透明質酸(Hyaluronan)(透明質酸(hyaluronic acid):HA)是一種糖胺聚糖,發現於許多細胞的細胞外基質中,尤其是結締組織中。透明質酸亦主要發現於哺乳動物的皮膚、軟骨和滑液中。透明質酸亦是眼睛玻璃體液的主要成分。透明質酸在各種生理過程諸如水和血漿蛋白的體內平衡中發揮作用(Laurent TC等人(1992) FASEB J 6: 2397-2404)。某些疾病與透明質酸的表達和/或產生有關。透明質酸酶是一種分解透明質酸的酶。透明質酸酶可用於透過催化透明質酸的水解來治療與透明質酸或其他糖胺聚糖積累相關的疾病或疾患。此外,因為透明質酸是皮下皮膚屏障的主要成分,透明質酸酶可用于增加組織滲透性,因此,當皮下注射時,可增加治療劑的分散和遞送。
基於天然PH20多肽的各種透明質酸酶(例如,Hydase™、Vitrase™、Wydase™)已經與其他治療劑結合使用,通常作為分散劑。許多這些透明質酸酶包含來源於綿羊或牛睾丸的PH20。
人PH20蛋白由509個胺基酸組成,並且已知為存在於精子的質膜中的糖基磷酸肌醇脂質錨定蛋白。雖然血液中存在的透明質酸酶Hyal1、Hyal2、Hyla3或Hyal4僅在酸性pH下有活性,而PH20即使在中性pH下亦有活性,所以PH20已被開發成與藥物混合用於皮下注射,並已在工業上使用。

同時,人PH20,一種在六個位點被N-連接糖基化的糖蛋白,具有非常複雜的三維結構,並且為了生產工業上有用的酶,在動物細胞諸如CHO細胞中表達後需要複雜的純化過程。因此,對於人PH20的工業實用性,根據其在發酵培養基中的表達水準提高含有人PH20基因的動物細胞的生產率是非常重要的。
在這一技術背景下,本發明的發明人已經確定N-末端或N-和C-末端的切割可以提高可溶性重組PH20多肽的生產率,並完成了本發明。

具體地,根據本發明的其中一個或多個胺基酸殘基從成熟動物野生型PH20的N-末端缺失的重組PH20多肽可被表徵為選自但不限於由以下組成的組:

本發明提供了編碼重組PH20多肽的核酸和包含該核酸的重組表達載體。
本發明還提供了用該重組表達載體轉化的宿主細胞,以及生產N-末端和/或C-末端切割的可溶性PH20多肽的方法,包括培養宿主細胞的步驟。
除非另有定義,否則本文中使用的所有技術和科學術語具有與本發明所落入領域的具有通常知識者通常理解的相同的含義。通常,本文所用的術語是本領域公知的且常用的。
為了擴大在PH20的C-末端處切割並轉化為可溶性形式的重組PH20多肽的工業實用性,本發明透過進一步切割PH20的N-末端,提供了生產率顯著提高的N-末端和/或C-末端切割的PH20多肽。本文使用的術語「可溶性」是指具有在水溶液中具有活性而不會聚集的三維結構的形式。
因此,本發明涉及重組PH20多肽,其中從成熟動物野生型PH20的N-末端缺失1至7個胺基酸殘基、優選1至6個胺基酸殘基、優選1至5個胺基酸殘基、優選1至4個胺基酸殘基、優選1至3個胺基酸殘基、優選1至2個胺基酸殘基、優選1個胺基酸殘基。
本發明中的成熟動物野生型PH20可以指表現出所需功能形式的重組PH20多肽。根據本發明的成熟動物野生型PH20可以指,例如,其中促進成熟動物野生型PH20分泌到細胞外的訊息肽在分泌過程中被切割的狀態。
動物可以是,例如但不限於哺乳動物、囓齒動物等,諸如人、大鼠、小鼠、倉鼠、兔、豬、牛、鹿、羊、猴等。
根據本發明的動物野生型PH20可以包括,例如,但不限於:

