Abstract

 従来達成し得なかったような高光学純度で乳酸を製造する。 乳酸とグリセロールとが併存した場合に乳酸のラセミ化反応が進行する結果、乳酸の光学純度が低下していることを見出した。乳酸を加熱濃縮する前にグリセロール量を低減させることで、加熱濃縮後の乳酸の光学純度を高く維持することができる。

Description

乳酸の製造方法 技術分野
本発明は、 ポリ乳酸等を製造する際の材料となる乳酸を、 高光学純度で製造す ることができる乳酸の製造方法に関する。 明
背景技術
ポリ乳酸は、 生体内で分解されるポリマーであり機械的特性等にも優れている 書
ことから医療分野で利用されてきたが、 自然環境下においても分解されるので環 境保護の観点から種々の用途に利用されることが期待されている。 '
ポリ乳酸の製造方法としては、 原料である乳酸を直接脱水縮合させる方法、 乳 酸エステルを脱アルコール縮合させる方法、 及びラクチドを開環重合させる方法 が挙げられる。 いずれの方法においても、 光学純度の高い乳酸を使用することに よって優れた物性のポリ乳酸を製造することができる。
ところで、 乳酸を製造する方法としては、 乳酸生合成系を有する微生物、 或い は乳酸生合成系を付与した微生物を用いた発酵法を挙げることができる。 発酵法 によれば、 遺伝子の構造上、 L-乳酸又は D-乳酸のいずれか一方のみを生産する菌 株を使用することによって、 上述したように、 光学純度の高い乳酸を原料とした ポリ乳酸の製造を達成する.ことができると考えられている。
しかしながら、 ポリ乳酸に対して、 より優れた物性が求められる場合、 従来の 発酵法において L-乳酸又は D-乳酸のいずれか一方のみを生産する菌株を使用し たとしても、 十分な光学純度の乳酸を準備することが困難であった。 発明の開示
そこで、 本発明は、 上述したような実状に鑑み、 例えば、 優れた物性を持つポ リ乳酸の原料としても利用可能な高光学純度で乳酸を製造できる乳酸の製造方法 を提供することを目的としている。 上記目的を達成するため、 本発明者らが鋭意検討した結果、 乳酸とグリセロー ルとが併存した場合に乳酸のラセミ化反応が進行する結果、 乳酸の光学純度が低 下していることを見出し、 本発明を完成するに至った。
すなわち、 本発明は以下を包含する。
(1) グリセロール量を減少させた溶液で、 当該溶液中の乳酸を加熱濃縮するェ 程を含む乳酸の製造方法。
(2) グリセ口ール生産能を低下させた微生物を使用した乳酸発酵によって上記 溶液を調製する工程を更に含むことを特徴とする (1) 記載の乳酸の製造方法。
(3) 上記微生物は、 グリセロール生産に関連する遺伝子のうち少なく とも 1つ の遺伝子の発現を抑制した変異体であることを特徴とする (2) 記載の乳酸の製 造方法。
(4)上記変異体は、 グリセロール- 3-リン酸脱水素酵素をコードする遺伝子を破 壊した乳酸生産菌であることを特徴とする (3) 記載の乳酸の製造方法。
(5) 上記乳酸生産菌は、 サッカロマイセス属に属する微生物であることを特徴 とする (4) 記載の乳酸の製造方法。
(6) 上記溶液は、 乳酸量に対するグリセロール量が 3.5重量%以下、 より好ま しくは 0.1重量%以下であることを特徴とする (1) 記載の乳酸の製造方法
(7) 微生物を使用した乳酸発酵によって上記溶液を調製する工程と、 上記溶液 からグリセロールを除去する工程とを更に含むことを特徴とする (1) 記載の乳 酸の製造方法。
(8) 上記グリセロールを除去する工程では、 上記溶液中の乳酸量に対するダリ セロール量を 3.5重量%以下、 より好ましくは 0.1重量%以下とすることを特徴 とする (7) 記載の乳酸の製造方法。
(9) 乳酸生産菌に対してグリセロール生産量が低減するような変異を導入して なる変異体。
(1 0) 上記乳酸生産菌がサッカロマイセス属に属する微生物であることを特徴 とする (9) 記載の変異体。
( 1 1)グリセロール- 3-リン酸脱水素酵素をコードする遺伝子を破壊することで グリセロール生産量を低減させたことを特徴とする (9) 記載の変異体。 (1 2) 乳酸生産菌に対して、 乳酸量に対するグリセロール生産量を 3.5重量% 以下、 より好ましくは 0.1重量%以下に低減するような変異を導入してなる ( 9 ) 記載の変異体。
また、 本発明者は、 乳酸発酵においてグリセロール生産能を低下させた微生物 を使用することによって、 乳酸の生産効率が向上することを見出し、 本発明を完 成するに至った。 すなわち、 本発明は以下を包含する。
(1 3) グリセロール生産能を低下させた微生物を使用した乳酸発酵によって、 乳酸を製造する工程を含む乳酸の製造方法。
(1 4) 上記微生物は、 グリセロール生産に関連する遺伝子のうち少なく とも 1 つの遺伝子の発現を抑制した変異体であることを特徴とする (1 3) 記載の乳酸 の製造方法。
(1 5)上記変異体は、 グリセロール- 3-リン酸脱水素酵素を破壊した乳酸生産菌 であることを特徴とする (14) 記載の乳酸の製造方法。
(1 6) 上記乳酸生産菌は、 サッカロマイセス属に属する微生物であることを特 徴とする (1 5) 記載の乳酸の製造方法。
(1 7) 上記乳酸生産菌は、 乳酸量に対するグリセロール生産量を 3.5重量%以 上、 より好ましくは 0.1重量%以上低減するような変異が導入されたものである ことを特徴とする (1 5) 記載の乳酸の製造方法。

発明を実施するための最良の形態
以下、 図面を参照して本発明をより詳細に説明する。
本発明に係る乳酸の製造方法は、 グリセロールを低減させた溶液で乳酸を加熱 濃縮する工程を含むものである。 特に本発明は、 発酵法による乳酸の製造に適用 される。 発酵法とは、 培地中の糖質が微生物の作用を受けて乳酸を生成する現象 を意味する。 以下の説明において、 乳酸を生成する能力を有する微生物及び乳酸 を生成する能力が付与された微生物を併せて 「乳酸生産菌」 と称する。
また、 本発明において、 「グリセロールを低減させる」 とは、 発酵法による乳酸 生産菌のグリセロール生産能を低下させること、 乳酸生産菌が生産したグリセ口 ールを除去及び/又は分解することのいずれか一方及び両方を意味する。乳酸とグ リセロールとが共存する場合には、 以下の反応によって、 乳酸のラセミ化が進行 することが判明した。

また、 上記反応は、 生成した乳酸を加熱濃縮する工程や加熱エステル化するェ 程及び生成した乳酸を加熱蒸留する工程において加えられる熱エネルギによって 進行し、 光学純度が低下してしまう。 よって、 生成した乳酸を加熱する工程に先 立って、 グリセロールを低減させることにより高光学純度の乳酸を得ることがで きる。
以下、 グリセロールを低減させる方法として、 発酵法による乳酸生産菌のダリ セロール生産能を低下させる方法 (方法 D 及び乳酸生産菌が生産したグリセ口 ールを除去及び/又は分解する方法 (方法 2 ) を順に説明する。
方法 1
乳酸生産菌のグリセロール生産能を低下させる場合、
1)乳酸生産菌が有するグリセ口ール生産に関連する遺伝子を破壊する
2)グリセ口ール生産に関連する遺伝子の発現を抑制する
3)グリセロール生産に関連する遺伝子によりコードされるタンパク質の活性を阻 害する
