Abstract
Die Erfindung betrifft die Verwendung eines Polynukleotids zur Modulation von Stammzellen. Erfindungsgemäß kodiert das Polynukleotid für a) ein EGFL7 Polypeptid oder für dessen gleichwirkende Polypeptidfragmente, oder b) ein Polypeptid, das mindestens 80 % Sequenzidentität zu den unter a) genannten Polypeptid oder Polypeptidfragmenten aufweist und welches zu dem EGFL7 Polypeptid gleichwirkend ist, oder c) ein Polypeptid, das mit einem unter a) und b) genannten Polypeptid oder Polypeptidfragment inhibierend in Wechselwirkung tritt.
Description
Verwendung von EGFL7 zur Modulation von Zellen
Die Erfindung betrifft die Verwendung von für das EGFL7 Polypeptid kodierenden Polynukleotiden, sowie die Verwendung von EGFL7 Polypeptiden und/oder von Modulatoren von EGFL7 zur Modulation von Stammzellen. Weiterhin betrifft die Erfindung entsprechende pharmazeutische Zusammensetzungen zur Modulation von Stammzellen.
Hintergrund der Erfindung
Der Entwicklung des Gehirns liegt ein genau reguliertes Zusammenspiel verschiedener Faktoren zu Grunde, die die Proliferation und Differenzierung von neuronalen Stammzellen im sich entwickelnden Embryo kontrollieren. Im Gegensatz dazu besitzt das adulte Gehirn ein lediglich eingeschränktes Potential zur Regeneration von Nervenzellen und Zellverluste können gewöhnlich nicht ausgeglichen werden. Dieser Befund wurde bis vor kurzem damit erklärt, dass das adulte Gehirn keine Stammzellen enthält, die eine Regeneration von verlorenen oder beschädigten Nervenzellen ermöglichen.
In den letzten Jahren konnte nachgewiesen werden, dass auch im adulten Gehirn von Säugetieren ausgehend von Stammzellen eine Neurogenese stattfindet und beschädigte Zellen ersetzt werden können. Weiterhin konnte gezeigt werden, dass in den Regionen des Gehirns, in denen Stammzellen lokalisiert worden sind, sogenannte Notch Rezeptoren exprimiert werden.
Die Notch Proteine umfassen eine Gruppe von membranständigen Rezeptoren, die Teil eines Signalweges in den Zellkern darstellen, mit dessen Hilfe die Expression bestimmter Gene reguliert wird. Die Notch Signalkaskade ist in allen bisher untersuchten Metazoen vorhanden. In Säugetieren werden vier verschiedene Notch Rezeptoren unterschieden, Notch 1 bis Notch 4.
Der vorliegenden Erfindung lag die Aufgabe zugrunde, Moleküle bereitzustellen, die die Modulation, also das Wachstum (Proliferation) und die Differenzierung von Stammzellen, insbesondere von neuralen Stammzellen beeinflussen.
Überraschender Weise wurde als ein derartiges Molekül EGFL7 identifiziert, dass aus der US 2003/0166907 Al unter dem Namen Zneul bekannt ist. In dieser Schrift wurde für EGFL7 gemutmaßt, dass es möglicher Weise mit dem Notch Rezeptor interagieren könnte (siehe Absätze [0096] bis [0098] der US 2003/0166907 Al). Belege für diese Spekulation gab es jedoch nicht. Insbesondere wurde die Verwendung von EGFL7 zur Modulation von Stammzellen nicht beschrieben.
Beschreibung der Erfindung
Die vorliegende Erfindung stellt die Verwendung eines isolierten Polynukleotids zur Modulation von Stammzellen bereit, wobei das Polynukleotid für mindestens eines der nachfolgend genannten Polynukleotide kodiert:
- Für ein EGFL7 Polypeptid gemäß SEQ ID NO 2 oder für beliebige Polypeptidfragmente des EGFL7 Polypeptids, die die gleiche Wirkung zeitigen, wie das EGFL7 Polypeptid.
- Für ein Polypeptid, welches mindestens 80% Sequenzidentität zu dem EGFL7 Polypeptid (SEQ ID NO 2) oder zu dessen gleichwirkenden Polypeptidfragmenten aufweist, wobei auch die Fragmente mit mindestens 80% Sequenzidentität eine zu dem EGFL7 Polypeptid gleiche Wirkung zeitigen.
- Für ein Polypeptid, dass mit einem der oben genannten Polypeptide inhibierend in Wechselwirkung treten kann.
Das Polynukleotid gemäß SEQ ID NO 1 kodiert für das EGFL7 Polypeptid.
Die Wirkung des EGFL7 Polypeptides ergibt sich für den Fachmann insbesondere aus den in den Beispielen dargestellten Versuchsergebnissen und der hierin enthaltenen Darstellung, insbesondere in Bezug auf das Protein Notch. Der Begriff „gleichwirkend" umfasst dabei alle zur Wirkung von EGFL7 qualitativ ähnlichen Ausprägungen, ohne dabei auf eine bestimmte Quantität in der Wirkung beschränkt zu sein. Ein Polypeptid tritt mit einem anderen Polypeptid „inhibierend in Wechselwirkung", wenn es das andere Polypeptid an der Bindung oder Aktivierung oder Inaktivierung eines weiteren Proteins hindert.
In einer bevorzugten Ausgestaltung der Erfindung beträgt die oben genannte Sequenzidentität zu dem EGFL7 Polypeptid oder zu dessen gleichwirkenden Polypeptidfragmenten 85 %, 88 % oder 90 %, besonders bevorzugt 95 % zu dem EGFL7 Polypeptid oder dessen Fragmenten.