人體的透明質酸酶存在於血漿或其他器官中,並且包括在酸性pH下有活性的Hyal1、Hyal2、Hyal3和Hyal4,以及在精子頂體中表達並在受精中起作用的PH20。
具有SEQ ID No. 1的胺基酸序列的人PH20多肽由M1至T35的訊息序列、L36至S490的透明質酸酶活性位點、以及A491至L509的C-末端GPI(糖基-磷脂醯肌醇)錨定序列組成,並且轉化為缺乏所有C-末端GPI錨定序列的可溶性形式的重組PH20多肽用作體內治療劑或分散劑。特別是,據報導,即使當C-末端胺基酸序列的某一部分(即I465至L509之間的任何部分)被切割時,酶活性得以保留,同時保持溶解性。對於商業產品,C-末端終止於第482位。然而,這種情況保持了除訊息序列之外的從N-末端的L36開始的序列(WO 2010/077297A等)。
最近,提出了具有從N37、F38、R39、A40、P41或P42以及L36開始的經切割的N-末端的透明質酸酶變體具有酶活性,並且在此基礎上,已經開發了具有良好熱穩定性和酶活性的透明質酸酶變體(WO 2020/022791A等人)。
猴(長鼻猴)的PH20多肽與人PH20多肽具有93.1%的序列同一性。因此,基於圖4中多重序列分析的結果,可以預期具有終止於Y483的C-末端的猴(長鼻猴)的PH20多肽是具有活性和可溶性的PH20。
牛(家牛)的PH20多肽和綿羊(綿羊(Ovis aries))的PH20多肽彼此之間有90.6%的序列同一性,分別與人PH20多肽有63.6%和63.5%的序列同一性。對於牛(家牛)的PH20多肽,Meyer等人(1997)報導了一種可溶性多肽具有終止於H478的C-末端。基於圖4中多重序列分析的結果,可以預期具有終止於H477的C-末端的綿羊(綿羊(Ovis aries))的PH20多肽是具有活性和可溶性的PH20。
本發明透過進一步切割N-末端,即進一步從成熟的天然PH20多肽缺失胺基酸殘基,提供了生產率提高的N-末端和/或C-末端切割的重組PH20多肽。本文使用的術語「N-末端和/或C-末端切割的重組PH20多肽」實質上是指PH20變體。
在具有SEQ ID No: 1的胺基酸序列的野生型PH20中,在選自由N37至P42組成的組的胺基酸殘基之前切割根據本發明的重組PH20多肽,導致一個或多個N-末端胺基酸殘基的缺失。
具體地,根據本發明的重組PH20多肽可以包括在選自由N37至P42組成的組的胺基酸殘基之前被切割的序列,並且起始於SEQ ID No: 1的胺基酸序列的N37、F38、R39、A40、P41或P42。更具體地,根據本發明的重組PH20多肽可以包含起始於SEQ ID No: 1的胺基酸序列的N37或F38的序列。
在選自由N37至P42組成的組的胺基酸殘基之前的切割表示在N-末端處緊接在選自由N37至P42組成的組的胺基酸殘基之前的所有的一個或多個胺基酸殘基都被切割和缺失。
例如,切割發生在胺基酸殘基N37、F38、R39、A40、P41或P42之前的說法是指,在SEQ ID No. 1的胺基酸序列中,緊接在N37之前的殘基1至36、緊接在F38之前的殘基1至37、緊接在R39之前的殘基1至38、緊接在A40之前的殘基1至39、緊接在P41之前的殘基1至40以及緊接在P42之前的殘基1至41分別被切割和除去。
根據本發明的重組PH20多肽可以進一步包括在C-末端處一些胺基酸殘基的缺失。具體地,根據本發明的重組PH20多肽可以在選自由SEQ ID No: 1的胺基酸序列的F468至Y482組成的組的胺基酸殘基之後被切割,導致C-末端處胺基酸殘基的缺失。更具體地,根據本發明的重組PH20多肽可以包含終止於SEQ ID No: 1的胺基酸序列的F468或Y482的序列。
