Abstract
Description
The invention relates to veterinary medicine and can be used in animal husbandry to stimulate metabolism, the immune system, the prevention and treatment of infectious gastrointestinal diseases of calves of the milk growing period.
А. Экологические особенности заболеваний пищеварительной системы новорожденных телят//One of the economically significant problems of modern industrial livestock is the production and rearing of young animals. From the first days of life, newborns are sensitive to environmental factors (V. Mishchenko et al. Environmental features of diseases of the digestive system of newborn calves //
№3. С. 34-38). 2012. №7.С. 26-30).Veterinary medicine, 2005. No. 3. S. 34-38). Their immune system is poorly developed and protection against infectious pathogens is provided by colostrum immunity. Their body is most sensitive to intestinal infections. As a rule, already in the first days, newborn calves have digestive problems and develop diarrheal syndrome, followed by an energy-deficient state. The treatment of diarrheal syndrome is especially acute in herds of stationary dysfunctional but viral and bacterial intestinal infections. The causative agents of viral infections are not sensitive to antibiotics, and in Escherichia coli - the leading pathogen, extremely high drug resistance (Efanova L.I. et al. Bacterial and viral pathogens in calves with the syndrome of diarrhea and pneumonia // Veterinary Medicine. 2012. No. 7.C . 26-30).
For the treatment of diarrheal syndrome, chemo-antibiotic therapy is widely practiced. In a certain range of doses, these substances ensure the death of pathogenic microorganisms, without causing much harm to the animal organism. However, drug resistance is rapidly developing for antibiotics. In addition, normal intestinal microflora is more sensitive to the biocidal effect of antibiotics. Ultimately, this inevitably leads to the development of dysbiosis. A serious problem of chemotherapy is the toxicity and side effects of drugs.
The prophylactic use of pro- and prebiotics to stimulate digestion and prevent diarrheal diseases is often associated with the development of dysbiosis and does not provide the necessary therapeutic effect for diarrhea of viral and bacterial etiology.
- 2009. - №1 (28) - С. 76-79).The possibility of overcoming drug resistance of pathogenic microorganisms in case of local infectious processes has been shown in a number of scientific studies (Evlevsky A.A. et al. New approaches to the treatment of acute postpartum endometritis and mastitis in cows // Veterinary pathology. - 2009. - No. 1 (28) - S. 76-79).
In veterinary medicine, it is known to use low concentrations of formalin for the prevention of mastitis in cows in the dry period (RF Patent No. 2239422 of 11/10/2004). The intracisternal administration of 0.3% formalin to cows with infected udder lobes provides for the rapid neutralization of microorganisms and the formation of local mammary immunity in relation to all associations of potential mastitis pathogens.
It is known to use low concentrations of formalin for the treatment of endometritis in cows (RF Patent No. 2219816 from 12/20/2003). The method includes intrauterine administration of a 0.3% formalin solution in a volume of 50 ml per administration 3-4 times with an interval of 12 hours. This provides a persistent clinical therapeutic effect and the formation of immunity against the association of microbial pathogens that caused the infectious process.
In these developments, formalin is used as the main antimicrobial and detoxification component, which provides a high therapeutic effect and local formation of immunity.
It is known to use low concentrations of formalin in a preparation comprising highly dispersed silica - 2.0-5.0%, formaldehyde - 0.5%, liquid nutrient medium - the rest (RF Patent No. 2158128 of 10.27.2000). The drug is used to treat diarrheal syndrome with intestinal infections. Along with high therapeutic and prophylactic effectiveness, the composition of this drug includes very expensive, complex in composition and deficient nutrient media Needle, Earl, Hanks, Tyrode, Dulbekko, 199 or other similar environments. This significantly reduces the possibility of widespread use of this development in the practice of veterinary medicine.
A complex immunometabolic drug is known - an amber biostimulant comprising succinic acid and an ASD of the second fraction in its composition (RF Patent No. 2303979 of 08/10/2007). The composition of this drug is most suitable for stimulating digestion processes and enhancing factors of natural resistance. However, this drug does not contain an energy component, which does not allow an effective course of stimulating therapy for hypotrophy. The absence of an antimicrobial component in its composition cannot have a positive result with intestinal infections.
A complex immunometabolic drug is known comprising 1% succinic acid, 4% ASD of the second fraction, 0.4% formaldehyde (RF Patent 2361579 dated 07/20/2009 "Method for producing a complex immunotropic antiseptic drug for the treatment and prevention of infectious diseases of animals"). The drug has antiseptic, immunomodulatory and trophicostimulating activity with the parenteral route of administration, provides an increase in the effectiveness of treatment and prevention of infectious diseases of animals. The drug does not cause resistance of pathogenic microorganisms to it, which allows you to use it repeatedly without side effects.
