生体触媒の存在下でアクリロニトリルを反応させて、アクリルアミドを製造する方法において、該反応を、管式熱交換器を備えた反応容積1m3以上の反応器を用い、かつ該熱交換器に導入する冷却水の温度と反応温度との差を5~20℃に保持して行うことを特徴とする、前記方法。
本発明の方法において、前記反応としては、例えば、連続反応が挙げられる。また、当該連続反応は、例えば、反応器の入口と出口とにおける反応混合物中アクリルアミドの濃度差(重量%)を、冷却水の温度と反応温度との差(℃)で除した値(重量%/℃)が、1.5~2.5の範囲内となるように保持して行うことができる。
固定化に用いる固定化担体としては、例えば、ガラスビーズ、シリカゲル、ポリウレタン、ポリアクリルアミド、ポリビニルアルコール、カラギーナン、アルギン酸、寒天、ゼラチン等が挙げられる。
生体触媒の使用量は、用いる生体触媒の種類や形態によって異なるが、反応器中に添加する生体触媒の活性が、反応温度10℃で乾燥菌体1mg当たり50~200U程度となるように調整することが好ましい。ただし、前記単位U(ユニット)とは、1分間にアクリロニトリルからアクリルアミドを1マイクロモル生成させる酵素量を意味し、製造に用いるアクリロニトリルを用いて測定した値である。
また、本発明の製造方法における反応時間は、限定はされないが、例えば、1~50時間であることが好ましく、より好ましくは3~20時間である。
本発明の製造方法に用いる管式熱交換器としては、例えば、多管式熱交換器及び2重以上のコイル式熱交換器等が好ましく挙げられる。
多管式熱交換器としては、例えば、U字管式熱交換器及び直管式熱交換器等が好ましく挙げられ、中でも、U字管式熱交換器がより好ましい。
U字管式熱交換器とは、伝熱管をU字形に曲げ管端を管板に取り付けた熱交換器である。U字管の曲げの最小半径は、伝熱管外径の1~3倍が好ましく、より好ましくは1.5倍~2.5倍である。
本発明で用いる多管式熱交換器(特にU字管式熱交換器)においては、例えば、流路数が、2~6であることが好ましく、より好ましくは2~4である。通常、熱交換器の伝熱管における冷却水の出入口には、仕切り室があり、その内部には冷却水を導くための仕切り板が取り付けられているが、流路数とは、この仕切り板によって仕切り室が仕切られた数(以下、Nと表記する。)を意味する(例えば、プロセス機器構造設計シリーズ1 熱交換器 化学工学協会編 P23(丸善株式会社)を参照)。具体的には、例えば、Nが2のときは流路数2であり、Nが3のときは流路数4であり、Nが4のときは流路数6である。なお、構造上、U字管式熱交換器の流路数は、2以上である。流路数が6より大きくなると、仕切り数が多くなり、熱交換の構造が複雑になるため、安価に製造又は入手することができない。
本発明の製造方法に用いる反応器が、多管式熱交換器を(内部に)備えたものである場合は、反応原料が供給される反応器の内部で、反応混合物と冷却水との熱交換が管式熱交換器の伝熱管を介して行われる。攪拌などによる混合操作で、反応混合物の流れは多管式熱交換器が障害物となり乱されるため、伝熱管の伝熱係数は増大する。これにより、ジャケット式冷却や反応器内部に設置した1重コイル式熱交換器に比べて、より効率的な熱交換を行うことができる。
前記2重以上のコイル式熱交換器とは、2本以上の伝熱管をコイル状に巻いた管束を反応器内に備えたものであり、蛇管式熱交換器とも言う。コイルは、通常、銅、鋼及び特殊鋼などの管をらせん状に巻いた構造である。
本発明で用いるコイル数は、2重以上であればよく、特に限定はされないが、例えば、2~3重が好ましく、より好ましくは2重である。同一の伝熱面積で比較して、コイル数が1つ(1重コイル)では、必要な伝熱面積を確保するために、らせん状に巻かれた高さ方向の隣り合う伝熱管の距離(コイルピッチ)が小さくなる。そのため、攪拌などによる混合操作で、らせん状に巻かれたコイルの内側と外側との反応混合物の流れが抑制されるため効率的な熱交換を行うことができない。
本発明で使用する反応器は、管式熱交換器を内部に設置できる形状であればよく、例えば槽型反応器が挙げられる。
本発明で使用する反応器は、反応容積が1m3以上であり、好ましくは2m3以上、より好ましくは5m3以上である。ここで、反応容積とは、反応器内に導入(仕込み)可能な反応混合物の正味容量を意味し、多槽連続反応器の場合、1槽当たりの反応混合物の正味容量を意味する。
本発明の製造方法において、冷却水とは、反応の際に生成する反応熱を除熱する目的で、管式熱交換器に(詳しくは該熱交換器の伝熱管に)導入するために用いられるものであり、その種類は特に限定されるものではなく、例えば塩化カルシウム液、塩化ナトリウム液、塩化マグネシウム液などの塩化物溶液や、エチレングリコール液などのアルコール液、工業用水などを使用することができる。