在選自由SEQ ID No. 1的胺基酸序列的F468至Y482組成的組的胺基酸殘基之後的切割,以及C-末端處胺基酸殘基的缺失意味著緊接在選自由F468至Y482組成的組至C-末端的胺基酸殘基之後的所有胺基酸殘基都被切割和缺失。例如,殘基F468或Y482之後的切割意味著SEQ ID No. 1的胺基酸序列的F468或Y482之後的所有殘基都被切割和缺失。

特別地,根據本發明的用於治療癌症的組合物的特徵在於用於與其他抗癌劑的聯合療法。
用於聯合療法的優選抗癌劑包括但不限於化療劑、抗體型抗癌劑、RNAi或細胞療法。
可用于聯合療法的抗癌劑是但不限於免疫抗癌劑,更優選免疫檢查點抑制劑。
在另一方面,本發明涉及一種編碼根據本發明的重組PH20多肽的核酸,所述重組PH20多肽在成熟動物野生型PH20的N-末端和/或C-末端處具有胺基酸殘基的缺失。
本文使用的核酸可以存在於細胞或細胞裂解物中,或者可以以部分純化或基本上純淨的形式存在。當透過標準技術,包括堿/SDS處理、CsCl顯帶(CsCl banding)、柱層析、瓊脂糖凝膠電泳和本領域公知的其他技術,從其他細胞成分或其他污染物諸如來自其他細胞的核酸或蛋白質中純化核酸時,核酸是「分離的」或「基本上純淨的」。本發明的核酸可以是,例如,DNA或RNA。
在另一方面,本發明涉及一種包含核酸的載體,特別是重組表達載體。對於根據本發明的重組PH20重組多肽的表達,編碼PH20重組多肽的DNA可以透過標準分子生物學技術(例如,PCR擴增或使用表達PH20重組多肽的雜交瘤的cDNA選殖)獲得,並且該DNA可以「可操作地連接」到轉錄和轉譯控制序列並插入到表達載體中。

此外,根據本發明的宿主細胞可以來源於植物或哺乳動物。可以優選使用猴腎細胞7 (COS7)、NSO細胞、SP2/0、中國倉鼠卵巢(CHO)細胞、W138、幼倉鼠腎(BHK)細胞、MDCK、骨髓瘤細胞系、HuT 78細胞或HEK293細胞,但不限於此。特別優選地,可以使用CHO細胞。
將核酸或載體轉染到宿主細胞中。對於「轉染」,可以使用通常用於將外源核酸(DNA或RNA)引入原核或真核宿主細胞的許多不同技術,諸如電泳、磷酸鈣沉澱、DEAE-葡聚糖轉染或脂質體轉染。各種表達宿主/載體組合可以用於表達根據本發明的PH20重組多肽。用於真核宿主的合適表達載體包括但不限於來源於SV40、牛乳突瘤病毒、腺病毒、腺相關病毒、巨細胞病毒和反轉錄病毒的表達控制序列。可在細菌宿主中使用的表達載體包括從大腸桿菌獲得的細菌質粒,諸如pET、pRSET、pBluescript、pGEX2T、pUC載體、col E1、pCR1、pBR322、pMB9或其衍生物;宿主範圍更廣的質粒,諸如RP4;噬菌體DNA,其可以由多種多樣的噬菌體λ衍生物作為示例,諸如λgt10和λgt11或NM989;和其他DNA噬菌體,諸如M13或絲狀單鏈DNA噬菌體。對酵母細胞有用的表達載體是2℃質粒及其衍生物。對昆蟲細胞有用的載體是pVL941。
在另一方面,本發明涉及一種生產根據本發明的重組PH20重組多肽的方法,包括培養宿主細胞以表達根據本發明的重組PH20重組多肽的步驟。
當將能夠表達PH20重組多肽的重組表達載體引入哺乳動物宿主細胞中時,可透過將宿主細胞培養足以在宿主細胞中表達的一段時間,或更優選足以允許PH20重組多肽分泌到培養宿主細胞的培養基中的一段時間,來產生PH20重組多肽。
