背景技術
[0002] ケモカインは生体内での主たる細胞遊走因子であり、細胞運動の亢進や細胞接着 分子の活性化を介して、リンパ球の組織浸潤を制御している。ケモカインは、その最 初の 2つのシスティン残基の配列により、 CC、 CXC、 C、 CXXXCの 4つのサブファミリ 一に分類される。 CC、 CXC、 Cケモカインのメンバーは、約 70アミノ酸力もなる分泌蛋 白質であり、それ自身には接着分子としての活性はないが、細胞接着を誘導すること ができる。分泌されたケモカインは、標的細胞表面上の 7回膜貫通型受容体に結合し て、三量体 G蛋白質を介してインテグリンを活性化し、細胞の接着や遊走を誘導する
[0003] 最近になって、従来の細胞遊走機構に加えて、新規の簡潔なリンパ球浸潤機構が 同定された。この機構は活性化された内皮細胞上に発現するフラクタル力インと、血 流中の単球、 NK細胞、そして T細胞の一部に発現する 7回膜貫通型受容体 CX3CR1 により媒介される。フラクタル力インは CXXXCケモカインの唯一のメンバーであり、そ の構造と機能にぉ 、て、他のケモカインには見られな 、際だった特徴を有して 、る。 フラクタル力インは、ケモカインドメイン、ムチンドメイン、細胞膜貫通領域、細胞質内 領域を有する膜結合型として細胞表面上に発現する。膜結合型フラクタル力インは C X3CR1と結合することで、生理的血流速存在下においても、セレクチンやインテグリ ンの介在なしに、単独で強固な接着を媒介することが可能である。すなわち、セレク チンやインテグリンを介する多段階の細胞浸潤機構と同様な機能を、フラクタルカイ ン -CX3CR1細胞浸潤系は一段階の反応で媒介する。さらに、膜結合型フラクタル力 インカゝらシエディングによって分泌される分泌型フラクタル力インは、 CX3CR1に結合 して、従来のケモカインと同様に、インテグリンの活性化や細胞遊走を誘導する。
[0004] また、フラクタル力インは、血管内皮細胞を炎症性サイト力インの TNFや IL-1で処理 すると発現が誘導される。一方 CX3CR1は単球や、 NK細胞のほとんどと、 T細胞の一 部に発現している力 好中球には発現していない。したがって、フラクタル力イン- CX 3CR1細胞浸潤系は、損傷を受けた組織の内皮細胞上、又は組織内に、ある種の免 疫細胞を動員するための、きわめて効率の良い機構であると考えられる。フラクタル 力インは上記のように炎症時に血管内皮細胞に誘導され、 CX3CR1は多種類の白血 球に存在していることから、フラクタル力イン- CX3CR1細胞浸潤系は、炎症性疾患に おける病態形成および進展への関与も強く示唆されている。実際にフラクタル力イン -CX3CR1細胞浸潤系と炎症性疾患における関与については数多く報告されており、 関節リューマチ (非特許文献 1)、潰瘍性大腸炎やクローン病に代表される炎症性腸 疾患 (非特許文献 2)、乾癬およびアトピー性皮膚炎 (非特許文献 3、非特許文献 4)、 喘息 (非特許文献 5)、動脈硬化症 (非特許文献 6)、急性呼吸促迫症候群 (非特許 文献 7)、など数多くの疾患について報告がなされている。一方では CX3CR1ノックァ ゥトマウスを用いた解析 (動脈硬化症 (非特許文献 8)、虚血性再灌流による組織障 害 (非特許文献 9) )、抗フラクタル力インモノクローナル抗体を用いた炎症性疾患の 病態モデル解析 (マウス Π型コラーゲン誘発関節炎、実験的自己免疫性脳髄膜炎 ( 特許文献 1)、コンカナパリン A誘発肝障害 (特許文献 1)、抗 CX3CR1抗血清を用い た病態モデル解析 (WKYラット半月体形成性腎炎 (非特許文献 10)、ァロ心臓移植 片の拒絶 (非特許文献 11) )、あるいはフラクタル力イン抑制変異体を用いた炎症性 疾患の病態モデル解析 (MRL/lprループス腎炎(2004年 4月 日本リュウマチ学会)) から、炎症性疾患において、フラクタル力イン- CX3CR1細胞浸潤系を阻害することで 病態進展抑制及び改善効果が期待され、炎症性疾患における新たな治療体系を構 築できるものと期待されている。し力しながらフラクタル力インと CX3CR1の相互作用を 阻害することにより発揮される、これら病態改善作用の詳細なメカニズムは未だ不明 であるのが現状である。
これまでに白血球の分類は、数多くの細胞表面マーカーによりリンパ球、単球、顆 粒球それぞれの集団が細力べ分類されてきている。最近ではケモカイン受容体の分 布から、さらに詳細な分類がなされてきており、単一集団と思われていた集団がいく つかの亜集団から形成される集合であることが判明しつつある。マウスの解析力も CX 3CR1は、上述のように単球、 NK細胞、そして T細胞の一部に発現することが報告され ているが、その中でも単球には CX3CR1を強発現し CCR2を発現しない集団(CX3CR lhighCCR2-)と CX3CR1が低発現で CCR2を強発現する集団(CX3CR11<>WCCR2 + )の 2 つが存在することが明らかとなりつつある。 CX3CR1ノックアウトマウスや CCR2ノックァ ゥトマウスの解析から、 CX3CR11<>WCCR2 +は炎症時に炎症部位に誘導され、炎症性 サイト力インである TNF aや強力な NO (—酸化窒素)合成酵素の iNOS (inducible Nitr ogen Oxide Synthetase)を産生し、組織障害に寄与することが示唆されている(非特 許文献 12、非特許文献 13)。

