション効率補正のために導入されたウミシイタケルシフェラーゼ活性にて除することで補正した。その後、NFATシグナル阻害率を以下の式にて算出した。
下記計算により、Ionomycin刺激によるNFATシグナル活性化をテストサンプルが何%阻害したかについて算出することができる。
NFATシグナル阻害率(%)=100-(試験サンプル及びIonomycin添加群-無刺激群)/(Ionomycinのみ添加群-無刺激群)×100
結果
NFATシグナル阻害率を算出した結果を図4に示す。図4から判るように、実施例1で得られた成分A、成分B及び成分C並びに実施例3で調製した成分Aの脱アシル体は、いずれもNFATシグナルを阻害した。すなわち、成分A、成分B及び成分Aの脱アシル体は、NFATにより正に制御される転写を抑制することができる。したがって、成分A、成分B及び成分Aの脱アシル体は、優れたNFATシグナル阻害剤であり、例えば免疫抑制剤、乾癬治療剤、アトピー性皮膚炎治療剤、(心)筋肥大抑制剤及び抗リウマチ薬等の候補物質として同定されたこととなる。
NFATシグナル阻害率を算出した結果を図4に示す。図4から判るように、実施例1で得られた成分A、成分B及び成分C並びに実施例3で調製した成分Aの脱アシル体は、いずれもNFATシグナルを阻害した。すなわち、成分A、成分B及び成分Aの脱アシル体は、NFATにより正に制御される転写を抑制することができる。したがって、成分A、成分B及び成分Aの脱アシル体は、優れたNFATシグナル阻害剤であり、例えば免疫抑制剤、乾癬治療剤、アトピー性皮膚炎治療剤、(心)筋肥大抑制剤及び抗リウマチ薬等の候補物質として同定されたこととなる。
〔実施例6〕育毛効果
本例では、対象物質の育毛効果を検証できるin vitroの実験系を用いて実施例1で得られた成分A及び成分Bの育毛効果を検証した。
ブタ毛包器官培養による毛伸長評価
食肉用豚種の臀部皮膚を適当な大きさに切り分け、余分な脂肪組織を除いた。豚皮は無菌下でヒビテン液(5%ヒビテン液(住友製薬)を水で5~20倍に希釈)に5分~10分浸することにより消毒を行った後にD-PBSで数回洗浄した。その後、実体顕微鏡下にて洗浄した豚皮から毛包を単離してWilliam's Medium E培地(Invitrogen)培地中に集めた。
単離した毛包は、1ウェルあたり400μlのWilliam's Medium E培地(1% Penicillin-Streptomycin 溶液(Invitrogen)添加)が入った24ウェル培養プレートに1本ずつ入れた。毛包は37℃、5% CO2条件下にて8日間培養し、その間、1日または2日おきに培地の交換を行った。
上記の培地に実施例1で得られた成分A、成分Bを添加し、溶媒コントロールであるエタノール添加群とともに8日間培養した。培養を開始日(0日目)および8日目の実体顕微鏡画像をCCDカメラ(pixera model.No.PVC 100C)で撮影し、その画像から毛球部基底部から毛幹先端までの長さを測定した。その後、溶媒コントロール群の培養前後における毛伸長量を100%として実施例1で得られた成分A及び成分Bの毛伸長率を算出した。
本例では、対象物質の育毛効果を検証できるin vitroの実験系を用いて実施例1で得られた成分A及び成分Bの育毛効果を検証した。
ブタ毛包器官培養による毛伸長評価

上記の培地に実施例1で得られた成分A、成分Bを添加し、溶媒コントロールであるエタノール添加群とともに8日間培養した。培養を開始日(0日目)および8日目の実体顕微鏡画像をCCDカメラ(pixera model.No.PVC 100C)で撮影し、その画像から毛球部基底部から毛幹先端までの長さを測定した。その後、溶媒コントロール群の培養前後における毛伸長量を100%として実施例1で得られた成分A及び成分Bの毛伸長率を算出した。
結果
毛伸長率を算出した結果を図6に示す。図6から判るように、実施例1で得られた成分A及び成分Bの添加群においては、溶媒コントロール群と比較して有意な毛伸長が認められた。この結果から、実施例1で得られた成分A及び成分BCは育毛効果を有することが明らかとなった。すなわち、実施例1で得られた成分A及び成分Bは、育毛効果を示す活性成分であり、育毛剤として、或いは外用剤として使用できることが明らかとなった。
毛伸長率を算出した結果を図6に示す。図6から判るように、実施例1で得られた成分A及び成分Bの添加群においては、溶媒コントロール群と比較して有意な毛伸長が認められた。この結果から、実施例1で得られた成分A及び成分BCは育毛効果を有することが明らかとなった。すなわち、実施例1で得られた成分A及び成分Bは、育毛効果を示す活性成分であり、育毛剤として、或いは外用剤として使用できることが明らかとなった。
Claims (19)
