Abstract
Carbonsäureester der allgemeinen Formel (I): R1-O-C(O)-(CH¿2?)x-O-C6H4-(CH2)y-C(O)-O-R?2¿; in der R?1 und R2¿ unabhängig voneinander Alkylgruppen mit 1-4 Kohlenstoffatomen darstellen, x eine ganze Zahl von 1 bis 4 und y 1 oder 2 ist, sind entzündungshemmende Wirkstoffe, die zur Herstellung topischer pharmazeutischer und kosmetischer Mittel geeignet sind.

"Entzündungshemmende Wirkstoffe für kosmetische Präparate"
Die Erfindung betrifft die Verwendung von Carbonsäureestern als entzündungshemmende Wirkstoffe zur Herstellung von topischen pharmazeutischen- und kosmetischen Mitteln.
Es ist bekannt, die Wirkung kosmetischer und pharmazeutischer Präparate, insbesondere solcher zur Verwendung auf infolge starker Lichteinstrahlung strapazierter Haut, durch Zugabe entzündungs¬ hemmender Substanzen zu erweitern.
In der Deutschen Offenlegungsschrift 26 17 817 wurde beispiels¬ weise vorgeschlagen, kosmetischen Präparaten, insbesondere Son¬ nenschutz- und Sonnenbrandbekämpfungsmitteln, bestimmte Alk- oxybenzoesäureester als Entzündungshemmer beizugeben.
Die Eignung von bestimmten Derivaten der Phenylessigsäure, insbe¬ sondere 4-Hydroxyphenylessigsäure, und deren Estern als entzün¬ dungshemmende Wirkstoffe ist aus der japanischen Offenlegungs¬ schrift 56/18936 bekannt.
Schließlich sind in der belgischen Patentschrift 74 24 84 Carbonsäure-Derivate mit entzündungshemmenden Eigenschaften offenbart.
Wenngleich durch die Zugabe dieser Verbindungen bereits eine deutliche Entzündungshemmung erzielt wird, besteht doch ein Be¬ dürfnis nach Substanzen mit einer höheren Wirksamkeit bei gleichen oder niedrigeren Anwendungskonzentrationen.
Es wurde nun gefunden, daß sich bestimmte Carbonsäureester der allgemeinen Formel (I),
R1-0-C(0)-(CH2)x-0-C6H4-(CH )y-C(0)-0-R2 (I)
in der R- und V\~- unabhängig voneinander Alkylgruppen mit 1-4 Koh¬ lenstoffatomen darstellen, x eine ganze Zahl von 1 bis 4 und y = 1 oder 2 ist, durch eine deutlich größere entzündungshemmende Wir¬ kung auszeichnen. Diese Verbindungen lassen sich problemlos in übliche pharmazeutische und kosmetische Grundlagen für topische Applikationen einarbeiten; sie sind gut kombinierbar mit den an¬ deren üblichen Inhaltsstoffen solcher Zubereitungen und weisen keinen oder nur einen minimalen Eigengeruch auf.
Gegenstand der Erfindung ist somit die Verwendung von Carbon¬ säureestern der allgemeinen Formel (I) als entzündungshemmende Wirkstoffe zur Herstellung von topischen pharmazeutischen und kosmetischen Mitteln.
Die Verbindungen gemäß Formel (I) sind mit Hilfe bekannter che¬ mischer Umsetzungen aus leicht zugänglichen Ausgangsprodukten herstellbar.
Ein bevorzugt verwendetes Herstellungsverfahren ist die Umsetzung von 4-Hydroxyphenylalkancarbonsäure-alkylestern der Formel H0-CöH4-(CH )y-C(0)-0-R2 mit Halogenalkancarbonsäure-alkylestern der Formel R1-0-C(0)-(CH2)χ-Cl in einem organischen Lösungsmittel wie beispielsweise Aceton, 1,2-Dimethoxyethan, Diethylenglykoldi- methyl- oder -diethylether bei Temperaturen von etwa 50 °C bis 150 °C. Als Hilfsmittel zum Binden des Halogenwasserstoffs können bei¬ spielsweise Soda, Pottasche, Natrium- oder Kaliumalkoholate ver¬ wendet werden.
