Claims (47)
1. Способ связывания полипептида, где полипептид содержит пептидную основную цепь и одну или более реакционноспособных тиольных групп, включающий в себя проведение реакции между реакционноспособной тиольной группой полипептида и ен-соединением, содержащим одну или более реакционноспособных ен-групп, в условиях, которые способствуют протеканию опосредованной радикальными группами тиол-ен-реакции, где реакционноспособная тиольная группа полипептида имеет реакционную способность в отношении опосредованной радикальными группами тиол-ен-реакции, которая, по меньшей мере, в 2 раза превышает реакционную способность цистеин-тиольной группы.
2. Способ по п. 1, где способ включает в себя проведение реакции между реакционноспособной тиольной группой полипептида и радикальным инициатором и ен-соединением.
3. Способ по п. 1 или 2, где реакционноспособная тиольная группа полипептида отделена от пептидной основной цепи полипептида двумя или более атомами углерода.
4. Способ по п. 3, где реакционноспособная тиольная группа полипептида представляет собой тиольную группу остатка гомоцистеина полипептида или тиольную группу остатка 2-амино-5-меркаптопентановой кислоты полипептида.
5. Способ по п. 3, где реакционноспособная тиольная группа полипептида представляет собой тиольную группу, прикрепленную к аминогруппе боковой цепи остатка лизина посредством линкера.
6. Способ по любому из пп. 2-5, где радикальный инициатор представляет собой фотоинициатор, выбранный из группы, состоящей из фенил-2,4,6-триметилбензоилфосфината лития (LAP), фенил-2,4,6-триметилбензоилфосфината натрия (NAP), 2-гидрокси-1-[4-(2-гидроксиэтокси)фенил]-2-метил-1-пропанона, 1-гидрокси-циклогексил-фенил-кетона и 2,2-диметокси-1,2-дифенилэтан-1-она.
7. Способ по п. 6, где тиол-ен-реакцию инициируют путем подвергания фотоинициатора воздействию света, имеющего длину волны, совпадающую с длиной волны возбуждения фотоинициатра.
8. Способ по п. 6 или 7, где способ дополнительно включает в себя регулирование количества фотоинициатора, интенсивности света и/или периода времени, в течение которого фотоинициатор подвергают воздействию света.
9. Способ по п. 8, где тиол-ен-реакция достигает приблизительно 70%-ного завершения - приблизительно 95%-ного завершения.
10. Способ по любому из пп. 1-9, где полипептид содержит две или более реакционноспособных групп, каждая из которых отделена от пептидной основной цепи полипептида двумя или более атомами углерода.
11. Способ по п. 10, где ен-соединение содержит две или более реакционноспособных ен-групп.
12. Способ по п. 11, где полипептид содержит n реакционноспособных тиольных групп, ен-соединение содержит m реакционноспособных ен-групп, где n и m независимо означают целое число ≥ 2, и n+m ≥ 5.
13. Способ по п. 12, где ен-соединение представляет собой ен-модифицированный биосовместимый мономер.
14. Способ по п. 13, где ен-соединение представляет собой норборнен-модифицированный полиэтиленгликоль (PEG).
15. Способ по п. 13 или 14, где полипептид дополнительно содержит пептидную последовательность, заведомо чувствительную к протеазе.
16. Способ по п. 15, где тиол-ен-реакция обеспечивает биосовместимый сшитый разлагаемый гидрогелевый полимер.
17. Способ по любому из пп. 1-9, где ен-соединение содержит одну реакционноспособную ен-группу.
18. Способ по п. 17, где ен-соединение включает в себя полимер, фрагмент захвата, метку, углевод, нуклеиновую кислоту, или второй полипептид.
19. Способ по п. 17, где ен-соединение включает биологически активный компонент.
20. Способ по любому из пп. 17-19, где полипептид содержит одну реакционноспособную тиольную группу, отделенную от пептидной основной цепи полипептида двумя или более атомами углерода.
21. Способ по любому из пп. 17-20, где полипептид выбирают из пептидных или белковых гормонов, факторов роста, цитокинов, интерлейкинов, рецепторов, солюбилизированных рецепторов, терапевтических белков, ферментов и структурных белков.
22. Способ по любому из пп. 1-9, где полипептид содержит одну реакционноспособную тиольную группу, отделенную от пептидной основной цепи полипептида двумя или более атомами углерода, и ен-соединение содержит две или более реакционноспособных ен-групп.