在某些情況下,所表達的PH20重組多肽可以從宿主細胞中分離並純化至均質。PH20重組多肽的分離或純化可以透過任何常用於蛋白質的分離或純化方法(諸如層析)來進行。層析可以是例如但不限於選自親和層析、離子交換層析或疏水層析的一種或多種組合。除了層析之外,過濾、超濾、鹽析或透析可以結合使用。
在下文中,給出實施例以進一步描述本發明。提供以下實施例僅是為了說明本發明,對於本領域具有通常知識者來說,顯然本發明的範圍不限於這些實施例。
實施例1.N-末端和/或C-末端切割的重組PH20透明質酸酶的選殖
對於PH20多肽的表達,透過Genscript (韓國)來合成編碼野生型人PH20多肽的胺基酸L36至S509的序列、編碼來自猴(長鼻猴)的PH20多肽(Uniprot ID: H2DJA7)的胺基酸M1至L510的DNA序列、編碼來自牛(家牛)的PH20多肽(Uniprot ID: F1MTV1)的胺基酸L36至H478的DNA序列、以及編碼來自綿羊(綿羊(Ovis aries))的PH20多肽(Uniprot ID: W5NSU1)的胺基酸L36至H477的DNA序列。
透過聚合酶連鎖反應(下文稱為「PCR」)擴增每個合成的PH20多肽基因,並將其插入pcDNA3.4-TOPO載體的XhoI或NotI限制性酶位點。為了在ExpiCHO細胞中表達,使用了人血清白蛋白訊息肽。對於使用HisTrap柱的蛋白質純化,將His標籤的DNA序列放置於PH20 cDNA的3'末端處。對N-末端和/或C-末端切割的PH20多肽進行PCR,並透過DNA測序證實胺基酸置換。
為了產生N-末端切割的人PH20多肽的變體,製備了含有L36-Y482的質粒,並將其用作範本以產生四種變體,其中胺基酸從N-末端依次一個接一個地被除去,並且對於每種變體,四種變體的C-末端終止於F468。
為了產生N-末端切割的猴(長鼻猴)PH20多肽的變體,製備了含有L36至Y483的質粒,並將其用作範本以產生五種變體,其中胺基酸從N-末端依次一個接一個地被除去。
為了產生N-末端切割的牛(家牛)PH20多肽的變體,製備了含有L36至H478的質粒,並將其用作範本以產生五種變體,其中胺基酸從N-末端依次一個接一個地被除去。
為了產生N-末端切割的綿羊(綿羊(Ovis aries))PH20多肽的變體,製備了含有L36至H477的質粒,並將其用作範本以產生五種變體,其中胺基酸從N-末端依次一個接一個地被除去。
實施例2.N-末端和/或C-末端切割的重組PH20透明質酸酶的表達和純化
透過使用ExpiCHO表達系統進行N-末端和/或C-末端切割的PH20多肽的表達。當ExpiCHO細胞的細胞密度達到6×10
6個細胞/mL時,使用ExpiFectamine CHO試劑將含有插入pcDNA3.4-TOPO載體的N-末端和C-末端切割的PH20多肽的cDNA的質粒轉染到ExpiCHO細胞中。作為細胞培養基,使用ExpiCHO表達培養基(100至500 mL)。轉染後,將ExpiCHO細胞以130 rpm振盪並培養總共6天,在此期間,將細胞在37℃下溫育1天,並進一步在32℃的較低溫度下溫育5天。溫育完成後,透過以10,000 rpm離心30分鐘收集細胞上清液。
