Description
本發明係關於包含迷迭香酸或含有迷迭香酸之萃取物之組合物及係關於水解酶或能夠產生水解酶之微生物在此等組合物中之用途。本發明亦係關於改良含有迷迭香酸及其衍生物之組合物的生物效應之方法。
迷迭香酸為在大量天然來源中發現之多酚,該等天然來源包括迷迭香、牛至、甘牛至草、百里香、胡椒薄荷、康福利草(comfrey)、歐芹、香蜂草、鼠尾草、牛膝草、羅勒、薰衣草等。此等植物已報導為具有抗氧化、消炎、抗癌及護肝之性質的草本藥物。此等生物效應與多酚,例如特別迷迭香酸、黃酮及黃烷醇(在迷迭香葉中鑑別)之存在相關。
此項技術中尚未描述關於改良包含迷迭香酸或包含該迷迭香酸之植物萃取物的組合物之生物效應的方式。
因此,仍需要改良包含迷迭香酸或包含該迷迭香酸及其衍生物之植物萃取物的組合物之生物效應。
因此,此目標藉由獨立請求項之特徵來解決。附屬請求項進一步發展本發明之中心思想。
因此,在一第一態樣中,本發明係關於一種包含迷迭香酸及/或其衍生物以及水解酶及/或含有或產生該酶之微生物的組合物。
在一第二態樣中,本發明係關於一種包含迷迭香酸及/或其衍生物以及其水解產物之組合物。
如請求項1至15中任一項之用於改良或預防對皮膚之損害的組合物亦為本發明之部分。
此外,本發明包含如請求項1至15中任一項之組合物之化妝品用途。
在另一態樣中,本發明係關於水解酶或含有或產生該酶之微生物用於水解迷迭香酸之用途。
水解酶或含有或產生該酶之微生物在包含迷迭香酸及/或其衍生物之組合物中用於改良該組合物之生物效應的用途屬於本發明之又一態樣。
此外,本發明亦提供一種製備包含迷迭香酸及/或其衍生物之具有增加之生物效應的組合物之方法,其包含以下步驟:a.提供迷迭香酸及/或其衍生物,b.提供水解酶或含有或產生該酶之微生物,及c.將該迷迭香酸及/或其衍生物及水解酶或含有或產生該酶之微生物與其他成份混合以產生該組合物,以使迷迭香酸之水解發生在組合物中或活體內。
最終,提供一種用於改良受檢者健康之方法,其包含投與如請求項1至15中任一項之組合物之步驟。
本發明係關於包含迷迭香酸及其衍生物之組合物。組合物可為"食品組合物"或"局部組合物"。"食品組合物"意謂可用於達成營養、醫藥及/或化妝目的之任何可消費產品。"局部組合物"意謂可塗覆於受檢者之皮膚、皮毛或毛髮以達成醫藥及/或化妝目的之任何組合物。組合物較佳為食品組合物。
迷迭香酸(圖1)及其衍生物通常在植物中見到,諸如迷迭香、牛至、甘牛至草、百里香、胡椒薄荷、康福利草、歐芹、香蜂草、鼠尾草、牛膝草、羅勒、薰衣草、紫蘇等。
迷迭香酸之"衍生物"意謂包含至少一迷迭香酸部分之任何化合物。此等化合物包括迷迭香酸之寡聚物、聚合物、糖苷等。
迷迭香酸及其衍生物可以純化合物形式提供或藉由各種藥草,諸如紫蘇(香蘇葉,Perilla frutescens L.
)、鼠尾草(洋蘇草,Salvia officinalis L.
)、薄荷(蘇薄荷,Mentha arvense L.
)、羅勒(羅勒香,Ocimum basilicum L.

)等來提供。其較佳藉由迷迭香(迷迭香,Rosmarinus officinalis L.