23. Способ по п. 22, где ен-соединение содержит две или более реакционноспособных ен-групп.
24. Способ по п. 22, где способ дополнительно включает в себя проведение реакции между вторым тиольным соединением и ен-соединением, содержащим две или более реакционноспособных ен-групп, где второе тиольное соединение содержит две или более реакционноспособных тиольных групп.
25. Способ по п. 24, где второе тиольное соединение содержит j реакционноспособных тиольных групп, ен-соединение содержит k реакционноспособных ен-групп, где j и k независимо представляют собой целое число ≥ 2, и j+k ≥ 5.
26. Способ по п. 24 или 25, где второе тиольное соединение включает полимерный фрагмент, выбранный из группы, состоящей из поли(молочной кислоты) (PLA); полигликолида (PGA); сополимера PLA и PGA (PLGA); поли(винилового спирта) (PVA); поли(этиленгликоля) (PEG); поли(этиленоксида); поли(этиленоксид)-co-поли(пропиленоксид)ного блоксополимера; полоксамина; полиангидрида; сложного поли(орто)эфира; поли(гидроксикислот); полидиоксанона; поликарбоната; полиаминокарбоната; поли(винилпирролидон)а; поли(этилоксазолин)а; полиуретана; карбоксиметилцеллюлозы; гидроксиалкилированной целлюлозы; полипептида; нуклеиновой кислоты; модифицированной нуклеиновой кислоты; запертой нуклеиновой кислоты; полисахарида; углевода; гепаран-сульфата; хондроитин-сульфата; гепарина, альгината; и их комбинации.
27. Способ по п. 24 или 25, где второе тиольное соединение включает пептидную основную цепь, разлагаемый пептид и две или более реакционноспособных тиольных групп, каждая из которых отделена от пептидной основной цепи двумя или более атомами углерода.
28. Способ по п. 27, где второе тиольное соединение включает пептидную последовательность, заведомо чувствительную к протеазе, и два остатка гомоцистеина, фланкирующие пептидную последовательность.
29. Способ по п. 24 или 25, где второе тиольное соединение включает полиэтиленгликоль и две концевые тиольные группы, прикрепленные к полиэтиленгликолю.
30. Способ по любому из пп. 22-29, где ен-соединение включает полимерный фрагмент, выбранный из группы, состоящей из поли(молочной кислоты) (PLA); полигликолида (PGA); сополимера PLA и PGA (PLGA); поли(винилового спирта) (PVA); поли(этиленгликоля) (PEG); поли(этиленоксида); поли(этиленоксид)-co-поли(пропиленоксид)ного блоксополимера; полоксамина; полиангидрида; сложного поли(орто)эфира; поли(гидроксикислот); полидиоксанона; поликарбоната; полиаминокарбоната; поли(винилпирролидон)а; поли(этилоксазолин)а; полиуретана; карбоксиметилцеллюлозы; гидроксиалкилированной целлюлозы; полипептида; нуклеиновой кислоты; модифицированной нуклеиновой кислоты; запертой нуклеиновой кислоты; полисахарида; углевода; гепаран-сульфата; хондроитин-сульфата; гепарина, альгината; и их комбинации.
31. Способ по любому из пп. 1-30, дополнительно включающий в себя получение полипептида, содержащего одну или более реакционноспособных тиольных групп.
32. Способ по п. 31, где получение полипептида, содержащего одну или более реакционноспособных тиольных групп, включает в себя синтез полипептида с использованием защищенной(-ого) тиол-содержащей(-его) аминокислоты или аналога аминокислоты, где тиольная группа отделена от атома углерода, смежного с карбоксигруппой, посредством двух или более атомов углерода, как например, защищенного гомоцистеина или 2-амино-5-меркаптопентановой кислоты.
33. Способ по п. 31, где получение полипептида, содержащего одну или более реакционноспособных тиольных групп, включает в себя модифицирование полипептида с использованием тиолирующего агента.
34. Способ по п. 31, где получение полипептида, содержащего одну или более реакционноспособных тиольных групп, включает в себя химическое или ферментативное превращение остатка метионина в полипептиде в остаток гомоцистеина.
35. Связанный полипептид, полученный способом по любому из пп. 1-34.