反応で生じる発熱量はΔCに比例し、熱交換器の除熱量はΔTに比例する。ΔC/ΔTの値が2.5よりも大きいと、反応器当たりのアクリルアミド生産性が高過ぎるため、除熱に過大な伝熱面積が必要となるだけでなく、反応効率が低下して触媒の供給量が増大し、安価にアクリルアミドを製造することができなくなる。
本発明の製造方法においては、原料化合物であるアクリロニトリルから目的生成物であるアクリルアミドへの変換率は、例えば、重量濃度基準で、85%以上であることが好ましく、より好ましくは90%以上、さらに好ましくは95%以上である。
本発明の製造方法において、反応後のアクリルアミドの回収及び精製は、例えば、濃縮操作(蒸発濃縮等)、活性炭処理、イオン交換処理、ろ過処理及び晶析操作等により行うことができる。
ニトリルヒドラターゼ活性を有するロドコッカス・ロドクロウス Rodococcus rhodochrousJ1株(受託番号:FERM BP-1478として独立行政法人 産業技術総合研究所 特許生物寄託センター(日本国茨城県つくば市東1丁目1番地1中央第6)に1987年9月18日に国際寄託されている)を、グルコース2%、尿素1%、ペプトン0.5%、酵母エキス0.3%、塩化コバルト0.05%(何れも質量%)を含む培地(pH7.0)により30℃で好気的に培養した。これを遠心分離機及び50mMリン酸緩衝液(pH7.0)を用いて、集菌洗浄し、生体触媒としての菌体懸濁液(乾燥菌体15質量%)を調製した。
(反応容積2m3、U字管式熱交換器でのアクリルアミド生成反応、ΔT=20℃)
SUS製ジャケット付反応器(内径1.3m、高さ2.0m、底面1/4半楕円)にU字管式熱交換器(伝熱管外径8mm、伝熱管本数50本、流路数4、伝熱面積3.0m2)を2器設置した。
反応器に、50mMリン酸緩衝液(pH7.0)を800L/hr、アクリロニトリルを350L/hr、上記調製後の菌体懸濁液を2.5L/hrで連続的に添加し、反応器内の反応混合物の液量が2m3となるように、反応器出口の抜き出し流量をコントロールした。
反応開始から10時間後、反応器から流出してくる反応液(25℃)を、ガスクロマトグラフィー(カラム:Waters社製 PoraPak-PS、1m、180℃、キャリアガス:ヘリウム、検出器:FID)及び屈折計(ATAGO、RX-1000)により測定し、予め測定した検量線により、反応液中のアクリロニトリル及びアクリルアミド濃度を算出したところ、アクリロニトリル1.1重量%、アクリルアミド33.4重量%(ΔC/ΔT=1.7)であり、反応器に供給したアクリロニトリルのうちの96%以上がアクリルアミドに変換された。
U字管式熱交換器を2重コイル式熱交換器(伝熱管外径34.0mm、外側コイル:コイル巻き径1.0m-10段巻き、内側コイル:コイル巻き径0.7m-10段巻き、伝熱面積6.0m2)へ変更した以外は、実施例1と同様にして、アクリロニトリルからアクリルアミドへの反応を行った。
反応開始から10時間後、反応器から流出してくる反応液(25℃)を、ガスクロマトグラフィー及び屈折計で分析したところ、アクリロニトリル1.3重量%、アクリルアミド33.2重量%(ΔC/ΔT=1.7)であり、反応器に導入したアクリロニトリルのうちの95%以上がアクリルアミドに変換された。
2重コイル式熱交換器を反応器から取り外し、反応器のジャケット(伝熱面積6.0m2)に冷却水を流した以外は、実施例2と同様にして、アクリロニトリルからアクリルアミドへの反応を行った。
反応開始から10時間後、ジャケット式熱交換器では伝熱係数が低いため十分に反応熱を除熱できなかったため、反応温度は28℃(ΔT≒23℃)まで上昇した。反応器から流出してくる反応液を、ガスクロマトグラフィー及び屈折計で分析した。温度上昇により生体触媒が失活したため、アクリロニトリル4.8重量%、アクリルアミド28.5重量%(ΔC/ΔT=1.2)となり、反応器に供給したアクリロニトリルのうちの82%しかアクリルアミドに変換されなかった。
反応器内に1重コイル式熱交換器(伝熱管外径34.0mm、コイル巻き径1.0m-18段巻き、伝熱面積6.0m2)を設置し流した以外は、実施例2と同様にして、アクリロニトリルからアクリルアミドへの反応を行った。
反応開始から10時間後、比較例1と同様に、反応温度を25℃に維持することができず、反応温度は27℃(ΔT≒22℃)まで上昇した。反応器から流出してくる反応液を、ガスクロマトグラフィー及び屈折計で分析した。温度上昇により生体触媒が失活したため、アクリロニトリル4.1重量%、アクリルアミド29.4重量%(ΔC/ΔT=1.3)であり、反応器に供給したアクリロニトリルのうちの84%しかアクリルアミドに変換されなかった。