In einer bevorzugten Ausgestaltung der erfindungsgemäßen Verwendung umfasst das inhibierende Polypeptid eine EGF-ähnliche Domäne oder ein Fragment davon. Insbesondere kann das inhibierende Polypeptid eine EGF-ähnliche Domäne eines Notch Rezeptors oder ein Fragment davon umfassen. Bevorzugter Weise ist das inhibierende Polypeptid ein Polypeptid gemäß SEQ ID NO 7 bis 10.
Die Modulation der Stammzellen bezieht sich insbesondere auf deren Wachstum (Proliferation) und/oder deren Differenzierung. Bevorzugter Weise sind die Stammzellen neurale Stammzellen des zentralen und/oder peripheren Nervensystems.
Die vorliegende Erfindung stellt weiterhin die Verwendung eines isolierten Polypeptids und/oder eines Modulators zur Modulation von Stammzellen bereit, insbesondere von neuralen Stammzellen des zentralen und/oder peripheren Nervensystems, wobei das Polypeptid ausgewählt ist aus der Gruppe bestehend aus:
Einem EGFL7 Polypeptid gemäß SEQ ID NO 2 und einem beliebigen Polypeptidfrag- ment dieses EGFL7 Polypeptids, wobei alle Polypeptidfragmente in Bezug auf das EGFL7 Polypeptid die gleiche Wirkung zeitigen.
Polypeptide die zu den genannten EGFL7 Polypeptid oder dessen genannten Polypeptidfragmenten mindestens eine Sequenzidentität von 80 %, bevorzugt von 85 % bzw. von 90%, besonders bevorzugt von 95%, aufweisen und welche in Bezug auf das EGFL7 Polypeptid die gleiche Wirkung zeitigen. Dies umfasst insbesondere die Bindung von EGFL7 an Notch und die Aktivierung der Signalkaskade.
- Ein Polypeptid, das mit einem der oben genannten Polypeptide inhibierend in Wechselwirkung treten kann. Dadurch wird insbesondere die Aktivierung der Notch Signalkaskade verhindert.
Der genannte Modulator ist ausgewählt aus einer Gruppe bestehend aus Aktivatoren und Inhibitoren der EGFL7 Polypeptidwirkung.
Um den Notch Rezeptor in ausreichender Weise zu aktivieren, sollen die Polypeptidfragmente von EGFL7 eine Länge von 10 bis 240 Aminosäurereste aufweisen. Bevorzugt sind Polypeptidfragmente mit einer Länge von 20 bis 200 Aminosäureresten, besonders bevorzugt sind Polypeptidfragmente mit einer Länge von 50 bis 150 Aminosäureresten.
Wie Versuche gezeigt haben, wird die Wirkung des EGFL7 Polypeptids auf den Notch Rezeptor insbesondere über bestimmte Domänen des EGFL7 Polypeptids vermittelt (vgl. Fig. 3 und die Beschreibung hierzu). Damit ein erfindungsgemäßes Polypeptidfragment in ausreichender Weise mit dem Notch Rezeptor interagiert und eine entsprechende Signalkaskade auslöst, ist es daher von Vorteil, wenn die verwendeten Polypeptidfragmente bestimmte Domänen umfassen.
Eine ausreichende Aktivierung des Notch Rezeptors erfolgt insbesondere durch solche Fragmente des EGFL7 Polypeptids, welche die die Bindung zum Notch Rezeptor vermittelnde EMI Domäne umfassen (AS 27 bis 104 des humanen EGFL7, SEQ ID NO 2). In dieser EMI- Domäne ist die Sequenz Y R T I Y R T A Y R R S P G enthalten (AS 64 bis 78 des humanen EGFL7, SEQ ID NO 2), die mit der in bekannten Notch Liganden enthaltenen Sequenz (Y)YYG korreliert. Demgemäß sollte die EMI-Domäne die Sequenz YxxxYxxxYxxxxG aufweisen, wobei x für jede beliebige geeignete Aminosäure steht.
Vorteilhafter Weise können die folgenden Polypeptidfragmente des EGFL7 Polypeptids zur Inhibierung des Notch Rezeptors verwendet werden: das Signalpeptid (Aminosäuren (AS) 1 bis 23 von EGFL7, SEQ ID NO 3), die EMI-Domäne (AS 27 bis 104 von EGFL7, SEQ ID NO 4), die EGF-ähnliche Domäne 1 (AS 103 bis 135 von EGFL7, SEQ ID NO 5) oder die EGF-ähnliche Domäne 2 (AS 137 bis 177 von EGFL7, SEQ ID NO 6).
Zur Inhibierung von EGFL wird vorteilhafter Weise die EGF-ähnliche Domäne 9-12 der Notch Rezeptoren 1, 2, und 4 (SEQ ID NO 7, 8, 10) und/oder die EGF-ähnliche Domäne 8-11 des Notch Rezeptors 3 (SEQ ID NO 9) verwendet. Die jeweilige EGF-ähnliche Domäne des Notch Rezeptors bindet an EGFL7 und verhindert somit seine Wechselwirkung mit dem Notch Rezeptor. Dadurch kann der Notch Rezeptor von anderen Liganden aktiviert werden.
Weiterhin kann zur Aktivierung von EGFL7 ein Aktivator verwendet werden. Derartige Aktivator sind Zytokine, insbesondere solche, die ausgewählt sind aus der Gruppe bestehend
aus vasculären endothelialen Wachstumsfaktoren (VEGF), Fibroblasten Wachstumsfaktoren (FGF), Thrombocyten-abgeleiteten Wachstumsfaktoren (PDGF). Auch Chemokine, Inter- leukine und Hypoxia gehören zu den Aktivatoren.