て、炎症時にお ける機能や重要性にっ ヽては言及されておらず、むしろ非炎症時の構成的な組織 マクロファージの供給に必要であり、これら細胞の機能を阻害することは好ましくない と言われている。
[0006] 一方、ヒト末梢血における単球においても細胞表面マーカー(CD16、 CD62Lなど) と CX3CR1の発現により詳細な分類がなされている。その一つの集団である CD16+C D62L-の単球は炎症性疾患において、末梢血中に増加することが報告されており、 病態進展との関与が強く示唆されて 、る。 CD16+CD62L-の単球は CX3CR1を高発 現していることが報告されており、先にあげたマウス CX3CRlhighCCR2-とほぼ同様の 性質をもっと予想される(非特許文献 14)。また CD16+CD62L-の単球は TNF aや iN OSを強く産生することがすでに報告されており、炎症性疾患における末梢血中の増 カロと相まって、病態進展に強く関与していることが考えられる。しかし CX3CR1の機能 を阻害することで、これら細胞からの TNF aや iNOS産生抑制が認められるかどうか具 体的な報告は存在しない。
[0007] 最も高 、NO合成能力を持つ iNOSは、 eNOS (endothelial NOS)および nNOS (neura 1 NOS)のように定常的には発現しておらず、刺激因子 (例えば炎症性サイト力イン及 び Z又はリポ多糖体など)によって誘導され、一過性に大量に産生されて細菌、ウイ ルス、真菌、寄生虫の排除などの生体防御反応に関わっている。しかしながら過剰な NOの産生は糸且織障害的に作用するため、病変部における iNOSの過剰な産生は病 態を進展させる大きな要因となると考えられている。実際に NOが病態形成や進行に 関わると考えられている疾患としては、炎症性疾患 (例えば、リウマチ性炎症、関節リ ユーマチ、変形性関節炎、クローン病、潰瘍性大腸炎、乾癬、動脈硬化症、自己免 疫疾患、又は急性炎症など)、アレルギー疾患 (喘息、アトピー性皮膚炎)、虚血性障 害 (例えば、梗塞若しくは虚血などに起因する各種の心臓障害若しくは脳障害、又は 虚血後の再灌流障害など)、ショック (例えば、内毒性ショック、出血性ショック、又は 心臓性ショックなど)、病的な血圧低下 (例えば、サイト力インを用いる癌治療における 血圧低下、又は敗血症、出血性ショック、若しくは肝硬変において生じる血圧低下な ど)、移植拒絶反応、神経系障害 (例えば、アルツハイマー病、てんかん、又は片頭 痛など)、腫瘍、又はインシュリン依存性糖尿病などが挙げられる。
[0008] 一方では病態モデルにおける iNOS活性の阻害あるいは iNOSノックアウトマウスの 解析から、関節リューマチ (非特許文献 15)、変形性関節症 (非特許文献 16)、潰瘍 性大腸炎やクローン病に代表される炎症性腸疾患 (非特許文献 17、非特許文献 18 )、コンカナパリン A誘発肝障害 (非特許文献 19)、喘息 (非特許文献 20)、エンドトキ シン誘導急性肺障害 (非特許文献 21)、動脈硬化症 (非特許文献 22)、虚血性障害 (非特許文献 23、非特許文献 24、非特許文献 25)、移植拒絶反応 (非特許文献 22) などにお!、て病態の改善が報告されて!、る。
[0009] このように実際に数多くの疾患モデル動物にぉ 、て iNOS活性の阻害あるいは iNOS ノックアウトマウスによる病態改善の報告がなされている力 未だ有望な iNOS活性を 阻害する薬剤は上巿されて 、な 、。
[0010] さらに、 iNOSの酵素活性阻害剤による過剰な NO産生の抑制は、敗血症ショックに 典型的な血圧低下や血管神経麻痺などの組織破壊や臓器障害を改善する一方で、 血圧 ·血流の調節と!/ヽぅ NOの根幹的な生理機能を遮断する危険性が示唆されて ヽ る(非特許文献 26)。特に iNOSノックアウトマウスを用いた敗血症における心筋機能 の解析では、産生する細胞の種類による iNOSの役割の違いが指摘されている。心筋 細胞で発現する iNOSは敗血症時のアドレナリン作動性刺激による心筋細胞の短縮と V、う有益な反応に必須である一方、心筋細胞の近傍に浸潤した炎症性細胞で発現 する iNOSは心筋細胞の障害と 、う有害な反応に関与して 、る(非特許文献 27)。
[0011] したがって、細胞種選択的な iNOS活性の阻害という新しいアプローチ、例えば iNO Sの酵素阻害ではなく炎症性細胞における iNOSの酵素阻害や産生抑制などによる薬 剤の提供が待ち望まれて ヽる。

発明の開示
[0012] 本発明は、新しい概念に基づぐ炎症性疾患治療剤、具体的には炎症性腸疾患( 特に潰瘍性大腸炎、クローン病)、乾癬、アトピー性皮膚炎、喘息、動脈硬化症、虚 血再灌流における組織障害、急性呼吸促迫症候群などの治療剤の提供を目的とす る。
[0013] 本発明者は、これまでの知見から、 CX3CR1の機能を阻害することで、 iNOSの産生 が抑制され病態の改善を示した報告がな力つたことに注目し、この機能解明により新 しい概念に基づく炎症性疾患治療剤の創出に成り得るとの仮説を構築し、鋭意検討 を行った。
[0014] その結果、フラクタル力インと CX3CR1の相互作用を阻害し、 CX3CR1の機能を阻害 する抗体を用いて、後述する実施例に記載の各種病態モデルで検討した結果、 iNO Sの産生が抑制され、かつ、病態が改善されることを初めて見出した。さらに、フラクタ ルカインと CX3CR1の相互作用を阻害し、 CX3CR1の機能を阻害する抗体が ConA肝 炎に有効であることは報告されている力 今般、 ConA肝炎での肝臓における iNOS m RNAの発現抑制と、炎症性細胞での iNOSの産生が抑制されることを初めて見出した 。これらのことから、フラクタル力インと CX3CR1の相互作用を阻害し、 CX3CR1の機能 を阻害する抗体または CX3CR1の機能を阻害する化合物(以下「CX3CR1アンタゴ- スト」と略記する場合もある)は、フラクタル力イン- CX3CR1細胞浸潤系が関与する疾 患であって、 iNOS活性または炎症性細胞による過剰な NOの産生が原因となる炎症 性疾患 [炎症性腸疾患 (特に潰瘍性大腸炎、クローン病)、乾癬、アトピー性皮膚炎、 喘息、動脈硬化症、虚血再灌流における組織障害、急性呼吸促迫症候群など]の治 療に有用であることを明らかにし、本発明を完成した。
[0015] すなわち、本発明は以下のとおりである。
( 1)フラクタル力インと CX3CR1との相互作用を阻害する抗体または CX3CR1アンタゴ 二ストを含有することを特徴とする、炎症性疾患治療剤。
(2)抗体が、抗フラクタル力イン抗体であることを特徴とする、(1)記載の剤。
(3)抗フラクタル力イン抗体力 モノクローナル抗体であることを特徴とする、(2)記載 の剤。
(4)受託番号 FERM BP-10372のハイブリドーマによって産生されるモノクローナル 抗体 5H8-4、または受託番号 FERM BP-10371のハイブリドーマによって産生される モノクローナル抗体 #126であることを特徴とする、 (3)記載の剤。
(5)炎症性疾患が炎症性腸疾患である(1)〜 (4)の 、ずれか一項に記載の剤。

[0020] 哺乳動物(例えばマウス、ハムスター又はゥサギ)は、該哺乳動物において免疫応 答を引き起こす免疫原の形態の CX3CR1又はフラクタル力イン又は蛋白質断片(例え ばペプチド断片)で免疫することができる。
[0021] CX3CR1又はフラクタル力インの遺伝子(例えば、 GenBank NM_001337又は NM_00 2996参照)を組み込んだ発現ベクターを宿主細胞、例えば細菌、哺乳類細胞株又は 昆虫細胞株中で発現させ、培養液又は菌体 ·細胞力 標準的な方法に従って CX3C R1又はフラクタル力インを精製することができる。また、例えば GST等との融合蛋白質 として発現させ、 GSTとの融合蛋白質の場合はダルタチオンカラムにより精製しても構 わない。