4)グリセ口ールの代謝分解能力を向上させる
5)グリセ口ールの細胞膜外への排出を抑える
6)グリセロールの細胞膜内への取り込みを促進させる
7)グリセロールの生産量が低下した突然変異株を取得する
8)グリセ口ールの生産量が低下する化合物を培養培地に添加する
といった 1)〜8)の方法を挙げることができる。これら 1)〜8)のいずれか 1つの方 法で乳酸生産菌のグリセロール生産能を低下させても良いし、 1)〜8)のうち複数 の方法を組み合わせた方法で乳酸生産菌のグリセ口ール生産能を低下させても良 レ、。
ここで、 本発明に係る乳酸の製造方法に使用可能な乳酸生産菌としては、 乳酸 を生成する能力を有する微生物である細菌、 酵母、 カビが挙げられる。 細菌とし て ίま、 ラク 卜ノ チノレス (Lactobaci l lus)属、ス トレプ卜コッカス (Streptococcus) •属、 ノ チノレス (Bac i l lus) 属、 ロイコノス卜ック (Leuconostoc) 属、 ぺディ才: π ッカス (Pediococcus) 属のものを例示できる。 酵母としては、 クルベルマイセス (Kluyveromyces) 属のものを例示できる。 カビとしては、 リゾプス (Rhyzopus) 属、 ァスペルギルス (Aspergi l lus) 属のものを例示できる。 この乳酸発酵微生物 としては、 特に、 ホモ乳酸発酵能を有する微生物を使用するのが好ましい。
また、 乳酸を生成する能力を付与した微生物とは、 本来的に乳酸生成能を有し ないが、 遺伝子工学的手法によって乳酸を生成する能力を有するように改変され た微生物を意味する。 例えば、 サツカロマイセス ' セレピシェに対して、 乳酸生 成に関連する遺伝子を導入した変異酵母を挙げることができる。 また、 変異酵母 以外にも、 乳酸生成能を持たない細菌、 酵母、 カビに乳酸生成に関連する遺伝子 を導入する こ とで、 使用可能である。 具体的にはサッカ ロマイ セス (Saccharomyces)、 シゾサッカロマづ セス (Schi zosaccharomyces)、 クノレへノレマ イセス(Kluyveromyces)、 ピキア(Pichia)、 ハンゼヌラ(Hansenula)、 カンジダ (Candida) , 卜リクスポロン(Trichosporon)、 またはヤマダジマ(Yamadazyma)属の ものを例示できる。 また細菌では大腸菌 (Escherichia col i)、 ザィモモナス属 (Zymomonas) やコリネ型細菌を例示できる。 カビではリゾプス (Rhyzopus) 属、 ァスペルギルス (Aspergi l lus) 属、 ムコール (Mucor) 属などを例示できる。 乳酸生成に関連する遺伝子としては、 乳酸脱水素酵素活性を有するたんぱく質 をコードする遺伝子 (LDH遺伝子) が挙げられる。 乳酸脱水素酵素 (LDH) として は、 生物の種類に応じてあるいは生体内において各種同族体が存在する。 本発明 において使用する乳酸脱水素酵素としては天然由来の LDHの他、 化学合成的ある いは遺伝子工学的に人工的に合成された LDHも包含している。 LDH としては、 好 ましくは、 ラク トバチルス 'ヘルべテイクス、 ラク トバチルス ·力ゼィ、 クルべ ノレマイセス . サーモトレランス、 トノレラスポラ . デノレブノレツキ、 シゾサッカロマ イセス ·ボンべ、 リゾプス ·ォリゼなどの原核生物もしくは力ビなどの真核生物 由来であり,より好ましくは、 植物、 動物、 昆虫などの高等真核生物由来である。 例えば、 ゥシ由来の LDH (L-LDH) である。 これら遺伝子を、 上述した酵母を始め とする本来的に乳酸生成能を有しない微生物に導入することによって、 当該微生 物に乳酸生成能を付与することができる。 本発明に係る乳酸の製造方法では、 こ のようにして得られた乳酸を生成する能力を付与した微生物を広く使用すること ができる。
1)乳酸生産菌が有するグリセ口ール生産に関連する遺伝子を破壊する方法 グリセロール生産とは、 微生物における解糖経路で生成するァセトアルデヒ ド をアルコール脱水素酵素の反応系外におくことで、 NADH の酸化がグリセロール -3 -リン酸脱水素酵素で行われるように発酵変換を誘導し、その結果、 グリセロー ルが生成、蓄積されることを意味する。グリセロール生産に関連する遺伝子とは、 上述したグリセロール生産の各反応に寄与する酵素をコードする遺伝子を意味す る。
グリセロール生産に関連する遺伝子としては、例えば、 グリセロール- 3-リン酸 脱水素酵素遺伝子、 グリセ口ール -1-リン酸脱リン酸化酵素遺伝子、 グリセ口ール キナーゼ遺伝子を挙げることができる。 より具体的には、 サッカロマイセス .セ レビシェにおいては、 GPD1遺伝子及ぴ GPD2遺伝子 (グリセロール- 3-リン酸脱水 素酵素遺伝子)、 RHR2遺伝子及び H0R2遺伝子 (グリセロール- 1-リン酸脱リン酸 化酵素遺伝子)、 GPP1遺伝子及び GPP2遺伝子 (グリセロールキナーゼ遺伝子) を 挙げることができる。
なお、 サッカロマイセス ' セレピシェにおける GPD1遺伝子及び GPD2遺伝子に ついては、 それぞれの遺伝子を単独で破壊した株及び両遺伝子を破壊した株にお いて、 グリセロール生産を低減することができる (Nissen, T. L.,ら、 Yeast 16, 463-474 (2000) ) 0 サッカロマイセス ·セレビシェにおける RHR2遺伝子及び H0R2 遺伝子については、 両遺伝子を破壊した株において、 グリセロール生産を低減す ることができる (Pahlman, A. K.,ら、 J. Biol. Chem. 276, 3555 - 3563 (2001) )。 乳酸生産菌において、 上述した遺伝子を破壊する方法としては、 特に限定され ないが、 当該遺伝子をゲノムから欠失させる方法、 当該遺伝子に外来 DNA断片を 揷入する方法等を挙げることができる。
以上のようにして、 グリセロール生産に関連する遺伝子を破壊することによつ て、 乳酸生産菌におけるグリセ口ールの生産を抑制することができる。
2)グリセロール生産に関連する遺伝子の発現を抑制する方法
グリセロール生産に関連する遺伝子の発現を抑制する方法とは、 上述した遺伝 子を破壊する方法を除く方法であって、 当該遺伝子の発現を抑制する方法を意味 する。 遺伝子の発現を抑制する方法としては、 上述した遺伝子の転写を抑制する 方法、 上述した遺伝子の転写後において当該遺伝子の翻訳を阻害する方法、 当該 遺伝子の mRNAを選択的に分解する方法等を挙げることができる。
より具体的に、 上述した遺伝子の転写を抑制する方法としては、 上述した遺伝 子の転写制御領域をゲノムから欠失させる方法、 当該遺伝子の転写制御領域に外 来 DNA断片を揷入する方法を挙げることができる。 また、 RNAデコイをコードす る核酸を細胞内に導入することによって、 上述した遺伝子の発現を転写レベルで 抑制することができる。 当該 RNAデコイとは、 転写因子の結合タンパク質をコー ドする遺伝子あるいは転写因子の結合部位の配列又は類似の配列を有する RNAを 指し、 これらを 「おとり」 として細胞内に導入することによって転写因子の作用 を抑制するものをいう。
