[0002] 1.血液流動性及びその測定方法
血管内の血液の流れやすさは、もちろん血管の収縮にも影響される力 血液自体 の流体としての特性に大きく影響される。血液は赤血球、白血球、血小板といった血 球成分と、さまざまなタンパク質や脂質、無機質などが含まれている血しょう成分と〖こ 分けることができる。血液の流動性、言い換えれば血液の粘度は、特に赤血球の容 積や変形能 (しなやかに変形して狭いところを通過する能力)、血しょうフイブリノーゲ ン、白血球、血小板などの影響が大きいと考えられている。現在は採取した血液の流 動性を測定する装置(MC- FAN: Micro Channel array Flow Analyzer)が開発されて おり、これにより、血液の流動性を測定することができる(非特許文献 1〜3)。

[0004] また、黒酢を長距離ランナー 10人に 20ml、 30日間、連日で飲用させ、飲用開始の 前日と最終飲用の翌日に血管機能検査と採血を行ったところ、全血通過時間、血管 内皮成長因子 (VEGF)と MCHCは黒酢の飲用により有意に低下し、 MCVは有意に増 大したことが報告されている。黒酢の赤血球変形能の亢進により微小循環が改善さ れ、黒酢による血液流動性の亢進は活動筋への血流量、すなわち酸素輸送量を増 加させ、持久性運動能力の向上に寄与すると考えられたと説明されている (非特許文 献 5)。
[0005] 更に、特許文献 1には、 L-グルタミン 8〜27質量部、 L-アルギニン 6〜21質量部、 L- ロイシン 8〜25質量部、 L-イソロイシン 6〜20質量部及び L-パリン 6〜19質量部の 5種 のアミノ酸を必須成分として含有(L-グルタミン 0.34g、 L-アルギニン 0.34g、 L-口イシ ン 0.28g、 L-イソロイシン 0.18g、 L -パリン 0.18g、 L-リジン 0.14g、 L-メチォニン 0.12g、 L -スレオ-ン 0.18g、 L-ヒスチジン 0.10g、 L-プロリン 0.10g、 L-フエ-ルァラニン 0.01g、 L -トリブトファン 0.03gを含有)する経口アミノ酸組成物を、服用期間を 10週間として、 1 日 3回、毎食後に 1袋ずつ(必須成分である 5種のアミノ酸として 3.96g)摂取したところ 、血液流動性が改善されたことが記載されている。

[0008] 本願発明者らは、特定のアミノ酸組成物が、通常の健常者の経口摂取直後に、血 液粘性低下効果を示すことを見出して、本願発明を完成させた。

発明を実施するための最良の形態
[0012] 1.血液流動性又は血液粘度と疾病
血液粘度は心臓血管系疾患に対する危険因子であるとされている。動脈系では、 血液粘度が増すと、組織への血流量を維持するために血圧が上昇する。静脈系で は、血液粘度の増大に伴い、血流量が減れば、ズリ速度が低下するので血液粘度が
増して一層血流量が減る。このように血液粘度の上昇によって引き起こされる循環障 害を過粘性症候群 (hyperviscosity syndrome)といい、さまざまな疾患の底辺に横たわ つて ヽることが多!、と!/ヽわれて!/ヽる (日本生理学雑誌、第 66卷、 pp.234-244)。
[0013] 本発明の血流改善剤又は血液粘度低下剤及び血液流動性改善剤又は血液粘度 低下剤を含有する飲食品は、高血圧、血栓症をはじめとする心臓血管系、肩凝り、冷 え性、梗塞症、疲労、高脂血症、動脈硬化症などの末梢あるいは中心の血行障害- 血流障害による疾患を、血流流動性を改善することにより、予防及び/又は治療する ことができる。
[0014] なお、血液の流動性は例えば、液流動性測定装置(MC- FAN: Micro Channel Arra y Flow Analyzer,株式会社ェムシ一研究所)を用いて計測できる。本装置は半導体 微細加工技術を用いて加工されたシリコンチップ上の微細な溝と、ガラス基盤を圧着 してできた流路(マイクロチャネルアレイ)に血液を流すことにより、流路を流れる細胞 の状態を顕微鏡で直接、観察および記録するものである(同社ホームページ)。