36. Способ получения биосовместимого сшитого разлагаемого гидрогелевого полимера, включающий в себя проведение реакции между реакционноспособным тиольным соединением и радикальным инициатором и реакционноспособным ен-соединением, где реакционноспособное тиольное соединение включает пептидную основную цепь, разлагаемый пептид и две или более реакционноспособных тиольных групп, каждая из которых отделена от пептидной основной цепи двумя или более атомами углерода; и реакционноспособное ен-соединение представляет собой ен-модифицированный биосовместимый мономер, содержащий две или более реакционноспособных ен-групп.
37. Способ по п. 36, где полипептид содержит n реакционноспособных тиольных групп, ен-соединение содержит m реакционноспособных ен-групп, где n и m независимо представляют собой целое число ≥ 2, и n+m ≥ 5.
38. Способ по п. 36, дополнительно включающий в себя добавление биологически активного компонента в реакционную смесь, где биологически активный компонент соединяют с полимерной матрицей биосовместимого сшитого разлагаемого гидрогелевого полимера.
39. Способ по п. 36 или 38, где реакционноспособные тиольные группы в реакционноспособном тиольном соединении независимо выбирают из тиольной группы остатка гомоцистеина полипептида, тиольной группы остатка 2-амино-5-меркаптопентановой кислоты полипептида и тиольной группы, прикрепленной к аминогруппе боковой цепи остатка лизина посредством линкера.
40. Способ по любому из пп. 36-39, где радикальный инициатор представляет собой фотоинициатор, выбранный из группы, состоящей из фенил-2,4,6-триметилбензоилфосфината лития (LAP), фенил-2,4,6-триметилбензоилфосфината натрия (NAP), 2-гидрокси-1-[4-(2-гидроксиэтокси)фенил]-2-метил-1-пропанона, 1-гидрокси-циклогексил-фенил-кетона и 2,2-диметокси-1,2-дифенилэтан-1-она.
41. Способ по любому из пп. 36-40, где реакционноспособное ен-соединение включает полимерный фрагмент, выбранный из группы, состоящей из поли(молочной кислоты) (PLA); полигликолида (PGA); сополимера PLA и PGA (PLGA); поли(винилового спирта) (PVA); поли(этиленгликоля) (PEG); поли(этиленоксида); поли(этиленоксид)-co-поли(пропиленоксид)ного блоксополимера; полоксамина; полиангидрида; сложного поли(орто)эфира; поли(гидроксикислот); полидиоксанона; поликарбоната; полиаминокарбоната; поли(винилпирролидон)а; поли(этилоксазолин)а; полиуретана; карбоксиметилцеллюлозы; гидроксиалкилированной целлюлозы; полипептида; нуклеиновой кислоты; модифицированной нуклеиновой кислоты; запертой нуклеиновой кислоты; полисахарида; углевода; гепаран-сульфата; хондроитин-сульфата; гепарина, альгината; и их комбинации.
42. Способ по любому из пп. 38-41, где биологически активный компонент представляет собой ткань, клетку, белок, пептид, низкомолекулярное лекарственное средство, нуклеиновую кислоту, инкапсулированную нуклеиновую кислоту (например, инкапсулированную в липидную наночастицу), липид, углевод, или соединение сельскохозяйственного назначения.
43. Способ по п. 42, где биологически активный компонент инкапсулируют в полимерную матрицу биосовместимого сшитого разлагаемого гидрогелевого полимера.
44. Способ по п. 42, где биологически активный компонент ковалентно связывают с полимерной матрицей биосовместимого сшитого разлагаемого гидрогелевого полимера.
45. Способ по п. 44, где биологически активный компонент представляет собой биологически активный полипептид, ковалентно связанный с полимерной матрицей посредством связи на основе простого тиоэфира, образованной в результате реакционного взаимодействия реакционноспособной тиольной группы биологически активного полипептида с ен-группой, где тио-группа связи на основе простого тиоэфира отделена от основной цепи биологически активного полипептида двумя или более атомами углерода.
46. Биосовместимый сшитый разлагаемый гидрогелевый полимер, полученный способом по любому из пп. 35-45.
47. Биосовместимый сшитый разлагаемый гидрогелевый полимер по п. 46, дополнительно содержащий биологически активный компонент, соединенный с полимерной матрицей биосовместимого сшитого разлагаемого гидрогелевого полимера.
