Description
プロスタグランジン含有経鼻投与製剤組成物 技術分野
本発明は、プロスタグランジン誘導体を有効成分として含有する経鼻投与製剤組成 物に関する。 背景技術
プロスタグランジンは微量で種々の薬理学的 ·生理学的作用を発揮し、 医薬への応 用を企図して多くの誘導体が合成され、 その生物活性が検討されてきた。そして、 あ る種のプロスタグランジン (以下、 P Gと略記する。 ) は睡眠誘発作用を有し、 これ を経鼻投与した場合に優れた作用を奏することから、当該 P Gを有効成分として含有 する経鼻投与製剤が開発された (PCT/JP00/08239) 。
前記 P G含有経鼻投与製剤は睡眠誘発剤として十分な効果を発揮したが、その後の 製造適性の検討に至り、製造中に水溶性高分子の粒子同士が付着 ·凝集するという問 題を生じる場合のあることがわかった。 この粒子同士の付着 '凝集という現象は、 製 造機器が大型化するにつれ、すなわち、製造スケールが大きくなるにつれて発生の頻 度が多くなつた。
本発明の目的は、製造時に付着 ·凝集を生じないプロスタグランジン含有経鼻投与 製剤を提供することである。 発明の開示
本発明者は前記目的を解決すべく鋭意検討を重ねた結果、 P G及び水溶性糖類を、 これらを溶解しうる溶媒中に溶解させ、次に、該溶液を造粒機中に流動又は攪拌流動 させたヒドロキシプロピルセルロース等の水溶性高分子の粉体に噴霧して P Gを水 溶性高分子の粒子に付着 ·担持させ、 これを必要に応じ乾燥するという方法により、 製造中に粉体同士の付着 ·凝集を生ずることなく、 P G含有経鼻投与製剤を製造しう ることを見出した。
また、 P Gを塩基性の溶媒に溶解させ、該溶液を造粒機中に流動又は攪拌流動させ たヒドロキシプロピルセルロース等の水溶性高分子の粉体に噴霧して P Gを水溶性 高分子の粒子に付着 ·担持させ、 これを必要に応じ乾燥するという方法により、 製造 中に粉体同士の付着 ·凝集を生ずることなく、 P G含有経鼻投与製剤を製造しうるこ とを見 した。
すなわち、 かかる知見をもとに完成した本発明の一つの態様は、 式 (I )

。のシクロアルキル 基を示し、 mは 0〜3の整数、 nは 1〜4の整数を示す。) で表されるプロスタグラ ンジン誘導体及び水溶性糖類を溶媒に溶解させ、該溶液を流動又は攪拌流動中の水溶 性高分子に噴霧し、式(I )で表されるプロスタグランジン誘導体を水溶性高分子の 表面に担持させたことを特徴とする経鼻投与製剤組成物である。
本発明の他の態様は、 式(A)で表されるプロスタグランジン誘導体及び水溶性糖 類を溶媒に溶解させ、該溶液を流動又は攪拌流動中の水溶性高分子に噴霧し、式(A) で表されるプロスタグランジン誘導体を水溶性高分子の表面に担持させたことを特 徵とする経鼻投与製剤組成物である。

本発明の他の態様は、 前記式(I ) 又は (A) で表されるプロスタグランジン誘導 体を塩基性溶媒に溶解させ、 該溶液を流動又は攪拌流動中の水溶性高分子に噴霧し、 式(I ) 又は(A) で表されるプロスタグランジン誘導体を水溶性高分子の表面に担 持させたことを特徴とする経鼻投与製剤組成物である。
本発明の他の態様は、 前記式(I ) 又は (A) で表されるプロスタグランジン誘導 体の 1質量部に対して、水溶性糖類が 1〜 2 0質量部、水溶性高分子が 9 0〜 9 9 8 質量部である前記経鼻投与製剤組成物である。