一方、 上述した遺伝子の翻訳を阻害する方法としては、 アンチセンス RNA法を 挙げることができる。 アンチセンス RNA法とは、 mRNAの一部又は全部にハイブリ ダイズするアンチセンス RNAを導入する力 \ 当該アンチセンス RNAをコードする DNA断片を宿主ゲノムに導入する方法を意味する。 アンチセンス RNA とは、 対象 となる mRNAに相補的な塩基配列を有し、これと 2本鎖を形成することによって該 mRNAにコードされる遺伝子の発現を翻訳レベルで抑制するものをいう。 なお、 ァ ンチセンス RNAの代わりに、 アンチセンス DNAを使用することによって、 新規遺 伝子の発現を転写レベルで抑制することもできる。 何れの場合であってもアンチ センス配列は、 遺伝子の翻訳又は転写をブロックする限り任意の核酸物質を使用 することができる。 例えば、 DNA、 RNA, 又は任意の核酸擬似物が挙げられる。 従 つて、 発現を抑制させようとする新規遺伝子の一部の配列に相補的となるように アンチセンス核酸 (オリゴヌクレオチド) 配列を設計すればよい。 なお、 アンチ センスオリゴヌクレオチドの分子類似体も使用することができる。分子類似体は、 高安定性、 分布特異性などを有するものである。 分子類似体には、 化学的に反応 性である基、 例えば鉄結合エチレンジァミン四酢酸をアンチセンスオリゴヌクレ ォチドに結合させたものが挙げられる。
さらにまた、 リボザィムを使用することによって、 上述した遺伝子の発現を翻 訳レベルで抑制することができる。 リボザィムとは、 特定のタンパク質の mRNA を切断するものをいい、 これら特定のタンパク質の翻訳を阻害するものをいう。 リボザィムは特定のタンパク質をコードする遺伝子配列より設計することができ, 例えば、ハンマーへッド型リボザィムとしては、 FEBS Letter, 228; 228-230 ( 1988) に記載の方法を用いることができる。 また、 ハンマーヘッド型リボザィムだけで はなく、 ヘアピン型リボザィム、 デルタ型リボザィムなどの、 特定のタンパク質 の mRNAを切断するものであって、これら特定のタンパク質の翻訳を阻害するもの であれば本発明において使用しうる。
上述した遺伝子の mRNAを選択的に分解する方法としては、 RNA干渉を利用する 方法を挙げることができる。 RNA干渉とは、二本鎖を形成している RNA (以下、 「二 本鎖 RNA」 又は 「dsRNA」 ともいう) が細胞内に存在する場合に、 その RNAの塩基 配列に相同的な内在性の mRNAが分解される結果、 当該 mRNAからの遺伝子発現が 特異的に抑制される現象である。 RNA 干渉は、 RNA インターフェアランス、 RNAi とも称される。 RNA 千渉の原理を利用するための遺伝子としては、 ヘアピン型 dsRNA等の宿主内で二本鎖 RMを形成するように、 発現を抑制したい遺伝子の塩 基配列に基づいて設計される。
以上のようにして、 グリセロール生産に関連する遺伝子の発現を抑制すること によって、 乳酸生産菌におけるグリセロールの生産を抑制することができる。
3)グリセロール生産に関連する遺伝子によりコードされるタンパク質の活性を阻 害する方法
上述したグリセロール生産に関連する遺伝子によりコードされる酵素の活性を 阻害することによって、 乳酸生産菌におけるグリセロールの生産を抑制すること ができる。 具体的には、 前記酵素に対する抗体を使用したり、 前記酵素に特異的 に作用する物質を使用することができる。
抗体は、 従来公知の手法を適用して取得することができ、 前記酵素の活性を阻 害するものであれば、 その由来、 種類 (モノクローナル、 ポリクローナル) およ び形状を問うものではない。 例えば、 前記酵素を抗原とし、 当該抗原に結合する 限り、前記抗体としては特に制限はなく、マウス抗体、 ラット抗体、 ゥサギ抗体、 ヒッジ抗体等を適宜用いることができる。 抗体は、 ポリクローナル抗体であって もモノクローナル抗体であってもよいが、 均質な抗体を安定に生産できる点でモ ノクローナル抗体が好ましい。 ポリクローナル抗体およびモノクローナル抗体は 当業者に周知の方法により作製することができる。
モノクローナル抗体を産生するハイプリ ドーマは、 基本的には公知技術を使用 し、 以下のようにして作製できる。 すなわち、 所望の抗原や所望の抗原を発現す る細胞を感作抗原として使用して、 これを通常の免疫方法にしたがって免疫し、 得られる免疫細胞を通常の細胞融合法によって公知の親細胞と融合させ、 通常の スク リーニング法により、 モノクローナルな抗体産生細胞 (ハイブリ ドーマ) を スクリーニングすることによって作製できる。 ハイブリ ドーマの作製は、 たとえ ίま *、 ミノレスティンらの方法 (Kohler. G. and i lstein, C., Methods Enzymol. ' (1981) 73 : 3-46 ) 等に準じて行うことができる。
一方、 阻害剤としては、 上述したグリセロール生産に関連する遺伝子によりコ 一ドされる酵素の活性を特異的に阻害する機能を有する物質を使用することがで さる。
以上のようにして、 グリセロール生産に関連する遺伝子によりコードされる酵 素の活性を阻害することによって、 乳酸生産菌におけるグリセロールの生産を抑 制することができる。
4)グリセロールの代謝分解能力を向上させる
グリセ口ールの代謝分解能力を向上させる場合、 グリセ口ールの代謝に関連す る遺伝子を過剰発現する方法を挙げることができる。 グリセ口ールの代謝に関連 する遺伝子としては、 例えば、 グリセロールリン酸化酵素遺伝子、 グリセロール -3 -リン酸脱水素酵素遺伝子を挙げることができる。 また、 サッカロマイセス *セ レビシェにおいては、 GUT1 遺伝子 (グリセロールリン酸化酵素)、 GUT2 (グリセ ロール- 3-リン酸脱水素酵素遺伝子)、 GCY1 (グリセロール脱水素酵素)、 DAK1 (ジ ヒ ドロアセトンリン酸化酵素) を挙げることができる。
乳酸生産菌において、 上述した遺伝子を導入する方法は特に限定されないが、 当該遺伝子を組み込んだ線状等の DNA断片、プラスミ ド(DNA)、 ウィルス (DNA)、 レトロ トランスポゾン (DNA)、 人工染色体 (YAC) を、 外来遺伝子の導入形態 (染 色体外あるいは染色体内) 等に応じて選択して組換えベクターを作成し、 乳酸生 産菌に導入させることができる。
以上のようにして、 グリセロールの代謝分解能力を向上させることによって、 乳酸生産菌におけるグリセロールの生産を抑制することができる。
5)グリセロールの細胞膜外への排出を抑える
グリセ口ールの細胞膜外への排出を抑える場合、 グリセ口ールの細胞膜外への 排出チャンネルをコードする遺伝子を破壊する方法を挙げることができる。 グリ セロールの細胞膜外への排出チャンネルをコードする遺伝子としては、 サッカロ マイセス 'セレピシェにおいて、 FPS1遺伝子を挙げることができる。