使用AKTA prime設備或類似設備(GE Healthcare)透過兩步柱層析純化從ExpiCHO細胞產生的在C末端處具有His標籤的PH20多肽。對於人PH20和猴PH20,使用Q瓊脂糖凝膠(其是陰離子交換層析),因為pI接近6。對於牛或綿羊PH20多肽,pI大於8,因此使用Capto S柱(其是陽離子交換層析)進行第一次純化,使用HisTrap HP柱(其是His標籤親和層析)進行每種蛋白質的第二次純化。
為了透過使用Q瓊脂糖凝膠柱純化蛋白質,製備了緩衝液A (20 mM磷酸鈉,pH 7.5)和緩衝液B (20 mM磷酸鈉,pH 7.5,0.5 M NaCl)。將培養基的pH和電導率調節至與緩衝液A相同,並將培養基透過孔徑為0.22 μm的膜過濾。將蛋白質結合到Q瓊脂糖凝膠柱上,使5CV的緩衝液A流動以除去非特異性結合的蛋白質,並使5CV的緩衝液B以0至100%的濃度梯度流動,以便洗脫蛋白質。
為了使用Capto S柱純化蛋白質,製備了緩衝液A (20 mM磷酸鈉,15 mM NaCl,pH 6.0)和緩衝液B (20 mM磷酸鈉,500 mM NaCl,pH 6.0)。將培養基的pH和電導率調節至與緩衝液A相同,並將培養基透過孔徑為0.22 μm的膜過濾。然後將蛋白質結合到Capto S柱上,使3倍柱體積(CV)的緩衝液A流過以除去非特異性結合的蛋白質。使4CV的緩衝液B順序流動以洗脫靶蛋白質。
為了使用HisTrap HP柱純化蛋白質,製備了緩衝液A (20 mM磷酸鈉,500 mM NaCl,pH 7.5)和緩衝液B (20 mM磷酸鈉,500 mM NaCl,500 mM咪唑,pH 7.5)。在蛋白質樣品與HisTrap HP柱結合後,7 CV的7%緩衝液B流動以除去非特異性結合的蛋白質,並且使3 CV的40%緩衝液B流動以洗脫靶蛋白質。使用透析緩衝液(20 mM磷酸鈉,100 mM NaCl,pH 7.0)透析柱洗脫液。
實施例3.N-末端和/或C-末端切割的重組PH20透明質酸酶的N-末端測序
純化的N-末端和/或C-末端切割的PH20多肽以每7.5% SDS-PAGE凝膠的泳道10 μg的量上樣,並進行電泳(150 V,1小時)。然後將含有經電泳展開的蛋白質的凝膠引入具有PVDF膜的印跡試劑盒中,並在100V下轉移90分鐘,透過麗春紅S染色確認轉移。最後,使用PPSQ-53A蛋白質測序儀(Shimadzu,日本)對透過切取每個蛋白質條帶獲得的樣品從N-末端開始的5個胺基酸進行測序。
作為N-末端測序的結果,證實了其他PH20多肽具有預期的序列,但是在一些具有N37-Y482或N37-F468的人PH20多肽中,發現N-末端處的胺基酸是天冬胺酸而不是天冬醯胺,導致了去醯胺化。基於這一結果和圖4的多重序列分析,表明天冬胺酸被發現作為牛和綿羊中相應的胺基酸,可以推斷天冬胺酸作為該位置處的胺基酸在穩定蛋白質結構中起作用。
實施例4.重組N-末端和/或C-末端切割的PH20透明質酸酶活性的測量
透明質酸酶的活性透過濁度測定法來測量,濁度是由透明質酸與白蛋白(BSA)混合時產生的沉澱引起的,其是透過吸光度來測量的。當透明質酸被PH20多肽水解時,透過混合透明質酸和白蛋白產生的沉澱的濁度/吸光度降低。該測定通常在pH 5.3下進行,如下所述。將具有已知活性(單位)的透明質酸酶標準品稀釋至6、8、10、12、15和20單位/mL,並在每個試管中製備。透過調節稀釋因數使其符合標準曲線的範圍,在緩衝液(20 mM磷酸鈉,pH 5.3,77 mM氯化鈉和0.01% (w/v)牛血清白蛋白)中稀釋經純化的蛋白質樣品。將50 μl的經稀釋的樣品分配到96孔盤的每個孔中,並在37℃下經受加熱反應10 min。