Abstract
Die vorliegende Erfindung betrifft die Verwendung von Phenoxazinon zur Inhibition von Enzymen mit Histondeacetylase-Aktivität, sogenannten HDAC-Inhibitoren (HDACi), und die Verwendung von solchen neuen HDACi zur Behandlung und Prophylaxe von Tumoren.
Description
AMINOPHENOXAZINONS ALS ANTITUMOR- UND ANTIENTZÜNDLICHES MITTEL
Die vorliegende Erfindung betrifft die Verwendung von Phenoxazinon zur Inhibition von Enzymen mit Histondeacetylase-Aktivität, sogenannten HDAC-Inhibitoren (HDACi), und die Verwendung von solchen neuen HDACi zur Behandlung und Prophylaxe von Tumoren und Entzündungen.
Das Krankheitsbild „Krebs" umfasst eine Gruppe von verschiedenen Krankheiten, die durch unkontrolliertes Wachstum einer stark heterogenen malignen Zellpopulation charakterisiert sind. Die geschätzte weltweite Inzidenz von verschiedenen Krebstypen liegt bei ca. 10 Millionen, etwa die Hälfte hiervon ist Entwicklungsländern zuzurechnen. Trotz einer großen Zahl von gegenwärtig zur Verfügung stehenden chemothera-
peutischen Substanzen, steht der große Erfolg in der Behandlung von Krebs noch aus. Es besteht nach wie vor ein großer medizinischer Bedarf an zielgerichtet wirksamen und nebenwirkungsreduzierten Substanzen zur Behandlung von Krebs. Deshalb gibt es derzeit große Bemühungen, um neue tumorspezifische Zielstrukturen zu identifizieren und hiergegen antitumorale Substanzen zu entwickeln.
Histondeacetylasen (HDACs) wurden bereits früh mit der Entstehung von verschiedenen Tumoren in Verbindung gebracht, da in verschiedenen Tumoren eine verstärkte Acetylierung von nucleosomalen Histonen beobachtet wird. So wird der Acetylierungsstatus von Lysinen von nucleosomalen Histonen, die die Chroma- tinstruktur modulieren und die Transkriptionsaktivität regulieren, durch die antagonistische Aktion von zwei Enzymfamilien kontrolliert, den Histondeacetylasen (HDACs) und den Histonacetyltransferasen (HATs).
Im Menschen kennt man mehrere HDAC-Unterklassen. Die Klasse-I-Enzyme, HDACl, -2, -3 und -8 weisen ein Molekulargewicht von 42 bis 45 kDa auf und haben Homologien in ihren katalytischen Abschnitten. Klasse-II-Enzyme, HDAC4, -5, -6, -9 und -10, weisen Molekulargewichte in einem Bereich von 120 bis 130 kDa auf, wobei HDAC4, -5,-7 und -9 Homologien in der C-terminalen katalytischen Domäne und der N-terminalen regulatorischen Domäne zeigen. HDACI l ist ein weiteres Mitglied der HDAC-Enzyme. Es weist lediglich niedrige Homologie mit den Enzymen aus den Klassen I und II auf und wird deshalb in eine eigene Klasse eingruppiert. Die SIRC2- Familie von Proteinen bildet eine neue Klasse von Enzymen, die zu den HDACs gezählt werden. Diese Enzyme sind hinsichtlich ihrer Aktivität abhängig von NAD+, während die Klasse-I- und Klasse-II-HDACs einen zinkabhängigen Mechanismus verwenden.
In der Zwischenzeit wurde eine Vielzahl von HDAC-Inhibitoren (auch HDACi genannt) entwickelt, die zum Teil bereits in Studien der klinischen Phase I und II getestet werden; vgl. Minucci & Pelicci. HDAC-Inhibitoren lassen sich in mehrere Gruppen klassifizieren. Sie umfassen die so genannten Hydroxaminsauren, die Nicht-Hydroxamin-HDAC-Inhibitoren, die kurz- kettigen Fettsauren, die Benzamide, die cychschen Peptide und die so genannten SIR2-Inhibitoren Einen Überblick über diese HDAC-Inhibitoren findet sich in Weinmann & Ottow Histone deacetylase Inhibitors, a survey of recent patents Expert Opin Ther Patents 2005, 15 1677-1690
Inwiefern einzelne HDAC-Inmbitoren bei bestimmten Tumorarten eingesetzt werden können, ist bisher nicht vergleichend untersucht worden Es ist davon auszugehen, dass für unterschiedliche Tumorarten auch HDAC-Inhibitoren aus unterschiedlichen Substanzklassen ein optimales Wirkungs-Nebenwirkungsprofil aufweiesen werden.