)來提供。
此外,萃取含有迷迭香酸來源之植物可使用此項技術中已知之方法進行。實際上,本發明之組合物較佳包含植物萃取物,該等植物萃取物包含迷迭香酸及/或其衍生物。萃取物較佳以用乾物質計之0.01 g至20 g之範圍內的量存在於組合物中。
因此,本發明提供一種組合物,該組合物包含迷迭香酸及/或其衍生物以及能夠水解迷迭香酸或其衍生物的水解酶(例如,酯酶)或含有或產生水解酶之微生物。
由於酶對迷迭香酸及/或其衍生物具有水解作用,所以組合物可進一步包含在該酶下迷迭香酸及/或其衍生物之水解產物。此等水解產物可包括咖啡酸、3,4-二羥基苯乳酸等,及其混合物。
本發明之組合物可為無水、濕潤或半濕潤組合物。"無水"意謂水活性低於0.6之組合物。"半濕潤"意謂水活性在0.6與0.9之間的組合物,且"濕潤"意謂水活性在0.9以上之組合物。
組合物可包含合適之食品或醫藥載劑。此等載劑包括牛奶、優格(yogurt)、乳酪、醱酵牛奶、基於牛奶之醱酵產品、冰淇淋、基於醱酵穀物之產品、基於牛奶之粉末、兒童及嬰兒配方乳品、動物(尤其寵物)食品、錠劑或口含劑、液體細菌懸浮液、呈無水形式之經口增補劑及呈液體形式之經口增補劑。
因此,組合物可為呈以下形式之食品組合物:完全營養食品、飲料、礦泉水、湯、食品增補劑及替代食品、溶液、噴霧、散劑、錠劑、膠囊、營養棒、液體細菌懸浮液、糖食、基於牛奶或基於醱酵牛奶之產品、優格、基於牛奶之粉末、腸內營養產品、用於兒童及/或嬰兒之組合物、基於穀物之產品或基於醱酵穀物之產品、基於大豆之產品、冰淇淋、巧克力、咖啡、"烹調"產品(諸如,蛋黃醬、番茄醬或沙拉調料)、寵物食品等。因此,組合物亦可意欲用於動物。
為攝取,經口組合物及尤其食品增補劑之許多實施例皆為可能。其係藉助於用於製造糖衣錠劑、明膠膠囊、凝膠、乳液、錠劑、膠囊或溶液常用之方法而調配。詳言之,包含迷迭香酸及/或其衍生物結合水解酶或含有或產生該酶之微生物的組合物可併入任何其他形式之食品增補劑或強化營養食品中,例如食品棒或壓實或未壓實粉末。粉末可用水稀釋至發泡飲料、乳製品或大豆衍生之產品中或可併入食品棒。
或者,組合物可為呈以下形式之局部組合物:水性、水性-醇或油性溶液;溶液類型之分散液或洗液或漿液類型之分散液;具有液體或半液體稠度之乳狀物類型之乳液,藉由脂肪相分散於水相(O/W)或水相分散於脂肪相(W/O)而獲得;或具有軟、半固體或固體稠度之乳膏、水性凝膠或無水凝膠類型之懸浮液或乳液;或微乳液;微膠囊或微粒;或離子及/或非離子類型之泡狀分散液。
本發明之組合物可包含常用賦形劑及組份(例如脂肪及/或水性組份)、濕潤劑、增稠劑、防腐劑、紋理劑、芳香劑及/或塗佈劑、抗氧化劑、染料,該等物質在食品及/或局部領域常用。
存在於組合物中之水解酶較佳為酯酶。
本發明之根本原理在於某些水解酶能夠將迷迭香酸(圖1)裂解成咖啡酸(圖2)及3,4-二羥基苯乳酸(圖3),而此等物質比迷迭香酸易於為生物利用。因此,包含迷迭香酸或植物萃取物(包含迷迭香酸)之組合物的生物效應得以改良。
藉由提供包含該水解酶之組合物,本發明允許上述迷迭香酸轉變成咖啡酸及3,4-二羥基苯乳酸之過程在食品基質自身中、在用於局部塗覆之產品自身中或在活體內消化過程之早期或在局部塗覆後發生,因此增加包含迷迭香酸及/或其衍生物之組合物的生物效應。
因此,含有迷迭香酸及/或其衍生物之本發明之組合物具有改良之生物效應。
或者或另外,本發明之組合物可包含含有或產生該水解酶之微生物。