將50 μl的0.06%透明質酸另外分配到每個孔中。0.06%透明質酸處於溶解在300 mM的pH為5.3的磷酸鈉緩衝液中的狀態。將樣品和0.06%透明質酸在37℃下反應45分鐘。反應後,將40 μl的酶-底物反應溶液分配到200 μl的酸性白蛋白溶液中,並在室溫下靜置19 min。然後使用分光光度計在600 nm處測量吸光度。酸性白蛋白溶液由溶解在pH為3.75的緩衝液中的0.1%白蛋白(BSA)組成,該緩衝液包含24 mM的乙酸鈉和79 mM的乙酸。使用活性標準品,透過標準曲線將經測量樣品的吸光度值轉化為活性。
當在pH 7.0下進行上述步驟時,蛋白質樣品緩衝液是pH為7.0的20 mM的磷酸鈉、77 mM的氯化鈉和0.01% (w/v)牛血清白蛋白,並且對於0.06%透明質酸水溶液是將透明質酸溶解於pH為7.0的20 mM磷酸鈉緩衝液和70 mM的氯化鈉中。
這些活性測量亦可以在培養基中進行。在這種情況下,由於低於300單位/mL的值是不可靠的,所以將定量極限(LOQ)設置為300單位/mL,低於該值的值被標記為無效,即0。此外,在使用經純化的蛋白質樣品進行活性測量的情況下,將定量極限(LOQ)設定為15單位/µg。
除了SEQ ID No. 1的野生型PH20多肽的M1至T35的訊息序列之外,將N-末端調整為L36、N37、F38、R39或A40,並選擇C-末端為F468或Y482,透過實施例1的方法產生總共10種N-末端和/或C-末端切割的PH20多肽。在透過動物細胞的暫態轉染培養獲得的培養基中純化這些選殖中的每一個,並透過實施例2的方法比較pH 5.3和pH 7.0下的酶活性。
如圖1所示,從比較每個培養基中人PH20活性的結果來看,令人驚訝的是,培養基中N37-Y482的活性比具有野生型N-末端的成熟L36-Y482的培養基中的活性高3倍以上,並且培養基中其中N-末端另外缺失兩個胺基酸殘基的F38-Y482的人PH20活性亦比具有野生型N-末端的成熟L36-Y482的培養物中的活性高2倍以上。
然而,缺少三個N-末端胺基酸殘基的R39-Y482導致酶的幾乎不表達,並且缺少四個N-末端胺基酸殘基的A40-Y382亦導致培養物中酶的幾乎不表達。
總之,對於人透明質酸酶PH20,發現在成熟人透明質酸酶PH20中缺少一個或兩個N-末端胺基酸殘基的變體在培養基中表現出顯著增加的表達。特別地,考慮到用於注射的人透明質酸酶PH20的工業可用性,表明透過生產其中一個或兩個N-末端胺基酸被切割的變體酶,可以獲得工業上和經濟上優異的變體。
此外,如圖2和圖3所示,L36-Y482、N37-Y482和F38-Y482的比活性之間幾乎沒有差異,證實了圖1中人透明質酸酶PH20活性的差異是因為生產率的差異。此外,在具有比其C-末端終止於Y482的蛋白質短14個C-末端胺基酸的PH20變體中,只有缺失一個N-末端胺基酸的N37-F468在培養基中有活性。
特別地,圖2示出了每種經純化的多肽在pH 5.3下的比活性,證實了當C-末端是Y482時L36-Y482、N37-Y482和F38-Y482之間幾乎沒有差異,但當C-末端為F468時,N37-F468和F38-F468具有比L36-F468高1.5至2倍的比活性。在pH 7.0下的比活性顯示出相同的模式。
