Abstract

Description
本發明屬於生物醫藥領域,具體地說,本發明涉及抗BCMA抗體偶聯物及其醫藥用途。
B細胞是淋巴細胞的一種,其在自適應體液免疫和特異性識別抗原的抗體的產生中發揮重要的作用。B細胞具有三種亞類,分別是幼稚B細胞、記憶B細胞和漿細胞。VDJ重組過程中,DNA的兩種或三種片段選自基因組文庫,並且重組可產生抗體可變結構域的組合陣列,藉由由不同譜系的B細胞編碼的可變結構域的進一步改變,可產生至多109種獨特的B細胞譜系,各譜系產生具有獨特靶特異性的抗體。多種疾病涉及B細胞,B細胞的惡性轉化導致癌症,包括一些淋巴瘤,如多發性骨髓瘤和霍奇金氏淋巴瘤。自身免疫性疾病也會涉及到B細胞,包括系統性紅斑狼瘡(SLE)和IgA腎病。涉及B細胞的癌症和自家免疫性疾病可被認為B細胞功能異常,因此控制這類疾病的可能的策略是使用靶向病理B細胞的抗體。
BCMA是TNF受體超家族的成員,其是對配體BAFF(B細胞激活因子)和APRIL(增殖誘導配體)的非糖基化的內在膜受體。BCMA和它的
相應配體具有調節體液免疫、B細胞發育和穩態等不同作用。BCMA由扁桃體記憶B細胞和由生髮中心B細胞表達,在脾臟、淋巴結、胸腺、腎上腺和肝臟均可以檢測到,並且多種B細胞系的分析表明在BCMA在成熟後的表達水平增加。

在本發明一個較佳的方案中,該BCMA介導的疾病或病症選自癌症或自身免疫疾病,其中該癌症較佳為表達BCMA的癌症,更佳淋巴瘤、白血病或骨髓瘤;該自身免疫疾病選自紅斑狼瘡、IgA腎病或風濕性關節炎。
本發明的抗體藥物偶聯物及其可藥用鹽或溶劑化合物具有顯著的抗腫瘤效果和良好的安全性,同是具有良好的體內代謝活性,體內藥效時間長,臨床應用前景廣闊。
[發明詳述]
一、術語
為了更容易理解本申請,以下具體定義了某些技術和科學術語。除顯而易見在本文件中的它處另有明確定義,否則本文使用的所有其它技術和科學術語都具有本申請所屬技術領域具有通常知識者通常理解的含義。
本申請所用胺基酸三字母代碼和單字母代碼如J.Biol.Chem,243,p3558(1968)中所述。
本申請所述的術語“抗體”指免疫球蛋白,是由兩條相同的重鏈和兩條相同的輕鏈藉由鏈間二硫鍵連接而成的四肽鏈結構。免疫球蛋白重鏈恆定區的胺基酸組成和排列順序不同,故其抗原性也不同。據此,可將免疫球蛋白分為五類,或稱為免疫球蛋白的同種型,即IgM、IgD、IgG、IgA和IgE,其相應的重鏈分別為μ鏈、δ鏈、γ鏈、α鏈和ε鏈。同一類Ig根據其鉸鏈區胺基酸組成和重鏈二硫鍵的數目和位置的差別,又可分為不同的亞類,如IgG可分為IgG1、IgG2、IgG3、IgG4。輕鏈藉由恆定區的不同分為κ鏈或λ鏈。五類Ig中第每類Ig都可以有κ鏈或λ鏈。
在本申請中,本申請所述的抗體輕鏈可變區可進一步包含輕鏈恆定區,該輕鏈恆定區包含人源或鼠源的κ、λ鏈或其變體。

術語“抗原呈遞細胞”或“APC”是在其表面上展示與MHC複合的外來抗原的細胞。T細胞利用T細胞受體(TCR)識別這種複合物。APC的實例包括但不限於樹突細胞(DC)、外用血單個核細胞(PBMC)、單核細胞、B淋巴母細胞和單核細胞衍生的樹突細胞。
術語“抗原呈遞”是指APC捕獲抗原和使它們能夠被T細胞識別的過程,例如作為MHC-I/MHC-II偶聯物的組分。
術語“BCMA”包括由細胞天然表達的BCMA的任何變體或同種型。本申請的抗體可與得自非人物種的BCMA交叉反應。作為另一種選擇,該抗體也可以是人BCMA特異性的,可不表現出與其他物種的交叉反應性。BCMA或其任何變體或同種型可從天然表達它們的細胞或組織中分離而得,或使用本領域通用以及本文所述的那些技術藉由重組技術產生。較佳地,抗BCMA抗體靶向具有正常糖基化模式的人源BCMA。