存在於本發明之組合物中之酶或微生物的量為使得至少10%,較佳至少30%,更佳至少50%之迷迭香酸(圖1)可水解成咖啡酸(圖2)及3,4-二羥基苯乳酸(圖3)之量。此可易於藉由熟習此項技術者藉由此項技術中已知之方法來測定。
視應用而定,水解酶可以0.01至1000 U/g之範圍內的量存在於組合物中。
當微生物用於組合物時,其較佳以1×103

用於本發明之組合物中之微生物可選自細菌、酵母或真菌。微生物較佳為益生微生物。更佳地,其係選自約氏乳桿菌(Lactobacillus johnsonii
,CNCM I-1225)、長雙岐桿菌(Bifidobacterium longum
,ATCC BAA-999)、乳雙岐桿菌(Bifidobacterium lactis
,CNCM I-3446)或其混合物。
在本發明之另一態樣下,提供一種包含迷迭香酸或包含迷迭香酸及/或其衍生物之植物萃取物以及其水解產物之組合物。較佳地,水解產物藉由酶裂解迷迭香酸及/或其衍生物而獲得。該水解產物可選自咖啡酸、3,4-二羥基苯乳酸等及其混合物。
本發明之組合物尤其適用於改良或預防對皮膚之損害。實際上,已發現其尤其適用於抑制基質金屬蛋白酶之表現。基質金屬蛋白酶較佳為1型。圖8展示在構成MMP1釋放中水解對包含迷迭香酸之萃取物之影響。可見與對照、阿魏酸(ferulic acid)及包含迷迭香酸之萃取物(未水解)相比,至少部分水解之迷迭香酸具有對構成MMP1釋放顯著之作用,此使其尤其適用於改良及/或預防對皮膚之損害。
根據一實施例,本發明之組合物可用於化妝上。在此實施例中,組合物可經設計以用於經口消耗或用於局部塗覆。
"化妝用途"意謂可改良人類或寵物之皮膚、皮毛及/或毛髮之美學方面或舒適的非治療用途。
在此背景下,化妝用途可包括預防對人類或寵物之皮膚、皮毛及/或毛髮的損害及/或改良人類或寵物之皮膚、皮毛及/或毛髮。該等損害尤其包括皮膚之光化及老化損害,諸如乾燥、不規則色素澱積(特別斑點、雀斑、滴狀黑色素過少症及持久性色素沈著過度)、皺紋(特別細微表面皺紋及深皺紋)、星狀假疤、彈性組織變性、無彈性、毛細血管擴張、靜脈湖、粉刺、皮脂腺增生、軟垂疣及皮脂溢性角化症。
化妝用途亦可對毛髮及皮毛特別有用,諸如改良毛髮或皮毛密度、纖維直徑、顏色、含油性、光澤性、皮脂產生,且可有助於防止毛髮或皮毛損失。
當用於化妝上時,本發明之組合物可呈現任何形式之上述食品組合物。其較佳呈可為液體或無水形式之食品增補劑形式,諸如溶液、噴霧、錠劑、膠囊、明膠膠囊、口含劑、散劑、凝膠、乳液等。其更佳呈膠囊形式。
或者,組合物可呈現可呈以下形式之局部組合物形式:水性、水性-醇或油性溶液;溶液類型之分散液或洗液或漿液類型之分散液;具有液體或半液體稠度之乳狀物類型之乳液,藉由脂肪相分散於水相(O/W)或水相分散於脂肪相(W/O)而獲得;或具有軟、半固體或固體稠度之乳膏、水性凝膠或無水凝膠類型之懸浮液或乳液;或微乳液;微膠囊或微粒;或離子及/或非離子類型之泡狀分散液。
根據熟習此項技術者已知之常用方法製備此等組合物。在一較佳實施例中,此等局部組合物經設計使得酶及/或微生物僅在局部塗覆後與迷迭香酸相互作用。
用於局部塗覆之組合物可尤其構成面部、手、足、重大組織皺紋或身體之清潔、保護、治療或護理乳霜(例如,日霜、晚霜、卸妝霜、粉底霜、防曬霜);化妝品,諸如粉底液、卸妝乳、身體之防護或護理乳、曬後乳、護膚液、護膚啫哩或泡沫,諸如清潔或殺菌洗劑、防曬液、人工著色液、沐浴組合物、含有殺菌劑之除臭組合物、鬚後啫哩或鬚後水、脫毛膏或用於蟲啶之組合物、肥皂、潔膚皂、潔膚霜、潔膚啫哩、潔膚乳液或慕斯、用於毛髮之氣霧劑組合物等。