Today, 4: 72, 1983)、 EBV-ハイブリドーマ法(Cole et al., Monoclonal Anti body inCancer Therapy, 1985, Allen R. Bliss, Inc., pages 77— 96)、コンビナトリアル 抗体ライブラリーのスクリーニング(Huse et al., Science, 246: 1275, 1989)等他の方 法により作製しても良い。
[0024] また別法として、 CX3CR1を発現させた昆虫細胞をそのまま哺乳類動物に免疫して 、該哺乳動物のリンパ球よりハイプリドーマを作製し、産生される抗体のスクリーニン グを、 CX3CR1を発現させた哺乳類細胞 (昆虫細胞との交叉免疫性が低ぐ昆虫細 胞由来の蛋白質に対する抗体が結合しない細胞)で行う方法も許される。また、特開 2002-345454号公報の方法により調製することもできる。

本発明は、フラクタル力インと CX3CR1との相互作用を阻害する抗体力 フラクタル 力イン- CX3CR1細胞浸潤系が関与する疾患であって、 iNOS活性または炎症性細胞 による過剰な NOの産生が原因となる炎症性疾患の治療に有用であることを見出した ことにより完成したものである。したがって、フラクタル力インと CX3CR1の相互作用を 阻害し、 CX3CR1の機能を阻害する化合物、 CX3CR1アンタゴ-ストも本発明に使用 することができる。前記作用を有する化合物であれば特に限定されず、公知な化合 物であってもよぐ新規な化合物であってもよい。さらに、後述するスクリーニング方法 によって得られる化合物であってもよ 、。

CX3CR1とフラクタル力インの相互作用を阻害する抗体または CX3CR1アンタゴニス トは、フラクタル力イン又は膜結合型フラクタル力インを発現する細胞に対し、 CX3CR 1陽性細胞が遊走する力否かによりスクリーニングできる。以下に CX3CR1陽性細胞 が遊走する力否かによりスクリーニングする具体的な方法について記載する力 本発 明はこれに限定されるものではない。
[0029] フラクタル力インに対する遊走は、例えばトランスゥエルカルチャーインサート(コー スター社製)を用いて、柳』定することができる。
[0030] トランスゥエルカルチャーインサートに、フラクタル力インを発現していない細胞例え ば ECV304細胞を培養し、カルチャーインサート上面に単層の細胞層を形成させる。 フラクタル力インを、適当な濃度好ましくは 10 nMの濃度になるように、遊走溶液 (例え ば RPMI- 1640: M199 = 1 : 1, 0.5% BSA, 20 mM HEPES, pH7.4)で希釈し、 24穴のゥ エルプレートに加える。 ECV304細胞を培養したトランスゥヱルカルチャーインサートを 24穴のトランスゥエルに取りつけ、遊走溶液に懸濁した適当な個数、好ましくは 106個 の末梢血単核球細胞をトランスゥエルカルチャーインサートにカ卩える。適当な条件好 ましくは 37°Cで 4時間培養した後、 ECV304細胞を通過して、ゥエルプレートに遊走し てきた細胞を回収し、細胞表面マーカーや細胞内抗原により同定する。好ましくは蛍 光標識した該細胞表面マーカーや細胞内抗原に対する抗体で蛍光染色した後、 FA CScal¾urを用いて定量する。

[0033] < 5 >iNOSの産生抑制
本明細書において「iNOSの産生抑制」、「iNOSの産生が抑制され」とは、 iNOSの mR NA発現量が抑制されること、もしくは iNOSタンパク質の産生量が抑制されることを意 味する。 iNOSの産生抑制は、リアルタイム PCR法、ウェスタンブロッテイング法、 ELISA 法(固相酵素免疫測定法)、 iNOS酵素活性測定法、いずれ力を用いて測定すること ができる。例えば、リアルタイム PCRは具体的には、以下の様に行う。トータル RNAを 組織及び細胞から常法により精製し、逆転写酵素と適当なプライマーを用いて cDNA を作製する。作製した cDNAをもとに、各分子に特異的なプライマーと DNAポリメラー ゼを用いることで各分子を特異的に増幅させることができる。各分子は、増幅させる 際に適当な蛍光色素を取り込ませることで、特定の機器 (PRISM 7700 Sequence Det ector (Applied Biosystems社製)など)を用いて増幅曲線をモニターすることができる。 内部標準遺伝子 (ダリセルアルデヒド- 3-リン酸デヒドロゲナーゼ遺伝子など)により各 分子の補正を行うことで、各分子の組織及び細胞における mRNA発現量を定量的に 柳』定することができる。
[0034] < 6 >フラクタル力インと CX3CR1との相互作用を阻害する抗体または CX3CR1アンタ ゴニストの用途
本発明は、フラクタル力インと CX3CR1との相互作用を阻害する抗体または CX3CR1 アンタゴニストを含有する炎症性疾患治療剤を提供するものである。前記治療剤にお いては、前記抗体はフラクタル力インと結合するものであることが好ましい。抗体を含 有する本発明の治療剤をヒトに適用する場合には、以下の態様が好ましい。 [0035] ヒト以外の動物、例えばマウスを免疫動物として作製されたマウスモノクローナル抗 体は、ヒトに投与した場合異種蛋白質として認識されて、モノクローナル抗体に対す る免疫応答を生じさせてしまうことが多い。この問題点を回避する一つの方法はキメラ 抗体、すなわち抗原結合領域がマウスモノクローナル抗体由来、それ以外の領域が ヒト抗体由来の抗体である。本発明におけるフラクタル力インと CX3CR1との相互作用 を阻害する抗体はキメラ抗体も含むものである。キメラ抗体としては、抗原結合領域と してマウスモノクローナル抗体の可変領域全体を使ったキメラ抗体(Morrison et al., Proc. Natl.Acad. Sci. USA, 81: 6851, 1985、 Takeda et al., Nature, 314: 452, 1985) 、また抗原結合領域としてヒト由来のフレームワーク領域とマウスモノクローナル抗体 由来の超可変領域を組み合わせて使ったキメラ抗体(Teng et al., Pro Natl. Acad. Sci. USA, 80: 7308-12, 1983、 Kozbar et al., Immunol. Today, 4: 7279, 1983)が挙 げられる力 本発明はこれに限定されるものではない。

