Изобретение относитс к фармацевтической промышленности и касаетс получени биологически активных веществ .
Известен способ совместного получени {1нсулина и панкреатина из поджелудочной железы свиньи путем экстракции органическим растворителем с последующим центрифугированием и выделением из экстракта инсулина и из осадка-панкреатина 1.
Однако известный способ не обеспечивает высокого выхода и высокой активности препаратов.
Целью изобретени вл етс повышение выхода и активности препаратов ,
Эта цель достигаетс тем, что экстракцию исходного сырь осзтдествл ют при рН 5-7 40-80%-ным ацетоном в присутствии нейтральных солей, в полученном после центрифугировани осадке устанавливают рН более 5,6 и отдел ют примеси экстракцией 60-70%ным ацетоном, кроме того, экстракцию инсулина из исходного сырь ocyi4ecTвл ют ацетоном при его содержании 5060% ,
Способ осуществл ют следующим образом .
Свежую или консервированную путем быстрого замораживани поджелудочную железу свиньи размельчают и суспендируют в смеси, например., из 24 об.% воды и 76 об.%: ацетона, к которой в случае необходимости добавл ют нейтральные соли, а также соли многоосновных кислот. Дл экстракции и последующего отделени нерастворимых составных частей желез поддерживают концентрацию ацетона в жидкой фазе между 40 и 80 об,%, чтобы было возможным растворение инсулина из ткани желез и дл предотвращени активации проферментов благодар автоматическим процессам в исходной смеси.
Содержание твердого вещества в суспензии составл ет 10-20%, К этой смеси добавл ют соль кальци , лучше всего в форме, водного раствора. Суспензию довод т до.значени -рН 5,7, предпочтительно 5,4-5,6, перемешивают. Врем экстракции составл ет 0,5-1,5 часа. Затем твердые вещества отдел ют путем центрифугировани .
Твердое вещеатво можно экстрагировать многократно, однако дл хорошего