乳酸生産菌において、 上述した遺伝子を破壊する方法としては特に限定されな いが、 当該遺伝子をゲノムから欠失させる方法、 当該遺伝子に外来 DNA断片を挿 入する方法、 当該遺伝子の発現タンパクの活性を低下させる変異を導入する方法 等を挙げることができる。
また、 上記 「2) グリセロール生産に関連する遺伝子の発現を抑制する方法」 で 説明した方法に準じて、 グリセ口ールの細胞膜外への排出チャンネルをコードす る遺伝子の発現を抑制する方法を採用することもできる。 同様に、 上記 「3) ダリ セロール生産に関連する遺伝子によりコードされるタンパク質の活性を阻害する 方法」 で説明した方法に準じて、 グリセロールの細胞膜外への排出チャンネルを コードする遺伝子によりコードされるタンパク質の活性を阻害する方法を採用す ることもできる。
以上のようにして、 グリセ口ールの細胞膜外への排出を抑えることによって、 乳酸生産菌におけるグリセ口ールの生産を抑制することができる。
6)グリセロールの細胞膜内への取り込みを促進させる
グリセ口ールの細胞膜内への取り込みを促進させる場合、 グリセ口一ルの取り 込みポンプをコードする遺伝子を過剰発現する方法を挙げることができる。 ダリ セロールの取り込みポンプをコードする遺伝子としては、 サッカロマイセス 'セ レビシェにおいて、 GUP1、 GUP2遺伝子を挙げることができる。
以上のようにして、 グリセロールの細胞膜内への取り込みを促進させることに よって、 乳酸生産菌におけるグリセ口ールの生産を抑制することができる。
7)グリセロールの生産量が低下した突然変異株を取得する
グリセロールの生産量が低下した突然変異株を取得する場合、 変異酵母を得る ための変異方法は、 いかなる方法でも良い。 変異の物理的方法としては、 紫外線 照射、 放射線照射などがあり、 化学的方法としては、 変異剤、 例えば、 ェチルメ タンスルホネート、 N —メチル一N —ニトログァニジン、 亜硝酸、 アタリジン 系色素などの溶液に懸濁させる変異方法がある。 また、 取得頻度は低くなるが、 自然変異においても目的とする変異酵母を取得することができる。
以上のようにして取得されたグリセロールの生産量が低下した突然変異株にお いては、 グリセロールの生産が抑制されたものとなる。 ここで、 突然変異株とし ては、 グリセロール生合成に関与する遺伝子に対する変異の結果としてグリセ口
—ル生産量が低下した株であっても良いし、グリセロール代謝に関与する遺伝子、 グリセロールの細胞外への排出に関与する遺伝子、 グリセロールの細胞膜内への 取り込みに関与する遺伝子煮たい汁変異の結果としてグリセロール生産量が低下 した株であっても良く、 変異の導入部位について何ら限定されるものではない。
8)グリセロールの生産量が低下する化合物を培養培地に添加する
グリセ口ールの生産量が低下する化合物を培養培地に添加する場合、 サッカロ マイセス ·セレビシェにおいては、 イノシトール、 カテキン、 二亜硫酸ナトリ ウ ム、 抗酸化剤等を培養培地に添加することで、 グリセロール生産量が低下するこ とカ知られてレヽる (Caridi, A. (2002) . Protect ive agents used to reverse the metabol ic changes induced in wine yeasts by concomitant osmotic and thermal stress. Lett Appl Microbiol 35, 98—101)。 また、 これら以外のク、、リセローノレの 生産量が低下する化合物を添加しても良い。
以上のようにして、 グリセロールの生産量が低下する化合物を培養培地に添加 することによって、 乳酸生産菌におけるグリセロールの生産を抑制することがで さる。
なお、上述した 1 )〜8)の方法において、乳酸生産菌の培養条件及び培地組成は、 特に限定されず、通常の培養条件及び培地組成を適用することができる。例えば、 乳酸生産菌の一例として、 乳酸生産能を付与したサッカロマイセス 'セレビシェ TC38株 (GPD1遺伝子、 GPD2遺伝子破壊株) を用いた場合、 培養条件としては、 通常、震盪培養または通気攪拌培養等の好気条件下、 25〜38°Cで 12〜80時間行う。 培養期間中、 pHは 2. 0〜7. 0に保持することが好ましい。 また、 pHの調整は、 無 機あるいは有機酸、 アルカリ溶液等を用いて行うことができる。 培養中は必要に 応じてハイグロマイシン、 G418などの抗生物質を培地に添加することができる。 また、 培地組成としては、 微生物が資化可能な炭素源、 窒素源、 無機塩類を含 有している培地であれば、天然培地、合成培地のいずれも使用することができる。 炭素源、と しては、 グルコース、 フルク トース、 スクロース、 デンプン、 セノレロー ス等の炭水化物、 酢酸、 プロピオン酸等の有機酸、 エタノール、 プロパノール等 のアルコールの他、糖蜜、木質バイオマスの加水分解物等を用いることができる。 窒素源としては、 アンモニア、 塩化アンモニゥム、 硫酸アンモニゥム、 酢酸アン モニゥム、 リン酸アンモニゥム等の無機塩もしくは有機酸のアンモニゥム塩また はその他の含窒素化合物の他、 ペプトン、 肉エキス、 コーンスティープリカ一、 酵母エキス等を用いることができる。 無機物としては、 リン酸第一カリ ウム、 リ ン酸マグネシウム、 硫酸マグネシウム、 塩化ナトリ ウム、 硫酸第一鉄、 硫酸マン ガン、 硫酸銅、 炭酸カルシウム、 などを用いることができる。 また、 チアミン、 ピオチン、 葉酸、 ナイァシン、 リボフラビン、 ピリ ドキシン、 パントテン酸等の ビタミン類も培地に添加することができる。
また、他の菌を用いる場合の培養条件は、 細菌発酵 に関しては、通常、 約 30°C 〜約 60°C、酵母発酵 に関しては、通常、約 20°C〜約 45°Cの温度範囲で行われる。 菌類発酵 に関しては、 その温度範囲は広範であるが、 約 25°C〜約 50°Cの範囲内 であることが多い。 なお、 培養期間中、 pHは 2. 0〜7. 0に保持することが好まし い。 培地組成は上記記載の培地を用いることができる。
—方、上述した 1)〜8)の方法によって乳酸生産菌のグリセロール生産能を低下 させる場合、 生成した乳酸を含む溶液 (例えば、 乳酸生産菌を培養した培地) に 含まれるグリセロール量は、 当該溶液に含まれる乳酸量に対して 3. 5重量%以下 であることが好ましく、 0. 4重量%以下であることがより好ましく、 0. 1重量%以 下であることが最も好ましい。
また、上述した 1)〜8)の方法によって乳酸生産菌のグリセロール生産能を低下 させる場合、 生成した乳酸を含む溶液 (例えば、 乳酸生産菌を培養した培地) に 含まれる、 乳酸量に対するグリセロール量は、 グリセロール生産能を低下させて いない乳酸生産菌が生成する、 乳酸量に対するグリセロール量と比較して有意に 低下していることが必須であるが、 35%以上低下していることが好ましく、 90% 以上低下していることがより好ましく、 95%以上低下していることが最も好まし レ、0
方法 2
乳酸生産菌が生産したグリセ口ールを除去する方法とは、 上記化学反応式に示 す化学反応の進行を阻止すべく、 乳酸生産菌を用いた発酵法によって生産された グリセロールを除去する工程を含む方法である。 なお、 以下の説明において、 乳 酸生産菌の培養液から菌体を除いた溶液を粗乳酸水溶液と称する。