[0037] 本発明の治療剤は、フラクタル力イン- CX3CR1細胞浸潤系が関与する疾患であつ て、 iNOS活性または炎症性細胞による過剰な NOの産生が原因となる炎症性疾患、 具体的には炎症性腸疾患 (特に潰瘍性大腸炎、クローン病)、乾癬、アトピー性皮膚 炎、喘息、動脈硬化症、虚血再灌流における組織障害、急性呼吸促迫症候群など の治療が必要な患者に対して投与できる。炎症性疾患は、自己免疫疾患またはその 他の炎症性疾患であってもよい。また、炎症性疾患は、腎炎、心筋炎、自己免疫性 肝障害、多発性硬化症、リウマチ、または、その他の炎症性疾患であってもよい。
[0038] 本発明の治療剤の投与は、注射 (皮下、静脈内など)などの常法により行うことがで きる。
[0039] 治療剤の形態は、投与方法により適宜選択され、医薬的に許容可能な担体と組み 合わせた医薬組成物であってもよぐ例えば、注射用途に適した医薬組成物としては 、滅菌水溶液 (水溶性の場合)又は分散液および滅菌注射溶液又は分散液を即座 に調製するための滅菌粉末が挙げられる。注射用途に適した医薬組成物はいずれ の場合にぉ 、ても滅菌されて 、なければならず、容易な注射器操作が可能な程度に 流体でなければならな 、。該組成物は製造および貯蔵条件下で安定でなければな らず、細菌や真菌などの混入微生物の作用力も保護されていなければならない。担 体は、例えば、水、エタノール、ポリオール(例えば、グリセロール、プロピレングリコー ル、およびポリエチレングリコールなど)、およびこれらの適当な混合物を含む溶媒で あるか又は分散媒体であってよい。適当な流動性は、例えば、レシチンなどのコーテ イングを使用することによって、分散液の場合は必要な粒径を維持することによって、 および界面活性剤を使用することによって維持することができる。微生物の作用から の保護は、種々の抗菌剤および抗真菌剤、たとえばパラベン、クロロブタノール、フエ ノール、ァスコルビン酸、チメロサールなどにより行うことができる。多くの場合等張剤 、例えば糖、ポリアルコール、例えばマン-トール、ソルビトール、塩化ナトリウムなど が組成物中に含まれているのが好ましいであろう。注射用組成物の持続吸収は、吸 収を遅らせる薬剤、例えばモノステアリン酸アルミニウムやゼラチンなどを組成物中に 酉己合すること〖こより行うことができる。
[0040] 注射用溶液の調製は、必要なら上記成分の 1又はその組合せとともに所要量のフ ラタタルカインと CX3CR1との相互作用を阻害する抗体を適当な溶媒中に配合し、つ いで滅菌濾過することにより行うことができる。一般に分散液の調製は、基本的な分 散媒体と上記から選ばれた必要な他の成分を含む滅菌媒体中に活性化合物を配合 することにより行う。滅菌注射溶液調製のための滅菌粉末の場合は、好ましい調製法 は真空乾燥および凍結乾燥であり、これにより活性成分と前もって滅菌濾過した所望 の追加成分との粉末が得られる。
[0041] 本発明の治療剤の投与量は、抗体の選択、投与対象、患者の年齢、性差、薬剤に 関する感受性、投与方法、疾患の履歴など、そして医師の判断に変化し得るが、適 当な投与量範囲は患者の体重 lkgあたり例えば約 0. 01〜30mg、好ましくは約 0. 1 〜10mg程度を投与するのが好ましい範囲である。投与経路の効率が異なることを考 慮すれば、必要とされる投与量は広範に変動することが予測される。例えば経口投 与は静脈注射による投与よりも高い投与量を必要とすると予想される。こうした投与量 レベルの変動は、当業界でよく理解されているような、標準的経験的な最適化手順を 用いて調整することができる。
[0042] さらに、 CX3CR1に対する抗体と、その抗体に結合した細胞毒性物質とを含むィムノ トキシンとすることもできる。
[0043] 毒性物質としては、サボリン、リシン、 Pseudomonas外毒素、ジフテリア毒素、化学療 法剤などが挙げられる。抗体と毒性物質の結合は、従来のィムノトキシンの作製に用 いられる方法によって行うことができる。このィムノトキシンは、 CX3CR1発現細胞特異 的に増殖抑制を示す。
[0044] CX3CR1アンタゴニストを含有する炎症性疾患治療剤を、ヒトに適用する場合には、 以下の態様が好ましい。 CX3CR1アンタゴ-ストは塩を形成してもよぐ薬学的に許容 し得る酸または塩基などとの塩が挙げられる。従って、 CX3CR1アンタゴニストまたは その塩は、フラクタル力イン- CX3CR1細胞浸潤系が関与する疾患であって、 iNOS活 性または炎症性細胞による過剰な NOの産生が原因となる炎症性疾患、具体的には 炎症性腸疾患 (特に潰瘍性大腸炎、クローン病)、乾癬、アトピー性皮膚炎、喘息、 動脈硬化症、虚血再灌流における組織障害、急性呼吸促迫症候群などの治療に用 いることができる。得られた物質を単独で用いることも可能である力 薬学的に許容さ れ得る担体と配合して医薬品組成物として用いることもできる。この時の有効成分の 担体に対する割合は、 1〜90重量%の間で変動され得る。また当該治療剤は、経口 または非経口(例えば静脈注射、筋肉注射、皮下投与、直腸投与、経皮投与)のい ずれかの投与経路で投与することができる。
[0045] 従って、 CX3CR1アンタゴニストまたはその塩を含有する医薬組成物は、投与経路 に応じて適当な剤形とされ、具体的には錠剤、カプセル剤、顆粒剤、散剤、あるいは シロップ剤等による経口剤、または注射剤、点滴剤、リボソーム剤、坐薬剤等による非 経口剤を挙げることができる。これらの製剤は通常用いられている賦形剤、増量剤、 結合剤、湿潤化剤、崩壊剤、表面活性化剤、滑沢剤、分散剤、緩衝剤、保存剤、溶 解補助剤、防腐剤、矯味矯臭剤、無痛化剤、安定化剤等を用いて常法により製造す ることができる。使用可能な無毒性の上記添加剤としては、例えば乳糖、果糖、ブドウ 糖、デンプン、ゼラチン、ステアリン酸マグネシウム、メチルセルロース、またはその塩 、エタノール、クェン酸、塩ィ匕ナトリウム、リン酸ナトリウムなどが挙げられる。 [0046] それらの投与形態としては、また、必要な投与量範囲は、得られる化合物の選択、 投与対象、投与経路、製剤の性質、患者の状態、そして医師の判断に左右される。 しかし、適当な投与量は患者の体重 lkgあたり例えば約 1. 0〜1, 500 g、好ましく は約 10〜500 g程度を投与するのが好ましい範囲である。投与経路の効率が異な ることを考慮すれば、必要とされる投与量は広範に変動することが予測される。例え ば経口投与は静脈注射による投与よりも高い投与量を必要とすると予想される。こう した投与量レベルの変動は、当業界でよく理解されているような、標準的経験的な最 適化手順を用いて調整することができる。

[0048] 本発明で用いられるフラクタル力インと CX3CR1との相互作用を阻害する抗体また は CX3CR1アンタゴ-ストは、 iNOSの産生抑制の作用を有するので、 CX3CR1アンタ ゴニストを有効成分とする iNOS産生抑制剤も提供される。 iNOS産生抑制剤は、上述 した治療剤と同様に組成物としてもよぐ iNOS産生抑制が必要な患者に、上述した治 療剤と同様に投与することができる。