In den erfindungsgemäßen Verbindungen steht R1 für Methyl-, Ethyl-, n-Propyl-, Isopropyl-, n-Butyl-, 2-n-Butyl- und tert-Butyl-Gruppen. Die Methyl- und die Ethylgruppe sind bevor¬ zugte Gruppen R- .
R2 steht ebenfalls für Methyl-, Ethyl-, n-Propyl-, Isopropyl-, n-Butyl-, 2-n-Butyl- und tert.-Butyl-Gruppen. Die n-Prbpyl- und die iso-Propylgruppe sind bevorzugte Gruppen R2.
Bevorzugte Werte für x sind 1 und 2. Da y=l ebenfalls bevorzugt ist, sind die erfindungsgemäß bevorzugten Verbindungen der allge¬ meinen Formel (I) Derivate der Phenylessigsäure.
Ethoxy- und Methoxycarbonylmethoxyphenylessigsäure-n-propyTester und die entsprechenden iso-Propylester sind besonders bevorzugte Verbindungen.
Erfindungsgemäß werden die Carbonsäureester der allgemeinen Formel (I) bevorzugt in Mengen von 0,01-10 Gew.-%, insbesondere in Mengen von 0,5-5 Gew.-%, bezogen auf das gesamte Mittel,
eingesetzt. Besonders bevorzugt werden die Verbindungen der all¬ gemeinen Formel (I) in Mengen von 2-5 Gew.-%, insbesondere in Mengen von etwa 3 Gew.-%, eingesetzt.
Besonders vorteilhaft ist die Verwendung der Verbindungen der allgemeinen Formel (I) zur Herstellung von Sonnenschutz- und Son¬ nenbrandbekämpfungsmitteln. Dabei können die Carbonsäurederivate sowohl in Mitteln, die vor als auch in solchen, die nach einem Sonnenbad auf die Haut aufgebracht werden, Verwendung finden.
Die erfindungsgemäßen Carbonsäureester weisen im Erythem-er- zeugenden UV-B-Bereich von etwa 300-320 nm selbst keine Absorp¬ tionsbande auf. Zur Herstellung von Mitteln, die vor einem Son¬ nenbad auf die Haut aufgebracht werden, ist es daher bevorzugt, sie in Kombination mit üblichen UV-Filtersubstanzen einzusetzen.
Geeignete UV-Filtersubstanzen sind beispielsweise:
- Salicylsäurederivate, wie die Ester des Menthols, des Homomenthols, des Fenchols, des Bo neols, des Glycerins, des Benzylalkohols, des Phenols, des Ethylenglykols, des 2-Ethylhexanols und des tert.-Butanols sowie das Triethanola moniumsalz der Salicylsäure,
- Zimtsäureester, wie p-Methoxy-, p-Amino- und p-Dimethylamino-zimtsäureester, beispielsweise p-Methoxyzimtsäure-3-ethoxyethylester, p-Methoxyzimtsäure-2-ethylhexylester sowie p-Acetamidozimtsäure-isopropylester,
- p-Aminobenzoesäure und ihre Derivate, wie p-Aminobenzoesäure-
ethyl-, -prόpyl-, -butyl-, -isobutyl-, -monoglycerin- und -dipropylenglykolethylester, p-Dimethylaminobenzoesäureethyl- und -a ylester sowie p-Diethylaminobenzoesäureethyl- und amylester,
- substituierte Benzophenone, wie
2,2'-Dihydroxy-4,4'-di ethoxybenzophenon, 2-Hydroxy-4-n-octoxy-benzophenon, 4-Pheny1benzophenon, 2-Hydroxy-4-methoxy-benzophenon-5-sulfonsäure und 4-Phenylbenzophenon-2-carbonsäure-isooctylester,
- Cumarin-Derivate, wie 4-Methylcumarin, 6-Methoxycumarin, 7-Ethylamino-4-methyl-cumarin, 7,8-Dihydroxycumarin, 6,7-Dihydroxycumarin, 7-Hydroxycumarin, 4-Methyl-7-hydroxycumarin und 3-Phenylcumarin,
des weiteren 2-Phenylbenzimidazol-5-sulfonsäure, Natrium-3,4-di- methoxyphenylglyoxylat, Butylbenzalaceton, Benzalacetophenon, 3-To1uy1iden-D,L-campher, 3-Benz 1iden-D,L-campher, 3-(p-Methy1- benzyliden)-D,L-campher, Urocaninsäure und deren Salze, Benzimida- zol-Derivate, Digalloyltrioleat, Sulfonamide, Hydrazone, Kondensa¬ tionsprodukte des Tannins mit Ethylenoxid sowie als Naturprodukte Extrakte von Zimtrinde, Ratanhia und Süßholz.
