Definitions

  • the present invention relates to a method for efficiently introducing a gene into a plant material via a bacterium belonging to the genus Agrobacterium.
  • Gene transfer by the agrobacterium method is a method for transforming a plant utilizing the function of agrobacterium.
  • T DNA which is part of the Ti (tumor inducing) plasmid involved in the pathogenesis of Agrobacterium tumemi, is integrated into the plant genome. It has a function to be performed.
  • the method for transforming plants by the agrobacterium method is to prepare a transformation plasmid in which the TDNA region of the Ti plasmid has been replaced with a gene desired to be introduced into the plant genome, and to transform the transformation plasmid into the Ti plasmid. This is a method in which a gene desired to be introduced into the plant genome is introduced into the plant genome by utilizing the function of the above-mentioned agrobacterium using agrobacterium prepared as a substitute.
  • Agrobacterium bacteria are said to use only dicotyledonous plants as hosts and do not infest monocotyledonous plants
  • the plant transformation method using the Agrobacterium territory method was primarily used for dicotyledonous plants. It was developed as a method for transforming plants. After that, various attempts have been made to introduce genes into monocotyledonous plants by the agrobacterium method, and a super-noary vector having a part of the virulence gene of the strongly pathogenic agrobacterium has been developed. It has been reported that the method using E. coli can stably and relatively efficiently transform monocotyledonous plants such as rice and corn (for example, Patent No. 2,649,287; Patent No. 3).
  • Transformation methods using agrobacterium are generally highly efficient, have a low copy number of the gene to be introduced, and can be introduced without fragmenting a specific region of T DNA. It has many excellent features, such as the fact that transformants can be obtained by short-term cultivation, so that there are few variations in culture. For this reason, it is now widely used as the most useful means of transformation for many plant species, both dicotyledonous and monocotyledonous.
  • the composition of a test material and a culture medium are different depending on the plant species.
  • the agrobacterium suspension is brought into contact with a tissue to be a material in any plant.
  • the common point is that transformed cells are selected after co-culture to produce transformed plants.
  • the plant tissue that is the source of the material is infected with Agrobacterium territory without the need to sterilize or otherwise treat it as needed (eg, Rogers, S. G "et al.
  • Transformation with Agrobacterium is the power reported in many plant species. The transformation efficiency varies greatly depending on the species, genotype, and tissue of the plant (eg, Potrykus, I., et. al., (1998), Agricultural Biotechnology, NY: See Mercel Dekker Inc., p.119-159).
  • tissue of the plant eg, Potrykus, I., et. al., (1998), Agricultural Biotechnology, NY: See Mercel Dekker Inc., p.119-159.
  • Transformation methods that do not depend on plant species or genotype are extremely useful for efficiently cultivating commercial varieties.
  • the development of a transformation method that does not depend on the plant tissue used as the material is also necessary for efficient transformation.
  • Various attempts to improve the transformation efficiency of corn by the bacterium method include selection of transformed cells on N6 basal medium (for example, Zhao, ZY., Et al., (2001), Mol. Breed., Vol. 8, p. 323-333), addition of silver nitrate and carbecillin to the medium (eg, Zhao, ZY., Et al "(2001), Mol. Description

一般に、材料となる植物組織は、必要に応じ滅菌処理がなされ る力 それ以外に特別な処理をすることなくァグロバタテリゥムの感染が行われる(例 えば、 Rogers, S. G" et al., (1988), Method for Plant Molecular Biology, p.423- 436, CA: Academic Press Inc.; Visser, R. G. F., (1991), Plant Tissue Culture Manual, B5: l- 9, Kluwer Academic Publishers; McCormick, S., (1991), Plant Tissue Culture Manual, B6:ト 9, Kluwer Academic Publishers;および、 Lindsey, K., et al" (1991), Plant Tissue Culture Manual, B7: l- 13, Kluwer Academic Publishers ^を参照)。
Breed., Vol.8, p.323-333; Frame, B. R., et al., (2002), Plant Physiol, Vol.129, p.13-22;お よび、 Ishida, Y" et al., (2003), Plant Biotechnology, Vol.14, p.57- 66、を参照)。 Ghaemiら( uhaemi, M., et al., (1994), Plant Cell, Tissue and urgan culture, Vol.36,

[0041] もう一つは、スーパーバイナリーベクター('super— binary' vector) (Hiei, Y., et al., (1994), The Plant Journal, Vol.6, p.271-282; Ishida, Y., et al., (1996), Nature Biotechnology, Vol.4, p.745-750; Komari, T. and Kubo T., (1999), Methods of Genetic Transformation: Agrobacterium tumefaciens. In Vasil, I. K. (ed.)
例えば、実施例で使用した LS— AS培地、 n N6— As培地、あるいはその他、 N6S3— AS培地、 2N6— AS培地(Hiei, Y., et al, (1994), The Plant Journal, Vol.6, p.271-282を参照)等の培地が知られている。
Amoah, B" et al" (2001), Journal of
Experimental Botany, Vol.52, p.1135- 1142)
