Abstract
1. Способ лечения рака у субъекта, включающий в себя стадию введения субъекту фармацевтически эффективной дозы ингибитора активности E2EPF-UCP (белок-носитель убиквитина). ! 2. Способ по п.1, в котором ингибитор активности UCP уменьшает стабильность HIF. ! 3. Способ по п.1, в котором ингибитор активности UCP ингибирует экспрессию фактора роста эндотелия сосудов (VEGF). ! 4. Способ по любому из пп.1-3, в котором ингибитор активности UCP выбирают из группы, состоящей из антисмыслового олигонуклеотида, комплементарно связывающегося с мРНК UCP, специфичной к гену UCP малой интерферирующей РНК, инактивированного UCP-подобного белка или его фрагмента, связывающегося с UCP пептида, специфичного к UCP антитела, соединения, ингибирующего транскрипцию UCP или трансляцию мРНК UCP, и соединения, ингибирующего функциональные возможности UCP. ! 5. Способ по п.4, в котором малая интерферирующая РНК представлена SEQ. ID. NO: 6 или SEQ. ID. NO: 8. ! 6. Противораковое средство, содержащее ингибитор активности UCP в качестве эффективного ингредиента. ! 7. Claims (31)
1. Способ лечения рака у субъекта, включающий в себя стадию введения субъекту фармацевтически эффективной дозы ингибитора активности E2EPF-UCP (белок-носитель убиквитина).
2. Способ по п.1, в котором ингибитор активности UCP уменьшает стабильность HIF.
3. Способ по п.1, в котором ингибитор активности UCP ингибирует экспрессию фактора роста эндотелия сосудов (VEGF).
4. Способ по любому из пп.1-3, в котором ингибитор активности UCP выбирают из группы, состоящей из антисмыслового олигонуклеотида, комплементарно связывающегося с мРНК UCP, специфичной к гену UCP малой интерферирующей РНК, инактивированного UCP-подобного белка или его фрагмента, связывающегося с UCP пептида, специфичного к UCP антитела, соединения, ингибирующего транскрипцию UCP или трансляцию мРНК UCP, и соединения, ингибирующего функциональные возможности UCP.
5. Способ по п.4, в котором малая интерферирующая РНК представлена SEQ. ID. NO: 6 или SEQ. ID. NO: 8.
6. Противораковое средство, содержащее ингибитор активности UCP в качестве эффективного ингредиента.
7. Противораковое средство по п.6, в котором ингибитор активности UCP выбирают из группы, состоящей из антисмыслового олигонуклеотида, комплементарно связывающегося с мРНК UCP, специфичной к гену UCP малой интерферирующей РНК, инактивированного UCP-подобного белка или его фрагмента, связывающегося с UCP пептида, специфичного к UCP антитела, соединения, ингибирующего транскрипцию UCP или трансляцию мРНК UCP, и соединения, ингибирующего функции UCP.
8. Противораковое средство по п.7, в котором малая интерферирующая РНК представлена SEQ. ID. NO: 6 или ID. NO: 8.
9. Способ лечения ишемических сосудистых заболеваний у субъекта, включающий в себя стадию введения субъекту фармацевтически эффективной дозы усилителя активности UCP.
10. Способ по п.9, в котором усилитель активности UCP увеличивает стабильность HIF.
11. Способ по п.9, в котором усилитель активности UCP увеличивает экспрессию VEGF.
12. Способ по любому из пп.9-11, в котором увеличение активности или уровня UCP опосредовано индуктором экспрессии мРНК UCP, стимулирующим промотор UCP, и плазмидным или вирусным носителем гена, индуцирующим экспрессию UCP.
13. Фармацевтический агент для лечения ишемических сосудистых заболеваний, содержащий терапевтически эффективное количество усилителя активности UCP.
14. Фармацевтический агент по п.13, где усилитель активности UCP представляет собой индуктор экспрессии мРНК UCP, стимулирующий промотор UCP, плазмидный или вирусный носитель гена, индуцирующий экспрессию UCP.
15. Способ скрининга стимулятора ангиогенеза, включающий выбор соединения, усиливающего экспрессию или активность UCP.
16. Способ по п.15, в котором экспрессию UCP определяют путем исследования взаимодействия РНК-РНК, ДНК-ДНК, ДНК-РНК или путем полуколичественной/количественной ПЦР и ПЦР в реальном времени.
17. Способ анализа для диагностики, лечения и прогнозирования рака, включающий в себя стадию измерения экспрессии UCP при помощи одного или нескольких веществ, реагирующих с UCP, в диагностическом образце субъекта.
18. Способ анализа для диагностики, лечения и прогнозирования рака по п.17, в котором одно или несколько веществ, реагирующих с UCP, представляют собой антитела или нуклеиновые кислоты, комплементарные гену UCP.
19. Способ скрининга кандидата на роль противоракового агента, который является ингибитором UCP, включающий в себя выбор соединения, ингибирующего экспрессию или активность UCP.
20. Способ по п.19, в котором экспрессию UCP определяют путем исследования взаимодействия РНК-РНК, ДНК-ДНК, ДНК-РНК или путем полуколичественной/количественной ПЦР и ПЦР в реальном времени.
21. Способ по п.19, в котором активность UCP определяют путем исследования взаимодействия между UCP и соединением.
22. Способ по п.19, в котором активность UCP определяют путем исследования того, ингибирует ли соединение активность белка фон Гиппеля-Линдау (VHL) или фактора, индуцируемого гипоксией, (HIF), или путем исследования того, ингибирует ли соединение убиквитинирование VHL или HIF, когда соединение инкубируют с UCP и VHL или HIF, соответственно.
23. Способ по п.22, в котором активность UCP определяют путем использования клеточной линии, экспрессирующей UCP и HIF или HIF.
24. Диагностический набор для рака, который включает в себя одно или несколько веществ, которые реагируют с UCP.
25. Диагностический набор для рака по п.24, в котором одно или несколько веществ, реагирующих с UCP, представляют собой антитела или нуклеиновые кислоты, комплементарные гену UCP.
26. Способ по п.1, в котором ингибитор активности UCP увеличивает активность VHL.
27. Способ по п.9, в котором усилитель активности UCP уменьшает активность VHL.
28. Способ по п.9, в котором ишемические сосудистые заболевания выбирают из группы, состоящей из критической ишемии конечностей (CLI), неоперабельного заболевания коронарной артерии (CAD), деменции, бокового амиотрофического склероза (ALS), диабетической нейропатии и инсульта.
29. Фармацевтический агент по п.13, где ишемические сосудистые заболевания выбирают из группы, состоящей из критической ишемии конечностей (CLI), неоперабельного заболевания коронарной артерии (CAD), деменции, бокового амиотрофического склероза (ALS), диабетической нейропатии и инсульта.
30. Способ по п.15, в котором активность UCP определяют путем исследования того, ингибирует ли соединение активность VHL или HIF, или путем исследования того, ингибирует ли соединение убиквитинирование VHL или HIF, когда соединение инкубируют с UCP и VHL или HIF, соответственно.
31. Способ по п.15, в котором активность UCP определяют путем использования клеточной линии, экспрессирующей UCP и VHL или HIF.