However, this drug does not contain an energy component. In clinical trials, it was found that a 0.4% concentration of formaldehyde in the drug does not provide the desired positive effect with enteral use in the clinic for the treatment of diarrhea syndrome.
The objective of the present invention is to develop a complex enteral composition that provides a clinically pronounced effect of stopping diarrheal syndrome with intestinal infections in calves of the milk period of cultivation while stimulating the digestion, metabolism, and immune system.
The specified task is implemented as follows. Composition for the treatment of diarrheal syndrome with intestinal infections of calves in the milk period of cultivation, including in 100 ml of solution: 1.0 g of succinic acid; 4 ml of ASD of the second fraction; 2.0 ml of 38-39% formalin; 30 ml of glycerin; 2 g of sodium chloride; drinking water - the rest.
Succinic acid (UC) is used as the main metabolite in the enteral composition. UC and its salts (succinates) have a wide spectrum of action on various mechanisms of regulation of the metabolic activity of cells. The object of the action of UC are cells and tissues that are in a state of excitement or pathologically altered. UC is a powerful stimulator of energy production in cells, which is especially important in all kinds of pathophysiological conditions, including toxicosis, when the body does not have enough energy for the normal provision of vital functions. Enteral use of UC allows you to provide energy to the cells of the gastric mucosa and increase the secretion of gastric juice.
В качестве средства стимуляции пищеварения и повышения факторов естественной резистентности (иммунитета) используется антисептик стимулятор Дорогова второй фракции (АСД второй фракции).As a means of stimulating digestion and increasing factors of natural resistance (immunity), an antiseptic Dorogov stimulator of the second fraction (ASD of the second fraction) is used.
26.12.2008).When administered enterally, the second fraction of the second fraction has an activating effect on the central and autonomic nervous system, stimulates the secretion of digestive glands, increases the activity of digestive tissue enzymes, helps normalize digestion, assimilation of substances and increases the body's resistance (Instructions for the use of ASD. Register. No. PVR- 2-1.1 / 00937 dated 12/26/2008).
As an easily digestible glucoplastic energy source, glycerol can be used. Glycerin is a trihydric alcohol, a colorless, syrupy liquid, with a sweet taste. Glycerin attracts water, which is important for the prevention of dehydration. Easily absorbed through the gastric mucosa. (Mozgov I.E. Pharmacology. - M., 1974. - S. 148-149). In the liver, glycerol is converted to glucose through gluconeogenesis.
Е. - М., 1974. - С. 263).As a means of improving digestion, sodium chloride is used. The metabolism of sodium chloride is closely associated with metabolic processes. With enteral use, sodium chloride enhances the secretion of digestive glands, promotes the formation of pepsin and the production of gastric juice (I. Mozgov Pharmacology. - M., 1974. - S. 263).
- С. 29-30).Formaldehyde in the form of an aqueous solution of 38-40% formalin has long been used in biotechnology for the manufacture of inactivated vaccines. At a concentration of 0.3-0.5%, it effectively neutralizes pathogenic microorganisms and inactivates toxic products of their vital activity (G. Ramon. 40 years of research work. M: Medicine, 1962. - P. 29-30).
Formalin is used as the main antimicrobial component. In vitro experimental experiments, it was found that the addition of 1 ml of 0.6% formalin to 10 ml of the abomasum of calves of patients with colibacteriosis, followed by an hour exposure at 37 ° C, ensures the death or loss of pathogenicity factors of the causative agent of colibacteriosis E. coli. This makes it possible to use formalin at a given concentration in a volume of 100.0 ml for the treatment of diarrhea syndrome in newborn calves.
The method of obtaining the composition for stimulating immunometabolic processes and the method of its use for the prevention and treatment of diarrheal syndrome with intestinal infections in calves of the milk growing period is illustrated by the following examples.
Composition by prescription. A sample of 10 g of succinic acid, 20 g of sodium chloride was added to the glass flask, 500 ml of drinking water was added. When heated to 80 ° C, complete dissolution of the components was achieved. While cooling to 50 ° C, 300 ml of glycerol and 40 ml of second-stage ASD and 20 ml of formalin were added. The addition of drinking water brought the volume of liquid to 1000.0 ml. After mixing, they were packaged in 100 ml vials and sealed. The presence of glycerol and formalin ensures a long shelf life (at least 12 months).