Im Gegensatz zu den membranständigen Liganden des Notch Rezeptors Jagged 1 und Jag- ged 2, die bekannter Weise eine in Bezug auf den Notch Rezeptor aktivierende Wirkung haben, wirkt das lösliche EGFL7 auf den Notch Rezeptor inhibierend. Um die inhibierende Wirkung des EGFL7 Polypeptids zu vermindern bzw. auszuschalten, kann ein geeigneter Inhibitor von EGFL7 verwendet werden.
Als derartige geeignete Inhibitoren kommen monoklonale oder polyklonale Antikörper in Frage, deren Herstellungsweise dem Fachmann bekannt ist. Ebenfalls in Frage kommen siR- NA Moleküle. Zur Inhibition kann beispielsweise die cDNA Sequenz des EGFL7 (SEQ ID NO 1) in einen geeigneten Adenovirus kloniert werden und als antisense Molekül exprimiert werden (vgl. Figuren 7 und 8). Ebenso können antisense Peptide und Antagonisten als Inhibitoren verwendet werden, beispielsweise kann durch Notch aktivierende Moleküle ein Verringerung der Expression von EGFL7 erreicht werden.
Wie die in den Beispielen gezeigten Versuche belegen (siehe z. B. Figur 12), führt die Aktivierung der Notch Signalkaskade zur vermehrten Differenzierung der neuralen Stammzellen in Astrozyten, wohingegen die Ausdifferenzierung von Neuronen und Oligodendrozyten vermindert wird. Demgemäß kann die erfindungsgemäße Verwendung zur Steuerung der Bildung von Neuronen und/oder Oligodendrocyten genutzt werden, und zwar sowohl in vivo als auch in vitro.
Die erfindungsgemäße Verwendung kann zur Therapie von einer Reihe von Erkrankungen des Nervensystems, insbesondere zu Therapie degenerative Erkrankungen eingesetzt werden. Solche Erkrankungen schließen Demenz, Morbus Alzheimer, Morbus Parkinson, Multiple Sklerose, Morbus Creutzfeldt- Jakob, die Lewy-Körperchen-Erkrankung, die Lobärathrophie, Chorea-Huntington und Morbus Wilson mit ein.
Ebenso können die genannten Stoffe zur Behandlung oder zur Stimulation der Neurogenese bei Verletzungen oder Schädigungen des Nervensystems verwendet werden. Darunter fallen
beispielsweise der Schlaganfall, die Querschnittslähmung, sowie die Schädigung durch Stoffwechselerkrankungen, Nervengifte oder HIV.
Ebenfalls ist es möglich, erfindungsgemäße Verwendung zur Manipulation von nicht pathologischen Vorgängen, etwa zur Stimulation von Lern- und Gedächtnisprozessen einzusetzen.
Die Verwendung der genannten Stoffe umfaßt auch die Manipulation von neuralen Stammzellen in vitro, wie beispielsweise bei der Stammzelltherapie.
Die vorliegende Erfindung umfaßt ebenfalls pharmazeutische Zusammensetzungen zur Modulation von Stammzellen, insbesondere neuraler Stammzellen des zentralen und/oder peripheren Nervensystems. Solche pharmazeutischen Zusammensetzungen enthalten ein Polynukleotid gemäß der obigen Beschreibung.
Solch ein Polynukleotid kann in Form eines Plasmids, Cosmids oder eines Virus vorliegen und mittels bekannter Transfektionsverfahren, wie der Lipofektion oder des ballistischen Transfers in eine Zelle eingeführt werden.
Die pharmazeutische Zusammensetzung kann auch ein Polypeptid und/oder einen Modulator gemäß der obigen Beschreibung enthalten.
Die vorliegende Erfindung betrifft auch ein Verfahren zur Modulation von Stammzellen. Bei einem derartigen Verfahren werden neurale Stammzellen mit einem Polypeptid und/oder einem Modulator in ihrer Differenzierung beeinfmsst. Das dabei verwendete Polypeptid ist ausgewählt aus der Gruppe bestehend aus: a) einem EGFL7 Polypeptid (SEQ ID NO 2), und dessen gleichwirkenden Polypeptidfrag- mente, b) Polypeptiden, die mindestens 80 % Sequenzidentität zu den unter a) genannten Polypeptid oder Polypeptidfragmenten aufweist und welche zu dem EGFL7 Polypeptid gleichwirkend sind, und c) Polypeptiden, die mit einem unter a) und b) genannten Polypeptid inhibierend in Wechselwirkung treten; und wobei der Modulator ausgewählt ist aus der Gruppe bestehend aus Aktivatoren und Inhibitoren der EGFL7 Polypeptid- Wirkung.
Die vorliegende Erfindung stellt weiterhin ein Verfahren zur Differenzierung von neuralen Stammzellen in Neuronen und/oder Oligodendrozyten bereit. Das Verfahren zeichnet sich durch die Gabe des EGFL7 Polypeptids oder dessen Aktivierung aus. Die Aktivierung von EGFL7 kann dabei in einer der oben beschriebenen und sich aus den Beispielen ergebenden Arten erfolgen.
Dem gegenüber wird mit einem Verfahren, bei dem das EGFL7 Polypeptid reduziert oder inhibiert wird, die Differenzierung von neuralen Stammzellen in Astrozyten erreicht. Die Reduktion oder Inhibition von EGFL7 kann dabei in einer der oben beschriebenen und sich aus den Beispielen ergebenden Arten erfolgen.
Die beschriebene Erfindung bezieht sich auf die Modulation von Stammzellen aus Säugetieren, insbesondere aus Stammzellen von Mäusen, Ratten, Affen und Menschen.
Die erfindungsgemäß modulierten Stammzellen können im Rahmen einer Therapie einem Patienten verabreicht bzw. in den Körper eines Patienten eingebracht werden. Dieser Patient kann ein Säugetier sein, insbesondere eine Maus, eine Ratte, ein Affe oder ein Mensch.