在另一態樣中,本發明係關於水解酶或含有或產生水解酶之微生物用於水解迷迭香酸(圖1)之用途。參考圖4至7,迷迭香酸水解成咖啡酸(圖2)及3,4-二羥基苯乳酸(圖3)可視所用酶量、進行水解之pH值及溫度及存在之迷迭香酸量而定。
因此,本發明提供水解酶或含有或產生該水解酶之微生物在包含迷迭香酸及/或其衍生物之組合物中用於改良該組合物之生物效應的用途。
當微生物用於此目的時,其較佳選自細菌、酵母或真菌。其更佳為益生微生物。其最佳選自約氏乳桿菌(CNCM I-1225)、長雙岐桿菌(ATCC BAA-999)、乳雙岐桿菌(CNCM I-3446)或其混合物。
根據本發明之另一實施例,給出製備包含迷迭香酸及/或其衍生物之具有增加之生物效應的組合物之方法。
該方法包含提供迷迭香酸及/或其衍生物之第一步。此可藉由各種藥草來提供,諸如迷迭香、牛至、甘牛至草、百里香、紫蘇、胡椒薄荷、康福利草、歐芹、香蜂草、鼠尾草、牛膝草、羅勒、薰衣草等或其混合物,或藉由萃取天然來源來提供。
迷迭香酸較佳以用乾物質計之0.01 mg至10 g之範圍內的量提供。
其次,提供水解酶或含有或產生該水解酶之微生物。
當酶如此使用時,其較佳以0.01-1000 U/g範圍內之量提供。
若使用微生物,其較佳以1×103
至1×1013
cfu/g範圍內之量提供。微生物可選自細菌、酵母或真菌。根據一較佳實施例,微生物為益生微生物。更佳地,其係選自約氏乳桿菌(CNCM I-1225)、長雙岐桿菌(ATCC BAA-999)、乳雙岐桿菌(CNCM I-3446)或其混合物。
接著該方法之第三步在於將迷迭香酸及/或其衍生物及水解酶及/或含有或產生該酶之微生物與其他成份混合以產生該組合物,以使迷迭香酸之水解發生在組合物中或活體內。
因此,上述迷迭香酸轉變成咖啡酸及3,4-二羥基苯乳酸之過程可發生在食品基質自身中、在用於局部塗覆之產品自身中或在活體內消化過程之早期或在局部塗覆後,因此增加包含迷迭香酸及/或其衍生物之該等組合物的生物效應。
其他成份可包括用於製造食品、局部組合物之任何成份及功能性成份。此等包括碳水化合物、蛋白質、脂質、纖維、維生素、益菌助生質、礦物質、植物萃取物等。
所得組合物可呈諸如上述彼等組合物之任何形式。
本發明進一步包含一種用於改良受檢者健康之方法,其包含投與如請求項1至15中任一項之組合物之步驟。投藥可為局部或經口。
本發明之組合物實際上可充當抗氧化劑、消炎劑及抗高血壓劑。此外,其在預防或治療心血管及腦血管疾病、預防2型糖尿病及腦退化性疾病上具有積極作用。
此外,本發明之組合物可經投與以治療及/或預防皮膚損害,該等損害例如藉由應力情形,例如藉助於化學、生物或物理應力,例如藉由暴露於氧化劑或致癌劑,暴露於細菌、病毒、真菌、衍生自周圍細胞之脂質及/或微生物或曝露於紫外線照射而產生。
此等損害進一步包含光化性角化病、紫癜、櫻桃狀血管性水腫、基底細胞癌及鱗狀細胞癌、皮膚燒傷及/或起泡、表皮增生、發炎、免疫抑制及癌症,例如非黑素瘤及黑素瘤皮膚癌。
本發明之組合物對人類或寵物之皮膚的作用可藉由習知方法來量測,包括最小紅斑量(MED)、比色法、經表皮水分丟失、DNA修復、量測介白素及蛋白聚糖產生或膠原酶活性、障礙函數或細胞更新。因此,本發明之組合物適用於改良及預防對皮膚之損害。