次に、 本発明の式 (I ) 化合物における置換基の意義を説明する。
Xで表される 「ハロゲン原子」 とは、 フッ素原子、 塩素原子、 臭素原子又はヨウ素

「薬学的に許容される塩」 とは、 例えば、 ナトリウム、 カリウムなどのアルカリ金 属との塩、 カルシウム、 マグネシウムなどのアルキル土類金属との塩、 アンモニア、 メチルァミン、 ジメチルァミン、 シクロペンチルァミン、 ベンジルァミン、 ピベリジ ン、モノェ夕ノ—ルァミン、ジェ夕ノ—ルァミン、モノメチルモノエタノ—ルァミン、 トロメタミン、 リジン、 テトラアルキルアンモニゥム、 トリス (ヒドロキシメチル) ァミノメタンなどとの塩である。
本発明において用いられる式 (A) 化合物 (以下、 適宜に 「薬物 A」 と呼ぶ。 ) の 製造方法は、 特開平 7— 2 3 3 1 4 4号公報に記載されている。式(A)化合物には 「薬学的に許容される塩」 が包含される。
本発明における 「水溶性糖類」 としては、 例えば、 マンニトール、 キシリトール、 エリスリトール、 乳糖が挙げられ、 少なくとも 1種が用いられる。
水溶性糖類の配合量は、 式(I ) 又は (A) で表されるプロスタグランジン誘導体 (以下、 適宜に 「薬物」 と呼ぶ。 ) 1質量部に対して 1〜2 0質量部であり、 1〜5 質量部であることが好ましく、 さらに 1 . 5〜2 . 5質量部であることが好ましい。 本発明において用いられる 「溶媒」 は、 薬物及び水溶性糖類を溶解させることがで きる溶媒であれば特に限定はないが、 例えば、 エタノールと水の混液が挙げられる。 本発明において用いられる「塩基性溶媒」は、 p H 7以上の塩基性溶媒で前記式( I ) 又は(A)で表されるプロスタグランジン誘導体を溶解することができるものをいう。 例えば、水一エタノール溶液にリン酸ニ水素ナトリゥム水和物及びリン酸水素ナトリ ゥムを溶解させて p Hを 8に調整した液が該当する。
本発明において 「水溶性高分子」 とは、 水に溶ける高分子を意味し、 例えば、 架橋 ビニル重合体及び水溶性セルロースェ一テルが挙げられ、少なくとも 1種が用いられ る。
「水溶性高分子」 の配合量は、 薬物 1質量部に対して 9 0〜 9 9 8質量部であり、 9 2〜9 8質量部であることが好ましく、さらに 9 5〜9 7質量部であることが好ま しい。
「架橋ビニル重合体」 は、 架橋ポリビニルアルコール (例えば、 ポリビニルアルコ ール) 、 架橋ポリアクリル酸及びその塩 (例えば、 ポリアクリルアミド) 、 ポリビニ
ルピロリドンを包含する。
「水溶性セルロースエーテル」とは、具体的には、ヒドロキシプロピルセルロース、 ヒドロキシアルキルメチルセルロース (例えば、 ヒドロキシプロピルメチルセルロー ス) 、 メチルセルロース、 ヒドロキシェチルセルロース、 ェチルヒドロキシェチルセ ルロ一ス、力ルポキシメチルヒドロキシェチルセルロース、カルボキシメチルセル口 ースナトリウム、 カチオン性セルロースエーテルを包含する。
製剤上の簡便さ及び経鼻投与製剤の薬効の点で、好ましくは、 ヒドロキシプロピル セルロース、 ヒドロキシプロピルメチルセルロース、ポリビニルピロリドン及びメチ ルセルロースの少なくとも 1種が用いられる。特に好ましいのは、 ヒドロキシプロピ ルセルロースである。
ここで用いる 「水溶性高分子」 は、 経鼻投与製剤組成物の製造原料として用いると いう点から、 その平均粒子径は、 15〜700 m、 かつ比重は 0. 4〜0. 6 gZ mLであることが好ましく、 さらに 75〜250 m、 かつ比重は 0. 5〜0. 6 g /mLであることが好ましい。 この条件を充たすものであれば、市販品でも合成品で もよく、 医薬製剤に使用可能な純度を満たす限り、 任意に選択できる。
ここに、平均粒子径は、マイクロトラック粒度分布計で測定した値であり、比重は、 A.B.D.粉体特性測定器で測定した値である。
また、 鼻腔内に付着滞留させるためには、 高粘度のもの、 例えば、 2%水溶液の粘 度が約 400 c p s (40 OmP a · s) 以上のものが好ましく、 約 l O O O c p s (100 OmP a · s.) 以上のものがさらに好ましい。
ここで、 粘度とは、 動粘度のことであり、 37°Cにおいて、 ウベローデ粘度計によ つて求めた値である。
その表面に 「担持」 とは、 薬物が、 鼻粘膜等の薬物吸収部位で吸収されるように水 溶性高分子によって支持されている状態をいい、 付着している状態も包含する。 薬物が水溶性高分子に「付着」 しているとは、薬物が直接水溶性高分子に付着して いる場合だけでなく、結合剤などを介して間接的に付着している場合も含まれる。 こ のときの結合剤としては、粒子同士の付着を生じないようにするため、前記と異なり、 比較的低粘度のものが好ましい。例えば、 2 %水溶液の粘度が約 400 c p s (40 OmP a * s) 未満のヒドロキシプロピルセルロースが該当する。