也就是說,基於圖2和圖3中所示的結果,商業上使用的PH20的較短變體(L36-F468,其中C-末端胺基酸被進一步切割至第468位的末端)在動物細胞中幾乎不表達。作為純化其中表達的非常少量的L36-F468變體並測量其比活性的結果,酶的比活性與L36-Y482相比大約為60%。基於這些發現,當從成熟的人透明質酸酶PH20缺失一個或兩個額外的N-末端胺基酸殘基時,PH20多肽的透明質酸蛋白酶活性或生產率顯著增加。
總之,儘管由L36-F468組成的人透明質酸酶PH20變體酶的比活性與L36-Y482 PH20變體相比沒有顯著降低,但其在重組細胞中的表達極低,使其不適合工業使用。
另一方面,對於N37-F468變體,在細胞中的表達顯著增加,這與L36-Y482變體的表達類似,但是N37-F468(其進一步缺失一個N-末端胺基酸)在細胞中的表達,與L36-F468相比,顯著增加了至少100倍。
鑒於PH20的分子模型和本發明的實驗結果,在重組PH20蛋白在細胞中表達時的轉錄和易位期間PH20的N-末端處的胺基酸和C-末端處的胺基酸之間的相互作用影響穩定性、折疊速率等,此外,極大地影響從細胞分泌的重組蛋白的表達。這顯著影響了PH20及其變體的生產率。這些結果不僅可以應用於人PH20,還可以應用于哺乳動物來源的PH20,諸如牛或綿羊,並且進一步,預期可以應用於具有與人PH20相似的結構或物理化學特性的其他PH20。
此外,發現在哺乳動物PH20中保持了這些特性,所述哺乳動物PH20具有與人PH20多肽相似的序列結構和相同的生物功能,特別是表1中列出的猴(長鼻猴)、牛(家牛)和綿羊(綿羊(Ovis aries)) PH20,如圖5和圖6所示。特別是,當N-末端起始于A40時,發現綿羊PH20多肽會高度地表達。
總之,本發明的結果表明,對於成熟的PH20蛋白,特別是動物PH20諸如人PH20,除去N-末端處額外的一至四個,優選一或兩個胺基酸殘基可以顯著增加重組細胞中的表達率,從而增加蛋白生產率。
對於人PH20,當C-末端終止于Y482時,從成熟PH20中除去了一些額外的N-末端胺基酸殘基的PH20顯示出顯著增加的表達,而透明質酸酶的內在活性沒有顯著變化,當C-末端終止于F468時,與具有完整N-末端的成熟蛋白質相比,從成熟PH20中除去了一個或兩個額外的N-末端胺基酸殘基的變體反而顯示出增加的酶的比活性。雖然已經嘗試透過在人PH20的胺基酸序列的特定位點置換不同的胺基酸來增加蛋白質的比活性,但是還沒有報導透過除去N-末端的胺基酸同時保持或增加其活性來顯著增加其在細胞中的表達的情況。據預測,經缺失N-末端的變體在免疫原性方面優於其中一個或多個胺基酸在一個或多個特定位點被替換的PH20,甚至在人體內長期給藥後亦是如此。
已經有報導表明了當成熟蛋白的N-末端被切割時表達增加,諸如關於人α1-抗胰蛋白酶(H. Johansen等人, Mol. Biol. Med. 1987, 4:291-305)和人乳突瘤病毒L1蛋白(M. Wei等人, Emerging Microbes & Infections, 2018, 7:160)的研究,但是很難在N-末端位點的胺基酸位置、截短數量等方面找到彼此的相似性。例如,在人α1-抗胰蛋白酶中,5至10個N-末端胺基酸的缺失增加了細胞中的表達,但沒有改變酶的活性,而在人乳突瘤病毒L1蛋白中,10或15個N-末端胺基酸的切割沒有改變蛋白的整體表達,但在某些情況下增加了蛋白的溶解性。特別是,對於透明質酸酶諸如PH20,還沒有進行過這樣的嘗試,亦沒有發現像本發明中那樣切割一個或兩個N-末端胺基酸顯著增加表達和活性的情況。