この工程は、 乳酸生産菌を用いた発酵法によって得られた粗乳酸水溶液に含ま れるグリセロールを除去するものであっても良いし、 乳酸生産菌の培養液に含ま れるグリセロールを除去するものであっても良い。 これらの工程は、 上記化学反 応式に示す化学反応が進行する前に行うことが望ましい。 具体的に、 当該化学反 応は、 反応に必要な熱エネルギーが、 グリセロールと乳酸とが共存する系に加え られることによって進行する。 例えば、 乳酸の製造工程において、 乳酸生産菌を 用いた発酵法によって得られた粗乳酸水溶液を加熱濃縮する場合には、 加熱濃縮 する前に粗乳酸水溶液中のグリセロールを除去することが好ましい。
このとき、 グリセロールを除去する工程によって、 乳酸量に対してグリセロー ル量が 3. 5重量%以下であることが好ましく、 0. 4重量%以下であることがより 好ましく、 0. 1 重量。ん以下であることが最も好ましい。 乳酸量に対してグリセ口 ール量が 3. 5重量%以下である場合には、 上記化学反応の進行を確実に阻害する ことができ、 その結果、 最終的に得られる乳酸の光学純度を非常に高くすること ができる。一方、乳酸量に対してグリセロール量が 3. 5重量%を超える場合には、 上記化学反応が進行する結果、 最終的に得られる乳酸の光学純度が低下する虞が 生じる。
より具体的に、 粗乳酸水溶液又は培養液に含まれるグリセロールを除去する手 法としては、 例えば、 電気透析法、 イオン交換法、 クロマトグラフ法、 抽出法 (溶 媒抽出法)、遠心分離法及び沈殿しやすい物質に変化させて分離する方法を挙げる ことができる。 但し、 粗乳酸水溶液又は培養液に含まれるグリセロールを除去す る手法としては、 これらの方法に限定されるものではない。 例えば、 粗乳酸水溶 液又は培養液に含まれるグリセロールを除去する手法としては、 グリセロールを 化学反応させて他の物質とする方法を挙げることができる。
ここで、 電気透析法とは、 粗乳酸水溶液又は培養液中に一対の電極を配設し、 直流電圧を印加することによって乳酸とグリセロールとを異なる電極近傍に分離 する方法である。 電気透析法を適用する場合には、 粗乳酸水溶液又は培養液中に 含まれる乳酸を分離しやすくするため、 アル力リによって当該乳酸を乳酸塩とし ておくことが望ましい。 イオン交換法とは、 粗乳酸水溶液又は培養液をイオン交 換樹脂にアプライし、 イオン性物質を当該イオン交換樹脂に吸着させる作用を利 用して、 グリセロールと乳酸とを分離する方法である。 クロマトグラフ法は、 展 開液とともに粗乳酸水溶液又は培養液をカラムにアプライし、 グリセロール及び 乳酸の移動速度の差を利用してこれらグリセロール及び乳酸を分離する方法であ る。 抽出法とは、 溶媒を用いて、 粗乳酸水溶液又は培養液に含まれる成分物質を 溶解させる分離する方法である。 遠心分離法とは、 粗乳酸水溶液又は培養液に遠 心力を与え、 グリセロール及び乳酸の比重差を利用してこれらグリセロール及び 乳酸を分離する方法である。 沈殿しやすい物質に変化させて分離する方法とは、 例えば、 粗乳酸水溶液又は培養液に濃硫酸又は発煙硫酸を加えることによってグ リセロールをスルホン化し、 スルホン化したグリセロールを沈殿させた後、 粗乳 酸水溶液又は培養液をろ過することによって乳酸とグリセロールとを分離する方 法を挙げることができる。 或いは、 沈殿しやすい物質に変化させて分離する方法 としては、 粗乳酸水溶液又は培養液に水酸化カルシウム又は炭酸カルシウムを加 えて乳酸を中和した後、 冷却することにより乳酸を乳酸カルシウムとして沈殿さ せ、 粗乳酸水溶液又は培養液をろ過することによって乳酸とグリセロールとを分 離する方法を挙げることができる。
一方、 グリセロールを化学反応させて他の物質とする方法としては、 例えば、 酸性条件下でグリセロール分子内での脱水反応を進行させる方法ゃグリセロール とカルボニル化合物 (アルデヒ ド化合物、 ケトン化合物) を反応させてァセター ルを生成する方法を挙げることができる。
以上で説明した方法 1及び 2によれば粗乳酸水溶液や培養液等の溶液に含まれ るグリセ口ール量を低減させることができる。 本発明に係る乳酸の製造方法にお いて、 溶液中の乳酸を加熱濃縮する工程では、 例えば、 上記方法 1によって得ら れた培養液から菌体を除去した溶液或いは上記方法 2によってグリセロールを除 去した溶液を、 当該溶液に含まれる乳酸を特に限定されないが約 6 0〜 7 0質 量%の濃度となるまで減圧下で加熱濃縮する。 本方法においては、 溶液中のダリ セロール量が低減しているため、 上記化学反応式に示す化学反応が生じず、 加熱 濃縮を行っても高光学純度の乳酸を製造することができる。
特に、 本方法において、 い乳酸生産能を有する乳酸生産菌を用いた発酵法によ つて乳酸を製造する場合、 最終的な乳酸の光学純度を 9 9 %以上にすることがで きる。 既存の方法によって高光学純度の乳酸を製造したとしても、 光学純度が 9 9 %以上の乳酸を製造することは不可能であり、 本発明において目的とする高光 学純度を達成することはできなかった。 このように、 9 9 %以上の光学純度の乳 酸は、 優れた生分解性をもつポリ乳酸の原料として、 また優れた物性を示すポリ 乳酸の原料として好適である。
また、 以上で説明した方法 1及び 2によれば粗乳酸水溶液や培養液等の溶液に 含まれるグリセロール量を低減させることによって、 高光学純度の乳酸を製造で きるだけでなく、 乳酸の生産性を向上させることができる。 例えば、 乳酸脱水素 酵素遺伝子を導入した酵母 (乳酸生産菌の一例) においては、 酵母本来のェタノ ール発酵が存在するため必ずしも乳酸収率は高くなく、 乳酸収率を向上させる目 的でアルコール発酵を抑制する試みがなされてきた。 しかしながら、 アルコール 発酵を抑制した乳酸生産酵母においては、 乳酸収率に加え、 発酵速度や培養速度 等の面で十分に満足できる菌株を取得できていなかった。
これに対して、 以上で説明した方法 1及び 2によれば、 粗乳酸水溶液や培養液 等の溶液に含まれるグリセロール量を低減させることでエタノール生産を低減さ せることができ、 その結果、 乳酸収率を向上させることができる。 従って、 本発 明に係る乳酸の製造方法によれば、 高生産性、 高収率で光学純度に優れた乳酸を 製造することができる。
なお、 本発明に係る乳酸の製造方法では、 乳酸生産菌を用いた発酵法によって 乳酸を製造する既知の方法と同様の処理工程を含んでいてもよい。 例えば、 発酵 法では、 培養液及び粗乳酸水溶液に含まれる乳酸成分をアンモニアで中和して乳 酸アンモニゥムとしているが、 本発明に係る乳酸の製造方法においても、 培養液 及び粗乳酸水溶液に含まれる乳酸成分をアンモニアで中和して乳酸アンモ-ゥム としてもよい。 培養液及び粗乳酸水溶液に乳酸アンモニゥムを含む場合には、 上 述した加熱濃縮の後、 ブタノール等のアルコールによってエステル化し、 乳酸ブ チル等の乳酸エステルの形で蒸留して乳酸成分を分離する。 その後、 分離した乳 酸エステルを加水分解し、 更に、 濃縮することによって乳酸を製造する。 なお、 乳酸成分をアンモニアで中和せずに、 乳酸のかたちで培養液及び粗乳酸水溶液に 含まれる場合には、 当該培養液及び粗乳酸水溶液から直接蒸留することで乳酸を 製造することもできる。 以下、 実施例を用いて本発明をより詳細に説明するが、 本発明の技術的範囲は 以下の実施例に限定されるものではなレ、。