Insbesondere ist ein Verfahren zum Klonen von menschlichen Lebewesen oder ein Verfahren zur Veränderung der genetischen Identität der Keimbahn des menschlichen Lebewesens in einer Ausfuhrungsform der Erfindung nicht umfasst.
Die Erfindung betrifft weiterhin einen Kit zur Durchführung einer Verwendung oder zur Durchführung eines Verfahrens der hier beschriebenen Art. Ein derartiger Kit umfasst insbesondere einen der hier beschriebenen Stoffe zur Modulation von Stammzellen.
Die Erfindung wird nun unter Verweis auf die Beispiele anhand der Figuren näher erläutert.
Es zeigen:
Figur 1 : Mittels eines Yeast Two-Hybrid Systems, bakterieller EGFL7-GST Fusionsproteine (GST pull-down Assays) und Koimmunopräzipitationen konnte gezeigt werden,
dass EGFL7 an Notch Rezeptoren bindet. In diesem Experiment bindet EGFL7 an Notch 1 bis 3, nachdem diese Proteine in HEK293 Zellen überexprimiert wurden.
Figur 2: Die Interaktion von EGFL7 mit Notch 1 bis Notch 3wurde mit endogen exprimier- ten Rezeptoren verifiziert, nachdem rekombinant gereinigtes EGFL7 (His-EGFL7) ins Zellmedium appliziert wurde. Nach Koimmunopräzipitation wurde eine Interaktion zwischen Notch 1 bis Notch 3 und EGFL7 festgestellt (HUVEC: humane Endothelzellen, MRC5: humane Fibroblasten, hAoSMC: humane glatte Muskelzellen).
Figur 3: Die Analyse der Bindungsstelle in EGFL7 durch verschiedene EGFL7-GFP Fusionsproteine zeigte, dass die EMI-Domäne von EGFL7 eine besonders starke Bindung an Notch Rezeptoren vermittelt und dass die EGF-ähnlichen Domänen 1 und 2 zur Bindung beitragen können, aber alleine schwächer binden. Alle drei Domänen zusammen vermitteln die Bindung an Notch Rezeptoren, was auch im Yeast Two-Hybrid System und durch GST pull-down Assays gezeigt werden konnte. Außerdem ist das Signalpeptid (SS) von EGFL7 für eine optimale Bindung an Notch Rezeptoren wichtig. Vermutlich werden EGFL7 und Notch innerhalb der Zelle über den gleichen Syntheseweg transportiert, wo sie sich treffen und inter- agieren.
Figur 4: Durch Koimmunopräzipitation von EGFL7 und Notch-GFP Fusionsproteinen konnte nach Überexpression in HEK293 Zellen gezeigt werden, dass EGFL7 innerhalb der Notch Rezeptoren an die EGF-ähnlichen Domänen 9-12 (in Notch 1, 2 und 4) bzw. 8-11 (in Notch 3) bindet. Dieser Bereich ist für die Aktivierung von Notch Rezeptoren besonders wichtig.
Figur 5: Messung der Aktivierung von Notch Rezeptoren durch Aktivierung von Reportergenen (Luziferase, angeschaltet durch TPl, Hesl und Hes5 Promotoren), sowie Western Blots spezifisch für aktiviertes Notch 1, zeigte, dass EGFL7 ein Antagonist für die durch die bekannten Notch Agonisten Jagged 1 und 2 induzierte Notch 1 Aktivierung ist. Dies geht mit der Tatsache konform, dass bekannte Notch Agonisten membranständige Moleküle sind und lösliche Notch-Liganden als Antagonisten wirken.
Figur 6: Die Selbsterneuerung von neuralen Stammzellen in Kultur in Form von sogenannten Neurosphären hängt von einer intakten Interaktion von Jagged 1 und Notch 1 ab. Figur 5 zeigt, dass dieses Interface mit EGFL7 gestört wird. Konsequenterweise führt die Expression von EGFL7 in Neurosphären (nach Infektion mit verschiedenen EGFL7 kodierenden Adenoviren) zu einer Reduktion der Selbsterneuerung von neuralen Stammzellen (gemessen als sekundär gebildete Sphären %). Knockdown von endogenem EGFL7 mit einem antisense Konstrukt (EGFL7 as) hat einen gegenteiligen Effekt. Chemische Inhibition von Notch Signalen erfolgt durch den gamma-Sekretase Inhibitor DAPT (N-[N-(3,5-Difluorophenacetyl)-L-alanyl]- S-phenylglycine-t-butyl-ester.
Figur 7: Weitere Untersuchungen zeigten, dass der in Fig. 6 gezeigte Befund auf eine reduzierte Proliferation (gemessen durch BrdU-Inkorporation in die DNA von proliferierenden Zellen) der neuralen Stammzellen zurückzuführen ist. Die Apoptose und Nekrose von infizierten neuralen Stammzellen war nicht verändert, so dass ein toxischer Effekt von EGFL7 oder der Adenoviren ausgeschlossenen werden kann.
Figur 8: Infektion von neuralen Stammzellen mit adenoviralen Konstrukten und Messung der spezifischen Aktivierung der Notch 1 Signalgebung durch die quantitative PCR des spezifischen Genproduktes Hes5 zeigte, dass EGFL7 auch im Neurosphä- renmodell als Antagonist der Notch Signalgebung wirkt.
Figur 9: Mit den identischen Proben aus Fig. 8 und quantitativer PCR von murinem EGFL7 (mEGFL7, SEQ ID NO 11) in den Neurosphären zeigte sich im Umkehrschluß, dass Notch Signale EGFL7 herunterregulieren. Wird das Notch Signal durch den Inhibitor DAPT blockiert, steigt die EGFL7 Produktion um den Faktor 5,5 an. Dies impliziert einen negativen Feedback Loop, in dem Notch Rezeptoren die Produktion des eigenen Inhibors blockieren.