Trotz der intensiven Arbeiten steht nach wie vor die Suche nach neuen nebenwirkungsarmen, therapeutisch potenten HDAC-Inhibitoren im Mittelpunkt. Es besteht deshalb nach wie vor Bedarf an der Bereitstellung neuer HDAC-Inhibitoren, die diese Eigenschaften haben, ein hohes Maß an Verträglichkeit aufweisen und vorzugsweise im Großmaßstab hergestellt werden können.
Mittlerweile weiß man, dass HDAC-Inhibitoren auch antnnflammatoπsche, antian- giogene und antiartherogene Eigenschaften aufweisen.
Vor diesem Hintergrund ist die der Erfindung zugrunde liegende Aufgabe, einen neuen HDAC-Inhibitor bereitzustellen, der zur Behandlung und Prophylaxe von Tumoren und/oder Entzündungen eingesetzt werden kann, und möglichst nebenwirkungsarm ist.
Diese Aufgabe wird durch die Verwendung von Phenoxazmon zur Inhibition der Histondeacetylase (HDAC) bzw zur Behandlung und/oder Prophylaxe von Tumoren und/oder Entzündungen gelost.
Die Erfinder haben überraschenderweise erkannt, dass es sich bei Phenoxazinonen um potente HDAC-Inhibitoren handelt, die konzentrationsabhängig ihre Wirkung zeigen. Besonders vorteilhaft ist, dass Phenoxazinone selektiv cytotoxisch für Tumorzellen sind, gesunde Zellen jedoch nahezu unbeeinflusst lassen. Phenoxazinone sind deshalb zur nebenwirkungsarmen Behandlung von Tumoren und Entzündungen besonders geeignet. Vorteilhaft ist ferner, dass Phenoxazinone einfach herzustellen und in wässriger Lösung besonders stabil sind. Ein entsprechendes Arzneimittel ist deshalb ohne weiteres im Großmaßstab kostengünstig herstellbar.
Diese Eignung von Phenoxazinonen war insbesondere deshalb überraschend, da diese Substanzklasse bislang in völlig anderem Zusammenhang, nämlich in Bezug auf die so genannte Allelopathie diskutiert wird. Unter Allelopathie versteht man die Eigenschaft von Pflanzen, organische Verbindungen auszuscheiden, welche das Wachstum oder das Keimen anderer Pflanzen unterbinden oder hemmen. Eine der am besten untersuchten Substanzklassen in diesem Zusammenhang sind die Benz- oxazinoide, die in den Pflanzen der Familie Poaceae, zu der Weizen, Mais und Roggen zählen, als inaktives Glucosid in Vakuolen vorkommen. Diese glycosidischen Vorstufen werden bei einer Verletzung der Pflanze ausgeschieden und mittels einer im Plastid gespeicherten ß-Glucosidase u.a. in die aktiven Benzoxazinoide 2,4- Dihydroxy-l,4-benzoxazin-3-on (DIBOA) und 2,4-Dihydroxy-7-methoxy-l,4- benzoxazin-3-on (DIMBOA) umgewandelt. Aufgrund einer spontanen und mikrobiel- len Hydrolyse werden die aktiven Benzoxazinoide u.a. in die Benzoxazolinone Benzoxazolin-2-on (BOA) und 6-Methoxy-benzoxazolin-2-on (MBOA) überführt, die auch als sog. "parentale" Komponenten bezeichnet werden. Aus diesen parentalen Komponenten gehen durch mikrobielle Transformation über den Zwischenschritt der Aminophenole, wie bspw. dem 2-Aminophenol (AP), oder direkt über Metabolisie- rung durch Pilze und Bakterien die erfindungsgemäßen Phenoxazinone hervor. Diese Phenoxazinone wiederum können mikrobiell degradiert werden und durchlaufen dabei bspw. Acetylierungs- und Oxidierungsreaktionen.
Die aus den Pflanzen stammenden Benzoxazinoide, Benzoxazolinone, Aminophenole und Phenoxazinone und deren Degradationsprodukte zeigen eine allelopathische
Wirkung auf verschiedene andere Pflanzen, Pilze, Bakterien und Insekten. Die Wirkmechanismen der allelopathischen Wirkung sind bisher jedoch nur in Ansätzen aufgeklärt.