本発明の経鼻投与製剤組成物の平均粒子径は 2 5〜5 0 0 imであり、 7 5〜3 0 0 /imが好ましく、 1 5 0〜2 5 0 mがさらに好ましい。なぜなら、 2 5 xm未満 では、鼻腔内に付着'滞留せず、 肺にまで達することが懸念され望ましくないからで あり、 5 0 0 z mを超えると粒子が大きすぎて鼻腔内壁に付着せず落下してしまうか らである。
経鼻投与製剤組成物の比重は 0 . 4〜0 . 6 gZmLであり、 0 . 4 5〜 6 g /mLが好ましく、 0 . 5〜0 . 5 8 g /mLがさらに好ましい。 なぜなら、 0 . 4 g/mL未満では、鼻腔内に付着'滞留せず、肺にまで達することが懸念され好まし くないからであり、 0 . 6 gZmLを超えると粒子が大きすぎて鼻腔内壁に付着せず 落下してしまうからである。
薬物の水溶性高分子への 「担持」 は、 薬物及び水溶性糖類を溶解させた溶液を、 造 粒機中を流動又は攪拌している水溶性高分子の粉体にスプレーノズル等を介して噴 霧し、 必要に応じて乾燥工程により溶媒を除去することによって行うことができる。 水溶性高分子を 「流動」 させる機器としては、 流動層造粒機を用いることができ、
「攪拌流動」 させる機器としては、攪拌流動層造粒機を用いることができる。重質の 粒子を形成する点でマルチプレックス (バウレック社製)やスパイラフロー (フロイ ント産業社製) のような攪拌流動層造粒機が好ましい。
本発明の経鼻投与製剤組成物は、発明の効果に支障がない限り、一般的に用いられ ている添加剤を任意に加えて経鼻投与製剤とすることができる。
一般的に用いられている添加剤としては、 例えば、 香料、 着色剤、 保存剤などが挙 げられる。
本発明の経鼻投与製剤組成物は、例えば、 ゼラチン製の硬カプセルに充填して、 本 願の経鼻投与製剤の 1態様とすることができる。
本発明の経鼻投与製剤中の有効成分は、 製剤全質量あたり、 好ましくは、 約 0 . 0 5〜約 3質量%、 より好ましくは、 0 . 1〜2質量%含有する。
本発明の経鼻投与製剤は、 一般に、薬物を含有した溶液及び粉末 (粉末をカプセル に充填したものを含む。 )を適当な投与器、例えば、点鼻器あるいは噴霧器(例えば、 投与用デバイス) を用い、鼻腔内に定量的に噴霧して投与することができる。具体的 には、粉剤を充填したカプセルを、針を備えた専用のスプレーにセットして針を貫通
させ、 それにより、 カプセルの上下に微小な孔を開け、次いで空気をゴム球などで送 り込んで粉剤を噴出させる方法などがある。
投与量は、 患者の年齢、 体重、 症状などにより異なるが、 成人に対し、 有効成分の 量として 50 g〜lmgZ日が好ましい。 発明を実施するための最良の形態
以下に実施例及び試験例を挙げて本発明をさらに詳細に説明する。
実施例 1
薬物 A 3 g及びマンニ! ^一ル 6 gを 40 %ェタノ一ル溶液 1 50 gに溶解させ る。 60〜100メッシュの範囲で分級したヒドロキシプロピルセルロース(HPC 一 H) 291 gを攪拌流動層造粒機 (MP— 01 ;パゥレック社製) に仕込み、 流動 する HP C— Hの粉体に前記エタノール水溶液を毎分 3 gの液流量で 20秒噴霧、 4 0秒乾燥の設定で間欠スプレーを行い、 経鼻投与製剤組成物を調製した。
実施例 2
マンニトールを乳糖に置き換え、実施例 1と同様にして経鼻投与製剤組成物を調製 した。
実施例 3
リン酸ニ水素ナトリゥムニ水和物 0.2 g及びリン酸水素ナトリウム 3 gを 30% エタノール水溶液 70 gに溶解させ、 さらに薬物 Aを 3 g溶解させる。 60〜100 メッシュの範囲で分級した HPC— H 295 gを攪拌流動層造粒機に仕込み、流動 する HP C—Hの粉体に前記エタノール溶液を毎分 3 gの液流量で 30秒噴霧、 40 秒乾燥の設定で間欠スプレーを行い、 経鼻投与製剤組成物を調製した。
実施例 4
薬物 A 3 g、 マンニトール 5. 5 g及びヒドロキシプロピルセルロース (HP C 一 L) 0. 5 gを 40%エタノール水溶液 150 gに溶解させた。 実施例 1に準拠し て、 経鼻投与製剤組成物を調製した。
試験例 1
下表 1記載の処方で造粒液を調製し、 HP C— H 291 gに噴霧して経鼻投与製 剤組成物の製造を試み、 その造粒状態を観察した。製造機器、 製造条件は次の通りで
ある。
製造機器:攪拌流動層造粒機 (MP— 01 ;パゥレック社製)
製造条件:風量: 30m3Zh
吸気温度: 40°C
排気温度: 25°C
スプレーガン X 1
給液流量: 3 gZmin
スプレー: 2 Osec
ドライ : 30 sec 処方及び造粒状態
* 30 %ェ夕ノ一ル水溶液にリン酸ニ水素ナトリウム 0. 2 g, リン酸水素ナ トリウム 3 gを溶解させて p Hを 8に調整した。 試験例 2
前記試験例 1で調整した実験例 1〜 3の経鼻投与製剤組成物の含量均一性を日本 薬局方第十四改正 ·一般試験法 ·含量均一性試験法を参考にして以下の試験方法によ り調べた。
製剤から 10ポイントずつサンプリングした検体の PG含量を液体クロマトダラ フ法により測定した。 また、 比重、 粒度分布も併せて測定した。 結果を表 2に示す。 判定値 = I M-X I +ks, X= (X 1 +X2+ · · · + Xn) Zn