Figur 10: Quantitative PCR von murinem EGFL7 in verschiedenen Geweben des Mäusehirns zeigt eine große Menge an EGFL7 RNA im Hippocampus und im Kortex. In der Subventrikulärzone (SVZ), in der neurale Stammzellen gebildet werden, sinkt die Menge jedoch stark ab und ist in aus der SVZ präparierten Neurosphären,
kaum noch zu detektieren. Da aus den vorhergehenden Daten hervorgeht, dass EGFL7 die Bildung von neuralen Stammzellen herunterreguliert, erscheint es stimmig, dass das Molekül in Bereichen des Hirns weniger produziert wird, wo neurale Stammzellen proliferieren sollen. Da außerdem das Überleben der Neuro- sphären in Kultur von intakter Selbsterneuerung abhängt, sollte die Konzentration von EGFL7 dort besonders gering sein, was auch der Fall ist. Dieses Ergebnis indiziert, dass die vorhergehenden Beobachtungen physiologisch relevant sind. (EOMA: Positivkontrolle, viel mEGFL7; Fibroblasten MEFs3 NIH 3T3, wenig mEGFL7; M6363 Maus-Adenokarzinom, Negativkontrolle, kein mEGFL7).
Figur 11 : Aus der Literatur ist das gezeigte Konzept bekannt (Louvi und Artavanis- Tsakonas. Notch signalling in vertebrate neural development. Nat Rev Neurosci. 2006, 7(2): 93-102), welches auch auf die Differenzierung von neuralen Stammzellen in Neurosphären übertragbar ist. Ein erhöhtes Notch Signal führt unter Differenzierungsbedingungen zur Bildung von mehr Astrozyten und weniger Neuronen sowie weniger Oligodendrozyten.
Figur 12: Das Konzept gemäß Fig. 11 wurde wie folgt untersucht. Coverslips wurden mit 10 μg/ml bovinem Serumalbumin (linker Balken eines Grautons) oder 10 μg/ml rekombinant hergestelltem EGFL7 (jeweiliger rechter Balken eines Grautons) beschichtet. Junge Neurosphären wurden auf die so beschichteten Coverslips ausgebracht und zur Differenzierung angeregt. Nach Färbung spezifischer Marker wurde ausgezählt, wie viele Astrozyten (Marker GFAP), wie viele Neuronen (Marker NF 160) und wie viele Oligodendrozyten (Marker 04) sich gebildet haben. Die auf EGFL7 beschichteten Coverslips differenzierten neuralen Stammzellen bildeten signifikant mehr Neuronen und Oligodendrozyten, aber weniger Astrozyten. Dieses Experiment zeigt, dass EGFL7 die Differenzierung von neuralen Stammzellen durch Inhibition der Notch Signalgebung beeinflußt.

Zusammenfassung
Unter Verwendung eines Hefe 2-Hybrid System Screens konnten gezeigt werden, dass EGFL7 in Notch 3 an die extrazelluläre Region bindet, die für die Aktivierung des Rezeptors benötigt wird (die so genannte DSL-Domäne). Nach Koimmunopräzipitation von endogenem und rekombinantem Notch, beziehungsweise der DSL-Domänen aus Notch, konnten verifiziert werden, dass EGFL7 an alle vier Notch Rezeptoren bindet. Ebenfalls wurde herausgefunden, dass in EGFL7 die sogenannte EMI-Domäne die Bindung vermittelt und dass die auf die EMI-Domäne folgenden beiden EGF-ähnlichen Domänen die Bindung verstärken.
In der EMI-Domäne wurde die Sequenz YxxxYxxxYxxxxG (Y = Tyrosin, G = Glyzin, x = beliebige Aminosäure) ermittelt, welche die Bindung von EGFL7 und Notch Rezeptoren wahrscheinlich vermittelt. Eine strukturell ähnliche Sequenz YYYG, gefolgt von EGF- ähnlichen Domänen, ist bereits aus existierenden Notch-spezifischen Liganden (z. B. Jagged 1) bekannt. Die modifizierte Sequenz in EGFL7 war bisher allerdings unbekannt. Die Quantifizierung der Bindung voll EGFL7 an Notch 1 unter Verwendung der Biacore-Technik ergab eine Dissoziationskonstante von größer 150 nmol/1, was einer starken Bindung entspricht. Nach Messung der spezifischen, durch Notch Rezeptoren induzierten Signaltransduktion durch sogenannte Reporter Gene Assays fanden wir, dass die durch Jagged 1 und Jagged 2 induzierte Aktivierung von Notch Rezeptoren herabgesetzt wird.
Um diese Wirkung im biologischen System zu testen, wählten wir die Selbsterneuerung von neuralen Stammzellen. Diese sogenannten Neurosphären wurden aus Mäusehirnen isoliert und ausgewählt, da ihre Selbsterneuerung in Zellkultur von einer intakten Jagged 1 - Notch 1 Interaktion abhängt. Wird diese Bindung unterbrochen, erneuern sich die neuralen Stammzellen schlecht bis gar nicht im Neurosphären-Modell.
Da EGFE7 und Jagged 1 an dieselbe Region im Notch 1 binden und EGFL7 die Notch- induzierte Signaltransduktion inhibiert, wurden diese Neurosphären mit einem Adenovirus infiziert, der EGFL7 exprimiert. Es zeigte sich, dass in den Neurosphären, welche EGFL7 ex-
primierten, die Expression von Hes 5 heruntergesetzt war (einem spezifisch durch Notch 1 aktivierten Gen) und das diese Zellen sich signifikant schlechter selbst erneuerten.