Die Erfinder haben völlig überraschend festgestellt, dass die Phenoxazinone als Inhibitoren der zellulären Histondeacetylase wirken.
Die der Erfindung zugrunde liegende Aufgabe wird hiermit vollkommen gelöst.
Dabei ist es erfindungsgemäß bevorzugt, wenn das Phenoxazinon ein Aminopheno- xazinon ist.
Diese Maßnahme hat den Vorteil, dass solch ein Phenoxazinon bereitgestellt wird, das sich nach den Erkenntnissen der Erfinder als HDAC-Inhibitor und damit zur Behandlung und/oder Prophylaxe von Tumoren und Entzündungen besonders eignet.
Weiter ist es bevorzugt, wenn das Phenoxazinon ausgewählt ist aus der Gruppe bestehend aus: 2-Amino-3H-phenoxazin-3-on (APO), 2-Amino-7-methoxy-3H- phenoxazin-3-on (AMPO), 2-Acetylamino-7-hydroxy-3H-phenoxazin-3-on (AAHPO), 2-Acetylamino-7-methoxy-3H-phenoxazin-3-on (AAMPO), 2-Acetylamino-3H- phenoxazin-3-on (AAPO), 2-Amino-7-hydroxy-3H-phenoxazin-3-on (AHPO), , 2- Amino-4, 6, 7-trimethoxy-3H-phenoxazin-3-on (AM3PO), 2-(2-Hydroxyacetyl)amino- 3H-phenoxazin-3-on (HAAPO), 7-Hydroxy-2-(2-hydroxyacetyl)amino-3H-phen- oxazin-3-on (HHAAPO), 2-(N-Hydroxy)acetylamino-3H-phenoxazin-3-on (NHAAPO).
Diese Maßnahme hat den Vorteil, dass eine Vielzahl von Phenoxazinonen bereitgestellt werden, die APO und AMPO sowie deren mikrobiellen Degradationsprodukte erfassen und allesamt als potenzielle HDAC-Inhibitoren in Frage kommen.
Weiter ist es bevorzugt, wenn die erfindungsgemaße Verbindung folgende Struktur aufweist
wobei Rl ausgewählt ist aus der Gruppe bestehend aus: H, OH, OCH3, wobei R2 ausgewählt ist aus der Gruppe bestehend aus: H, CH2OH, wobei R3 ausgewählt ist aus der Gruppe bestehend aus: H, CH2OH, wobei R4 ausgewählt ist aus der Gruppe bestehend aus: O, OH, wobei R5 ausgewählt ist aus der Gruppe bestehend aus: H, COCH3, COCH2OH, und wobei R6 ausgewählt ist aus der Gruppe bestehend aus: H, OH.
Diese Maßnahme hat den Vorteil, dass dem Fachmann das Grundgerust bereitgestellt wird, das den Aminophenoxazinonen APO und AMPO gemein ist, die sich lediglich hinsichtlich des Restes Rl voneinander unterscheiden. Durch Variationen der Reste Rl, R2, R3, R4, R5 und R6 können die therapeutischen bzw. inhibitorischen Eigenschatten der Verbindung optimiert werden. Insbesondere kann durch Variation des Restes R2 die Loslichkeit der Verbindung erhöht werden.
Erfindungsgemaß ist es bevorzugt, wenn bei der vorstehenden Struktur der Rest R2 Glucuronsaure aufweist.
Diese Maßnahme hat den Vorteil, dass die erfindungsgemaße Verbindung dadurch in ihrer Loslichkeit erhöht wird. Die Kopplung von Glucuronsaure an die Verbindung an Position Rl erfolgt durch dem Fachmann bekannte Maßnahmen, bspw. enzyma- tisch unter Verwendung der Glucoronyltransferase. Glucuronsaure bzw. Glucuronat ist ein Derivat der Glucose, von der es sich durch die Oxidationsstufe am 6. Kohlenstoff unterscheidet, wo eine Carboxylgruppe statt einer Hydroxymethylgruppe
vorliegt. Glucuronsäure löst sich gut in Wasser und Alkohol, ihr Schmelzpunkt liegt bei 1650C. In der kristallinen Form zeigt sich Mutarotation.
Besonders bevorzugt ist es, wenn das Arzneimittel zur Behandlung von Lebertumoren vorgesehen ist.
