Abstract

Description
出於以下原因中之一或多者,諸如聚碸及聚醚碸之芳族疏水性聚合物作為薄膜聚合物為有吸引力的:較高玻璃轉移溫度、非晶形玻璃態、熱及氧化穩定性、極好的強度及可撓性、對極端pH之耐性及甚至在高溫下之低蠕變。然而,此等聚合物固有地為疏水性且因此由此等聚合物製成之多孔性薄膜不可藉由水來潤濕。

本發明提供一種自潤濕多孔性薄膜,其包含芳族疏水性聚合物及包含式A-B或A-B-A之共聚物的潤濕劑,其中A為包含聚合單體或單體之混合物的親水性片段,其中單體具有式(I):CH2=C(R1)(R2) (I),其中R1為氫或烷基,且R2選自經取代或未經取代之雜環基、經取代或未經取代之雜芳基、甲醯胺基、甲醯胺基烷基、胺基羰基、烷基羰氧基及經兩性離子基取代之胺基羰基烷基,且B為聚醚碸,其中片段B及A經由氧原子連接。本發明亦提供製備此類自潤濕多孔性薄膜之方法。
本發明之自潤濕多孔性薄膜具有較高水可濕性,如由其71達因/平方公分之高臨界潤濕表面張力(CWST)證明。潤濕添加劑具有與芳族疏水性聚合物之高度相容性且因此不濾出或僅最低限度地濾出。潤濕添加劑亦自身均勻分佈在多孔性薄膜中。多孔性薄膜具有較高水滲透率,例如在330-620LMH範圍內。多孔性薄膜亦具有由低積垢程度表示之低非特異性蛋白質結合(BSA)。
在一具體實例中,本發明提供一種自潤濕多孔性薄膜,其包含芳族疏水性聚合物及包含式A-B或A-B-A之共聚物的潤濕劑,其中A為包含聚合單體或單體之混合物的親水性片段,其中單體具有式(I):CH2=C(R1)(R2) (I),其中R1為氫或烷基,且R2選自經取代或未經取代之雜環基、經取代或未經取代之雜芳基、甲醯胺基、甲醯胺基烷基、胺基羰基、烷基羰氧基及經兩性離子基取代之胺基羰基烷基,且B為聚醚碸,其中片段B及A經由氧原子連接。「A」進一步包含端基。
如本文所用之術語「雜環基」指單環雜環基或雙環雜環基。單環雜環為含有至少一個獨立地選自由O、N、N(H)及S組成之群的雜原子的三員、四員、五員、六員或七員環。三員或四員環不含或含有一個雙鍵及一個選自由O、N、N(H)及S組成之群的雜原子。五員環不含或含有一個雙鍵及一個、兩個或三個選自由O、N、N(H)及S組成之群的雜原子。六員環含有零個、一個或兩個雙鍵及一個、兩個或三個選自由O、N、N(H)及S組成之群的雜原子。七員環含有零個、一個、兩個或三個雙鍵及一個、兩個或三個選自由O、N、N(H)及S組成之群的雜原子。單環雜環可未經取代或經取代且經由單環雜環內含有之任何可取代碳原子或任何可取代氮原子連接至母分子部分。
雜環基之實例包括吡啶基、哌啶基、哌基、吡基、吡咯
基、哌喃基、四氫哌喃基、四氫硫代哌喃基、吡咯啶基、吡咯啶酮基、呋喃基、四氫呋喃基、噻吩基、四氫噻吩基、嘌呤基、嘧啶基、噻唑基、噻唑啶基、噻唑啉基、唑基、三唑基、四唑基、四基、苯并唑基、嗎啉基、硫代嗎啉基、喹啉基及異喹啉基。
術語「雜芳基」指具有五至十個環原子之環狀芳基,其中至少一個原子為O、S或N,且其餘原子為碳。雜芳基之實例包括吡啶基、吡基、嘧啶基、吡咯基、吡唑基、咪唑基、噻唑基、唑基、異唑基、噻二唑基、二唑基、噻吩基、呋喃基、喹啉基及異喹啉基。
雜環基或雜芳基部分上之取代基可為烷基、鹵基、羥基、硝基、羧基、酮基、側氧基、胺基、硫氧基、磺醯基、磺醯基氧基、磷醯基、膦醯基或其任何組合。舉例而言,環CH2基團可經C=O基團置換。取代可在碳原子上或在雜原子(諸如環氮)上,例如環氮上之烷基提供四級銨基。
根據一具體實例,烷基較佳為C1-C6烷基。烷基之實例包括甲基、乙基、正丙基、異丙基、正丁基、第二丁基、異丁基、第三丁基、正戊基、異戊基、正己基及其類似者。