Im Folgenden werden die durchgerührten Experimente mit Verweis auf die jeweiligen Figuren detailliert beschrieben:
Immunpräzipitationsstudien ('s. Figur 1)
HEK293 Zellen wurden transfiziert und nach zwei Tagen in eiskaltem Triton X-100 Lysepuf- fer lysiert (50 mmol/1 HEPES (pH 7,4), 150 mmol/1 NaCl, 1 % Triton X-100, 10 % Glyzerin, 1.5 mmol/1 MgC125 1 mmol/1 EDTA, 1 mmol/1 EGTA, 1 mmol/1 Natriumorthovanadat, 20 mmol/1 NaF, 1 mmol/1 PMSF, 10 μg/ml Aprotinin, 2 μg/ml Leupeptin, und 10 μmol/1 ZnCl2). Die Lysate wurden bei 4 °C durch Zentrifugation bei 16200 g für 20 Minuten gereinigt und dann für zwei Stunden bei 4 °C unter Rotation mit Antikörper inkubiert. Protein- Antikörper Komplexe wurden durch Inkubation mit Protein A-Agarose (Roche) für weitere zwei Stunden bei 4 0C gesammelt, dann zentrifugiert und drei Mal mit Lysepuffer gewaschen. Anschließend wurden Western blot Analysen durchgerührt. Die Präzipitate wurden bei 95 °C fünf Minuten lang gekocht und die Proteine durch Standard SDS-PAGE getrennt. Daraufhin wurden die Proteine auf eine Nitrozellulose-Membran geblottet und mit den angezeigten Antikörpern inkubiert. Gebundene Antikörper-Komplexe wurden durch Meerrettich Peroxidase- gekoppelte Sekundärantikörper detektiert und durch ECL Substrat (Santa Cruz Biotechnolo- gy) visualisiert.
Immunpräzipitationsstudien (s. Figur 2)
Frisch präparierte HUVEC Zellen aus Nabelschnurvenen waren eine gute Quelle für endogenen Notch 1 Rezeptor sowie Jagged 1 and 2. Humane MRC5 Fibroblasten enthielten beträchtliche Mengen an Notch 2 Rezeptor und humane glatte Muskelzellen aus Aorta lieferten detek- tierbare Mengen an Notch 3. Rekombinantes humanes EGFL7 gereinigt aus Sf9 Insektenzellen wurde im Medium bei einer Konzentration von 1 μg/ml appliziert und nach der Zelllyse wurden Koimmunopräzipitationsstudien (siehe oben) mit spezifischen Antikörpern durchgeführt. Anti-Erk2 Antikörper diente als Negativkontrolle, da er wie die anderen Antikörper, die in diesem Experiment verwendet wurden, polyklonal sowie von Santa Cruz Biotechnology war und aus dem Kaninchen stammte, allerdings gegen ein intrazelluläres Kontrollprotein ge-
richtet ist. Unter diesen Bedingungen assoziierte EGFL7 mit den getesteten Notch Rezeptoren und Jagged 2 aber nicht mit Jagged 1.
Analyse der Bindungsstellen in EGFL7 (s. Figur I)
Um die Bindungsstelle in EGFL7 an Notch Rezeptoren zu bestimmen wurden verschiedene EGFL7 Konstrukte mit GFP fusioniert und zusammen mit Notchl in HEK293 Zellen übe- rexprimiert. Analyse siehe Fig. 1.
Analyse der Bindungsstellen in Notch (s. Figur 1)
Um die Bindungsstelle in Notch Rezeptoren an EGFL7 zu bestimmen wurden die extrazellulären EGF-Domänen, die für die Aktivierung von Notch Rezeptoren verantwortlich sind, mit GFP fusioniert und zusammen mit EGFL7 in HEK293 Zellen überexprimiert. Analyse siehe Fig. 1.
Reportergen Experimente (s. Figur 5)
Transaktivierung von Notch-sensitiven Elementen wurde wie von Kurode et al. 2003 beschrieben durchgeführt. HEK293 Zellen wurden mit den angezeigten Plasmiden plus 200 ng 12xCBFl TPl -Leuchtkäfer Luziferase Reportergen Plasmid pGa981-6 plus 50 ng pSV-ß-gal Kontroll Plasmid (von Ursula Zimber-Strobl und Lothar Strobl; es kann aber auch ein anderes dem Fachmann bekanntes Plasmid verwendet werden) durch Lipofectamin 2000 (Invitrogen) transfiziert. 24 Stunden nach Transfektion wurden die Zellen in SDS-freiem Lysepuffer Iy- siert und die intrazelluläre Luziferase-Konzentration mit dem Chemiluminscent Luciferase Reporter Gene Assay (Roche) gemessen. Ebenso wurde die ß-Galaktosidase Aktivität in den Zellextrakten mit dem Chemiluminescent ß-gal Reporter Gene Assay (Roche) bestimmt. Die Proben wurden in einem Victor3 microplate reader (Perkin Eimer) analysiert.
Kulturen und Assavs mit Neurosphären (s. Figuren 6 und 7)
Primäre Kulturen wurden aus erwachsenen C57bl6 Mäusen (6 bis 8 Wochen alt) durch Präparation der lateralen Wand des lateralen Ventrikels präpariert. Gewebe wurde enzymatisch durch 0,8 mg/ml Papain (Worthington) in Leibovitz L- 15 Medium mit 400 U DNase I und
0,5 mmol/1 EDTA dissoziiert. Zellen wurden in Neurosphären Medium (DMEM/Hams-F12 1:1 + Glutamax, B27 Supplement, 10 mmol/1 HEPES (pH 7,4) und 2 mmol/1 L-Glutamin) mit 20 ng/ml EGF, 20 ng/ml FGF, 100 U/ml Penizillin und 100 μg/ml Streptomizin unter Standardbedingungen kultiviert.