[0015] 2.血液流動性又は血液粘度の改善
上記のように、血液粘度は心臓血管系疾患など種々の疾患に対する危険因子であ るから、近年増大する傾向にある心臓血管系疾患をはじめとする、これら疾患の治療 及び Z又は予防のためには、血液粘度を低下させる組成物又は血液流動性を改善 させる組成物の開発は有用である。血液粘度が増大する要因としては、赤血球の形 態変形能を低下、赤血球集合のなどが挙げられる。例えば、赤血球内の Caの蓄積と これに起因する膜タンパク質間相互作用の変化は、本態性高血圧症の一因であると いわれている。他方、血液粘度を低下させる要因として例えば、 NO生成物質が赤血 球変形能を増大させるといわれている(日本生理学雑誌、第 66卷、 No.9、 pp.287-297 ) oまた、前記のように、果実酸や黒酢を摂取することにより、スポーツ選手のスポーツ 中での血液粘性が低下することが報告されて 、る。
[0016] 3.アミノ酸の組成(以下、アミノ酸は 3文字コードで示すことがある。 )
本願発明には、スレオニン、プロリン、グリシン、ノ リン、イソロイシン、ロイシン、チロ シン、フエ二ルァラニン、リジン、メチォニン、トリプトファン、ヒスチジン及びアルギニン を含むアミノ酸組成物を有効成分として含有する血液流動性改善剤又は血液粘度
低下剤を包含する。
[0017] また、本願発明には、スレオニン、プロリン、グリシン、ノ リン、イソロイシン、ロイシン 、チロシン、フエ二ルァラニン、リジン、メチォニン、トリプトファン、ヒスチジン及びアル ギニンに加えて、ァスパラギン酸、セリン、グルタミン酸及びァラニンを含むアミノ酸組 成物を有効成分として含有する血液流動性改善剤又は血液粘度低下剤を包含する
[0018] 本願発明には、具体的には、スレオニン、プロリン、グリシン、パリン、イソロイシン、 ロイシン、チロシン、フエ二ルァラニン、リジン、メチォニン、トリプトファン、ヒスチジン及 びアルギニンからなるアミノ酸組成物を有効成分として含有する血液流動性改善剤 又は血液粘度低下剤を包含する。
[0019] また、本願発明には、具体的には、スレオニン、プロリン、グリシン、パリン、イソロイ シン、ロイシン、チロシン、フエ二ルァラニン、リジン、メチォニン、トリプトファン、ヒスチ ジン、アルギニン、ァスパラギン酸、セリン、グルタミン酸、及びァラニン力 なるアミノ 酸組成物を有効成分として含有する血液流動性改善剤又は血液粘度低下剤を包含 する。
[0020] 本願発明のアミノ酸組成物を構成する各アミノ酸は、遊離のアミノ酸でもよぐまた、 薬学上又は食品上許容される塩でもよい。例えば、各アミノ酸は、塩酸塩、乳酸塩な どであってもよい。
[0021] 本願アミノ組成物としては次のものをあげることができる。
[0022] 3- 1.スレオニン、プロリン、グリシン、ノ リン、イソロイシン、ロイシン、チロシン、フエ 二ルァラニン、リジン、メチォニン、トリプトファン、ヒスチジン及びアルギニンからなるァ ミノ酸組成物を有効成分として含有する血液流動性改善剤又は血液粘度低下剤で ある。
[0023] 本願発明には、有効成分として、スレオニン、プロリン、グリシン、パリン、イソ口イシ ン、ロイシン、チロシン、フエ二ルァラニン、リジン、メチォニン、トリプトファン、ヒスチジ ン及びアルギニン力もなるアミノ酸組成物を、アミノ酸としては重量比で、スレニオンを 2〜15モル、プロリンを 4〜30モル、グリシンを 7〜20モル、ノ リンを 4〜8モル、イソ口 イシンを 3〜9モル、ロイシンを 2〜12モル、チロシンを 1〜9モル、フエ二ルァラニンを
0.5〜5モル、リジンを 5〜11モルの割合で含有し、かつそれぞれ 5モル以下の割合で 、メチォニン、トリプトファン、ヒスチジン、アルギニンを含むことを特徴とする血液流動 性改善剤又は血液粘度低下剤が包含される。
