[0002] 不斉炭素上にヒドロキシメチル基を有する多くの有用化合物やその中間体が知ら れている。従来、これら光学活性ヒドロキシメチルイ匕化合物の合成法として、入手が容 易な光学活性化合物からの誘導化法 (非特許文献 1)や光学分割法 (非特許文献 2, 3)、ジァステレオ選択的不斉合成反応 (非特許文献 4)などが用いられてきた。近年 、不斉合成法の進歩により触媒的不斉ヒドロキシメチル化反応の報告も増えつつある 力 基質一般性、収率、立体選択性などに問題が残されている (非特許文献 5〜7、 特許文献 1)。
ところで、ホルムアルデヒドは、有機合成において 1炭素増炭素反応の最も重要な 求電子剤であり、ヒドロキシメチルイ匕反応にぉ 、てもホルムアルデヒドをルイス酸で活 性ィ匕する方法がしばしば用いられてきた。し力しながら、有機溶媒中で反応を行うた めには、ホルムアルデヒド重合体力 熱分解により発生させる必要があり、安全性や 利便性の点で問題が大きい。また、ホルムアルデヒドの水溶液であるホルマリンは安 価で取り扱いも容易である力 通常のルイス酸は加水分解を受けやすいため、ホル マリンをルイス酸によって活性ィ匕することは困難である。

一方、ビスマス塩は高いルイス酸性を有しており、反応の種類によっては (特に水溶 液中の反応)、スカンジウムよりも優れた触媒活性を示すことが知られている。しかも、
ビスマスカチオンはほとんど無毒であり、スカンジウムよりも安価である。しかしながら 、ビスマス塩を用いる触媒的不斉反応としては、塩化メチレン中でのアルデヒドのトリ メチルシリルシア-ドによるシァノ化反応が唯一知られているに過ぎない(非特許文 献 10)。

課題を解決するための手段
[0006] このような課題を解決するために、本発明者らは、ホルマリン等のホルムアルデヒド を求電子剤としたケィ素エノラートとのアルドール反応を検討した結果、ビスマス塩と 光学活性なビビリジン化合物とから調整される不斉触媒を用いることにより、該反応が 高収率かつ高立体選択的に進行することを見出し、本発明の完成に至った。
[0007] 即ち、本発明は、水溶液中又は水と有機溶媒との混合溶媒中で、キラルなビビリジ ン化合物力もなる配位子又はその対称体と、 BiYで表されるルイス酸 (式中、 Yはノヽ

ロゲン原子、 OAc、 OCOCF、 CIO、 SbF、 PF又は OSO CFを表す。)とを混合
させて得られる触媒の存在下で、下式 (式 1)
(式中、 R5〜R7は水素原子、脂肪族、炭化水素基、単環又は多環の脂環式炭化水 素基、単環又は多環の芳香族もしくは芳香脂族炭化水素基、あるいは複素環基を表 し、 R5と R7はそれぞれ異なり、 R6は水素原子以外であり、 R8は、それぞれ同じであつ ても異なってもよぐメチル基、ェチル基、又はイソプロピル基を表す)で表されるケィ 素エノラートとホルムアルデヒドとを反応させることを含む、光学活性ヒドロキシメチル 化化合物の製法である。

発明の効果
[0010] この発明によれば、例えば医薬品やそのリードィ匕合物等の原料または合成中間体 として有用な、不斉炭素にヒドロキシメチル基が結合したィ匕合物を、安価で安全な原
料であるホルムアルデヒド(例えばホルマリン)とビスマス塩とを用いることで、水系溶 媒中で高収率かつ高立体選択的に得ることができる。
発明を実施するための最良の形態
[0011] 本発明で用いる触媒はキラルなビビリジンィ匕合物力もなる配位子又はその対称体と 、 BiYで表されるルイス酸(式中、 Yはハロゲン原子、 OAc、 OCOCF、 CIO、 SbF

[0012] ビビリジンィ匕合物力 なる配位子又はその対称体は、水酸基の結合した 2つの不斉 炭素を有し、 Bi塩の水中での触媒活性を調整するキラルな配位子となる。ビビリジン 化合物は Bi塩への配位能が適切であるため、ルイス酸性を低減させず、又、 Biと配 位子力もなる錯体力 カチオンを放出させることも少なぐ触媒の立体選択性が保た れる。
特に、下式 (化 2)

(式中、 R1は炭素数 4以下のアルキル基またはフエ-ル基を表す)で表されるビビリジ ン化合物を用いると、ルイス酸性及び立体選択性の点で好ま 、。
[0013] ビスマス塩として、 BiYで表されるルイス酸を用いる。 Yはハロゲン原子、 OAc、 O