應瞭解本發明之概念可同樣用作佐劑治療,幫助目前使用之藥物治療。因為本發明之組合物可易於與食品物質一起經口投與,所以可投與含有高量本發明之物質之特定臨床食品。
藉助於下述非限制性實例進一步說明本發明。
經口投與之化妝品
呈硬膠囊形式之組合物具有以下調配物:
組合物可以每日2至3粒膠囊之量投與個體。
藉由以1×105
至1×1013
cfu之細菌對1 g至10 g之迷迭香萃取物(或迷迭香酸)的比例混合迷迭香萃取物(或迷迭香酸)與約氏乳桿菌(Lal)來製備食品配方。所得混合物添加或摻合有任何合適之載劑,例如醱酵牛奶、優格、新鮮乳酪、凝乳、糖食棒、早餐穀物薄片或棒、飲料、奶粉、基於大豆之產品、非乳醱酵產品或用於臨床營養之營養增補劑。
迷迭香酸藉由酶之裂解酶量之影響
在40℃下使用不同量之酶(每毫克迷迭香酸0.01 U、0.02 U及1 U酶)在磷酸鹽緩衝液(50 mM,pH 6.6)中執行試驗。如圖4所示,反應速率與酶量成比例且當氯原酸酯酶以每毫克受質1 U使用時,在僅30 min反應時間後迷迭香酸完全轉變成咖啡酸及3,4-二羥基苯乳酸(資料未圖示)。
pH值之影響
研究pH值對迷迭香酸之酶水解(0.02 U/mg)的影響。圖5展示在pH值5.0、6.0、7.0及8.0下咖啡酸之產生。在pH 8.0下,迷迭香酸在2 h反應時間後完全轉變而在pH 6.0下反應速率極低且在pH 5.0下未觀測到水解。
溫度之影響
為研究溫度對反應速率及產物穩定性之影響,在30℃、40℃及50℃下使用每毫克受質0.02 U酶在pH 6.6下於磷酸鹽緩衝液(50 mM)中執行試驗。如圖6所示,當反應在40℃下或50℃下進行時未觀測到顯著差別,而在30℃下反應速率減慢許多。
受質濃度之影響
在40℃、pH 6.6下使用0.02單元酶及1 mg、2 mg、4 mg或6 mg迷迭香酸,研究受質濃度對反應速率及產率之影響。圖7展示反應速率及產率受酶/受質之比率強烈影響。
實際上,當使用1 mg受質時90%以上之迷迭香酸在4 h後水解,而當在相同條件下使用6 mg受質時觀測到僅40%迷迭香酸水解。
用微生物裂解迷迭香酸1.與約氏乳桿菌(CNCM I-1225)一起培育
使細菌在37℃下缺氧狀態下MRS培養基中生長16小時。離心後,用10 ml磷酸鈉緩衝液(50 mM,pH 8)洗滌細胞(菌絲球)3次。將細胞(0.60 g)懸浮於1 ml磷酸鈉緩衝液(50 mM,pH 8)中且添加迷迭香酸(5 mg/mL)且在45℃下培育混合物在24 h間。每半小時取出一等分試樣用於動力學研究。離心後,藉由HPLC分析上清液。培育4 h後,所有迷迭香酸轉變成咖啡酸及3,4-羥基苯乳酸。
2.與乳雙岐桿菌(CNCM I-3446)一起培育
使細菌在37℃下缺氧狀態下MRS+0.05%半胱胺酸培養基中生長16小時。離心後,用10 ml磷酸鈉緩衝液(50 mM,pH 8)洗滌細胞(菌絲球)3次。將細胞(0.60 g)懸浮於1 ml磷酸鈉緩衝液(50 mM,pH 8)中且添加迷迭香酸(5 mg/mL)且在45℃下培育混合物在24 h間。每半小時取出一等分試樣用於動力學研究。離心後,藉由HPLC分析上清液。培育24 h後,50%以上之迷迭香酸轉變成咖啡酸及3,4-二羥基苯乳酸。
測試條件,MMP-1(基質金屬蛋白酶)劑量(ELISA)處理及照射
將纖維母細胞置放於96孔盤中之培養基中。預培養24 h後,用含有待測試之產物之培養基(或無產物以進行參考)替代該培養基。一式三份地進行各實驗條件(使用3個培養孔)。