(実験例)
本発明を適用した実施例を説明するに先立って、 上記化学反応式が実際に起こ ることを検証する。 本実験例では、 グリセロールと乳酸の間の化学反応、 及び、 エチレンダリコールと乳酸との間の化学反応が実際に起こり うることを検証した。 先ず、 L-乳酸とグリセ口ール若しくはエチレングリコールとを 1対 2の割合(モ ル比) で混合した溶液を調製した。 次に、 当該溶液に P-トルエンスルホン酸を添 加して常圧で水をとばしながら加熱 ( 1 5 0 °Cで 1 5時間) した。
反応終了後、 溶液をクロ口ホルムに溶解し (1〜 1 0質量%) GC— MSで分析を 行った。 GC— MS分析では、 日本電子社製の四極子型質量分析装置(JMS-AM SUN200) 及び J & W Scientif ic社製のカラム DB- 1を使用し、 ィンジェクション温度を 3 0 0 °C、 カラム温度を 5 0〜 3 0 0 °Cとし、 昇温速度を 5 °C/minとし、 ヘリウム 流量を 1 ml/min とした。
上記化学反応式に示したグリセロールと乳酸の間の化学反応が進行している場 合、 上記化学反応式における環状化合物を検出することができる。 また、 ェチレ ングリコールと乳酸の間には、 下記の化学反応式に示す化学反応が進行している と考えられ、 下記化学反応式における環状化合物を検出することができる。 下記 化学反応式における環状化合物の生成に至る反応が進行する程、 L -乳酸の光学純 度がより低下することとなる。

グリセロールと乳酸の間の化学反応による環状化合物は、 分子ィオンピークと して 1 4 6及び 1 1 5を同時に検出できるものとして観測されることとなる。 こ れは、上記環状化合物と構造が類似するグリセロール 2量体の MSスぺク トルにお いて、 側鎖のヒ ドロキシメチル基が取れた分子イオンピークが観測されることに 基づいている。 また、 参考として検証したエチレングリコールと乳酸との間の化 学反応による環状化合物は、 分子イオンピークとして 1 1 6及び 7 3が同時に検 出できるものとして観測されることとなる (Macromolecules, 2001 , 34, 8641)。
L -乳酸とグリセロールを含む溶液を用いた実験の結果、 分子イオンピークとし て 1 4 6と 1 1 5に同時に存在する化合物が、 保持時間 1 4 . 5分に観測するこ とができた (図 1及び 2参照)。 また、 L-乳酸とエチレングリコールを含む溶液を 用いた実験の結果、 分子イオンピークとして 7 3と 1 1 6に同時に存在する化合 物が、 保持時間 7分に観測することができ (図 3及び 4参照)、 ピーク強度比も文 献値 (1 1 6の分子イオンピークと 7 3の分子イオンピークとの強度比が 2 3 : 1 0 0 (Macromolecules, 2001, 34, 8641) ) とほぼ同じであることを確認した。 これらの結果より、 乳酸とグリセロール又はエチレンダリコールとが共存した 系に対して熱エネルギーを与えることによって、 上記化学反応式に示す化学反応 が進行することが確認できた。本実験例より、溶液中の乳酸を加熱濃縮する前に、 当該溶液中のグリセロール量を減少させることで高光学純度の乳酸を製造できる ことが示唆された。
〔実施例 1〕
上述した実験例では、 溶液中の乳酸を加熱濃縮するに前に、 当該溶液中のダリ セ口一ル量を減少させることで高光学純度の乳酸を製造できることが示唆された。 そこで、 本実施例では、 グリセローノ 生産に関連する遺伝子を破壊した乳酸生産 菌を用いた発酵法により、 高光学純度の乳酸を製造できることを実証した。
遺伝子破壊株の作出
GPD1破壊株の作製
特開 2003-259878号公報 (特願 2002- 65879) で作製した乳酸生産能を持つ酵母 を胞子形成培地 (1% リン酸カリウム、 0. 1% イーストエキストラタ ト、 0. 05% ブ ドウ糖、 2% 寒天) で胞子を形成させ、 ホモタリック性を利用して 2倍化を行い、 2 倍体である染色体両方に LDH 遺伝子が導入されている株を取得し、 これを KCB-27- 7株とした。
大腸菌 K12株をテンプレートとして、 ハイグロマイシン耐性遺伝子 (以下、 HPH 遺伝子) の DNA断片を PCRで増幅した。 HPH遺伝子の DNA塩基配列は GENBANKデ —タベースに V01499で登録されており、 HPH遺伝子の両末端のプライマー HPH - U
(5, -ATG AAA AAG CCT GAA CTC ACC- 3 ' (配列番号 1 ) ) と HPH - D (5 ' -CTA TTC CTT TGC CCT CGG ACG-3 ' (配列番号 2 ) ) を使用した。
酵母 IF02260株 (社団法人発酵研究所に登録されている菌株) のゲノム DNAを テンプレートとして、 TDH3プロモーター領域の DNA断片を PCRで増幅した。 TDH3 遺伝子の DNA塩基配列は GENBANKデータベースに Z72977で登録されており、プラ イマ一 TDH3P - U (5 ' -ATA TAT GGA TCC TAG CGT TGA ATG TTA GCG TCA AC - 3 ' ; TDH3 プロモーター配列に BamHIサイ トを付加 (配列番号 3 ) )、 TDH-3P- D (5 ' -ATA TAT CCC GGG TTT GTT TGT TTA TGT GTG TTT ATT CG-3 ' ; TDH3 プロモーター配列に Smalサイ トを付加 (配列番号 4 ) ) を使用した。

Bacteriol. , 153, 163-168 (1983))にて形質転換した。 形質転換後、 200μ g/ml ハイグロマイシンを含む YPD培地のプレートにまいて、 30°Cで 2日間培養し、 形 質転換体を得た。 形質転換体よりゲノム DNAを調製し、 PCR法により挿入 DNA断 片の外側のプライマーである GPD1- 295F (5' - TGC TTC TCT CCC CTT CTT- 3' (配 列番号 8))、 GPD1+1472R (5' - CAG CCT CTG AAT GAG TGG T-3' (配列番号 9)) を 用いて、 HPH遺伝子が GPD1遺伝子領域の染色体に組込まれていることを確認した。 この株を胞子形成培地で胞子を形成させ、 ホモタリック性を利用して 2倍化を 行った。 2倍体である染色体の GPD 1遺伝子領域の両方に HPH遺伝子が組み込まれ GPD1遺伝子が破壊されている株を取得した。 これを TC20株とした。
GPD2破壊株の作製