術語“嵌合抗體(chimeric antibody)”,是將鼠源性抗體的可變區與人抗體的恆定區融合而成的抗體,可以減輕鼠源性抗體誘發的免疫應答反應。建立嵌合抗體,要選建立分泌鼠源性特異性單抗的融合瘤,然後從小鼠融合瘤細胞中選殖可變區基因,再要據需要選殖人抗體的恆定區基因,將小鼠可變區基因
與人恆定區基因連接成嵌合基因後插入人載體中,最後在真核工業系統或原核工業系統中表達嵌合抗體分子。人抗體的恆定區可選自人源IgG1、IgG2、IgG3或IgG4或其變體的重鏈恆定區,較佳包含人源IgG1、IgG2或IgG4重鏈恆定區,或者使用胺基酸突變後增強ADCC(antibody-dependent cell-mediated cytotoxicity,抗體依賴的細胞介導的細胞毒作用)毒性的IgG1重鏈恆定區。
術語“抗原結合片段”是指抗體的抗原結合片段及抗體類似物,其通常包括至少部分母體抗體(parental antibody)的抗原結合區或可變區(例如一個或多個CDR)。抗體片段保留母體抗體的至少某些結合特異性。通常,當基於莫耳來表示活性時,抗體片段保留至少10%的母體結合活性。較佳地,抗體片段保留至少20%、50%、70%、80%、90%、95%或100%或更多的母體抗體對靶標的結合親和力。抗原結合片段實例包括但不限於:Fab、Fab’、F(ab’)2、Fv片段、線性抗體(linear antibody)、單鏈抗體、奈米抗體、結構域抗體和多特異性抗體。工程改造的抗體變體綜述於Holliger和Hudson,2005,Nat.Biotechnol.23:1126-1136中。
“Fab片段”由一條輕鏈和一條重鏈的CH1及可變區組成。Fab分子的重鏈不能與另一個重鏈分子形成二硫鍵。
“Fc”區含有包含抗體的CH2和CH3結構域的兩個重鏈片段。兩個重鏈片段由兩個或多個二硫鍵並藉由CH3結構域的疏水作用保持在一起。

術語“單鏈抗體”是由抗體的重鏈可變區VH和輕鏈可變區VL藉由一段連接肽連接而成的單鏈重組蛋白,它是具有完全抗原結合位點的最小抗體片段。
術語“結構域抗體片段”是僅含有重鏈可變區或輕鏈可變區鏈的具有免疫學功能的免疫球蛋白片段。在某些情況下,兩個或多個VH區與肽接頭共價連接以形成二價結構域抗體片段。二價結構域抗體片段的兩個VH區可靶向相同或不同抗原。
本申請的術語“與BCMA結合”,指能與人BCMA相互作用。
本申請的術語“抗原結合位點”指本申請抗體或抗原結合片段識別的三維空間位點。

本申請所用的術語“特異性結合”、“選擇性結合”是指抗體與預定的抗原上的表位結合。通常,當使用人BCMA作為分析物並使用抗體作為配體,在儀器中藉由表面電漿共振(SPR)技術測定時,抗體以大約低於10-7M或甚至更小的平衡解離常數(KD)與預定的抗原結合,並且其與預定抗原結合的親和力是其與預定抗原或緊密相關的抗原之外的非特異性抗原(如BSA等)結合的親和力的至少兩倍。術語“識別抗原的抗體”在本文中可以與術語“特異性結合的抗體”互換使用。

本申請工程化的抗體或抗原結合片段可用常規方法製備和純化。相應抗體的cDNA序列可以選殖並重組至GS表達載體。重組的免疫球蛋白表達載體可以穩定地轉染CHO細胞。作為一種更推薦的現有技術,哺乳動物類表達

