Description
СПОСОБ ПРОИЗВОДСТВА КАЗЕИНА И УСТРОЙСТВО ДЛЯ ЕГО
ОСУЩЕСТВЛЕНИЯ
Изобретение может быть использовано в различных сферах промышленности, среди которых наиболее значительными являются пищевая, бумажная, химическая, а также фармацевтическая области промышленности. В молочной промышленности казеин используется для производства сыров. Но в настоящее время основной областью использования казеинов являются казеинаты, получаемые при добавлении в казеин щелочных веществ. В зависимости от этих добавляемых веществ могут быть получены казеинаты натрия, калия или кальция. Казеинаты широко используются в молочной промышленности для производства аналогов сыров, шоколадного молока, маргарина и многих других продуктов.
Казеин может быть получен двумя различными способами, а именно, кислотным осаждением и сычужным осаждением. Кислотный способ получения казеина является традиционным и основан на достижении точки осаждения казеина с помощью кислоты. Кроме того, разные кислоты определяют различную структуру осажденного казеина. Например, казеин, осажденный молочной кислотой, более зернистый и рыхлый, в то время как осаждение соляной кислотой определяет более вязкую и тягучую структуру казеина. Сычужный казеин производят, используя молоко свертывающие ферменты и придерживаясь оптимальных условий свертывания и формирования сгустка. Готовый сгусток измельчают, нагревают до 58-60 0C при перемешивании, промывают водой и высушивают. Надо упомянуть, что в отличии от кислотного осаждения сычужный способ осаждения казеина является необратимым процессом. Сычужным способом осажденный казеин в своем составе имеет много кальция и фосфатов, а кислотный казеин содержит ограниченное количество этих солей. Однако, оба вида казеина отличаются стойкостью к высоким температурам и имеют высокую пищевую ценность. Более подробно технологические способы производства казеина описаны в книге J.Dukstаs, D.Касеrаuskаs ,,Pieno реrdirbimо tесhпоlоgijа", 1994 г., Вильнюс.
Выше упомянутые способы производства казеина также описаны в патентах Российской Федерации ÷Nb 1600671, JVs 1692505, JVb 2199233, JVb 2201099. В молочной промышленности широко используются мембранные процессы фильтрации (микро-, ультра-, нанофильтрация и обратный осмос) - сепарационные (разделительные) процессы, которые протекают под давлением с использованием пористых полимерных или неорганических материалов. Эти процессы за последние 30 лет нашли широкое применение в различных отраслях промышленности для очистки
или концентрирования жидких сред. С помощью мембранных технологий перерабатывают цельное молоко, обезжиренное молоко, предварительно сквашенное молоко, а также сыворотка. В отличие от обычной фильтрации, которая используется при выделении взвешенных частиц больше чем 10 мкм, мембранные процессы фильтрации позволяют отделить частицы, размер которых меньше чем 10 микрон. Мембранная фильтрация позволяет концентрировать выделяемые частицы в меньшем объеме относительно первоначального объема жидкости. Таким образом могут быть получены концентраты сывороточных белков, не содержащие жиры и бактерии. Информацию о мембранных технологиях можно найти на сайте www. gеаfiltrаtiоп.соm. Известными способами полученный казеин имеет следующие недостатки:
1. Казенны нерастворимы в воде, что затрудняет их непосредственное использование в различных отраслях промышленности. Использование казеинов требует их конвертации в казеинаты или другие приемлемые формы.
2. Поскольку сычужный способ осаждения казеина является необратимым процессом, полученный нерастворимый казеин, в отличие от кислотного казеина, не может быть восстановлен в растворимую коллоидную форму. Это свойство сильно ограничивает возможности его применения.
3. Произведенный сычужный казеин не может быть повторно переработан.
4. Кислотным способом полученный казеин является кислым продуктом, кроме того, во время производства получаемая сыворотка также является кислой. Перед использованием, например, в пищевой промышленности, такие кислые продукты необходимо нейтрализовать до значения пригодного к употреблению.
Целью изобретения является производство водорастворимого казеина, не используя кислоты, щелочи или какие-либо другие химические вещества. Цель может быть достигнута путем мембранных микро-, ультрафильтрационных систем, с помощью которых белки молока разделяют на сывороточные белки и белок казеина. Белок казеина далее обрабатывают в мембранном ультрафильтрационном концентраторе, при этом для выделения белка не используя каких-либо связывающих веществ. Предлагается мембранный способ производства казеина, в начале которого цельное коровье молоко сепарируют в сепараторе, где отделяют жиры и обезжиренное молоко. Новым является то, что обезжиренное молоко после сепаратора из промежуточной емкости пастеризируют в пластинчатом пастеризаторе при 50-80 0C температуре с выдержкой 15-75 с, после чего охлаждают до 4-10 0C и направляют в
промежуточную балансовую емкость. Из этой емкости обезжиренное молоко подают в мембранный фильтр микрофильтрации-фракционирования белков, в котором молочный белок разделяют на казеин и сывороточный белок. Отделенный белок казеина-ретентат микрофильтрации через промежуточную емкость подают в мембранный фильтр ультрафильтрации+дефильтрации, где образовывают ретентат ультрафильтрации - 14- 36 % сухих веществ концентрат казеина. Полученный концентрат направляют в сушилку, где высушивают до 4-6 % влажности, а полученный в воде растворимый порошок казеина направляют сначала в охлаждающий циклон и далее в фасовочный цех. В состав оборудования производства казеина входит мембранный фильтр микрофильтрации-фракционирования белков, мембранный фильтр ультрафильтрации + дефильтрации концентрирования белка казеина, мембранный фильтр ультрафильтрации концентрирования сывороточных белков.