對人PH20的三維晶體結構尚不清楚。然而,根據使用程式對PH20的三維結構的預測,預測儘管胺基酸F38、R39和Y434位於PH20的N-末端處第37位的天冬醯胺側鏈附近,並且在它們之間具有適當的電荷相互作用,但是成熟蛋白質中的胺基酸Leu36不與周圍的胺基酸形成任何特定的鍵,並且疏水性胺基酸暴露於水溶液,從而不利地影響結合速度或穩定性,進而影響蛋白質的表達。
在成熟的野生型動物PH20蛋白質中,具有不會顯著改變蛋白質三維結構的胺基酸置換的變體可以顯示與本發明相同的結果。由於在分別與人PH20具有63.6%和63.5%同一性的牛和綿羊PH20中,以及與人PH20具有超過93.1%同一性的靈長類PH20中亦證實了其效果,對於與人PH20具有60%、70%、80%、90%、91%、92%、93%、94%、95%、96%、97%、98%或99%同一性的PH20,在蛋白質的三維結構沒有被顯著修飾的範圍內,可以預測相同的結果。
實施例5.具有不同訊息肽的N-末端和/或C-末端切割的重組PH20透明質酸酶的製備以及活性的測量
製備了九種用人PH20、人血清激素或Igκ的單一肽替換的變體,以鑒定訊息肽的變化。訊息肽的序列列於下表2。
在前面的實施例中,訊息肽來自人血清白蛋白(HSA),並且測試了表2中列出的其他訊息肽是否亦獲得了相同的結果。
除了應用表2中的訊息肽之外,如實施例1和2中那樣製備重組人PH20的L36-Y482、N37-Y482和F38-Y482,並如實施例4中那樣進行活性測定,結果示於圖7中。
如圖7所示,即使當使用人PH20或Igκ的訊息肽時,依賴於成熟PH20的N末端缺失的生產率變化傾向於與使用人血清激素的訊息肽的情況相似。在這點上,證實了具有在N-末端胺基酸進一步的缺失的成熟PH20中表達和生產率的增加與訊息肽的類型關係不大。
與常規重組PH20多肽相比,本文所述的具有改善的酶活性和生產率的N-末端和/或C-末端切割的PH20多肽表現出優異的表達和更高的酶活性,這可使得治療成本的降低,因為工業使用的生產成本更低。
前面已經詳細描述了本發明的某些方面。對於本領域具有通常知識者來說,顯然這些特定的實施例僅僅是優選的實施方式,並不意圖限制本發明的範圍。因此,本發明的實質範圍由所附請求項及其等同物限定。

Claims (13)
- 一種重組PH20多肽,其中,成熟動物野生型PH20的N-末端處缺失1至7個胺基酸殘基。
- 如請求項1所述的重組PH20多肽,其特徵在於,選自由以下組成的組:(a)重組PH20多肽,其中在具有SEQ ID No. 1的胺基酸序列的人野生型PH20中,在選自由N37至P42組成的組的胺基酸殘基之前,透過切割缺失一個或多個N-末端胺基酸殘基;(b)重組PH20多肽,其中在具有SEQ ID No. 2至8中任一個的胺基酸序列的靈長類野生型PH20中,在選自由N37至P42組成的組的胺基酸殘基之前,透過切割缺失一個或多個N-末端胺基酸殘基;(c)重組PH20多肽,其中在具有SEQ ID No. 9的胺基酸序列的牛野生型PH20中,在選自由D37至P42組成的組的胺基酸殘基之前,透過切割缺失一個或多個N-末端胺基酸殘基;以及(d)重組PH20多肽,其中在具有SEQ ID No. 10的胺基酸序列的綿羊野生型PH20中,在選自由D37至P42組成的組的胺基酸殘基之前,透過切割缺失一個或多個N-末端胺基酸殘基。

- 一種編碼如請求項1至8中任一項所述的重組PH20多肽的核酸。
- 一種包含如請求項9所述的核酸的重組表達載體。
- 一種用如請求項10所述的重組表達載體轉染的宿主細胞。