兩性離子基包含帶正電基團及帶負電基團,其由適合之間隔原子(諸如碳、氧或硫)或諸如烷基部分之部分分離。帶正電基團之實例包括銨及四級銨基團。帶負電基團之實例包括羧酸鹽、磺酸鹽及膦酸鹽。

用以形成薄膜之芳族疏水性聚合物可選自聚碸(PSU)、聚伸苯基醚碸(PPES)、聚醚碸(PES)、聚碳酸酯(PC)、聚醚醚酮(PEEK)、聚(酞酮醚碸酮)(PPESK)、聚苯硫醚(PPS)、聚苯基醚(PPE)、聚苯醚(PPO)及聚醚醯亞胺(PEI),其具有以下結構:
芳族疏水性聚合物可具有任何適合的分子量,例如數目平均分子量為約25kDa至約250kDa,較佳約50kDa至約100kDa。芳族疏水性聚合物中「n」之值可為約30至約300,較佳約50至約250。
共聚物之芳族疏水性聚合片段B為用一個或較佳兩個酚部分封端之聚醚碸。此類經酚取代之部分可藉由在對雙(4-氯苯基)碸(或其氟類似物)為過量之雙酚S存在下進行聚縮合引入,如熟習此項技術者所已知。
以上芳族疏水性片段B中之每一者內的重複單元之數目n可為約10至約250,較佳約20至約200,且更佳約30至約100。
嵌段共聚物之數目平均分子量在約5,000至約50,000公克/莫耳,較佳約10,000至約40,000公克/莫耳範圍內。
根據一具體實例,共聚物具有式(Ia)或(Ib):

舉例而言,1-乙烯基吡咯啶-2-酮單體可在預先形成之聚合片段B上聚合。市售可得之聚合片段B之一實例為具有羥基端基之Solvay之雙遠螯聚醚碸VW-10700RP。1-乙烯基吡咯啶-2-酮在聚合物上如以下所說明聚合:

本發明進一步提供藉由上文所描述之方法製備之自潤濕多孔性薄膜。
根據本發明之一具體實例,自潤濕多孔性薄膜為多孔性薄膜,例如奈米多孔性薄膜,例如孔直徑在1nm與100nm之間的薄膜或孔直徑在1μm與10μm之間的微孔薄膜。
形成薄膜之聚合物溶液藉由將聚合物溶解於溶劑或溶劑之混合物中來製備。多種聚合物適用作本發明中之形成薄膜之聚合物,且在此項技術中已知。適合之聚合物可包括諸如以下之聚合物:聚碸(PSU)、聚醚碸(PES)、聚苯基醚(PPE)、聚伸苯基醚碸(PPES)、聚苯醚(PPO)、聚碳酸酯(PC)、聚(酞酮醚碸酮)(PPESK)、聚醚醚酮(PEEK)、聚醚酮酮(PEKK)及聚醚醯亞胺(PEI)及其摻合物。
除一或多種聚合物之外,典型的聚合物溶液包含至少一種溶劑,且可進一步包含至少一種非溶劑。適合之溶劑包括例如N,N-二甲基甲醯胺(DMF)、N,N-二甲基乙醯胺(DMAC)、N-甲基吡咯啶酮(NMP)、二甲亞碸(DMSO)、甲基亞碸及其混合物。適合之非溶劑包括例如水、各種聚乙二醇(PEG,例如PEG-200、PEG-300、PEG-400、PEG-1000)、各種聚丙二醇、各種醇(例如甲醇、乙醇、異丙醇(IPA)、戊醇、己醇、庚醇及辛醇)。
典型地,在薄膜澆鑄溶液中以約3至約20wt%,較佳在約4至約15wt%範圍內的量包括本發明之共聚物A-B-A。
薄膜澆鑄溶液典型地包括約10至約30wt%芳族疏水性聚合物、約0至約10wt%添加劑(諸如PEG)、呈至多約90wt%的量的極性溶劑或溶劑之混合物(諸如NMP、DMF及/或DMAc)。亦可包括非溶劑(諸如水及/或醇)。
可在形成自潤濕薄膜中利用之典型淬滅浴包括(但不限於)含有非溶劑(諸如水、醇及其類似者)自身或非溶劑與一或多種極性溶劑之組合的浴。
溶液之適合的組分在此項技術中已知。包含聚合物之說明性溶液及說明性溶劑及非溶劑包括例如在美國專利4,340,579、4,629,563、4,900,449、4,964,990、5,444,097、5,846,422、5,906,742、5,928,774、6,045,899、6,146,747及7,208,200中所揭示之彼等物。