So hat sich vorteilhafterweise gezeigt, dass Phenoxazinon und insbesondere APO und AMPO besonders proliferationshemmend und cytotoxisch auf Hepatomzellen wirken, wobei überraschenderweise primäre humane Hepatocyten nicht geschädigt werden.
Ein weiterer Gegenstand der vorliegenden Erfindung betrifft deshalb ein Verfahren zur Behandlung und/oder Prophylaxe eines Tumors bei einem Lebewesen, das folgende Schritte aufweist: (1) Verabreichung eines Phenoxazinon in das Lebewesen, und (2) ggf. Wiederholung des Schrittes 1.
Es versteht sich, dass die vorstehend genannten und die nachstehend noch zu erläuternden Merkmale nicht nur in der jeweils angegebenen Kombination, sondern auch in anderen Kombinationen oder in Alleinstellung verwendbar sind, ohne den Rahmen der vorliegenden Erfindung zu verlassen.
Die Erfindung wird nun anhand von Ausführungsbeispielen veranschaulicht, die rein exemplarisch sind und die Reichweite der Erfindung nicht einschränken. Dabei wird Bezug genommen auf die beigefügten Figuren.
Fig. 1 zeigt, dass APO und AMPO eine inhibitorische Aktivität auf die Histon- deacetylase aufweisen. Mittels eines auf Fluoreszenz basierenden HDAC-Aktivitäts-/Inhibitions-Screening-Assays wurde jeweils die prozentuale Inhibition von HDAC-Enzymen in zellulären Extrakten aus HeLa-Zellen standardisiert gemessen. 100 % Inhibition wird erreicht, wenn kein HDAC-Substrat umgesetzt werden kann, da alle im HeLa-
Kernextrakt vorhandenen Enzyme inhibiert sind. Dargestellt sind die Mittelwerte von drei unabhängigen Experimenten und die Standardabweichung. Suberoylanilid-Hydroxaminsäure (SAHA) als etablierter HDAC-Inhibitor wurde als Positivkontrolle verwendet.
Fig. 2 zeigt die Akkumulierung von acetyliertem Histon H3 (17 kDa) in
Hepatomzellen nach Inkubation mit APO. Hep3B-Zellen wurden für 24 h ohne HDAC-Inhibitor (Kontrolle) oder mit 5 μM APO (APO) oder 2 μM SAHA (SAHA) behandelt. Als Nachweis für die gleiche Proteinbeladung wurde Vinculin (116 kDa) simultan im Blot nachgewiesen.
Fig. 3 zeigt den anti-proliferativen Effekt von APO und AMPO auf Tumorzelllinien. Die Hepatomzelllinien Hep3B, HepG2 und HuH7 wurden 5 Tage mit verschiedenen Konzentrationen von APO und AMPO inkubiert. Die Zellviabilität wurde photometrisch mittels SRB-Assay bestimmt. Dargestellt sind die Mittelwerte von drei unabhängigen Experimenten und die Standardabweichung.
Fig. 4 zeigt die cytotoxische Wirkung von APO und AMPO auf Tumorzellen.
Hepatomzellen wurden exemplarisch für 48 h mit verschiedenen Konzentrationen von APO und AMPO behandelt. Die auf der y-Achse dargestellte relative LDH-Aktivität wurde anhand der Überstände spektrophotometrisch bestimmt und in Bezug zu unbehandelten Hepatomzellen gesetzt. Dargestellt sind die Mittelwerte von drei unabhängigen Experimenten und die Standardabweichung.
Fig. 5 zeigt die fehlende Toxizität von APO und AMPO auf primäre humane
Hepatocyten (PHH). PHH von drei unterschiedlichen Spendern wurden für 48 h mit verschiedenen Konzentrationen von APO und AMPO behandelt. Die Inkubation mit 40 μM TSA als Positivkontrolle führte zu 100 % Zelltod (schwarzes Dreieck). Die LDH-Aktivität wurde anhand
der Überstände spektrophotometrisch bestimmt und in Bezug zu unbehandelten Hepatocyten gesetzt. Dargestellt sind die Mittelwerte von drei unabhängigen Experimenten und die Standardabweichung.