Eine Zusammenstellung geeigneter UV-Filtersubstanzen findet sich auch in der Deutschen KosmetikVerordnung (Anlage 7 zu § 3b).
Die genannten UV-Filtersubstanzen werden üblicherweise in Mengen von 1-10 Gew.-%, bezogen auf das gesamte Mittel, eingesetzt. Men¬ gen von 2-6 Gew.-% sind bevorzugt.
Weiterhin ist es erfindungsgemäß bevorzugt, den erfindungsgemäßen Mitteln Antioxidation zuzufügen. Besonders bevorzugt ist hierbei die Verwendungen von natürlichen oder naturidentischen Verbindun¬ gen wie beispielsweise Tocopherolen. Die genannten Antioxidantien sind in den erfindungsgemäßen Mitteln in Mengen von 0,1-5 Gew.-%, insbesondere von 0,5-2 Gew.-%, bezogen auf das gesamte Mittel, enthalten.
Die erfindungsgemäßen Mittel können als flüssige, pastöse oder feste Zubereitungen formuliert werden, beispielsweise als wäßrige oder alkoholische Lösungen, wäßrige Suspensionen, Emulsionen, Salben, Cremes, Öle, Pulver oder Stifte. In Abhängigkeit von der gewünschten Formulierung können die Carbonsäurederivate in phar¬ mazeutischen und kosmetischen Grundlagen für topische Applikati¬ onen eingearbeitet werden, die als weitere Komponenten beispiels¬ weise Ölkomponenten, Fette und Wachse, Emulgatoren, anionische, kationische, ampholytische, zwitterionische und/oder nichtionogene Tenside, niedere ein- und mehrwertige Alkohole, Wasser, Konser¬ vierungsmittel, Puffersubstanzen, Verdickungsmittel, Duftstoffe, Farbstoffe und Trübungsmittel enthalten.
B E I S P I E L E
Die nachfolgenden Beispiele dienen zur weiteren Erläuterung der Erfindung, ohne diese darauf zu beschränken.
1. Herstellung der Carbonsäureester
a) 4-Ethoxycarbonylmethoxy-phenylessigsäure-isopropylester
Eine Mischung aus 19,4 g (0,1 Mol) 4-Hydroxyphenylessig- säureisopropylester, 18,4 g (0,11 Mol) Bromessigsäure- eth Tester, 13,8 g (0,1 Mol) Kaliumcarbonat und 200 ml Aceton wurde 12 Stunden unter Rückfluß erhitzt. Danach wurde der Niederschlag abfiTtriert, das Aceton abdestiTliert und die verbleibende Flüssigkeit einer fraktionierten Destillation unterworfen. Die Ausbeute an 4-Ethoxycarbonylmethoxy-phenyl- essigsäure-isopropylester betrug 26,6 g (95 % d. Th.). Das Produkt hatte einen Siedepunkt von 183-185 °C bei einem Druck von 1,2 mbar und einen Brechungsindex no20 = 1,4961.
Nach analogen Synthesevorschriften wurden erhalten:
b) 4-Methoxycarbonylmethoxy-pheny1essigsäure-isopropy1ester
Siedepunkt: 155-156 °C / 0,05 mbar ; ΠQ20 = 1,5021.
c) 4-Ethoxycarbonylmethoxy-phenylessigsäure-n-propylester
Siedepunkt: 147 °C / 0,05 mbar ; nD 20 = 1,5091.
d) 4-Ethoxycarbonylmethoxy-phenylessigsäure-n-butylester
Hier erfolgte die Reinigung säulenchromatographisch.
nD 20 = 1,4940
e) 4-EthoxycarbonyTmethoxy-phenyTessigsäure-isobutyTester
nD 20 = 1,4909.