COCF、 CIO、 SbF、 PF又は OSO CF (OTf)を表す。中でも Bi(TOf) 3が効果

的である。また、ビスマスカチオンは、毒性が極めて弱ぐスカンジウムよりも安価であ るという特徴がある。
[0014] この配位子と BiYとを溶媒中で混合すると、 Bi塩が配位子に配位し、触媒を形成

する。この溶媒としては、水と混和しやすい非プロトン性極性溶媒、及びこれらの溶媒 と水との混合溶媒を例示することができる。非プロトン性極性溶媒としては DME (ジメ トキシェタン)ゃジグリム(ジエチレングリコーノレジメチノレエーテノレ)等のエーテル、プ
口ピオ-トリル等の-トリル、アセトン等のケトンが挙げられる。これらの有機溶媒は、 例えば、水 1に対し、これらの溶媒 1〜19 (体積比)の割合で混合することができる。 溶媒中の配位子と Bi (OTf) 等の BiYの各濃度は 0.01〜0.1mol/l程度が好ましい

[0015] 本発明にお 、ては、この触媒を下記のホルムアルデヒドとケィ素エノラートとの不斉 ヒドロキシメチルイ匕反応 (ィ匕 3)に用いる。

R5〜R7は水素原子、脂肪族、炭化水素基、単環又は多環の脂環式炭化水素基、 単環又は多環の芳香族もしくは芳香脂族炭化水素基、あるいは複素環基を表し、こ れらは置換基を有していてもよい。この炭化水素基あるいは複素環基としては、たと えば、メチル、ェチル、プロピル、イソプロピル、ブチル等のアルキル基、シクロへキシ ル基、フエ-ル基、フエ-ルェチル基、フエ二ルビニル基、ナフチル基、フリル基、チ ェニル基等が例示される。またこれらの有してもよい置換基としては、ハロゲン原子、 アルコキシ基、チォアルコキシ基、炭化水素基等の各種のものであってよい。
R5〜R7は好ましくは以下のとおりである。
R5は水素原子又はアルキル基を表し、 R6はアルキル基、アルキルァリール基、ァリ ール基又はスルフイドを表し、但し、 R5と R6は共にその一部が芳香族環を形成してい てもよい炭素及び任意にヘテロ原子、好ましくは炭素原子力 成る 5〜6員環を形成 してもよく、 R7は水素原子、アルキル基、アルキルァリール基、又はァリール基を表す 又、 R5と R7はそれぞれ異なる。
[0016] R8は炭化水素基を表す。これらはそれぞれ同じであっても異なってもよいが、好ま しくは同一である。 R8はメチル基、ェチル基、又はイソプロピル基である。
[0017] この反応は、水溶液中又は水と有機溶媒との混合溶媒中において行われる。この
際、水との混合溶媒として用いられる有機溶媒としては、水と容易に混ざり合う、ジメト キシェタン(DME)、テトラヒドロフラン (THF)、ァセトニトリル、ジォキサン等が挙げら れるが、好ましくは DME、 THF、ァセトニトリル、ジォキサンが例示される。有機溶媒 と水との混合比については特に限定はないが、一般的には水を 1重量%以上、より 好ましくは 5重量%以上含む。
[0018] 水溶液又は混合溶媒の使用量は、適宜に考慮されるものである力 通常は、原料 物質並びに触媒の溶解に必要とされる量として、たとえばこれらの 2〜50重量倍の割 合での使用が考慮される。
[0019] 反応液中の HCHOZケィ素エノラートのモル比は好ましくは 1〜50、より好ましくは 1〜: LO程度である。また触媒は、ケィ素エノラートに対して 1〜50モル%、より好ましく は 5〜20モル0 /0使用する。
反応温度は— 30°C〜常温、より好適には— 15〜0°Cの範囲である。
反応時間は、適宜定めてもよぐ例えば、 0. 5〜50時間である。
[0020] この反応により、光学活性ヒドロキシメチルイ匕化合物が生成する。
[0021] 本発明の製法においては、(前記キラルなビビリジンィ匕合物からなる配位子又はそ の対称体 Z前記ルイス酸)で表されるモル比を 2. 5以上とするのが好ましぐ 3以上と することがより好ましい。前記モル比が 2. 5未満の場合、生成物の収率や選択性が 低くなる傾向にある。最も好ましくは前記モル比が 3〜4である。
[0022] 又、本発明の製法において、さらに 2, 2'—ビビリジンを添加剤としてカ卩えることが好 ましい。 2, 2'—ビビリジンをカ卩えて上記反応を行うと、収率や選択性を低下させること なく触媒量を減らすことが可能である。 2, 2'—ビビリジンの添加量としては、例えば 前記ビスマス塩 1モル当り、 5モル以上とすることが好ましい。 2, 2'—ビビリジンの添 加量が 3モル以下であると、生成物の収率が充分に向上しない傾向にあり、又、 5モ ルを超えても効果が飽和する。
[0023] 以下、実施例にて本発明を例証するが本発明を限定することを意図するものでは ない。
実施例 1
[0024] 下式 (ィ匕 2)の構造を有するキラルなビビリジンを、非特許文献 3に記載の方法により
作成した
真空下 200°Cで 1時間乾燥した金属塩 MXn (以下の表 1中の各化合物、 0.020mmol ) に DME (0.50 mL)を加える。この溶液に上記配位子 la (上記式 (ィ匕 2)の化合物に おいて!?1 = tert-Bu, 0.022mmol)を加え、透明になるまで室温で撹拌した。 0°Cまで 降温した後に HCHO水溶液(35%,86mg,1.0mmol)と、ケィ素エノラート 2(以下の化学 式 (化 4)中の化合物 (0.20 mmol)をカ卩えた。 4時間撹拌した後に飽和重曹水を加え、 水層から CH C1で 3回抽出した。有機層を Na SOで乾燥した後、溶媒を減圧下留去