“細胞毒性藥物”在用於本發明時指抑制細胞的功能和/或引起細胞死亡或破壞的物質。

7、8、9、10的均值。可用常規方法如UV/可見光光譜法、質譜、ELISA試驗和HPLC特徵鑑定偶聯反應後每個ADC分子的藥物品均數量。
本發明的一些實施方式中,細胞毒性藥物藉由連接單元偶聯在配體的N端胺基和/或賴胺酸殘基的ε-胺基上,在本發明的另一些實施方式中,細胞毒性藥物藉由連接單元偶聯在配體的巰基上。一般地,偶聯反應中能與抗體偶聯的藥物分子數將小於理論上的最大值。
可以用以下非限制性方法控制配體細胞毒性藥物偶聯物的載量,包括:

實施例1-1化合物a的合成
取原料a-1(4.1g,9.71mmol)、原料a-2(4.3g,8.09mmol,含4%胺基異構體雜質)置於250mL反應瓶中,氮氣保護下加入DCM(54mL),MeOH(18mL)攪拌並冷至0℃,加入DMTMM(3.6g,12.1mmol),三乙胺(2.5g,
24.2mmol),並保持0℃攪拌反應1h,HPLC中控顯原料原料a-2反應完全,減壓(<25℃)蒸乾反應液,加入MTBE(120mL)攪拌打漿(泥狀物),倒出溶液過濾,泥狀物再次加入120mL MTBE打漿(固體),過濾,濾餅用水(60mL×2)洗,晾乾得粗品,粗品用二氯甲烷和甲醇溶解,濕法上樣管柱層析兩次(沖提劑DCM:MeOH=40:1-20:1)分離得到化合物a純品(6.2g,7.37mmol),純度:99.3%,收率91%
實施例1-2化合物b的合成
取化合物a(5.7g,6.77mmol)置於500mL三口瓶中,氮氣保護下加入乾燥的THF(114mL),攪拌溶解並冷卻內溫至-10℃左右,加入DBU(3.09g,20.31mmol),滴加過程中保持內溫-10至-5℃,5min加畢,滴畢後,保持內溫-10至-5℃,反應2.5h,固體析出。
冷卻內溫至-20℃,加入MTBE(114mL),期間保持內溫-20至-10℃,產物完全析出,過濾,濾餅用MTBE(57mL×2)洗,抽乾後得到化合物b粗品6g,-78℃保存待用。
實施例1-3化合物e的合成
將化合物c(651mg,1mmol)溶於10mL的DCM裡,冰浴冷卻下開啟攪拌,滴入DBU(456mg,3mmol)。冰浴下反應一個小時後反應完成,依次加入化合物d(257mg,1mmol)和HATU(420mg,1.1mmol),在冰浴下攪拌30分鐘後,LCMS顯示反應完成,反應液在25℃條件下濃縮,殘留物用過管柱機反相管柱純化(ACN in H2O,50%出產物)得到化合物e,紅棕色固體,130mg,收率19%。
MS:691.3[M+23]。
實施例1-4化合物f的合成
將化合物e(130mg,0.19mmol)溶於DCM,加入苯甲醚(62mg,0.57mmol)和二氯乙酸(245mg,1.9mmol),反應在室溫下攪拌過夜,共16小時,取樣LC-MS中控,顯示原料完全消耗,停止反應,將反應液在25℃條件下濃縮,殘留物用過管柱機反相管柱純化(ACN/H2O,30%出產物),得化合物f,粉色固體,53mg,收率54%。
MS:519.2[M+1]。
實施例1-5化合物D的合成
將化合物f(23mg,0.044mmol)和化合物b(27mg,0.044mmol)溶於DCM(3mL)和MeOH(1mL),在氮氣保護下冷卻到-30℃。加入DMTMM(20mg,0.067mmol),反應在-20℃~-10℃控溫反應1小時,取樣LC-MS中控,顯示原料完全消耗。控制溫度在-10℃,加入10mL水淬滅反應,加入30mL的DCM分層。水相用DCM/MeOH=10/1(50mL)萃取。合併有機相,用無水硫酸鈉乾燥,在25℃條件下減壓濃縮,殘留物經製備純化(ACN/H2O/0.05%FA),得化合物D,白色固體,5.8mg,收率12%,HPLC純度98.87%。
MS:1120.3[M+1]。
實施例2:抗原準備
編碼帶His標簽的人BCMA(BCMA-His)蛋白由SinoBiologics公司合成(Cat No.:Cat No.:10620-H08H)。