Ausführungsbeispiele
Beispiel 1: Aktivität von APO und AMPO als Histondeacetylase-Inhibitor CHDACO
Ein kommerzielles Testsystem steht zur Verfügung, mit dem Substanzen daraufhin untersucht werden können, ob sie eine inhibitorische Aktivität gegenüber Histonde- acetylasen aufweisen: Cayman Chemical HDAC Activity/Inhibitor Screening Assay Kit (Biozol, Eching, Deutschland). In einem ersten Schritt wurden mit diesem Testsystem APO und AMPO untersucht. Hierbei wurden HeLa-Zellkernextrakte mit verschiedenen Konzentrationen von APO und AMPO (1, 5, 10, 20, 50 und 100 μM) und einem testspezifischen HDAC-Substrat inkubiert. Diese auf Fluoreszenz basierende Methode erlaubt eine prozentuale Angabe der HDAC-Inhibition.
Das Ergebnis dieses Experimentes ist in Fig. 1 dargestellt. Dabei zeigt sich eine konzentrationsabhängige HDACi-Aktivität, sowohl für APO (-■-) als auch für AMPO (-A-). Als Positivkontrolle wurde Suberoylanilid-Hydroxaminsäure (SAHA) verwendet, ein bekannter HDACi, der bereits in der klinischen Onkologie eingesetzt wird (-
Als zusätzlicher und unabhängiger Nachweis für eine inhibitorische Wirkung von Aminophenoxazinonen auf HDAC-Enzyme wurden Hepatomzellen (Hep3B und HepG2) mit 5 oder 10 μM APO bzw. AMPO für 24 h inkubiert und mittels Western- Blotting der Acetylierungsstatus von Histon-H3-Proteinen untersucht (Anti-Acetyl- Histon-H3-Antikörper AcH3, 1:20.000, Biomol Hamburg, Deutschland; Anti- Vinculin-Antikörper, 1:5.000, Sigma-Aldrich). Eine Akkumulierung acetylierter Histonproteine ist ein Surrogatparameter für die Hemmung der Acetylgruppen- abspaltenden HDAC-Enzyme.
Das Ergebnis ist in Fig. 2 dargestellt. Dabei zeigt sich eine Akkumulierung von acety- liertem Histon H3 im Vergleich zur Kontrolle sowohl bei APO als auch bei AMPO in beiden Hepatomzelllinien (her exemplarisch die Behandlung mit APO und SAHA bei Hep3B dargestellt. Als Positivkontrolle fungierte die Behandlung mit 2 μM SAHA.
Beispiel 2: Proliferationshemmende Wirkung von APO und AMPO auf Tumorzellen
Für die Bestimmung eines inhibitorischen Effektes auf das Proliferationsverhalten von Tumorzellen wurden exemplarisch Hepatomzellen mit den Aminophenoxazino- nen APO bzw. AMPO inkubiert und ein Sulforhodamin-B-Cytotoxizitätsassay (SRB- Assay) durchgeführt. Hierfür wurden Hepatomzellen (Hep3B, HepG2 und HuH7) mit verschiedenen Konzentrationen von APO und AMPO (5, 10, 20 und 50 μM) für fünf Tage behandelt. Es erfolgte nach Fixierung eine Färbung der Zellen mit SRB (Sulfo- rhodamin-B-Natriumsalz, Sigma-Aldrich). Photometrisch wurde die Färbeintensität gemessen und die Viabilität (anhand unbehandelter Zellen) berechnet.
Das Ergebnis dieses Experimentes ist in Fig. 3 dargestellt. Dabei zeigte sich für alle drei Zelllinien, dass sowohl APO als auch AMPO das Wachstum signifikant und konzentrationsabhängig hemmen konnten.
Beispiel 3: Selektive Cytotoxizität von APO und AMPO auf Tumorzellen
Als Surrogatparameter für eine Zellschädigung von Tumorzellen durch die Substanzen APO und AMPO wurden Hepatomzelllinien (Hep3B, HuH7) für 48 h mit verschiedenen Konzentrationen von APO und AMPO (5, 50 und 100 μM) inkubiert. Nachfolgend wurden die Überstände spektrophotometrisch auf Aktivität des Enzyms Laktatdehydrogenase (LDH) gemessen (Sigma TOX7 In Vitro Toxicology Assay Kit, Sigma-Aldrich). Ein Anstieg der LDH gilt als sicheres Zeichen für eine schwerwiegende Zellschädigung mit Verlust der Membranintegrität.
Das Ergebnis dieses Experimentes ist in Fig. 4 dargestellt. Dabei zeigte sich bei beiden Hepatomzelllinien ein konzentrationsabhängiger Einfluss von APO und AMPO auf die Aktivität der LDH. Das heißt, mit zunehmender Konzentration von APO und AMPO werden die untersuchten Tumorzellen vermehrt abgetötet.
