Abstract

Das Verfahren zur Herstellung von Peptiden, Peptidmimetika und Proteinen ist dadurch charakterisiert, dass man als Biokatalysator ein Enzym zusammen mit einem Substratmimetikum einsetzt. Die native Spezifität des Enzyms wird durch chemische oder gentechnische Manipulationen verändert, Substratmimetikum ist eine Carboxylkomponente mit einer Esterabgangsgruppe, deren Spezifitätsdeterminante der Erkennungsregion des Enzyms angepasst ist. Beansprucht wird ferner die Trypsinvariante D 189K,K60E und ihre Verwendung als CN-Ligase bei Segmentkondensationen.