Мембранный фильтр микрофильтрации-концентрирования белков составляют мембраны, которые отделяют частицы размером 0,05-10 мкм, площадь их поверхности от 50 до 310 м2, скорость фильтрации 50-120 л/cм2/ч, рабочая температура 5-28 0C.
Мембранный фильтр ультрафильтрации+дефильтрации состоит из мембран, отделяющих частицы размером 0,001-0,2 мкм, площадь поверхности которых 50-310 м2, скорость фильтрации от 50 до 120 л/cм2/ч, а рабочая температура 5-28 0C. Предлагаемое изобретение описывается следующими схемами: рис. 1 - общая схема производства водорастворимого казеина из коровьего молока;
На производство доставленное цельное коровье молоко из молоковоза 1 по приемочному трубопроводу направляют в промежуточную емкость 2, из которой в нагреватель 3, где подогревают до 5-58 0C температуры. Подогретое молоко подается в отделитель жиров молока - сепаратор 4, с помощью которого отделяют обезжиренное молоко и жиры. Во время сепарации полученное обезжиренное молоко направляют в промежуточную емкость 5, затем в пластинчатый пастеризатор, где пастеризируют при 50-87 0C температуре с выдержкой от 15 до 75 с. Далее в пластинчатом холодильнике 7 обезжиренное молоко охлаждают до 5-28 0C и через промежуточную емкость 8 направляют в мембранный фильтр микрофильтрации-фракционирования белков 9. Ретентат микрофильтрации (белок казеина) через промежуточную емкость 15 подают в мембранный фильтр ультрафильтрации+дефильтрации 16, из которого полученный ретентат ультрафильтрации, - 14-36 % сухих веществ концентрат белка казеина,
направляют в сушилку 17 и высушивают до 4-6 % влажности. Таким образом полученный порошок имеет неповрежденную белковую структуру и является водорастворимым.
Характеристика водорастворимого казеина: Белок казеина 74 - 85 %
Жиры 0,8 - 2 %
Лактоза+минеральные вещества 18 - 7 % Влажность 4 - 6 %.
При производстве казеина выше описанным способом получают дополнительные продукты, а именно, концентрат сывороточных белков, сливки (рис. 1). Пермеат микрофильтрации (сывороточные белки) через промежуточную емкость 10 поступает в пластинчатый холодильник 11, где его охлаждают до 5-28 0C. Охлажденный пермеат подают в мембранный фильтр ультрафильтрации 12, из которого выходит ретентат ультрафильтрации, имеющий 83 % сывороточных белков, 16-36 % сухих веществ. Ретентат ультрафильтрации в сушилке 13 высушивают до 4-6 % влажности, что позволяет получить концентрат сывороточных белков, состав которого следующий: Сывороточные белки 85 %
Жиры 2 %
Лактоза+минеральные вещества 7 - 9 % Влажность 4 - 6 %.
Сливки 60 %-нoй жирности из сепаратора 4 поступают в нормализатор 18, в который из промежуточной емкости 10 поступает необходимое количество пермеата микрофильтрации. В нормализаторе 18 сливки нормализуют, например до 40 %-нoй жирности, а затем подают в пастеризатор сливок 19, где пастеризуют при температуре 95-105 0C. Полученные сливки направляются для дальнейшего использования.
Преимущество растворимого казеина, полученного мембранным способом, в том, что не требуется дополнительной конвертации в казеинаты, которая необходима для казеина, полученного традиционным способом. Поскольку казеин является основным молочным белком производства сыров, мембранным способом полученный казеин для производства сыров может быть использован непосредственно. Это позволяет в 3 раза уменьшить производственные затраты.
Также большим преимуществом водорастворимого казеина является то, что в процессе производства не используются ни кислоты, ни щелочи, ни какие-либо другие связывающие вещества, что дает возможность получить чистый экологический продукт.

ФОРМУЛА ИЗОБРЕТЕНИЯ
5 1. Способ производства казеина, когда сырое коровье молоко сепарируют в сепараторе, где отделяют жиры и обезжиренное молоко, отличающийся тем, что обезжиренное молоко после сепаратора из промежуточной ёмкости пастеризируют в пастеризаторе при температуре 50 - 80 0C, выдерживая 15-75 секунд, после чего охлаждают до температуры 4 - 10 0C и направляют в
Ю промежуточную балансную ёмкость, из которой подают в микрофильтрационный — мембранный филтр фракционирования белков, где молочный белок разделяют на белок казеина и белок сыворотки, а отделенный белок казеина - ретентат микрофильтрации подают через промежуточную ёмкость в мембранный фильтр ультрафильтрации + дефильтрации, где получают
15 ретентат ультрафильтрации - концентрат казеина 14-36 % сухого материала, которого направляют в сушилку, где высушивают до 4-6 % влажности, а полученную водорастворимую муку казеина направляют в циклон очлаждения и на расфасовку.
2. Устройство для производства казеина, включающее сепаратор, 0 отличающееся тем, что оно состоит из микрофильтрационного мембранного фильтра фракционнирования белков и мембранного фильтра ультрафильтрации + дефильтрации сыворотки протеина и белка казеина.
3. Устройство для производства казеина по п. 2, отличающееся тем, что микрофильтрационный мембранный фильтр фракционирования белков состоит 5 из мембран, разделительная способность которых от 0,05 до 5 мкм, площадь поверхности 50-310 м2, скорость фильтрации 50-120 л/cм2/ч, а рабочая температура 5-280C.
4. Устройство для производства казеина по п. 2, отличающееся тем, что мембранный фильтр ультрафильтрации + дефильтрации сываротки протеина и 0 белка казеина состоит из мембран, разделительная способность которых от 0,001 до 0,5 мкм, площадь поверхности 50-310 м2, скорость фильтрации 50-120 л/cм2/ч, а рабочая температура 5-28 0C.