[0024] 3- 2.スレオニン、プロリン、グリシン、ノ リン、イソロイシン、ロイシン、チロシン、フエ ニノレアラニン、リジン、メチォニン、トリプトファン、ヒスチジン、ァノレギニン、ァスパラギ ン酸、セリン、グルタミン酸、及びァラニンからなるアミノ酸組成物を有効成分として含 有する血液流動性改善剤又は血液粘度低下剤である。
[0025] 本願発明には、アミノ酸としては重量比で、スレニオンを 2〜 15モル、プロリンを 4〜 30モル、グリシンを 7〜20モル、ノ リンを 4〜8モル、イソロイシンを 3〜9モル、ロイシン を 2〜12モル、チロシンを 1〜9モル、フエ二ルァラニンを 0.5〜5モル、リジンを 5〜11 モルの割合で含有し、かつそれぞれ 5モル以下の割合で、メチォニン、トリプトファン 、ヒスチジン、アルギニンを含み、更に 1モル以下の割合のァスパラギン酸、 5モル以 下の割合のセリン、 4モル以下の割合のグルタミン酸、 12モル以下の割合のァラニン を含むアミノ酸組成物を有効成分とする血液流動性改善剤又は血液粘度低下剤が 包含される。
[0026] より具体的には、本願発明には、スレニオンが 6.92重量%、プロリンが 16.79重量%、 グリシンが 11.60重量%、ノ リンが 5.56重量%、イソロイシンが 4.79重量%、ロイシンが 6.53 重量%、チロシンが 6.93重量%、フエ-ルァラニンが 5.12重量%、リジンが塩酸リジンとし て 12.75重量%、メチォニンが 0.64重量%、トリプトファンが 3.63重量%、ヒスチジンが 3 .23重量0 /0、アルギニンが 4.96重量0 /0、ァスパラギン酸がァスパラギン酸 Naとして 0.22 重量%、セリンが 2.11量%、グルタミン酸が 3.81重量%、及びァラニンが 4.41重量%から なるアミノ酸組成物を有効成分とする血液流動性改善剤又は血液粘度低下剤が包 含される。
[0027] 4.本願発明の組成物の使用の形態
本願発明のアミノ酸組成物の使用形態は特に限定されないが、医療用としても、飲 食用、栄養補助飲食用としても用いることができる。
[0028] 医療用につ 、ては、経口投与、直腸投与、注射、輸液による投与等の一般的投与 経路により使用することができる。経口投与の場合には、上記組成物自体あるいは薬
学上許容される担体、賦形剤、希釈剤等とともに混合し、散剤、顆粒剤、錠剤、カブ セル剤、トローチ剤、シロップ剤等とすることもできる。また、注射剤としては、適当な 緩衝剤、等張剤等を添加し、滅菌蒸留水に溶解したものを用いればよい。
[0029] 本願発明の組成物は極めて安全であるので、その投与量は非常に広範に設定で き、更に、投与方法、使用目的により異なるが、通常 1回に 0.5〜5g、 1日投与量として l〜20g、好ましくは 5〜10gとすることができる。これらの溶液剤とする場合には、 0.5〜 10wt%溶液として 10〜1000ml、好ましくは 1〜4重量%溶液として 100〜400mlを 1回投 与量とすればよい。
[0030] 食用とする場合には、本願アミノ酸組成物は、そのまま食用できるが各種食品への 添加物とすることもできる。また、本願発明の組成物は水に溶解して飲料とすることも でき、その場合には、他の栄養成分、例えば、水溶液ビタミン類、タウリンなどを更に 添加することもできる。また、嗜好性の改善のために、塩ィ匕ナトリウムなどの塩類、タエ ン酸等の酸類及び Z又は他の適当な風味を加えて飲料とすることもできる。安定性 のために、 pH調整剤、キレート剤を更に添加することもできる。
実施例
[0031] [実施例 1]アミノ酸飲料 (VAAM)を飲用した際の血流改善効果の確認
試料としてアミノ酸飲料 (VAAM、 190g、明治乳業 (株))を用い、プラセボとして水(19 0 g)を用いた。アミノ酸飲料 1には、アミノ酸として、 AspNa-H 0が 7mg、 Thrが 207mg、