實施例3:鼠融合瘤及抗體序列的獲得
用人抗原BCMA-His進行動物免疫,共5隻Balb/c和5隻A/J小鼠,雌性,10週齡,使用Sigma完全弗氏佐劑(CFA)和Sigma不完全弗氏佐劑(IFA),免疫原和免疫佐劑以1:1的比例充分混合乳化,製成穩定“油包水”液體;注射劑量25μg/200μL/小鼠。
對免疫小鼠血清使用如實施例3該間接ELISA法評估血清效價及結合細胞表面抗原的能力,對照效價檢測情況(大於10萬倍稀釋度)決定啟動細胞融合。選擇血清效價、親和力和FACS結合強的免疫小鼠進行一次終免疫
後處死小鼠,取脾細胞和SP2/0骨髓瘤細胞融合後鋪板獲得融合瘤,藉由間接ELISA篩選到目標融合瘤,並藉由有限稀釋法建株為單株細胞株。得到的陽性抗體株進一步使用間接ELISA進行篩選,從而選定結合重組蛋白的融合瘤。收集對數生長期融合瘤細胞,用Trizol(Invitrogen,15596-018)提取RNA並反轉錄(PrimeScriptTM Reverse Transcriptase,Takara #2680A)。將反轉錄得到的cDNA採用mouse Ig-Primer Set(Novagen,TB326 Rev.B 0503)進行PCR擴增後測序,最終得到鼠源抗體M1的序列。

實施例4:抗體的體外結合活性檢測方法
(1)體外間接ELISA結合實驗:
用pH7.4的PBS將BCMA His蛋白(Sino Biological Inc.,cat# 10428-H08H)稀釋至1μg/ml濃度,以100μl/孔的體積加入96孔高親和力酶標板中,於4℃冰箱孵育過夜(16-20小時)。用PBST(pH7.4 PBS含0.05%Tween-20)洗板4次後,加入用PBST稀釋的3%牛血清白蛋白(BSA)封閉液150μl/孔,室溫孵育1小時進行封閉。封閉結束後,棄去封閉液,並用PBST緩衝液洗板4次。
用含3%BSA的PBST稀釋待測抗體,1μM起始,10倍梯度,10個劑量,以100μl/孔加到酶標板中,放於室溫孵育1小時。孵育結束後用PBST洗板4次,加入100μl/孔用含3%BSA的PBST稀釋的HRP標記羊抗人二抗(Abeam,cat#ab97225),室溫孵育1小時。用PBST洗板4次後,加入100μl/孔TMB顯色受質(Cell Signaling Technology,cat#7004S),於室溫避光孵育1分鐘,加入100μl/孔Stop Solution(Cell Signaling Technology,cat#7002S)終止
反應,用酶標儀(BioTek,型號Synergy H1)在450nm處讀取吸收值,分析數據。做濃度信號值曲線分析結果,如下表所示:
(2)體外細胞結合實驗:
收集培養好的BCMA高表達細胞(過表達BCMA的HEK-293T細胞和表達BCMA的腫瘤細胞,NCI-H929),調節細胞密度後分鋪於96孔U底板,每孔1×105至2×105個細胞。1200g,5min離心,去上清,添加100ul已梯度稀釋的抗體溶液或小鼠免疫血清,4℃度孵育60min;1200g,5min離心,去上清,PBS洗細胞2次後,添加螢光標記二抗(PE標記山羊抗人單抗或PE標記山羊抗鼠單抗)每孔100uL,4℃孵育60min。1200g,5min離心去上清。PBS洗細胞2次後,再重新懸浮於PBS,使用流式細胞計數儀檢測信號,並作濃度曲線分析結果。
實施例5:小鼠抗體人源化實驗
鼠源抗人BCMA單株抗體人源化如本領域許多文獻公示的方法進行。簡言之,使用人恆定結構域替代親本(鼠源抗體)恆定結構域,根據鼠源抗體和人抗體的同源性選擇人種抗體序列,本申請將鼠源抗體M1進行人源化。
在所獲得的鼠源抗體VH/VL CDR典型結構的基礎上,將重、輕鏈可變區序列與人源抗體種系數據庫比較,獲得同源性高的人種系模板。
將鼠源抗體M1的CDR區移植到選擇好的相應人源化模板上。然後,以鼠源抗體的三維結構為基礎,對包埋殘基、與CDR區有直接相互作用的殘基,以及對VL和VH的構象有重要影響的殘基進行回復突變,並對CDR區化學不穩定胺基酸殘基優化,經表達測試和回復突變數量對比,選擇出設計了人源化重鏈可變區HCVR的序列,序列如下:

根據以上各人源化抗體輕鏈和重鏈的胺基酸序列合成cDNA片段,插入到pcDNA3.1表達載體(Life Technologies Cat.No.V790-20)中。將表達載體和轉染試劑PEI(Polysciences,Inc.Cat.No.23966)以1:2的比例轉染HEK293細胞(Life Technologies Cat.No.11625019),並置於CO2孵育箱中孵育4-5天。收取細胞培養液,離心過濾後上樣到抗體純化親和管柱,經磷酸緩衝液洗管柱、甘胺酸鹽酸緩衝液(pH2.7 0.1M Gly-HCl)沖提、1M Tris鹽酸pH 9.0中和、以及磷酸緩衝液透析,得到本申請的人源化抗體蛋白。
實施例6:體外結合親和力和動力學實驗
使用實施例4(1)體外間接ELISA結合實驗測定的各人源化抗體對人BCMA抗原的親和力(EC50)如下表所示:
使用實施例4(2)體外細胞結合實驗測定的各人源化抗體對NCI-H929腫瘤細胞的親和力(EC50)如下表所示:
實施例7:抗體的內吞作用
檢測本發明抗體結合BCMA後是否能夠和人BCMA共同內吞入細胞內,用NCI-H929進行評估。NCI-H929細胞使用胰酶消化(先用PBS清洗一遍,37℃、2min左右),收集細胞並用預冷的FACS緩衝液重新懸浮,調整細胞濃度為1×106/mL。取EP管,加入1mL細胞懸液,1500rpm離心5分鐘後去上清,加入1mL已經配製好的待測抗體重新懸浮細胞,抗體的終濃度均為20μg/ml,4度搖床孵育1h,離心棄上清(4℃、1500rpm×5min),FACS緩衝液洗滌兩次,去上
清。每管加入100μL螢光二抗工作液重新懸浮細胞,4℃搖床孵育30min,離心棄上清(4℃、1500rpm×5min),FACS緩衝液洗滌兩次,去上清。每管加入1.0mL預熱的NCI-H929細胞完全培養基重新懸浮細胞並混勻,分裝為4管,每管200μL,分別為0min組,blank組,30min組和2h組,取出0min及blank置於冰上,其餘放置於37℃培養箱,分別內吞30min、2h,在相應時間點取出EP管,置於冰上預冷5min,所有處理組離心棄上清(4℃、1500rpm×5min),用FACS緩衝液洗滌一次,去上清。去除0min組外所有處理組EP管中加入250μL strip緩衝液,室溫孵育8min,離心棄上清(4℃、1500rpm×5min),FACS緩衝液洗滌兩次,去上清。所有處理組加入100μL免疫染色固定液,4℃放置30min以上,用流式細胞儀DxFlex進行檢測。BCMA抗體內吞百分比=(各個時間點螢光強度值-零點時的平均螢光強度值)/零點時的平均螢光輕度值。結果見下表:
實施例8:抗體偶聯藥物的製備
ADC2的製備
在37℃條件下,向抗體Ab2的PBS緩衝水溶液(pH=6.5的0.05M的PBS緩衝水溶液;7.3ml,13.8mg/ml,0.681μmol)加入配置好的三(2-羧乙基)膦的水溶液(10mM,0.347mL,3.47μmol),置於水浴振盪器,於37℃振盪反應3小時,停止反應;將反應液用水浴降溫至25℃,稀釋至14.0ml,並取出3.3ml溶液往下反應。