実施例 1 [0050] マウス CD4陽性 CD45RB強陽性 (CD4+CD45RBhigh) Tリンパ球移入炎症性腸疾患モ デルにおける抗フラクタル力イン抗体 (5H8-4)の効果
(1)抗フラクタル力イン抗体 (5H8-4)の調製
抗フラクタル力イン抗体 (5H8-4)は、以下の方法により調製した (特開 2002-345454 号公報)。抗原にはマウスフラクタル力イン (R&D社製)を用いた。抗原は TiterMaxァ ジュバンドと混合した後、アルメニアハムスターに免疫し、以降抗原のみで追加免疫 を行った。血清中の抗体価は ELISAを用いて測定した。抗体価が上昇したアルメニア ハムスター力もリンパ球を分離し、リンパ球: P3ミエローマ細胞の比率力 1になるよう に混合し、 PEG (ベーリンガー社製)を用いて細胞融合を行った。ハイプリドーマは、 R PMI-1640/10% FCS/HAT/10% Origen HCF (ISGN社製)を用いて、 96-ゥエルプレー トで 1週間培養した。そして、培養上清を用いて、 ELISAを実施し、陽性ゥヱルを同定 した。抗マウスフラクタル力イン抗体を産生するハイブリドーマは、限界希釈を 2回行 い、クロー-ングを行った。モノクローナル抗体は、不完全フロイントアジュバントを投 与した SCIDマウスにハイプリドーマを接種して作製した腹水から、プロテイン Aカラム を用いて精製した。中和活性は、 CX3CR1発現細胞のマウスフラクタル力インに対す る遊走を抑制することを指標にして測定し、中和抗体 (5H8-4)を得た。この中和抗体 を産生するハイブリドーマを Ham ®mFKN5H8-4と名づけた。

CD4陽性 CD45RB強陽性 (CD4+CD45RBhigh) Tリンパ球移入炎症性腸疾患モデル の作製については Powrie et al. , Int. Immunol, 5, 1461-1471, 1993を参考にした。 雌性、 8〜10週齢の Balb/cマウス(日本チャールズ 'リバ一)の脾臓を摘出し、孔径 100 μ mのセルストレーナ一 (ファーミンジェン社製)上で組織をすり潰し脾臓細胞を分離 した。分離した脾臓細胞は、脾臓一個あたり 5mlの塩ィ匕アンモ-ゥム溶液 (0.75%塩ィ匕 アンモ-ゥム、 16mMトリス緩衝液、 pH7.4)をカ卩ぇ 15分室温に放置し赤血球を溶解し た。脾臓細胞溶液に PBSを 2倍容量加え、 1500rpmで 5分遠心して沈殿を回収した。 分離した脾臓細胞から CD4 T cell Isolation kit (ミルテュー社製)により CD4 Tリンパ 球を精製した。 CD4陽性 CD45RB強陽性 Tリンパ球を分離するために、精製した CD4 Tリンパ球に対して、フィコエリスリン (PE)標識抗 CD4抗体 (eBioscience社製)、フルォ レセインイソチオシァネート(FITC)標識抗 CD45RB抗体(eBioscience社製)を用い二 重染色を行った。二重染色の後、 FACSAria (ベタトンディッキンソン社製)を用い CD4 陽性 CD45RB強陽性細胞をソーティングし目的の細胞を回収した。回収された細胞 は PBSで洗浄後、 2xl06/mlの細胞濃度に PBSにて懸濁した。雌性、 8〜10週齢の SCI Dマウス(日本クレア)の腹腔に、上記で調製した CD4陽性 CD45RB強陽性細胞を 200 μ 1ずつ、すなわち 4xl05/mouseで移入を行った。各群 7匹の CD4陽性 CD45RB強陽 性細胞を移入した SCIDマウスに、 500 μ gのコントロール抗体(ノヽムスター IgG)、 500 μ gの抗フラクタル力イン抗体 (5Η8-4) (抗体は PBS溶液)を、細胞移入後 2週間目より、 3日に 1回投与した。なお投与は尾静脈力も行った。また陰性対照群として、 CD4 T cell Isolation kit (ミルテュー社製)により精製し、 CD45RBの発現強度による分離をし て ヽな 、マウス CD4 Tリンパ球(トータル CD4)を 1.2xl06/mouseで移入した SCIDマウ ス 4匹を用いた。各抗体投与後 2週間後に剖検を行い体重減少、大腸内便性状をス コア化したもの、大腸肥厚、 iNOS mRNA発現解析、病理学的観察により評価を行つ た。便性状のスコアは表 1に示すようにデキストラン硫酸ナトリウム誘発大腸炎で用い られている便性状のスコア(Cooper et al., Lab. Invest., 69, 238-249, 1993)を使用し た。

[0054] また iNOS等の mRNA発現は、大腸より RNeasy mini kit (キアゲン社製)を用いて精製 した全 RNA (500 ng)を AMV逆転写酵素(TAKARA社製)とランダムへキサマー(TAK ARA社製)を用いて逆転写して得た cDNAをテンプレートとして、リアルタイム PCRにて 解析した。リアルタイム PCRは、各種プライマーと QuantiTect SYBR Green PCR kit ( Qiagen社製)の試薬、ゥラシル- DNA-グリコシラーゼ(Invitrogen社製)を混合した反応 液を調製し、 ABI PRISM 7700 Sequence Detector (Applied Biosystems社製)を用レヽ て行った。 PCRは、 50°C 2分、 95°C 15分の反応を行った後、 95°C 15秒および 60°C 1 分の反応を 35サイクル実施した。使用したプライマーのセットは以下のとおりである。

コントロール抗体投与群に対して、抗フラクタル力イン抗体 (5H8-4)投与群は、体重 減少が抑制された(図 1)。またコントロール抗体投与群に対して、抗フラクタル力イン 抗体 (5H8-4)投与群は、大腸内便性状スコアおよび大腸肥厚にぉ 、て改善を示し た(図 2)。大腸部位の形態的観察においても、コントロール抗体投与群では結腸か ら直腸部位において肥厚が観察され、抗フラクタル力イン抗体 (5H8-4)投与により改 善が認められた(図 3)。組織切片染色にお!ヽてコントロール抗体群では大腸粘膜層 に非常に多くの白血球 (Tリンパ球、単球、マクロファージなど)が浸潤し、粘液を産生 する杯細胞の消失や大腸上皮組織の損傷、過形成が認められるのに対し、抗フラタ タルカイン抗体 (5H8-4)投与群では、白血球の浸潤が抑制され大腸上皮細胞の損 傷も僅かであった(図 4)。一方 mRNA発現に関してもコントロール抗体投与群では iN OSの発現が上昇し、抗フラクタル力イン抗体 (5H8-4)によりその発現が顕著に抑制さ れた。また、パーフォリン、 Fas、 FasLなどの細胞障害性因子、 IFN y , TNF aなどの サイト力インの mRNAに関してもコントロール抗体投与群では発現が上昇し、抗フラタ タルカイン抗体 (5H8-4)投与群では発現の抑制が認められた(図 5)。以上の結果か ら、フラクタル力イン- CX3CR1経路力 CD4陽性 CD45RB強陽性(CD4+CD45RBhigh) Tリ ンパ球移入炎症性腸疾患モデルにおいて重要な役割を果たすことが明らかとなった 。その作用は iNOS産生を抑制することで過剰な NOの産生を抑制し、組織障害を軽 減しているものと推察された。つまりフラクタル力イン- CX3CR1の相互作用阻害に基 づく iNOS産生抑制は、炎症性腸疾患に有用な治療体系であることが示唆された。 実施例 2
マウスォキサゾロン誘発炎症性腸疾患モデルにおける抗フラクタル力イン抗体 (5H8- 4)の効果
(1)方法
マウスォキサゾロン誘発炎症性腸疾患モデルについては Iijima et al. , J. Exp. Med. , 199, 471-482, 2004を参考にした。雄性 8〜10週齢 Balb/cマウス(日本チャールズ' リバ一)の腹部を約 2cm四方剃毛した。 3% 4-エトキシメチレン- 2-フエ-ル- 2-ォキサ ゾリン- 5-オン(以下ォキサゾロン、シグマ社製)を含む 100%エタノール溶液を 150 μ 1 ずつ各マウスに塗布した。ォキサゾロン感作して力も 4日目に絶食し、 5日目にジェチ ルエーテル麻酔下のマウスの肛門力 約 3cmの部位に、 0.5%ォキサゾロンを含む 50% エタノール生理食塩水溶液を 100 1ずつマウスに腸注した。なお陰性対照群として 正常 Balb/cマウスに 50%エタノール生理食塩水溶液を 100 μ 1ずつ腸注したマウス 5匹 を設定した。各群 7匹のォキサゾロン腸注したマウスに、 500 /z gのコントロール抗体( ハムスター IgG)、 500 /z gの抗フラクタル力イン抗体(5H8-4) (抗体は PBS溶液)を投与 した。なお抗フラクタル力イン抗体 (5H8-4)投与は感作 5日目(ォキサゾロン腸注直前 )に行った。なお抗体投与は尾静脈力も行った。評価項目は疾患活動指数 [Disease Activity Index] (以下 DAI ;便の硬度、血の含有度合い、そして体重増減をスコア化し て DAI値として算出)、大腸の短縮化、大腸肥厚および病理学的観察により評価を行 つた。なお DAIは表 3に示すようにデキストラン硫酸ナトリウム誘発大腸炎で用いられ ている DAI (Cooper et al., Lab. Invest., 69, 238-249, 1993)を使用した。