実施例1と同様のU字管式熱交換器を用いて、反応器に、50mMリン酸緩衝液(pH7.0)を1200L/hr、アクリロニトリルを140L/hr、上記調製後の菌体懸濁液を0.9L/hrで連続的に添加し、反応器内の反応混合物の液量が2m3となるように、反応器出口の抜き出し流量をコントロールした。
U字管式熱交換器に冷却水(10%エチレングリコール水溶液)を導入し、熱交換器の入口側及び出口側に設置した温度計により、冷却水の温度を測定した。反応器内の液温(反応温度)が25℃に保たれるように、冷却水流量をコントロールした。なお、ジャケットには冷却水を流していない。熱交換器の入口及び出口における冷却水の温度は、それぞれ17℃及び22℃であった(ΔT=5℃)。アクリロニトリルからアクリルアミドへの反応を行った。
実施例2と同様の2重コイル式熱交換器を用いた以外は、実施例3と同様にして、アクリロニトリルからアクリルアミドへの反応を行った。
反応開始から10時間後、反応器から流出してくる反応液(25℃)を、ガスクロマトグラフィー及び屈折計で分析したところ、アクリロニトリル0.6重量%、アクリルアミド10.7重量%(ΔC/ΔT=2.1)であり、反応器に導入したアクリロニトリルのうちの93%以上がアクリルアミドに変換された。
2重コイル式熱交換器を反応器から取り外し、反応器の外部に反応液の循環路を設け、循環路内に循環ポンプ及び固定管板式熱交換器(伝熱管外径13.9mm、長さ2.5m、伝熱管本数55本、伝熱面積6.0m2)を設置し、反応液を伝熱管内部に流入させ、冷却水を胴側に流した以外は実施例4と同様にして、アクリロニトリルからアクリルアミドへの反応を行った。
反応開始から10時間後、反応器から流出してくる反応液(25℃)を、ガスクロマトグラフィー及び屈折計で分析した。反応液の循環により生体触媒が失活したため、アクリロニトリル1.2重量%、アクリルアミド9.9重量%(ΔC/ΔT=2.0)であり、反応器に供給したアクリロニトリルのうちの86%しかアクリルアミドに変換されなかった。
また、反応液を循環させるポンプや配管などの付帯設備費や循環動力コストは、実施例3及び4の場合よりも大幅に増加した。
SUS製ジャケット付反応器(内径25cm、高さ40cm、ジャケット伝熱面積2360cm2)
50mMリン酸緩衝液(pH7.0)を9.0L/hr、アクリロニトリルを1.05L/hr、実施例1で作成した菌体懸濁液を50mMリン酸緩衝液で100倍希釈した希釈液を200g/hrで連続的に添加し、反応器内の反応混合物の液量が15Lとなるように、反応器出口の抜き出し流量をコントロールした。
ジャケットの入口及び出口における冷却水の温度をそれぞれ17℃及び22℃とした(ΔT=5℃)。反応開始から10時間後、反応器から流出してくる反応液(25℃)を、ガスクロマトグラフィー及び屈折計で測定したところ、アクリロニトリル0.3重量%、アクリルアミド11.1重量%(ΔC/ΔT=2.2)であり、反応器に供給したアクリロニトリルのうちの95%以上がアクリルアミドに変換された。
一方、比較例1及び2では、2m3反応において、反応熱を十分に除熱することができず、アクリロニトリルからアクリルアミドへの変換率も低下した。
比較例3では、2m3スケールの反応において、反応熱を除熱することができたが、反応液の循環により触媒が失活したため、アクリロニトリルからアクリルアミドへの変換率が低下した。また、反応液を外部循環させるための設備コストや循環エネルギーコストが増加し、実施例3及び4の場合に比べて、アクリルアミド製造コストが大幅に増加した。
比較例4では、工業スケールよりかなり小さな15Lスケールの反応であったため、ジャケット式冷却でも反応熱を除熱することができた。

Claims (9)
- 生体触媒の存在下でアクリロニトリルを反応させて、アクリルアミドを製造する方法において、該反応を、管式熱交換器を備えた反応容積1m3以上の反応器を用い、かつ該熱交換器に導入する冷却水の温度と反応温度との差を5~20℃に保持して行うことを特徴とする、前記方法。
- 前記反応が連続反応である、請求項1記載の方法。
- 前記反応器の入口と出口とにおける反応混合物中アクリルアミドの濃度差(重量%)を、前記冷却水の温度と前記反応温度との差(℃)で除した値が、1.5~2.5の範囲内となるように保持して前記反応を行う、請求項2記載の方法。
- 前記管式熱交換器が2重以上のコイル式熱交換器である、請求項1~3のいずれか1項に記載の方法。
- 前記管式熱交換器が多管式熱交換器である、請求項1~3のいずれか1項に記載の方法。
- 前記多管式熱交換器がU字管式熱交換器である、請求項5記載の方法。
- 前記U字管式熱交換器の流路数が2~6である、請求項6記載の方法。
- 前記U字管式熱交換器が、前記反応器内に縦置き型で設置されている、請求項6又は7記載の方法。
- 前記U字管式熱交換器の管束が取り外しできる構造である、請求項6~8のいずれか1項に記載の方法。