將化合物D(3.0mg,3.72μmol)溶解於0.15mL DMSO中,加入到上述3.3ml溶液中,置於水浴振盪器,於25℃下振盪反應3小時,停止反應。將反應液用Sephadex G25凝膠管柱脫鹽純化(沖提相:pH為6.5的0.05M的PBS緩衝水溶液,含0.001M的EDTA),得到Ab2抗體偶聯物的示例性產物ADC2的PBS緩衝液(1.35mg/mL,13mL),於4℃冷凍儲液。
採用紫外法測定平均值y。將裝有琥珀酸鈉緩衝液的比色皿分別置於參比吸收池和樣品測定吸收池中後,扣除溶劑空白後,再將裝有供試品溶液的比色皿置於樣品測定吸收池中,測定280nm和370nm處吸光度。
數據處理:
藉由建立標準曲線,測定280nm波長下的吸收,確定抗體含量CAb,測定370nm波長下的吸收,確定小分子含量CDrug。
藥物載量平均值y=CDrug/CAb。
藉由以上方法測定示例性產物ADC2的y值為4。藉由UV-HPLC純化獲得ADC2(y=4)樣品。
實施例9抗體藥物偶聯物的細胞殺傷活性
為檢測本公開的抗體-藥物偶聯物對腫瘤細胞的殺傷作用,採用BCMA高表達水平細胞株NCI-H929(ATCC保藏號CRL-9068)進行評估。收集NCI-H929細胞,離心計數後用完全培養基調整細胞密度為1×105個/mL,鋪於白色96孔板中間60個孔,每孔100μL,細胞數為10000細胞/孔,邊緣孔加入100μL/孔DPBS,細胞板放入37℃,5%CO2培養箱培養過夜。次日,用完全培養基在96孔V型底板中配製抗體-藥物偶聯物工作溶液,133uM起始,5倍稀釋,9個濃度,配製完成後加入到白色96孔板中,每孔80μL,兩複孔,將細胞板放入37℃,5% CO2培養箱中繼續培養72小時。實驗第五天,檢測讀數:取出細胞培養板,平衡至室溫後,每孔加入90μl CellTiter-Glo®細胞活力檢測試劑(Promega,Cat#:G7573),振盪混勻後放於暗處靜置10分鐘後使用酶標儀的發光程序進行檢測。使用GraphPad Prims軟體計算IC50值。使用陰性抗體IgG1與化合物D偶聯的抗體-藥物偶聯物(y=4)作為陰性對照。實驗結果如下表所示:
實驗結果表明:ADC2(y=4)具有較好的腫瘤細胞的體外殺傷活性。
實施例10:抗體藥物偶聯物的腫瘤殺傷活性
為進一步研究抗體-藥物偶聯物對體內形成的腫瘤的增殖抑制效果,在小鼠體內用NCI-H929細胞(ATCC保藏號CRL-9068)形成移植瘤後,評估本公開抗體-藥物偶聯物的抗腫瘤效果。將10x106個NCI-929細胞接種於到6-8週齡的免疫缺陷的雌性小鼠(NOD SCID)背部皮下,14天後開始藉由靜脈注射抗體-藥物偶聯物,每1週尾靜脈注射給藥1次,連續給藥兩週,給藥劑量為3mg/kg。對照組採用人IgG1 isotype control蛋白,劑量為3mg/kg,對照組或給藥組每組5隻小鼠。藉由測量腫瘤體積計算抑瘤率,計算公式如下:
抑瘤率TGI %=100%-(第14天給藥組腫瘤體積-第0天給藥組腫瘤體積)/(第14天對照組腫瘤體積-第0天對照組腫瘤體積)。
實驗結果如下表所示:
實驗結果表明:抗體-藥物偶聯物ADC2(y=4)顯示出良好的體內腫瘤抑制效果。