2. Testverfahren für die entzündungshemmende Wirkung
Zur Bestimmung der entzündungshemmenden Wirkung der Substanzen wurde ein Test an haarTosen Mäusen auf Hautschädigung durch UV- Licht durchgeführt.
Die haarTosen Mäuse wurden am Rücken mit einer UV-Lampe (Ultravi- talux-La pe, 0SRAM) aus 45 cm Entfernung 3 Minuten lang bestrahlt, wodurch eine meßbare Hautentzündung (Ödem) ausgelöst wurde. Un¬ mittelbar nach Ende der Bestrahlung und 1, 2 und 3 Stunden danach wurden die PrüfSubstanzen in Form einer ethanoTischen Lösung (2 x 20 μT pro Auftragung mittels einer automatischen Pipette) aufge¬ tragen und mit einem Glasstab sanft einmassiert. Als Vergleichs¬ versuch wurde bei in gleicher Weise bestrahlten Mäusen eine Nachbehandlung mit dem Trägerstoff Ethanol (Kontrolle) durch-
geführt. Nach '30 Stunden wurde der Grad der Ödembildung durch Messung der Hautfaltendicke bestimmt. Durch Vergleich der Ver¬ änderung der Hautfaltendicke aufgrund der Bestrahlung bei be¬ handelten und nur mit dem Trägerstoff behandelten Tieren wurde der Grad der Hemmung des Ödems bestimmt.

Claims
1. Verwendung von Carbonsäureestern der allgemeinen Formel (I),
R1-0-C(0)-(CH2)χ-0-C6H4-(CH2)y-C(0)-0-R2 (I)
in der R1 und R2 unabhängig voneinander Alkylgruppen mit 1-4 Kohlenstoffatomen darstellen, x eine ganze Zahl von 1 bis 4 und y = 1 oder 2 ist, als entzündungshemmende Wirkstoffe zur Herstellung von topischen pharmazeutischen und kosmetischen Mitteln.
2. Verwendung nach Anspruch 1, dadurch gekennzeichnet, daß in den Carbonsäureestern der allgemeinen Formel (I) R* eine Methyl¬ gruppe oder eine Ethylgruppe ist.
3. Verwendung nach einem der Ansprüche 1 oder 2, dadurch gekenn¬ zeichnet, daß in den Carbonsäureestern der allgemeinen Formel (I) R2 eine Isopropyl-Gruppe ist.
4. Verwendung nach einem der Ansprüche 1 bis 3, dadurch gekenn¬ zeichnet, daß die Carbonsäureester der allgemeinen Formel (I) Derivate der Phenylessigsäure sind.
5. Verwendung nach einem der Ansprüche 1 bis 4, dadurch gekenn¬ zeichnet, daß die Carbonsäureester der allgemeinen Formel (I) in einer Menge von 0,01 bis 10 Gew.-%, insbesondere von 0,5 bis 5 Gew.-%, bezogen auf das gesamte Mittel, eingesetzt wer¬ den.
6. Verwendung nach einem der Ansprüche 1 bis 5, dadurch gekenn¬ zeichnet, daß die^ pharmazeutischen und kosmetischen Mittel Sonnenschutz- und Sonnenbrandbekämpfungsmittel sind.
7. Verwendung nach einem der Ansprüche 1 bis 6, dadurch gekenn¬ zeichnet, daß die Carbonsäureester der allgemeinen Formel (I) in Kombination mit üblichen UV-FilterSubstanzen eingesetzt werden.
8. Verwendung nach Anspruch 7, dadurch gekennzeichnet, daß die UV-Filtersubstanzen in Mengen von 1 bis 10 Gew.-%, insbeson¬ dere 2 bis 6 Gew.-%, bezogen auf das gesamte Mittel, einge¬ setzt werden.
9. Verwendung nach einem der Ansprüche 1 bis 8, dadurch gekenn¬ zeichnet, daß die Carbonsäureester der allgemeinen Formel (I) in Kombination mit Antioxidantien eingesetzt werden.
10. Verwendung nach Anspruch 9, dadurch gekennzeichnet, daß die Antioxidantien in Mengen von 0,1 bis 5 Gew.-%, insbesondere von 0,5 bis 2 Gew.-%, bezogen auf das gesamte Mittel, einge¬ setzt werden.
