Claims (30)
1. Способ лечения человека, страдающего почечноклеточной карциномой, причем функция почек пациента нарушена, предусматривающий
a) введение суточной дозы, составляющей 1-52 ММЕ IL-2, 1-3 дозами в сутки в течение 3-6 суток в неделю, повторяемое в течение 1-24 недель; и
b) отсутствие введения IL-2 в течение 1-4 недель.
2. Способ лечения человека, страдающего почечноклеточной карциномой, причем функция почек пациента нарушена, предусматривающий
a) введение дозы, составляющей 1-52 ММЕ IL-2 через каждые 5 суток - 1 месяц, повторяемое в течение 1-24 недель, причем IL-2 ковалентно конъюгирован с полиэтиленгликолем или с полиоксиэтилированным многоатомным спиртом; и
b) отсутствие введения IL-2 в течение 1-4 недель.
3. Способ лечения человека, страдающего почечноклеточной карциномой, предусматривающий
a) введение суточной дозы, составляющей 9-18 ММЕ IL-2, 1-3 дозами в сутки в течение 3-6 суток в неделю, повторяемое в течение 1-24 недель;
b) отсутствие введения IL-2 в течение 1-4 недель;
c) введение суточной дозы, составляющей 9 ММЕ IL-2, 1-3 дозами в сутки в течение 3-6 суток в неделю, повторяемое в течение 1-24 недель; и
d) отсутствие введения IL-2 в течение 1-4 недель.
4. Способ по п.3, причем способ предусматривает
a) во-первых, введение суточной дозы, составляющей 18 ММЕ IL-2, в течение 5 суток на протяжении одной недели;
b) во-вторых, введение суточной дозы, составляющей 9 ММЕ IL-2, в течение 2 суток с последующим введением суточной дозы, составляющей 18 ММЕ IL-2, в течение 3 суток каждой недели, повторяемые в течение 5 недель;
c) в-третьих, отсутствие введения IL-2 в течение 3 недель;
d) в-четвертых, введение суточной дозы, составляющей 9 ММЕ IL-2, в течение 5 суток каждой недели, повторяемое в течение 6 недель; и
e) в-пятых, отсутствие введения IL-2 в течение 3 недель.
5. Способ по любому из пп.1-3, в котором IL-2 представляет собой рекомбинантно продуцированный IL-2, содержащий аминокислотную последовательность, характеризующуюся, по меньшей мере, 70%-ной идентичностью последовательности по отношению к аминокислотной последовательности IL-2 человека.
6. Способ по п.5, в котором указанный IL-2 содержит аминокислотную последовательность, характеризующуюся, по меньшей мере, 80%-ной идентичностью последовательности по отношению к аминокислотной последовательности IL-2 человека.
7. Способ по п.6, в котором указанный IL-2 содержит аминокислотную последовательность, характеризующуюся, по меньшей мере, 90%-ной идентичностью последовательности по отношению к аминокислотной последовательности IL-2 человека.
8. Способ по п.7, в котором указанный IL-2 содержит аминокислотную последовательность, характеризующуюся, по меньшей мере, 95%-ной идентичностью последовательности по отношению к аминокислотной последовательности IL-2 человека.
9. Способ по любому из пп.1-3, в котором указанный IL-2 представляет собой мутеин IL-2.
10. Способ по п.9, в котором указанный IL-2 представляет собой дез-аланил-1, серин-125 человеческий интерлейкин-2 (алдезлейкин).
11. Способ по п.9, в котором указанный IL-2 выбран из группы, состоящей из Ala104 Ser125 IL-2; дез-Ala1 дез-Pro2 дез-Thr3 дез-Ser4 Ala104 Ser125 IL-2; и дез-Ala1 дез-Pro2 дез-Thr3 дез-Ser4 дез-Ser5 дез-Ser6 IL-2.
12. Способ по п.2, в котором IL-2 конъюгирован с полиэтиленгликолем.
13. Способ по п.12, в котором полиэтиленгликоль характеризуется средней молекулярной массой от 1000 до 40000 Да.
14. Способ по п.12, в котором полиэтиленгликоль характеризуется средней молекулярной массой от 2000 до 20000 Да.
15. Способ по п.12, в котором полиэтиленгликоль характеризуется средней молекулярной массой от 3000 до 12000 Да.
16. Способ по п.2, в котором IL-2 ковалентно конъюгирован с полиоксиэтилированным многоатомным спиртом.
17. Способ по п.16, в котором полиоксиэтилированный многоатомный спирт выбран из группы, состоящей из полиоксиэтилированного сорбита, полиоксиэтилированной глюкозы и полиоксиэтилированного глицерина.
18. Способ по п.17, в котором полиоксиэтилированный многоатомный спирт представляет собой полиоксиэтилированный глицерин.
19. Способ по п.18, в котором полиоксиэтилированный глицерин характеризуется средней молекулярной массой от 1000 до 40000 дальтон.
20. Способ по любому из пп.1-3, в котором указанная почечноклеточная карцинома является метастатической.
21. Способ по п.3, в котором у указанного субъекта проводится множество циклов лечения в течение периода времени, достаточного для достижения, по меньшей мере, частичного опухолевого ответа.
22. Способ по п.3, дополнительно предусматривающий проведение множества циклов лечения, состоящих из
a) введения суточной дозы, составляющей 9 ММЕ IL-2, 1-3 дозами в сутки в течение 3-6 суток в неделю, повторяемое в течение 1-24 недель; и
b) отсутствия введения IL-2 в течение 1-4 недель;
проводимых у указанного субъекта в течение периода времени, достаточного для достижения, по меньшей мере, частичного опухолевого ответа.
23. Способ по п.21 или 22, в котором период времени составляет, по меньшей мере, 6 месяцев.
24. Способ по п.21 или 22, в котором период времени составляет, по меньшей мере, 12 месяцев.
25. Способ по п.21 или 22, в котором достигается полный опухолевый ответ.
26. Способ по любому из пп.1-3, в котором указанный IL-2 вводят путем подкожного, внутрибрюшинного, внутримышечного, внутривенного, перорального, легочного, назального, местного или чрескожного введения или введения с помощью инфузии или суппозиториев.
27. Способ по п.26, в котором указанный IL-2 вводят подкожно.
28. Способ по п.3, в котором функция почек указанного пациента нарушена.
29. Способ по п.3, в котором указанный пациент не переносит лечение высокими дозами IL-2.
30. Применение IL-2 для производства лекарственного препарата для лечения человека, страдающего почечноклеточной карциномой, в соответствии с любым из предшествующих пунктов.