芳族疏水性聚合物、共聚物及溶劑及其他添加劑首先混合在一起且隨後在約55至約75℃,較佳約60至約65℃之溫度下加熱,直至獲得均質溶液。
可在適合之基板(諸如玻璃)上藉由使用刮刀澆鑄具有適合之厚度(例如約0.005至約0.006吋)之膜來製備平坦薄片薄膜。將澆鑄膜凝聚在淬滅浴中且於水中洗滌,持續足以濾出不直接結合於疏水性-基質聚合物之任何材料的時間段。藉由熟習此項技術者已知之方法乾燥薄膜。
除製備平坦薄片薄膜以外,本發明之自潤濕薄膜可使用此項技術中熟知之製程形成為摺疊薄膜、空心纖維薄膜、管狀薄膜及其類似者。
本發明進一步提供過濾流體之方法,該方法包含使流體穿過自潤濕多孔性薄膜。
根據本發明之具體實例的自潤濕多孔性薄膜可用於各種應用,包括例如血液透析、微過濾及超過濾應用。另外其亦可用於診斷應用(包括例如樣品製備及/或診斷側流裝置)、噴墨應用、微影(例如作為基於HD/UHMW PE之介質之替代)、為醫藥工業過濾流體、金屬移除、產生超純水、處理工業及地表水、為醫學應用(包括為家庭及/或患者使用,例如靜脈內應用,亦包括例如過濾生物流體,諸如血液(例如病毒移除))過濾流體、為電子工業過濾流體、為食物及飲料工業過濾流體、啤酒過濾、澄清、過濾含有抗體及/或蛋白質之流體、過濾含有核酸之流體、細胞偵測(包括就地)、細胞收集及/或過濾細胞培養流體。或者,或另外,根據本發明之具體實例之多孔性薄膜可用於過濾空氣及/或氣體,及/或可用於排氣應用(例如允許空氣及/或氣體但不允許液體穿過其間)。根據本發明之具體實例之多孔性薄膜可用於多種裝置中,包括手術裝置及產品,諸如眼科手術產品。
根據本發明之具體實例,自潤濕多孔性薄膜可具有多種組態,包括平面、平坦薄片、摺疊、管狀、螺旋形及空心纖維。
根據本發明之具體實例之自潤濕多孔性薄膜典型地安置於包含至少一個入口及至少一個出口且界定入口與出口之間的至少一個流體流動路徑之殼體中,其中至少一個本發明薄膜或包括至少一個本發明薄膜之過濾器跨越流體流動路徑,以提供過濾裝置或過濾模組。在一具體實例中,提供包含以下之過濾裝置:包含入口及第一出口且界定入口與第一出口之間的第一流體流動路徑的殼體;及至少一個本發明薄膜或包含至少一

較佳地,對於交叉流應用,至少一個本發明多孔性薄膜或包含至少一個本發明薄膜之過濾器安置於包含至少一個入口及至少兩個出口且界定入口與第一出口之間的至少一個第一流體流動路徑及入口與第二出口之間的第二流體流動路徑之殼體中,其中本發明薄膜或包含至少一個本發明薄膜之過濾器跨越第一流體流動路徑以提供過濾裝置或過濾模組。在說明性具體實例中,過濾裝置包含交叉流過濾模組、殼體,該殼體包含入口、包含濃縮物出口之第一出口及包含滲透物出口之第二出口且界定入口與第一出口之間的第一流體流動路徑及入口與第二出口之間的第二流體流動路徑,其中至少一個本發明薄膜或包含至少一個本發明薄膜之過濾器跨越第一流體流動路徑安置。
過濾裝置或模組可滅菌。可利用具有適合形狀且提供入口及一或多個出口之任何殼體。
殼體可由任何適合的剛性不透水材料製造,包括任何不透水之熱塑性材料,其與處理中之流體相容。舉例而言,殼體可由金屬,諸如不鏽鋼或由聚合物,例如透明或半透明聚合物,諸如丙烯酸、聚丙烯、聚苯乙烯或聚碳酸酯樹脂製造。
以下實施例進一步說明本發明,但當然不應將其解釋為以任何方式限制其範疇。
材料及方法:起始物質為Solvay之雙遠螯聚醚碸VW-10700 RP,其描述為具有末端酚部分,密度如藉由滴定所測定,為每公克樹脂0.187