Serが 63mg、 Gluが 114mg、 Gly力 ¾47mg、 Alaが 132mg、 Valが 166mg、 Metが 19mg、 lie 力 l43mg、 Leuが 196mg、 Tyr力 ¾08mg、 Pheが 153mg、 Trpが 109mg、 LysHClが 382mg、 Hisが 97mg、 Argが 149mg、及び Proが 503mgで含有されている。
[0032] 10名の被験者力 試料の飲用前に一度、採血した。採血直後に試料を飲用させ、 1 時間経過後に再度、採血した。この 1時間を被験者は安静状態とした。実験のタイム テーブルは図 1に示した。試料の飲用前後での血液通過時間を測定し、比較するこ とで、血流改善効果を確認した。 10名の被験者のうちの 4名にて更にプラセボ試験( 水飲用)を行った。
[0033] なお、採血は真空採血管に予めへパリン 0.25ml (全血に対し 5%量)を添加しておき 、被験者より 5mlを採血する方法で行なった。得られた鮮血は直ちに測定試料として
用いた。測定には MC- FAN (Micro Channel Array Flow Analyzer)を用いた。 MC- FA Nでは流路系に 20cm水柱の圧力をかけ、この一定圧の下に全血 100 μ 1の流れる速 度を測定した。各測定時におけるバラツキは、各々の測定時に生理食塩水 100 1の 流速を測定することで補正した。 MC- FANに装着するマイクロチップには、 Bloody 6- 7 (チップサイズ: 15mm X 15mm,厚さ: 500 μ m、流路部:幅 X長さ X深さ =7 m X 30 μ m X 4.5 iu m)を用いた。
[0034] (結果)
10名の被験者につ!ヽてアミノ酸飲料 (VAAM)の血流改善効果を評価した。その結 果、 8名において血流時間は短縮し、残りの 2名では変化が見られなかった(図 2)。プ ラセボ群の 3名において血流時間の変化は認められなかった力 1名において大幅な 延長が認められた。この 1名については明らかにイレギュラーな測定値だと考えるの が妥当である。飲料摂取前の 100 μ 1の血流時間は 45.4±4.4秒 (mean士 S.D.)であつ たのに対して、摂取 1時間後では 41.6±3.2秒であり、有意に短縮された(student' s t -検定)(図 3)。他方、水飲用では血液流動性に影響を及ぼさなかった。つまり、アミ ノ酸飲料 (VAAM)の単回摂取により認められた血流改善効果は、 VAAMの有効成分 となるアミノ酸混合物によるものだと推察できる。
[0035] [実施例 2] アミノ酸混合物 (VAAM Mix.)を摂取した場合の血液流動性改善効果の 確認
試料としてアミノ酸混合物 (VAAM Mix.) (3.0 g、明治乳業 (株))を用いた。 VAAM M ix.には、アミノ酸として Aspが 7mg、 Thr力 S207mg、 Serが 63mg、 Gluが 114mg、 Glyが 347 mgゝ Alaが 132mg、 Valが 166mg、 Metが 19mg、 lieが 143mg、 Leuが 196mg、 Tyr力 208mg 、 Pheが 153mg、 Trpが 109mg、 LysHClが 382mg、 Hisが 97mg、 Argが 149mg、及び Pro 力 S503mgで含有されている。これを、オブラート(ビップトウキヨゥ (株)製)に包んで、 50g の軟水で 6名の被験者に飲用させた。 VAAM Mix.の組成(重量%)は表 1のとおりで ある。
[0036] 6名の被験者から試料の飲用前に一度、採血した。採血直後に試料を飲用させ、 1 時間経過後に再度、採血した。この 1時間を被験者は安静状態とした。試料の飲用 前後での血液通過時間を測定し、比較することで、血流改善効果を確認した。なお、
各血液試料にっ 、て 2回ずつ測定した。