Im Vergleich zu Hepatomzellen wurde nachfolgend eine Charakterisierung von nicht-malignen Parenchymzellen, nämlich primären humanen Hepatozyten von drei unterschiedlichen Spendern durchgeführt.
Das Ergebnis ist in Fig. 5 dargestellt. Dabei zeigte sich überraschend, dass weder die Behandlung mit APO noch mit AMPO, selbst bei hohen Konzentrationen von bis zu 100 μM, zu einer relevanten Zellschädigung führt. Als Positivkontrolle fungierten 40 μM TSA (bekannter "toxischer" HDACi, der in hohen Konzentrationen Zelltod auslösen kann).
Beispiel 4: Schlussfolgerung
Die Erfinder konnten durch Experimente erstmals für die pflanzlichen Produkte der Phenoxazinone, exemplarisch vertreten durch APO und AMPO, eine HDACi-Aktivität zeigen. Sowohl die Ergebnisse des HDAC-Aktivitäts/Inhibitor-Screening-Assays als auch die der Western-Blot-Analysen zeigten eine HDACi-Aktivität im μM- Konzentrationsbereich. Zusätzlich konnte eine proliferationsinhibierende und cytotoxische Wirkung auf Tumorzellen gezeigt werden. Im Gegensatz hierzu tolerieren nicht-maligne primäre humane Hepatozyten diese Substanzen selbst in hohen Konzentrationen.

Verwendung eines Phenoxazinons zur Inhibition der Histondeacetylase (HDAC).
Verwendung nach Anspruch 1, dadurch gekennzeichnet, dass das Phenoxazi- non ein Aminophenoxazinon ist.
Verwendung nach Anspruch 1 oder 2, dass das Phenoxazinon ausgewählt ist aus der Gruppe bestehend aus: 2-Amino-3H-phenoxazin-3-on (APO), 2-Ami- no-7-methoxy-3H-phenoxazin-3-on (AMPO), 2-Acetylamino-7-hydroxy-3H- phenoxazin-3-on (AAHPO), 2-Acetylamino-7-methoxy-3H-phenoxazin-3-on (AAMPO), 2-Acetylamino-3H-phenoxazin-3-on (AAPO), 2-Amino-7-hydroxy- 3H-phenoxazin-3-on (AHPO), , 2-Amino-4,6,7-trimethoxy-3H-phenoxazin-3- on (AM3PO), , 2-(2-Hydroxyacetyl)amino-3H-phenoxazin-3-on (HAAPO), 7- Hydroxy-2-(2-hydroxyacetyl)amino-3H-phenoxazin-3-on (HHAAPO), 2-(N- hydroxy)acetylamino-3H-phenoxazin-3-on (NHAAPO).

Verwendung von Phenoxazinon zur Herstellung eines Arzneimittels zur Behandlung und/oder Prophylaxe von Tumoren und/oder Entzündungen.
Verwendung nach Anspruch 6 dadurch gekennzeichnet, dass das Phenoxazinon ein Aminophenoxazinon ist.
Verwendung nach Anspruch 6 oder 7, dadurch gekennzeichnet, dass das Phenoxazinon ausgewählt ist aus der Gruppe bestehend aus: 2-Amino-3H- phenoxazin-3-on (APO), 2-Amino-7-methoxy-3H-phenoxazin-3-on (AMPO), 2-Acetylamino-7-hydroxy-3H-phenoxazin-3-on (AAHPO), 2-Acetylamino-7- methoxy-3H-phenoxazin-3-on (AAMPO), 2-Acetylamino-3H-phenoxazin-3-on (AAPO), 2-Amino-7-hydroxy-3H-phenoxazin-3-on (AHPO), 2-Amino-4,6,7- trimethoxy-3H-phenoxazin-3-on (AM3PO), , 2-(2-Hydroxyacetyl)amino-3H- phenoxazin-3-on (HAAPO), 7-Hydroxy-2-(2-hydroxyacetyl)amino-3H-phen- oxazin-3-on (HHAAPO), 2-(N-Hydroxy)acetylamino-3H-phenoxazin-3-on (NHAAPO).

12. Verfahren zur Behandlung und/oder Prophylaxe eines Tumors bei einem Lebewesen, das folgende Schritte aufweist:
(1) Verabreichung eines Phenoxazinon in das Lebewesen, und
(2) ggf. Wiederholung des Schrittes 1.