毫當量OH。聚合物之Mw=21332,如藉由GPC分析在DMAc中所測定(用PS標準)。所有引用之資料已提供於製造商之分析憑證中。在DMF中用0.05N LiBr及PMMA作為標準物進行之內部分析提供Mw=31578及PDI=1.62之值。
將乙烯基單體真空蒸餾或通過含有抑制劑移除劑之管柱。
此實施例說明製備根據具體實例之共聚物A-B-A之一般方案。
將VW-10700 RP溶解於無水NMP中,隨後添加乙烯系單體及作為自由基誘發劑之AIBN。所得混合物在環境溫度下用氬氣淨化至少2-5分鐘,隨後在80℃下加熱16-24小時。之後反應混合物對空氣開放且添加至過量之經劇烈攪拌非溶劑,通常至少10體積之2-丙醇中。過濾出(或在一些情況下,離心)所得沉澱物,用2-丙醇充分沖洗且在真空烘箱中在70℃下乾燥隔夜。藉由GPC(具有0.05N LiBr之DMF,用PMMA分子量標準物校準)分析所獲得之物質且組合物藉由1H NMR分析(在DMSO-d6中)及元素分析測定。
此實施例說明根據本發明之具體實例製備PVP-b-PES-b-PVP之方法,其中(m1+m2)/n=0.35:
將VW-10700RP(0.5g;0.093mmol OH)、1-乙烯基吡咯啶-2-酮(1.0mL;9mmol)及AIBN(8mg;0.047mmol)溶解於無水NMP(3mL)中且根據一般方案共聚合,產生0.39g產物。
1H NMR分析指出35莫耳% VP單元,如藉由比較7.80-8.20ppm之間(4H PES單元)及1.20-2.40ppm之間(6H VP單元)的信號之積分所測定。元素分析(N%=2.46;S%=10.71)指出34莫耳% VP單元。GPC:Mw=31370;PDI=1.55。
實施例3
此實施例說明根據本發明之具體實例製備PVP-b-PES-b-PVP之方法,其中(m1+m2)/n=0.50。
將VW-10700RP(0.5g;0.093mmol OH)、1-乙烯基吡咯啶-2-酮(2.0mL;18mmol)及AIBN(16mg;0.094mmol)溶解於無水NMP(2mL)中且根據一般方案共聚合,產生0.66g產物。
1H NMR分析指出50莫耳% VP單元,如藉由比較7.80-8.20ppm之間(4H PES單元)及1.20-2.40ppm之間(6H VP單元)的信號之積分所測定。元素分析(N%=4.09;S%=8.56)指出52莫耳% VP單元。GPC:Mw=32600;PDI=1.58。
此實施例說明根據本發明之具體實例製備
PVAc-b-PES-b-PVAc之方法,其中(m1+m2)/n=0.16:
將VW-10700RP(0.5g;0.093mmol OH)、乙酸乙烯酯(1.75mL;18.7mmol)及AIBN(15mg;0.092mmol)溶解於無水NMP(3mL)中且根據一般方案共聚合,不同之處在於使用甲醇作為非溶劑來沉澱及洗滌產物(0.57g)。
1H NMR分析指出16莫耳% VAc單元,如藉由比較7.80-8.20ppm之間(4H PES單元)及4.60-4.80ppm之間(1H VAc單元)的信號之積分所測定。GPC:Mw=30225;PDI=1.59。
此實施例說明根據本發明之具體實例製備P4VP-b-PES-b-P4VP之方法,其中(m1+m2)/n=0.11:
將VW-10700RP(0.5g;0.093mmol OH)、4-乙烯基吡啶(1.1mL;10.2mmol)及AIBN(15mg;0.092mmol)溶解於無水NMP(3mL)中且根據一般方案,使用甲醇作為非溶劑共聚合,產生0.41g產物。