し、残渣をシリカゲル薄層クロマトグラフィー (hexane:AcOEt = 2:1)で精製した。なお、 水 ZDMEの体積比は 1Z9であった。

得られた結果を表 1に示す。表中、 traceは、ほとんど検出されな力つたことを示す。 また、 Eeはェナンチォマー過剰率を示す。

実施例 2
[0027] 金属塩としてビスマストリフラートを 3mol%用い、配位子 laを 9mol%用い、さらに D MEの代わりに表 2に示す各溶媒を用いたことの他は、実施例 1とまったく同様にして 反応を行った。

[0030] ビスマストリフラート及び配位子 laの量を変化させ、又、反応温度を変化させたこと の他は、実施例 1とまったく同様にして反応を行った。但し、ケィ素エノラートの濃度を 0.36Mとした。

[0033] ビスマストリフラートに対する配位子 laの量を変化させたことの他は、実施例 1とまつ たく同様にして反応を行った。

実施例 5
[0037] ビスマス塩に対して、さらに 5倍モルの 2, 2 '—ビビリジンを添加剤として加え、ビス マストリフラート及び配位子 laの配合割合、並びに反応時間を変化させたことの他は 、実施例 1とまったく同様にして反応を行った(実験例 43, 44)。

[0039] 表 5にお 、て、触媒量を減らすと収率及び Eeも低下して 、る(実験例 40〜42)。実 験例 42では、一方の基質であるケィ素エノラートが反応開始 4時間後に消失し、反 応の進行が停止した (収率 63%) .—方、 Xと yの値が実験例 42と同一である力 さら に 2, 2 '—ビビリジンを加えた実験例 43の場合、反応開始後 21時間までケィ素ェノラ ートが残存しつつ反応が進行し、収率が大幅に向上し、 Eeも高くなつた。触媒量 (表 中の Xで表される)をさらに 0. 5 mol%に減らした実験例 44の場合でも、収率が若干低 下したものの選択性は維持された。
実施例 6
[0040] 以上の結果を踏まえ、種々の基質を用い、 2, 2'—ビビリジンを添加剤としてカ卩えた ことの他は、実施例 1とまったく同様にして反応を行った。但し、ビスマストリフラートの 量を lmol%とし、配位子 la量を 3mol%とし、 2, 2 '—ビビリジンの量を 5mol%とし、溶 媒として水 ZDMEの体積比を 1Z4とした。又、反応時間を基質に応じて変化させた [化 10]

Claims
(式中、 R5〜R7は水素原子、脂肪族、炭化水素基、単環又は多環の脂環式炭化水 素基、単環又は多環の芳香族もしくは芳香脂族炭化水素基、あるいは複素環基を表 し、 R5と R7はそれぞれ異なり、 R6は水素原子以外であり、 R8は、それぞれ同じであつ ても異なってもよぐメチル基、ェチル基、又はイソプロピル基を表す)で表されるケィ 素エノラートとホルムアルデヒドとを反応させることを含む、光学活性ヒドロキシメチル 化化合物の製法。
前記キラルなビビリジンィ匕合物力もなる配位子は、下式