実施例 3
[0061] マウスォキサゾロン誘発炎症性腸疾患モデルにおける抗フラクタル力イン抗体 (#126 )の効果
(1)抗フラクタル力イン抗体 (#126)の調製
マウスフラクタル力イン(Genzyme社製)と TiterMax™Goldアジュバンドを混合した後 、アルメニアハムスターに複数回免疫し、さらに最終免疫をマウスフラクタル力インの みで行った。血清中の抗体価を固相化したフラクタル力インを用いた ELISAで測定し 、抗体価が上昇したアルメニアハムスター力 リンパ球を分離し、リンパ球: P3ミエロー マ細胞の比率が 5:1になるように混合し、 PEG (Rosh社製)を用いて細胞融合を行った 。 ノヽイブリドーマは、 RPMI— 1640/10% FCS/HAT/10% Origen HCF (ISGN社製)を用い て、プレートで 1週間培養した。そして、培養上清を用いて固相化したフラクタルカイ ンを用いた ELISAを実施し、陽性ゥエルを同定した。抗フラクタル力イン抗体を産生す るハイブリドーマは、 2回の限界希釈によりクローユングを行った。モノクローナル抗体 は、プリスタンを投与した SCIDおよびヌードマウスにハイプリドーマを接種して作製し た腹水から、プロテイン Aカラムを用いて精製した。得られた抗体の中和活性は、 CX3 CR1発現細胞のマウスフラクタル力インに対する遊走を抑制することを指標にして測 定し、中和活性を有する #126抗体を得た。この中和抗体を産生するハイプリドーマを Ham ®mFKN#126.1.1と名づけた。

実施例 2と同様の方法を用い検討を行った。評価項目は大腸短縮および大腸肥厚 とした。各群 7匹のォキサゾロンを腸注したマウスに、 500 gのコントロール抗体(ノヽム スター IgG)、 500 gの抗フラクタル力イン抗体 (#126) (抗体は PBS溶液)を投与した。
[0063] (3)結果 抗フラクタル力イン抗体 (#126)は抗フラクタル力イン抗体 (5H8-4)と同様に、コント ロール抗体投与群に比較し、大腸短縮および大腸肥厚を改善した(図 10)。以上の 結果から、異なる 2種類の抗フラクタル力イン抗体がォキサゾロン誘発大腸炎モデル において効果を示したことは、フラクタル力イン- CX3CR1細胞浸潤系の阻害が炎症 性腸疾患治療に有用であることが強く示唆された。
実施例 4
[0064] ConA肝炎における iNOSの発現と抗フラクタル力イン抗体(5H8-4)の iNOS産生に対 する効果
(1)方法
1群 4匹の C57BL/6マウスに、 500 μ gのコントロール抗体(ノヽムスター IgG)、 500 g の抗フラクタル力イン抗体 (5H8-4) (抗体は PBS溶液)を静脈内投与し、直後にコンカ ナノくリン A(ConA、シグマアルドリッチ社製)を 12 mg/kgで静脈内投与した。その 2時 間後に肝臓を摘出し、 RNA抽出を行った。なお陰性対照群として C57BL/6マウスに P BS (リン酸バッファー)を静脈内投与したマウス 4匹を設定した。 iNOSの mRNA発現は 、実施例 1と同様の方法にて RNAを回収し、 cDNAを合成し、リアルタイム PCRにて測 定した。また、コントロール抗体あるいは抗フラクタル力イン抗体と ConA、陰性対照群 として PBSを静脈内投与して 12時間後の肝臓を摘出し、凍結組織切片を作製した。 抗 iNOS抗体と抗 CX3CR1抗体で免疫組織染色を行い、 iNOS, CX3CR1の陽性細胞 の数を測定した。

ConAの投与により上昇した iNOS mRNA発現は抗フラクタル力イン抗体の投与によ り顕著に減少した。また、グランザィム B、 Fasなどの細胞障害性因子、 IFN y , TNF a 、 IL-4などのサイト力インの mRNAに関しても、 ConAの投与により発現が上昇し、抗フ ラタタルカイン抗体 (5H8-4)の投与により発現の減少が認められた(図 11)。また、免 疫組織染色の結果から、 iNOS陽性の炎症性細胞は ConAの投与により増加し、抗フ ラタタルカイン抗体の投与により顕著に減少していた (図 12)。また、 CX3CR1陽性の 炎症性細胞も ConAの投与により増加し、抗フラクタル力イン抗体 (5H8-4)の投与によ り顕著に減少していた (図 12)。以上の結果力 フラクタル力インと CX3CR1の相互作 用を阻害することで、 ConA肝炎での肝臓における iNOS mRNAの発現抑制と、炎症 性細胞での iNOS産生が抑制されることが明ら力となり、炎症性細胞による過剰な NO の産生が阻害されることで、肝臓の組織障害が軽減されることが示唆された。
実施例 5
[0066] ConA肝炎における CX3CR1および iNOS発現細胞の解析
免疫組織染色の結果を詳細に解析した。 CX3CR1陽性細胞は主に丸く小型で、核 は馬蹄形で細胞質は少なぐ門脈 (portal vein)や中心静脈 (central vein),類洞 (sinus oid)に隣接しており、一部は壊死部 (necrotic region)の辺縁部でも認められた。 iNOS 陽性細胞は、主に大きく進展しており、核は馬蹄形で細胞質は大きぐ類洞に隣接し ており、壊死部の辺縁部に加え壊死部の中でも見られた力 門脈や中心静脈近傍で は見られなかった。このように、 CX3CR1陽性細胞と iNOS陽性細胞の形態や局在に は異なる点が見られたので、 CX3CR1および iNOS陽性細胞の特徴を詳細に解析した