1H NMR分析指出11莫耳% 4-VP單元,如藉由比較7.20-7.30
ppm之間(4H PES單元)及6.50-6.70ppm之間(2H 4-VP單元)的信號之積分所測定。元素分析(N%=1.11;S%=12.90)指出16莫耳% 4-VP單元。GPC:Mw=32080;PDI=1.60。
此實施例說明製備根據本發明之具體實例之自潤濕薄膜的方法。
將含有30wt% PES E6020、65wt% DMF及5wt% NMP之5g聚合物溶液與12.5g在40wt%下懸浮於DMF中之160nm Li摻雜之經HMDS處理之二氧化矽粒子(於他處由Harton等人描述)組合。向此混合物中依序添加PEG1000(0.23g)、PVP-K90(0.06g)及實施例3中描述之PVP-b-PES-b-PVP(0.12g),隨後在30,000rpm下均質化5分鐘且在200毫巴下除氣30分鐘。使用刮塗澆鑄棒將所得混合物澆鑄在預先形成的PVOH薄膜(在玻璃板上由在80℃下乾燥2h之10%儲備溶液製備)上,在60℃下置放於烘箱中8分鐘,隨後立即置放於在80℃下之水浴中1h。最終,將膜浸泡在1N HCl中30分鐘,用水洗滌,且浸泡在10% KOH水溶液中隔夜。所得薄膜用水洗滌2h且隨後在70℃下乾燥1h。
為了比較,製備具有含WE3(Ashland之PlasdoneTM S-630 copovidone)而非本發明共聚物之調配物的薄膜作為標準物。就可濕性、水通量、粒子保留率及BSA溶液通量及輸送量而言比較樣品。結果呈現在表1中。

- 如申請專利範圍第1項至第5項中任一項之自潤濕多孔性薄膜,其中該共聚物具有式(Ia):
- 如申請專利範圍第1項至第6項中任一項之自潤濕多孔性薄膜,其中該共聚物具有下式:
- 一種製備自潤濕多孔性薄膜之方法,其包含:(i)將包含溶劑、芳族疏水性聚合物及式A-B或A-B-A之共聚物的溶液澆鑄成所需形狀,其中A為包含聚合單體或單體之混合物的親水性片段,其中該單體具有式(I):CH2=C(R1)(R2) (I),其中R1為氫或烷基,且R2選自經取代或未經取代之雜環基、經取代或未經取代之雜芳基、甲醯胺基、甲醯胺基烷基、胺基羰基、烷基羰氧基及經兩性離子基取代之胺基羰基烷基,且B為聚醚碸,其中片段B及A經由氧原子連接;(ii)使該澆鑄溶液經受相轉換以形成該自潤濕多孔性薄膜;及(iii)視情況用水沖洗該自潤濕多孔性薄膜。
- 如申請專利範圍第8項之方法,其中R1為氫或甲基,且R2選自吡咯啶酮基、吡啶基、咪唑基、N-甲基咪唑基、甲醯胺基、甲醯胺基甲基、胺基羰基、甲基羰氧基及經兩性離子基取代之胺基羰基丙基。
- 如申請專利範圍第8項或第9項之方法,其中A之該單體選自1-乙烯基吡咯啶-2-酮、N-[3-(二甲胺基)丙基]甲基丙烯醯胺、乙酸乙烯酯、1-乙烯咪唑、1-乙烯基-3-甲基咪唑、1-乙烯基-2-吡啶、1-乙烯基-4-吡啶、 丙烯醯胺、N-乙烯基甲醯胺及N-烯丙基甲醯胺及3-(甲基丙烯醯胺基丙基)二甲基(3-磺丙基)銨內鹽。
- 如申請專利範圍第8項至第10項中任一項之方法,其中該芳族疏水性聚合片段B為聚醚碸。
- 如申請專利範圍第8項至第11項中任一項之方法,其中該芳族疏水性聚合物選自聚碸、聚醚碸、聚伸苯基醚、聚伸苯基醚碸、聚苯醚、聚碳酸酯、聚(酞酮醚碸酮)、聚醚酮、聚醚醚酮、聚醚酮酮、聚醯亞胺、聚醚醯亞胺及聚醯胺-醯亞胺。
- 如申請專利範圍第8項至第12項中任一項之方法,其中該芳族疏水性聚合物為聚碸或聚醚碸。
- 如申請專利範圍第8項至第12項中任一項之方法,其中該共聚物具有式(Ia)或(Ib):