Preservation of physico-chemical properties and sterility allows you to pack and store the compositions in food plastic bottles or cans, which makes it possible to manufacture the composition in industrial volumes.
The clinical efficacy of the energy metabolism composition is illustrated by the following examples.
Example 1. Evaluation of the clinical effectiveness of the composition in diarrhea syndrome of newborn calves.
20.07.2009.This series of experiments was carried out in February-March 2017 in the Experimental selection station of the Lgovsky district of the Kursk region. The etiology of diarrheal syndrome is caused by drinking colostrum (milk) from cows with mastitis. As a comparison, we used a complex immunotropic antiseptic drug for the treatment and prevention of infectious diseases of animals, including 1% succinic acid, 4% ASD of the second fraction, 0.4% formalin, made as described in RF patent 2361579 dated July 20, 2009.
In the experimental and compared groups included calves with the presence of clinically pronounced diarrhea syndrome. The inventive composition and the known drug was drunk 30 minutes before feeding with milk. The course of use is until the termination of diarrheal syndrome.
1.In the course of clinical observations it was found that a pronounced reduction of diarrheal syndrome was achieved after one - two-fold drinking of the claimed and compared formulations. However, the effectiveness of relief of diarrheal syndrome when drinking the claimed composition was higher. So, the clinical recovery of 44 of the 57 calves of the experimental group was achieved after 3-4 feedings, which amounted to 59.7%. A weak degree, not hazardous to the health of calves, diarrhea was observed in 7 calves of the experimental group on 3 and in 2 on 4 days. The effect of reducing diarrheal syndrome of the compared group was noticeably weaker. So, on day 2, clinical recovery was diagnosed in 17 (31%) calves. In general, the picture of the clinical recovery of calves of both groups for clarity is presented in table 1.
Кишечная палочка была выделена в непатогенной форме, что указывало на возрастание ее роли в физиологическом процессе пищеварения (таблица 2).When bacteriological examination feces from 5 patients with diarrhea of calves of the first experimental group (were selected twice: - with the clinical manifestation of diarrhea and after its attenuation), that oral administration of the claimed composition provided complete displacement of staphylococci, Pseudomonas aeruginosa and pathogenic form of E. coli. E. coli was isolated in a non-pathogenic form, which indicated an increase in its role in the physiological process of digestion (table 2).
The use of the claimed composition provided a more pronounced activation of metabolic processes. In calves of the first experimental group of total protein, the levels of total calcium, reserve alkalinity and inorganic phosphorus on day 10 were closer to the background values of clinically healthy peers.
4.A pronounced tendency to normalize the content of total protein, calcium, phosphorus and reserve alkalinity of the blood is important in the matter of increasing the overall resistance of the body. The results of studies evaluating the impact of the claimed composition on the humoral factors of natural resistance are presented in table 4.
The dynamics of immunological parameters indicates that the claimed composition provided a more pronounced tendency to increase the phagocytic activity of neutrophils, bactericidal and lysozyme activity of blood serum.
Example 2. Clinical efficacy of the composition for diarrheal syndrome of viral and bacterial etiology in calves during the dairy period of cultivation.
Evaluation of the effectiveness of the use of the composition was carried out in the conditions of the Amosovsky farm in the Kursk region during the period of mass incidence of diarrhea in newborn calves. In the biomaterial from the dead calves, the following were distinguished: the diarrhea virus genome and pathogenic forms of E. coli (E. coli). This indicated a viral-bacterial etiology. The effectiveness of antibiotic use was extremely low or completely absent.
20.07.2009.In a clinical trial, we used a well-known complex immunotropic antiseptic drug for the treatment and prevention of infectious diseases of animals, including 1% succinic acid, 4% ASD of the second fraction, 0.4% formalin, made as described in RF patent 2361579 dated 20.07 .2009.
50%.The order of application. The inventive composition and the known drug was drunk 30 minutes before feeding with milk. In cases of severe intoxication, feeding with milk was completely replaced by drinking the composition or a known drug. In such cases, 100 ml of the composition or preparation was added in 900 ml - 1 liter of saline 1% aqueous solution. The results of clinical observations are presented in table 5. Despite the complex viral and bacterial background of diarrheal syndrome, the use of the inventive composition provided a sufficiently pronounced therapeutic effect superior to the well-known drug. All experimental calves survived. The death of calves before applying the inventive composition reached 50%.
Thus, based on the results of clinical trials, it was found that the claimed composition showed a higher therapeutic effect when stopping diarrheal syndrome of infectious etiology, has a more pronounced metabolic and immunostimulating activity.