在待測試之產物存在(缺乏)下培養24小時後,將培養基移除且藉由緩衝溶液(EBSS)替代且使細胞遠離任何光源。
在培育結束時,EBSS溶液由含有待測試之化合物之培養基替代且將培養盤置放於37℃、5% CO2
下。
處理48 h後,收集培養基上清液以進行ELISA MMP-1劑量。接著藉由根據標準程序量測活細胞之脫氫酶對MTT之還原,在細胞層上評估細胞存活力。
腸原酶之定量(MMP-1)
根據供應者之說明,使用"BIOTRACK人類MMP-l,ELISA系統,Amersham RPN2610"套組藉由ELISA獲得MMP-1劑量(人類間質膠原酶)。
資料處理
傳送計數資料且用PRISM軟體(Graph Pad Software)處理計數資料。藉由方差分析(ANOVA)使用鄧恩氏多重比較測試(Dunnett's multiple comparison test)進行組間比較。
下表展示藉由纖維母細胞之構成MMP-1釋放。*
說明相對於參考文獻之顯著差別(p<0.01)。Naturex為市售之包含迷迭香酸之天然萃取物。
藉由用約氏乳桿菌之益生菌株處理包含迷迭香酸之天然萃取物來獲得經水解之Naturex。在培育期間,迷迭香酸至少部分轉變成咖啡酸及3,4-二羥基苯乳酸。
上表中展示之結果證明與未經水解之迷迭香酸相比,至少部分水解之迷迭香酸提供皮膚益處。更特定言之,其展示本發明之組合物對MMP-1表現之抑制作用。

Claims (25)
- 一種組合物,其包含a)迷迭香酸,及b)選自氯原酸酯酶之水解酶,及/或含有或產生水解酶之益生微生物,其中該水解酶為酯酶,其中該益生微生物係選自約氏乳桿菌(Lactobacillus johnsonii,CNCM I-1225)、長雙岐桿菌(Bifidobacterium longum,ATCC BAA-999)、乳雙岐桿菌(Bifidobacterium lactis,CNCM I-3446)或其混合物,其中水解酶能夠將迷迭香酸水解成咖啡酸及3,4-二羥基苯乳酸,及其中該組合物包含組份a)經由該酶之水解產物,或其中迷迭香酸之水解發生於活體內。
- 如請求項1之組合物,其經調配以用作營養、醫藥或化妝產品。
- 如請求項1之組合物,其中該組合物為營養組合物或食品組合物。
- 如請求項3之組合物,其中該食品組合物呈以下形式:完全營養食品、飲料、礦泉水、湯、食品增補劑及替代食品、溶液、噴霧、散劑、錠劑、膠囊、營養棒、液體細菌懸浮液、糖食、基於牛奶或基於醱酵牛奶之產品、優格(yogurt)、基於牛奶之粉末、腸內營養產品、用於兒童及/或嬰兒之組合物、基於穀物之產品或基於醱酵穀物 之產品、基於大豆之產品、冰淇淋、巧克力、咖啡、"烹調"產品、寵物食品等。

- 如請求項17之方法,其中該迷迭香酸由各種藥草,藉由萃取天然來源或其混合物來提供。
- 如請求項18之方法,其中該藥草係選自由迷迭香、牛至(oregano)、甘牛至草(marjoram)、百里香(thyme)、紫蘇(perilla)、胡椒薄荷(peppermint)、康福利草(comfrey)、歐芹(parsley)、香蜂草(balm)、鼠尾草(sage)、牛膝草(hyssop)、羅勒(basil)及薰衣草(lavender)所組成之群。
- 如請求項17至19中任一項之方法,其中該等其他成份係選自碳水化合物、蛋白質、脂質、維生素、益菌助生 質、礦物質、植物萃取物、纖維、賦形劑、芳香劑。