- 上記式(I)中、R1がアンゲロイル基、イソバレロイル基及びセネシオイル基からなる群から選ばれる少なくとも1種の官能基であり、R2はアンゲロイル基、イソバレロイル基、2メチルブチル基及びセネシオイル基からなる群から選ばれる少なくとも1種の官能基である請求項1記載のNFATシグナル阻害剤。
- 上記一般式(1)で表されるクマリン誘導体が、アルカンジェリシン、3’-angeloyloxy-4’-isovaleryloxy-2’,3’-dihydrooroselol、3’-angeloyloxy-4’-senecioyloxy-2’,3’-dihydrooroselol及び3’-hydroxy-4’-angeloyloxy-2’,3’-dihydrooroselolからなる群より選択される少なくとも1種の化合物である請求項1記載のNFATシグナル阻害剤。
- 上記式(I)中、R1がアンゲロイル基、イソバレロイル基及びセネシオイル基からなる群から選ばれる少なくとも1種の官能基であり、R2はアンゲロイル基、イソバレロイル基、2メチルブチル基及びセネシオイル基からなる群から選ばれる少なくとも1種の官能基である請求項4記載の育毛剤。
- 上記一般式(1)で表されるクマリン誘導体が、アルカンジェリシン、3’-angeloyloxy-4’-isovaleryloxy-2’,3’-dihydrooroselol、及び3’-hydroxy-4’-angeloyloxy-2’,3’-dihydrooroselolからなる群より選択される少なくとも1種の化合物である請求項4記載の育毛剤。
- 上記式(I)中、R1がアンゲロイル基、イソバレロイル基及びセネシオイル基からなる群から選ばれる少なくとも1種の官能基であり、R2はアンゲロイル基、イソバレロイル基、2メチルブチル基及びセネシオイル基からなる群から選ばれる少なくとも1種の官能基である請求項8記載の使用。
- 上記一般式(1)で表されるクマリン誘導体が、アルカンジェリシン、3’-angeloyloxy-4’-isovaleryloxy-2’,3’-dihydrooroselol、3’-angeloyloxy-4’-senecioyloxy-2’,3’-dihydrooroselol及び3’-hydroxy-4’-angeloyloxy-2’,3’-dihydrooroselolからなる群より選択される少なくとも1種の化合物である請求項8記載の使用。
- 上記式(I)中、R1がアンゲロイル基、イソバレロイル基及びセネシオイル基からなる群から選ばれる少なくとも1種の官能基であり、R2はアンゲロイル基、イソバレロイル基、2メチルブチル基及びセネシオイル基からなる群から選ばれる少なくとも1種の官能基である請求項11記載の使用。
- 上記一般式(1)で表されるクマリン誘導体が、アルカンジェリシン、3’-angeloyloxy-4’-isovaleryloxy-2’,3’-dihydrooroselol、及び3’-hydroxy-4’-angeloyloxy-2’,3’-dihydrooroselolからなる群より選択される少なくとも1種の化合物である請求項11記載の使用。
- 上記式(I)中、R1がアンゲロイル基、イソバレロイル基及びセネシオイル基からなる群から選ばれる少なくとも1種の官能基であり、R2はアンゲロイル基、イソバレロイル基、2メチルブチル基及びセネシオイル基からなる群から選ばれる少なくとも1種の官能基である請求項14記載のNFATシグナル阻害方法。
- 上記一般式(1)で表されるクマリン誘導体が、アルカンジェリシン、3’-angeloyloxy-4’-isovaleryloxy-2’,3’-dihydrooroselol、3’-angeloyloxy-4’-senecioyloxy-2’,3’-dihydrooroselol及び3’-hydroxy-4’-angeloyloxy-2’,3’-dihydrooroselolからなる群より選択される少なくとも1種の化合物である請求項14記載のNFATシグナル阻害方法。
- 上記式(I)中、R1がアンゲロイル基、イソバレロイル基及びセネシオイル基からなる群から選ばれる少なくとも1種の官能基であり、R2はアンゲロイル基、イソバレロイル基、2メチルブチル基及びセネシオイル基からなる群から選ばれる少なくとも1種の官能基である請求項17記載の育毛方法。
- 上記一般式(1)で表されるクマリン誘導体が、アルカンジェリシン、3’-angeloyloxy-4’-isovaleryloxy-2’,3’-dihydrooroselol、及び3’-hydroxy-4’-angeloyloxy-2’,3’-dihydrooroselolからなる群より選択される少なくとも1種の化合物である請求項17記載の育毛方法。