Claims (15)
- 一種通式(I)所示的抗體藥物偶聯物或其藥學上可接受的鹽或溶劑化合物,其中,W選自-(CReRf)g-X1-(CReRf)u-X2-(CReRf)h-,Re或Rf各自獨立地選自氫、氘、羥基、胺基、烷基、鹵素、鹵烷基、氘代烷基或羥烷基;較佳地,Re或Rf各自獨立地選自氫、氘,更佳為氫,X1或X2各自獨立地選自N、H、O或S;較佳地,X1或X2各自獨立地選自S;更佳為O,g、u或h各自獨立地選自1、2、3或4;較佳地,g、u或h各自獨立地選自1、2、3;更佳為2,y為1~20;較佳4~10;更佳4、6、8或10,Ab為抗BCMA抗體或其抗原結合片段,該抗BCMA抗體或其抗原結合片段包含重鏈可變區和輕鏈可變區,該重鏈可變區包含SEQ ID NO:3所示的HCDR1、SEQ ID NO:4所示的HCDR2和SEQ ID NO:5所示的HCDR3;以及該 輕鏈可變區包含SEQ ID NO:6所示的LCDR1、SEQ ID NO:7所示的LCDR2和SEQ ID NO:8所示的LCDR3。
- 如請求項1所述的抗體藥物偶聯物或其藥學上可接受的鹽或溶劑化合物,其中,該抗BCMA抗體或其抗原結合片段選自鼠源抗體、嵌合抗體、人抗體或人源化抗體。
- 如請求項2所述的抗體藥物偶聯物或其藥學上可接受的鹽或溶劑化合物,其中,該抗BCMA抗體或其抗原結合片段進一步包含人IgG1、IgG2、IgG3或IgG4或其變體的重鏈恆定區,較佳地,該抗BCMA抗體或其抗原結合片段進一步包含人IgG1、IgG2或IgG4的重鏈恆定區;更佳地,該抗BCMA抗體或其抗原結合片段進一步包含胺基酸突變後具有增強的ADCC毒性的IgG1重鏈恆定區。或者,該抗BCMA抗體或其抗原結合片段進一步包含如SEQ ID NO:22所示的重鏈恆定區。
- 如請求項2所述的抗體藥物偶聯物或其藥學上可接受的鹽或溶劑化合物,其中,該抗BCMA抗體或其抗原結合片段進一步包含人抗體κ鏈、λ鏈或其變體的輕鏈恆定區;較佳地,該抗BCMA抗體或其抗原結合片段進一步包含人抗體κ鏈的輕鏈恆定區;進一步佳地,該抗BCMA抗體或其抗原結合片段進一步包含如SEQ ID NO:23所示的輕鏈恆定區。
- 如請求項2所述的抗體藥物偶聯物或其藥學上可接受的鹽或溶劑化合物,其中,該抗BCMA抗體或其抗原結合片段包含選自以下序列所示的 重鏈可變區,或與以下序列相比具有至少70%、75%、80%、85%、90%、95%或99%同一性的重鏈可變區:SEQ ID NO:9、SEQ ID NO:10或SEQ ID NO:11;和/或,選自以下序列所示的輕鏈可變區,或與以下序列相比具有至少70%、75%、80%、85%、90%、95%或99%同一性的輕鏈可變區:SEQ ID NO:12、SEQ ID NO:13或SEQ ID NO:14。
- 如請求項5所述的抗體藥物偶聯物或其藥學上可接受的鹽或溶劑化合物,其中,該抗BCMA抗體或其抗原結合片段包含SEQ ID NO:9所示的重鏈可變區和SEQ ID NO:12所示的輕鏈可變區;或,該抗BCMA抗體或其抗原結合片段包含SEQ ID NO:10所示的重鏈可變區和SEQ ID NO:13所示的輕鏈可變區;或,該抗BCMA抗體或其抗原結合片段包含SEQ ID NO:11所示的重鏈可變區和SEQ ID NO:14所示的輕鏈可變區。

- 一種如請求項1至11中任一項所述的抗體藥物偶聯物或其藥學上可接受的鹽或溶劑化合物或如請求項13所述的醫藥組成物在製備用於治療或預防BCMA介導的疾病或病症的藥物中的用途。
- 如請求項14所述的用途,其中,該BCMA介導的疾病或病症選自癌症或自身免疫疾病,其中該癌症較佳為表達BCMA的癌症,更佳淋巴瘤、白血病或骨髓瘤;該自身免疫疾病選自紅斑狼瘡、IgA腎病或風濕性關節炎。