実施例 4と同様に ConA静脈内投与 12時間後の肝臓 (ConA肝炎肝組織)を用い、 凍結組織切片を作製した。抗 CX3CR1抗体あるいは抗 iNOS抗体(BD Bioscience社 製)と抗 BM8抗体(BMA社製)、抗じ0111)抗体(68 3(^^6社製)、あるいは抗 MCP- 1 抗体(Gengyme Techne社製)で蛍光二重染色を行い、 CX3CR1陽性細胞および iNO S陽性細胞における BM8、 CDllb、 MCP- 1の陽性率を測定した。
[0068] 上記の抗 CX3CR1抗体は、抗原としてマウス CX3CR1 C末ペプチド(CSILSSFTHYT SEGDGSLLL (配列番号 31))を用いてゥサギで作製した抗体であった。具体的には 以下のようにして得た。配列番号 31の合成ペプチドを KLHと結合させ、ゥサギ (JW) にアジュバントと共に 1回あたり 250 g/羽免疫した。アジュバントは、初回のみフロイ ンド完全アジュバントを使用し、残りはフロインド不完全アジュバントを用いた。計 5回 免疫し、抗体価の上昇を、ペプチドを固相化した ELISAにより確認し、血清を回収し た。上記の配列のペプチドを Thiopropyl Sepharose 4B(Amersham Bioscience社製) に結合させたものを用いてァフィユティー精製を行 、、回収した血清カも抗 CX3CR1 抗体を得た。 [0069] さらに、末梢血(PBL)おける CX3CR1陽性細胞、および、 C57BL/6雄 9週令の骨髄 細胞を M-CSF (50ng/ml)で 5日間培養し、骨髄細胞力も分ィ匕させたマクロファージ (B MM φ )における CX3CR1陽性細胞を、マクロファージおよび単球に特異的な F4/80、 ならびに、未熟骨髄系細胞の表面マーカーである Ly6Cについて、抗 F4/80抗体 (CA LTAG社製)および抗 Ly6C抗体 (BMA社製)を用いて、フローサイトメトリー (FACS)に より解析した。また、抗 Ly6C抗体による ConA肝炎肝組織の組織染色、および、蛍光 標識したマクロファージを浸潤させた ConA肝炎肝組織の、抗 MCP-1抗体による組織 染色を行った。

代表的な染色像を図 13に示す。図中、写真の左および上の文字は、蛍光染色に 用いた抗体が認識するマーカーを示す。また、バーの長さは 20 /z mである。また、各 マーカーの陽性率の測定結果を図 14に示す。図中、 BM8+は BM8陽性、 CDllb+は CDllb陽性、 MCP1+は、 MCP- 1陽性を意味する。
[0071] CX3CR1陽性細胞は、丸く小型の CDllb陽性が 68.9 ±2.7% (n=3)、大型で進展した BM8陽性が 53.4±2.0% (n=3)であった。一方、 iNOS陽性細胞は、大型で進展した BM 8陽性が 81.6± 1.8% (n=3)、 CDllb陽性が 12.6±3.0% (n=3)であった。したがって、 CX 3CR1陽性細胞は iNOS陽性細胞と必ずしも同じ細胞ではなぐ一部は別の細胞集団 で発現していることが明ら力となった。最近、 iNOS産生細胞の浸潤には、 CCR2が関 与することが細菌感染モデルマウスにおいて報告されている(非特許文献 13)。そこ で、 CCR2のリガンドである MCP-1の発現を解析した。 MCP-1陽性細胞は、 CX3CR1 と同様に丸く小型であった。 CX3CR1陽性細胞の 72.5±3.7% (n=3)が MCP-1を発現し ており、逆に MCP-1陽性細胞のほとんどが CX3CR1陽性であった。したがって、 ConA 肝炎では、 CX3CR1陽性細胞は、 MCP-1を発現する主な細胞であることが明ら力とな つた。一方、 MCP- 1陽性細胞と iNOS陽性細胞は、形態が異なることから予想されるよ うに、 iNOS陽性細胞の 14.6±2.2% (n=3)しか MCP-1を発現しておらず、ほとんど一致 しなかった。したがって、 iNOS陽性細胞の多くは、 CX3CR1/MCP-1陽性細胞と異な ることが明ら力となった。興味深いことに、肝臓の類洞血管 (sinusoidal vessel)や壊死 部では、 iNOS陽性細胞と MCP-1陽性細胞は高頻度で隣接して存在して 、た。 [0072] CX3CR1陽性細胞の FACSによる解析結果を図 15に示す。また、組織染色の結果 を図 16に示す。
[0073] 末梢血(PBL)で CX3CR1陽性細胞を FACSで解析した結果、 CX3CR1は F4/80陽性 の単球で発現しており、発現量は Ly6C-成熟単球で強ぐ Ly6C+未熟単球で弱かつ た。 ConA投与 12時間後の肝臓を未熟骨髄系細胞の表面マーカーである Ly6Cで組 織染色した結果、 iNOS陽性の F4/80陽性細胞は Ly6C陽性であり、未熟単球由来で あることが示唆された。次に、 C57BL/6雄 9週令の骨髄細胞を M- CSF (50ng/ml)で 5 日間培養し、骨髄細胞力も分ィ匕させたマクロファージ (BMM φ )を得た。付着した BM M φを ImM EDTA/PBSで回収し FACSで解析を行った結果、末梢血成熟型単球と同 様の CX3CR1W F4/80+ Ly6C- /lowであった。そこで、 BMM φを CFSE 10 μ Μで蛍光 標識して 2.5χ106個/ 250 1の細胞をマウスに静注し、さらに 10分後に ConA (15mg/k g)を静注して 12時間後に肝臓を回収して浸潤を検討した。 ConA投与により、 ΒΜΜ の肝臓への浸潤は著しく増加した。また、浸潤した蛍光標識細胞は、 ConA投与によ り MCP-1を産生していた。 iNOSは、蛍光標識細胞では発現せず、宿主由来であった
[0074] 以上の結果から、炎症時には CX3CR1陽性成熟型単球は炎症局所に浸潤して MC P-1を発現し、 iNOS産生未熟型単球の炎症局所への浸潤を誘導することで病態を形 成することが示唆された。
実施例 6

(1)方法
実施例 4で作製した ConAを静脈内投与 2時間後の肝臓由来 cDNAを用い、実施例 1と同様の方法にて、ケモカインの mRNA発現をリアルタイム PCRにて測定した。使用 したプライマーのセットは以下のとおりであった。コントロール遺伝子として、ダリセル アルデヒド- 3-リン酸デヒドロゲナーゼ (G3PDH)遺伝子を用いた。