pCAT 3- Basic Vector (プロメガ製)をテンプレートとして、 クロラムフエニコ ール耐性遺伝子 (以下、 CAT遺伝子) の DNA断片を PCRで増幅した。 CAT遺伝子の DNA塩基配列は GENBANKデータベースに M16323で登録されており、 CAT遺伝子の 両末端のプライマー CAT-U (5, -ATA TAT CCC GGG ATG GAG AAA AAA ATC ACT GGA TAT AC- 3' (配列番号 1 0))、 CAT- D (5, -ATA TAT AAG CTT TTA CGC CCC GCC CTG CCA CTC ATC- 3' (配列番号 1 1 )) を使用した。
CAT 遺伝子断片を大腸菌プラスミ ド pBluescriptll (プロメガ社製) の EcoRV 部位に挿入した。 このプラスミ ドを pBCATと命名した。 本プラスミ ドを BamHI と Smal部位で切断後、 TDH3プロモーター断片を挿入し、 このプラスミ ドを pBCAT- P と命名した。 さらに本プラスミ ドを Hindlll と Sail部位で切断後、 CYC1 ターミ ネーター断片を挿入し、 このプラスミ ドを pBCAT- PTと命名した。

この株を胞子形成培地で胞子を形成させ、 ホモタリック性を利用して 2倍化を 行った。 2倍体である染色体の GPD2遺伝子領域の両方に CAT遺伝子が組み込まれ GPD2 遺伝子が破壊されている株を取得した。 このうち、 KCB27- 7 株由来の GPD2 破壊株を TC21株、 TC20株由来の同遺伝子破壊株を TC38株と命名した。
発酵試験 1
上記で得られた形質転換体について、 発酵培地 (スクロース 14. 4%、 糖蜜 0. 6%) 500mlに菌体濃度が 0. 3%になるように接種し、 34°C、 pH5. 0 (アンモニアで中和)、 通気量 0. 6Vvm、 3 日間発酵を行い、 L-乳酸とグリセロールの生産量を検討した。 L-乳酸、 エタノール、 グリセロール濃度は王子計測機器株式会社製バイオセンサ 一 BF- 4により測定した。 L-乳酸の対糖収率は L-乳酸生産量を醱酵前の糖含量で割 ることにより計算した。 結果を表 1に示す。

KCB27- 7株 (LDH導入株) 8.6 0.64 表 1に示すように、発酵終了時の培養液における L-乳酸濃度及びグリセロール 濃度は、 それぞれ 9 . 1重量% (乳酸アンモ-ゥムで 1 0 . 8重量%) 及び〇. 0 0 8 2重量0 /。であった。 このグリセロール濃度は、 L-乳酸に対して 0 . 1 %以 下であった。 また、 D-乳酸の濃度をロッシュ社製 F-キッ トを用いて測定し、 L -乳 酸の光学純度を下記式に従って算出した。 なお、 下記式において、 D-乳酸の濃度 を 「D」 と表記し、 L-乳酸の濃度を 「L」 と表記する。

発酵試験 2
上記で得られた形質転換体について、 発酵培地 (グルコース 4%、 ィーストェキ ス トラク ト 1%) 50ml を入れた 100ml三角フラスコに菌体濃度が 0. 3%になるよう に接種し、 32°Cで震盪 (回転数 80rpm;震盪幅 70mm)、 2〜3 日間発酵を行い、 L- 乳酸とエタノールとグリセロールの生産量を検討した。 その結果を表 2に示す。

その結果、 GPD1破壊株及ぴ GPD2破壊株は、 KCB27- 7株と比較して L-乳酸生産 量が増加、 エタノール及びグリセロール生産量が低減し、 L-乳酸の対糖収率が向 上したことを見出した。 GPD 1、 GPD2の 2重破壊株は、 単独破壊株よりさらにェ タノール及びグリセロール生産量が低下し、 対糖収率が向上した。
詳細には、 KCB27- 7株による乳酸量に対するグリセロール量 (5. 3重量%) と比 較すると、 TC20 株においては乳酸量に対するグリセロール量 (0. 37 重量%) が 93. 0%低下しており、 TC21株においては乳酸量に対するグリセロール量 (3. 2重 量%) が 39. 6%低下しており、 TC38株においては乳酸量に対するグリセロール量 (0. 065重量%) が 98. 8%低下していた。 '
以上のことから、 GPD1破壊株及び/又は GPD2破壊株におけるグリセ口ール生産 量低減は、 エタノール生産性の低減と乳酸生産性の向上にも寄与することも確認 できた。 そして、 L-乳酸の対糖収率も 5%以上、 好ましくは 10%以上向上すること が示された。
L-乳酸の精製
先ず、 上述した発酵法によって得られた培養液からフィルター (旭化成ケミカ ルズ社製、 商品名 microza) を用いて菌体を分離して粗乳酸水溶液を調製した。 次に、 得られた粗乳酸水溶液を、 大気圧、 1 2 4 °C (熱源温度 1 6 0 °C) になる まで加熱濃縮した。 この加熱濃縮によって、 粗乳酸水溶液に含まれる L-乳酸濃度 を約 7 0 %とした。
次に、 加熱濃縮された粗乳酸水溶液に対して乳酸比でブタノールを 3倍モル添 加した後、 大気圧、 1 1 0〜 1 2 0 °C (熱源温度 1 6 0 °C) にて 1 2時間反応さ せた。これにより、粗乳酸水溶液に含まれる乳酸アンモユウムをエステル化した。 次に、 乳酸プチルを含む反応液を、 圧力 2 O torr、 1 2 0 °C (熱源温度 1 6 0 °C) の条件下で蒸留することによって、 乳酸ブチルを分離精製した。
次に、 分離精製した乳酸ブチルに対して、 乳酸ブチル比で水を 1 6倍モル添加 した後、 大気圧、 1 0 0 °C (熱源温度 1 6 0 °C) にて 8時間反応させた。 これに より、 乳酸ブチルを加水分解した。 最後に、 反応液を大気圧、 1 2 8 °C (熱源温 度 1 6 0 °C) になるまで加熱濃縮した。 この加熱濃縮によって、 精製乳酸水溶液 に含まれる 乳酸濃度を約 9 0 %とした。
以上の工程を経て得られた L-乳酸を最終製品とした。得られた最終製品におけ る L -乳酸の光学純度は、 9 9 . 5 1 %であった。 また、 以上の工程における乳酸 の回収率は 7 6 . 0 %であった。
〔実施例 2〕
本実施例では、 乳酸生産菌が生産したグリセロールを除去した後、 乳酸を加熱 濃縮することで高光学純度の乳酸を製造できることを実証した。
乳酸生産菌及び発酵法
本実施例において乳酸生産菌としては、 G P D 1遺伝子及び G P D 2遺伝子を 破壊していない以外は実施例 1で使用した乳酸産生能力を付与されたサッカロマ イセス ·セルピシェを使用した。 また、 本実施例において発酵法は、 実施例 1 と 同様の条件で行った。
発酵終了時の培養液における L-乳酸濃度及びグリセロール濃度は、 それぞれ 8 . 6重量% (乳酸アンモニゥムで 1 0 . 2重量。 /0) 及び 0 . 7重量%であった。 ま た、 L-乳酸の光学純度は 9 9 . 7 1 %であった。
グリセ口ール除去
本例では、 先ず、 上述した発酵法によって得られた培養液からフィルター (旭 化成ケミカルズ社製、 商品名 microza) を用いて菌体を分離して粗乳酸水溶液を 調製した。 次に、 得られた粗乳酸水溶液を電気透析にかけ、 溶液中のグリセロー ルを分離して除去した。具体的には、旭化成ケミカルズ社製、電気透析装置 MICRO ACILYZER S3、 カートリ ッジ AC-110-550を用い、 希釈側に粗乳酸水溶液、 濃縮側 に蒸留水を仕込み、 印可電圧 15Vにて電気透析を実施した。 電気透析は、 希釈側 の電導度が 0. 5mSになるまで実施した。これにより、濃縮側に乳酸を移動させた。 さらに、 電導度の低下した希釈側の粗乳酸水溶液を廃棄し、 再度、 希釈側に粗乳 酸水溶液を仕込み、 繰り返し透析を実施した。