Zur Bestimmung des Selbsterneuerungspotentials der neuralen Stammzellen wurden drei Tage alte Sphären mit verschiedenen Adenoviren oder dem Notch Signalinhibitor DAPT (N-[N- (3,5-Difluorophenacetyl)~L-alanyl]-S-phenylgrycine t-butyl Ester) inkubiert. Danach wurden die Zellen dissoziiert und bei klonaler Dichte (100 Zellen/cm2 oder 0,5 Zellen/μl) ausgesät. Nach fünf Tagen wurden die gebildeten Neurosphären gezählt (Singec et al., 2006). Proliferation wurde durch Inkorporation von Bromdesoxiuridin (BrdU) für zwölf Stunden und FACS Analyse bestimmt (FACS-Calibur, Beckton-Dickinson).
Konstruktion adenoviraler Expressionsvektoren
Human und Maus EGFL7 sowie korrespondierende anti-sense Sequenz wurden in das Trans- ferplasmid pDC515 (von Andreas Simm) kloniert. Es kann aber auch ein anderes dem Fachmann bekanntes Transferplasmid verwendet werden. Infektiöse Partikel wurden durch Ko- transfektion mit dem adenoviralen pBHGfrt(del)El,3FLP in Ela-transformierte HEK293 Zellen (Bett et al., 1994) kreiert. Nach 14 bis 20 Tagen wurden die Partikel gesammelt und titriert (1 pfu pro 500 Partikel).
Quantitative RT-PCR (s. Figuren 8 bis IQ^)
RNA von Zelllinien und Neurosphären wurden mit Trizol (Invitrogen) isoliert. RNA von Gewebeproben wurde mit dem RNeasy Kit (Qiagen) isoliert. cDNA (SEQ ID NO 11) wurde von 1 μg RNA pro Probe durch das Thermoscript RT-PCR System (Invitrogen) synthetisiert. Für quantitative real-time PCR wurden folgende Primer verwendet (Nyfeler et al., 2005):
Maus RNA-Polymerase II

5'-AAGTAC CGT GGC GGT GGA GAT GC-3' (forward) (SEQ IDNO 16) und
5'-CGC TGGAAG TGGTAAAGC AGC TT-3' (reverse) (SEQ IDNO 17).
Real-time PCR wurde mit iQ SYBR Green Supermix (Bio-Rad) durchgeführt. Resultate wurden zu RNA Polymerase II mit iQ5 Optical System Software (Bio-Rad) normalisiert.
Differenzierung von Neurosphären (Fig. 12)
Frisch dissoziierte Neurosphären-Zellen wurden auf Objektträger 10 μg/ml BSA oder rekom- binantem EGFL7 überführt. Dann wurden die Zellen zwölf Tage in Neurosphären-Medium plus 1 % FCS ohne EGF und FGF inkubiert. Zum Schluß wurden die Zellen in 4 % PFA 10 Minuten bei RT fixiert und mit den Zelltyp-spezifischen Markern anti-GFAP (Astrozyten), anti-beta(III)tubulin and anti-NF 160 (Neuronen) und anti-O4 (Oligodendrozyten) gefärbt. Danach wurde die Verteilung der einzelnen Zelltypen durch Zählung in 15 unabhängigen Sektoren unter dem Fluoreszenzmikroskop bestimmt.
Claims
1. Verwendung eines Polynukleotids zur Modulation von Stammzellen, dadurch gekennzeichnet, dass das Polynukleotid kodiert für a) ein EGFL7 Polypeptid oder für dessen gleichwirkende Polypeptidfragmente, oder b) ein Polypeptid, dass mindestens 80 % Sequenzidentität zu den unter a) genannten Polypeptid oder Polypeptidfragmenten aufweist und welches zu dem EGFL7 Polypeptid gleichwirkend ist, oder c) ein Polypeptid, das mit einem unter a) und b) genannten Polypeptid oder Polypeptidfragment inhibierend in Wechselwirkung tritt und eine EGF-ähnliche Domäne oder ein Fragment davon umfasst.
2. Verwendung gemäß Anspruch 1, wobei die EGF-ähnliche Domäne von einem Notch Rezeptor stammt.
3. Verwendung gemäß Anspruch 1, wobei das inhibierende Polypeptid ein Polypeptid gemäß SEQ ID NO 7 bis 10 ist.
4. Verwendung gemäß einem der vorangehenden Ansprüche, zur Modulation des Wachstums und/oder der Differenzierung von Stammzellen.
5. Verwendung gemäß einem der vorangehenden Ansprüche, wobei die Stammzellen neurale Stammzellen des zentralen und/oder peripheren Nervensystems sind.
6. Verwendung eines Polypeptids und/oder eines Modulators zur Modulation von Stammzellen, wobei das Polypeptid ausgewählt ist aus der Gruppe bestehend aus: a) EGFL7 Polypeptid, und dessen gleichwirkende Polypeptidfragmente, b) einem Polypeptid, das mindestens 80 % Sequenzidentität zu dem unter a) genannten Polypeptid oder Polypeptidfragmenten aufweist und welche zu dem EGFL7 Polypeptid gleichwirkend sind, und c) einem Polypeptid, das mit einem unter a) und b) genannten Polypeptid oder Polypeptidfragment inhibierend in Wechselwirkung tritt; wobei der Modulator ausgewählt ist aus der Gruppe bestehend aus:
Aktivatoren der EGFL7 Polypeptid- Wirkung und Inhibitoren der EGFL7 Polypeptid-
Wirkung.