[0037] また、採血は真空採血管に予めへパリン 0.25ml (全血に対し 5%量)を添加しておき 、被験者より 5mlを採血する方法で行なった。得られた鮮血は直ちに測定試料として 用いた。測定には MC- FAN (Micro Channel Array Flow Analyzer)を用いた。 MC- FA Nでは流路系に 20cm水柱の圧力をかけ、この一定圧の下に全血 100 μ 1の流れる速 度を測定した。各測定時におけるバラツキは、各々の測定時に生理食塩水 100 1の 流速を測定することで補正した。 MC- FANに装着するマイクロチップには、 Bloody 6- 7 (チップサイズ: 15mm X 15mm,厚さ: 500 μ m、流路部:幅 X長さ X深さ =7 m X 30 μ m X 4.5 iu m)を用いた。
[0038] (結果)
VAAM Mix.を飲用した場合、 6名すベての被験者において血流時間は短縮した( 図 4)。試料摂取前の 100 1の血流時間は 45.0± 1.9秒(mean士 S.D.)であったのに対 して、摂取 1時間後では 42.8± 1.8秒であり、有意に短縮された(student' s t_検定) ( 図 5)。これまでの検討ではアミノ酸飲料 (VAAM、明治乳業 (株))の血流改善効果を 確認している。今回の検討において、アミノ酸混合物 (Mix.)の飲用により血流改善効 果が認められたことで、アミノ酸が VAAMの血流改善効果の原因物質として寄与して いると考えられる。

VAAM Mix.の組成 (重量0 /o)
[0039] 試料としてアミノ酸混合物(BCAA及び CAAM) (3.0g、明治乳業 (株)支給)を用いた 。試料はオブラート (ビップトウキヨゥ (株)製)に包み、 50gの軟水で 6名の被験者に飲 用させた。なお、 BCAAでは、ノリンが 750mg、ロイシンが 1500mg、及びイソロイシンが 750mgであった。 CAAMの糸且成(重量0 /0)は表 2のとおりである。
[0040] 試料の飲用前に一度、採血した。採血直後に試料を飲用させ、 1時間経過後に再 度、採血した。この 1時間を被験者は安静状態とした。試料の飲用前後での血液通過 時間を測定し、比較することで、血流改善効果を確認した。なお、採血は真空採血管 に予めへパリン 0.25ml (全血に対し 5%量)を添カ卩しておき、被験者より 5mlを採血する 方法で行った。得られた鮮血は直ちに測定試料として用いた。

CAAMを飲用した場合、 3名の被験者において血流時間は短縮し、 3名において延 長した(図 8)。試料摂取前の 100 /z lの血流時間は 41.6±3.0秒であったのに対して、 摂取 1時間後では 41.4± 1.2秒であり、血流時間に有意差は認められな力つた(図 9)

Claims
[1] アミノ酸糸且成物であって、スレオニン、プロリン、グリシン、ノ リン、イソロイシン、ロイ シン、チロシン、フエ二ルァラニン、リジン、メチォニン、トリプトファン、ヒスチジン、アル ギニンを含むことを特徴とする血液流動性改善剤又は血液粘度低下剤。
[2] 更にァスパラギン酸、セリン、グルタミン酸、ァラニンを含む請求項 1記載の血液流 動性改善剤又は血液粘度低下剤。
[3] スレ-オンを 2〜15モル、プロリンを 4〜30モル、グリシンを 7〜20モル、ノ リンを 4〜
8モノレ、イソロイシンを 3〜9モル、ロイシンを 2〜12モル、チロシンを 1〜9モル、フエ二 ルァラニンを 0.5〜5モル、リジンを 5〜11モルの割合で含有し、かつそれぞれ 5モル 以下の割合で、メチォニン、トリブトファン、ヒスチジン、アルギニンを含む請求項 1記 載の血液流動性改善剤又は血液粘度低下剤。
[4] 更に 1モル以下の割合のァスパラギン酸、 5モル以下の割合のセリン、 4モル以下の 割合のグルタミン酸、 12モル以下の割合のァラニンを含む請求項 3記載の血液流動 性改善剤又は血液粘度低下剤。