結果を図 17に示す。図中、「normal」は陰性対照、「control」はコントロール抗体投 与群、「ant卜 FKN」は抗フラクタル力イン投与群を意味する。
ConAの投与により 2時間後に肝臓での MCP- 1、 KC、 MIP- 2、 IP- 10、 MIP-1 a、 MIP -1 βの mRNAの発現が上昇した。これらのケモカインの mRNA発現は抗フラクタル力 イン抗体の投与により減少した。以上の結果から、フラクタル力インと CX3CR1の相互 作用を阻害することで、多種類のケモカインの産生が抑制され、炎症の初期誘導反 応である炎症性細胞浸潤と活性化の増幅回路が広範囲に作動しなくなることが示唆 された。

コントロール抗体投与群では CD4、 T- bet、 F4/80、 M-CSF receptor (M-CSF R)、 L y-6G、 CDllcなどの各種白血球マーカーの mRNA発現が亢進していた(図 18)。これ らの結果から、実施例 1の組織切片染色における、コントロール抗体投与群でのおび ただしい数の浸潤した白血球は CD4 + T細胞(特に Thlタイプ CD4+ T細胞)や単球/ マクロファージ、好中球、樹状細胞など多種類の白血球力も構成されている可能性 が示唆された。また各々のケモカイン-ケモカイン受容体、 KC、 MIP- 2、 CXCR2、 MC P— 1、 MCP— 3、 CCR2、 MIP— 1 a、 MIP— 1 β、 CCR5、 IP— 10、 I— TAC、 CXCR3、 TARC、 MDC、 CCR4の mRNA発現亢進がコントロール抗体投与群において認められたことか ら、浸潤した白血球はこれらケモカインーケモカイン受容体機構を介して浸潤したも のと推察された(図 19、図 20)。さらに一連の炎症性サイト力イン、 IL-1 β、 IL-6、 IL- 12 a、 11-12 β、 11-23 a、 11-17, RANKLや組織破壊に関与するプロテアーゼ MMP- 2 、 MMP-9、 MMP-14さらに炎症部位での血管新生に関与する bFGFや VEGF Recepto r 2の mRNA発現もコントロール抗体投与群にお 、て顕著な亢進が認められた(図 21 、図 22)。また榭状細胞などの活性ィ匕受容体である CD40や TLR2、活性化 T細胞で 発現し、榭状細胞などを活性化する分子 CD40L、榭状細胞の活性ィ匕により発現が増 強する CD80、 CD86などの T細胞活性化補助シグナル分子群および MHC Class IIの 発現亢進が認められたことから、 T細胞および榭状細胞などの抗原提示細胞が病変 部位において活性化され、抗原特異的な獲得免疫系が作動している可能性が示唆 された(図 23)。これら一連の白血球マーカー、ケモカインーケモカイン受容体、サイ トカイン、組織破壊分子、血管新生関連分子、 T細胞 '榭状細胞活性化分子の mRNA 発現は、抗フラクタル力イン抗体投与により顕著に抑制されていた(図 18〜図 23)。 以上の結果から、フラクタル力インと CX3CR1の相互作用を阻害することで、炎症性サ イトカインゃケモカインの産生が抑制され、炎症性細胞浸潤と活性ィ匕の増幅回路が 広範囲に作動しなくなり、炎症性腸疾患の病態進展が抑制される可能性が示唆され た。
実施例 8
マウスォキサゾロン誘発炎症性腸疾患モデルにおける、細胞表面マーカー、サイト力 イン、ケモカイン、細胞活性化および細胞障害性分子の mRNA発現と抗フラクタル力 イン抗体 (5H8-4)の細胞表面マーカー、サイト力イン、ケモカイン、細胞活性化および 細胞障害性分子の mRNA発現に対する効果
( 1)方法
実施例 2と同様の方法により炎症性腸疾患モデルを作製した。コントロール抗体、 坑フラクタル力イン抗体 (5H8-4)を実施例 2の方法に従 ヽ投与した。ォキサゾロン腸 注後 24時間および 3日目に剖検を行い、大腸を摘出後、大腸組織より実施例 1と同様 の方法を用い RNAを抽出した。陰性対照群には 50%エタノール生理食塩水を腸注し た。 RNAを抽出後、実施例 1と同様の方法を用い cDNAを合成しサイト力イン、ケモカ インの mRNA発現を TaqMan PCRにて測定した。使用したプライマーのセットは以下 のとおりである。

ォキサゾロン腸注 24時間後において、コントロール抗体投与群では IL-1 β、 IL-6な どの炎症性サイト力イン、 KC、 MIP-2, MCP-1 , MCP-3, MIP- 1 a、 MIP-1 βなどのケ モカイン、ケモカイン受容体 CXCR2の mRNA発現の亢進が認められ、この発現亢進 は抗フラクタル力イン抗体の投与により顕著に抑制された(図 24)。ォキサゾロン腸注 3日後においては、 Iし- 1 β、 IL- 6、 KC、 MIP- 2、 MCP- 1、 MCP- 3、 CXCR2に加え、好 中球のマーカーである Ly-6Gの mRNA発現の亢進が認められ、この発現亢進は抗フ ラタタルカイン抗体の投与により顕著に抑制された(図 25)。以上の結果から、坑フラ クタルカイン抗体のォキサゾロン誘発炎症性腸疾患モデルにおける病態改善効果の 一部は、 IL-1 βや IL-6の炎症性サイトカインゃケモカインの産生が抑制されること、 好中球の浸潤が抑制されることで発揮されることが示唆された。
産業上の利用の可能性
炎症性細胞による iNOS活性の産生抑制による、過剰な NO産生を選択的に阻害す るという新しいアプローチに基づく炎症性疾患治療剤が提供される。

Claims

請求の範囲
[I] フラクタル力インと CX3CR1との相互作用を阻害する抗体または CX3CR1アンタゴニス トを含有することを特徴とする、炎症性疾患治療剤。
[2] 抗体が、抗フラクタル力イン抗体であることを特徴とする、請求項 1記載の剤。
[3] 抗フラクタル力イン抗体力 モノクローナル抗体であることを特徴とする、請求項 2記 載の剤。
[4] 抗フラクタル力イン抗体が、受託番号 FERM BP-10372のハイブリドーマによって産 生されるモノクローナル抗体 5H8-4、または受託番号 FERM BP-10371のハイブリド 一マによって産生されるモノクローナル抗体 #126であることを特徴とする、請求項 3記 載の剤。
[5] 炎症性疾患が炎症性腸疾患である請求項 1〜4の 、ずれか一項に記載の剤。
[6] 炎症性疾患が潰瘍性大腸炎またはクローン病である請求項 5記載の剤。
[7] 炎症性疾患が乾癬である請求項 1〜4の 、ずれか一項に記載の剤。
[8] 炎症性疾患がアトピー性皮膚炎である請求項 1〜4の 、ずれか一項に記載の剤。
[9] 炎症性疾患が喘息である請求項 1〜4の 、ずれか一項に記載の剤。
[10] 炎症性疾患が動脈硬化症である請求項 1〜4の 、ずれか一項に記載の剤。
[II] 炎症性疾患が虚血再灌流における組織障害である請求項 1〜4のいずれか一項に 記載の剤。