7. Verwendung gemäß Anspruch 6, wobei die Polypeptidfragmente ein Signalpeptid, eine EMI-Domäne und/oder eine EGF-ähnliche Domäne aufweisen.
8. Verwendung gemäß Anspruch 6 oder 7, wobei die Polypeptidfragmente 10 bis 240 Aminosäurereste aufweisen.
9. Verwendung gemäß Anspruch 6, wobei die inhibierenden Polypeptide eine EGF- ähnliche Domäne oder ein Fragment davon umfassen.
10. Verwendung gemäß Anspruch 9, wobei die inhibierenden Polypeptide eine EGF- ähnliche Domäne eines Notch Rezeptors oder ein Fragment davon umfassen.
11. Verwendung gemäß Anspruch 9 oder 10, wobei das inhibierende Polypeptide ein Polypeptid gemäß SEQ ID NO 7 bis 10 ist.
12. Verwendung gemäß einem der Ansprüche 6 bis 11, wobei die Stammzellen neurale Stammzellen des zentralen und/oder peripheren Nervensystems sind.
13. Verwendung gemäß einem der Ansprüche 6 bis 12, wobei der Aktivator ausgewählt ist aus der Gruppe bestehend aus VEGF, FGF, PDGF, Hypoxia, Chemokinen und Zytokinen.
14. Verwendung gemäß einem der Ansprüche 6 bis 13, wobei der Inhibitor ausgewählt ist aus der Gruppe der Antikörper, siRNA Moleküle, antisense Peptide und Antagonisten.
15. Verwendung gemäß einem der Ansprüche 6 bis 14, zur Steuerung der Bildung von Neuronen und/oder Gliazellen, insbesondere der Manipulation von Stammzellen aus Hirngewebe und/oder zur Neurogenesemodulation.
16. Verwendung gemäß einem der Ansprüche 6 bis 15, zur Therapie von Erkrankungen des Nervensystems, wie beispielsweise Demenz, Alzheimer, Parkinson, Multiple Sklerose, Creutzfeld- Jakob, Lewy-Köφerchen-Erkrankung, Lobärathrophie, Chorea Huntington und Morbus Wilson.
17. Verwendung gemäß einem der Ansprüche 6 bis 16, zur Behandlung oder Stimulation der Neurogenese bei Verletzungen oder Schädigungen des Nervensystems, wie beispielsweise Schlaganfall, Querschnittslähmung, Schädigung durch Stoffwechseler- krankungen, Nervengifte und HIV.
18. Verwendung gemäß einem der Ansprüche 6 bis 17, zur Manipulation von nichtpathologischen Vorgängen, wie Lern- oder Gedächtnisprozessen.
19. Pharmazeutische Zusammensetzung zur Modulation von Stammzellen, insbesondere neuralen Stammzellen des zentralen und/oder peripheren Nervensystems, enthaltend
A) ein Polynukleotid, wobei das Polynukleotid kodiert für aa) ein EGFL7 Polypeptid oder für dessen gleichwirkende Polypeptidfragmente, bb) ein Polypeptid, dass mindestens 80 % Sequenzidentität zu den unter a) genannten Polypeptid oder Polypeptidfragmenten aufweist und welches zu dem EGFL7 Polypeptid gleichwirkend ist, oder cc) ein Polypeptid, dass mit einem unter aa) und ab) genannten Polypeptid oder
Polypeptidfragment inhibierend in Wechselwirkung tritt und eine EGF- ähnliche Domäne oder ein Fragment davon umfasst, oder
B) ein Polypeptid und/oder einen Modulator, wobei das Polypeptid ausgewählt ist aus der Gruppe bestehend aus: dd) EGFL7 Polypeptid, und dessen gleichwirkende Polypeptidfragmente, ee) einem Polypeptid, das mindestens 80 % Sequenzidentität zu dem unter a) genannten Polypeptid oder Polypeptidfragmenten aufweist und welche zu dem EGFL7 Polypeptid gleichwirkend sind, und ff) einem Polypeptid, das mit einem unter dd) und ee) genannten Polypeptid oder
Polypeptidfragment inhibierend in Wechselwirkung tritt; und wobei der Modulator ausgewählt ist aus der Gruppe bestehend aus: Aktivatoren und Inhibitoren der EGFL7 Polypeptid- Wirkung.
20. Verfahren zur Modulation von Stammzellen, gekennzeichnet durch die Beeinflussung der Differenzierung von neuralen Stammzellen mit einem Polypeptid und/oder einem Modulator zur Modulation von Stammzellen.
21. Verfahren gemäß Anspruch 20, wobei die Modulation von Stammzellen eine Differenzierung von neuralen Stammzellen in Neuronen und Oligodendrozyten ist, gekennzeichnet durch die Gabe oder Aktivierung von EGFL7.
22. Verfahren gemäß Anspruch 20, wobei die Modulation von Stammzellen eine Differenzierung von neuralen Stammzellen in Astrozyten ist, gekennzeichnet durch die Reduktion oder Inhibierung von EGFL7.
23. Verfahren zur Therapie oder Prophylaxe eines Individuums, insbesondere eines Menschen, wobei eine nach Anspruch 21 bis 23 modulierte Stammzelle dem Individuum verabreicht wird.
24. Verfahren nach Anspruch 23, wobei die Therapie oder Prophylaxe degenerative und akute Erkrankungen umfasst, insbesondere Demenz, Morbus Alzheimer, Morbus Parkinson, Multiple Sklerose, Morbus Creutzfeldt-Jakob, die Lewy-Körperchen- Erkrankung, die Lobärathrophie, Chorea-Huntington, Morbus Wilson, Schlaganfall, Querschnittslähmung und die Schädigung durch Stoffwechselerkrankungen, Nervengifte und/oder HIV, sowie die Stimulation von Lern- und Gedächtnisprozessen.