Claims
請求の範囲
1. 式 (I)
(式中、 Xはハロゲン原子を示し、 R1は C3_10のシクロアルキル基、 のアル キル基で置換された C 0のシクロアルキル基又は C 4^ 3のシクロアルキルアル キル基を示し、 R 2は水素原子、 C^ 。のアルキル基又は C 3^ Qのシクロアルキル 基を示し、 mは 0〜3の整数、 nは 1〜4の整数を示す。 ) で表されるプロス夕ダラ ンジン誘導体及び水溶性糖類を溶媒に溶解させ、該溶液を流動又は攪拌流動中の水溶 性高分子に噴霧し、式(I)で表されるプロスタグランジン誘導体を水溶性高分子の 表面に担持させたことを特徴とする経鼻投与製剤組成物。
2.式(A)で表されるプロスタグランジン誘導体及び水溶性糖類を溶媒に溶解させ、 該溶液を流動又は攪拌流動中の水溶性高分子に噴霧し、式(A)で表されるプロス夕 グランジン誘導体を水溶性高分子の表面に担持させたことを特徴とする経鼻投与製 剤組成物。

4. 前記式 (I ) 又は(A) で表されるプロスタグランジン誘導体の 1質量部に対し て、水溶性糖類が 1〜2 0質量部、水溶性高分子が 9 0〜9 9 8質量部である請求の 範囲第 1項又は第 2項記載の経鼻投与製剤組成物。
5 . 水溶性糖類がマンニトール、 キシリトール、 エリスリトール及び乳糖の少なくと も 1種である請求の範囲第 1項又は第 2項記載の経鼻投与製剤組成物。
6 .溶媒がエタノールと水の混液である請求の範囲第 1項又は第 2項記載の経鼻投与 製剤組成物。
7 .塩基性溶媒が塩基性に調整された水一エタノール溶液である請求の範囲第 3項記 載の経鼻投与製剤組成物。
8 .水溶性高分子が架橋ピニル重合体及び水溶性セルロースエーテルの少なくとも 1 種である請求の範囲第 1項〜第 3項の何れか 1項に記載の経鼻投与製剤組成物。
9 .水溶性高分子がヒドロキシプロピルセルロース、 ヒドロキシプロピルメチルセル ロース、ポリビニルピロリドン及びメチルセルロースの少なくとも 1種である請求の 範囲第 1項〜第 3項の何れか 1項に記載の経鼻投与製剤組成物。
1 0 .水溶性高分子がヒドロキシプロピルセルロースである請求の範囲第 1項〜第 3 項の何れか 1項に記載の経鼻投与製剤組成物。
1 1 . 平均粒子径が 2 5〜5 0 0 x m、 かつ、 比重が 0 . 4〜0 . 6 g /mLである 請求の範囲第 1項〜第 1 0項の何れか 1項に記載の経鼻投与製剤組成物。

1 3 . 前記式 (I ) 又は式 (A) で表されるプロスタグランジン誘導体及び水溶性糖 類を溶媒に溶解させ、該溶液を流動又は攪拌流動中の水溶性高分子に噴霧し、式( I ) 又は(A)で表されるプロスタグランジン誘導体を水溶性高分子の表面に担持させた ことを特徴とする経鼻投与製剤組成物の製造方法。