電気透析後の粗乳酸水溶液に含まれる L-乳酸濃度及びグリセロール濃度を測 定したところ、 それぞれ 2 1 . 6重量%及び 0 . 0 2重量%であった。 このグリ セロール濃度は、 L-乳酸に対して 0 . 1 %以下であった。
L-乳酸の精製
上述したようにグリセロールを除去した粗乳酸水溶液を、 Buchi社製 Rotavapor
R-220 を用いて、 大気圧、 1 2 4 °C (熱源温度 1 6 0 °C) になるまで加熱濃縮し た。 この加熱濃縮によって、 粗乳酸水溶液に含まれる L-乳酸濃度を約 6 5 %とし た。
次に、 加熱濃縮された粗乳酸水溶液に対して乳酸比でブタノールを 3倍モル添 加した後、 大気圧、 1 1 0〜 1 20°C (熱源温度 1 6 0°C) にて 1 2時間反応さ せた。これにより、粗乳酸水溶液に含まれる乳酸アンモニゥムをエステル化した。 次に、 乳酸ブチルを含む反応液を、 圧力 2 Otorr、 1 20 °C (熱源温度 1 6 0°C) の条件下で蒸留することによって、 乳酸ブチルを分離精製した。
次に、 分離精製した乳酸ブチルに対して、 乳酸ブチル比で水を 1 6倍モル添加 した後、 大気圧、 1 00°C (熱源温度 1 60°C) にて 8時間反応させた。 これに より、 乳酸ブチルを加水分解した。 最後に、 反応液を大気圧、 1 28°C (熱源温 度 1 60°C) になるまで加熱濃縮した。 この加熱濃縮によって、 精製乳酸水溶液 に含まれる 乳酸濃度を約 90%とした。
以上の工程を経て得られた L-乳酸を最終製品とした。得られた最終製品におけ る L-乳酸の光学純度は、 99. 1 6%であった。 また、 以上の工程における乳酸 の回収率は 64. 8%であった。
〔比較例〕
比較のために、 実施例 2で使用した乳酸生産菌を用いて、 実施例 2と同様に発 酵法を実施した。 本比較例では、 粗乳酸水溶液に含まれるグリセロールを除去せ ずに、 その後の L-乳酸の精製を実施した。 その結果、 発酵終了後の培養液に含ま れる L-乳酸の光学純度は 99. 7 1 %であった。 培養液に含まれるグリセロール 濃度は、 乳酸に対して 1 %であった。 L-乳酸の精製後、 乳酸の光学純度は 9 8. 40%であった。 また、 L-乳酸の回収率は 70. 4%であった。
〔結果〕
以上、 実施例 1及び実施例 2の結果から分かるように、 L-乳酸を加熱濃縮する 前に、 グリセロール量を低減させることによって、 高光学純度の L-乳酸を製造で きることが実証された。 より具体的には、 グリセロールが 1 %存在する系におい て L-乳酸を加熱濃縮する場合 (比較例 1 ) には L-乳酸の光学純度が 98. 40 % であったのに対して、 グリセロールが 0. 1 %以下の系において L-乳酸を加熱濃 縮する場合(実施例 1及び 2 )には L-乳酸の光学純度が 99 %以上となっていた。 このように、 実施例 1及び 2によって、 9 9%以上といった高光学純度で L-乳酸 を製造できる方法を確立することができた 本明細書で引用した全ての刊行物、 特許および特許出願をそのまま参考として 本明細書にとり入れるものとする。 産業上の利用可能性
本発明により、 優れた物性を持つポリ乳酸の原料としても利用可能な高光学純 度で乳酸を製造できる乳酸の製造方法を提供することができる。 配列表フリーテキスト

Claims

請求の範囲
1 . グリセロール量を減少させた溶液で、 当該溶液中の乳酸を加熱濃縮する 工程を含む乳酸の製造方法。
2 . グリセロール生産能を低下させた微生物を使用した乳酸発酵によって上 記溶液を調製する工程を更に含むことを特徴とする請求項 1記載の乳酸の製造方 法。
3 . 上記微生物は、 グリセロール生産に関連する遺伝子のうち少なく とも 1 つの遺伝子の発現を抑制した変異体であることを特徴とする請求項 2記載の乳酸 の製造方法。
4 . 上記変異体は、 グリセロール- 3-リン酸脱水素酵素をコードする遺伝子を 破壊した乳酸生産菌であることを特徴とする請求項 3記載の乳酸の製造方法。
5 . 上記乳酸生産菌は、 サッカロマイセス属に属する微生物であることを特 徴とする請求項 4記載の乳酸の製造方法。
6 . 上記溶液は、 乳酸量に対するグリセロール量が 3. 5重量%以下であるこ とを特徴とする請求項 1記載の乳酸の製造方法。
7 . 上記溶液は、 乳酸量に対するグリセロール量が 0. 1重量%以下であるこ とを特徴とする請求項 6記載の乳酸の製造方法。
8 . 微生物を使用した乳酸発酵によって上記溶液を調製する工程と、 上記溶 液からグリセ口ールを除去する工程とを更に含むことを特徴とする請求項 1記載 の乳酸の製造方法。
9 . 上記グリセロールを除去する工程では、 上記溶液中の乳酸量に対するグ リセロール量を 3. 5重量%以下とすることを特徴とする請求項 8記載の乳酸の製 造方法。
1 0 . 上記グリセロールを除去する工程では、 上記溶液中の乳酸量に対する グリセロール量を 0. 1重量%以下とすることを特徴とする請求項 9記載の乳酸の 製造方法。
1 1 . 乳酸生産菌に対してグリセロール生産量が低減するような変異を導入 してなる変異体。
1 2 . 上記乳酸生産菌がサッカロマイセス属に属する微生物であることを特 徴とする請求項 1 1記載の変異体。
1 3 . グリセロール- 3-リン酸脱水素酵素をコードする遺伝子を破壊すること でグリセロール生産量を低減させたことを特徴とする請求項 1 1記載の変異体。
1 4 . 乳酸生産菌に対して、乳酸量に対するグリセロール生産量を 3. 5重量。 /0 以下に低減するような変異を導入してなる請求項 1 1記載の変異体。
1 5 . 乳酸生産菌に対して、乳酸量に対するグリセロール生産量を 0. 1重量% 以下に低減するような変異を導入してなる請求項 1 4記載の変異体。
1 6 . グリセロール生産能を低下させた微生物を使用した乳酸発酵によって、 乳酸を製造する工程を含む乳酸の製造方法。
1 7 . 上記微生物は、 グリセロール生産に関連する遺伝子のうち少なく とも 1つの遺伝子の発現を抑制した変異体であることを特徴とする請求項 1 6記載の 乳酸の製造方法。
1 8 . 上記変異体は、 グリセロール- 3-リン酸脱水素酵素を破壊した乳酸生産 菌であることを特徴とする請求項 1 7記載の乳酸の製造方法。
1 9 . 上記乳酸生産菌は、 サッカロマイセス属に属する微生物であることを 特徴とする請求項 1 8記載の乳酸の製造方法。
2 0 . 上記乳酸生産菌は、 乳酸量に対するグリセロール生産量を 3. 5重量% 以上低減するような変異が導入されたものであることを特徴とする請求項 1 8記 載の乳酸の製造方法。
2 1 . 上記乳酸生産菌は、 乳酸量に対するグリセロール生産量を 0. 1重量% 以上低減するような変異が導入されたものであることを特徴とする請求項 2 0記 載の乳酸の製造方法。
